Abstract
본 발명은 세포 배양계, 포유류 세포 배양계 또는 돌연변이 동물에서 발현하기 어려운 것으로 알려진 말라리아 표면 단백질 MSP-1의 mRNA량 및 단백질 발현을 증가시키는 변형된 재조합 핵산 서열(바람직하게는 DNA) 및 방법에 관한 것이다. 본 발명의 재조합 기법을 사용한 발현용으로 바람직한 단백질 후보는 천연 MSP-1 유전자에 비하여 총 AT 함량 또는 AT 풍부 영역 및/또는 mRNA 불안정성 모티프 및/또는 희귀 코돈이 감소된 서열을 암호화하는 DNA로부터 발현된 MSP-1 단백질이다.
Description
시험관내 세포 배양계 또는 생체내 재조합체 생성계에서 일부 이종 유전자 생성물의 재조합체를 생성하는 것이 어려울 때가 종종 있다. 예를 들어, 여러 연구진들은 원래 단백질이 유래한 세포와 상이한 세포 배양계, 특히 포유류 세포 배양계에서 박테리아, 기생충 및 바이러스에서 유래한 단백질을 발현하는 것이 어렵다는 것을 발견하였다. 포유류 세포가 생성하기 어려운 치료학적으로 중요한 단백질의 일례로는 말라리아 유충체 표면 단백질(MSP-1)이 있다.
말라리아는 열대 지방에서는 심각한 건강 문제이다. 기존 약물에 대한 내성이 급속하게 확산되고 있어, 백신의 필요성이 절박하다. 피.팔시파럼(P. falciparum)의 생활환 과정에서 발현되는 다수의 항원 중, MSP-1이 가장 광범위하게 연구되어 왔으며 예방 접종용으로 가장 성공적인 후보인 것으로 보인다. 피.팔시파럼에 노출된 개체는 MSP-1에 대한 항체를 형성하고, 연구 결과 MSP-1에 대해 자연적으로 획득된 면역 반응과 말라리아에 의한 사망율 감소 사이에 상관 관계가 있다는 것을 밝혀냈다. 여러 연구에서, 정제된 천연 MSP-1 또는 단백질의 재조합 단편을 이용하여 면역화시키면 최소한 부분적으로 기생충으로부터의 보호를 유발함을 확인하였다[Diggs et al., (1993) Parasitol. Today 9:300-302). 따라서, MSP-1은 피.팔시파럼에 대한 백신 개발에 가장 중요한 표적이다.
MSP-1은 190∼220 kDa 당단백질이다. C 말단 영역은 백신으로서 사용하기 위한 재조합체 생성에 있어서 중심이 된다. 그러나, 백신으로서 MSP-1을 개발하는데 있어서 주요 문제는 친화도 정제된 천연 단백질에 대한 면역학적 효능이 동일한 박테리아 또는 효모 발현계에서[Chang et al., (1992) J. Immunol. 148-:548-555], 백신을 생성할 수 있을 만큼 충분한 양으로 재조합 단백질을 얻기가 어렵다는 것이다.
충분한 양의 MSP-1의 발현을 증강시키는 개선된 절차가 유리하다.
본 발명은 이종 유전자 발현에 관한 것이다. 더욱 구체적으로, 본 발명은 고등 진핵 세포계에서의 말라리아 유전자 발현에 관한 것이다.
도 1은 MSP-142의 동일한 단백질 아미노산 서열을 유지하면서 AT 함량을 낮추고 mRNA 불안정성 모티프가 제거되도록 306개 뉴클레오티드 위치를 대체한 본 발명에 따라 변형된 MSP-142의 cDNA 서열을 도시한다. 대문자는 뉴클레오티드 치환체를 나타낸다.

도 5의 패널 A는 노던 분석으로서, 작제물 GTC 627은 본 발명에 따라 변형된 신규 MSP-142유전자를 포함하고, GTC 479는 천연 MSP-142유전자를 포함하는 작제물이며, 작제물 GTC 469는 음성 대조군 DNA이다.

바람직한 양태들의 상세한 설명
본원에서 언급한 특허 및 과학 문헌은 당업자들이 이용가능한 지식을 수록하고 있다. 본원에서 인용한 간행된 미국 특허, 허가된 특허 출원, 공개된 외국 공보와 참조 문헌은 참고로 도입한 것이다. 이들 참조 문헌과 본원에 개시된 내용간에 상충되는 것이 있으면 본원에 개시된 내용으로 해석해야 한다.
