本発明では、組換えヒトPD-1タンパク質を使用してマウスを免疫し、次いで、組換えヒトPD-1に高親和性で特異的に結合するM944 scFv抗体クローンをファージ抗体ライブラリースクリーニングにより得た。次いで、M944 scFv抗体の重及び軽鎖可変領域をコードするヌクレオチド配列を、マウスIgG1重鎖定常領域及びマウスカッパ軽鎖定常領域をそれぞれコードするヌクレオチド配列を用いたPCRにより構築し、生じる構築した配列を、一過性発現ベクターに挿入し、HEK-293上で培養して発現させた。高純度マウス抗体PD1-M944を、プロテインA精製カラムを使用して精製した。ELISAにより、マウス抗体PD1-M944がPD-1のリガンドへの結合を遮断可能であったことが示された。
また、本発明は、本発明の抗体又はその部分をコードする核酸分子に関する。このような核酸分子の配列は、配列番号3~7及び26~33を含むが、これらに限定されない。
また、本発明では、1つ又は複数の本発明のヌクレオチド配列を含む組換え構築物を提供する。本発明の組換え構築物は、ベクターと共に使用することができ、このベクターは、例えば、プラスミド、ファージミド、ファージ又はウイルスベクターであり、本発明の抗体をコードする核酸分子をこのベクターに挿入する。
このPD1-H944抗体の特性及び機能解析を実施した。解析では、本発明の抗体が、次の利点を有することが示された。(1)ヒトPD-1に高親和性及び特異性で結合することができ解離速度が低いため、良好な抗腫瘍有効性がもたらされる。(2)ニボルマブよりも完全にPD-L1のPD-1への結合を遮断することができる。(3)ニボルマブと比較した場合、より感受性かつより特異性に組換えヒトPD-1に結合し、マウスPD-1には同様に結合しないが、組換えサルPD-1には同等に交差結合する。(4)組換えヒトPD-1のリガンドPd-L1及びPd-L2への結合を有効に遮断可能である。(5)免疫抑制されたT細胞を有効に再活性化、及び組換えヒトPD-1受容体細胞系を活性化可能である。(6)MC38結腸がん担がんヒト化PD-1マウスモデルにおいて、ニボルマブ(Sino Biological, Inc.社)のそれよりも遥かに良好かつペムブロリズマブ(Sino Biological, Inc.社)のそれと同等に、優れた腫瘍抑制作用を示す。(7)低いADCC及びCDCの両活性を示し、その上、ADCC活性は、ニボルマブのそれよりも低い。
本発明の抗体を使用し、異常発現するPD-1受容体を特異的に標的として、多様な腫瘍若しくはがんを治療するか又は多様な腫瘍若しくはがんの治療のための医薬を調製することができる。この腫瘍又はがんの非限定的な例としては、結腸がんが挙げられる。
本発明の抗体は、少なくとも1つの他の薬剤(例えば、安定化化合物)を用いて調製し、本発明の抗体及び1つ又は複数の薬学的に許容される担体、希釈剤又は賦形剤を含む医薬組成物を形成し得る。
また、本発明は、1つ又は複数の容器を含む、医薬パッケージ及びキットに関し、この容器は、前述の本発明の医薬組成物を含む。このような容器に付随するものは、薬物又は生物製剤の製造、使用又は流通を管理する政府機関により指定された形態の、この製剤を製造、使用又は流通させる代理店によるヒト投与の認可を反映する仕様書であり得る。
本発明の医薬組成物は、当技術分野において公知の方法、例えば、従来の混合、溶解、造粒、錠剤調製、粉砕、乳化、封入、包埋又は凍結乾燥方法により調製することができる。
また、上記の本発明の抗体を含む医薬組成物は、1つ又は複数の他の治療薬、例えば、抗悪性腫瘍薬と組み合わせ、この場合、生じる組合せ物により許容不可能な有害作用は発生しない。
ファージ抗体提示ライブラリーを使用した、マウス由来抗体によるPD-L1/PD-L2へのPD-1結合の遮断についてのスクリーニング
1.1 マウスの免疫
組換えヒトPD-1タンパク質(Sino Biological, Inc社、Cat. 10377-H08H)を使用してマウスを免疫した。このヒトPD-1タンパク質(UniProtKB Q15116)の細胞外領域のアミノ酸配列は、Met1-Gln167(配列番号1)である。
