Abstract
본 발명은 VH-VL 작제물의 형태로 연결되어 궁극적으로는 펩타이드를 통하여 함께 연결되어 있는, 면역글로불린의 중쇄 및 경쇄의 다양한 가변 영역을 지니고 있는 단쇄의 다중 항원-결합성 분자, 이의 제조 방법, 및 약제 또는 진단 보조제로서의 이의 용도에 관한 것이다.
Description
본 발명은 VH-VL 작제물의 형태로 연결되어 궁극적으로는 펩타이드를 통하여 함께 연결되어 있는, 면역글로불린의 중쇄 및 경쇄의 다양한 가변 영역을 지니고 있는 단쇄의 다중 항원-결합성 분자, 이의 제조 방법, 및 약제 또는 진단 보조제로서의 이의 용도에 관한 것이다.
2가지 상이한 항원, 예를 들면, 종양 세포 표면 항원과 효능기(effector) 분자를 인식하는 이중 특이적(bispecific) 항체가 실험적인 면역 치료법에 광범위하게 사용되고 있다[참조: Fanger et al., Crit. Rev. Immunol. 12, 101-124(1992); van de Winkel et al., Immunol. Today 18, 562-564(1997)]. 이와 같이 이중 특이적 항체에 의해 점증되는 효능기 작용물로는, 예를 들면, 세포독성 세포 및 식세포, 보체 성분, 사이토킨, 및 혈전 붕괴성 및 섬유소 용해성 효소와 같이 체내에서 천연적으로 발생되는 것들 뿐만 아니라 외인성 효능기 분자, 예를 들면, 독소, 프로드럭(prodrug)-전환 효소 및 방사성핵종이 있다. 따라서, 예를 들면, 이종 이식된 종양(xenotransplanted tumor)에서 호지킨(Hodgkin) 종양 관련 항원 CD30과 T-세포 항원 CD3 및 CD28에 대한 이중 특이적 항체를 주입하면, 이는 세포독성 T 세포를 점증시키고 자극하여 항암 활성을 유도시킨다[참조: Renner et al., Science 264, 833, 1994]. 또 다른 접근법에서는, CD30과 알칼린 포스파타제에 대한 이중 특이적 항체를 사용하여, 상기 효소를 상기 종양 부위에 점증시킴으로써 약물의 비독성 전구체를 독성 약물로 전환시킨다[참조: Sahin et al., Cancer Res. 50, 6944-6948, 1990].
정제된 이중 특이적 항체를 주입하는 것에 대한 대체 방안은 시험관내 또는 생체내에서 형질감염된 세포에 의해 이들 이중 특이적 항체를 발현 및 분비시키는 것이다. 이러한 방법의 이점은 형질도입된 세포에 의한 이중 특이적 항체의 생산이 생체내에서 일어나기 때문에 주입 전에 이중 특이적 항체를 정성들여 생산하여 정제시킬 필요가 없다는 점이다. 또한, 적합한 발현 시스템을 선택함으로써, 이중 특이적 항체의 발현을 기관 또는 특정 종양에서 국부적으로 조절하거나 전신 조절하는 것이 가능하다.
이중 특이적 항체는, 예를 들면, 화학적 가교결합시킴으로써 제조할 수 있다[참조: Nisonoff et al., Nature 194, 355, 1962]. 동물 기원의 모노클로날 또는 폴리클로날 항체 분자를 화학적으로 가교결합시키면, 어떠한 적은 부분도 불활성화되지 않는다. 또한, 이종이량체(heterodimer)와 동종이량체(homodimer) 모두가 생성된다. 동종이량체는 정교한 공정에 의해 필요로 하는 이종이량체로부터 분리시켜야만 한다. 이중 특이적 항체를 생산하는, 하이브리드 하이브리도마로 불리우는 하이브리도마 세포는 비교적 정교한 방식으로만 생산될 수 있는데, 이는 이를 2가지 상이한 하이브리도마와 함께 융합시킬 필요가 있기 때문이다[참조: Milstein et al., Nature 305, 537, 1983]. 작용성 이종이량체의 비율이 이론상 단지 10% 정도로 비교적 낮은데, 이는 상기 2가지 항체 분자의 중쇄와 경쇄가 어떠한 방식으로든 서로 결합될 수 있기 때문이다. 사용된 출발 물질은 사람에 대한 면역원성인 쥐의 모노클로날 항체를 주로 포함한다.
각종 2가 또는 이중 특이적 항체 분자를 또한 재조합 기술을 이용하여 제조하고 세균성 또는 진핵성 세포에서 발현시킬 수 있다[참조: WO 93/06217]. 쥐 및 사람 기원 모두의 출발 분자를 사용하여 이들을 제조할 수 있다는 것은 모든 재조합 항체 분자에 해당된다. 이중 특이적 재조합 항체 분자를 가능한 한 효율적으로 제조하기 위한 각종 방법이 개발되었다. 이들 방법을 사용하여 각종 그룹의 분자를 제조할 수 있다.
한 그룹의 분자에서는, 이량체화를 달성하기 위해서, 항체의 가변 부분을 면역글로불린 불변 영역(Fc, CH3, CL)에 융합시킨다[참조: Hu et al., Cancer Res. 56, 3055-3061(1994); Hayden et al., Ther. Immunol. 1, 3-15(1994)]. 그러나, 이러한 경우에는, 이종이량체성 분자를 선별할 필요가 없기 때문에, 이러한 방식으로는 주로 2가 동종이량체가 생성된다. 더욱이, 이들 분자를 작용성 형태로 발현시키는 것은 진핵성 세포에 한정된다.
