Description
Foreliggende oppfinnelse angår et nytt hydrat av 5-[4-[2-(N-metyl-N-{2-pyridyl)amino)etoksy]benzyl]tiazolidin-2,4-dion, maleinsyresalt, en fremgangsmåte for fremstilling og anvendelse derav, samt farmasøytisk preparat inneholdende hydratet.
International Patentsøknad, Publikasjonsnummer WO94/05659 beskriver visse tiazolidindion-derivater som har hypoglycemisk og hypolipidemisk aktivitet omfattende 5-[4-[2-(N-metyl-N-(2-pyridyl)amino)etoksy]benzyl]tiazolidin-2,4-dion, maleinsyresalt (nedenfor også referert til som "Forbindelse (I)").
Forbindelse (I) er beskrevet kun som en vannfri form. Det er nå oppdaget
at Forbindelse (I) foreligger i en ny hydratisert form som er spesielt egnet for bulk-fremstilling og håndtering. Dette kan fremstilles ved en effektiv, økonomisk og reproduserbar prosess spesielt egnet for fremstilling i stor skala.
Det nye hydrat også har nyttige farmasøytiske egenskaper, og spesielt er
det angitt å være nyttig for behandling og/eller forebygging av diabetes mellitus, tilstander forbundet med diabetes mellitus og visse komplikasjoner derav.
Følgelig tilveiebringer foreliggende oppfinnelse et hydrat av 5-[4-[2-(N-metyl-N-(2-pyridyl)amino)etoksy]benzyl]tiazolidin-2,4-dion, maleinsyresalt ("Hydratet")karakterisert vedat Hydratet:
(i) omfatter vann i området fra 0,3 til 0,6 molekvivalenter; og
(ii) gir et infrarødt spektrum inneholdende topper ved 1757,1331,1290,1211
og 767 cm-''; og/eller
(iii) gir et Ramanspektrum inneholdende topper ved 1758,1610,1394,1316 og 1289 cnrr1; og/eller (iv) gir et faststoff-tilstand kjernemagnetisk resonansspektrum inneholdende kjemiske skift hovedsakelig som angitt i Tabell I; og/eller
(v) gir et røntgenpulverdiffraksjon(XRPD)mønster hovedsakelig som angitt i
Figur IV.
Hensiktsmessig er vanninnholdet av Hydratet i området fra 0,3 til 0,5 molekvivalenter, for eksempel 0,4 molekvivalenter
I et foretrukket aspekt gir Hydratet et infrarødt spektrum hovedsakelig i henhold til Figur i.
I et foretrukket aspekt gir Hydratet et Ramanspektrum hovedsakelig i
henhold til Figur II.
I et foretrukket aspekt gir Hydratet et faststoff-ttlstand kjernemagnetisk resonansspektrum hovedsakelig i henhold til Figur III.
Fortrinnsvis gir Hydratet et røntgenpulverdiffraksjons(XRPD)mønster hovedsakelig som vist på Figur IV.
Hydratet kan foreligge i visse dehydratiserte former som reversibelt omdannes til Hydratet når det kommer i kontakt med vann, enten i væske- eller dampform. Foreliggende oppfinnelse omfatter alle slike reversibelt rehydratiserbare former av Hydratet.
Foreliggende oppfinnelse omfatter Hydratet isolert i ren form, eller når det er blandet med andre materialer, for eksempel den kjente vannfri form av Forbindelse I, ovennevnte reversibelt rehydratiserbare former eller hvilken som helst andre materialer.
Således tilveiebringes i ett aspekt Hydratet i isolert form.
I et ytterligere aspekt tilveiebringes Hydratet i ren form.
I enda et ytterligere aspekt tilveiebringes Hydratet i krystallinsk form.
Oppfinnelsen tilveiebringer også en fremgangsmåte for fremstilling av Hydratet,karakterisert vedat 5-[4-[2-(N-metyl-N-(2-pyridyl)amino)etoksy]benzyl]-tiazolidin-2,4-dion, maleinsyresalt blir krystallisert fra vandig etanol, hensiktsmessig vandig denaturert etanol.
