Abstract
本发明公开了一种具有降糖功能的分子精华固体饮料及其制备方法,其特征在于,所述的分子精华固体饮料包括以下重量份数的原料制成:1~10份人参,5~20份桑椹,5~25份枸杞子,5~20份黄精,10~30份山药,5~25份茯苓,5~20份葛根,15~35份玉米须,3~15份玉竹和1~10份甘草。本发明主要功效是降血糖,预防糖尿病,提高人体免疫力。

技术领域
本发明涉及保健品领域,特别涉及一种具有降糖功能的分子精华固体饮料及其制备方法。
背景技术
糖尿病是一组以高血糖为特征的代谢性疾病。高血糖则是由于胰岛素分泌缺陷或其生物作用受损,或两者兼有引起。长期存在的高血糖,导致各种组织,特别是眼、肾、心脏、血管、神经的慢性损害、功能障碍。糖尿病已成为一个全球性的严重公共卫生问题,在中国糖尿病流行形势尤为严峻。在经济高速发展和工业化进程加速、生活方式的改变和老龄化等多种因素驱动下, 我国糖尿病患病率呈逐年递增趋势,患病人数高居世界第一。糖尿病已成为继心脑血管疾病、肿瘤之后另一个严重危害我国人民健康的重要慢性非传染性疾病,糖尿病及其并发症严重影响患者生存质量和预期寿命, 为患者及社会带来了极大的医疗和经济负担。
目前,治疗糖尿病的药物较多,临床上使用的西药虽具有一定的疗效,但多数存在较大的不良反应,如出现停药后反弹,患者肝肾损害、横纹肌融解症等副作用,治标不治本。中药不仅可以通过其药理作用,实现治疗疾病的有益效果,且因其具有毒性小、副作用少的特点,已被人们广泛应用于调养身体、延年益寿的养生药物。但目前针对糖尿病的中药多偏于单方、验方,缺乏***完善的临床药效研究。单方的降糖作用的有葛根、枸杞子、茯苓、桑葚等。
发明内容
针对现有技术存在的不足,本发明的目的在于提供一种具有降糖功能的分子精华固体饮料及其制备方法,将原料做成速溶固体饮料形式分子保健品,增强了吸收利用度,又解决了携带不便和饮用麻烦等问题,起到提高人体免疫力,有效预防糖尿病的发生。
本发明的上述技术目的是通过以下技术方案得以实现的:
一种具有降糖功能的分子精华固体饮料,所述的分子精华固体饮料包括以下重量份数的原料制成:1~10份人参,5~20份桑椹,5~25份枸杞子,5~20份黄精,10~30份山药,5~25份茯苓,5~20份葛根,15~35份玉米须,3~15份玉竹和1~10份甘草。
优选地,所述的一种具有降糖功能的分子精华固体饮料包括以下重量份数的原料制成:3份人参,10份桑椹,10份枸杞子,10份黄精,19份山药,10份茯苓,10份葛根,19份玉米须,6份玉竹和3份甘草。
优选地,一种具有降糖功能的分子精华固体饮料的制备方法,包括以下步骤:
(1)物料称取、粉碎、溶解;
(2)细胞破壁萃取;
(3)高速离心;
(4)浓缩;
(5)真空干燥。
优选地,步骤(1)的粉碎为将人参、桑椹、枸杞子、黄精、山药、茯苓、葛根、玉米须、玉竹和甘草按照重量份数进行称取,粉碎至30目。
优选地,步骤(1)的溶解为将粉碎后的物料混合在一起,用15-25倍水做为溶媒,在水温50℃-60℃下浸泡20-60分钟,然后将各种原料用乳化泵进行乳化均匀混合。
优选地,步骤(2)的细胞破壁萃取在低压3Mpa-5MPa,高压18MPa-20MPa的条件下进行细胞破壁萃取。
优选地,步骤(3)选用孔径为200nm-500nm的陶瓷膜进行分级微滤。
优选地,步骤(4)先将微滤清液进行反渗透浓缩,浓缩成一定浓度的浓液,最后把浓液在50℃-60℃条件下减压浓缩至相对密度为30-35°Bé的浸膏。
