Det ble nylig vist at ekspresjon av varianter av overflate-glykoproteinet CD44 er nødvendig og tilstrekkelig for å utløse såkalte spontane metastatiske virkninger både i en ikke-metastaserende pankreasadenokarsinom-cellelinje hos rotte og i en ikke-metastaserende fibrosarkom-cellelinje hos rotte (Guntert et al., 1991). Mens den minste CD44 isoform, standardformen CD44s (eller CD44std), eksprimeres ubikvitært i en rekke forskjellige vev, derunder epitelceller, eksprimeres bestemte spleisevarianter av CD44 (CD44v, CD44var) bare på en undergruppe av epitelceller. CD44-variantene dannes ved alternativ spleising, slik at sekvensene fra 10 ekson (vl-vlO) skjæres fullstendig ut i CD44s, men kan forekomme i de større variantene i forskjellige kombinasjoner (Screaton et al., 1992; Tolg et al., 1993; Hofmann et al., 1991). Variantene atskiller seg ved at forskjellige aminosyresekvenser innsettes på et bestemt sted i den ekstracellulære del av proteinet.
Slike varianter kan påvises i forskjellige tumorceller hos mennesker og i tumorvev hos mennesker. Således ble nylig ekspresjonen av CD44-variantene undersøkt i løpet av den kolorektale karsinogenese (Heider et al., 1993a). Ekspresjonen av CD44-variantene mangler i normalt tykktarmsepitel hos mennesker, og bare en svak ekspresjon er påviselig i de formerende celler av krypter. I senere stadium av tumorforløpet, f.eks. i adeno-karsinomer, eksprimerer alle maligne avart varianter av CD44. Vevsekspresjon av variant CD44 på et høyt nivå kunne også påvises i aggresive non-Hodgin-lymfomer (Koopman et
al. 1993).
Antistoffet Var3.1 mot CD44v6 beskrives i The Journal of Cell Biology, Bd. 122, No. 2, July 1993, M. Salmi et al. Det beskrives videre epitoper innenfor aminosyresekvensen som kodes av ekson v6-varianten av CD44 genet.

D. Jackson et al. beskriver også antistoffer mot CD44 ekson v6. Det beskrives videre antistoffer med forbedret biologisk aktivitet i forhold til kjente antistoffer. De siterte antistoffene 2F10 og 8G5 er ikke tilstrekkelig underbygget idet verken epitopen eller fremgangsmåten for fremstilling av de nevnte antistoffene med samme spesifisitet er beskrevet.
Ekson v6 synes å spille en spesiell rolle ved den metastatiske utbredelse (Rudy et al. 1993). I dyremodell kunne antistoffer mot v6-spesifikke epitoper forhindre at metastase-ende celler slår seg ned, og vekst av metastaser (Seiter et al. 1993). I tykktarmskarsinomer korrelerer v6 ekspresjon med tumorprogresjon (Wielenga et al., 1993). I magekarsinomer er v6-ekspresjonen en viktig diagnostisk markør ved differensieringen mellom tumorer av intestinale og slike av diffus type (Heider et al., 1993b). V6-ekspresjonen ble målt i begge de sistnevnte arbeider ved hjelp av antistoffer mot v6 spesifikke epitoper.
Monoklonale antistoffer mot epitoper som kodes for av ekson v6 er tidligere kjent (Hofmann et al., 1991, Wielenga et al., 1993). På grunn av den høye potensielle nytte som slike antistoffer kan ha i diagnose og terapi, består et stort behov for antistoffer med forbedrede egenskaper.
Oppgaven for den foreliggende oppfinnelse var å tilveiebringe et antistoff som har signifikant bedre egenskaper i forhold til de kjente v6-spesifikke antistoffer.
Denne oppgaven kunne løses med den foreliggende oppfinnelse. Foreliggende oppfinnelse vedrører følgelig antistoff mot en epitop innenfor aminosyresekvensen som kodes for av det variable ekson v6 i CD44-genet, kjennetegnet ved at det er antistoffet VFF-18 som dannes av hybridomcellelinjen med deponerings-nummer DSM ACC2174.
