Claims (27)
1. Farmaceutska formulacija koja sadrži aktivnu tvar u krutoj disperziji s matričnim polimerom, naznačena time da aktivna tvar je 4-[3-(4-ciklopropankarbonil-piperazin-1-karbonil)-4-fluoro-benzil]-2H-ftalazin-1-on ili njegova sol ili solvat, te matrični polimer ispoljava nisku higroskopnost i visoku temperaturu omekšavanja.
2. Formulacija prema zahtjevu 1, naznačena time da je aktivna tvar u stabilnom amorfnom obliku.
3. Formulacija prema zahtjevu 2, naznačena time da je barem 90% aktivne tvari u amorfnom obliku.
4. Formulacija prema zahtjevu 2, naznačena time da je barem 95% aktivne tvari u amorfnom obliku.
5. Formulacija prema zahtjevu 2, naznačena time da je 100% aktivne tvari u amorfnom obliku.
6. Formulacija prema bilo kojem od zahtjeva 1 do 5, naznačena time da je matrični polimer odabran iz skupa koji čine: kopovidon, hidroksipropil metilceluloza ftalat (HPMCP), hidroksipropil metilceluloza acetat sukcinat (HPMCAS), 2-hidroksipropil -β-ciklodekstrin (HPBCD), hidroksipropil metilceluloza (Hipromeloza, HPMC), polimetakrilati, hidroksipropil celuloza (HPC), te celuloza acetat ftalat (CAP).
7. Formulacija prema bilo kojem od zahtjeva 1 do 5, naznačena time da matrični polimer je kopovidon.
8. Formulacija prema zahtjevu 7, naznačena time da kopovidon je ko-polimer 1-vinil-2-pirolidona i vinil acetata u masenom omjeru od 6:4.
9. Formulacija prema bilo kojem od zahtjeva 1 do 8, naznačena time da je težinski omjer aktivne tvari: matričnom polimeru od 1:0.25 do 1:10.
10. Formulacija prema zahtjevu 9, naznačena time da je težinski omjer aktivne tvari: matričnom polimeru 1: ≥2 do 1:10.
11. Formulacija prema zahtjevu 9, naznačena time da je težinski omjer aktivne tvari: matričnom polimeru 1:2 do 1:4.
12. Formulacija prema bilo kojem od prethodnih zahtjeva naznačena time da količina aktivne tvari po jedinici doziranja je barem 20% težinski.
13. Formulacija prema zahtjevu 12 naznačena time da količina aktivne tvari po jedinici doziranja je od 20% do 30% težinski.
14. Formulacija prema bilo kojem od prethodnih zahtjeva, naznačena time da kruta disperzija sadrži površinski aktivno sredstvo i/ili plastifikator.
15. Formulacija prema zahtjevu 14, naznačena time da je površinski aktivno sredstvo odabrano iz skupa koji čine: natrijev dodecil sulfat (natrijev lauril sulfat); natrijev dokuzat; cetrimid; benzetonij klorid; cetilpiridinij klorid; laurinska kiselina; polioksietilen alkil eteri; polioksietilen sorbitan esteri masne kiseline, npr. polisorbati 20, 40, 60 i 80; polioksietilen derivati ricinusovog ulja, npr. Cremophor RH40™; polioksietilen stearati i poloksameri.
16. Formulacija prema bilo kojem od prethodnih zahtjeva naznačena time da je ukupna količina aktivne tvari u formulaciji od 25 mg do 400 mg.
17. Formulacija prema zahtjevu 1, naznačena time da matrični polimer je kopovidon i formulacija dodatno sadrži sredstvo za pospješivanje klizivosti, topivo punilo, te lubrikant.
18. Formulacija prema zahtjevu 17, naznačena time da sredstvo za pospješivanje klizivosti je silicijev dioksid, topivo punilo je manitol, i lubrikant je natrijev stearil fumarat.
19. Formulacija prema zahtjevu 1, naznačena time da matrični polimer je kopovidon, težinski omjer aktivne tvari: matričnom polimeru je 1:2.3 i ukupna količina aktivne tvari je 25% težinski.
20. Formulacija prema zahtjevu 19, naznačena time da formulacija također sadrži 1.8% težinski koloidnog silicijevog dioksida, 14.7%% težinski manitola i 1% težinski natrijevog stearil fumarata.
21. Formulacija prema bilo kojem od prethodnih zahtjeva, naznačena time da se kruta disperzija je proizvodi pomoću isparavanjem otapala ili ekstruzijom taline.
22. Formulacija prema zahtjevu 21, naznačena time da se kruta disperzija proizvodi ekstruzijom taline.

Dnevna farmaceutska doza 4-[3-(4-ciklopropankarbonil-piperazin-1-karbonil)-4 fluoro-benzil]-2H-ftalazin-1-ona za liječenje raka kod pacijenta, naznačena time da doza sadrži 10 do 1000 mg 4-[3-(4-ciklopropankarbonil-piperazin-1-karbonil)-4-fluoro-benzil]-2H-ftalazin-1-ona u krutoj disperziji s matričnim polimerom koji ispoljava nisku higroskopnost i visoku temperaturu omekšavanja.
25. Dnevna farmaceutska doza prema zahtjevu 24, naznačena time da matrični polimer je kopovidon i težinski omjer 4-[3-(4-ciklopropankarbonil-piperazin-1-karbonil)-4-fluoro-benzil]-2H-ftalazin-1-ona u odnosu na matrični polimer je 1:2 do 1:4.
26. Dnevna farmaceutska doza prema zahtjevima 24 ili 25, naznačena time da se dnevna farmaceutska doza je daje u manje od 4 jedinice dnevno.
27. Postupak proizvodnje krute amorfne disperzije 4-[3-(4-ciklopropankarbonil-piperazin-1-karbonil)-4-fluoro-benzil]-2H-ftalazin-1-ona naznačen time da sadrži korake:
(i) miješanje odgovarajuće količine 4-[3-(4-ciklopropankarbonil-piperazin-1-karbonil)-4-fluoro-benzil]-2H-ftalazin-1-one ili njegove farmaceutski prihvatljive soli ili solvata sa željenom količinom barem jednog matričnog polimera, pri čemu matrični polimer ispoljava nisku higroskopnost i visoku temperaturu omekšavanja;
(ii) povećanje temperature smjese da se dobije talina; i
(iii) ekstrudiranje taline da se dobije kruti produkt.