또 다른 그룹의 분자에서는, 항체의 가변 부분을 다른 단백질로부터의 펩타이드 또는 단백질 영역에 융합시켜 2가 또는 이중 특이적 분자를 제조한다[참조: Pluckthun and Pack, Immunotechnol. 3, 83-105(1997)]. 이러한 경우 역시, 랜덤한 결합으로 인해, 통상적으로 동종이량체와 이종이량체가 형성된다. 또한, 이들 분자는 일정 비율(몇몇 경우에서는 상당한 비율)의 외래 서열을 함유하므로, 현저한 면역원성이 예상될 수 있다.
세 번째 그룹의 분자에서는, 재조합 Fv 단편을 약간 변형시킨 것, 일반적으로 단쇄 Fv 단편(scFv)을 사용하여 2가 또는 이중 특이적 항체를 제조한다[참조: Holliger and Winter, Curr. Opin. Biotechnol. 4, 446-449(1993)]. 이러한 변형에는 scFv 쇄의 C 말단에서 부가의 시스테인을 통하여 이량체화하는 것이 포함된다[참조: MacCartney et al., Protein Engin. 8, 301-314(1994)]. 그러나, 이로써, 동종이량체와 이종이량체 모두가 생성되며, 세균성 세포에서의 발현시에는 비-작용성 집합체가 생성된다. 구조물 scFv(A)-링커-scFv(B)의 세로 일렬의 scFv 분자[참조: Mallender and Voss, J. Biol. Chem. 269, 199-206, 1994]를 세균 세포와 진핵 세포 모두에서 발현시킬 수 있다. 그러나, 이들 중 몇몇 경우에서는, 4개의 가변 영역이 비-작용적으로 결합될 수도 있다.
재조합 항체 기술은 최근 수 년간 신규의 작은 2가 또는 이중 특이적 항체 단편을 개발시켰다. 이러한 유형의 분자의 예는 "디아보디(diabody)"이다[참조: Holliger et al., Proc. Natl. Acad. Sci. USA 90, 6444-6448, 1993]. 이들은 매우 짧은 링커에 의해 함께 연결되어 있는, 면역글로불린의 가변 VH 영역과 VL 영역을 포함한다. 이러한 링커는 너무 짧아서, 단쇄 Fv 단편의 경우에 발생된 것과 같이, 특정 쇄의 VH 영역과 VL 영역이 결합하지 못한다. 이는 두 쇄의 VH 영역과 VL 영역이 결합되어 이량체를 형성하므로, 2개의 결합 부위를 갖는 분자가 생성된다는 것을 의미한다[참조: Perisic et al., Structure 2, 1217-1226, 1994]. 이중 특이적 "디아보디"는 구조물 VH(A)-VL(B) 및 VH(B)-VL(A)의 두 쇄를 한 세포에서 발현시킴으로써 생성시킨다. 이러한 경우, VL은 면역글로불린의 경쇄(L)의 가변(V) 영역을 의미하고 VH는 면역글로불린의 중쇄(H)의 가변(V) 영역을 의미하며, 이들 가변 영역은 항원(A) 또는 (B)에 결합된다. VH 부분을 VL 부분과 결합시키면 작용적으로 활성인 결합 부위를 갖는 이종이량체성 단편이 생성된다. 세균을 이용하여 발현시킨 이중 특이적 "디아보디"는 각종 효능기 분자, 면역글로불린, C1q 또는 효소 또는 효능기 세포, 예를 들면, 세포독성 T 임파구를 점증시키는데 성공적으로 사용된 바 있다[참조: Kontermann et al., Nature Biotechnol. 15, 629(1997); Holliger et al., Protein Engin. 9, 299(1996); Nature Biotechnol. 15, 632(1997); Zhu et al., BioTechnol. 14, 192(1996); FitzGerald et al., Protein Engin. 10, 1221(1997); Krebs et al., J. Biol. Chem. 273, 2858(1998)].
그러나, "디아보디"는 다음과 같은 단점을 지니고 있다: 두 VH(A)-VL(B) 및 VH(B)-VL(A) 쇄는 더 이상 물리적으로 함께 연결되지 않기 때문에, 동종이량체와 이종이량체가 동등한 정도로 생성될 수 있으며,이로써 이종이량체에 필요한 극히 정교한 정제 공정이 수반되야 한다. 또한, 기존에 scFv 단편에 대해 나타난 바와 같이, 이량체의 분리가 일어난다[참조: Glockshuber et al., Biochem. 29, 1362-1367(1990)]. 이러한 문제점을 해결하기 위해서, 디설파이드-안정화된 "디아보디"[참조: FitzGerald et al., Protein Engin. 10, 1221-1225, 1997] 또는 "크놉 인투 홀 디아보디(knob into hole diabody)"[참조: Zhu et al., Protein Sci. 6, 781-788, 1997]이 개발되었다. 그러나, 이의 제조 방법은 상당히 복잡하다. 또한, 이중 특이적 "디아보디"를 유전공학적으로 발현시키기 위해서는, 각 쇄에 대한 시그날 서열과 리보솜 결합 부위가 요구되는데, 이 역시 매우 복잡하다. 비-작용성 동종이량체의 비율을 증가시키는, 등몰량이 아닌 양의 가변 영역을 발현시키는 것이 또한 가능하다.