본 발명은 세포 배양계, 포유류 세포 배양계 또는 돌연변이 포유류에서 말라리아 표면 항원 MSP-1의 mRNA량 및 단백질 발현을 증가시키는 개선된 재조합 DNA 조성물 및 절차를 제공한다. 본 발명의 발현계에서 발현용으로 바람직한 단백질 후보는 재조합 발현계에 비하여 AT 함량이 감소되고, mRNA 불안정성 모티프 및 희귀 코돈이 제거된 서열을 암호화하는 DNA를 보유하는 MSP-1의 C 말단 유도체이다. 따라서, 본 발명의 제1 목적은 서열 번호 2로 표시되는 서열에서 유래된 DNA 서열을 제공하는 것이다. 이 유도 서열은 서열 번호 1에 도시되어 있다.
바람직한 양태에서, 본 발명의 핵산은 포유류 세포 배양계 또는 돌연변이 포유류에서 천연 유전자에 의한 발현량의 25% 이상, 바람직하게는 50% 이상, 더욱 더 바람직하게는 100% 이상의 양으로 동일한 조건하에 포유류 세포 배양계 또는 돌연변이 포유류에서 MSP-1을 발현할 수 있다.
본원에서 ″발현″은 단백질 발현을 초래하는 mRNA 전사를 의미한다. 해당 단백질에 특이적인 항체를 사용하는 방법을 비롯하여 당업계에 공지된 다수의 기법을 사용하여 발현을 측정할 수 있다. ″천연 유전자″(natural gene 또는 native gene)는 MSP-1의 야생형 형태를 암호화하고 변형된 핵산이 유래된 유전자 서열, 또는 이의 단편(자연 발생적 대립 유전자 변형체 포함)을 의미한다. ″바람직한 코돈″은 변형된 MSP-1 유전자를 발현하고자 하는 세포 또는 조직형, 예컨대 유방 조직에 의해 더욱 우세하게 사용되는 코돈을 의미한다. 코돈 대체 결과 최소한 마우스 유방 조직이 인식되는 코돈이 되는 한 본원에 개시된 모든 코돈을 바람직한 코돈으로 변경해야 하는 것은 아니다. ″AT 함량 감소″는 A(아데닌) 또는 T(티민) 함유 뉴클레오티드 위치나 A 및/또는 T 함유 코돈을 마우스 유방 조직이 인식하지만 표적 단백질의 아미노산 서열은 변경시키지 않는 뉴클레오티드 또는 코돈으로 대체함으로써 천연 MSP-1 유전자에 비하여 A 또는 T를 보유한 뉴클레오티드의 전체 백분율이 더 낮아지는 것을 의미한다.

이론에 구애되지 않는다면, 앞서의 연구는 GM-CSF mRNA의 3' 비해독된 영역에서 유래한 보존된 AU 서열(AUUUA)이 선택적 mRNA 분해를 매개한다고 제시하였다[Shaw, G. and Kamen, R. Cell 46:659-667). 과거에는 유전자의 비해독 영역내 불안성성 모티프의 존재에 주목하였다. 본 발명은 MSP-1 유전자의 암호화 영역에서 불안정성 서열을 제거하는데 있어서 장점을 인식한 최초의 발명이다.
본 발명의 제3 목적은 본 발명의 변형 MSP-1 핵산과 염소 베타 카제인 프로모터 및 시그널 서열을 포함하는 벡터와, 본 발명의 핵산으로 형질전환시킨 숙주 세포를 제공하는 것이다.
