Abstract
Un compuesto derivado de pirazolo[3,4-d]pirimidina que tiene la fórmula (1), mezclas racémicas-diastereoméricas, isómeros ópticos, sales aceptables para uso farmacéutico, profármacos o metabolitos biológicamente activos de los mismos donde: G es un compuesto de fórmula (2), donde Z100 es un resto de fórmula (3) o un grupo opcionalmente sustituido con R1 seleccionado del grupo formado por alquilo, cicloalquilo, pirrolidinilo, quinolinilo, quinoxalinilo, quinazolinilo, isoquinolinilo, ftalazinilo, imidazo[1,2-a]pirimidinilo, 1H-imidazo[1,2-a]imidazolilo, imidazo[2,1-b][1,3]tiazolilo, naftilo, tetrahidronaftilo, benzotienilo, furanilo, tienilo, benzoxazolilo, benzoisoxazolilo, benzotiazolilo, un resto del grupo de fórmulas (4), tiazolilo, benzofuranilo, 2,3-dihidrobenzofuranilo, indolilo, isoxazolilo, tetrahidropiranilo, tetrahidrofuranilo, piperidinilo, pirazolilo, pirrolilo, pirrolopiridinilo, H-piridinona, oxazolilo, isotiazolilo, oxadiazolilo, tiadiazolilo, indolinilo, indazolilo, imidazo[1,2-a]piridinilo, benzoisotiazolilo, 1,1-dioxibenzoisotiazolilo, pirido-oxazolilo, pirido-tiazolilo, pirimido-oxazolilo, pirimido-tiazolilo y bencimidazolilo; Z110 es un enlace covalente, o un C1-6 opcionalmente sustituido que es opcionalmente sustituido con uno o más sustituyentes seleccionados del grupo formado por alquilo, CN, OH, halógeno, NO2, COOH, amino sustituido o insustituido y fenilo sustituido o insustituido; Z111 es un enlace covalente, un C1-6 opcionalmente sustituido o un -(CH2)n-cicloalquil-(CH2)n opcionalmente sustituido; donde los grupos opcionalmente sustituido son opcionalmente sustituidos con 1 o más sustituyentes seleccionados del grupo formado por alquilo, CN, OH, halógeno, NO2, COOH, amino sustituido o insustituido y fenilo sustituido o insustituido; Ra y R1 representan cada uno, uno o más sustituyentes para cada caso independientemente seleccionado del grupo formado por hidrógeno, halógeno, -CN, -NO2, -C(O)OH, -C(O)H, -OH, -C(O)O-alquilo, -C(O)O-arilo, -C(O)O-heteroarilo, -C(O)-alquilo, -C(O)-arilo, -C(O)-heteroarilo, carboxamido sustituido o insustituido, tetrazolilo, trifluorometilcarbonilamino, trifluorometilsulfonamido, alquilo sustituido o insustituido, cicloalquilo sustituido o insustituido, alcoxi sustituido o insustituido, arilo sustituido o insustituido, heteroarilo sustituido o insustituido, alquenilo sustituido o insustituido, ariloxi sustituido o insustituido, heteroariloxi sustituido o insustituido, heteroarilalcoxi sustituido o insustituido, arilalcoxi sustituido o insustituido, alquil-S(O)p- sustituido o insustituido, alquil-S- sustituido o insustituido, aril-S(O)p- sustituido o insustituido, heteroaril-S(O)p- sustituido o insustituido, arilalquilo sustituido o insustituido, heteroarilalquilo sustituido o insustituido, cicloalquilalquilo sustituido o insustituido, alquinilo sustituido o insustituido, amino sustituido o insustituido, amino sustituido o insustituido alquilo, grupos amido sustituidos o insustituidos, heteroariltio sustituido o insustituido, ariltio sustituido o insustituido, -Z105-C(O)N(R)2, -Z105-N(R)-C(O)-Z200, -Z105-N(R)-S(O)2-Z200, -Z105-N(R)-C(O)-N(R)-Z200, Rc y CH2ORc, donde Rc para cada caso es independientemente hidrógeno, alquilo sustituido o