따라서, 본 발명의 목적은 앞서 기재된 소위 "디아보디"의 단점을 지니고 있지 않고 간단한 방식에 의해 주로 균일한 형태로 제조될 수 있는 다중 항원-결합성 분자를 개발하는 것이다.

도 6은 CEA 및 이. 콜라이(E. coli) β-갈락토시다제(CEAGal)에 대한 디아보디(diabody)와 비교해서, 본 발명에 따르는 단쇄의 이중 항원-결합성 단백질(scDb-CEAGal)을 통하여 플라스틱-결합된 CEA 상에 β-갈락토시다제를 점증시키는 것을 도시한 것이다. 미세역가판을 음성 대조군으로서 BSA로 피복시킨다.
도 7은 포유류 세포에 의해 분비된 scDb-CEAGal을 통하여 β-갈락토시다제를 점증시키는 것을 도시한 것인데, 여기서 상이한 양의 scDb-CEAGal와 β-갈락토시다제가 CEA-피복된 미세역가판에 이용된다. 이용된 기질은 o-니트로페닐 β-D-갈락토피라노시드이다. 미세역가판을 음성 대조군으로서 BSA로 피복시킨다.
도 8A는 본 발명에 따르는 단쇄의 이중 항원-결합성 단백질(scDb), β-갈락토시다제(β-Gal), 다우노마이신 β-D-갈락토피라노시드(프로드럭) 및/또는 다우노마이신(약물)의 존재하에서 LoVo 세포의 사멸을 도시한 것이다.
도 8B는 CEA-음성 세포주(A549)를 이용한 대조 실험을 도시한 것이다.
따라서, 본 발명의 한 국면은
a) 제1 특이성(A)을 지닌 면역글로불린의 중쇄의 가변 영역(VH) 및 이의 작용성 부분,
b) 제2 특이성(B)을 지닌 면역글로불린의 경쇄의 가변 영역(VL) 및 이의 작용성 부분,
c) 특이성(B)을 지닌 면역글로불린의 중쇄의 가변 영역(VH) 및 이의 작용성 부분, 및
d) 특이성(A)을 지닌 면역글로불린의 경쇄의 가변 영역(VL) 및 이의 작용성 부분을 포함하고, 상기 VH 영역과 VL 영역이 VH-VL 작제물 또는 VL-VH 작제물의 형태로 연결되어 있으며 2개의 VH-VL 작제물이 펩타이드(P)를 통하여 연결되어 있는 단쇄의 다중 항원-결합성 분자이다.
또 다른 양태에 있어서, 본 발명에 따르는 분자는 2개 초과의 상기 VH-VL 작제물(예: VH(B)-펩타이드-VL(B))을 포함한다. 이로써, 여러 개의 특이성 (A), (B), (C) 등을 지니고 있는 분자를 수득할 수 있다.
바람직한 양태에서는, 개개의 VH-VL 작제물을 동일한 특이성을 지닌 이들의 가변 영역을 통하여 결합시킨다. 본 발명에 따르는 분자의 특히 바람직한 구조가 도 1에 도시되어 있다.
또 다른 바람직한 양태에서는, 개개의 특이성이 본질적으로 동일하다. "본질적으로 동일한"이란 용어는 본 발명에 따르면, 상기 영역의 분자 구조는 상이하지만, 이들의 특이성, 즉 특이적 항원-결합 기능을 유지하는 것이 완벽하게 가능하다는 것을 의미한다. 이러한 결과는 본 발명에 따르는 분자가 면역글로불린의 중쇄 또는 경쇄의 완전한 가변 영역을 포함할 수 있을 뿐만 아니라 이의 작용성 부분, 즉 이들의 특이적 항원-결합 특성이 유지된 부분을 포함할 수 있다는 것이다. 예를 들면, 본 발명의 목적상, 면역글로불린의 가변 영역의 개개의 CDR 부분("상보적 결정 영역")으로 구성된 인위적인 작제물도 본 발명에 포함된다[참조: Jones, P.T. et al., Nature, 321, 522, 1986; Riechmann, L. et al., Nature 332, 323, 1988; Verhoeyen, M. et al., Science, 239, 1534, 1988].
바람직한 양태에서는, VH 영역과 VL 영역이 VH-L-VL 작제물 또는 VL-L-VH 작제물의 형태로 펩타이드 링커(L)를 통하여 연결된다. 이러한 경우, 상기 링커는 일반적으로 가능한 한 짧아야 하고, 바람직하게는 약 1 내지 20개 아미노산, 특히 약 1 내지 5개 아미노산 길이이다. 서열 GGGGS을 갖는 링커가 특히 바람직하다.
링커(L)와는 대조적으로, 연결성 펩타이드(P)는 적합한 어떠한 길이를 가질 수 있다. 그러나, 펩타이드(P)는 바람직하게는 약 12 내지 40개 아미노산, 특히 약 12 내지 20개 아미노산, 특히 약 14개 아미노산을 갖는다. 특히 바람직한 펩타이드(P)는 아미노산 서열 GGGGSGGRASGGGS 또는 GGGGSGGRASGGGGS을 포함하고 상기 아미노산 서열로 이루어진 펩타이드(P)가 특히 바람직하다.
