Abstract
Ligandos derivados de piperidina de alta afinidad para el receptor de nociceptina ORL-1, caracterizados porque comprenden un compuesto representado por la formula (1) o una sal o solvato del mismo farmacéuticamente aceptable donde: la línea de puntos representa un doble enlace opcional; X1 es R5-alquilo C1-12, R5-cicloalquilo C3-12, R7-arilo, R8-heteroarilo, R10-heterocicloalquilo C3-7; X2 es -CHO, -CN, -NHC(=NR26)NHR26, -CH(=NOR26), -NHOR26, R7-arilo, R7-arilalquilo C1-6, R7-arilalquenilo C1-6, R7-arilalquinilo C1-6, -(CH2)vOR13, -(CH2)vCOOR27, -(CH2)vCONR14R15, -(CH2)vNR21R22 o -(CH2)vNHC(O)R21, donde v es 0, 1, 2 o 3; o X1 es del grupo de formulas (2) y X2 es hidrogeno; o X1 y X2 conjuntamente forman un grupo espiro que tiene la formula del grupo de formulas (3); m es 1 o 2; n es 1, 2 o 3, con la condicion de que cuando n es 1, uno de R16 y R17 es -C(O)R28; p es 0 o 1, Q es -CH2-, -O-, -S-, -SO-, -SO2- o -NR17-; R1, R2, R3 y R4 son independientemente seleccionados del grupo que consiste en hidrogeno y alquilo C1-6, o (R1 y R4) o (R2 y R3) o (R1 y R3) o (R2 y R4) conjuntamente pueden formar un puente alquileno de 1 a 3 átomos de carbono; R5 representa de 1 a 3 substituyentes, independientemente seleccionados del grupo que consiste en H, R7-arilo, R6-cicloalquilo C3-12, R8-heteroarilo, R10-heterocicloalquilo C3-7, -NR19R20, -OR13 y S(O)0-2R13; R6 es de 1 a 3 substituyentes independientemente seleccionados del grupo que consiste en H, alquilo C1-6, R7-arilo, -NR19R20, -OR13, -SR13; R7 representa de 1 a 3 substituyentes seleccionados del grupo que consiste en hidrogeno, halo, alquilo C1-6, R25-arilo, cicloalquilo C3-12, -CN, -CF3, -OR19, -alquilo C1-6-OR19, -OCF3, -NR19R20, -alquilo C1-6-NR19R20, -NHSO2R19, -SO2N(R26)2, -SO2R19, -SOR19, -SR19, -NO2, -CONR19R20, -NR20COR19, -COR19, -COCF3, -OCOR19, -OCO2R19, -COOR19, -alquilo C1-6-NHCOOC(CH3)3, -alquilo C1-6-NHCOCF3, -alquilo C1-6-NHSO2-alquilo C1-6, -alquilo C1-6-NHCONH-alquilo C1-6- o un resto de formula (4), donde f es 0 a 6; o los substituyentes R7 en los átomos de carbono adyacentes del anillo pueden formar conjuntamente un anillo metilendioxi o etilendioxi; R8 representa de 1 a 3 substituyentes independientemente seleccionados del grupo que consiste en hidrogeno, halo, alquilo C1-6, R25-arilo cicloalquilo C3-12 CF3, -OR19, -alquilo C1-6-OR19, -OCF3, -NR19R20, -alquilo C1-6-NR19R20, -NHSO2R19, -SO2N(R26)2, -NO2, -CONR19R20, -NR20COR19, -COR19, -OCOR19, -OCO2R19, -COOR19, R9 es hidrogeno, alquilo C1-6, halo, -OR19, -NR19R20, -NHCN, -SR19 o -alquilo C1-6-NR19R20; R10 es H, alquilo C1-6, -OR19, -alquilo C1-6-OR19, -NR19R20, o alquilo C1-6-NR19R20; R11 está independientemente seleccionado del grupo que consiste en H, R5-alquilo C1-6, R6-cicloalquilo C3-12, -alquilo C1-6cicloalquilo C3-12, -alquilo C1-6-OR19, -alquilo C1-6-NR19R20 y un resto de formulas (5) donde q y a son tal como se han definido más arriba; R12 es H, alquilo C1-6, halo, -NO2, -CF3, -OCF3, -OR19, -alquilo C1-6-OR19, -NR19R20, o alquilo C1-6-NR19R20; R13 es H, alquilo C1-6, R7-arilo, -alquilo C1-6-OR19, -alquilo C1-6-NR19R20, -alquilo C1-6-SR19 o arilalquilo C1-6; R14 y R15 está independientemente seleccionados del grupo que consiste en H, R5-alquilo C1-6, R7-arilo y el resto de formula (6) donde q es 1 a 3 y a es 1 o 2; R16 y R17 son independientemente seleccionados del grupo que consiste en hidrogeno, R5-alquilo C1-6, R7-arilo, cicloalquilo C3-12, R8-heteroarilo, R8-heteroarilalquilo C1-6-C(O)R28, -alquil C1-6-heterocicloalquilo C3-7, -alquilo C1-6-OR19 y -alquilo C1-6-SR19; R19 y R20 son independientemente seleccionados