Abstract
본 발명은 황화수소 검출용 형광 프로브 및 이의 제조방법에 관한 것이다. 본 발명에 의한 황화수소를 검출할 수 있는 형광 센서는 신속하고 정확하게 황화수소만을 선택적으로 검출이 가능한 효과를 가진다. 또한, 생체 내 비침습적인 영상을 이용하여 세포 및 조직의 깊은 곳에서 진행되는 생물학적인 현상을 실시간으로 확인이 가능하다.
Description
본 발명은 하기 화학식 1로 표시되는 황화수소 검출용 형광 프로브 및 이의 제조방법에 관한 것이다.
황화수소(H2S)는 새롭게 인식된 전달자로서, 시스타티오닌-γ-리아제(Cystathinone-γ-lyase, CSE) 및 시스타티오닌-β-신티아제(Cystathionin-β-synthase, CBS)와 같은 효소의 활동을 통해 시스테인(Cysteine) 및 호모시스테인(Homocysteine)으로부터 포유류의 조직에서 합성된다.
이러한 황화수소(H2S)는 산화질소(Nitric oxide)와 일산화탄소(Carbon monoxide)와 더불어 3차 기체신호전달물질(3th gasotransmitter) 또는 기체(gasomediator)로서 다양한 생리현상에 참여하고, 세포 면역 시스템에 중요한 역할을 하는 것으로 알려져 있다. 특히, K-ATP의 개시자(opener) 역할을 하여 심혈관 시스템의 항상성에 기여하며 손상된 심혈관 근육의 치유 및 신경계의 산화 스트레스로부터 보호 역할 등에 직접 관여하는 것으로 알려져 있어, 여러 연구 분야에서 각광을 받고 있다. 이와 관련하여 종래 한국등록특허 제1,404,369호에서는 황화수소 농도를 실시간으로 검출하는 황화수소 센서를 구비하는 심장 허혈 및 재관류에 대한 실시간 생체신호 측정 장치가 개시된 바 있다.
또한, 인간의 혈액 내 혈장(plasma)의 황화수소 농도는 50 ~ 100㎛ 수준으로 알려져 있으므로, 황화수소의 농도 수준을 정량적으로 검출할 수 있다면 다운 증후군(Down syndrome), 알츠하이머 병(Alzheimer's disease), 당뇨병 및 간경변증 등의 질병 진단이 가능하다.
최근에, 살아있는 세포 또는 조직에서 황화수소(H2S)를 검출하는 방법으로는 가스 크로마토그래피, 전기화학 및 비색법 등이 있으나, 이러한 방법들은 조직 및 세포 용해물의 처리 및/또는 파괴를 필요로 한다.
또한, 미토콘드리아 내부에는 전체 황화물의 30%가 저장되어 있고 황화수소 역량의 황화물이 존재하고 있는데, 이러한 황화수소(H2S)를 선택적이고 정량적으로 검출하는 방법이 요구되는 실정이다.
이에, 최근 들어 다양한 형광탐침법(Fluorescent probe)들이 개발되고 있다. 예를 들면, 종래 황화수소(H2S) 검출을 위한 형광 프로브로서 2,4-디니트로벤젠 설포닐 플루오로세인 화합물이 개시된 바 있으나, 상기 화합물의 경우 시간이 지남에 따라 술폰산 에스테르가 가수 분해하여 형광 강도가 변화한다는 문제점이 발생한다(Analytica Chimica Acta, 631, 91, 2009).
본 발명은 상기와 같은 문제점을 해결하기 위하여 안출된 것으로, 생체 내에서 비침습적인 영상을 이용하여 황화수소(H2S)를 검출할 수 있는 형광 프로브와 이의 제조방법을 제공하는데 그 목적이 있다.
또한, 본 발명은 세포 및 생체 내에 존재하는 황화수소(H2S)를 검출하는 방법을 제공하는데 그 목적이 있다.
본 발명의 적절한 실시 형태에 따르면, 황화수소 검출용 형광 프로브는 하기 화학식 1로 표시되는 것을 특징으로 한다.

또한, 상기 황화수소 검출용 형광 프로브는, 생체 내의 황화수소와 반응하여 550 내지 680nm 및 695 내지 770nm에서 형광으로 가변하는 것을 특징으로 한다.
본 발명의 다른 적절한 실시 형태에 따르면, 황화수소 검출용 형광 프로브의 제조방법은, 화학식 2로 표시되는 화합물을 4-클로로레시놀과 반응시켜 화학식 3으로 표시되는 화합물로 제조하는 단계; 및 상기에서 제조된 화학식 3으로 표시되는 화합물을 화학식 4로 표시되는 화합물과 반응시켜 화학식 1로 표시되는 화합물을 제조하는 단계를 포함하는 것을 특징으로 한다.

