Claims (19)
1. Способ обогащения популяции клеток, экспрессирующих маркеры, характерные для линии первичной кишечной трубки, включающий стадии:
а. дифференцирования популяции плюрипотентных стволовых клеток в популяцию клеток, экспрессирующих маркеры, характерные для линии дефинитивной энтодермы;
b. дифференцирования популяции клеток, экспрессирующих маркеры, характерные для линии дефинитивной энтодермы, в клетки, экспрессирующие маркеры, характерные для линии первичной кишечной трубки;
с. дифференцирование популяции клеток, экспрессирующих маркеры, характерные для линии первичной кишечной трубки, в популяцию клеток, экспрессирующих маркеры, характерные для линии панкреатической энтодермы; и
d. обогащения популяции клеток, которые экспрессируют маркеры, характерные для линии первичной кишечной трубки, путем контактирования популяции клеток, которые экспрессируют маркеры, характерные для линии первичной кишечной трубки, с реагентом, который способен связывать Lif-рецептор.
2. Способ по п. 1, где способ дополнительно включает культивирование популяции плюрипотентных клеток.
3. Способ по п. 1 или 2, где плюрипотентные стволовые клетки являются плюрипотентными стволовыми клетками человека.
4. Способ по п. 3, где плюрипотентные стволовые клетки человека являются эмбриональные стволовыми клетками человека.
5. Способ по п. 1, где клетки, экспрессирующие маркеры, характерные для линии дефинитивной энтодермы, являются клетками дефинитивной энтодермы.
6. Способ по п. 1 или 5, где клетки, экспрессирующие маркеры, характерные для линии первичной кишечной трубки, являются клетками первичной кишечной трубки.
7. Способ получения панкреатических эндокринных клеток-предшественников, включающий дифференцирование обогащенной популяции клеток задней части передней кишки, полученных способом по п. 1, в панкреатические эндокринные клетки-предшественники.
8. Способ обогащения популяции клеток первичной кишечной трубки, включающий контактирование популяции клеток первичной кишечной трубки с реагентом, который способен связывать Lif-рецептор.
9. Способ по п. 8, где клетки первичной кишечной трубки получают последовательным дифференцированием плюрипотентных стволовых клеток человека.
10. Способ по п. 8 или 9, где плюрипотентные стволовые клетки человека являются эмбриональными стволовыми клетками человека.
11. Способ по п. 9, где клетки первичной кишечной трубки получают способом, включающим:
а. дифференцирование популяции плюрипотентных стволовых клеток человека в популяцию клеток дефинитивной энтодермы; и
b. дифференцирование популяции клеток дефинитивной энтодермы в клетки первичной кишечной трубки.
12. Способ по п. 11, где способ дополнительно включает культивирование популяции плюрипотентных стволовых клеток.
13. Способ по п. 8, дополнительно включающий дифференцирование обогащенной популяции полученных клеток задней части передней кишки в панкреатические эндокринные клетки-предшественники.