Oppfinnelsen vedrører videre en hybridomacellelinje som utskiller disse antistoffer, og som er deponert under deponeringsnumer DSM ACC2174 hos DSM-Deutsche Sammlung fur Mikroorganismen und Zellkulturen GmbH, Mascheroder Weg lb, D-38124 Braunschweig, Tyskland.
Det er videre beskrevet ifølge foreliggende oppfinnelse rekombinant antistoffmolekyl, kjennetegnet ved at det har idiotypen til et antistoff ifølge krav 1.
I det følgende forstås under begrepet antistoffer eller antistoffmolekyler ikke bare fullstendige immunoglobuliner, men også ekvivalente substanser med hensyn til bindingsspesifitet og -affinitet såsom de beskrevne antistoflf-derivater og rekombinanate antistoffmolekyler.
Antistoffet VFF-18 ifølge oppfinnelsen ble fremstilt ved en fremgangsmåte ifølge eksempel 1. Antistoffet kan videre oppnås fra den deponerte hybridomcellelinje. Det ligger innenfor en gjennomsnittsfagmanns kunnskaper å fremstille derivater av antistoffet ifølge oppfinnelsen ut fra en sekvensanalyse av dette antistoff og/eller under anvendelse av hybridom-cellelinjen, som produserer dette antistsoff, å fremstille rekombinante antistoffmolekyler med den samme idiotop, dvs. antistoffmolekyler som har den samme aminosyre-sekvens i området til antigenbindingssetet (complementarity-determining regions, CDR) som antistoffet VFF-18. Slike derivater og rekombinante antistoffer er derfor uttrykkelig omfattet av oppfinnelsen.
Fra det fullstendige immunoglobulin VFF-18 antistoff kan f.eks. Fab- eller F(ab')2-fragmenter eller andre fragmenter fremstilles (Kxeitman et al., 1993). For diagnostiske metoder kan VFF-18-antistoffmolekyler, fragmenter derav eller rekombinante antistoffmolekyler med den samme idiotop, f.eks. knyttes til radioaktive isotoper såsom 1311,111 In, "Tc eller radioaktive forbindelser (Larson et al., 1991; Thomas et al., 1989; Srivastava, 1988) , enzymer såsom peroksidase eller alkalisk fosfatase (Catty et Raykundalia, 1989), med fluorescensfargestoffer (Johnson, 1989) eller biotinmolekyler (Guesdon et al., 1979). For terapeutiske anvendelser kan VFF-18 eller VFF-18-avledede antistoffmolekyler knyttes til toksiner (Vitetta et al., 1991; Vitetta et Thorpe, 1991; Kreitman et al., 1993; Theuer et al., 1993), zytostatika (Schrappe et al., 1992), prodrugs (Wang et al., 1992; Senter et al., 1989) eller radioaktive substanser. Antistoffet kan videre knyttes til et cytokin eller et annet immunomodulatorisk polypeptid, f.eks. til tumornekrosefaktor eller Interleukin-2.
Fagmannen er videre istand til, etter analyse av aminosyresekvensen til antistoffet VFF-18 og/eller under anvendelse av hybridoma-cellelinjen som produserer dette antistoffet, spesielt den deri foreliggende genetisk informasjon, å fremstille rekombinante antistoffmolekyler med den samme idiotype som VFF-18. Tilsvarende fremgangsmåter er kjent teknikk. Slike rekombinante antistoffmolekyler kan f.eks. være humaniserte antistoffer (Shin et al., 1989; Gtissow et Seemann, 1991), bispesifikke antistoffer (Weiner et al., 1993; Goodwin, 1989), single-chain-antistoffer (scFv, Johnson et Bird, 1991), fullstendige eller fragmentariske immunoglobuliner (Coloma et al., 1992; Nesbit et al., 1992; Barbas et al., 1992), eller antistoffer fremstilt ved chain shuffling (Winter et al, 1994). Humaniserte antistoffer kan f.eks. fremstilles ved CDR-poding (EP 0239400). Også rammeverk-regioner kan modifiseres (EP 0519596). For humanisering av antistoffer kan idag metoder såsom PCR (se f.eks. EP 0368684; EP 0438310; WO 9207075) eller computer-modellering ( se f.eks. WO 9222653) anvendes. Det kan også fremstilles fusjonsproteiner, f.eks. single-chain-antistoffer/toksin-fusjonsproteiner (Chaudhary et al., 1990; Friedman et al., 1993). Slike antistoffmolekyler er derfor likeledes en del av oppfinnelsen.