TriPure Isolation Reagent (Roche社)を使用してRNAをマウス脾臓組織から抽出し、逆転写キット(Invitrogen社)を使用した逆転写によりcDNAを得た。マウス抗体の軽及び重鎖可変領域をコードする各ヌクレオチド配列を増幅し、このscFvをコードするヌクレオチド配列をオーバーラップ伸長PCR方法により構築した。この場合、軽及び重鎖可変領域をコードする各ヌクレオチド配列を、リンカーTCTAGTGGTGGCGGTGGTTCGGGCGGTGGTGGAGGTGGTAGTTCTAGATCTTCC(配列番号2)を介して結合させ(参照: Rapid PCR-cloning of full-length mouse immunoglobulin variable regions; Cloning immunoglobulin variable domains for expression by the polymerase chain reaction T JonesらBio/Technolgy 9(6):579,July 1991)、次いで、制限エンドヌクレアーゼSfi I(Fermentas社)によりファージベクターpComb3x(Sino Biological, Inc.社)に酵素的にライゲーションし、次いで、コンピテントX-Blueに電気的形質転換を行って免疫マウスのファージ提示scFv抗体ライブラリーを構築した。組換えヒトPD-1タンパク質をELISAプレート上にコーティングすることにより、ファージ抗体パニングのプロセス後に抗PD-1陽性抗体濃縮ファージライブラリーを得た(参照: Antibody Phage Display: Methods and Protocols, Philippa M. O'Brien and Robert Aitken (Eds), Humana Press, ISBN: 9780896037113)。モノクローナルファージを濃縮ライブラリーから選択して発現させ、次いで、これらの組換えヒトPD-1タンパク質への結合をELISAにより試験した。組換えヒトPD-1に特異的に結合する、高度に結合する抗体M944 scFvのクローンを選択し、シーケンシングを行う会社に委託してM944 scFv抗体のヌクレオチド配列を配列決定した(配列番号3)。
M944 scFv抗体の重及び軽鎖可変領域のヌクレオチド配列(配列番号4/5)を、それぞれマウスIgG1重鎖定常領域のヌクレオチド配列(配列番号6)及びマウスカッパ軽鎖定常領域のヌクレオチド配列(配列番号7)を用いてPCRにより構築した。次いで、生じるヌクレオチド配列をHind III及びXba I(Fermentas社)により酵素的に切断して一過性発現ベクターpSTEP2(Sino Biological, Inc社)に挿入し、プラスミドを抽出してHEK-293細胞に7日間トランスフェクトした。培養上清をプロテインAカラムにより精製して高純度抗体を得た。
(1)組換えヒトPD-1のPD-L1への結合の遮断:組換えヒトPD-1タンパク質を1μg/mLの濃度で1ウェルあたり100μLの96ウェルプレート上に一晩4℃でコーティングした。翌日プレートを洗浄し、室温で1時間ブロッキングした。10μg/mLのビオチン化タンパク質PD-L1-Fc-ビオチン(SinoBiological, Inc.社)100μLを種々の濃度(0.4μg/mL、2μg/mL及び10μg/mL)のPD1-M944又はニボルマブ(Sino Biological, Inc社)と同時インキュベートした。プレートを洗浄して非結合抗体を除去した。プレートを抗生物質ストレプトアビジン/HRP(Beijing Zhong Shan -Golden Bridge Biological Technology Co., Ltd社)とインキュベートし、次いで、繰り返し洗浄し、基質発色溶液を加えて発色させた。終結後にOD450を検出した。結果は、組換えPD-L1タンパク質が、コーティングしたPD-1タンパク質に有効に結合可能であり、組換えPD-L1タンパク質のPD-1への結合が、種々の濃度のPD1-M944又はニボルマブの追加により有効に阻害されたことを示した(図1A)。この結果は、マウス抗体PD1-M944が、PD-1のリガンドPD-L1への結合を遮断する良好な機能を有することを示した。
マウス抗体PD1-M944の配列のヒト化してヒト化抗体PD1-H944の配列を生成する
2.1 マウス抗体PD1-M944の軽及び重鎖のそれぞれについての3つのCDRの判定
M944 scFv抗体のヌクレオチド配列を実施例1.2において判定し、これによりPD1-M944 scFv抗体の重及び軽鎖可変領域のアミノ酸配列を推定した。配列番号8/9を参照されたい。