优选地,步骤(5)的真空干燥温度为40-60℃。
综上所述,本发明对比于现有技术的有益效果为:
1、本发明分子精华固体饮料的制备方法是将所有原料都先经过乳化均匀混合,再进行细胞破壁提取,采用膜分级过滤,使原料中的有效成分最大限度的提取出来;
2、本发明将组方中的各种原料做成固体速溶固体饮料的形式,增强了吸收利用度,具有冲饮携带方便,不含农药残留等优点;
3、本发明的十种原料(人参,桑椹,枸杞子,黄精,山药,茯苓,葛根,玉米须,玉竹,甘草)组合在一起,含有玉竹多糖、人参皂苷、枸杞多糖、黄精多糖、姜黄素,黄芪多糖等多种有效成分,起到相辅相成的作用。本饮品组方的主要功效是降血糖,预防糖尿病,提高人体免疫力。
具体实施方式
实施例1
一种具有降糖功能的分子精华固体饮料,所述的分子精华固体饮料包括以下重量份数的原料制成:3份人参,10份桑椹,10份枸杞子,10份黄精,19份山药,10份茯苓,10份葛根,19份玉米须,6份玉竹和3份甘草。
本实施例的预防糖尿病的分子精华固体饮料的制备方法如下:
(1)将人参,桑椹,枸杞子,黄精,山药,茯苓,葛根,玉米须,玉竹和甘草按重量份数称取,进行粉碎至30目,用15倍水做为溶媒,在水温50℃下浸泡30分钟,然后将各种原料用乳化泵使其乳化均匀混合;
(2)混匀后的物料在低压3MPa,高压18MPa的条件下进行细胞破壁萃取;
(3)高速离心,选用孔径为200nm的陶瓷膜进行分级微滤;
(4)对微滤清液进行反渗透浓缩,浓缩成一定浓度的浓液,最后把浓液在50℃条件下减压浓缩至相对密度为30°Bé的浸膏,在 40℃的温度下将浸膏用真空干燥机进行干燥,得干燥品。
实施例2
一种具有降糖功能的分子精华固体饮料,所述的分子精华固体饮料包括以下重量份数的原料制成:10份人参,6份桑椹,12份枸杞子,5份黄精,12份山药,11份茯苓,20份葛根,15份玉米须,8份玉竹和1份甘草。
本实施例的预防糖尿病的分子精华固体饮料的制备方法如下:
(1)将人参,桑椹,枸杞子,黄精,山药,茯苓,葛根,玉米须,玉竹和甘草按重量份数称取,进行粉碎至30目,用20倍水做为溶媒,在水温60℃下浸泡20分钟,然后将各种原料用乳化泵使其乳化均匀混合;
(2)混匀后的物料在低压5MPa,高压20MPa的条件下进行细胞破壁萃取;
(3)高速离心,选用孔径为500nm的陶瓷膜进行分级微滤;
(4)对微滤清液进行反渗透浓缩,浓缩成一定浓度的浓液,最后把浓液在60℃条件下减压浓缩至相对密度为35°Bé的浸膏,在 60℃的温度下将浸膏用真空干燥机进行干燥,得干燥品。
实施例3
一种具有降糖功能的分子精华固体饮料,所述的分子精华固体饮料包括以下重量份数的原料制成:2份人参,20份桑椹,5份枸杞子,20份黄精,10份山药,5份茯苓,8份葛根,20份玉米须,4份玉竹和6份甘草。
本实施例的预防糖尿病的分子精华固体饮料的制备方法如下:
(1)将人参,桑椹,枸杞子,黄精,山药,茯苓,葛根,玉米须,玉竹和甘草按重量份数称取,进行粉碎至30目,用25倍水做为溶媒,在水温55℃下浸泡60分钟,然后将各种原料用乳化泵使其乳化均匀混合;
(2)混匀后的物料在低压4MPa,高压19MPa的条件下进行细胞破壁萃取;
(3)高速离心,选用孔径为300nm的陶瓷膜进行分级微滤;
(4)对微滤清液进行反渗透浓缩,浓缩成一定浓度的浓液,最后把浓液在55℃条件下减压浓缩至相对密度为33°Bé浸膏,在 60℃的温度下将浸膏用真空干燥机进行干燥,得干燥品。
实施例4
