Abstract
1. Растение трансгенной кукурузы, которое имеет геном, включающий последовательность SEQ ID NO:29.2. Семя кукурузы, которое имеет геном, включающий вариант DAS-40278-9 AAD-1, присутствующий в семени, депонированном в Американскую коллекцию типовых культур (АТСС) под номером доступа РТА-10244.3. Семя кукурузы, которое имеет геном, включающий последовательность SEQ ID NO:29.4. Растение кукурузы, полученное в результате выращивания семени по п.2, которое включает последовательность SEQ ID NO:29.5. Потомство растения кукурузы по п.4, включающее вариант DAS-40278-9 AAD-1.6. Потомство растения кукурузы по п.1, устойчивое к гербициду, которое включает последовательность SEQ ID NO:29.7. Растение трансгенной кукурузы, включающее вставку трансгена в хромосомальном сайте-мишени кукурузы, расположенном на хромосоме 2 на расстоянии примерно 20 сМ между маркерами простых повторов последовательности (SSR) UMC1265, частично амплифицируемым последовательностями SEQ ID NO:30 и SEQ ID NO:31, и ММС0111, частично амплифицируемым последовательностями SEQ ID NO:32 и SEQ ID NO:33, при этом указанный сайт-мишень включает гетерологичную нуклеиновую кислоту.8. Claims (26)
1. Растение трансгенной кукурузы, которое имеет геном, включающий последовательность SEQ ID NO:29.
2. Семя кукурузы, которое имеет геном, включающий вариант DAS-40278-9 AAD-1, присутствующий в семени, депонированном в Американскую коллекцию типовых культур (АТСС) под номером доступа РТА-10244.
3. Семя кукурузы, которое имеет геном, включающий последовательность SEQ ID NO:29.
4. Растение кукурузы, полученное в результате выращивания семени по п.2, которое включает последовательность SEQ ID NO:29.
5. Потомство растения кукурузы по п.4, включающее вариант DAS-40278-9 AAD-1.
6. Потомство растения кукурузы по п.1, устойчивое к гербициду, которое включает последовательность SEQ ID NO:29.
7. Растение трансгенной кукурузы, включающее вставку трансгена в хромосомальном сайте-мишени кукурузы, расположенном на хромосоме 2 на расстоянии примерно 20 сМ между маркерами простых повторов последовательности (SSR) UMC1265, частично амплифицируемым последовательностями SEQ ID NO:30 и SEQ ID NO:31, и ММС0111, частично амплифицируемым последовательностями SEQ ID NO:32 и SEQ ID NO:33, при этом указанный сайт-мишень включает гетерологичную нуклеиновую кислоту.
8. Способ получения растения трансгенной кукурузы, который включает инсерцию гетерологичной нуклеиновой кислоты в положение на хромосоме 2 на расстоянии примерно 20 сМ между маркерами простых повторов последовательности (SSR) UMC1265, частично амплифицируемым последовательностями SEQ ID NO:30 и SEQ ID NO:31, и ММС0111, частично амплифицируемым последовательностями SEQ ID NO:32 и SEQ ID NO:33.
9. Часть растения по п.4, выбранная из группы, состоящей из пыльцы, семяпочки, цветков, семенных коробочек, линта, побегов, корней и листьев, которая включает последовательность SEQ ID NO:29.
10. Растение трансгенной кукурузы, включающее вставку трансгена в геномной последовательности или фланкированную геномной последовательностью, выбираемой из группы, состоящей из остатков 1-1873 SEQ ID NO:29 или остатков 6690-8557 SEQ ID NO:29.
11. Выделенная полинуклеотидная молекула, которая включает по меньшей мере 15 нуклеотидов и остается гибридизированной в строгих условиях промывки с последовательностью нуклеиновой кислоты, выбираемой из группы, состоящей из остатков 1-1873 SEQ ID NO:29, остатков 6690-8557 SEQ ID NO:29, и комплементарных им остатков.
12. Выделенный полинуклеотид, который включает нуклеотидную последовательность, выбираемую из группы, состоящей из SEQ ID NO:1-28 и 30-33.
13. Выделенный полинуклеотид, который гибридизируется в строгих условиях промывки с нуклеотидной последовательностью, выбираемой из группы, состоящей из остатков 1863-1875 SEQ ID NO:29, остатков 6679-6700 SEQ ID NO:29, и комплементарных им остатков.
14. Полинуклеотид по п.13, который является ампликоном, полученным при помощи полимеразной цепной реакции.
15. Способ обнаружения варианта кукурузы в образце, содержащем ДНК кукурузы, включающий контактирование указанного образца с по меньшей мере одним полинуклеотидом, позволяющим диагностировать вариант DAS-40278-9 кукурузы AAD-1, присутствующий в семени, депонированном в Американскую коллекцию типовых культур (АТСС) под номером доступа РТА-10244.
16. Способ по п.15, который включает контактирование указанного образца с:
а) первым праймером, который связывается с фланкирующей последовательностью, выбираемой из группы, состоящей из остатков 1-1873 SEQ ID NO:29, остатков 6690-8557 SEQ ID NO:29, и комплементарных им остатков; и
b) вторым праймером, который связывается с последовательностью вставки, включающей остатки 1874-6689 SEQ ID NO:29, или комплементарные им остатки;
выполнение полимеразной цепной реакции указанного образца и анализ ампликона, полученного между указанными праймерами.
17. Способ по п.16, в котором указанные праймеры выбирают из группы, состоящей из SEQ ID NO:1-28.
18. Способ по п.15, в котором указанный полинуклеотид, включающий по меньшей мере 30 нуклеотидов, гибридизируется в строгих условиях с последовательностью, выбираемой из группы, состоящей из остатков 1863-1875 SEQ ID NO:29, остатков 6679-6700 SEQ ID NO:29, и комплементарных им остатков; где указанный способ дополнительно включает гибридизацию в строгих условиях указанного образца и указанного полинуклеотида и анализ указанного образца на предмет гибридизации указанного полинуклеотида с указанной ДНК.
19. Набор для обнаружения ДНК, который включает первый праймер и второй праймер по п.17.
20. Набор для обнаружения ДНК, который предназначен для осуществления способа по п.18.
21. Набор для обнаружения ДНК, который включает полинуклеотид по п.13.
22. Полинуклеотид, содержащий SEQ ID NO:29.
23. Способ получения полинуклеотида по п.22.
24. Способ инсерции трансгена в сегмент ДНК генома кукурузы, в котором указанный сегмент ДНК содержит 5'-конец, включающий нуклеотидные остатки 1-1873 SEQ ID NO:29, и 3'-конец, включающий нуклеотидные остатки 6690-8557 SEQ ID NO:29.
25. Способ селекции растения кукурузы, который включает скрещивание первого растения кукурузы, включающего SEQ ID NO:29, со вторым растением кукурузы с целью получения третьего растения кукурузы, имеющего геном, и анализ указанного третьего растения кукурузы на наличие SEQ ID NO:29 в указанном геноме.
26. Способ интрогрессии признака устойчивости к гербициду в растение кукурузы, который включает скрещивание первого растения кукурузы, включающего SEQ ID NO:29, со вторым растением кукурузы с целью получения третьего растения кукурузы, имеющего геном, и анализ указанного третьего растения кукурузы на наличие SEQ ID NO:29 в указанном геноме.