또한, 상기 화학식 4로 표시되는 화합물은 하기 반응식 2에 의하여 합성되는 것을 특징으로 한다.

여기서, 상기 반응식 2에서 (a)는 4-아미노벤질 알코올, 아질산나트륨(NaNO2), 아지드화나트륨(NaN3), HCl, 물이고, (b)는 염화술푸릴(SO2Cl2), 디클로로메탄(DCM)이다.
본 발명의 또 다른 적절한 실시 형태에 따르면, 황화수소의 검출 방법은, 황화수소 검출용 형광 프로브를 생체 내에 주입하는 단계; 상기 황화수소 검출용 형광 프로브가 생체 내 황화수소와 반응하여 형광을 나타내는 단계; 및 상기 형광을 확인하는 단계를 포함하는 것을 특징으로 한다.
본 발명에 의한 황화수소를 검출할 수 있는 형광 프로브는 신속하고 정확하게 황화수소만을 선택적으로 검출이 가능한 효과를 가진다.
또한, 생체 내 비침습적인 영상을 이용하여 세포 및 조직의 깊은 곳에서 진행되는 생물학적인 현상을 실시간으로 관찰이 가능하다.

이하, 본 발명에 대하여 상세히 설명하기로 한다. 이에 앞서, 본 명세서 및 청구범위에 사용된 용어나 단어는 통상적이거나 사전적인 의미로 한정해서 해석되어서는 아니되며, 발명자는 그 자신의 발명을 가장 최선의 방법으로 설명하기 위해 용어의 개념을 적절하게 정의할 수 있다는 원칙에 입각하여 본 발명의 기술적 사상에 부합하는 의미와 개념으로 해석되어야만 한다. 따라서, 본 명세서에 기재된 실시예에 기재된 구성은 본 발명의 가장 바람직한 일 실시예에 불과할 뿐이고 본 발명의 기술적 사상을 모두 대변하는 것은 아니므로, 본 출원시점에 있어서 이들을 대체할 수 있는 다양한 균등물과 변형예들이 있을 수 있음을 이해하여야 한다.
본 발명에서, 용어 '프로브'는 '센서'라고도 하며 생체 내/외 표적을 검출하거나, 영상화할 수 있는 것으로 정의된다. 일반적으로 영상화제, 조영제, 방사성 의약품 등의 용어로 대체하여 사용되고 있다.
본 발명의 황화수소 검출용 형광 프로브는 하기 화학식 1로 표시되는 화합물이다.

여기서, 상기 화학식 1로 표시되는 화합물은 아자이드기를 포함하여 황화수소를 향한 접근성을 향상시킬 수 있다.
상기 황화수소 검출용 형광 프로브는 근적외선 형광물질(Near-Infrared Fluorescence, NIRF)로, 생체 내에 주입하게 되면 미토콘드리아 또는 세포질에 선택적으로 위치될 수 있다. 상기 황화수소 검출용 형광 프로브가 생체 내에 주입하게 되면, 550 내지 680nm 및 695 내지 770nm에서 형광의 세기가 변하게 되어 이를 측정함으로써 황화수소를 검출할 수 있다.(도 1 참조)
상기 황화수소 검출용 형광 프로브를 적용하면 바람직하게는 세포 또는 조직과 같은 생체 내 100 내지 200㎛의 투과 깊이에서 황화수소의 탐지가 가능하고, 또한 실시간으로 영상화할 수 있다. 따라서, 황화수소와 관련된 생명과학 연구와 질병의 조기 진단, 진단 시약 및 치료제의 개발에 크게 기여할 수 있다.
또한, 본 발명의 황화수소 검출용 형광 프로브의 제조방법은, 하기 반응식 1을 따르는 것을 특징으로 한다.

상기 반응식 1은 화학식 2로 표시되는 화합물을 4-클로로레시놀과 반응시켜 화학식 3로 표시되는 화합물로 제조하는 단계; 및 상기에서 제조된 화학식 3로 표시되는 화합물을 화학식 4로 표시되는 화합물과 반응시켜 화학식 1로 표시되는 화합물로 제조하는 단계를 포함하는 것을 특징으로 한다.
하기에, 본 발명의 화학식 1로 표시되는 화합물의 제조방법을 단계별로 더욱 상세히 설명하기로 한다.
우선, 하기 화학식 2로 표시되는 화합물을 준비한다.