본 발명의 제4 목적은 배선이 본 발명의 핵산을 포함하는 돌연변이된 비인간 포유류를 제공하는 것이다. 돌연변이 동물을 생성하는 일반적인 원리는 당업계에 공지되어 있다. 예를 들어, Hogan 등의 문헌[Manipulating the Mouse Embryo: A Laboratory Manual, Cold Spring Harbor Laboratory, (1986)], Simons 등의 문헌[Bio/Technology, 6:179-183, (1988)], Wall 등의 문헌[Bio Reprod. 32:645-651, (1985)], Buhler 등의 문헌[Bio/Technology, 8:140-143(1990)], Ebert 등의 문헌[Bio/Technology, 9:835-838(1991)], Krimenfort 등의 문헌[Bio/Technology, 9:844-847(1991), Wall 등의 문헌[J.Cell. Biochem. 49:113-120(1992)] 참조. 포유류 및 그 배세포내로 외래 DNA 서열을 도입하는 기법은 본래 마우스에서 개발되었다. 예컨대 Gordon 등의 문헌[Proc.Natl.Acad.Sci. USA 77:7380-7384, (1980)], Gordon and Ruddle, Science 214:1244-1246 (1981)], Palmiter 및 Brinster 등의 문헌[Cell 41:343-345, 1985], Brinster 등의 문헌[Proc.Natl.Acad.Sci. USA 82:4438-4442(1985)] 및 Hogan 등의 문헌[동일 문헌] 참조. 따라서, 이들 기법은 젖소 및 염소를 비롯한 거대 동물에 사용하는데 적당하다. 매우 최근까지, 돌연변이 마우스 또는 돌연변이 가축 형성에 가장 널리 사용되는 절차는 돌연변이 발현 작제물의 형태로 수 백개의 해당 DNA 선형 분자를 수정된 난의 전핵 중 하나로 주사하는 것이다. 접합체의 세포질내로 DNA를 주사하는 방법도 널리 사용된다. 가장 최근에 수정되지 않은 난에 주사할 수 있는 전체 돌연변이 세포주를 클로닝할 수 있게 되었다[KHS Campbell et al., Nature 380 64-66(1996)].
유선 발현계는 고 발현량, 저 비용, 옳바른 프로세싱 및 접근성의 장점을 갖는다. 소 및 인간 알파 락트알부민과 같은 기지의 단백질을 젖 분비 돌연변이 동물에서 생성할 수 있음이 일부 연구자들에 의해서 밝혀졌으며[Wright et al., Bio/Technology 9:830-834(1991); Vilotte et al., Eur.J.Biochem., 186:43-48(1989); Hochi et al., Mol Reprod. And Devel. 33:160-164(1992); Soulier et al., FEBS Letters 297(1,2):13-18(1992)], 이 계는 다량의 단백질을 발현하는 것으로 확인되었다.
본 발명의 제5 목적은 본 발명의 변형 MSP-1 유전자를 포함하는 DNA 백신을 제공하는 것이다. 이러한 DNA 백신은 캡슐화시키지 않고 전달할 수 있다. 또는 리포좀의 일부나 바이러스 게놈의 일부로서 전달할 수도 있다. 일반적으로, 이러한 백신은 변형 MSP-1 유전자를 발현시키고 DNA 백신을 수용한 인간을 비롯한 동물에서 항체 반응을 유발할 수 있는 유효량으로 전달한다. 제1 전달이 있은 지 1 주일 후에 후속 전달하여 항체 반응을 증가시킬 수 있다. 바람직한 전달 경로는 점막의 막 뿐 아니라 비경구 투여를 통한 도입을 포함한다.
발명의 개요
본 발명은 세포 배양계, 포유류 세포 배양계 또는 돌연변이 포유류에서 말라리아 표면 항원 MSP-1의 mRNA량 및 단백질 발현을 증가시키는 개선된 재조합 DNA 조성물 및 절차를 제공한다. 본 발명의 발현계에서 발현용으로 바람직한 단백질 후보는 재조합 발현계에 비하여 AT 함량이 감소되고, mRNA 불안정성 모티프 및 희귀 코돈이 제거된 서열을 암호화하는 DNA를 보유하는 MSP-1의 C 말단 유도체이다. 따라서, 본 발명의 제1 목적은 서열 번호 2로 표시되는 서열에서 유래된 DNA 서열을 제공하는 것이다. 이 유도 서열은 서열 번호 1에 도시되어 있다.

