Abstract
본 발명은 실리콘 치환 아미노-파이로닌 화합물의 유도체, 이를 이용한 세포 또는 조직의 영상화 방법, 이를 이용한 암진단 방법 및 이를 제조하는 방법 등에 관한 것으로서, 더욱 구체적으로는 상기 실리콘 치환 아미노-파이로닌 화합물의 유도체는 이광자 흡수 형광체로, 종래 일광자 흡수 형광체에 비해 625 nm 이상의 적색 또는 근적외선 영역에서의 더 긴 흡수 및 방출 파장을 가지고 있어, 생체 영상화 연구에 있어서 자가 형광에 의한 영향을 최소화할 수 있는 바 깊은 조직에서의 고해상도 영상화에 적합할 것으로 기대된다. 상기 형광체 플랫폼을 기반으로 개발된 형광 프로브는 생체 내 특정 물질 분석 및 영상화에 있어 그 활용범위를 확장시킬 수 있을 것으로 기대된다.
Description
본 발명은 아미노-실라-파이로닌 화합물 기반 일광자 또는 이광자 흡수 형광체 및 이의 용도에 관한 것으로, 보다 자세하게는 상기 형광체의 세포 영상화에의 응용에 관한 것이다.
형광(fluorescence) 신호를 바탕으로 한 생체 영상화(bioimaging) 기술은 세포 소기관(organelle), 조직(tissue) 등을 시각화할 수 있는 방법으로 널리 활용되고 있으며, 이 중 형광 신호를 발하는 형광 프로브(fluorescent probe)의 개발은 생체 내 특정 물질 분석 및 영상화에 있어 그 활용범위를 확장시키게 된다. 현재 생체 영상화에 활용되고 있는 대부분의 형광 물질 및 프로브는 일광자 흡수(one-photon absorption) 형광체이며, 일광자 광학현미경(one-photon microscopy, OPM)에 의해 생체 영상화가 구현된다.
그러나 최근에는 빛 산란의 영향에 민감하지 않으면서, 보다 깊은 조직에 대해서도 해상도 높은 영상화를 가능하게 하는 이광자 광학현미경(two-photon microscopy, TPM)을 이용한 생체 영상화 연구에 대한 관심이 증가되고 있는 추세이다. 이광자 광학현미경 영상화법은 900 nm 파장대에 가까운 근적외선(near infrared, NIR) 영역에서 여기(excitation)가 가능하며, 동시에 높은 조직 투과성과 함께 초점(focal point) 부분만 여기 시킬 수 있기 때문에 생체 조직에 대한 낮은 광-손상(photo-damage)과 형광체에 대한 낮은 광-퇴색(photo-bleaching) 등의 생체 영상화 분야에 있어서 다양한 장점을 가지고 있다.
이광자 현미경을 이용한 생체 영상화를 위해서 현재 이용되고 있는 이광자 흡수 형광체로는 대표적으로 아세단(acedan), 나프탈이미드(napthalimide) 유도체 등이 있다. 이들 형광체들은 주로 녹색 영역에서 형광을 발하는데, 이 영역은 생체 내 물질(innate biomolecules)로부터 방출되는 자가 형광(autofluorescence) 영역과 겹치기 때문에 조직 영상화 결과의 신뢰도를 저하시키는 단점을 가지고 있다. 따라서 생체 내 물질에 의한 자가 형광 영역을 벗어난 625 nm 이상의 장파장 영역(적색 또는 근적외선)에서도 강한 형광을 방출하는 새로운 형광체를 개발한다면, 기존의 이광자 흡수 물질이 지니는 한계점을 극복할 수 있을 것으로 기대된다.
한편, 형광 프로브는 여기 파장과 방출 파장의 차이(Stokes shift)가 크면 클수록 형광 물질 또는 프로브가 자체적으로 광을 흡수(self-absorption)하여 소광(quenching)시키는 것을 감소시킬 수 있다. 또한, 서로 다른 파장 영역에서 여기시킬 수 있으며 동시에 서로 다른 파장 영역에서 방출을 하는 형광 프로브의 경우에는 비율 기반 형광 신호(ratiometric fluorescence signal)를 얻을 수 있으며, 외부 환경에 따라 형광 세기가 매우 민감하게 감응하는 특성을 극복할 수 있어서 신뢰성 있는 정량적 분석이 가능하다. 따라서, 이와 같은 특성을 가지는 형광 프로브는 생체 내 특정 기질 감지 및 생체 영상화 연구에 매우 유용하다(국내등록특허 10-1662427).
따라서, 장파장 영역에서도 강한 형광을 방출하며, 서로 다른 파장 영역에서 흡수 및 방출을 하는 광물리적 특성을 나타내는 형광 프로브의 개발은 기존 한계점을 극복하여 다양한 생체 영상화 분야에 폭넓게 적용할 수 있으며, 신뢰성 높은 생체 영상화 분석 기법을 제공할 수 있을 것으로 기대된다.
본 발명은 상기와 같은 종래 기술상의 문제점을 해결하기 위해 안출된 것으로, 자가 형광의 간섭을 최소화하여 높은 해상도로 조직 영상화를 구현할 수 있는 새로운 형광체 화합물인, 아미노-실라-파이로닌(Amino Si-pyronin) 화합물 기반 일광자 또는 이광자 흡수 형광체 및 이의 제조 방법, 이의 용도 등을 제공하는 것을 그 목적으로 한다.
그러나 본 발명이 이루고자 하는 기술적 과제는 이상에서 언급한 과제에 제한되지 않으며, 언급되지 않은 또 다른 과제들은 아래의 기재로부터 본 발명이 속하는 기술 분야의 통상의 지식을 가진 자에게 명확하게 이해될 수 있을 것이다.
본 발명은 하기 화학식 1로 표시되는 아미노-실라-파이로닌(Amino Si-pyronin) 화합물 또는 이들의 약학적으로 허용가능한 염을 제공한다.