또 다른 바람직한 양태에 있어서, 본 발명에 따르는 분자는 추가의 성분으로서 효능기(E)를 포함한다. 이러한 효능기는 본 발명에 따르는 분자에 직접적으로 연결되거나, 또는 경우에 따라 연결기(B)를 통하여 연결될 수 있다. 연결기(B)가 프로테아제 절단 서열, 바람직하게는 PSA(전립선-특이적 항원), 카텝신, 플라스미노겐 및/또는 플라스미노겐 활성인자 절단 서열을 포함하는 것이 특히 바람직하다.
이러한 프로테아제 절단 서열이 특히 바람직한데, 이는 프로테아제의 사용에 의해 효능기를 본 발명에 따르는 분자로부터 분리시킬 수 있기 때문이다. 이러한 분리는 효능기의 활성이 연결기를 통하여 본 발명에 따르는 분자에 직접적으로 또는 간접적으로 연결되는 효능기로 인해 억제되는 경우에 특히 유리하다.
프로테아제는 특히 염증 발생 영역이나 종양에 존재하기 때문에, 프로테아제 절단 서열을 통하여 본 발명에 따르는 분자에 대한 결합으로 인해 바람직하게는 불활성화되는 효능기는 광범위하게 국소적으로 방출된다. 또한, 본 발명에 따르는 분자, 예를 들면, 종양 세포 상의 항원, 종양 관련 내피 세포 또는 염증성 세포, 예를 들면, 임파구 또는 매크로파지 상의 항원에 대한 특이성을 지니고 있는 분자에 적합한 표적 구조물을 선택함으로써, 효능기를 갖는 본 발명에 따르는 분자를, 예를 들면, 염증 발생 영역 또는 종양에 축적시키는 것이 가능하다.
본 발명에 따라서 수 개의 효능기를 본 발명에 따르는 분자에 직접적으로 또는 간접적으로 결합시킬 수 있다.
프로테아제 또는 관련 절단 서열은, 예를 들면, DE 1970430.1에 상세히 기재되어 있다.
연결기를 통하여 당해 분자에 결합되는 효능기를 갖는 본 발명에 따르는 바람직한 분자의 한 예가 도 2에 도식적으로 제시되어 있다.
본 발명에 따르는 분자 중의 개개의 성분에 대한 선택은 일반적으로 적용 분야에 따라 결정된다.
본 발명에 따르는 분자를 진단 보조제로서 사용하고자 하는 경우에는, 예를 들면, 추가의 양태에서, 제1 특이성(A)은 분석하고자 하는 분자에 대해 지시되고 제2 특이성(B)는 직접 또는 간접적으로 분석물에 대해 지시된다. 이러한 분석물은, 예를 들면, 방사능 분자, 형광성 분자, 또는 효소적 활성을 통하여 분석물의 전구체를 활성 분석물로 전환시켜 주는 효소일 수 있다.
또 다른 바람직한 양태에서는, 제1 특이성(A)이 분석하고자 하는 분자에 대해 지시되고 제2 특이성(B)은 분석하고자 하는 또 다른 분자에 대해 지시되며 효능기(E)가 분석물, 예를 들면, 방사능 분자, 형광성 분자 및/또는 앞서 설명된 바와 같은 효소이다.
그러나, 본 발명에 따르는 분자는 또한, 바이러스성 또는 비-바이러스성 벡터의 표적 세포-특이적 결합용 리간드로서 사용될 수 있다. 바람직한 양태에서는, 본 발명에 따르는 분자가 소위 다작용성 리간드로서 사용될 수 있다. 이를 위해서는, 펩타이드(P) 및/또는 효능기(E)가 퓨소제닉(fusogenic) 펩타이드를 포함하는 경우가 유리하다. 이러한 다작용성 리간드는 뉴클레오타이드 서열의 표적 세포-특이적 전이에 이용되고, 일반적으로 표적 세포-특이적 리간드, 유전자 작제물-특이적 리간드 및 퓨소제닉 펩타이드를 포함하는 단백질이다. 이러한 유형의 다작용성 리간드는 유전자 작제물, 즉 핵산 작제물을 표적 세포에 특이적으로 결합시키는데 사용될 수 있으며, 퓨소제닉 펩타이드는 이러한 핵산 작제물이 세포막을 통해 세포핵 내로 침투되어 엔도솜(endosome)으로부터 방출될 수 있게 해준다.
본 발명에 따르는 분자를 벡터용 리간드로서 사용하거나 다작용성 리간드로서 사용하는 경우에는, 제1 특이성(A)이 표적 세포에 대해 지시되고 제2 특이성(B)이 벡터에 대해 지시되는 것이 유리하다. 이러한 벡터는 일반적으로 핵산, 양이온성 펩타이드 또는 단백질, 양이온성 지질, 양이온성 펩타이드 또는 양이온성 포르피린이다. 본 발명의 특정 양태에서는, 상기 벡터가, 예를 들면, 아데노바이러스(AdV), 아데노-관련 바이러스(AAV), 백시니아 바이러스, RSV, HSV, 인플루엔자 바이러스 또는 렌티바이러스로부터 유도된 바이러스성 벡터이다.
표적 세포-특이적 리간드, 표적 세포 상의 막 구조물, 면역글로불린으로부터 유도된, 즉 VH 영역과 VL 영역을 포함하는 표적 세포-특이적 리간드 및 유전자 작제물-특이적 리간드의 예 뿐만 아니라 퓨소제닉 특성을 지닌 펩타이드의 예가 DE 19649645.4에 기재되어 있다.
본 발명에 따르는 분자는 또한 예방제 및/또는 치료제로서 적합하다.
