- Die vorliegende Erfindung betrifft die Verbindung 4-(1-Hydroxy-1-methyl-ethyl)furan-2-sulfonamid.
- Die vorliegende Erfindung betrifft ferner ein Verfahren zur Herstellung einer Verbindung, die ausgewählt ist aus:
1-(4-Chlor-2,6-diisopropyl-phenyl)-3-[4-(1-hydroxy-1-methyl-ethyl)-furan-2-sulfonyl]-harnstoff;
1-(8-Chlor-1,2,3,5,6,7-hexahydro-s-indacen-4-yl)-3-[4-(1-hydroxy-1-methyl-ethyl)-furan-2-sulfonyl]-harnstoff;
1-(1,2,3,5,6,7-Hexahydro-s-indacen-4-yl)-3-[4-(1-hydroxy-1-methyl-ethyl)-furan-2-sulfonyl]-harnstoff; und
1-(2,6-Diisopropyl-phenyl)-3-[4-(1-hydroxy-1-methyl-ethyl)-furan-2-sulfonyl]-harnstoff,
wobei das Verfahren die Reaktion der Verbindung 4-(1-Hydroxy-1-methyl-ethyl)furan-2-sulfonamid mit dem entsprechenden Isocyanat umfasst. - Detaillierte Beschreibung der Erfindung
- Das Sulfonamid der Erfindung kann in die entsprechende Sulfonylharnstoffverbindung durch Umsetzung mit einer Isocyanatverbindung in Gegenwart einer Base, wie Natriumhydrid, Natriumhydroxid, Triethylamin oder 1,8-Diazabicyclo[5.4.0]undec-7-en, und eines polaren Lösemittels, wie Tetrahydrofuran, Aceton oder Dimethylformamid, umgewandelt werden. Das auf diese Weise gebildete Reaktionsgemisch wird während einer Zeitspanne zwischen etwa 10 h und etwa 14 h, vorzugsweise etwa 12 h, unter Rückflusskühlung erhitzt.
- Die vorliegende Erfindung wird durch die folgenden Herstellungsbeispiele erläutert, ist jedoch nicht auf die Einzelheiten derselben beschränkt.
- HERSTELLUNGSBEISPIEL A
- 4-Chlor-2,6-diisopropylanilin
- Zu einer gerührten Lösung von 2,6-Diisopropylanilin (47 g) in N,N-Dimethylfomamid (886 ml) wurde N-Chlorsuccinimid (37,3 g) gegeben, und das Gemisch wurde über Nacht gerührt. Die gebildete dunkelrote Lösung wurde in Wasser (12 l) gegossen und mit Diethylether extrahiert. Die vereinigten Etherextrakte wurden mit Wasser und Kochsalzlösung gewaschen, über wasserfreiem Natriumsulfat getrocknet und unter Vakuum eingeengt. Das gebildete dunkelrote Öl wurde durch Filtration über Silicagel unter Elution mit 6 : 1 Hexan/Methylenchlorid gereinigt, wobei 32 g der Titelverbindung erhalten wurden. 1H-NMR δ 7,02 (s, 2), 3,71 (br, s, 2), 2,91 (qq, 2, J = 6,9 Hz), 1,27 (d, 6, J = 6,9 Hz), 1,27 (d, 6, J = 6,9 Hz).
- HERSTELLUNGSBEISPIEL B
- 4-Chlor-2,6-diisopropylphenylisocyanat
- Zu einer gerührten Lösung von 4-Chlor-2,6-diisopropylanilin (32 g) und Triethylamin (7,8 ml) in Tetrahydrofuran (505 ml) wurde Triphosgen (14,9 g) gegeben. Das Gemisch wurde 2 h unter Rühren unter Rückflusskühlung erhitzt. Das Tetrahydrofuran wurden dann unter Vakuum entfernt und das gebildete Öl wurde in Pentan aufgenommen und über Silicagel filtriert, wobei 33,3 g des Produkts erhalten wurden. 1H-NMR δ 7,18 (s, 2), 3,22 (qq, 2, J = 7,1 Hz), 1,25 (d, 6, J = 7,1 Hz), 1,25 (d, 6, J = 7,1 Hz).
- HERSTELLUNGSBEISPIEL C
- 3,5,6,7-Tetrahydro-2H-s-indacen-1-on
- Konzentrierte Schwefelsäure (550 ml) wurde tropfenweise unter Rühren über einen Zeitraum von 2 h zu 137 g 3-Chlor-1-indan-5-yl-propan-1-on gegeben. Die gebildete dicke schwarze Lösung wurde auf 90°C erhitzt, bis die Chlorwasserstoffentwicklung aufhörte (üblicherweise 1–4 h). Das Gemisch wurde dann auf Raumtemperatur gekühlt und auf 5 kg Eis gegossen. Die gebildete Aufschlämmung wurde über Nacht gerührt und filtriert. Der Feststoff wurde mit Wasser gewaschen, bis das Wasser klar durch das Filter lief. Der lohfarbene Feststoff wurde dann unter Vakuum getrocknet und aus Hexan umkristallisiert, wobei 90 g der Titelverbindung erhalten wurden. Fp 72,4–74,8°C.