CDR移植の古典的ヒト化方法を使用してマウス抗体のヒト化を実施した。マウス軽又は重鎖可変領域に最も近い、ヒト抗体軽又は重鎖可変領域を鋳型として選択し、マウス軽又は重鎖のそれぞれ由来の3つのCDR(Table1(表2))をヒト抗体の可変領域に挿入して、ヒト化軽鎖可変領域(VL)及び重鎖可変領域(VH)配列を得た。PD1-M944の軽鎖可変領域のヒト鋳型は、PD1-M944の軽鎖に対して73.48%相同なIGKV3-11*01であり、重鎖可変領域のヒト鋳型は、PD1-M944の重鎖に対して81.94%相同なIGHV3-21*02である。
CDR活性の支持に必須の、マウス由来フレームワーク領域内の鍵となる部位として、対応する部位をマウス抗体の配列に示す部位に復帰変異させ、この場合、軽鎖では89番目をMに、91番目をFに復帰変異させ、重鎖では44番目をRに、78番目をNに復帰変異させた。
ヒト化抗体PD1-H944の生成
PCR増幅後、逐次的に結合した重/軽鎖シグナルペプチドをコードするヌクレオチド配列(配列番号28/29)を含む、PD1-H944抗体を含むシグナルペプチドをコードする上記の各重/軽鎖ヌクレオチド配列(配列番号26/27)、ヒト化抗体の重/軽鎖可変領域(配列番号30/31)、及びヒトIgG4重鎖定常領域/ヒトカッパ軽鎖定常領域(配列番号32/33)をそれぞれ、制限エンドヌクレアーゼHind III及びXba I(Fermentas社)により二重分解し、次いで、市販のベクターpcDNA3(Invitrogen社)に挿入した。プラスミド抽出の後、pCDNA3軽鎖ベクターのNruI+NaeI+Dra I(Fermentas社)による三重分解により断片1.8kbを得、次いで、CHO/dhfr系発現ベクターpSSE(Sino Biological, Inc社)に挿入し、次いで、pCDNA3重鎖ベクターのNruI+NaeI+Dra I(Fermentas社)による三重分解により断片2.5kbを得た。次いで、これまでの工程において構築したpSSE(Sino Biological, Inc社)軽鎖ベクターにも挿入して、完全なベクターを得た。発現ベクターは、DNA増幅エレメントdhfr遺伝子、NeoR耐性遺伝子、及び抗体軽及び重鎖の発現エレメントを含む、真核細胞発現ベクターである。発現ベクターをdhfr-DG44細胞にトランスフェクトし、PD1-H944陽性細胞系をG418スクリーニングにより得、PD1-H944高発現細胞系をMTX段階圧力スクリーニングにより得、次いで、無血清培養による環境に適応させてクローンスクリーニングを行った。各工程では、抗体を高発現するクローンを、ELISAアッセイの結果と共に細胞増殖状態及び抗体医薬の鍵となる品質特性に基づいて選択した。
ヒト化抗体PD1-H944の特性決定
4.1 PD1-H944のヒト、マウス及びサルPD-1抗原への結合親和性アッセイ
(1)PD1-H944の組換えヒトPD-1タンパク質への結合能
間接ELISAを使用して、PD1-H944の組換えヒトPD-1タンパク質への特異的結合を検出した。種々の濃度(0.16ng/mL、0.49ng/mL、1.48ng/mL、4.44ng/mL、13.33ng/mL、40ng/mL、120ng/mL、360ng/mL、1080ng/mL、3240ng/mL及び9720ng/mL)の組換えヒトPD-1タンパク質を96ウェルプレート上に一晩4℃でコーティングした。翌日プレートを洗浄し、室温で1時間ブロッキングした。それぞれ2μg/mLのPD1-H944、ニボルマブ(Bristol-Myers Squibb社)及び陰性対照抗体H7N9-R1-IgG4(Sino Biological, Inc.社)100μLによるインキュベーション後、プレートを洗浄して非結合抗体を除去し、次いで、ヤギ抗ヒトIgG F(ab)2/HRP(JACKSON社)によりインキュベートして繰り返し洗浄し、基質発色溶液を加えて発色させてOD450を検出した。PD-1-H944及びニボルマブの組換えヒトPD-1タンパク質への結合のEC50は、それぞれ31.5ng/mL、R2=0.998及び179.0ng/mL、R2=0.997であった。これは、PD1-H944が組換えヒトPD-1タンパク質に、ニボルマブよりも有意に良好に結合することを示す(図2A~図2C)。
