(a)微生物のための栄養として同化可能な炭素源および同化可能な窒素 源を含む水性培地中、グルコアミラーゼ(アスペルギルスの場合)またはセロビ オヒドロラーゼ(トリコデルマの場合)プロモーターの制御下で異種フィターゼ 遺伝子を有するアスペルギルスまたはトリコデルマ属の微生物をフィターゼの組 換え体発現を可能にする条件下で培養し; (b)水性培地を濾過して微生物を除去して、水性濾液を得;次いで (c)(b)由来の濾液を限外濾過に付して、少なくとも14,000FTU /gのフィターゼ濃度を有する水性液体を得ることからなる、フィターゼを含む 水性液体の製法。 2. 微生物がアスペルギルス・ニガー、アスペルギルス・オリザエまたはトリ コデルマ・レッセイである請求項1記載の製法。 3. 微生物がグルコアミラーゼ(AG)遺伝子を有さないか、または発現しな い請求項1または2記載の製法。 4. 微生物が多コピーのフィターゼ遺伝子を有する前記請求項のいずれか1項 記載の製法。 5. 水性液体が実質上、タカ−アミラーゼを含有しない前記請求項のいずれか 1項記載の製法。 6. 実質上、培地中の全ての炭素および窒素源が(b)における濾過前に微生 物によって消費された前記請求項のいずれか1項記載の製法。 7. 水性液体が炭素および/または窒素源を含有しない前記請求項のいずれか 1項記載の製法。 8. フィターゼがグルコアミラーゼシグナル配列を有する微生物において発現 される前記請求項のいずれか1項記載の製法。 9. 水性濾液または水性液体のいずれも結晶化および/または着色除去工程に 付さない前記請求項のいずれか1項記載の製法。 10. 得られた水性液体が18,000FTU/g以上のフィターゼ活性を有 する前記請求項のいずれか1項記載の製法。 11. 少なくとも14,000FTU/gの濃度のフィターゼを含む前記請求 項のいずれか1項記載の製法によって調製できる水性液体。 12. フィターゼが発現された培養基から得られる請求項11記載の水性液体 。 13. 少なくとも15%(w/w)の食用炭水化物ポリマーおよび請求項11 または12記載の水性液体を加工処理して、フィターゼ含有顆粒を得ることから なる、動物飼料における使用に適するフィターゼ含有粒状物の製法。 14. 水性液体および担体を混合し、得られた混合物を練合する請求項13記 載の製法。 15. 次いで顆粒を乾燥する請求項14記載の製法。 16. a)フィターゼを含有する水性液体を固形担体と混合し; b)a)で得られた混合物を機械的に加工処理して酵素含有顆粒を得;次いで c)b)で得られた酵素含有顆粒を乾燥させることからなる、請求項13また は14記載の製法。 17. 加工処理が押出し成形、ペレット成形、高剪断造粒、膨張、流動層凝集 またはそれらの組み合わせからなる、請求項15記載の製法。 18. 請求項13〜17のいずれか1項記載の製法により調製できるフィター ゼ含有粒状物。 19. フィターゼおよび少なくとも15%(w/w)の食用炭水化物ポリマー を含む固形担体から形成された乾燥顆粒を含んでなる粒状物。 20. 顆粒が少なくとも1個の2価カチオンを含む請求項19記載の粒状物。 21. 顆粒が1以上の疎水性のゲル形成または水不溶性化合物を含む請求項1 9または20記載の粒状物。 22. 疎水性のゲル形成または水不溶性化合物が誘導体化セルロース、ポリビ ニルアルコール(PVA)または食用油を含む請求項21記載の粒状物。 23. 誘導体化セルロースがヒドロキシ−プロピル−メチル−セルロース、カ ルボキシ−メチル−セルロースまたはヒドロキシ−エチル−セルロースであり、 および/または食用油が大豆油もしくはカノーラ油である請求項22記載の粒状 物。 24. さらにエンド−キシラナーゼおよび/またはβ−グルカナーゼを含む請 求項19〜23のいずれか1項記載の粒状物。 25. 担体がデンプンを含む請求項19〜24のいずれか1項記載の粒状物。 26. フィターゼが熱耐性(熱安定性)フィターゼ以外である請求項19〜2 5のいずれか1項記載の粒状物。 27. フィターゼが真菌性フィターゼである請求項19〜26のいずれか1項 記載の粒状物。 28. 真菌性フィターゼがアスペルギルスまたはトリコデルマ種由来である請 求項19〜27のいずれか1項記載の組成物。 29. 請求項19〜28のいずれか1項記載のフィターゼ含有粒状物を1以上 の動物飼料物質または成分と混合することからなる、動物飼料、動物飼料のプレ ミックスまたは前駆物質の製法。 30. 飼料物質および組成物または粒状物の混合物を蒸気処理し、ペレット成 形し、所望により乾燥させる請求項29記載の製法 31. 請求項19〜28のいずれか1項記載の粒状物および/または少なくと も6,000FTU/gの活性を有するフィターゼ含有粒状物を含んでなる組成 物。 32. 食用飼料組成物である請求項31記載の組成物。 33. 動物飼料である請求項32記載の組成物。 34. 請求項19〜27のいずれか1項記載の粒状物と混合した1以上の飼料 物質または成分のペレットを含んでなる請求項32または33記載の組成物。 35. 請求項29または30記載の製法により調製できる動物飼料、または動 物飼料のプレミックスもしくは前駆物質である請求項31〜34のいずれか1項 記載の組成物。 36. 動物に請求項19〜27のいずれか1項記載の粒状物または請求項31 〜35のいずれか1項記載の組成物を含む食餌を与えることからなる、動物の成 長を促進する方法。 37. 動物飼料における、または動物飼料の成分としての、または動物用食餌 における使用のための請求項19〜27のいずれか1項記載の粒状物の使用。 38. フィターゼのペレット成形安定性を改善するために、少なくとも15% (w/w)の食用炭水化物ポリマーをフィターゼの担体として含んでなる組成物 の使用。