- HERSTELLUNGSBEISPIEL D
- 1,2,3,5,6,7-Hexahydro-s-indacen
- Ein Gemisch von 3,5,6,7-Tetrahydro-2H-s-indacen-1-on (90 g), Ethanol (1 l), 10% Palladium-auf-Kohle (1–2 g) und konzentrierte Salzsäure (50 ml) wurde auf einem Parr-Schüttler bei Raumtemperatur bis zum Aufhören der Wasserstoffaufnahme hydriert. Das Gemisch wurde über einen Celite-Pfropfen filtriert. Der Pfropfen wurde mit 1 l Diethylether gewaschen. Das Filtrat wurde mit Wasser verdünnt und die organische Phase wurde abgetrennt. Die wässrige Phase wurde mit 1 l Ether extrahiert, und die vereinigten Etherextrakte wurden mit Wasser, gesättigter Natriumbicarbonatlösung und Kochsalzlösung gewaschen. Die Etherextrakte wurden über wasserfreiem Natriumsulfat getrocknet und unter Vakuum eingeengt. Der gebildete blassgelbe Feststoff wurde aus Methanol umkristallisiert, wobei 61 g der Titelverbin dung als farblose Kristalle erhalten wurden. Fp 56,6–58,5°C.
- HERSTELLUNGSBEISPIEL E
- 1-(1,2,3,5,6,7-Hexahydro-s-indacen-4-yl)-ethanon
- Zu einer gerührten Lösung von 1,2,3,5,6,7-Hexahydro-s-indacen (30 g) und Acetylchlorid (14,2 ml) in 120 ml Benzol bei 0°C wurden 30 g Aluminiumchlorid über einen Zeitraum von 1 h gegeben. Das Kühlbad wurde entfernt, und die Lösung wurde auf Raumtemperatur erwärmt und 4 h gerührt. Das tiefrote Gemisch wurde dann auf ein Gemisch von 270 g Eis und 50 ml konzentrierte Salzsäure gegossen. Die Schichten wurden getrennt und die wässrige Phase wurde mit Diethylether extrahiert. Die vereinigten organischen Phasen wurden mit gesättigter Natriumbicarbonatlösung und Kochsalzlösung gewaschen, über wasserfreiem Natriumsulfat getrocknet und unter Vakuum eingeengt, wobei ein orangefarbener Feststoff erhalten wurde, der aus Hexan umkristallisiert wurde, wobei 34 g der Titelverbindung erhalten wurden. Fp 69,1–76,1°C.
- HERSTELLUNGSBEISPIEL F
- 1-(1,2,3,5,6,7-Hexahydro-s-indacen-4-yl)-ethanonoxid
- Ein Gemisch von 1-(1,2,3,5,6,7-Hexahydro-s-indacen-4-yl)-ethanon (33 g), Ethanol (250 ml), Hydroxylaminhydrochlorid (58,5 g) und Pyridin (80 ml) wurde über einen Zeitraum von 12 h unter Rückflusskühlung erhitzt. Das Gemisch wurde dann auf Raumtemperatur gekühlt und unter Vakuum eingeengt. Der Rückstand wurde dann mit 500 ml Wasser behandelt und mit Chloroform-Methanol extrahiert. Die organische Phase wurde über wasserfreiem Natriumsulfat getrocknet und unter Vakuum eingeengt, wobei 32 g der Titelverbindung als Gemisch der E- und Z-Isomere erhalten wurden. 178,6–182,3°C.
- HERSTELLUNGSBEISPIEL G
- 1-(1,2,3,5,6,7-Hexahydro-s-indacen-4-yl)-acetamid
- Ein Gemisch von 1-(1,2,3,5,6,7-Hexahydro-s-indacen-4-yl)-ethanonoxid (85 g) in 270 ml Trifluoressigsäure wurde tropfenweise zu einer gerührten rückfließenden Lösung von 90 ml Trifluoressigsäure über einen Zeitraum von 1/2 h gegeben. Die gebildete purpurfarbene Lösung wurde dann 1 h unter Rückflusskühlung erhitzt. Die Lösung wurde auf Raumtemperatur gekühlt, und die Trifluoressigsäure wurde unter Vakuum entfernt. Der dunkle Feststoff wurde mit Ethylacetat/Hexanen verrieben, wobei 83 g eines grauen Feststoffs erhalten wurden, der ohne weitere Reinigung verwendet wurde. Fp 257,4–259,1°C.

