- Die Erfindung betrifft ein neues Verfahren zur Herstellung von 1-Cyclopropyl-7- substituierten-6-fluor-4-oxo-1,4-dihydro-chinolin-3-carbonsäure-Derivaten und deren pharmazeutisch verträglichen Salzen.

- R eine Piperazinyl-, 4-Methyl-piperazinyl- oder 4-Ethyl- piperazinyl-Gruppe bedeutet, eine starke antibakterielle Aktivität aufweisen (Eur. J. Ciin. Microbiol. 1983,2, S. 111; J. Clin. Pharmacol. 1985 25, S. 82; Drugs Exptl. Clin. Res. 1985, 5, S. 317).
- Zur Herstellung der Chinolin-carbonsäuren der allgemeinen Formel I kann 1-Cyclopropyl-6- fluor-7-chlor-4-oxo-1,4-dihydro-chinolin-3-carbonsäure mit einem cyclischen Amin in Gegenwart eines Lösungsmittels bei einer Temperatur von 135 bis 140ºC 2 Stdn. (DOS 3 033 157; DOS 3 142 854) umgesetzt werden. Die Herstellung dieser Verbindungen unter Verwendung der gleichen Reagentien ist auch in der EP-A-0078362 beschrieben worden.
- Die JP-A 60/078986 betrifft die Verwendung von Bor-Zwischenprodukten bei der Herstellung von Amino-substituierten Oxazinen, und in der JP-A 59/122470 ist die Verwendung von BF&sub2;- Komplex-Zwischenprodukten bei der Herstellung von Amino-substituierten Chinolin-carbonsäure- Derivaten beschrieben.
- Die Erfindung betrifft ein neues Verfahren zur Herstellung von 1-Cyclopropyl-7- substituierten-6-fluor-4-oxo-1,4-dihydro-chinolin-3-carbonsäure-Derivaten der weiter oben angegebenen allgemeinen Formel I. Das Verfahren ist dadurch gekennzeichnet, daß eine Verbindung der allgemeinen Formel II:
- worin
- R¹ und R², die gleich oder verschieden sein können, und eine (gewünschtenfalls durch Halogen substituierte) aliphatische Acyloxy-gruppe mit 2-6 C-Atomen, oder eine aromatische Acyloxy-gruppe mit 7-11 C-Atomen bedeuten, mit einem cyclischen Amin der allgemeinen Formel III:
- worin
- R³ Wasserstoff, Methyl oder Ethyl bedeutet, oder einem ihrer Salze umgesetzt und die so erhaltene Verbindung der allgemeinen Formel IV:
- worin
- R, R¹ und R² die oben angegebene Bedeutung haben, der Hydrolyse unterworfen wird.
- Der Vorteil des Verfahrens der vorliegenden Erfindung besteht darin, daß es ermöglicht, die Verbindung der allgemeinen Formel I in einfächer Weise in hohen Ausbeuten und innerhalb einer kurzen Reaktionszeit herzustrellen.
- Nach einer bevorzugten Durchführungsfbrm des Verfahrens der Erfindung wird das Borat- Derivat der allgemeinen Formel IV ohne Isolierung in die gewünschte Chinolin-3-carbonsäure der allgemeinen Formel I übergeführt.
- Die Borat-Derivate der allgemeinen Formel IV sind neue Verbindungen.
- Die Umsetzung der Borat-Derivate der allgemeinen Formel II mit dem cyclischen Amin der allgemeinen Formel III kann in Gegenwart eines inerten organischen Lösungsmittels und eines Säurebinders durchgeführt werden.
- Als inerte organische Lösungsmittel werden vorzugsweise Säureamide (wie Dimethylformamid, Dimethylacetamid), Ketone (wie Aceton, Methyl-ethyl-keton), Ether (wie Dioxan, Tetrahydrofuran, Diethyl-ether), Ester (wie Ethylacetat, Methylacetat, Ethylpropionat), Suffoxide (wie Dimethylsulfoxid), Alkohole (wie Methnol,Ethanol, 1-Decanol, Butanol) verwendet.