Hensiktsmessig inneholder det vandige etanol fra 2% til 15 volum % vann, så som 5% til 15volum%, fortrinnsvis 7% til 12 volum% vann, helst 10% til 12 volum%, for eksempel 10%.
Andre vandige løsningsmidler kan også anvendes for å fremstille Hydratet, for eksempel isopropanol, acetonitril, tetrahydrofuran, metyletylketon, etylacetat eller eddiksyre eller blandinger derav. Den nøyaktige mengden vann anvendt i hver av de alternative løsningsmidler vil avhenge av det spesielle løsningsmiddel valgt, men typisk er det i området fra 2 til 15 volum% vann, for eksempel 3%. For visse løsningsmidler så som etylacetat, kan vannnivåer så lave som 1 volum% frembringe Hydratet (frembringe således et egnet område på 1 til 15 volum% vann i det egnete løsningsmiddel). Alternativt kan Hydratet oppnås ved krystallisering fra vann inneholdende en liten mengde (for eksempel 2 til 5 volum%) av en organisk syre så som eddiksyre.
Krystallisasjon og enhver omkrystallisering blir generelt utført ved lav temperatur til omgivelsestemperatur, så som i området mellom 0 til 30°C for eksempel 25°C; alternativt kan krystallisasjon igangsettes ved en forhøyet temperatur, så som i området mellom 30°C og 60°C, for eksempel 50°C og deretter fullføres ved å la temperaturen i løsningsmidlet avkjøles til omgivelses-eller lav temperatur, så som i området mellom 0 til 30°C, for eksempel 20°C.
Krystallisasjonen kan igangsettes ved poding med krystaller av Hydratet, men dette er ikke essensielt.
Forbindelse I blir fremstilt i henhold til kjente prosedyrer, så som de som er beskrevet i WO94/05659. Innholdet i WO94/05659 er inntatt her som referanse.
Når betegnelsen 'forebygging av tilstander forbundet med diabetes mellitus' anvendes her, omfattes behandlingen av tilstander så som insulinresistens, redusert glukosetoleranse, hyperinsulinaemi og gestasjonell diabetes.
Diabetes mellitus betyr fortrinnsvis Type II diabetes mellitus.
Tilstander forbundet med diabetes omfatter hyperglykemi og insulinresistens, spesielt ervervet insulinresistens og fedme. Ytterligere tilstander forbundet med diabetes omfatter hypertensjon, kardiovaskulær sykdom, spesielt aterosklerose! visse spiseforstyrrelser, spesielt reguleringen av appetitt og matinntak hos individer som har lidelser forbundet med for lite spising ,så som anoreksi nervosa og lidelser forbundet med for mye spising, så som fedme og anorektisk bulimi. Ytterligere tilstander forbundet med diabetes omfatter polycystisk ovariesyndrom og steroidfremkalt insulinresistens.
Komplikasjoner av tilstander forbundet med diabetes mellitus omfatter her nyresykdom, spesielt nyresykdom forbundet med utviklingen av Type II diabetes omfattende diabetisk nefropati, glomerulonefritt, glomerulær sklerose, nefrotisk syndrom, hypertensiv neurosklerose og sluttstadium-nyresykdom.
Som anvendt her angir 'vandig' med referanse til et gitt løsningsmiddel eller løsningsmiddelblanding et løsningsmiddel som inneholder tilstrekkelig vann til å gi Hydrat, dvs. som har fra 0,3 til 0,6 molekvivalenter vann.
Som nevnt ovenfor har forbindelsen ifølge foreliggende oppfinnelse nyttige terapeutiske egenskaper: Foreliggende oppfinnelse vedrører følgelig Hydratet for anvendelse som en aktiv terapeutisk substans.
Mer spesielt vedrører foreliggende oppfinnelse Hydratet for anvendelse i behandling og/eller forebygging av diabetes mellitus, tilstander forbundet med diabetes mellitus og visse komplikasjoner derav.
Hydratet kan administreres som sådant, eller fortrinnsvis som et farmasøytisk preparat som også omfatter en farmasøytisk akseptabel bærer. Preparatet av Hydratet og doser derav er generelt som beskrevet for Forbindelse (I) i Internasjonal Patentsøknad, Publikasjonsnummer WO94/05659.