Det ligger i området til gjennomsnittfagmannens kunnskaper å bestemme den nøyaktige epitop av VFF-18, og med disse kjennskaper å fremstille ekvivalente antistoffer med den samme bindingsspesifitet. Dette kan f.eks. gjøres ved peptidbindingsstudier som i Eksempel 2, såsom ved variasjon av peptidet Hul. Slike antistoffer er derfor likeledes en del av oppfinnelsen.
Et videre aspekt av oppfinnelsen er anvendelsen av VFF-18 eller VFF-18-avledede eller ekvivalente antistoffmolekyler i diagnose utenfor legemet til et menneske eller dyr.
Diagnostiske fremgangsmåter kan være i og for seg kjente fremgangsmåter under anvendelse av antistoffmolekylene ifølge oppfinnelsen, f.eks. enzymkoblede immuno-målinger (ELISA, Catty et Raykundalia, 1989), radioimmunoassays (Catty et Murphy, 1989), immunhistokjemiske fremgangsmåter (Heider et al., 1993b) eller Western blots. Slike fremgangsmåter kan hensiktsmessig utføres med vevsprøver eller kroppsvæsker som er tatt ut fra kroppen, f.eks. biopsier. Undersøkelsene kan gjøres kvalitativt, semi-kvantitativt eller kvantitativt. Antistoffet eller antistoffmolekylene kan derunder anvendes som f.eks. beskrevet i den kjente teknikk for andre v6-spesifikke antistoffer (WO9500851), hvorunder de fordelaktige egenskaper hos antistoffet ifølge oppfinnelsen eller antistoffmolekylene ifølge oppfinnelsen betyr en signifikant forbedring av slike fremgangsmåter.
Foreliggende oppfinnelse vedrører videre antistoff eller antistoffmolekyl ifølge ett av kravene 1 eller 3-12, for farmasøytisk anvendelse.
Det er videre beskrevet anvendelse av et antistoff eller antistoffmolekyl ifølge ett av kravene 1 eller 3-11, for fremstilling av et middel for diagnose in vivo.
De overlegne egenskapene til VFF-18 i forhold til andre anti-CD44v6-antistoffer viser eksemplene 2 til 4.

