Claims (15)
1
1. Fremgangsmåde til påvisning af et målmolekyle, som eventuelt er til stede i en testprøve, hvilken fremgangsmåde omfatter: 5 (a) at bringe en testprøve i kontakt med en aptamer med en specifik affinitet for målmolekylet, hvor et aptameraffinitetskompleks dannes ved en aptamers interaktion med dens målmolekyle, hvis målmolekylet er til stede i testprøven, hvor et aptameraffinitetskompleks er et ikke-kovalent kompleks dannet ved en aptamers interaktion med dens målmolekyle; 10 (b) efter dannelse af et aptameraffinitetskompleks i trin (a) at indføre et kon kurrentmolekyle i testprøven og/eller at indfange aptameraffinitetskomplekset på en fast bærer og efterfølgende at vaske med en opløsning med et konkurrentmolekyle, der er til stede i opløsningen, hvor konkurrentmolekylet er udvalgt blandt et oligonukleotid, en polyanion, en polydextran eller dNTPs; og 15 (c) at påvise og/eller kvantificere aptameren i aptameraffinitetskomplekset eller aptameraffinitetskomplekset, der er adskilt fra resten af testprøven.
2. Fremgangsmåde ifølge krav 1, hvor konkurrenten er en polyanion.
3. Fremgangsmåde ifølge krav 2, hvor polyanionen er dextransulfat.
4. Fremgangsmåde ifølge krav 2, hvor polyanionen er heparin eller polydextran.
5. Fremgangsmåde ifølge et af kravene 1 til 4, hvor aptameren er en enkelt- strenget nukleinsyre eller en dobbeltstrenget nukleinsyre.
6. Fremgangsmåde ifølge krav 5, hvor aptameren er DNA eller RNA.
7. Fremgangsmåde ifølge et af kravene 1 til 6, hvor målmolekylet er udvalgt fra gruppen bestående af et protein, et kulhydrat, en polysaccharid, et glycoprotein, et hormon, en receptor, et antigen, et antistof, en virus, et substrat, en metabolit, en cofaktor, en inhibitor, et lægemiddel, et farvestof, et næringsmiddel, en vækstfaktor og et væv. 35
8. Fremgangsmåde ifølge krav 7, hvor målmolekylet er et protein. 2
9. Fremgangsmåde ifølge krav 8, hvor proteinet er et glycoprotein.
10. Fremgangsmåde ifølge krav 8, hvor proteinet er en receptor eller et anti- 5 stof.
11. Fremgangsmåde ifølge krav 7, hvor målmolekylet er et kulhydrat.
12. Fremgangsmåde ifølge krav 11, hvor kulhydratet er en polysaccharid. 10

Fremgangsmåde ifølge et af kravene 1 til 12, hvor testprøven er en biologisk prøve fra gruppen bestående af helblod, leukocytter, perifere mononukleare blodceller, plasma, serum, spyt, ånde, urin, sæd, savl, hjernehindevæske, amniotisk væske, kirtelvæske, lymfevæske, brystvorteaspirat, bron- 15 kieaspirat, synovialvæske, ledaspirat, celler, et celleekstrakt, fæces, væv, en vævsbiopsi og cerebrospinalvæske.
14. Fremgangsmåde ifølge krav 13, hvor testprøven er en biologisk prøve af celler. 20
15. Fremgangsmåde ifølge krav 14, hvor cellerne er leukocytter eller perifere mononukleare blodceller.
