Description
Beispielsweise wird Natriumdodecylsulfat in einer solchen Menge verwendet, daß sich eine Konzentration von 0,5 bis 10 mM in der Reaktionsmischung ergibt. Natrium cholat wird in einer solchen Menge verwendet, daß sich eine Konzentration von 2 bis 30 mM in der Reaktions mischung ergibt. Schließlich werden Natriumfluorid, Phenanthrolin oder Hydroxylamin in einer solchen Menge verwendet, daß sich eine Konzentration von 0,5 bis 10 mM in der Reaktionsmischung ergibt. Die Einheit der Bilirubin-Oxydasen-Aktivität ist wie folgt festgelegt: Fünf mg Bilirubin-Kristalle werden in 250 ml einer 0,2 M Tris-HCL-Pufferlösung (pH 8,4) mit Äthylendiamintetraacetat-Säure (1 mM) gelöst. 2 ml dieser Lösung wird mit 0,2 ml der Enzymlösung ver mischt. Diese Mischung wird auf 37° C gehalten und ihre Absorptionsabnahme bei 440 nm wird dann gemessen. Die Enzymmenge, die 1 mM Bilirubin pro Minute oxidiert, wird als eine Einheit definiert.
Claims (3)
- a) Reagierenlassen der Reagenzien mit jeder Lösung der physiologischen Komponenten unterschiedlicher Konzentration, wobei die Reagenzien unter Anwendung des herkömmlichen Verfahrens zur Bestimmung der genannten Komponenten in Gegenwart von Bilirubin-Oxydase aus Mikroorganismen der Gattung Myrothecium oder der Gattung Coprinus und in Gegenwart von zumindest einem Additiv aus einer Verbindung aus der Gruppe Natriumcholat Taurocholsäure, Natriumdodecylsulfat, p-Toluolsulfonsäure, Salicylsäure und Sulfosalicylsäure, eine Eichkurve hervorbringen, welche die Beziehung zwischen der photometrischen Absorptionsfähigkeit jeder Reaktionsmischung und der Konzentration der genannten Komponente aufzeigt.
- b) Unterwerfen eines Blutserums mit einer unbekannten Konzentration der genannten Komponente der Reaktion in derselben Weise wie unter a), um die Absorptionsfähigkeit der sich ergebenden Reaktionsmischung zu messen, und
- c) Berechnen der Konzentration genannter Komponente in dem Blutserum durch Vergleich des in Schritt b) gemessenen Wertes mit denen der Eichkurve.