신규 변형 MSP-142유전자의 작제
일양태로서 MSP-1의 C 말단 단편을 암호하는 신규 변형 핵산을 제공한다. 본 발명의 포유류 세포에서 발현할 수 있는 MSP-1의 42 kD C-말단 부분(MSP-142)을 암호하는 본 발명의 신규 변형 핵산은 도 1에 도시하였다. 천연 MSP-142유전자(도 2)는 포유류 세포 배양물 또는 돌연변이 마우스에서 발현될 수 없다. 천연 MSP-142유전자를 분석해 본 결과 포유류 유전자와 다른 여러 특징이 있음이 암시되었다. 첫째, 총 AT 함량이 76%로 매우 높았다. 둘째, mRNA 불안정성 모티프, AUUUA가 이 1100 bp DNA 분절에서 10 배 높게 나타났다(도 2). 이와 같은 차이를 해명하기 위하여 새로운 MSP-142유전자를 디자인하였다. 천연 MSP-142유전자의 306개 위치에 침묵 뉴클레오티드 치환체를 도입하여 총 AT 함량을 49.7%로 감소시켰다. 또한, 코돈 사용을 변화시켜 천연 유전자 중의 10 AUUUA mRNA 불안정성 모티프를 각각 제거하였다. 코돈 사용을 변화시키기 위하여, 여러 마우스 및 염소의 유방 특이적 단백질을 사용하여 도 3에 기재한 유방 조직 특이적 코돈 사용표를 만들었다. 이 표를 사용하여 전술한 변형 MSP-142유전자의 코돈 사용을 선택하였다. 예컨대, 도 3의 표에 나타낸 바와 같이 천연 유전자에서는 Leu의 65%(25/38)가 TTA에 의해 암호되고 있는데, 이 코돈은 유선에서는 희귀한 코돈이다. 따라서, 변형 MSP-142유전자에서는 유선에 존재하는 Leu의 바람직한 코돈인 CTG에 의해 Leu이 100% 암호화되었다.
발현 벡터는 변형 MSP-142유전자를 사용하여, 염소 베타 카제인의 처음 26개 아미노산을 변형 MSP-142유전자의 N-말단에 융합시키고, 이 융합 유전자를 운반하는 SalI-XhoI 단편을 발현 벡터 pCDNA3의 XhoI 부위에 서브클로닝하여 작제하였다. 이 유전자 생성물을 친화성 정제할 수 있도록 MSP-142유전자의 3' 말단에 His6 태그를 융합시켰다. 그 결과 플라스미드 GTC627이 얻어졌다(도 4c).
천연 MSP-142유전자 작제물과 본 발명의 변형 MSP-142핵산을 비교하기 위하여, 다음과 같이 천연 MSP-142유전자의 발현 벡터를 작제하고, 이 유전자를 포유류 세포 배양물에 첨가한 뒤 마우스에게 주사하여 돌연변이 마우스를 제조하였다.
천연 MSP-142발현 벡터의 작제
플라스모듐 팔시파럼의 절두형 유충체 표면 단백질-1(MSP-1)을 분비형으로 만들기 위하여, MSP-1의 42 kD C 말단 부분(MSP-142)을 암호하는 야생형 유전자를 염소 베타 카제인의 처음 15개 아미노산을 암호하는 DNA 서열이나 처음 26개 아미노산을 암호하는 DNA 서열에 융합시켰다. 이것은, 먼저 프라이머 MSP1 및 MSP2를 사용하여 MSP-1 플라스미드(데이비드 카슬로우(NIH) 박사로부터 입수함)를 PCR 증폭시키고(도 6), 그 다음 PCR 생성물을 TA 벡터(Invitrogen)에 클로닝하여 제조하였다. PCR 생성물의 BglII-XhoI 단편을 발현 벡터 pCDNA3 내의 올리고 OT1 및 OT2와 결찰시켰다(도 6). 그 결과 플라스미드 GTC564(도 4b)를 얻었다. 이 플라스미드는 15개 아미노산 베타 카제인 시그널 펩티드, 성숙 염소 베타 카제인의 처음 11개 아미노산 및 후속되는 천연 MSP-142유전자를 암호한다. 올리고 MSP-8 및 MSP-2(도 6)를 사용하여 PCR로 MSP-1 플라스미드를 증폭시키고, 이 생성물을 그 다음 TA 벡터내로 클로닝하였다. 배양된 COS-7 세포 중에서의 천연 MSP 유전자의 발현은 순간 형질감염 분석법으로 분석하였다. GTC479 및 GTC564 플라스미드 DNA를 제조자의 프로토콜에 따라 리포펙트아민(Gibco-BRL)을 사용하여 COS-7 세포로 도입시켰다. 형질감염 2일 후 이 COS 세포로부터 총 세포 RNA를 분리하였다. 형질감염 2일 후 배양 배지에35S-메티오닌을 첨가하여 10 시간 동안 새로 합성된 단백질을 대사적 표지시켰다.