一种具有降糖功能的分子精华固体饮料,所述的分子精华固体饮料包括以下重量份数的原料制成:5份人参,5份桑椹,25份枸杞子,5份黄精,10份山药,5份茯苓,5份葛根,35份玉米须,3份玉竹和2份甘草。
本实施例的预防糖尿病的分子精华固体饮料的制备方法如下:
(1)将人参,桑椹,枸杞子,黄精,山药,茯苓,葛根,玉米须,玉竹和甘草按重量份数称取,进行粉碎至30目,用15倍水做为溶媒,在水温50℃下浸泡40分钟,然后将各种原料用乳化泵使其乳化均匀混合;
(2)混匀后的物料在低压3MPa,高压20MPa的条件下进行细胞破壁萃取;
(3)高速离心,选用孔径为250nm的陶瓷膜进行分级微滤;
(4)对微滤清液进行反渗透浓缩,浓缩成一定浓度的浓液,最后把浓液在55℃条件下减压浓缩至相对密度为32°Bé浸膏,在 45℃的温度下将浸膏用真空干燥机进行干燥,得干燥品。
实施例5
一种具有降糖功能的分子精华固体饮料,所述的分子精华固体饮料包括以下重量份数的原料制成:6份人参,5份桑椹,5份枸杞子,5份黄精,30份山药,25份茯苓,5份葛根,15份玉米须,3份玉竹和1份甘草。
本实施例的预防糖尿病的分子精华固体饮料的制备方法如下:
(1)将人参,桑椹,枸杞子,黄精,山药,茯苓,葛根,玉米须,玉竹和甘草按重量份数称取,进行粉碎至30目,用20倍水做为溶媒,在水温55℃下浸泡60分钟,然后将各种原料用乳化泵使其乳化均匀混合;
(2)混匀后的物料在低压3MPa,高压18MPa的条件下进行细胞破壁萃取;
(3)高速离心,选用孔径为200nm-500nm的陶瓷膜进行分级微滤;
(4)对微滤清液进行反渗透浓缩,浓缩成一定浓度的浓液,最后把浓液在55℃条件下减压浓缩至相对密度为35°Bé浸膏,在 60℃的温度下将浸膏用真干燥机进行干燥,得干燥品。
对比例1 公开号为CN111789176A中国发明专利申请中的实施例1中的一种预防和延缓糖尿病的补气健脾益肾茶。
试验例1 性能测试
(1)实验动物:SPF级成年雄性SD(Sprague-Dawley)大鼠,体质量300±20 g,由广东省医学实验动物中心提供,大鼠适应性饲养7 d,自然昼夜循环,非直接光照生活,通风良好,自由饮食、饮水。
(2)试验方法:
A、造模方法:大鼠一次性腹腔注射链脲佐菌素60mg/kg,72h后尾静脉采血测血糖值,以血糖浓度≥16.7mmol/L为糖尿病造模成功的标准。
B、分组:取糖尿病造模成功的小鼠60只,随机分为6组,每组10只,分别为模型组、阳性药组(格列苯脲),实施例1(高、中、低剂量组)、对比例1组;另取正常小鼠10只,为空白组。
(3)给药
造模成功24h后,阳性药组、实施例1(高、中、低剂量组)、对比例1组分别灌胃给予相应剂量的药液;空白及模型组给予等量的生理盐水。每日给药1次,固定时间,连续给药8周。
(4)检测
在第2、4、8周给药后,观察各组动物的血糖值。在第2、4周给药后剪尾取血测血糖值,第8周给药后,眼球静脉取血,离心,取血清,测定血糖值。

试验例2 预防糖尿病的分子精华固体饮料玉竹多糖、人参皂苷、枸杞多糖、黄精多糖含量测定
a.玉竹多糖含量测定
对照品溶液的制备取无水葡萄糖对照品适量,精密称定,加水制成每1ml含无水葡萄糖0.6mg的溶液,即得。