이를 위하여, 예를 들면, 제1 특이성(A)는 세포막, 예를 들면, 임파구, 매크로파지, 단구, 과립구, 조혈 세포, 내피 세포, 평활근 세포, 횡문근(striped muscle) 세포, 상피 세포, 간 세포, 신장 세포, 신경교 세포, 지지성 조직의 세포, 종양 세포 또는 백혈병 세포; 세포외 매트릭스의 단백질, 보체 시스템의 단백질, 응고 시스템의 단백질, 키닌 시스템의 단백질, 혈장의 단백질, 지지성 조직의 단백질; 사이토킨 또는 케모카인; 내인성 또는 외인성 독소; 약제, 예를 들면, 디지탈리스(digitalis) 및/또는 병원체, 예를 들면, 세균성, 바이러스성 및/또는 기생충 병원체에 대해 지시된다.
제2 특이성(B)은, 예를 들면, 임파구, 매크로파지, 단구 또는 과립구의 세포막에 대해 지시될 수 있으며, 그 결과 이러한 세포막이 표적 구조물과 가교결합되어 세포독성, 면역조절성 또는 염증성 진행 과정을 유발시킬 수 있다.

바람직한 양태에서는, 본 발명에 따르는 핵산이 출발 코돈의 5' 말단에 해독을 증진시킬 수 있는 서열 GCCACC 또는 GCCGCC를 포함한다.
본 발명의 또 다른 양태는 본 발명에 따르는 핵산을 포함하는 벡터에 관한 것이다. 이러한 경우, 상기 벡터는 특히 양이온성 지질, 양이온성 중합체, 양이온성 펩타이드 또는 양이온성 포르피린 중에서 선택되는, 바이러스성 또는 비-바이러스성 벡터, 바람직하게는 비-바이러스성 벡터일 수 있다.

그러나, 약물을 본 발명에 따르는 분자를 발현하는 세포의 세포막에 정착시키기 위해서, 글리코포스포리피드 앵커를 암호화하는 뉴클레오타이드 서열을 핵산 작제물에 삽입시킬 수도 있다.
글리코포스포리피드 앵커의 삽입은 일반적으로 본 발명에 따르는 분자를 암호화하는 뉴클레오타이드 서열의 3' 말단에서 일어나며, 시그날 서열의 삽입에 덧붙여 일어난다.
예를 들면, CEA, N-CAM 및 기타 막 단백질, 예를 들면, Thy-1에 대한 글리코포스포리피드 앵커가 기재되어 있다.
이러한 막관통 영역으로 인해, 특정한 세포가 세포막 상에 본 발명에 따르는 분자를 발현시키므로, 본 발명에 따르는 분자의 항원-결합 부분의 의해 인식되는 표적 또는 효능기 구조물에 대해 특이적인 "수용체"가 획득된다.

본 발명의 목적상 바람직한 프로모터 또는 활성인자 서열에는 특히 암세포 또는 육종 세포에서 생성되는 단백질을 암호화하는 유전자의 유전자-조절 서열 또는 요소가 포함된다. 따라서, 작은 세포 기관지 암종에 바람직하게 사용되는 것은 N-CAM 단백질의 프로모터이고, 난소 암종에 바람직하게 사용되는 것은 간염 성장 인자 수용체 또는 L-플라스틴의 프로모터이며, 췌장 암종에 바람직하게 사용되는 것은 L-플라스틴 또는 다형성 상피 점소(PEM)의 프로모터이다.
본 발명의 상당한 이점은 개개의 성분의 연결이 이종이량체성 결합에 유리하기 때문에 단쇄의 다중 항원-결합성 분자가 우점적으로 형성된다는 것이다. 또한, scFv 단편에 대해 제시된 바와 같이, 이량체의 분해가 감소된다[참조: Glockshuber et al., Biochem. 29, 1362-1367, 1990]. 따라서, 소위 "디아보디"가 디설파이드-안정화되거나 "크놉 인투 홀 디아보디"의 형태인 경우일지라도, 이러한 디아보디보다 덜 복잡하고 더 균일한 형태로 본 발명에 따르는 분자를 제조할 수 있다.

종양 치료법

선택된 활성 물질은 일반적으로, 병원체에 의해 생성되고 면역 반응물 주입에 의해, 즉 항체 결합 및/또는 세포독성 T-임파구를 통하여 병원체의 중화 및/또는 사멸을 유도하는 단백질의 DNA이다. 이러한 유형의 소위 중화 항원은 이미 백신 항원으로서 사용되고 있다[참조: Ellis, Adv. Exp. Med. Biol. 327, 263(1992)].

상기 세균을 파열시키고 단쇄의 이중 항원-결합성 단백질을 고정화된 금속 친화성 크로마토그래피(IMAC)하여 외질(periplasmic) 제제로부터 정제한다[참조: Hochuli et al., Bio/Techn. 6, 1321-1325(1988)]. 이러한 방법의 상세한 내역이 문헌[참조: Kontermann et al., (1997)]에 기재되어 있다.