De tre antistoffer VFF-7 (anti v6), VFF-8 (anti-v5) og VFF-18 (anti-v6) ble radioaktivt merket med N-succinimyl[2,3-<3>H] propionat og anvendt for bindingstester på forskjellige tumorcellelinjer. Følgende cellelinjer ble derunder anvendt: CHO-CD44var: rekombinante hamster-cellelinjer (Chinese Hamster Ovary), den humane variant CD44 (Eksons v3-vl0) eksprimert på overflaten; HCT-116, CX-1, HT-29, CaCo, COLO 205: humane tykktarm-karsinomlinjer; A431: human plateepitelkarsinomlinje. Den spesifikke binding av antistoffer på de forskjellige cellelinjer er vist. Mens bindingen av v6-spesifikke antistoffer VFF-7 og VFF-18 til den rekombinante cellelinje CHO-CD44 ligger i den samme størrelsesorden, viser antistoffene på tumorcellelinjene helt forskjellige bindingsforhold. I noen tilfeller observerer man bare for VFF-18 og i mindre omfang VFF-8-binding (HT-29, CaCo, COLO 205), ved de andre cellelinjer binder VFF-18 vesentlig bedre enn VFF-7. Figur 5: Påvisning av løselige CD44-varianter som inneholder eksonet v6 i humant normalserum. I en Sandwich-ELISA ble de tre v6 spesifikke antistoffer VFF-4, VFF-7, henholdsvis VFF-18 anvendt som besjiktingsantistoff. I alle tre tilfeller anvendtes som påvisningsantistoff et CD44std-spesifikt antistoff (BU-52, std = standard) konjugert med peroksidase. Signalet av to forskjellige humane normalsera ved forskjellige fortynninger er vist i disse testene ((A) og (B)). I begge tilfeller observeres med VFF-18 et vesentlig sterkere signal enn med de to andre antistoffene. Figur 6: Serumverdier for CD44-varianter som inneholder v6. Med to forskjellige ELISA'er bestemtes innholdet av løselig CD44-var i sera fra 6 friske givere. I en test ble anvendt VFF-7, i den andre VFF-18 som besiktingsantistoff; begge antistoffer gjenkjenner eksonet v6. Som sammenligningspreparat tjente i begge tilfeller en rekombinant løselig CD44-variant (Ekson v3-vl0) fremstilt i CHO-celler. VFF-18-ELISA'en viser i gjennom-
snitt 3,5 ganger høyere verdier enn for VFF-7-ELISA. Dette betyr at løselige CD44var som forekommer i serum gjenkjennes av VFF-18 bedre enn av VFF-7.
Eksempler
Eksempel 1: Fremstilling av det monoklonale antistoff VFF-18
Kloning av pGEX-fusjonsproteiner
Hele variantregionen av HPKII-typen av CD44v (Hofmann et al, 1991) ble amplifisert fra menneskers keratinozytt-cDNA med polymerase-kjedereaksjon (PCR). De to PCR-primere 5'-CAGGCTGGGAGCCAAATGAAGAAAATG-3', posisjoner 25-52, og 5' -TG AT AAGG AACG ATTG AC ATT AG AGTTGGA-3', posisjoner 1013 - 984 av LCLC97-variantregionen, som beskrives av Hofmann et al., inneholdt et EcoRI-gjen-kjennelsessete, som ble benyttet til å klone PCR-produktet direkte i vektoren pGEX-2T (Smith et al., 1988). Det resulterende konstruksjonsprodukt (pGEX CD44v HPKII, v3-vlO) koder for et fusjonsprotein på tilnærmet 70 kD bestående av glutation-S-transferase fra Schistosoma japonicum og eksons v3-vl0 fra humant CD44 (Fig. 1; Heider et al., 1993a). Fusjonsproteinet ble uttrykt i E. Coli og deretter affinitetsrenset over glutation-agarose (Smith et al., 1988).
Immunisering og screening
Balb/c-hunnmus ble immunisert intraperitonealt med affinitetsrenset fusjonsprotein etter det følgende skjema:
1. Immunisering: 90 jag fusjonsprotein i komplett Freund's adjuvans
2. og 3. Immunisering: 50 u.g fusjonsprotein ufullstendig Freund's adjuvans
Immuniseringene fant sted med 4 ukers mellomrom. 14 dager etter den siste imuni-sering ble dyrene immunisert igjen på 3 påfølgende dager med hver gang 10 ug fusjonsprotein i PBS. På den påfølgende dag ble miltcellene til et dyr med høyt antistoffinnhold fusjonert med P3.X63-Ag8.653-musemyelomceller ved hjelp av polyetylenglykol 4000. Hybridomcellene ble så seleksjonen i mikrotiterplater i HAT-medium (Kohler et Milstein, 1975;Kearneyetal., 1979).