- Als Säurebinder kann eine organische oder anorganische Base verwendet werden. Einige Beispiele aus der Gruppe der organischen Basen sind Trialkyl-amine (wie Triethylamin, Tributylamin), cyclische Amine (wie Pyridin, 1,5-Diazabicyclo(5.4.0)undec-5-en, 1,5-Diazabicyclo(4.3.0)non-5-en, 1,4-Diazabicyclo(2.2.2)octan); einige Beispiele für geeignete anorganische Basen sind die Alkalimetall- oder die Erdalkalimetall-hydroxide oder -carbonate. So wird als Säurebinder dem Kaliumcarbonat, Kalium-hydrogencarbonat, Natriumhydroxid, Calciumhydroxid, etc. der Vorzug gegeben; es kann aber auch das Amin der allgemeinen Formel III im Überschuß verwendet werden.
- Die Reaktion zwischen dem Bor-derivat der allgemeinen Formel II und dem cyclischen Amin der allgemeinen Formel III kann bei einer Temperatur im Bereich von 0 bis 200ºC durchgeführt werden, was vom eingesetzten Lösungsmittel abhängt. Die Reaktionsdauer kann zwischen einer halben Std. und 10 Stdn. variieren, was von der Reaktionstemperatur abhängt. Die Durchführung der Reaktion bei erhöhter Temperatur kann die Reaktionsdauer abkürzen. Wenn auch den oben angegebenen Reaktionsbedingungen der Vorzug gegeben wird, so sind aber auch andere Reaktionsbedingungen möglich.
- Die Borate der allgemeinen Formel IV können - nach oder ohne vorherige Isolierung - unter sauren oder basischen Bedingungen zu den gewünschten Chinolin-3carbonsäuren der allgemeinen Formel I hydrolysiert werden. Die Verbindung der allgemeinen Formel IV fällt aus der Reaktionsmischung aus (z.B. beim Kühlen) und kann - falls gewünscht - isoliert werden (z.B. durch Filtration oder Zentrifugieren).
- Die basische Hydrolyse wird vorzugsweise durch Erhitzen mit einer wässrigen Alkalimetallhydroxid- oder -carbonatlösung oder einer wässrigen Erdalkalimetall-hydroxidlösung durchgeführt. Der Vorzug wird einer wässrigen Lösung von Natrium-hydroxid, Kaliumhydroxid, Natriumcarbonat, Kaliumcarbonat, Calciumhydroxid, Kalium-hydrogen-carbonat gegeben. Es konnen aber auch organische Amine (wie Triethyl-amin) in der Hydrolysestufe verwendet werden.
- Zur sauren Hydrolyse wird vorzugsweise eine wässrige Mineralsäure verwendet. So kann die Hydrolyse eines Borats der allgemeinen Formel IV durch Erhitzen mit einer wässrigen Lösung von Salzsäure, Bromwasserstoff, Schwefelsäure oder Phosphorsäure durchgeführt werden. Zur Hydrolyse können auch organische Säuren, wie Essigsäure oder Propionsäure, verwendet werden.
- Die Hydrolyse der Verbindungen der allgemeinen Formel IV kann auch in einem wässrigen Medium in Gegenwart eines mit Wasser mischbaren organischen Lösungsmittels durchgeführt werden. Einige Beispiele hierfür sind Alkohole (wie Methanol, Ethanol), Ketone (wie Aceton), Ether (wie Dioxan), Säureamide (wie Formamid, Dimethylformamid), Sulfoxide (wie Dimethylsulfoxid) oder Pyridin.
- Die so erhaltene Chinolin-3-carbonsäure der allgemeinen Formel I kann isoliert werden, zum Beispiel durch Einstellen des pH-Wertes der wässrigen Lösung auf einen geeigneten Wert, und Abtrennen der ausgefallenen Kristalle, zum Beispiel durch Filtration oder Zentrifugieren, oder durch Lyophilisation der wässrigen Reaktionsmischung.