PD1-H944の組換えジャーカット/PD-1細胞への結合能を、組換えヒトPD-1安定発現細胞系ジャーカット/PD-1(SinoCellTech Ltd社)を実験材料として使用したフローサイトメトリーにより測定した。PD1-H944、ニボルマブ及び陰性対照抗体H7N9-R1-IgG4を種々の濃度(0.195μg/mL、0.391μg/mL、0.781μg/mL、1.562μg/mL、3.125μg/mL、6.25μg/mL、12.5μg/mL、25μg/mL、50μg/mL、100μg/mL及び200μg/mL)で、対数増殖期の組換えジャーカット/PD-1細胞5×105/チューブに加え、混合及び4℃でインキュベートし、次いで、洗浄及び遠心分離して非結合抗体を除去した。抗体の細胞への結合を、ヤギ抗ヒトIgG Fc-FITC(Sino Biological, Inc.社)を加えることにより検出した。結果は、PD1-H944はジャーカット/PD-1細胞にEC50of9.63μg/mL、R2=1.000で特異的に結合し、ニボルマブはジャーカット/PD-1細胞にEC50of10.18μg/mL、R2=0.994で特異的に結合し、陰性対照抗体H7N9-R1-IgG4はジャーカット/PD-1細胞に結合しなかったことを示した(図3)。これは、PD1-H944が、ジャーカット/PD-1細胞により発現したPD-1に結合する良好な能力を有し、この結合能が、ニボルマブのそれよりもわずかに良好であることを示す。
PD1-H944(0.90nM、1.79nM、3.66nM、7.24nM)、ニボルマブ(0.51nM、1.02nM、2.05nM、4.10nM)のPD-1に対する親和性をOctet Biomolecular Interaction Assay Systemを使用して陰性対照抗体としてのH7N9-R1-IgG4(13.30nM)と共に判定した。結果は、PD1-H944の組換えヒトPD-1タンパク質に対する平均KD結合親和性が64.8pM、平均結合速度定数konが3.01E+05 M-1s-1、平均解離速度定数koffが1.95E-05 s-1であり、ニボルマブのPD-1タンパク質に対する平均KD結合親和性が74.1pM、平均結合速度定数konが6.92E+05 M-1s-1、平均解離速度定数koffが5.12E-05 s-1であったことを示した(図4)。PD1-H944はニボルマブよりも強力な親和性を有し、これはニボルマブの親和性の約1.14倍であり、PD1-H944は低い解離速度を有するため、PD1-H944は、ニボルマブよりも強力なPD-1タンパク質への結合能を有する。
間接ELISAを使用して、PD1-H944の組換えサル及びマウスPD-1タンパク質への特異的結合を検出した。種々の濃度(0.16ng/mL、0.49ng/mL、1.48ng/mL、4.44ng/mL、13.33ng/mL、40ng/mL、120ng/mL、360ng/mL、1080ng/mL、3240ng/mL及び9720ng/mL)の組換えヒト、サル及びマウスPD-1タンパク質(Sino Biological, Inc.社)1ウェルあたり100μLを96ウェルプレート上に一晩4℃でコーティングし、翌日、洗浄して室温で1時間ブロッキングした。PD1-H944、ニボルマブ及び陰性対照抗体H7N9-R1-IgG4(すべて2μg/mL)100μLによるインキュベーション後、プレートを洗浄し、二次抗体のヤギ抗ヒトIgG F(ab)2/HRPを加えて発色させ、OD450を検出した。アッセイは、三つ組で行った。結果は、PD1-H944が、組換えヒトPD-1タンパク質及び組換えサルPD-1タンパク質に、それぞれ抗原EC50が25.8ng/mL(R2=0.999)及び32.7ng/mL(R2=0.997)で結合可能であり、その上、組換えマウスPD-1タンパク質に交差結合せず(図5A)、ニボルマブが、組換えヒトPD-1タンパク質及び組換えサルPD-1タンパク質に、それぞれEC50が113.2ng/mL(R2=0.997)及び80.2ng/mL(R2=0.997)で結合可能であり、その上、マウスPD-1タンパク質には結合せず(図5B)、陰性対照抗体が、組換えヒトPD-1タンパク質、組換えサルPD-1タンパク質又は組換えマウスPD-1タンパク質に結合しなかったことを示した(図5C)。PD1-H944のサルPD-1への良好な結合は、この薬物の安全性評価のためのサルの使用を支持する。