Bestemmelsen av antistoffinnholdet i serum henholdsvis screeningen av hybridom-supernatantene, ble utført ved hjelp av en ELISA. Ved denne testen ble først mikrotiterplater besjiktet med fusjonsprotein (GST-CD44v3-10) eller bare med glutation-S-transferase. Deretter ble det inkubert med seriefortynninger av serumprøver henholdsvis hybridomsupernatanter, og de spesifikke antistoffer med peroksidasekonjugerte antistoffer påvist mot muse-immunoglobulin. Hybridomer som bare reagerte med glutation-S-transferase ble kastet. De gjenværende antistoffer ble først karakterisert i en ELISA med områdespesifikke fusjonsproteiner (Ekson v3, ekson v5 + v6, ekson v6 + v7, ekson v8-vlO) (Koopman et al., 1993). Deres immunhistokjemiske reaktivitet ble testet på menneskehudsnitt. Antistoffet VFF-18 ble så identifisert gjennom binding til det syntetiske peptid Hul (QWFGN RWHEG YRQT). Sekvensen av Hul er et fragment av det humane CD44-eksons v6.
Eksempel 2: Binding av CD44v6-spesifikr antistoff til syntetiske peptider
Bindingen av CD44-spesifikt antistoff til syntetiske peptider ble bestemt i en ELISA. Løsningner:
Besjiktingsbuffer: 0,05 M natriumkarbonat pH 9,6
Målebuffer: PBS (fosfatbuffret saltvann

Peptidene (50 ug/ml i besjiktingsbuffer) ble immobilisert påNUNC Maxisorp immunoplater (1.1ASML) eller Acti-A-plater fra Fabrik Bio Products (VFF-antistoffer) ved 4°C natten over. Ved Acti-A-platene binder peptidet kovalent til platen. Så ble det vasket med PBS/0,05% Tween, frie adsorbsjonsseter på overflaten blokkert med målebuffer (1 time ved romtemperatur) og igjen vasket 1 gang med PBS/0,05% Tween 20. Acti-A-plater ble redusert etter blokkeringen med 10 mmol natriumborhydrid i 20 mmol natriumhydrogenkarbonat, pH 9,0 (rystet 1 time ved romtemperatur), og så vasket 3 ganger med PBS/0,05% Tween 20. Så ble hybridom-supernatanter henholdsvis antistoffløsninger i målebuffer brakt til konsentrasjoner mellom 0,02 og 10,0 ug/ml i glass og inkubert 2 timer ved romtemperatur i en plateryster. Så fulgte tre vasketrinn nmed PBS/0,05% Tween 20. Så ble 100 jxl/glass med pepperrotperoksidase-konjugert antimus-IgG-antistoff tilsatt til målebufferen i en egnet fortynning. Etter 2 timers inkubasjon ved romtemperatur på platerysteren, ble det vasket 3 ganger med PBS/0,05% Tween 20 og farget med substrat-løsning. Etter 10-15 timers utvikling, ble det stoppet med 20 M svovelsyre og absorpsjonen ved 450nm (referanse 690 nm) målt på fotometer.
I et første eksperiment ble bindingen av det for rotte-CD44v6 spesifikke antistoff 1.1ASML til peptidet Ral (KWFEN EWQGK NPPT) påvist. (Tabell 1, Figur 3 (A)):
I et videre forsøk syntetisertes et peptid homologt med Ral fra den humane CD44v6 sekvens (Hul, QWFGN RWHEG YRQT). Forskjellige anti-human-CD44v6-antistoffer ble bundet til Hul. Derunder viser det seg at VFF-18 har en betydelig høyere bindingsaffinitet enn alle andre anvendte antistoffer (Tabell 2, Figur 3(B)).
For kvantitativ eluering ble dessuten rensede anti-human-CDd44v6-antistoffer bundet i forskjellige konsentrasjoner til peptidet Hul. Også her viser det seg den tydelig bedre bindingsaffinitet sammenlignet med de andre antistoffer (Tabell 3, Figur 3 (C)).