- Die Verbindungen der allgemeinen Formel I können in an sich bekannter Weise in ihre pharmazeutisch verträglichen Salze übergeführt werden. So können Säureadditionssalze mit Halogenwasserstoffen, Sulfonsäuren, Schwefelsäure oder organischen Säuren gebildet werden. Vorzugsweise werden die Chloride, Bromide, 4-Methyl-phenyl-sulfonate, Methan-sulfonate, Maleate, Fumarate, Benzoate, etc. hergestellt. Die Verbindungen der allgemeinen Formel I bilden ebenfalls Salze mit Alkali- oder Erdalkalimetallen oder Metallionen. Infolgedessen können die Natrium-, Kalium-, Magnesium-, Silber-, Kupfer-salze, etc. hergestellt werden.
- Die Verbindungen der allgemeinen Formel I und deren pharmazeutisch verträglichen Salze können in an sich bekannnter Weise in ihre Hydrate (z.B. die Hemihydrate, Trihydrate) übergeführt werden.
- Die vorliegende Erfindung betrifft auch die neuen Verbindungen der allgemeinen Formel IV, in der R, R¹ und R² die weiter oben angegebene Bedeutung haben.
- Die Ausgangsverbindungen der allgemeinen Formel II können durch Umsetzung von 1- Cyclopropyl-6-fluor-7-chlor-4-oxo-1,4-dihydro-chinolin-3-carbonsäure (DOS 3 141 854) mit einem Bor-derivat, z.B. einer Verbindung der allgemeinen Formel V:
- worin R¹, R² die weiter oben angegebene Bedeutung haben und R&sup5; eine - gewünschtenfalls durch Halogen substituierte Acyloxygruppe mit 2-6 C-Atomen oder eine
- aromatische Acyloxy-gruppe mit 7-11 C-Atomen bedeutet, in einem wässrigen oder in einem organischen Medium hergestellt werden.
- Weitere Einzelheiten der vorliegenden Erfindung sind den nachfolgenden Beispielen zu entnehmen.
- 4,1 g (1-Cyclopropyl-6-fluor-7-chlor-1,4-dihydro-4-oxo-chinolin-3carboxylat-0³,0&sup4;)-bis- (acetato-0)-Bor und 2,8 g Piperazin-anhydrid erhitzt man in 16 ml Dimethylsuffoxid auf 110ºC unter Rühren, gibt 40 mi einer 3W/v %igen wässrigen Natriumhydroxidlösung der bräunlich-roten Lösung zu und erhitzt die Reaktionsmischung unter Rückfluß'eine Stunde zum Sieden. Die heiße, schwach gelbe Lösung filtriert man und stellt den pH-Wert durch Zusatz von 1,8 ml 96 %iger Essigsäure auf 7 ein. Die Reaktionsmischung kühlt man auf Raumtemperatur ab, filtriert die ausge&llenen weißen Kristalle ab, wäscht sie mit Wasser und Methanol und trocknet sie. Das Rohprodukt wird zur Reinigung in 10 ml Wasser zum Sieden erhitzt. So werden 2,99 g 1-Cyclopropy-6-fluor-7-(1-piperazinyl)-1,4- dihydro-4-oxo-chinolin-3-carbonsäure erhalten. Das Produkt zersetzt sich bei 255ºC.
- Analyse für die Formel C&sub1;&sub7;H&sub1;&sub8;FN&sub3;0&sub3;:
- berechnet: C=61,62% H=5,48% N=12,68%
- gefunden: C=61,58% H=5,50% N=12,61%.
- Durch Umsetzung von (1-Cyclopropyl-6-fluor-4-chlor-1,4-dihydro-4-oxo-chinolin-3- carboxylat-0³,0&sup4;)-bis-(acetat-0)-Bor mit N-Methyl-piperazin in der in Beispiel 1 angegebenen Weise wird 1-Cyclopropyl-6-fluor-7-(4-methyl-piperazin)-1,4-dihydro-4-oxo-chinolin-3-carbonsäure erhalten, die sich bei 248-250ºC zersetzt.