種々の濃度勾配のPD1-H944及びニボルマブのビオチン化サル及びマウスPD-1タンパク質(Sino Biological, Inc.社)に対する親和性(図6A~図6Bを参照)を、Octetを使用して測定し、解析してKD値を得た。結果は、組換えサルPD-1タンパク質によるPD1-H944の親和性KD値が108pMであり、組換えサルPD-1タンパク質によるニボルマブの親和性KD値が131pMであり、これらが同等であったことを示した(図6A)。PD1-H944及びニボルマブの両方は、組換えマウスPD-1タンパク質に結合しなかった(図6B)。
組換えヒトPD-1タンパク質1ウェルあたり100μLを96ウェルプレート上にコーティングし、一晩4℃で放置した。翌日プレートを洗浄し、室温で1時間ブロッキングした後、1μg/mLのヒトPD-L1-ビオチン(Sino Biological, Inc.社)又は2μg/mLのヒトPD-L2-ビオチン(Sino Biological, Inc.社)を加えた。次いで、種々の濃度(0.003μg/mL、0.008μg/mL、0.025μg/mL、0.074μg/mL、0.222μg/mL、0.667μg/mL、2μg/mL、6μg/mL、18μg/mL)のPD1-H944、ニボルマブ又は陰性対照抗体H7N9-R1-IgG4を加え、インキュベートし、抗生物質ストレプトアビジン/HRPを加え、インキュベートし、OD450を検出した。各郡では、二つ組で試験を実施する。結果は、ビオチン化組換えヒトPD-L1及びPD-L2タンパク質が、コーティングしたヒトPD-1タンパク質に有効に結合可能であり、種々の濃度のPD1-H944及びニボルマブの両追加により、組換えヒトPD-L1タンパク質(図7A)及び組換えヒトPD-L2タンパク質(図7B)のヒトPD-1への結合が有効に阻害されたことを示した。PD1-H944及びニボルマブは、組換えヒトPD-L1をIC50がそれぞれ0.116μg/mL(R2=0.995)及び0.129μg/mL(R2=0.997)で、組換えヒトPD-L2をIC50がそれぞれ0.446μg/mL(R2=0.994)及び0.486μg/mL(R2=0.996)で阻害した。ヒトPD-1のヒトPD-L1又はヒトPD-L2への結合を阻害するPD1-M944の能力は、ニボルマブの能力との有意差はなく、陰性対照抗体では、有意な阻害は観察されなかった。
組換えヒトPD-1を安定に発現する対数増殖期のジャーカット/PD-1細胞5×105細胞/チューブに種々の濃度(0.260μg/mL、0.390μg/mL、0.585μg/mL、0.878μg/mL、1.317μg/mL、1.975μg/mL、2.963μg/mL、4.444μg/mL、6.667μg/mL又は10.000μg/mL)のPD1-H944、ニボルマブ及び陰性対照抗体H7N9-R1-IgG4を加え、4℃でインキュベートした。0.4μg/mLのB7H1-Fc-ビオチン(Sino Biological, Inc.社)10μLを加え、PBSで洗浄し、非結合抗体を遠心分離により除去した。ストレプトアビジンAlexa Fluor(登録商標)488コンジュゲート(Life Technologies社)を加え、4℃で20分間インキュベートし、繰り返し洗浄して遠心分離し、PBSを200μL加えて細胞を再懸濁しフローサイトメーター上で検出した。結果は、組換えヒトPD-L1タンパク質が、ジャーカット/PD-1細胞に有効に結合可能であり、種々の濃度のPD1-H944及びニボルマブを加えた場合、組換えヒトPD-L1タンパク質のジャーカット/PD-1細胞への結合が、有効に阻害されたことを示した(図8)。阻害濃度IC50は、それぞれ1.78μg/mL(R2=0.994)及び2.48μg/mL(R2=0.989)であった。PD1-H944は、ヒトPD-1及びヒトPD-L1の結合をニボルマブよりもわずかに良好に阻害し、陰性対照抗体では、有意な阻害は観察されなかった。
親和性受容体CD16a(FcγRIIIa)タンパク質は、ADCCの作用を媒介する主要なFc受容体であり、V158 SNP部位を含むこのCD16aは、相対的に高い親和性を有する。PD1-H944の組換えCD16a(V158)タンパク質への結合をELISAにより測定して、抗体依存性細胞媒介細胞傷害性(ADCC)作用に関するPD1-H944の潜在力を評価した。