del grupo que consiste en hidrogeno, alquilo C1-6, cicloalquilo C3-12, arilo, y arilalquilo C1-6; R21 y R22 hidrogeno, alquilo C1-6, cicloalquilo C3-12,cicloalquil C3-12 alquilo C1-6-heterocicloalquilo C3-7, -alquilo C1-6-heterocicloalquilo C3-7, R7-arilo, R7-aril-alquilo C1-6-, R8-heteroarilo-alquilo C1-6, alquilo C1-6-OR19, -alquilo C1-6, NR19R20, -alquilo C1-6-SR19, y alquilo C1-6-NR18-alquilo C1-6-NR18-alquilo C1-6; R18 es hidrogeno o alquilo C1-6; Z1 y Z2 son cada uno R7-arilo; Z3 es hidrogeno o alquilo C1-6; R25 representa 1 a 3 substituyentes independientemente seleccionados del grupo que consiste en H, alquilo C1-6, alcoxi C1-6 y halo; R26 está independientemente seleccionado del grupo que consiste en H, alquilo C1-6 y R25-C6H4-CH2-; R27es H, alquilo C1-6, R7-arilalquilo C1-6, o cicloalquilo C3-12; R28 es alquilo C1-6, alquil C1-6cicloalquilo C3-12, R7-arilo, R7-aril-alquilo C1-6-R8-heteroarilo alquil C1-6-NR19R20, -alquilo C1-6-OR19, o alquilo C1-6-SR19 con la condicion de que cuando X1 y X2 juntos son un compuesto del grupo de formulas (7) y Z1 y Z2 son cada uno R7-fenilo, R7 no es -OCH2-arilo y al menos un es orto-halo u orto-alquilo C1-6; arilo (incluyendo la porcion arilo de arilalquilo) representa un grupo carbocíclico que contiene de 6 a 15 átomos de carbono y que tiene por lo menos un anillo aromático donde dicho grupo arilo puede estar opcionalmente fusionado con anillos arilo, cicloalquilo C3-7, heteroarilo o heteroarilo-cicloalquilo C3-7;y donde R7-arilo significa que cualquiera de los átomos de carbono y nitrogeno sustituibles disponibles en dicho grupo arilo y/o anillos fusionados está opcionalmente e independientemente sustituido, y donde el anillo arilo está sustituido con 1-3 grupos R7; heteroarilo representa grupos cíclicos que tienen uno a tres heteroátomos seleccionados de O, S y N, interrumpiendo dicho(s) heteroátomo(s) una estructura de anillo carbocíclico y teniendo una cantidad suficiente de electrones pi deslocalizados para proporcionar carácter aromático, con grupos heterocíclicos aromáticos que contienen de 5 a 14 átomos de carbono, donde dicho grupo heteroarilo puede estar opcionalmente fusionado con uno o más anillos arilo, cicloalquilo, heteroarilo o heterocicloalquilo; y donde cualquiera de los átomos de nitrogeno o carbono disponibles en dicho grupo heteroarilo y/o dicho(s) anillo(s) fusionado(s) puede estar opcional e independientemente sustituido y donde el anillo heteroarilo puede estar sustituido con 1-3 grupos R8; y heterocicloalquilo representa un anillo saturado que contiene de 3 a 7 átomos de carbono, preferiblemente de 4 a 6 átomos de carbono, interrumpidos por 1 a 3 heteroátomos seleccionados de -O-, -S- y NR21, donde R21 es tal como se definio antes, y en donde opcionalmente, dicho anillo puede contener uno o dos enlaces insaturados que no imparten carácter aromático al anillo; y en donde cualquiera de los átomos de carbono sustituibles disponibles en el anillo puede estar sustituido y donde el anillo heterocicloalquilo puede estar sustituido con 1-3 grupos R10; los grupos heterocicloalquilo representativos incluyen 2- o 3-tetrahidrotienilo, 1-, 2-, 3- o 4-piperidinilo, 2- o 3-pirrolidinilo, 1-, 2-, o 3-piperazinilo, 2- o 4-dioxanilo, morfolinilo, o las estructuras del grupo de formulas (8), en donde R17 es como se ha definido antes y t es 0, 1 o 2. Composiciones farmacéuticas que incluyen los mismos y el uso de dichos compuestos como inhibidores, receptores de nociceptina para la manufactura de medicamentos utiles para el tratamiento, ansiedad, tos, asma, depresion y abuso de alcohol.