이와 같이 정제된 단백질은 SDS 폴리아크릴아미드 겔 전기영동 및 겔 여과에 의해 제시된 바와 같이 분자량이 60kDa이다. 세균성 배양액 1리터 당 상기 분자 약 200 내지 300㎍을 정제하는 것이 가능하다. 세균적으로 발현된 단쇄의 이중 항원-결합성 단백질은 ELISA에서 CEA 및 β-갈락토시다제와 반응하고, o-니트로페닐 β-D-갈락토피라노시드의 전환에 의해 제시된 바와 같이, 상기 효소를 플라스틱-결합된 CEA에 점증시킬 수 있다(도 6). 또한, 이러한 단쇄의 이중 항원-결합성 단백질은 β-갈락토시다제를 점증시키고 기질 5-브로모-4-클로로-3-인돌릴 β-D-갈락토피라노시드(X-Gal)를 전환시킴으로써 고정된 및 생 CEA-발현 LoVo 세포를 염색시킬 수 있다. 이를 위하여, LoVo 세포를 적절한 항원-결합성 단백질 10㎍/ml, β-갈락토시다제 10㎍/ml 및 X-Gal(PBS, 3mM 시안화칼륨 철(Ⅲ), 3mM 시안화칼륨 철(Ⅱ) 중의 0.8mg/ml)와 함께 배양한다. 각종 대조군 세포(A549, HEK293)나 계란 라이소자임 및 β-갈락토시다제에 대한 디아보디(HELGal; Kontermann et al., (1997)]의 경우에는 염색이 전혀 관찰되지 않았다.
실시예 2
단쇄의 이중 항원-결합성 단백질의 진핵성 발현
진핵성 세포에서 발현하기 위하여, 단쇄의 이중 항원-결합성 단백질의 암호화 영역을 진핵성 발현 벡터(pSecTagA, Invitrogen) 내로 클로닝시키는데, 여기서 세균성 시그날 서열은 상기 벡터에 이미 존재하고 있는 Ig-κ 시그날 서열로 대체시킨다.
이를 위하여, 상기 단쇄 작제물을 프라이머 LMB2 및 pelB-Metminus를 사용하여 증폭시킴으로써, pelB 리더 내의 메티오닌(위치 21)을 트레오닌으로 대체시킨다. 이어서, Sfil 및 EcoRI로 가수분해시키고 벡터 pSecTagA 내로 클로닝시킨다. 이러한 성숙한 단쇄의 항원-결합성 단백질은 VHA 영역의 N 말단에 7개의 부가의 아미노산(AAQPATA)를 함유한다.
상기 플라스미드를 리포펙타민(Gibco)을 사용하여 진핵성 HEK 293 세포 내로 일시적으로 형질감염시킨다. 제오신의 존재하에서 안정한 세포를 선별한다. 이들 세포를 사용한 면역형광성 실험에서 소포체 ER 및 골기체(Golgi apparatus)의 염색을 검출하는 것이 가능하다. 또한,35S-Met-표지된 세포의 상등액으로부터의 60kDa 단백질의 면역침강법 및 고정화된 금속 친화성 크로마토그래피(IMAC)에 의한 정제에 의해 상기 단쇄의 이중 항원-결합성 단백질의 분비를 추가로 검출할 수 있다. 이와 같이 정제된 단백질은 β-갈락토시다제 및 CEA-피복된 미세역가판의 결합에 있어서 작용적으로 활성이고 또한 β-갈락토시다제를 플라스틱-결합된 CEA에 점증시키는데 있어서 작용적으로 활성이다(도 7).
실시예 3
단쇄의 이중 항원-결합성 단백질을 분비하는 세포와의 공동 배양에 의한 시험관내 효소 점증
당해 단쇄의 이중 항원-결합성 단백질을 생산하는 HEK 293 세포와 CEA-양성 LoVo 세포를 공동배양함으로써 점증을 시험관내에서 조사한다. 이를 위하여, 먼저, 본 발명에 따르는 단백질을 생산하는 세포를 트랜스웰 세포 배양 디쉬(Costar)에서 LoVo 세포와 함께 배양하는데, 여기서 2개의 세포주가 막에 의해 격리된다. 2일 후, β-갈락토시다제(10㎍/ml)를 가하고, 기질 X-Gal을 가함으로써 점증을 검출한다. 당해 단쇄의 이중 항원-결합성 단백질을 생산하는 세포와의 공동 배양에서는 특이적 염색이 발견된 반면, 형질감염되지 않은 HEK 293 세포를 사용한 대조 실험에서는 어떠한 염색도 나타나지 않았다.
LoVo 세포를 A549 세포(CEA-음성)로 대체시킨 실험, 및 당해 단쇄의 이중 항원-결합성 단백질을 생산하는 세포를 효소 및 기질과 함께 배양한 실험도 마찬가지로 음성이다. 후자는 당해 단쇄의 이중 항원-결합성 단백질을 생산하는 세포가 그 자체로는 상기 효소와 결합할 수 없다는 것을 예시하고 있다.
또 다른 실험에서는, 당해 단쇄의 이중 항원-결합성 단백질을 분비하는 HEK 293 세포, 및 LoVo 세포를 1:4 내지 1:24의 접종비로 직접적으로 공동배양한다. HEK 293 세포를 LoVo 세포와 구별하기 위해서는, LoVo 세포를 가하기 전에 HEK 세포를 CM-Dil(Molecular Probes)로 염색시킨다(레드 형광). 2일 후, β-갈락토시다제를 가하고, X-Gal(10㎍/ml)을 가한 다음 항-β-갈락토시다제 항체(Biotrend)로의 직접적인 면역형광에 의해 세포에 대한 결합을 검출한다. 이들 실험은 또한, 상기 효소가 LoVo 세포에 특이적으로 점증된다는 것을 나타내는 반면, HEK 293 세포는 전혀 염색되지 않았다. 형질감염시키지 않은 HEK 293 세포를 이용한 대조 실험은 음성이다. 이들 실험은 당해 단쇄의 이중 항원-결합성 단백질이 종양 세포를 특이적 및 선택적으로 인식한다는 것을 입증해준다.