- Eine Aufschlämmung von 1-(1,2,3,5,6,7-Hexahydro-s-indacen-4-yl)-acetamid (110 g) in YY ml 25%-iger Schwefelsäure wurde mit genügend Ethanol, um eine Lösung herzustellen, (etwa YY ml) behandelt. Die gebildete Lösung wurde über einen Zeitraum von 2 Tagen unter Rückflusskühlung erhitzt. Die gebildete schwarze Lösung wurde unter Rückflusskühlung mit Aktivkohle behandelt, heiß filtriert und auf 0°C gekühlt. Die Lösung wurde dann vorsichtig mit 20%-iger Natriumhydroxidlösung neutralisiert. Die gebildete Aufschlämmung wurde dann filtriert und mit Wasser gewaschen, bis das Filtrat neutral durchlief. Der lohfarbene Feststoff wurde dann isoliert und unter Vakuum getrocknet, wobei 80 g der Titelverbindung, Fp 94,5–96,6°C, erhalten wurden, die ohne weitere Reinigung verwendet wurden. Falls notwendig, kann die Titelverbindung aus Methanol umkristallisiert werden, wobei ein weißer Feststoff erhalten wird.
- HERSTELLUNGSBEISPIEL I
- 4-Isocyanato-1,2,3,5,6,7-hexahydro-s-indacen
- Zu einer gerührten Lösung von 1,2,3,5,6,7-Hexahydro-s-indacen-4-yl-amin (77 g) in Tetrahydrofuran (1,5 l) und Triethylamin (68,3 ml) wurde Triphosgen (43,9 g) in einer Portion gegeben. Das Gemisch wurde 1/2 h unter Rückflusskühlung erhitzt und dann auf Raumtemperatur gekühlt. Das Tetrahydrofuran wurde unter vermindertem Druck entfernt, und der Rückstand wurde in Pentan aufgenommen und durch einen Silicagelpfropfen filtriert. Entfernen des Pentans unter Vakuum ergab 80 g eines weißen Feststoffs. Fp 35,0–36,2°C.
- Die Herstellungsbeispiele J und K erläutern allgemeine Verfahren zur Herstellung der Verbindung der Erfindung.
- HERSTELLUNGSBEISPIEL J
- 3-(1-Hydroxy-1-methyl)ethylfuran
- Zu einer gerührten Lösung von 24,97 ml Methylmagnesiumbromid (3 M Lösung in Diethylether) bei 0°C wurden 4,82 ml Ethyl-3-furoat in Diethylether gegeben. Das Gemisch wurde unter Verwendung eines Warmwasserbads 30 min sanft erwärmt. Nach der Beendigung wurde das Gemisch in Eiswasser gegossen, unter Verwendung einer gepufferten Lösung vorsichtig angesäuert und mit Diethylether extrahiert. Die Etherextrakte wurden vereinigt, mit Kochsalzlösung gewaschen, über Natriumsulfat getrocknet und unter Vakuum eingeengt. Der tertiäre Furanalkohol wurde unter Verwendung von Flashsäulenchromatographie mit 6 : 1 Hexan/Ethylacetat gereinigt. Ausbeute: 2,89 g (64%). 1H-NMR (400 MHz, Aceton-d6) δ 1,45 (s, 3H), 1,46 (s, 3H), 3,89 (br s, 1H), 6,45 (br 2, 1H), 7,41 (br s, 1H), 7,42 (br s, 1H).
- HERSTELLUNGSBEISPIEL K
- 2-Aminosulfonyl-3-(1-hydroxy-1-methyl)ethylfuran
- Zu einem gerührten Gemisch von 2,89 g des tertiären Furanalkohols in THF bei –78°C wurden 17,19 ml Methyllithium (1,4 M Lösung in Diethylether) und 5 min später 18,51 ml sek-Butyllithium (1,3 M Lösung in Cyclohexanen) gegeben. Das Gemisch wurde 40 min bei –78°C weitergerührt und mit 5,02 ml flüssigem SO2 versetzt. Die Temperatur wurde 5 min bei –78°C gehalten und dann auf Raumtemperatur erhöht, wobei 2 h weitergerührt wurde. Das THF wurde dann unter Vakuum entfernt und das Lithiumsulfinat wurde in 76,4 ml Wasser gelöst und anschließend mit 7,78 g Hydroxylamin-o-sulfonsäure und 31 g Natriumacetat versetzt. Dieses Gemisch wurde über Nacht bei Raumtemperatur gerührt und mit Ethylacetat extrahiert. Die Ethylacetatextrakte wurden vereinigt, mit Kochsalzlösung gewaschen, über Natriumsulfat getrocknet und unter Vakuum eingeengt. Das Sulfonamid wurde unter Verwendung von Flashsäulenchromatographie mit 2 : 1 Hexan/Ethylacetat gereinigt. Ausbeute: 1,91 g (41%), Fp 110,1–111,6°C.
- BEISPIEL 1
- 4-(Hydroxy-1-methyl-ethyl)furan-2-sulfonamid
- Die Titelverbindung kann nach einem zu dem in Herstellungsbeispiel K beschriebenen analogen Verfahren hergestellt werden.
- Die Verbindung gemäß der Erfindung kann mit Isocyanatderivaten umgesetzt werden, wobei die Sulfonylharnstoffverbindungen der Beispiele 3–6 hergestellt werden. Beispiel 2 erläutert ein allgemeines Verfahren zur Herstellung von Sulfonylharnstoffverbindungen.
- BEISPIEL 2
- 1-(4-Chlor-2,6-diisopropyl-phenyl)-3-[3-(1-hydroxy-1-methylethyl)-benzolsulfonyl]harnstoff
- Zu einer gerührten Lösung von 2-(3-Aminosulfonylphenyl)-propan-2-ol (26,5 g) in Tetrahydrofuran wurde Natriumhydrid (5,2 g einer 60%-igen Dispersion in Mineralöl) in mehreren Portionen gegeben. Sobald die Wasserstoffentwicklung nachgelassen hatte, wurde 4-Chlor-2,6-diisopropylphenylisocyanat (30,8 g) in einer Portion zugegeben, und das gebildete Gemisch wurde 12 h unter Rückflusskühlung erhitzt. Das Gemisch wurde dann auf Raumtemperatur gekühlt und unter Vakuum eingeengt. Der gebildete Schaum wurde in Wasser gelöst, mit 1 N Natriumhydroxid basisch gemacht und mit zwei Portionen von Ether/Hexan im Verhältnis 1 : 1 extrahiert. Die wässrige Schicht wurde mit 1 N Salzsäure angesäuert, und der gebildete weiße Feststoff wurde abfiltriert, mit Wasser gewaschen und getrocknet. Dies ergab 50 g eines weißen Feststoffs, der aus feuchtem Ethylacetat/Hexan umkristallisiert wurde, wobei die Titelverbindung erhalten wurde. Schmelzpunkt 160,5–162,0°C.
- Die Titelverbindungen von Beispiel 3–6 wurden nach einem zu dem in Beispiel 2 beschriebenen analogen Verfahren unter Verwendung der angegebenen Reagentien hergestellt.

