Description

본 발명의 백신 제제

본 발명의 백신이 피부에, 보다 구체적으로는 진피에 투여될 경우, 백신은 낮은 액체 부피, 특히 약 0.05 ml 내지 0.2 ml로 사용된다.

C3H/HeJ 마우스 (6 내지 8주령)를 50 ㎕ CFA로 어쥬번트 처리된 15 ㎍ 단백질의 피하투여로 면역처리한 후, 3-4주 후에 IFA와 함께 15 ㎍ 단백질로 부스터 처리할 수 있다. 전신계 감염으로부터 수동 및 능동 보호를 위해 마우스에 대해 8 내지 10주차에 LD50이 15 내지 90인 폐렴구균의 복강내 주사에 의한 시험 전에 면역 혈청 또는 단백질을 복강내 투여할 수 있다. 추가로, 단백질은 마우스 비인두 군집 모델 (Wu et al Mcrobial Pathogenesis 1997; 23:127-137)에서 시험할 수 있다.
마우스 이외에, 유아 래트는 에스. 뉴모니애의 군집화 및 감염에 취약하다. 수동 보호 연구에서, 마우스 면역 혈청 (100 ㎕ i.p. 또는 1O ㎕ i.n.)을 2 내지 5일령의 유아 래트에게 에스. 뉴모니애 (10 ㎕) 비내 투여에 의한 시험 전에 투여할 수 있다. 군집화는 비내 세척물 (20-40 ㎕ 점적, 10 ㎕ 회수)을 도말하여 결정할 수 있다.
조합 백신의 단백질 성분 사이의 유리한 상호작용은 1가 백신에서는 그 보호 효과가 일정 수준 미만인 각 단백질의 일정 투여량을 투여하여 입증할 수 있다. 1가 백신보다 증가한 조합 백신의 보호 효능은 각 성분간의 유리한 상호작용에 의한 것일 수 있다.
본 발명은 하기 실시예에서 예시된다. 실시예는 다른 설명이 없으면 당업계에 공지된 통상의 표준 기술을 사용하여 수행된다. 하기 실시예는 본 발명을 예시 하고자 한 것으로서 본 발명을 한정하는 것이 아니다
본 발명은 폐렴구균 단백질을 기초로 한 방법에 의해 노년층의 폐렴구균성 감염 (예를 들어 폐렴) 및(또는) 유아의 폐렴구균성 감염 (예를 들어 중이염)의 예방 또는 치료를 위한 개선된 백신을 제공한다. 한 바람직한 실시태양에서, 백신은 노년층의 폐렴구균성 감염의 예방 또는 치료에 적합하다.