추가의 실험에서는, 비독성 다우노마이신 β-D-갈락토피라노시드를 세포독성 다우노마이신으로 전환시키는 것에 관해 조사하였다. LoVo 세포를 정제된 단쇄의 이중 항원-결합성 단백질(10㎍/ml)와 함께 1시간 동안 배양한 다음 연속적으로 β-갈락토시다제(1㎍/ml) 및 다우노마이신 β-D-갈락토피라노시드(2μM)와 37℃에서 1시간 동안 배양함으로써, 다우노마이신과의 직접적인 배양으로 인한 염색과 비교해서, 기질의 자가형광에 의해 핵에서 생성된 다우노마이신의 특이적인 국재화를 검출할 수 있다. 단쇄의 이중 항원-결합성 단백질 또는 β-갈락토시다제의 부재하에서 및 대조군 세포(HEK 293; 당해 단쇄의 이중 항원-결합성 단백질을 생산하는 HEK)를 사용한 경우에는 어떠한 핵 염색도 검출할 수 없었다. 이들 실험은 당해 단쇄의 이중 항원-결합성 단백질이 효소를 종양 세포에 점증시킬 수 있으며, 이러한 효소가 비독성 전구체를 독성 물질로 전환시키는데 이용될 수 있다는 것을 입증해준다.
이러한 효과를 추가의 실험에 사용하여 종양 세포를 특이적으로 사멸시킬 수 있다.
이를 위하여, LoVo 세포를 96웰 판에서 단쇄의 이중 항원-결합성 단백질(10㎍/ml)과 배양한 다음 37℃에서 1시간 동안 다우노마이신 β-D-갈락토피라노시드(5μM)와 배양한다. 2일 후에 WST 시험(Boehringer Mannheim)을 사용하여 세포의 사멸을 분석한다(도 8). 이러한 경우, 프로드럭이 약물로 전환할 때의 세포 사멸은 대략적으로 약물의 존재하에서 만큼이나 우수하다(도 8A). 한 성분의 부재하 또는 CEA-음성 A549 세포를 사용한 경우에는 어떠한 효과도 거의 발견되지 않았다(도 8B).
실시예 4
단쇄의 이중 항원-결합성 단백질의 세포내 발현을 위한 작제물의 제조
당해 단쇄의 이중 항원-결합성 단백질의 세포내 발현을 위해, 이러한 단쇄의 이중 항원-결합성 단백질에 대한 유전자의 DNA를 각종 프라이머로 증폭시킨다:
-막관통 단쇄의 이중 항원-결합성 단백질(TM-scDAP). 여기에는 상기 유전자의 3' 말단에 PDGFR의 막관통 영역을 삽입시키는 프라이머가 사용된다
-(LPFKVVVISAIIALVVLTIISLIILIMLWQKKPRYES)
-소포체에 국재된 단쇄의 이중 항원-결합성 단백질(ER-scDAP). 여기에는 상기 유전자의 3' 말단에 ER 보유 시그날(SEKDEL)을 삽입시키는 프라이머가 사용된다.
-세포질에 국재된 단쇄의 이중 항원-결합성 단백질(cyto-scDAP). 여기에는 상기 유전자의 5' 말단에서 시그날 서열을 메티오닌 및 최적 해독 개시용 코작 서열로 대체시키는 프라이머가 사용된다.
-핵에 국재된 단쇄의 이중 항원-결합성 단백질(nuc-scDb). 여기에는 상기 유전자의 5' 말단에서 시그날 서열을 메티오닌 및 최적 해독 개시용 코작 서열로 대체시키고 상기 유전자의 3' 말단에 핵 국재화 서열(PKKKRKVGGGT: 핵 국재화 서열은 밑줄쳐 있다)을 삽입시키는 프라이머가 사용된다.
이들 단편을 적합한 진핵성 발현 벡터(pSecTagA 및 pcDNA3; Invitrogen)에 클로닝시킨다. 이어서, 이로써 생성된 작제물(TM-scDAP, ER-scDAP, cyto-scDAP, nuc-scDAP, 및 sec-scDAP(=분비된 단백질; 실시예 2 참조))을 일시적으로 진핵성 세포(3T3)에 형질감염시키고, 항-Myc 에피토프 항체를 사용하여 면역형광성에 의해, 발현된 단백질의 국재화를 조사한다. 작제물 sec-scDAP, ER-scDAP 및 TM-scDAP에 대해서는, 분비 경로에 전형적인 염색을 검출할 수 있는 반면, cyto-scDAP는 세포질에서 산만한 국재화를 나타내고 nuc-scDAP는 핵에서 국재화를 나타낸다.
본 발명에 따라 간단한 방식에 의해 다중 항원-결합성 분자가 제공된다.
Claims (37)
- a) 제1 특이성(A)을 지닌 면역글로불린의 중쇄의 가변 영역(VH) 및 이의 작용성 부분,b) 제2 특이성(B)을 지닌 면역글로불린의 경쇄의 가변 영역(VL) 및 이의 작용성 부분,c) 특이성(B)을 지닌 면역글로불린의 중쇄의 가변 영역(VH) 및 이의 작용성 부분, 및d) 특이성(A)을 지닌 면역글로불린의 경쇄의 가변 영역(VL) 및 이의 작용성 부분을 포함하고, 상기 VH 영역과 VL 영역이 VH-VL 작제물 또는 VL-VH 작제물의 형태로 연결되어 있으며 2개의 VH-VL 작제물이 펩타이드(P)를 통하여 연결되어 있는 단쇄의 다중 항원-결합성 분자.