여기서, 상기 화학식 2로 표시되는 화합물은 임상 응용 염료 인도시아닌 그린(Indocyanine green, ICG)보다 높고 더 안정된 형광 강도를 가지는 근적외선(NIR) 형광 염료로써, 클로로-치환된 시아닌 염료이다.
상기 화학식 2의 화합물은 2-[2-[2-클로로-3-[1,3-디하이드로-3,3-디메틸-1-프로필-2H-인돌-2-일리덴)에틸리덴]-1-시클로헥산-1-yl]에틸]3,3-디메틸-1-프로필인돌륨 아이오드(2-[2-[2-Chloro-3-[(1,3-dihydro-3,3-dimethyl-1-propyl-2H-indol-2-ylidene)ethylidene]-1-cyclohexen-1-yl]ethenyl]-3,3-dimethyl-1-propylindolium iodide)이며, 근적외선(NIR) IR-780 아이오딘화물(Aldrich® Chemistry로부터 상업적으로 입수 가능)을 사용할 수 있다.
상기 화학식 2로 표시되는 화합물이 준비되면, 상기 화합물과 4-클로로레시놀을 혼합하여, 하기 화학식 3으로 표시되는 화합물로 제조한다.

여기서, 상기 화학식 3으로 표시되는 화합물을 제조할 경우, 바람직하게는 30 내지 70℃에서 반응시킬 수 있다.
또한, 상기 4-클로로레시놀 투입시 유기용매를 함께 투입할 수 있다. 상기 유기용매는 디메틸포름아미드(DMF), 트라이에틸아민(TEA), 에탄올, 디이소프로필에테르, 디에틸에테르, 디옥산, 테트라히드로퓨란(THF), 디메틸아세트아미드(DMA), 디메일설폭사이드(DMSO), 메틸렌클로라이드(MC), 아세토나이트릴(ACN), 클로로벤젠, 톨루엔 및 벤젠 중 적어도 하나 이상을 포함할 수 있으며, 바람직하게는 디메틸포름아미드(DMF) 및 트라이에틸아민(TEA) 중 적어도 하나 이상을 사용할 수 있다.
상기 화학식 3으로 표시되는 화합물을 제조하고 난 후, 상기 화합물과 하기 화학식 4로 표시되는 화합물을 반응시켜 화학식 1로 표시되는 화합물을 제조한다.

여기서, 상기 화학식 1의 화합물을 제조할 경우, 바람직하게는 30 내지 70℃ 반응시킬 수 있다.
또한, 상기 화학식 4의 화합물의 투입시 유기용매를 포함할 수 있다. 여기서, 상기 유기용매는 디메틸포름아미드(DMF), 트라이에틸아민(TEA), 에탄올, 디이소프로필에테르, 디에틸에테르, 디옥산, 테트라히드로퓨란(THF), 디메틸아세트아미드(DMA), 디메일설폭사이드(DMSO), 메틸렌클로라이드(MC), 아세토나이트릴(ACN), 클로로벤젠, 톨루엔 및 벤젠 중 적어도 하나 이상을 포함할 수 있으며, 바람직하게는 디메틸포름아미드(DMF) 및 트라이에틸아민(TEA) 중 적어도 하나 이상을 사용할 수 있으며, 바람직하게는 메틸렌클로라이드(MC) 및 아세토나이트릴(ACN) 중 적어도 하나 이상을 포함할 수 있다.
또한, 상기 유기용매는 탄산칼륨(K2CO3)을 더 포함할 수 있다.
또한, 상기 유기용매는 완충제를 더 포함할 수 있다. 상기 완충제는 바람직하게는 포스페이트-완충 염수("PBS"), 아세트산나트륨, 아세트 산암모늄, 아세트산, 시트르산, 시트르산나트륨, 트리스(히드록시메틸)아미노에탄("트리스"), N-2-히드록시에틸피페라진-N'-2-에탄술폰산("HEPES"), 3-(N-모르폴리노) 프로판술폰산 ("MOPS"), 2-(N-모르폴리노)에탄술폰산("MES"), N-(2-아세트아미도)이미노디아세트산 ("ADA"), 피페라진-N,N'-비스(2-에탄술폰산) ("PIPES"), 및 N-(2-아세트아미도)-2-아미노에탄술폰산 ("ACES") 중 적어도 하나를 포함할 수 있으며, 가장 바람직하게는 PBS를 포함할 수 있다. 또한, 상기 PBS는 pH가 7.4인 것이 바람직하다.
여기서, 상기 화학식 4로 표시되는 화합물은 하기 반응식 2에 의하여 합성된다.