- 1-(2,6-Diisopropyl-phenyl)-3-[4-(1-hydroxy-1-methyl-ethyl)-furan-2-sulfonyl]-harnstoff

- 4-(1-Hydroxy-1-methyl-ethyl)-furan-2-sulfonamid; 2,6-Diisopropyl-phenyl-isocyanat.
- Fp: 121,3–126,4°C.

- 1-(1,2,3,5,6,7-Hexahydro-s-indacen-4-yl)-3-[4-(1-hydroxy-1-methyl-ethyl)-furan-2-sulfonyl]-harnstoff

- 4-(1-Hydroxy-1-methyl-ethyl)-furan-2-sulfonamid; 4-Isocyanato-1,2,3,5,6,7-hexahydro-s-indacen.
- Fp: 153,8–154,4°C.

- 1-(8-Chlor-1,2,3,5,6,7-hexahydro-s-indacen-4-yl)-3-[4-(1-hydroxy-1-methyl-ethyl)-furan-2-sulfonyl]-harnstoff

- Fp: 127,4–129,2°C.
Claims (2)
- Die Verbindung 4-(1-Hydroxy-1-methyl-ethyl)-furan-2-sulfonamid und Salze derselben.
- Verfahren zur Herstellung einer Verbindung, die ausgewählt ist aus: 1-(4-Chlor-2,6-diisopropyl-phenyl)-3-[4-(1-hydroxy-1-methyl-ethyl)-furan-2-sulfonyl]-harnstoff; 1-(8-Chlor-1,2,3,5,6,7-hexahydro-s-indacen-4-yl)-3-[4-(1-hydroxy-1-methyl-ethyl)-furan-2-sulfonyl]-harnstoff; 1-(1,2,3,5,6,7-Hexahydro-s-indacen-4-yl)-3-[4-(1-hydroxy-1-methyl-ethyl)-furan-2-sulfonyl]-harnstoff; und 1-(2,6-Diisopropyl-phenyl)-3-[4-(1-hydroxy-1-methyl-ethyl)-furan-2-sulfonyl]-harnstoff, wobei das Verfahren die Reaktion der Verbindung nach Anspruch 1 mit dem entsprechenden Isocyanat umfasst.