- 4,1 g 1-Cyclopropyl-6-fluor-7-chlor-1,4-dihydro-4-oxo-chinolin-3-carboxylat-0³,0&sup4;)-bis- (acetato-0)-Bor und 3,7 g N-Ethyl-piperazin erhitzt man in 16 ml Dimethylsuffoxid auf 90ºC unter Rühren, gibt nach 10 Minuten 40 ml einer 3W/v %igen wässrigen Natriumhydroxidlösung zu und erhitzt die Reaktionsmischung eine Stunde unter Rückfluß zum Sieden. Die heiße Lösung wird filtriert und der pH-Wert mit 96 %iger Essigsäure auf 7 eingestellt. Nach dem Kühlen der Reaktionsmischung werden die ausgefällenen Kristalle abfiltriert und mit Wasser gewaschen. Es werden so 3,3 g 1- Cyclopropyl-7-(4-ethyl-piperazinyl)-6-fluor-1,4-dihydro-4-oxo-chinolin-3-carbonsäure erhalten. Schmp: 183-185ºC.
- Analyse für die Formel C&sub1;&sub9;H&sub2;&sub2;FN&sub3;0&sub3;:
- berechnet: C=63,35% H=6,17% N=11,69%
- gefunden: C=63,31% H=6,21% N=11,70%.
Claims (10)
1. Verfahren zur Herstellung von Verbindungen der allgemeinen Formel I:
worin
R eine Piperazinyl-, 4-Methyl-piperazinyl- oder 4-Ethyl-piperazinyl-gruppe bedeutet,
sowie deren pharmazeutisch verträglichen Salzen,
dadurch gekennzeichnet, daß man eine Verbindung der allgemeinen Formel II:
worin
R¹ und R² eine (gewünschtenfalls durch Halogen substituierte) aliphatische Acyloxy-gruppe
mit 2-6 C-Atomen, oder eine aromatische Acyloxy-gruppe mit 7-11 C-Atomen
bedeuten,
mit einem Piperazin-derivat der allgemeinen Formel III:
worin
R³ Wasserstoff, Methyl oder Ethyl bedeutet,
oder einem ihrer Salze umsetzt, die erhaltene Verbindung der allgemeinen Formel IV:
worin
R, R¹ und R² die oben angegebene Bedeutung haben,
(nach oder ohne vorheriger Isolierung) der Hydrolyse unterwirft und die so erhaltene Verbindung der
allgemeinen Formel I - falls gewünscht - in eines ihrer Salze überführt oder aus einem ihrer Salze die
Säure freisetzt.
2. Verfahren nach Anspruch 1, dadurch gekennzeichnet, daß man eine Verbindung der
allgemeinen Formel II mit einem Piperazinderivat der allgemeinen Formel III in Gegenwart eines
Säureamids, Suffoxids, Ketons, Alkohols, Ethers oder Esters als Lösungsmittel umsetzt.
3. Verfahren nach Anspruch 2, dadurch gekennzeichnet, daß man Dimethylsulfoxid als
Lösungsmittel verwendet.
4. Verfahren nach Anspruch 1, dadurch gekennzeichnet, daß man die Reaktion der
Verbindungen der allgemeinen Formeln II und III in Gegenwart eines Säurebinders durchführt.
5. Verfahren nach Anspruch 4, dadurch gekennzeichnet, daß man als Säurebinder ein Amin
oder eine im Überschuß eingesetzte Verbindung der allgemeinen Formel III verwendet.
6. Verfahren nach Anspruch 1, dadurch gekennzeichnet, daß man die Hydrolyse in einem
sauren Medium durchfit.
7. Verfahren nach Anspruch 6, dadurch gekennzeichnet, daß man als Säure Salzsäure,
Schwefelsäure oder Essigsäure verwendet.
8. Verfahren nach Anspruch 1, dadurch gekennzeichnet, daß man die Hydrolyse in einem
basischen Medium durchführt.
9. Verfahren nach Anspruch 8, dadurch gekennzeichnet, daß man als Base ein
Alkalimetallhydroxid, ein Erdalkalimetallhydroxid oder Triethylamin verwendet.
10. Verbindungen der allgemeinen Formel IV:
worin
R eine Piperazinyl-, 4-Methyl-piperazinyl- oder 4-Ethyl-piperazinyl-gruppe und
R¹ und R² eine (gewünschtenfälls durch Halogen substituierte) aliphatische Acyloxy-gruppe mit 2-6
C-Atomen oder eine aromatische Acyloxy-gruppe mit 7-11 C-Atomen bedeuten.