Følgelig vedrører foreliggende oppfinnelse også et farmasøytisk preparat omfattende Hydratet og en farmasøytisk akseptabel bærer for dette.
Hydratet administreres normalt i enhetsdoseform.
Den aktive forbindelse kan administreres ad hvilken som helst egnet vei, men gis vanligvis ad oral eller parenteral vei. For slik anvendelse vil forbindelsen normalt anvendes i form av et farmasøytisk preparat sammen med en farmasøytisk bærer, fortynningsmiddel og/eller tilsetningsmiddel, selv om den nøyaktige form av preparatet naturlig vil avhenge av administreringsmetoden.
Preparater blir fremstilt ved blanding, og blir hensiktsmessig tilpasset for oral, parenteral eller topisk administrering og kan som sådanne foreligge i form av tabletter, kapsler, orale flytende preparater, pulvere, granulater, pastiller, rekonstituerbare pulvere, injiserbare og infuserbare løsninger eller suspensjoner, suppositorier og transdermale anordninger. Oralt administrerbare preparater er foretrukket, spesielt formede orale preparater, ettersom de er mer hensiktsmessige for generell anvendelse.
Tabletter og kapsler for oral administrering blir vanligvis presentert i en enhetsdose og inneholder konvensjonelle tilsetningsmidler så som bindemidler, fyllmidler, fortynningsmidler, tabletteringsmidler, smøremidler, sprengingsmidler, fargemidler, smakstilsetninger og fuktemidler. Tablettene kan belegges i henhold til velkjente metoder på området.
Egnete fyllmidler for anvendelse omfatter cellulose, mannitol, laktose og andre lignende midler. Egnete sprengningsmidler omfatter stivelse, polyvinylpyrrolidon og stivelsesderivater så som natriumstivelsesglykolat. Egnete smøremidler omfatter for eksempel magnesiumstearat. Egnete farmasøytiske akseptable fuktemidler omfatter natriumlaurylsulfat.
Faststoff orale preparater kan fremstilles ved konvensjonelle blandingsmetoder, fylling, tablettering eller lignende. Gjentatte blandingsoperasjoner kan anvendes for å fordele det aktive midlet gjennom preparatene ved anvendelse av store mengder av fyllmidler. Slik operasjoner er selvfølgelig konvensjonelle på området.
Orale flytende preparater kan foreligge i form av for eksempel vandige eller oljeaktige suspensjoner, løsninger, emulsjoner, siruper eller eliksirer, eller kan presenteres som et tørt produkt for rekonstituering med vann eller andre egnete konstituenter før anvendelse. Slike flytende preparater kan inneholde konvensjonelle additiver så som suspenderingsmidler, for eksempel sorbitol, sirup, metylcellulose, gelatin, hydroksyetylcellulose, karboksymetylcellulose, aluminiumstearatgel eller hydrogenert spiselig fett, emulgeringsmidler, for eksempel lecitin, sorbitan-monooleat eller akasie; ikke-vandige konstituenter (som kan omfatte spiselige oljer), for eksempel mandelolje, fraksjonert kokosnøttolje, oljeaktige estere så som estere av glycerol, propylenglykol eller etylalkohol; konserveringsmidler, for eksempel metyl- eller propyl-p_-hydroksybenzoat eller sorbinsyre og om ønsket konvensjonelle smaksmidler eller fargemidler.
For parenteral administrering blir fluide enhetsdoseformer blir fremstilt inneholdende en forbindelse ifølge foreliggende oppfinnelse og en steril bærer. Forbindelsen kan avhengig av konstituenten og konsentrasjonen foreligge enten oppslemmet eller oppløst. Parenterale løsninger blir normalt fremstilt ved oppløsning av den aktive forbindelse i en konstituent og flItersterilisering før fylling i et egnet medisinglass eller ampulle og tetting. Fordelaktig er tilsetningsmidler så som et lokal-anaestetikum, konserveringsmidler og buffermidler også løst i konstituenten. For å forbedre stabiliteten, kan preparatet fryses etter fylling i medisinglasset og vannet fjernes under vakuum.