COS 세포에서의 MSP mRNA 발현률을 측정하기 위하여, 노던 블롯을 GTC479 유래의32P 표시된 DNA 단편으로 탐침하였다. GTC479 또는 GTC564 형질감염체에서는 MSP RNA가 전혀 검출되지 않았다(데이터는 도시 안함). 계속 노출시킨 결과, 분해된 MSP mRNA의 잔류 농도가 관찰되었다.35S 표지된 배양물 상청액과 용균물을 MSP에 대하여 유발된 폴리클로널 항체로 면역침전시켰다. 면역침전 실험 결과, GTC479 또는 GTC564 형질감염 세포의 상청액이나 용균물에서는 발현이 전혀 일어나지 않는 것으로 나타났다(데이터는 제시 안함). 이와 같은 결과를 통해, COS 세포 중에서는 천연 MSP-1 유전자가 발현되지 않음을 알 수 있었다.
돌연변이 마우스의 유선에서 발현되지 않는 천연 MSP-142유전자
염소 베타-카제인 시그널 펩티드의 15개 아미노산과, 염소 베타-카제인의 처음 11개 아미노산 및 천연 MSP-142유전자를 암호하는 GTC479의 SalI-XhoI 단편을, 벡터 BC350 중에서 발현되는 베타-카제인의 XhoI 부위에 클로닝하였다. 그 결과 플라스미드 BC574를 얻었다(도 7). BC574의 SalI-NotI 단편을 마우스 배에 주사하여 돌연변이 마우스를 제조하였다. 돌연변이 마우스 15 주를 제조하였다. 초기 마우스 암컷으로부터 유액을 수집하고, MSP에 대한 폴리클로널 항체를 사용하여 웨스턴 분석하였다. 분석된 7마리 마우스 중 어떤 마우스도 유액 중에 MSP-142단백질을 발현시키지 않았다. MSP-142의 mRNA가 유선 중에서 발현되는지를 추가 조사하기 위하여 11일째 유액분비 돌연변이 마우스로부터 총 RNA를 추출하고 노던 블롯으로 분석하였다. 분석된 BC 574주 중 어느 것에서도 MSP-142mRNA가 검출되지 않았다. 즉, MSP-142변이유전자는 돌연변이 마우스의 유선에서는 발현되지 않았다. 이를 종합해보면, 본 실험은 천연 기생 MSP-142유전자가 포유류 세포에서는 발현될 수 없으며, 방해 요인은 mRNA 다량의 농도임을 암시한다.
포유류 세포에서의 MSP 발현
순간 형질감염 실험을 실시하여 COS 세포에서의 본 발명의 변형 MSP-142유전자의 발현에 대해 평가하였다. COS-7 세포내로 GTC627 및 GTC479 DNA를 도입시켰다. 노던 분석을 위하여 형질감염후 48 시간 후에 총 RNA를 분리하였다. RNA를 고정화시킨 후 GTC627의32P 표지된 SalI-XhoI 단편으로 프로브하였다. GTC479와 GTC627 간에는 큰 차이가 관찰되었다. 전술한 바와 같이 GTC479 형질감염 세포에서는 어떤 MSP-142mRNA도 검출되지 않은 반면, GTC627에서는 다량의 MSP-142mRNA가 발현되었다(도 5, 패널 A). 음성 대조군으로 사용한 GTC469는 시판되는 클로닝 벡터인 PU19 중에 클로닝된 GTC564의 삽입체를 포함한다.35S 메티오닌으로 대사적 표지화한 후 MSP에 대한 폴리클로널 항체(캐슬로우 박사에게서 입수함, NIAID, NIH)로 면역침전시킨 결과, 형질감염된 COS 세포에 의해 MSP-142단백질이 합성된 것을 관찰하였다(도 5, 패널 B). 또한, 형질감염된 COS 상청액에서 MSP-142이 검출되었는데, 이것은 MSP-142단백질이 분비형임을 시사하는 것이다. 또, Ni-NTA 컬럼을 사용하여 GTC627 형질감염된 COS 상청액으로부터 MSP-142를 친화성 정제하였다.
이 결과는 기생체의 MSP-142유전자를 변형시키면 COS 세포 중에서 MSP mRNA가 발현된다는 것을 입증한다. 결과적으로, 포유류 세포에 의해 MSP-142생성물이 합성되고 분비되었다.