insustituido, arilo sustituido o insustituido, -CH2-NRdRe, -W-(CH2)t-NRdRe, -W-(CH2)t-O-alquilo, -W-(CH2)t-S-alquilo, o -W-(CH2)t-OH; Z105 para cada caso es independientemente un enlace covalente o C1-6; Z200 para cada caso es independientemente un C1-6 sustituido o insustituido, fenilo sustituido o insustituido o C1-6-fenilo sustituido o insustituido; Rd y Re para cada caso son independientemente H, alquilo, alcanoílo o SO2-alquilo; o Rd, Re y el átomo de nitrógeno al cual están unidos forman en conjunto un anillo heterocíclico de 5 o 6 miembros; t para cada caso es independientemente un entero entre 2 y 6; W para cada caso es independientemente un enlace directo ó O, S, S(O), S(O)2, o NRf, donde Rf para cada caso es independientemente H o alquilo; o R1 es un anillo carbocíclico sustituido o insustituido o un anillo heterocíclico fusionado con el anillo 2; R3 para cada caso es, independientemente, hidrógeno, hidroxi, alquilo sustituido o insustituido, -C(O)-alquilo sustituido o insustituido, un -C(O)-arilo sustituido o insustituido, o un -C(O)-heteroarilo sustituido o insustituido o alcoxi sustituido o insustituido; Rb para cada caso es independientemente H, alquilo sustituido o insustituido, arilalquilo sustituido o insustituido, cicloalquilo sustituido o insustituido o arilo sustituido o insustituido; p es 1 o 2; o en un grupo con contenido de fósforo, el átomo de nitrógeno, el átomo de fósforo, R y Rb juntos forman un anillo heterocíclico de 5 o 6 miembros; o A es NRSO2 y R, Ra y el átomo de nitrógeno juntos forman un anillo heterocíclico de 5 o 6 miembros sustituido o insustituido fusionado al anillo 1; o Z110-A-Z111 tomados juntos son un enlace covalente; y R2 es H o un grupo de la fórmula -Z101-Z102; Z101 es un enlace covalente, -(C1-6)-, -(C1-6)-O-, -(C1-6)-C(O)-, -(C1-6)-C(O)O-,-(C1-6)-C(O)-NH-, -(C1-6)-C(O)-N(C1-6)- o un grupo fenilo sustituido o insustituido; Z102 es hidrógeno, un grupo alquilo sustituido o insustituido, un grupo cicloalquilo sustituido o insustituido, un cicloalquenilo sustituido o insustituido, un grupo heterocíclico sustituido o insustituido, saturado o insaturado; o un grupo heterobicíclico sustituido o insustituido, saturado o insaturado; donde dicho alquilo sustituido, cicloalquilo sustituido, cicloalquenilo sustituido, heterocíclico sustituido y grupo heterobicíclico sustituido con uno o más sustituyentes cada uno independientemente seleccionado del grupo formado por hidroxilo, ciano, nitro, halo, C1-6 sustituido o insustituido, arilo sustituido o insustituido, C(O)-alquilo sustituido o insustituido, alcoxi sustituido o insustituido, -N(R)-(C1-6)-OR sustituido o insustituido, -N((C1-6)-OR)2 sustituido o insustituido, -N(R)-(C1-6)-C(O)2R sustituido o insustituido, -(C1-6)-N(R)-(C1-6)-OR sustituido o insustituido, -(C1-6)-N(R)-(C1-6)-N(R)2 sustituido o insustituido, -(C1-6)-C(O)N(R)-(C1-6)-N(R)2 sustituido o insustituido, sulfonamido sustituido o insustituido, ureido sustituido o insustituido, carboxamido sustituido o insustituido, amino sustituido o insustituido -N(R)-(C1-6)-OR sustituido o insustituido, oxo y un grupo heterocíclico saturado, insaturado o aromático, sustituido o insustituido que comprende uno o más heteroátomos seleccionados del grupo formado por N, O y S;