Eksempel 3: Binding av radioaktivt markert CD44v6-spesiflkt antistoff til
tumorcellelinjer
Radioaktiv markering av antistoff.
1 mCi N-succinimidyl-[2,3-<3>H]-propionat ([<3>H]-NSP, Amersham 1 mCi/ml) ble inndampet i et silikonisert prøverør ved 0°C på vannstrålevakuum nesten til tørrhet. 15 jag antistoff (1 mg/ml i PBS, pH 7,4) ble tilsatt og inkubert 48 timer ved 4°C. Så ble overskudd [<3>H]-NSP oppfanget ved reaksjon med 30ul IM glysin i PBS (20 min. ved romtemperatur). Separasjonen av det merkede antistoff av [<3>H]-glysin fant sted over en
Sephadex-G-25-M-søyle (søylevolum 15 ml), hvorunder det som eluentbuffer ble anvendt PBS/0,5% BSA. Det [<3>H]-merkede antistoff foreligger i eksklusjonsvolumet. Mengden av antistoff ble bestemt i en ELISA for påvisning av museimmunoglobulin, og den spesifikke aktiviteten utregnet.
Binding av radioaktivt merkede antistoffer til turmoceller.
De tre antistoffer VFF-7 (anti-v6), VFF-8 (anti-v5) og VFF-18 (anti-v6) ble radioaktivt merket med N-suksinimidyl-[2,3-<3>]propionat og anvendt for bindingstesten til forskjellige tumorcellelinjer. Følgende cellelinjer ble derunder anvendt: CHO-CD44var: rekombinant hamstercellelinje (Chinese Hamster Ovary), den humane variant CD44 (eksons v3-vl0) uttrykt på overflaten; HCT-116, CX-1, HT-29, CaCo, COLO 205: humane tykktarmkarsinomlinje; A431: human plateepitelkarsinom-linje.
Cellene ble oppstartet i 12-hulls vevskulturplater, inkubert natten over ved 37°C i CCVvekstskap, deretter vasket en gang med PBS og fiksert med etanol (1 min, ved romtemperatur). Så ble det vasket en gang med dyrkningsmedium (RPMI 1640/10% kalve-fosterserum), og det radioaktive antistoff (250 000 dpm/glass i dyrkningsmedium) bundet. Etter en inkubering på 25 timer ved romtemperatur på plateryster, ble det vasket 3 ganger med PBS/0,5% BSA, cellene solubilisert med 0,1M NaOH/1% Triton X-100, og radio-aktiviteten målt i scintillasjonsteller. Uspesifikk binding ble bestemt i nærvær av et 100 gangers overskudd umerket antistoff. Bindingen ble beregnet i forhold til et enhetlig celletall (400.000 celler). Etter bestemmelsen av den spesifikke aktiviteten til antistoffene, kan den bundne antistoffrnengde uttrykkes i fmol.
Tabell 4 og figur 4 viser den spesifikke binding av antistoffer til de forskjellige cellelinjer. Mens bindingen av det v6-spesifikke antistoff VFF-7 og VFF-18 er tilnærmet lik til den rekombinante cellelinje CHO-CD44var, viser antistoffene til rumorcellelinjene meget forskjellige bindingsforhold. I noen tilfeller observerer man bare VFF-18- og i liten grad VFF-8-binding (HT-29, CaCo, COLO 205), ved de andre cellelinjer binder VFF-18 vesentlig bedre enn VFF-7.

Peroksidase-løsning B (H2O2) 1:1.