标准曲线的制备 精密量取对照品溶液1.0ml、1.5ml、2.0ml、2.5ml、3.0ml,分别置50ml量瓶中,加水至刻度,摇匀。精密量取上述各溶液2ml,置具塞试管中,分别加4%苯酚溶液1ml,混匀,迅速加入硫酸7.0ml,摇匀,于40℃水浴中保温30分钟,取出,置冰水浴中5分钟,取出,以相应试剂为空白,照紫外-可见分光光度法(通则0401),在490nm的波长处测定吸光度,以吸光度为纵坐标,浓度为横坐标,绘制标准曲线。
取本品粗粉约1g,精密称定,置圆底烧瓶中,加水100ml,加热回流1小时,用脱脂棉滤过,如上重复提取1次,两次滤液合并,浓缩至适量,转移至100ml量瓶中,加水至刻度,摇匀,精密量取2ml,加乙醇10ml,搅拌,离心,取沉淀加水溶解,置50ml量瓶中,并稀释至刻度,摇匀,精密置取2ml,照标准曲线的制备项下的方法,自“加4%苯酚溶液1ml”起,依法测定吸光度,从标准曲线上读出供试品溶液中无水葡萄糖的重量(mg),计算,即得。
b.人参总皂苷含量测定
照高效液相色谱法(通则0512)测定:色谱条件与***适用性试验以十八烷基硅烷键合硅胶为填充剂;以乙腈为流动相A,以水为流动相B,按下表中的规定进行梯度洗脱;检测波长为203nm。理论板数按人参皂苷Rg1峰计算应不低于6000。

对照品溶液的制备精密称取人参皂苷Rg1对照品、人参皂苷Re对照品及人参皂苷Rb1对照品,加甲醇制成每1ml各含0.2mg的混合溶液,摇匀,即得。
供试品溶液的制备取本品粉末(过四号筛)约lg,精密称定,置索氏提取器中,加三氯甲烷加热回流3小时,弃去三氯甲烷液,药渣挥干溶剂,连同滤纸筒移入100ml锥形瓶中,精密加水饱和正丁醇50ml,密塞,放置过夜,超声处理(功率250W,频率50kHz)30分钟,滤过,弃去初滤液,精密量取续滤液25ml,置蒸发皿中蒸干,残渣加甲醇溶解并转移至5ml量瓶中,加甲醇稀释至刻度,摇匀,滤过,取续滤液,即得。
测定法分别精密吸取对照品溶液10μl与供试品溶液10~20μl,注入液相色谱仪,测定,即得。
c.枸杞多糖含量测定
对照品溶液的制备:取无水葡萄糖对照品25mg,精密称定,置250ml量瓶中,加水适量溶解,稀释至刻度,摇匀,即得(每1ml中含无水葡萄糖0.1mg)。
标准曲线的制备:精密量取对照品溶液0.2ml、0.4ml、0.6ml、0.8ml、1.0ml,分别置具塞试管中,分别加水补至2.0ml,各精密加入5%苯酚溶液1ml,摇匀,迅速精密加入硫酸5ml,摇匀,放置10分钟,置40℃水浴中保温15分钟,取出,迅速冷却至室温,以相应的试剂为空白,照紫外-可见分光光度法(通则0401),在490nm的波长处测定吸光度,以吸光度为纵坐标,浓度为横坐标,绘制标准曲线。
取本品粗粉约0.5g,精密称定,加***100ml,加热回流1小时,静置,放冷,小心弃去***液,残渣置水浴上挥尽***。加入80%乙醇100ml,加热回流1小时,趁热滤过,滤渣与滤器用热80%乙醇30ml分次洗涤,滤渣连同滤纸置烧瓶中,加水150ml,加热回流2小时。趁热滤过,用少量热水洗涤滤器,合并滤液与洗液,放冷,移至250ml量瓶中,用水稀释至刻度,摇匀,精密量取1ml,置具塞试管中,加水1.0ml,照标准曲线的制备项下的方法,自“各精密加入5%苯酚溶液1ml”起,依法测定吸光度,从标准曲线上读出供试品溶液中含葡萄糖的重量(mg),计算,即得。
d.黄精多糖含量测定
对照品溶液的制备:取经105℃干燥至恒重的无水葡萄糖对照品33mg,精密称定,置100ml量瓶中,加水溶解并稀释至刻度,摇匀,即得(每1ml中含无水葡萄糖0.33mg)。