상기 화학식 1에서, R1은 치환 또는 비치환된 탄소수 1 내지 18의 알킬기이며, 상기 비치환은 수소 원자가 작용기로 치환된 것을 의미하며, 상기 치환은 수소 원자 대신에 일반적인 작용기가 치환된 것을 의미한다. 이 때 작용기는 메틸기, 에틸기, 페닐기, 알릴기, 프로파질기, 아지도에틸기, 아지도프로필기, 벤질기 터트-부틸부티레이트기, 아미노부틸기, 히드록시에틸기, 알데하이드기, 카복실기, 카복실릭 에스터기, 하이드록시기, 머캅토기, 아지도기, 아민기, 비닐기, 할로겐기, 시아노기, 아릴기, 니트릴기, 카보네이트기 등이나, 이에 제한되지 않는다.

본 발명의 일 구체예에 있어서, 상기 화합물 또는 이들의 약학적으로 허용가능한 염은 일광자 흡수 형광체 또는 이광자 흡수 형광체일 수 있으나, 형광을 방출할 수 있는 형태라면 이에 제한되지 않는다.
본 발명의 다른 구체예에 있어서, 상기 화합물 또는 이들의 약학적으로 허용가능한 염은 바람직하게는 625 nm 이상의 장파장 영역 또는 근적외선 영역에서 형광을 방출하는 형광체이나, 세포 또는 조직을 영상화할 수 있는 영역이라면 이에 제한되지 않는다.
또한, 본 발명은 상기 화합물 또는 이들의 약학적으로 허용가능한 염을 세포 또는 조직에 처리하고 관측하는 단계를 포함하는 세포 또는 조직의 영상화 방법을 제공한다.
본 발명의 일 구체예에 있어서, 상기 관측하는 단계는 일광자 형광 현미경 또는 이광자 형광 현미경을 이용하는 것을 특징으로 한다.
또한, 본 발명은 상기 화합물 또는 이들의 약학적으로 허용가능한 염을 포함하는 진단용 조성물을 제공한다.
또한, 본 발명은 상기 화합물 또는 이들의 약학적으로 허용가능한 염을 체내 또는 조직에 투여하여, 진단용 형광 영상을 획득하는 방법을 제공한다.
또한, 본 발명은 상기 화합물 또는 이들의 약학적으로 허용가능한 염의 진단 용도를 제공한다.
본 발명의 일 구체예에 있어서, 상기 진단은 바람직하게는 암, 결핵 등의 질병을 진단하는 것으로서, 본 발명의 화합물 또는 이들의 약학적으로 허용가능한 염을 단독으로, 또는 특정 질병의 마커를 결합시킨 본 발명의 화합물 또는 이들의 약학적으로 허용가능한 염을 이용하여 진단하는 것으로서, 상기 질병에는 제한이 없다.
또한, 본 발명은 (a) 3,7-비스(디메틸아미노)-5,5-디메틸디벤조[b,e]실린-10(5H)-온에 트리플루오로메탄설폰산 무수물을 첨가하여 혼합물을 제조하는 단계; 및 (b) 상기 혼합물에 아민을 첨가하는 단계를 포함하는, 하기 화학식 1로 표시되는 아미노-실라-파이로닌(Amino Si-pyronin) 화합물의 제조 방법을 제공한다.