또한, 본 실험에서 사용한 폴리클로널 항체는 당해 기술 분야에 공지된 방법으로 제조할 수 있다[Antibodies: A Laboratory Manual, Ed Harlow and David Lane, eds. Cold Spring Harbor Laboratory, publishers(1988)]. 또, 문헌[Chang et al., Infection and Immunity(1996) 64:253-261 and Chang et al.,(1992) Proc.Natl.Acad.Sci. USA 86:6343-6347]에도 MSP 혈청 항체의 생산에 대해 기술되어 있다.
이 분석의 결과는 본 발명의 변형된 MSP-142핵산이 전혀 발현되지 않는 천연 단백질에 비해 상당히 고농도로 발현된다는 것을 보여주고 있다. 이와 같은 결과는 유전자 중의 AT%를 감소시키면 이종계에서 MSP 유전자의 발현이 유도된다는 최초의 실험 결과이며, 또한 MSP 암호 영역으로부터 AUUUA mRNA 불안정성 모티프를 제거하면 COS 세포에서 MSP 단백질이 발현된다는 것을 최초로 입증한 것이다. 도 5, 패널 A에 나타낸 노던 분석 결과(즉, 천연 유전자에 의해서는 RNA가 전혀 관찰되지 않고, 본 발명에 따른 변형된 DNA 서열에 의해서는 적당한 농도가 관찰됨), 역시 단백질 생성이 증가됨이 나타나고 있다.
다음 실시예는 돌연변이 마우스의 유액에서 발현되는 천연 비융합(및 비글리코실화) 단백질로서의 MSP1-42에 대한 것이다.
MSP 돌연변이 유전자의 작제
15개 아미노산 β-카제인 시그널 펩티드에 MSP1-42를 융합시키기 위하여, 15개 아미노산-카제인 시그널 및 말단이 ClaI 부위인 MSP1-42의 처음 5개 아미노산을 암호하는 올리고쌍 MSP203과 MSP204(MSP203 : ggccgctcgacgccaccatgaaggtcctcataa ttgcctgtctggtggctctggccattgcagccgtcactccctccgtcat, MSP204 :cgatgacggagggagt gacggctgcaatggccagagccaccagacaggcaattatgaggaccttcatggtggcgtcgagc)를 BC620의 ClaI-XhoI 단편(MSP1-42 유전자의 나머지를 암호함)에 결찰시켜 발현 벡터 pCDNA3의 XhoI 부위에 삽입하였다. 이 플라스미드(GTC669)의 XhoI 단편을 유액 특이적 발현 벡터 BC350의 XhoI 부위에 클로닝하여 BC670을 생성하였다(도 9).
돌연변이 마우스의 유액내에서의 MSP1-42의 발현
플라스미드 BC670으로부터 SalI-NotI 단편을 제조하여 마우스 배(胚)에 미량주입하여 돌연변이 마우스를 제조하였다. 돌연변이 마우스는, 꼬리 생검으로부터 마우스 DNA를 추출한 뒤, 올리고 GTC17과 MSP101(GTC17 : GATTGACAAGTAATACGCTGTTTCCTC, MSP101 : GGATTCAATAGATACGG)을 사용하여 PCR 분석하여 동정하였다. 기초 돌연변이 마우스 암컷 유래의 유액을 유액분비 7일과 9일째 수집하고 웨스턴 분석하여, 폴리클로널 항MSP 항체와 모노클로널 항MSP 항체 5.2(데이비드 카슬로우 박사, NIH)로 MSP-1-42의 발현률을 측정하였다. 그 결과, 돌연변이 마우스의 유액에 발현된 MSP-1-42의 발현 농도가 1 내지 2 ㎎/㎖인 것으로 나타났다(도 10).
MSP1-42 글리코실화 부위 결실 돌연변이체의 작제
MSP를 생성하는 유액의 분석 결과, 돌연변이 MSP 단백질은 N-글리코실화되었다. MSP1-42 유전자 중의 N-글리코실화 부위를 제거하기 위하여, 181번과 262번의 Asn(N)을 Gln(Q)으로 치환시켰다. 이 치환은, MSP1의 대응 영역에 어닐링하고 AAC 대신 CAG 돌연변이를 보유하는 DNA 올리고를 디자인하여 도입시켰다. 이 올리고를 그 다음 PCR 프라이머로서 사용하여 N 대신 Q 치환체를 암호하는 DNA 단편을 생성하였다.