标准曲线的制备:精密量取对照品溶液0.1ml、0.2ml、0.3ml、0.4ml、0.5ml、0.6ml,分别置10ml具塞刻度试管中,各加水至2.0ml,摇匀,在冰水浴中缓缓滴加0.2%蒽酮-硫酸溶液至刻度,混匀,放冷后置水浴中保温10分钟,取出,立即置冰水浴中冷却10分钟,取出,以相应试剂为空白。照紫外-可见分光光度法(通则0401),在582nm波长处测定吸光度。以吸光度为纵坐标,浓度为横坐标,绘制标准曲线。
取60℃干燥至恒重的本品细粉约0.25g,精密称定,置圆底烧瓶中,加80%乙醇150ml,置水浴中加热回流1小时,趁热滤过,残渣用80%热乙醇洗涤3次,每次10ml,将残渣及滤纸置烧瓶中,加水150ml,置沸水浴中加热回流1小时,趁热滤过,残渣及烧瓶用热水洗涤4次,每次10ml,合并滤液与洗液,放冷,转移至250ml量瓶中,加水至刻度,摇匀,精密量取1ml,置10ml具塞干燥试管中,照标准曲线的制备项下的方法,自“加水至2.0ml”起,依法测定吸光度,从标准曲线上读出供试品溶液中含无水葡萄糖的重量(mg),计算,即得。
采用上述测定方法,对实施例1-3制备的预防糖尿病的分子精华固体饮料的玉竹多糖、人参总皂苷、枸杞多糖、黄精多糖测定结果如下表1所示。

由上表可知,通过本发明的制备方法提取制备的有效成分得到富集,有效成分含量高,提取率高,功效性能好。

Claims (9)
1.一种具有降糖功能的分子精华固体饮料,其特征在于,所述的分子精华固体饮料包括以下重量份数的原料制成:1~10份人参,5~20份桑椹,5~25份枸杞子,5~20份黄精,10~30份山药,5~25份茯苓,5~20份葛根,15~35份玉米须,3~15份玉竹和1~10份甘草。
2.根据权利要求1所述的一种具有降糖功能的分子精华固体饮料,其特征在于,所述的分子精华固体饮料包括以下重量份数的原料制成:3份人参,10份桑椹,10份枸杞子,10份黄精,19份山药,10份茯苓,10份葛根,19份玉米须,6份玉竹和3份甘草。
3.根据权利要求1所述的一种具有降糖功能的分子精华固体饮料的制备方法,其特征在于,包括以下步骤:
(1)物料称取、粉碎、溶解;
(2)细胞破壁萃取;
(3)高速离心;
(4)浓缩;
(5)真空干燥。
4.根据权利要求3所述的一种具有降糖功能的分子精华固体饮料的制备方法,其特征在于,步骤(1)的粉碎为将人参、桑椹、枸杞子、黄精、山药、茯苓、葛根、玉米须、玉竹和甘草按照重量份数进行称取,粉碎至30目。
5.根据权利要求3所述的一种具有降糖功能的分子精华固体饮料的制备方法,其特征在于,步骤(1)的溶解是将粉碎后的物料混合在一起,用15-25倍水做为溶媒,在水温50℃-60℃下浸泡20-60分钟,然后将各种原料用乳化泵进行乳化均匀混合。
6.根据权利要求3所述的一种具有降糖功能的分子精华固体饮料的制备方法,其特征在于,步骤(2)的细胞破壁萃取在低压3Mpa-5MPa,高压18MPa-20MPa的条件下进行细胞破壁萃取。
7.根据权利要求3所述的一种具有降糖功能的分子精华固体饮料的制备方法,其特征在于,步骤(3)选用孔径为200nm-500nm的陶瓷膜进行分级微滤。
8.根据权利要求3所述的一种具有降糖功能的分子精华固体饮料的制备方法,其特征在于,步骤(4)先将微滤清液进行反渗透浓缩,浓缩成一定浓度的浓液,最后把浓液在50℃-60℃条件下减压浓缩至相对密度为30-35°Bé的浸膏。
9.根据权利要求3所述的一种具有降糖功能的分子精华固体饮料的制备方法,其特征在于,步骤(5)的真空干燥温度为40-60℃。