- 제1항에 있어서, 2개 초과의 VH-VL 작제물을 포함하는 단쇄의 다중 항원-결합성 분자.
- 제1항 또는 제2항에 있어서, VH-VL 작제물이 동일한 특이성을 지닌 영역을 통하여 연결되어 있는 단쇄의 다중 항원-결합성 분자.
- 제1항 내지 제3항 중의 어느 한 항에 있어서, 특이성(A)와 (B)가 본질적으로 동일한 단쇄의 다중 항원-결합성 분자.
- 제1항 내지 제4항 중의 어느 한 항에 있어서, VH 영역과 VL 영역이 VH-L-VL 작제물 또는 VL-L-VH 작제물의 형태로 펩타이드 링커(L)를 통하여 연결되는 단쇄의 다중 항원-결합성 분자.
- 제5항에 있어서, 링커(L)가 약 1 내지 20개 아미노산, 바람직하게는 약 1 내지 5개 아미노산 길이인 단쇄의 다중 항원-결합성 분자.
- 제5항에 있어서, 링커(L)가 아미노산 서열 GGGGS를 포함하는 단쇄의 다중 항원-결합성 분자.
- 제1항 내지 제7항 중의 어느 한 항에 있어서, 펩타이드(P)가 약 12 내지 40개 아미노산, 바람직하게는 약 12 내지 20개 아미노산, 특히 약 14개 아미노산 길이인 단쇄의 다중 항원-결합성 분자.
- 제8항에 있어서, 펩타이드(P)가 아미노산 서열 GGGGSGGRASGGGS 또는 GGGGSGGRASGGGGS를 포함하는 단쇄의 다중 항원-결합성 분자.
- 제1항 내지 제9항 중의 어느 한 항에 있어서, 추가의 성분으로서 하나 이상의 효능기(effector; E)(들)를 포함하는 단쇄의 다중 항원-결합성 분자.
- 제10항에 있어서, 효능기(E)가 연결기(B)를 통하여 분자에 연결되는 단쇄의 다중 항원-결합성 분자.
- 제11항에 있어서, 연결기(B)가 프로테아제 절단 서열, 바람직하게는 PSA, 카텝신, 플라스미노겐 및/또는 플라스미노겐 활성인자 절단 서열을 포함하는 단쇄의 다중 항원-결합성 분자.
- 제1항 내지 제12항 중의 어느 한 항에 있어서, 제1 특이성(A)이 분석하고자 하는 분자에 대해 지시되고, 제2 특이성(B)이 분석물에 대해 지시되는 단쇄의 다중 항원-결합성 분자.
- 제13항에 있어서, 분석물이 방사능 분자, 형광성 분자 및/또는 효소인 단쇄의 다중 항원-결합성 분자.
- 제10항 내지 제12항 중의 어느 한 항에 있어서, 제1 특이성(A)이 분석하고자 하는 분자에 대해 지시되고, 제2 특이성(B)이 분석하고자 하는 또 다른 분자에 대해 지시되며, 효능기(E)가 분석물인 단쇄의 다중 항원-결합성 분자.
- 제15항에 있어서, 분석물이 방사능 분자, 형광성 분자 및/또는 효소인 단쇄의 다중 항원-결합성 분자.
- 제1항 내지 제12항 중의 어느 한 항에 있어서, 펩타이드(P) 및/또는 효능기(E)가 퓨소제닉(fusogenic) 펩타이드를 포함하는 단쇄의 다중 항원-결합성 분자.
- 제1항 내지 제17항 중의 어느 한 항에 있어서, 제1 특이성(A)이 표적 세포에 대해 지시되고, 제2 특이성(B)이 벡터에 대해 지시되는 단쇄의 다중 항원-결합성 분자.
- 제18항에 있어서, 벡터가 핵산, 양이온성 펩타이드 또는 단백질, 양이온성 지질, 양이온성 중합체, 양이온성 포르피린, 또는 AdV, AAV, 백시니아, RSV, HSV, 인플루엔자 또는 렌티바이러스 벡터로 이루어진 그룹 중에서 선택된 바이러스성 벡터인 단쇄의 다중 항원-결합성 분자.
- 제1항 내지 제12항 중의 어느 한 항에 있어서, 제1 특이성(A)이 세포막, 특히 임파구, 매크로파지, 단구, 과립구, 조혈 세포, 내피 세포, 평활근 세포, 횡문근(striped muscle) 세포, 상피 세포, 간 세포, 신장 세포, 신경교 세포, 지지성 조직의 세포, 종양 세포 또는 백혈병 세포; 세포외 매트릭스의 단백질, 보체 시스템의 단백질, 응고 시스템의 단백질, 키닌 시스템의 단백질, 혈장의 단백질, 지지성 조직의 단백질; 사이토킨 또는 케모카인; 내인성 또는 외인성 독소; 약제, 특히 디지탈리스(digitalis) 및/또는 병원체, 예를 들면, 특히 세균성, 바이러스성 및/또는 기생충 병원체에 대해 지시되는 단쇄의 다중 항원-결합성 분자.