실시예
1. 항원의 제조 및 발현
NR1xR2
CbpA는 수개의 도메인으로 구성된 75 kDa의 표면 노출 단백질이다. N-말단 도메인은 2개의 고보존 반복체 (R1 및 R2)를 포함하고, C-말단 도메인은 10개의 탠덤(tandem)한 20개 아미노산의 직접 반복 서열을 포함한다. CbpA 트런케이트는 콜린 결합 도메인이 없는 NR1xR2를 제조하기 위해 제조하였다.
NR1xR2 유전자는 에스. 뉴모니애의 혈청형 4의 균주 (예를 들어 W097/41157 또는 W099/51266 참조)로부터 입수한 DNA로부터 PCR에 의해 증폭하였다. PCR은 Expand High Fidelity PCR System 또는 Hi-Fi (Roche)를 사용하여 수행하였다. 이것은 Taq 폴리머라제 및 프루프리딩(proofreading) 폴리머라제의 혼합물을 포함한다. 고유의 3'-5' 엑소뉴클레아제 프루프리딩 활성 때문에, Hi-Fi를 사용하면 Taq 폴리머라제에 비해 DNA 합성 신뢰도가 3배 증가한다.
PCR 단편을 pGEM-T Vector Systems (Promega)의 pGEM-T 벡터에 클로닝하였다. 상기 단계는 추후 라이게이션을 위한 PCR 단편의 제한 효소에 의한 분해를 촉진시키기 위해 필요하다. pGEM-T 벡터는 선형으로 제공되고, 3'-T 오버행(overhang)을 포함한다. 이들 오버행은 증폭된 단편의 3' 말단에 주형 독립적인 방식으로 단일 데옥시아데노신을 부가하는 열안정성 폴리머라제에 의해 생성되는 PCR 산물의 삽입을 촉진시킨다.
단편 및 벡터는 문헌 [Benore-Parsons et al, Nucleic Acids research, 23, 4926-4927, 1995]에 따라 효소에 의한 절단 (NdeI 및 XbaI 분해) 후에 정제하였다. 아가로스 부분은 3 내지 4시간 동안 완전히 동결건조시켰다. 1:1 에탄올-TE 용액을 동결건조된 겔에 첨가하였다. 시료를 1시간 동안 부드럽게 혼합하고, 아가로스를 압축한 후 원심분리에 의해 완전히 제거하였다. DNA는 에탄올 침전에 의해 용출액으로부터 회수하였다.
NR1xR2를 코딩하는 DNA를 파지 λ로부터의 긴 프로모터 L을 포함하는 벡터에 클로닝하였다. 목적 단백질은 AR58 이. 콜라이 균주에 존재할 경우에는 열에 의해, AR120 이. 콜라이 균주 내에서는 날리딕산에 의해 유도될 수 있다.
세균의 예비 배양은 30 ℃에서 철야 수행하였다. 상기 예비 배양액을 총 20 ml의 부피로 약 40배 희석하여 O.D.가 0.4 내지 0.6이 될 때까지 30 ℃에서 정치시켰다. 42 ℃에서 열 유도시켰다. 시료를 상이한 시점에서 채취하였다. 1 ml의 배양액을 7000 rpm에서 5분 동안 원심분리하였다. 배양 상등액을 -20 ℃에서 보관하고, 펠렛 (전체 추출물)을 500 ㎕의 시료 완충액 (웨스턴 블롯 또는 SDS-PAGE 분 석) 또는 500 ㎕의 용해 완충액에 현탁시켜 37 ℃에서 30분간 인큐베이션시켰다 (ELISA). 용해 완충액의 조성은 SDS 0.1%, 데옥시콜레이트 0.1%, Na 시트레이트 0.015 M이다.
시료를 4-20% 겔 상에 로딩하여 SDS-PAGE (Novex, Invitrogen)에서 이동시켰다. 200 V에서 이동시켰다. 쿠마시 블루 (Coomassie blue) 염색을 수행하였다. 시료를 웨스턴 블로팅을 위해 4-20% 겔 (Novex, Invitrogen)에 로딩하였다. 200 V에서 이동시켰다. 겔을 니트로셀룰로스에 이송하고, 토끼 α-NR1xR2 폴리클로날 항체 (제1 항체) 및 알칼린-포스파타제에 커플링된 α-토끼 항체 (2차 항체)를 사용하여 반점을 가시화시켰다.
약 55 kDa의 밴드가 SDS-PAGE 분석에서 관찰되었다. SDS-PAGE 분석을 기초로 하여 클론 28B2를 선택하고 발효를 위해 이송하였다. 상기 클론의 서열을 결정하고, 그 서열(아미노산 39 (즉, 시그날 서열 이후) 내지 446 = 406개 아미노산)을 확인하였다.
철야 유도된 세균의 용해 및 원심분리 후에 NR1xR2의 용해도를 연구하였다. SDS-PAGE 분석 및 ELISA 시험을 수행하였다. NR1xR2는 주로 가용성 분획으로 회수되는 것으로 나타났다 (> 95%).