본 발명의 아미노-실라-파이로닌 화합물 기반 일광자 또는 이광자 흡수 형광체는 구조적으로는 단순하나 적색 또는 근적외선 영역에서의 흡수 및 방출 파장을 가지고 있어 현재 널리 사용되고 있는 시아닌(cyanine) 계 형광체에 비해서 화학적 및 광 안정성이 뛰어나며, 생체 영상화 연구에 있어서 자가 형광에 의한 영향을 최소화할 수 있어 검사에 대한 신뢰성을 현저히 높일 수 있다. 또한, 본 발명의 형광체가 가진 이광자 흡수 특성을 이용하면 깊은 조직에서의 고해상도 영상화에도 사용가능하다. 따라서, 본 발명의 형광체 플랫폼은 이를 기반으로 하여 다양한 특성을 가지는 형광 프로브의 개발이 가능하며, 이를 이용하여 단독으로 또는 특정 질환의 마커에 대한 표적을 결합시켜, 암 세포뿐만 아니라 다양한 질환을 영상화하여 진단하는데 사용 가능할 뿐만 아니라, 이를 통하여 검사 정확성도 현저히 증가시킬 수 있을 것으로 기대된다. 또한, 이에 한정되지 않고 생체 내 특정 물질의 분석 및 영상화에 있어서 그 활용 범위를 다양하게 적용가능할 것으로 기대된다.

본 발명자들은 세포 또는 조직의 자가 형광의 간섭을 최소화하여 높은 신뢰성 및 높은 해상도를 가지는 세포 및/또는 조직의 영상화를 구현할 수 있는 새로운 형광체를 개발하고자 예의 연구한 결과, 본 발명을 완성하였다. 상기 형광체는 625 nm 이상의 장파장 영역 및 근적외선 영역에서 형광을 방출하는 형광체이며 아민기에 서로 다른 치환기를 도입함에 따라서 서로 다른 영역에서 흡수 및 방출을 하는 다양한 유도체로 변환시킬 수 있는 가능성을 가지고 있기 때문에 비율 기반 생체 영상화에 적합한 장점을 가진다.
본 발명의 일 실시예에서는 세포 또는 동물 모델의 조직에 본 발명의 화합물을 처리한 결과 종래의 이광자 흡수 형광체와 비교하여 큰 흡수 단면 값을 나타내며, 세포에 대하여 우수한 형광 영상을 제공할 수 있음을 확인하였다. 따라서, 본 발명의 화합물 또는 이의 화학적으로 허용가능한 염을 이용하여 세포 또는 조직을 영상화하는 방법을 제공할 수 있다. 또한, 본 발명의 화합물 또는 이의 약학적으로 허용가능한 염을 포함하는 진단용 조성물을 제공할 수 있다.
본 명세서에 있어서, “세포(cell) 또는 조직(tissue)”이란 인간 또는 비-인간을 포함하는 포유류의 세포 또는 조직일 수 있고, 상기 비-인간 포유류는 바람직하게는 마우스, 랫트, 개, 고양이, 말, 소, 양, 염소, 돼지, 토끼 등을 포함할 수 있으나, 이에 한정되지 않는다.
본 명세서에 있어서, “유기용매(organic solvent)”란 유기물질로 이루어진 다른 물질을 용해시키는 물질을 총칭하며, 바람직하게는 에탄올, 메탄올, 톨루엔, 에틸렌, 페톨, 벤젠, 크실렌, 디클로로메탄, 헥산, 메틸 아세테이트, 에틸 아세테이트, 테트라클로로에틸렌 등이나, 본 명세서의 화합물, 디이소프로필에틸아민 및 피리딘, 2,6-루티딘, 디이소프로필에틸아민 등을 용해시킬 수 있는 용매라면 이에 제한되지 않는다.
본 명세서에 있어서, “화합물(compound)”이란 2개 이상의 다른 원소들이 일정 비율로 구성된 순물질을 의미하는데, 탄소와 수소의 포함여부에 따라 유기화합물과 무기화합물, 원소의 결합방식에 따라 이온화합물과 분자화합물로 분류될 수 있으며, 상기 화합물은 바람직하게는 화학식 1 내지 26으로 표시되는 화합물들 중의 하나이다.