N262-Q 돌연변이를 도입하기 위하여, 올리고 쌍, MSPGYLYCO-3 (CAGGGAATGCTGCAGATCAGC) 및 MSP42-2 (AATTCTCGAGTTAGTGGTGGTGGTGGTGGTGATCGCAGAAA ATACCATG, 도 11)를 사용하여 플라스미드 GTC627(합성 MSP1-42 유전자 함유)을 PCR 증폭시켰다. 이 PCR 생성물을 pCR2.1 벡터(Invitrogen)에 클로닝하였다. 그 결과 플라스미드 GTC716을 얻었다.
N181-Q 돌연변이를 도입하기 위하여, 올리고 MSPGLYCO-1 (CTCCTTGTTCAGGAACTTGTAGGG) 및 MSPGLCO-2 (GTCCTGCAGTACACATATGAG, 도 4)를 사용하여 플라스미드 GTC627을 증폭시켰다. 이 PCR 생성물을 pCR2.1내로 클로닝하였다. 그 결과, 플라스미드 GTC700을 얻었다.
MSP 이중 글리코실화 돌연변이체는 다음과 같은 3 단계로 작제하였다. 먼저, BC670의 XhoI-BsmI 단편 및 GTC716의 BsmI-XhoI 단편을 벡터 pCR2.1의 XhoI 부위에 결찰시켰다. 그 결과, N262-Q 돌연변이를 보유한 MSP-1-42 유전자를 포함하는 플라스미드를 얻었다. 그 다음, 이 플라스미드의 EcoNI-NdeI 단편을 플라스미드 GTC716의 EcoNI-NdeI 단편으로 치환시켜 제2 돌연변이인 N181-Q를 도입시켰다. 마지막으로, 이 플라스미드의 XhoI 단편을 BC350내로 클로닝하여 BC718을 생성하였다(도 12).
비글리코실화 MSP1의 돌연변이적 발현
BC718은 다음과 같은 특성이 있다. 즉, 유액분비 동안 돌연변이 동물의 유선에서 발현될 수 있도록 β-카제인 프로모터의 제어하에 MSP1-42 유전자를 보유한다. 또한, N-말단에 어떤 부가 아미노산이 없는 MSP1-42가 유액으로 분비될 수 있도록 MSP1-42에 직접 융합된 15개 아미노산 β-카제인 리더 서열을 암호한다. 마지막으로, N-Q 치환으로 인하여, 이 작제물에 의해 돌연변이 동물의 유액에 생성되는 MSP는 N-글리코실화되지 않을 것이다. 이를 종합해 보면, BC718에 의해 유액에 생성되는 돌연변이 MSP는 기생체 MSP와 동일한 것이다.
플라스미드 BC718로부터 SalI/XhoI 단편을 제조한 뒤, 마우스 배로 미량주입하여 돌연변이 마우스를 제조하였다. 돌연변이 동물은 전술한 바와 같이 동정하였다. 기초 마우스 암컷 유래의 유액을 수집하고 항체 5.2를 사용하여 웨스턴 블롯 분석하였다. 그 결과는 도 13에 도시하였는데, 비글리코실화 MSP1 발현량이 0.5 내지 1 ㎎/㎖ 농도인 것으로 나타났다.
Claims (8)
- 서열 번호 1에 도시된 바와 같은 변형 MSP-1 핵산 서열.
- MSP-1을 암호화하는 쳔연 유전자의 전체 AT 함량을 낮추는 단계, 모든 mRNA 불안정성 모티프를 제거하는 단계 및 모든 희귀 코돈을 마우스 유선의 바람직한 코돈으로 대체하는 단계를 포함하여 변형 MSP-1 핵산을 제조하는 방법.
- 제2항에 기재된 방법으로 제조된 변형 MSP-1 핵산.
- 제1항에 기재된 변형 MSP-1 핵산과 염소 베타 카제인 프로모터 및 시그널 서열을 포함하는 벡터.
- 제4항에 기재된 벡터로 형질전환시킨 숙주 세포.
- 제4항에 기재된 벡터를 포함하는 인간을 제외한 돌연변이 포유류.
- 제1항에 기재된 변형 MSP-1 핵산을 포함하는 인간을 제외한 돌연변이 포유류.
- 제1항에 기재된 변형 핵산을 포함하는 DNA 백신.