또한, 상기 유전자를 클로닝하고, 서열을 결정하여 NR1xR2와 유사한 방법으로 발현시켰다. R1xR2는 CbpA (에스. 뉴모니애 혈청형 4N의)의 아미노산 177 내지 443를 포함하고, PhtD는 아미노산 21 (즉, 시그날 서열 다음의) 내지 최종 아미노 산 (에스. 뉴모니애 혈청형 4N의 아미노산 839)를 포함하고, Sp91은 아미노산 20 (VAA) 내지 최종 아미노산을 포함한다. 융합 단백질의 경우, R1xR2-Sp91 Cterm은 CbpA의 아미노산 177-446를 포함하고, 번역이 정지될 때까지 271개의 아미노산을 포함하고, NR1xR2-Sp91Cterm은 CbpA의 아미노산 39-446를 포함하고, 번역이 정지될 때까지 271개의 아미노산을 포함한다. 두 융합 단백질의 경우, 2개의 추가의 아미노산 (GS)가 (N)R1xR2과 Sp91Cterm 서열 사이에서 발견된다. 모든 구조체에서, ATG는 전사 및 번역을 가능하게 하기 위해 유전자의 5'에 도입되고, 이것은 상기 언급한 각 서열의 앞에 추가의 N-말단 메티오닌이 존재함을 의미한다.
실시예
2. 혈청학
임상 연구에서 사용된 혈청을 사용하여, 에스. 뉴모니애 단백질에 대해 자연발생한 항체 반응을 ELISA로 평가하였다.
2.1 실험 방법
ㆍ 혈청 시료
- 각각 2 내지 4개월 및 6 내지 12개월의 유아로부터 수거한 1쌍의 혈청 (N = 20, DTPa HBV 연구).
- 20세 이하의 성인 혈청 (N = 50).
- 65세 이상의 성인 혈청 (N = 140).
ㆍ ELISA 과정
면역-플레이트를 1 ㎍/ml의 각 단백질로 4 ℃에서 철야 코팅하였다. 혈청의 연속적인 2배 희석액 (1/10 희석액으로 출발)을 교반 하에 실온에서 1시간 동안 인 큐베이션하였다. 면역-검출은 4000배 희석된 퍼옥시다제-커플링된 항-인간 IgG 모노클로날 항체 (Strateck, HP6043)을 사용하여 수행하고, 교반 하에 실온에서 30분 동안 인큐베이션하였다. 가시화 후에, 중앙 역가를 SoftMaxPro로 계산하였다. 역가가 10 이상인 혈청은 양성인 것으로 간주하였다. 기하 평균 계산을 위해, 역가 5 (컷 오프의 1/2)를 임의로 음성 혈청으로 분류하였다.
㎍/ml로 표시한 IgG 농도는 폴리클로날 항-인간 IgG 염소 항체에 의해 플레이트에 포획되고 상기한 바와 동일한 퍼옥시다제 표지된 항체에 의해 가시화된 chromopure IgG (Jackson)의 광학 밀도 (OD) 곡선에 시료 OD를 비교하여 확립하였다.
2.2 결과
2.2.1 유아의 스트렙토코커스 단백질 혈청학
2 내지 4개월의 유아의 혈청에서 측정된 가장 높은 항체 역가 및 혈청 반응 양성 비율은 PhtD, PsaA, Sp128, NR1xR2에서 얻어졌고, 이보다 약간 낮은 수치가 Sp91 및 Ply에서 얻어졌다. Sp1O1 및 Sp13O에 대한 반응은 없거나 낮게 검출되었다. Sp46 및 PhtA는 시험되지 않았다 (물질 입수 문제).
*항체 반응은 6 내지 12개월의 동일한 대상에서 수집한 혈청에서 일반적으로 감소하고, 이것은 높은 역가가 주로 수동 전달된 모계 항체에 의한 것이었다.
그러나, 특정 유아에서는 아마도 폐렴구균에 대한 자연 노출의 결과로서 일부 단백질에 대한 면역 반응이 성장하면서 증가하였다. 이러한 혈청 전환에 주로 관련되는 항원은 명백하게 PsaA이었다. PhtD, NR1xR2, Sp128, Sp91 및 Ply에 대한 항체 수준의 증가도 밝혀졌다. 단지 Sp1O1 및 Sp13O에 대한 체액성 반응의 일부 변화만이 관찰되었다 (도 1 및 2 참조).
2.2.2 청년층의 스트렙토코커스 단백질 혈청학
기하 평균 역가에 따르면, PhtD, PhtA 및 NR1xR2는 평가된 청년층 집단에서 가장 면역원성이 높은 단백질이고, 그 다음으로 Sp128, Ply 및 Sp91의 면역원성이 높았다. 모든 대상은 상기 단백질에 대한 검출 가능한 항체를 보유하였다. 보다 낮은 반응이 Sp46, 특히 Sp13O 및 Sp1O1에서 나타났다. PsaA는 시험되지 않았다 (충분한 혈청을 입수할 수 없음) (도 3 및 4 참조).
2.2.3 장년층의 스트렙토코커스 단백질 혈청학
청년층에 비해 장년층의 스트렙토코커스 단백질에 대한 항체 수준이 분명하게 감소하였다. 장년층에서, 가장 면역원성이 높은 단백질은 PhtD이고, 그 다음으로 Sp128, NR1xR2, Sp91, Ply 및 PsaA의 면역원성이 높았다. Sp1O1 및 Sp13O에 대해서는 미약한 반응만이 측정되었다. Sp46 및 PhtA는 시험되지 않았다 (물질 입수 문제) (도 5 및 6 참조).