PD1-H944の組換えC1qタンパク質への結合をELISAにより測定し、これにより補体依存性細胞傷害性(CDC)作用に関するPD1-H944の潜在力を評価した。
PD1-H944の組換えヒトFc受容体(FcRn)タンパク質への結合をELISAにより測定し、これによりヒトにおけるPD1-H944の薬物動態を評価した。
ヒト化抗体PD1-H944の機能解析
5.1 PD1-H944により刺激された混合リンパ球応答によるCD4+T細胞の活性化
抗ヒトPD-1抗体の中和活性を、DCを用いたCD4+Tリンパ球の混合アッセイにより検出した。ヒト末梢血単核細胞(PBMC)をフィコール密度勾配遠心分離により単離し、次いで、単核細胞を接着培養方法により得、160ng/mLのrhIL-4(Sino Biological, Inc.社)及び20ng/mLのrhGM-CSF(R&D Systems社)を含む1640細胞培養培地(GIBCO社)(10%のFBS、100単位/mLのペニシリン、100μg/mLのストレプトマイシンを含む)を生じる単核細胞に加え、CO2インキュベーター内で3日目までインキュベートし、次いで、培養培地の半量を交換した。6日のインキュベーション後、懸濁する細胞を採取した。これは、DCであることがわかる。DCの密度を2×106細胞/mLに調整し、最終濃度50μg/mLのマイトマイシンを加え、37℃で20分間処理し、1640培地で3回洗浄して、さらなる工程を行った。
このアッセイでは、エフェクター細胞ジャーカット-NFAT-Luc2p-PD-1(SinoCellTech Ltd社)及び標的細胞CHO-K1-PD-L1-CD3E(SinoCellTech Ltd社)を実験材料として使用した。このような2つの細胞の同時培養により生じたPD-1/PD-L1相互作用により、TCRシグナル伝達及びNFAT-REにより媒介される生物発光が阻害される。PD-1抗体を加えると、PD-1/PD-L1相互作用が遮断され、これによりこの阻害が解除された。組換えヒトPD-1受容体細胞系におけるPD1-H944の活性化機能を、エフェクター細胞における生物発光(RLU)の強度を測定することにより判定した。
このアッセイでは、エフェクター細胞ジャーカット-NFAT-Luc2p-CD16A(SinoCellTech Ltd社)及び標的細胞CHO-PD-1(SinoCellTech Ltd社)をアッセイ材料として使用して、組換え高度活性CD16a(V158)受容体遺伝子系方法によりPD1-H944媒介ADCC作用を測定した。この機構は、2つの細胞を同時培養してPD1-H944を同時に加える場合、PD1-H944のFab断片がPD-1高発現標的細胞に結合し、これによりこのFc断片を、Fcγ受容体CD16Aを保有するエフェクター細胞に結合させ、結果的にエフェクター細胞ジャーカット-NFAT-Luc2p-CD16Aを活性化してNFAT-REにより媒介される化学発光を促進するものである。
PD-1高発現腫瘍細胞への結合の後、PD1-H944は、古典的補体経路を活性化して腫瘍細胞を殺傷し、この細胞死を生じ得る。このアッセイでは、PD1-H944のCDC作用を、WST-8方法を使用して検討した。
(1)MC38結腸がん皮下移植腫瘍を有するヒト化PD-1マウスモデルにおけるPD1-H944の薬力学試験I
対数増殖期のMC38細胞(Sun Ran Shanghai Biotechnology Co., Ltd.社)を腫瘍播種に使用した。PBS中に再懸濁したMC38細胞を5×105細胞/0.1mLでB-hPD-1ヒト化マウス(Biocytogen社)(C57BL/6マウスのPD-1遺伝子のエクソン2の部分をヒトPD-1ゲノムの対応する部分と置換することにより得た)においてマウス胸郭右側面の皮下に播種した。合計47マウスであった。腫瘍量が約100mm3に増えると、個々の腫瘍量に応じてマウスを選択し、エクセルソフトウェアを使用して5つの群にランダムに割り当てた。各郡は8匹であった。4群及び5群は、この適用には無関係の他の抗PD-1抗体を投与したため、これらのデータは本明細書には提示しない。投与は、群化した日に開始した。マウスには、腹腔内注射(I.P.)により3日毎に1回、6回連続で投与し、最終投与後10日で犠死させ、腫瘍組織を摘出して秤量した。特定の投与計画は、以下のTable3(表4)に示した。薬物の抗腫瘍作用を、腫瘍増殖阻害率TGI(%)を算出することにより評価した。TGI(%)<60%は無効であると考え、TGI(%)≧60%であり、溶媒対照群の腫瘍量よりも統計学的に有意に低かった(P<0.05)処置群の腫瘍量は、有効である、すなわち、腫瘍増殖に対する有意な阻害作用を有すると考えた。TGI(%)は、次のように算出した。