상기 화합물 2c를 제조하기 위하여, 실시예 2.2와 동일한 방법으로 획득된 최종 생성물(200 mg, 0.39 mmol)을 무수 테트라히드로푸란(tetra hydro furan, THF) 15 mL을 이용하여 용해시킨 후에, -78 ℃에서 0.84 mL의 2차-부틸리튬(1.4 M)을 첨가하였다. 그리고 2차-부틸리튬이 첨가된 혼합물을 2 시간 동안 교반시킨 후에 디메틸디클로로실레인(0.7 mmol) 85 μL를 첨가하였다. 그리고 다시 상온에서 3 시간 동안 교반시킨 후, 1 N의 염화수소를 이용하여 반응을 중지시켰다. 이 후, 포화 탄산수소나트륨을 이용하여 중화시킨 후 디클로로메탄(2 × 10 mL)과 증류수를 이용하여 추출하고, 유기층을 무수황산나트륨으로 건조시켰다. 그리고 40 mbar의 감압조건에서 용매를 완전히 제거한 후, 별도의 정제과정 없이 다음 단계에서 사용하였다.
2.4. 화합물 22의 제조
상기 화합물 22를 제조하기 위하여, 실시예 2.3과 동일한 방법으로 획득된 최종 생성물(160 mg, 0.39 mmol)을 아세톤 15 mL을 이용하여 용해시킨 후에, 0 ℃에서 185 mg의 과망간산칼륨(1.17 mmol)을 30분 동안 조금씩 첨가하였다. 그리고 3 시간 동안 교반시킨 후에 디클로로메탄을 이용하여 희석시켜 준 후에 디클로로메탄과 셀라이트를 혼합하여 첨가시킨 후에 40 mbar의 감압조건에서 용매를 제거하였다. 그리고 실리카겔(Merck-silicagel 60, 230-400 mesh)을 통과하는 관 크로마토그래피를 이용하여 정제하여 최종적으로 노란색 고체 화합물 22(33 mg, 20 %)를 획득하였다. 정제 시에는 전개액으로 3 % EtOAc/CH2Cl2를 사용하였다. 그리고 제조된 화합물 22를 NMR로 관찰한 결과 “1H NMR (CDCl3, 300MHz, 298K, δ: 8.07 (m, 2H), 3.29 (d, 8H), 2.9 (m 8H), 2.03 (m, 8H), 0.59 (m, 6H). 13C NMR (CDCl3, 500MHz, 298K, δ: 143.3, 135.9, 129.6, 128.4, 123.9, 122.6, 50.4, 49.9, 28.7, 28.1, 21.8, 21.5, 0.1.”로 확인되었다.

상기 화합물 14를 제조하기 위하여, 실시예 2와 동일한 방법으로 획득된 최종 생성물(35 mg, 0.082 mmol)을 무수 디클로로메탄 2 mL을 이용하여 용해시킨 후에 상온에서 트리플루오로메탄설폰산 무수물(0.16 mmol) 27 μL를 첨가하고 20 분 동안 교반시킨 후에 0.82 mmol의 프로파질 아민(propargyl amine) 52.5 μL를 첨가하였다. 그리고 다시 상온에서 4 시간 동안 교반시킨 후에 디클로로메탄(2 × 10 mL)과 증류수를 이용하여 추출하고, 유기층을 무수황산나트륨으로 건조시켰다. 그리고 40 mbar의 감압조건에서 용매를 제거한 후, 실리카겔(Merck-silicagel 60, 230-400 mesh)을 통과하는 관 크로마토그래피를 이용하여 정제하여 최종적으로 적색 고체 화합물 14(35.3 mg, 70 %)를 획득하였다. 정제 시에는 전개액으로 3 % MeOH/CH2Cl2를 사용하였다. 그리고 제조된 화합물 14를 NMR로 관찰한 결과 “1H NMR (CDCl3, 300MHz, 298K, δ: 7.58 (s, 2H), 4.37 (d, 2H), 3.36 (d, 8H), 2.84 (t, 8H), 2.53 (s, 1H), 2.02 (q, 10H), 0.56 (s, 6H). 13C NMR (CDCl3, 125 MHz, 298 K): d 175.4, 147.0, 128.6, 126.5, 123.0, 122.1, 119.6, 78.4, 74.5, 50.8, 50.2, 39.9, 29.9, 29.4, 27.9, 21.5, 21.1, 0.2, -0.1.”로 확인되었다.