Mikrotiterplater (Nunc-Immunoplate MaxiSorp F96) ble besjiktet med 5 u.g/ml av et av de CD44v6-spesifikke antistoffer (inkubasjon ved 4°C natten over), deretter ble det vasket med PBS/0,05% Tween, frie absorbsjonsseter på plateoverflatene blokkert med målebuffer (1 time v/romtemperatur), og igjen vasket en gang med PBS/0,05% Tween 20. Serumprøvene ble så igjen forfortynnet minst 1:5 i prøvefortynningsmiddel og videre-fortynnet i serielle 1:2 fortynninger i platen i prøvefortynningsmiddel. Så tilsattes 50 ul/ glass med pepperrotperoksydasekonjugert anti-CD44std antistoff (Klon BU-52, The Binding Site, Birmingham) i en egnet fortynning (1:3000 - 1:10 000) i målebuffer. Etter 3 timers inkubasjon ved romtemperatur på plateryster, ble det vasket 3 ganger med PBS/0,05% Tween 20 og farget med substratløsning. Etter 10-15 minutter utvikling, ble det avbrutt med 2M svovelsyre, og absorbsjonen ved 450 nm (referanse 690 nm) målt på fotometer.
For kvantifisering, ble en seriell fortynning i målebuffer av et løselig CD44-standardpreparat lagt på parallelt med serumprøvene. Dette preparatet ble renset fra en supernatant av rekombinante hamsterceller (CHO) som uttrykker løselig CD44v3-vl0. Ved CD44v3-vl 0 dreier det seg om en human CD44-variant som inneholder peptid-sekvensene som kodes for av eksonene v3 til v 10.
Tabell 5 og figur 5 viser påvisningen av løselige CD44-varianter som inneholder eksonet v6 i humant normalserum. I en sandwich-ELISA ble de tre v6-spesifikke antistoffer VFF-4, VFF-7 henholdsvis VFF-18 anvendt som beskjiktingsantistoffer. I alle tre tilfeller anvendtes som påvisningsantistoff et CD44-std-spesifikt antistoff (BU-52, std = standard) konjugert med peroksidase. Signalet fra 2 forskjellige human-antisera er vist ved forskjellige fortynninger i denne testen. I begge tilfeller ((A) og (B)) observeres med VFF-18 et vesentlig sterkere signal enn med de to andre antistoffene.
Tabell 6 og figur 6 viser serumverdier fra v6-holdige CD44 varianter. Med to forskjellige ELISA'er bestemtes innholdet av løselig CD44var i sera fra 6 sunne givere. I en test ble VFF-7, i en annen VFF-18 anvendt som besiktningsantistoff; begge antistoffer gjenkjenner ekson v6. Som sammenligningspreparat tjente i begge tilfeller en i CHO-celle fremstilt rekombinant løselig CD44-variant (Ekson v3-vl 0). VFF-18-ELISA viser i gjennomsnitt 3,5 ganger høyere verdier enn VFF-7-ELISA. Dette betyr at det i serum forekommende løselige CD44var gjenkjennes bedre sammenlignet med rekombinant protein av VFF-18 enn av VFF-7.

Claims (17)
1. Antistoff mot en epitop innenfor aminosyresekvensen som kodes for av det variable ekson v6 i CD44-genet, karakterisert ved at det er antistoffet VFF-18 som dannes av hybridomcellelinjen med deponeringsnnummer DSM ACC2174.
2. Hybridom-cellelinje, karakterisert ved at det har deponeringsnummer DSM ACC2174.
3. Antistoffmolekyl, karakterisert ved at det kan oppnås ved kjemisk eller enzymatisk modifikasjon av et antistoff ifølge krav 1.
4. Antistoffmolekyl ifølge krav 3, karakterisert ved at det er et fragment av et antistoff ifølge krav 1.
5. Antistoffmolekyl ifølge krav 4, karakterisert ved at det er et Fab- eller et F(ab'>2-fragment.
6. Antistoffmolekyl ifølge krav 3, karakterisert ved at antistoffet er knyttet sammen med et annet molekyl, hvor det andre molekylet er valgt fra gruppen av radioaktive isotoper, radioaktive forbindelser, enzymer, peroksydase, alkalisk fosfatase, fluorescent farvestoffer, biotinmolekyler, toksiner, cytostatika, cytokiner eller immunmodulatoriske polypeptider, tumomekrosefaktor eller interleukin-2.