상기 반응식 2에서 (a)는 4-아미노벤질 알코올, 아질산나트륨(NaNO2), 아지드화나트륨(NaN3), HCl, 물, (b)는 염화술푸릴(SO2Cl2), 디클로로메탄(DCM)이다.
본 발명의 황화수소 검출 방법으로는, 상기에서 제조된 황화수소 검출용 형광 프로브를 생체 내에 주입하는 단계; 상기 황화수소 검출용 형광 프로브가 생체 내 황화수소와 반응하여 형광을 나타내는 단계; 및 형광을 확인하는 단계를 포함하는 것을 특징으로 한다.
상기에서 제조된 황화수소 검출용 형광 프로브를 황화수소로 처리할 경우, 바람직하게는 pH가 5 내지 12에서 형광체를 확인할 수 있다.(도 2 참조)
여기서, 하기 화학식 1로 표시되는 황화수소 검출용 형광 프로브는 황화수소와 반응하면 하기 화학식 3으로 표시되는 화합물로 바뀌면서 형광을 나타내는 것을 특징으로 한다. 상기 형광 세기는 550 내지 680nm 및 695 내지 770nm에서 동시에 가변하는 것을 특징으로 한다.

합성예
1. 화학식 2로 표시되는 화합물의 합성
30mL의 물에 NaNO2(1.0g, 15.0mmol)를 용해시키기 위하여, 0℃에서 30분 동안 6N HCl(10mL) 하에서 4-아미노벤질알코올(1.23g, 10.0mmol)을 첨가하고 교반하여 적하시켰다.
박층크로마토그래피(TLC)에 의하여 반응을 살펴보았고, 두 시간 후에 반응을 정지시켰다. 에테르로 3회 추출한 후, 유기상은 포화 NaHCO3 수용액, 물 및 염수로 순차적으로 세척하였다.
유기상은 MgSO4에서 건조시키고, 진공에서 증발시켰다.
생성물은 황색 오일(1.33g, 수율 89%)인 화학식 2로 표시되는 화합물을 수득하기 위하여, 용출액으로써 에틸 아세테이트/n-헥산을 사용하여 실리카 컬럼 크로마토그래피로 정제하였다.
최종적으로 수득된 하기 화학식 2로 표시되는 화합물의 1H 및 13C NMR 스펙트럼 결과를 하기에 나타내었다.

합성예
2. 화학식 3으로 표시되는 화합물의 합성
4-클로로레조시놀(270mg, 1.87mmol)과 트리에틸아민(0.65mL, 4.67mmol), 건조 DMF(2.7mL)를 플라스크에 넣어서 혼합물을 제조하고, 이 혼합물을 30분 동안 아르곤 분위기 하에 실온에서 교반하였다. 그리고, R-780 아이오드 화합물(500mg, 0.75mmol)에 상기 혼합물에 첨가하고, 4시간 동안 50℃에서 교반하였다.
블루-그린 용액(283mg, 수율 66%)인 화학식 3으로 표시되는 화합물을 수득하기 위하여, 용출액 CH2Cl2/EtOH(30:1)을 사용하여 실리카 컬럼 크로마토그래피로 정제하였다.
최종적으로 수득된 하기 화학식 3으로 표시되는 화합물의 1H NMR 스펙트럼과 질량분석 결과를 하기에 나타내었다.

표시되는 화합물의 합성
17mL의 DCM 하에서 화학식 2로 표시되는 화합물(500mg, 3.35 mmol)을 용해시키기 위하여, 0℃에서 SO2Cl2(0.54mL, 6.7mmol)을 첨가하고 적하시켰다. 그리고, 반응을 위해 상온에서 밤새 교반하였다.
이후 TLC에 의하여 반응을 살펴보았고, 반응 혼합물을 EtOAc에서 희석하고, 포화된 NaHCO3 수용액, 물 및 염수로 순차적으로 세척하였다.
유기상은 MgSO4에서 건조시켰다.
황색 오일(440mg, 수율 78%)인 화학식 4로 표시되는 화합물을 수득하기 위하여, 용출액으로써 EtOAc/n-헥산(1:20)을 사용하여 실리카 컬럼 크로마토그래피로 정제하였다.
최종적으로 수득된 하기 화학식 4로 표시되는 화합물의 1H NMR 스펙트럼 결과를 하기에 나타내었다.