Parenterale suspensjoner blir fremstilt på i det vesentlige samme måte, bortsett fra at den aktive forbindelse blir oppslemmet i konstituenten istedenfor å bli løst og sterilisert ved eksponering for etylenoksyd før oppslemming i den sterile konstituent. Fordelaktig inngår et overflateaktivt middel eller fuktemiddel i preparatet for å lette jevn fordeling av den aktive forbindelse.
I tillegg kan slike preparater inneholde ytterligere aktive midler så som anti-hypertensive midler og diuretika.
Som vanlig praksis er, vil preparatene vanligvis være fulgt av skrevne eller trykte retningslinjer for anvendelse i den aktuelle medisinske behandling.
Som anvendt her omfatter betegnelsen 'farmasøytisk godtagbare' forbindelser, preparater og bestanddeler for både human og veterinærmedisinsk anvendelse: for eksempel omfatter betegnelsen 'farmasøytisk godtagbare salt' et veterinærmedisinsk akseptabelt salt.
Hensiktsmessig kan den aktive bestanddel administreres som et farmasøytisk preparat definert ovenfor, og dette farmasøytiske preparat utgjør et spesielt aspekt ved foreliggende oppfinnelse.
Ved behandling og/eller forebygging av diabetes mellitus, tilstander forbundet med diabetes mellitus og visse komplikasjoner derav, kan Hydrat tas i doser, så som dem beskrevet ovenfor.
Lignende doseringsopplegg er egnet for behandling og/eller forebygging av andre pattedyr.
I et ytterligere aspekt tilveiebringer foreliggende oppfinnelse anvendelse av Hydrat for fremstilling av et medikament for behandling og/eller forebygging av diabetes mellitus, tilstander forbundet med diabetes mellitus og visse komplikasjoner derav.
Ingen ugunstige toksikologiske effekter er påvist ved ovennevnte behandlinger med forbindelsene ifølge foreliggende oppfinnelse.
De følgende eksempler illustrerer oppfinnelsen, men begrenser den ikke på noen som helst måte.
Eksempel 1: Fremstilling av Hydrat av 5-[4-[2-(N-metyl-N-(2-pyridyl)amino)-etoksy]benzyl]tiazolidin-2,4-dion, maleinsyresalt.
5-[4-[2-(N-metyl-N-(2-pyridyl)amino)etoksy]benzyll tiazolidin-2,4-dion fri base (6,0
g) og maleinsyre (2,1 g) ble oppvarmet til 60 °C i denaturert etenol (140 ml)
inneholdende ytterligere vann (6,1 ml, dvs. et totalt vanninnhold på ca. 10 volum-%) og rørt ved denne temperaturen i 30 min., hvorunder en løsning ble oppnådd. Den resulterende løsning ble filtrert, gjenoppvarmet til 55°C og så avkjølt til 20-25° C og rørt i 18 timer. Produktet ble filtrert og tørket ved 50°C i vakuum, hvilket ga tittelforbindelsen (4,62 g, 58%).
KARAKTERISERENDE DATA: De følgende karakteriserende data ble opnådd for Hydratet:
A Vanninnhold
Dette ble bestemt til 1,55 vekt % (0,41 molekvivalenter) ved anvendelse av et Karl Fischer apparat.
B Infrarød
Det infrarøde absorpsjonspektrum av en mineraloljedispersjon av Hydratet ble oppnådd ved anvendelse av et Nicolet 710 FT-IR spektrometer ved 2 cm-'' spaltning. Data ble digitalisert ved 1 cm-<1>intervaller. Spektret oppnådd er vist i

Et Ramanspektrum av Hydratet ble registrert gjennom glass-medisinglass ved anvendelse av et Perkin Eimer 2000R spektrometer ved 4 cm-<1>spaltning og er vist i Figur II (1800 - 200 cm-<1>). Eksitasjon ble oppnådd ved anvendelse av en Nd:YAG laser (1064 nm) med en ytelse på 500 mW. Topp-posisjoner er som følger. 1758,1743,1703,1610,1586,1544,1468,1435,1394,1330,1316,1289, 1265, 1238, 1206, 1185, 1148, 1095, 1032, 1003, 976, 923, 903, 843, 825, 780, 741, 722, 664, 637, 606, 526, 471, 403, 331 og 293 cm-<1>.