腫瘍質量の阻害率IRTW(%)=(W溶媒対照-W処置群)/W溶媒対照×100、Wは腫瘍質量である。

PD1-H944により特異的に結合したアミノ酸部位の同定
実施例4.2からのデータは、PD1-H944が、PD-1タンパク質への結合に関してPD-L1(図7A)又はPD-L2(図7B)と有効に競合し、PD1-H944により標的とされるPD-1上の結合するエピトープ及びPD-L1又はPD-L2リガンドにより標的とされるPD-1上の結合するエピトープとの間のオーバーラップを実証することを示す。
PD1-H944-PD-1タンパク質界面間の相互作用を検討するために、この実施例では、PD1-H944及びPD-1タンパク質のZDOCKドッキングを実施した。PD1-H944により標的とされるPD-1タンパク質結合エピトープが、リガンドPD-L1により標的とされるPD-1タンパク質結合エピトープとオーバーラップすることに基づいて、PD1-H944の相同性モデリングをDS4.0(Accelrys Software Inc.社)による抗体モデルプログラムを使用して実施し、最終モデル構築物の妥当性をラマチャンドランマップにより試験した。PD-1タンパク質三次元構造をPDBデータベースからダウンロードし(PDB ID:4ZQK)、Protein Preparationプログラムにより初期化した。PD1-H944モデルの結合モデル及びPD-1構造をZDOCKプログラムによりドッキングさせた。次いで、点数化したトップテンをRDOCKにより最適化した。最良のモデルをProtein Interface Analysisプログラムにより更に解析した(図22)。ドッキングモデル界面相互作用により、PD1-H944によりPD-1タンパク質に結合した主要なペプチド配列がCC'及びFGシートであったことが示された(Table13(表14))。
PD1-H944の機能性エピトープを更に確認するために、PD1-H944のPD-1タンパク質への結合の主要な部位であると予想されたTable13(表14)のペプチド配列に基づいて、一連のアラニン変異PD-1タンパク質変異体を調製してELISAアッセイにより解析した。モデルの正確性を検証するために、実施例6.1のモデル以外のいくつかの部位をまた、この試験のために選択し(Table14(表15))、このような変異部位を、5つのエピトープに大まかに空間立体構造的に群化した(図23)。
Claims (23)
- 軽鎖可変領域若しくはその部分及び/又は重鎖可変領域若しくはその部分を含む、単離PD-1抗体又はその抗原結合断片であって、
前記軽鎖可変領域又はその部分が、配列番号10のアミノ酸配列を有する軽鎖CDR1、配列番号11のアミノ酸配列を有する軽鎖CDR2、及び配列番号12のアミノ酸配列を有する軽鎖CDR3を含み、
前記重鎖可変領域又はその部分が、配列番号13のアミノ酸配列を有する重鎖CDR1、配列番号14のアミノ酸配列を有する重鎖CDR2、及び配列番号15のアミノ酸配列を有する重鎖CDR3を含む、単離PD-1抗体又はその抗原結合断片。 - 配列番号23のPD-1抗体軽鎖可変領域配列に対する少なくとも90%、92%、95%、98%若しくは100%の配列同一性を有するアミノ酸配列、及び/又は配列番号22のPD-1抗体重鎖可変領域配列に対する少なくとも90%、92%、95%、98%若しくは100%の配列同一性を有するアミノ酸配列を含むか、これらからなるか、又はこれらから本質的になる、請求項1に記載の単離PD-1抗体又はその抗原結合断片。
- 軽鎖定常領域及び重鎖定常領域を含み、好ましくは、前記軽鎖定常領域が、配列番号25のカッパ軽鎖定常領域に対する少なくとも90%、92%、95%、98%若しくは100%の配列同一性を有するアミノ酸配列を有し、及び/又は前記重鎖定常領域が、配列番号24のIgG4重鎖定常領域に対する少なくとも90%、92%、95%、98%若しくは100%の配列同一性を有するアミノ酸配列を有する、請求項1又は2に記載の単離PD-1抗体。
- IgG抗体、好ましくは、IgG4抗体である、請求項1から3のいずれか一項に記載の単離PD-1抗体。