합성예
4. 화학식 1로 표시되는 화합물의 합성
화학식 3으로 표시되는 화합물(75mg, 0.13mmol)과 K2CO3(55mg, 0.39mmol)을 건조 CH2Cl2/CH3CN(1:1, 1.0mL)을 포함하는 플라스크에 넣어서 혼합물을 제조하고, 이 혼합물을 30 분동안 아르곤 분위기 하에서 실온에서 교반시켰다. 그리고, 화학식 4로 표시되는 화합물(89mg, 0.39mmol)을 첨가하여 밤새 반응시켰다.
블루 용액(47mg, 수율 51%)인 화학식 1로 표시되는 화합물을 수득하기 위하여, 용출액 CH2Cl2/EtOH(60:1)을 사용하여 실리카 컬럼 크로마토그래피로 정제하였다.
최종적으로 수득된 하기 화학식 1로 표시되는 화합물의 1H 및 13C NMR 스펙트럼과 질량분석 결과를 하기에 나타내었다.

1. 화학식 1로 표시되는 화합물의 황화수소 검출
실시예
1-1. 파장에 따른 형광 반응 평가
PBS 완충액(10 μM, pH 7.4)을 사용하여 합성예 4에서 제조된 화학식 1로 표시된 화합물을 제조하였다. 그리고, 37℃에서 NaHS(황화수소 소스)를 30분 동안 반응시켜 형광 스펙트럼을 조사하였다. 그 결과를 도 3의 a)에 나타내었다.
반응이 종료되고, 황화수소를 처리하였을 경우 화학식 3으로 표시되는 화합물의 수율은 0.72로 나타났다.
도 3의 a)를 살펴보면, 720nm에서 약 200배가 향상되었음을 알 수 있었다. 따라서, 화학식 1로 표시되는 화합물의 형광이 켜지는 것은 황화수소에 의해 유발된다는 것이 증명되었다.
실시예
1-2. 농도에 따른 형광 반응 평가
PBS 완충액(10 μM, pH 7.4)을 사용하여 합성예 4에서 제조된 화학식 1로 표시된 화합물을 제조하였다. 그리고, 37℃에서 이 화합물과 NaHS(황화수소 소스)를 반응시켰다. 여기서, 상기 화학식 1로 표시된 화합물은 720nm에서 형광을 측정한 것으로, NaHS(황화수소 소스)의 농도를 0 에서 50㎛로 변화시켜 농도에 따른 형광 반응을 조사하였으며, 그 결과를 도 3의 b)에 나타내었다.
도 3의 b)를 살펴보면, 0 에서 50㎛ 까지 황화수소를 투입함에 따라 형광강도가 선형적으로 증가하는 것으로 보아, 형광강도의 측정만으로 황화수소 농도의 정량적인 측정이 가능하다는 것을 알 수 있었다.
실시예
2. 화학식 1로 표시되는 화합물의 선택적인 황화수소 검출
분석물질은 표 1에 나타내었다. 여기서, 분석물질 1은 화학식 1로 표시되는 화합물만을 나타낸 것이고, 분석물질 2 내지 17은 화학식 1로 표시되는 화합물과의 혼합물을 나타낸 것이다.
분석물질의 제조는 37℃에서 PBS 완충액(10㎛, pH 7.4, 30% acetonitrile, v/v) 하에서 수행되었다.