D NMR
90,55MHz<13>C CP-MAS NMR spektret for Hydratet er vist nedenfor i Figur III. Kjemiske skift er tabulert i Tabell I. Data ble registrert ved omgivelsestemperatur og 10 kHz spinrifrekvens uten tidligere maling av prøven på et Bruker AMX360WB spektrometer, med 1,6ms krysspolarisering og en gjentagelseshastighet på 20 s. Kjemiske skift var referert til høy-feltsresonans av fast adamantan (38,4 ppm i forhold til tetrametylsilan) og anses som nøyaktig innenfor +/- 0,5 ppm. Topper ble ikke tilegnet.
E Røntgen pul verdiffraksjon (XRPD)
XRPD mønsteret til Hydratet er vist nedenfor i Figur IV og en sammenfatning av XRPD vinklene og beregnete gitteravstandskarakteristika for Hydratet er angitt i Tabell II.
Et PW1710 røntgenpulverdiffraktometer (Cu røntgenkilde) ble anvendt for å oppnå spektret ved anvendelse av de følgende innstillinger og betingelser:

Eksempel 2
Maleatsaltet av 5-t4-[2-(N-metyl-N-(2-pyridyl)amino)etoksy]ben2yl]tiazolidin-2,4-dion anhydrat (3,0 g) ble rørt og oppvarmet ved 55-60 °C i acetonitril (30 ml) inneholdende vann (1 ml) inntil fullstendig oppløsning ble oppnådd. Den resulterende løsning ble rørt og avkjølt til 20-25 °C og produktet ble filtrert, vasket med acetonitril (5 ml) og tørket ved 50 °C i vakuum, hvilket ga tittelforbindelsen (1,8 g, 60%). Vann- innholdet i produktet var 1,77%.
Eksempel 3
Metoden ifølge Eksempel 2 ble gjentatt ved anvendelse av tetrahydrofuran (15 ml) inneholdende vann (0,5 ml) som løsningsmiddel. Utbytte 1,8 g (60%), vanninnhold 1,60%.
Eksempel 4
Metoden ifølge Eksempel 2 ble gjentatt ved anvendelse av metyletylketon (30 ml) inneholdende vann (1 ml) som løsningsmiddel. Utbytte 2,05 g (68 %), vanninnhold 1,58%.
Eksempel 5
Metoden ifølge Eksempel 2 ble fulgt ved anvendelse av 2,0 g maleatsalt, oppvarmning til 65 °C i etylacetat (150 ml) inneholdende vann (1,5 ml) som løsningsmiddel. Utbytte 1,34 g (67 %). vanninnhold 1,61 %.
Eksempel 6
Metoden ifølge Eksempel 2 ble fulgt, oppvarming til 65-70 °C i isopropanol (33 ml) inneholdende vann (1 ml) som løsningsmiddel. Utbytte 2,4 g (80 %). Vanninnhold 1,58%.
Eksempel 7
Metoden ifølge Eksempel 2 ble gjentatt ved anvendelse av en blanding av vann (20 ml) og eddiksyre (1,0 g) som løsningsmiddel. Utbytte 0,76 g (38 %). Vanninnhold 1,78 %.
Claims (13)
1. Hydrat av 5-[4-[2-(N-metyl-N-(2-pyridyl)amino)etoksy]benzyl]tiazolidin-2,4-dion, maleinsyresalt,karakterisert vedat hydratet (i) har et vanninnhold fra 0,3 til 0,6 molekvivalenter: og videre erkarakterisertved minst ett av (ii) et infrarødt spektrum inneholdende topper ved 1757,1331.1290,1211 og 767 cm"<1>; (iii) et Ramanspektrum inneholdende topper ved 1758,1610,1394.1316 og 1289 cm-<1>;(iv) et faststoff-tilstand kjernemagnetisk resonansspektrum inneholdende kjemiske skift hovedsakelig som angitt i Tabell I; (v) et røntgenpulverdiffraksjon(XRPD)mønster hovedsakelig som angitt i Figur IV.