- モノクローナル抗体である、請求項1から4のいずれか一項に記載の単離PD-1抗体。
- KD平均値で20~200pM、好ましくは60~70pM、より好ましくは64.8pMの親和性で組換えヒトPD-1タンパク質に結合する、請求項1から5のいずれか一項に記載の単離PD-1抗体又はその抗原結合断片。
- 配列番号1のアミノ酸配列を含むPD-1タンパク質分子、又は配列番号1に対する少なくとも90%、92%、95%、98%若しくは100%の配列同一性を有するアミノ酸配列を有するタンパク質分子に特異的に結合する、請求項1から6のいずれか一項に記載の単離PD-1抗体又はその抗原結合断片。
- PD-1タンパク質細胞外領域60SESFV64、78KLAAFPEDRSQP89、128LAPKAQI134の少なくとも1つから選択されるペプチド配列に特異的に結合し、
好ましくは、PD-1タンパク質細胞外領域E61、K78、D85及びP130の少なくとも1つから選択されるアミノ酸残基に特異的に結合する、請求項1から7のいずれか一項に記載の抗体又はその抗原結合断片。 - PD-1タンパク質細胞外領域60SESFV64、78KLAAFPEDRSQP89、128LAPKAQI134の少なくとも1つから選択されるペプチド配列に特異的に結合し、
好ましくは、PD-1タンパク質細胞外領域E61、K78、D85及びP130の少なくとも1つから選択されるアミノ酸残基に特異的に結合する、単離PD-1抗体又はその抗原結合断片。 - 前記抗原結合断片が、Fv、Fab、Fab'、Fab'-SH、F(ab')2、Fd断片、Fd'断片、一本鎖抗体分子、又は単一ドメイン抗体の形態であり、前記一本鎖抗体分子が、好ましくはscFv、di-scFv、tri-scFv、ダイアボディ又はscFabである、請求項1から9のいずれか一項に記載の単離PD-1抗体又はその抗原結合断片。
- 請求項1から10のいずれか一項に記載の単離PD-1抗体又はその抗原結合断片と、低分子化合物及び/又は生体高分子から選択される別の分子とにより形成される、共有結合性若しくは非共有結合性コンジュゲート又は組換え多重標的融合薬物である、改変薬物分子。
- 請求項1から10のいずれか一項に記載の単離PD-1抗体又はその抗原結合断片をコードするヌクレオチド配列を含むか、これからなるか、又はこれから本質的になり、好ましくは、配列番号26及び/又は27のヌクレオチド配列を含むか、これらからなるか、又はこれらから本質的になる、単離核酸。
- 請求項12に記載の単離核酸を含むベクター。
- 請求項12に記載の単離核酸を含み、及び/又は請求項13に記載のベクターを含む、単離細胞。
- 請求項1から10のいずれか一項に記載の単離PD-1抗体又はその抗原結合断片を生成する方法であって、請求項14に記載の単離細胞を培養する工程、及び前記単離PD-1抗体又はその抗原結合断片を精製する工程を含む、方法。
- 腫瘍又はがんの治療のための医薬の調製における、請求項1から10のいずれか一項に記載の単離PD-1抗体若しくはその抗原結合断片、又は請求項11に記載の改変薬物分子の使用。
- 前記腫瘍又はがんが、結腸がんである、請求項16に記載の使用。
- 腫瘍又はがんの治療のための、請求項1から10のいずれか一項に記載の単離PD-1抗体若しくはその抗原結合断片、又は請求項11に記載の改変薬物分子の使用。
- 前記腫瘍又はがんが、結腸がんである、請求項18に記載の使用。
- 請求項1から10のいずれか一項に記載の単離PD-1抗体若しくはその抗原結合断片、又は請求項11に記載の改変薬物分子を含む、医薬組成物。
- 請求項20に記載の医薬組成物を1つ又は複数の他の治療薬と共に含む医薬組合せ物。
- 請求項1から10のいずれか一項に記載の単離PD-1抗体若しくはその抗原結合断片、請求項11に記載の改変薬物分子、請求項20に記載の医薬組成物、又は請求項21に記載の医薬組合せ物を含み、好ましくは、投与のための装置を更に含む、キット。
- 腫瘍又はがんを治療する方法であって、それを必要とする対象に、請求項1から10のいずれか一項に記載の治療有効量の単離PD-1抗体若しくはその抗原結合断片、又は請求項11に記載の改変薬物分子、又は請求項20に記載の医薬組成物、又は請求項21に記載の医薬組合せ物、又は請求項22に記載のキットを投与し、これにより腫瘍又はがんを治療する工程を含み、好ましくは前記腫瘍又はがんが結腸がんである、方法。