17개의 분석물질을 스크리닝한 결과를 도 4에 나타내었다.
도 4를 살펴보면, 황화수소 소스를 투입한 화학식 1로 표시되는 화합물인 분석물질 2에서만 형광강도가 높게 나타났음을 알 수 있었으며, 그 결과 화학식 1로 표시되는 화합물은 황화수소만을 선택적으로 탐지하는 것을 발견하였다.
실시예
3. 쥐의
대식세포에 대한 형광반응 평가
37℃에서 쥐의 대식세포를 Dulbecco’s modified Eagle’s 배지(DMEM)에 넣고, 5%의 CO2와 95% 공기의 분위기 하에서 FBS(fetal bovine serum) 10%를 보충하여 배양하였다.
24시간 후에, 커버슬립은 DPBS에서 배지를 제거하기 위해 3회 세척하고, 나중에 사용하기 위하여 DPBS에 배양시켰다.
실험 결과를 도출하기 위하여 5㎛의 화학식 1로 표시되는 화합물을 세포 시료에 첨가하여 30분 동안 배양시켰다. 그리고, NaHS를 0, 20, 40 및 80㎛로 투입하여 형광 강도를 알아보았다.
이미지 전에 세포를 DPBS에서 3회 세척 하였다. 형광 이미징은 공초점 현미경(Olympus Fluoview 1000)의 오일-이멀젼(oil-immersion) 40배 대물렌즈를 사용하여 얻었다.
실험 결과를 도 5에 나타내었다.
도 5를 살펴보면, 황화수소 소스인 NaHS의 농도를 증가시킬수록 형광강도가 강해지는 것을 알 수 있었다.
실시예
4.
헬라세포(HeLa cells)에
대한 세포독성검사(
CCK
-8 assays)
37℃, 5%의 CO2 하에서 헬라세포(HeLa cells)와 10%(v/v) FBS 및 1%의 페니실린/스트렙토마이신을 보충하여, 200μL의 DMEM(GIBCO, 11885) 내에 5,000 cell/well의 밀도의 96-well plates에 플레이팅하였다.
세포를 24시간 배양한 후, 세포 시료 플레이트에 첨가하였다. 그리고, 1시간 동안 배양한 후, 10μL의 CCK-8 용액(Dojindo, Japan)을 각각의 플레이트에 첨가하고, 세포를 추가로 30분 동안 배양하였다.
450nm에서 흡광도를 마이크로플레이트 리더(SpectraMax M2 / Molecular Devices)에서 측정하였다.
측정 결과를 도 6에 나타내었다.
도 6을 살펴보면, 화학식 1로 표시되는 화합물은 헬라세포(HeLa cells) 내에서 95% 이상 생존하고 있는 것으로 나타나 매우 낮은 독성을 나타내고 있음을 알 수 있었다.
실시예
5. 쥐를 대상으로 한 농도에 따른 형광반응 평가
Balb C 누드 쥐(Balb/c nude mice, 15-22g)에게 키시라진(xylazine, 10mg/kg)과 케타민(ketamine, 80mg/kg)를 복강에 주사함으로써 마취시켰다.
그리고, 쥐에게 화학식 1로 표시되는 화합물(50㎛, 20μL의 DMSO)을 복강에 주사시키고, NaHS(0, 50, 100, 200㎛, 100μL의 PBS)를 복강에 주사시켰다.
쥐는 675nm의 여기 필터와 695 - 770 nm의 발광 필터와 함께 생체 내 이미징 시스템에 IVIS 루미나 II을 사용하여 형광을 탐지하였다. 그 결과를 도면 7에 나타내었다.
도면 7을 살펴보면, 황화수소 소스인 NaHS의 주입량을 증가시킬수록 형광 강도가 강해지는 것을 알 수 있었다.
실시예
6. 쥐를 대상으로 한 시간에 따른 형광반응 평가
Balb C 누드 쥐(Balb/c nude mice, 15-22g)에게 키시라진(xylazine, 10mg/kg)과 케타민(ketamine, 80mg/kg)를 복강에 주사함으로써 마취시켰다.
그리고, 쥐에게 화학식 1로 표시되는 화합물(50㎛, 20μL DMSO)을 복강에 주사시키고, NaHS(100㎛, 100μL의 PBS)를 복강에 주사시켰다.
쥐를 675nm 여기 필터와 695 내지 770nm의 발광 필터와 695 내지 770nm의 발광 필터를 사용하여 다른 시간(0, 0.5, 1, 2, 3시간)에 생체 내 이미징 시스템에 IVIS 루미나 II을 사용하여 형광을 탐지하였다. 그 결과를 도면 8에 나타내었다.
도면 8을 살펴보면, 황화수소 소스인 NaHS를 주입하자마자 형광이 나타났으며 약 1시간 내지 2시간 이내에 형광이 최대로 발현하는 것을 알 수 있었다.
Claims (5)
- 제 1항에 있어서,상기 황화수소 검출용 형광 프로브는, 생체 내의 황화수소와 반응하여 550 내지 680nm 및 695 내지 770nm에서 형광으로 가변하는 것을 특징으로 하는 황화수소 검출용 형광 프로브.
- 제 1항에 기재된 황화수소 검출용 형광 프로브를 생체 내에 주입하는 단계;상기 황화수소 검출용 형광 프로브가 생체 내 황화수소와 반응하여 형광을 나타내는 단계; 및상기 형광을 확인하는 단계를 포함하는 것을 특징으로 하는 황화수소의 검출 방법.