상기 화합물 15를 제조하기 위하여, 실시예 2와 동일한 방법으로 획득된 최종 생성물(20 mg, 0.047 mmol)을 무수 디클로로메탄 2 mL을 이용하여 용해시킨 후에 상온에서 트리플루오로메탄설폰산 무수물(0.093 mmol) 16 μL를 첨가하고 20 분 동안 교반시킨 후에 0.47 mmol의 벤질 아민(benzyl amine) 51 μL를 첨가하였다. 그리고 다시 상온에서 4 시간 동안 교반시킨 후에 디클로로메탄(2 × 10 mL)과 증류수를 이용하여 추출하고, 유기층을 무수황산나트륨으로 건조시켰다. 그리고 40 mbar의 감압조건에서 용매를 제거한 후, 실리카겔(Merck-silicagel 60, 230-400 mesh)을 통과하는 관 크로마토그래피를 이용하여 정제하여 최종적으로 적색 고체 화합물 15(22 mg, 70 %)를 획득하였다. 정제 시에는 전개액으로 3 % MeOH/CH2Cl2를 사용하였다. 그리고 제조된 화합물 15를 NMR로 관찰한 결과 “1H NMR (CDCl3, 300MHz, 298K, δ: 7.39 (m, 7H), 4.91 (s, 2H), 3.35 (q, 8H), 2.85 (t, 8H), 2.00 (m, 8H), 0.56 (s, 6H). 13C NMR (CDCl3, 75 MHz, 298 K): d 176.1, 146.8, 137.3, 129.2, 128.1, 127.7, 126.3, 122.9, 118.7, 53.7, 50.7, 50.2, 29.9, 29.4, 27.9, 21.5, 21.1, 0.2, -0.1.”로 확인되었다.

상기 화합물 17을 제조하기 위하여, 실시예 12와 동일한 방법으로 획득된 최종 생성물 화합물 13(30 mg, 0.05 mmol)을 무수 디클로로메탄 5 mL을 이용하여 용해시킨 후에 0 ℃에서 0.75 mmol의 2,6-루티딘(2,6-lutidine) 87 μL를 첨가하고 10 분 동안 교반시킨 후에 0.5 mmol의 염화아세틸(acetyl chloride) 36 μL를 첨가하고 상온에서 12 시간 동안 교반시킨 후에 디클로로메탄(2 × 10 mL)과 증류수를 이용하여 추출하고, 유기층을 무수황산나트륨으로 건조시켰다. 그리고 40 mbar의 감압조건에서 용매를 제거한 후, 실리카겔(Merck-silicagel 60, 230-400 mesh)을 통과하는 관 크로마토그래피를 이용하여 정제하여 녹색 고체 화합물을 획득하였다. 정제 시에는 전개액으로 5 % MeOH/CH2Cl2를 사용하였다. 그리고 녹색 고체 화합물은 다시 HPLC(high performance liquid chromatography)로 정제하여 최종적으로 화합물 17(5 mg, 15 %)을 획득하였다. 제조된 화합물 17을 NMR로 관찰한 결과 “1H NMR (CD3OD, 500 MHz, 298 K): d 7.10 (s, 2H), 3.63-3.60 (m, 8H), 3.22 (s, 3H), 2.98-2.95 (m, 4H), 2.79-2.77 (m, 4H), 2.09-2.06 (m, 4H), 2.03-2.00 (m, 4H), 1.83 (s, 3H), 0.68 (s, 6H); 13C NMR (CD3OD, 125 MHz, 298 K): d 173.0, 160.2, 152.2, 142.6, 134.7, 134.0, 127.2, 125.4, 53.2, 53.1, 52.6, 39.1, 30.1, 28.9, 22.0, 21.9, -0.8.”로 확인되었다.

상기 화합물 20을 제조하기 위하여, 실시예 12와 동일한 방법으로 획득된 최종 생성물 화합물 13(30 mg, 0.05 mmol)을 무수 디클로로메탄 5 mL을 이용하여 용해시킨 후에 0.25 mmol의 디이소프로필에틸아민(Diisopropylethylamine, DIPEA) 44 μL와 0.5 mmol의 피리딘 40 μL를 첨가하고 상온에서 10 분 동안 교반시킨 후에 0.5 mmol의 4-니트로벤질클로로포르메이트 108 mg을 첨가하고 45 ℃에서 24 시간 동안 교반시켜 혼합하였다. 그리고 혼합된 용액을 디클로로메탄(2 × 10 mL)과 1 N의 염화수소를 이용하여 추출하고, 유기층을 무수황산나트륨으로 건조시켰다. 그리고 40 mbar의 감압조건에서 용매를 제거한 후, 실리카겔(Merck-silicagel 60, 230-400 mesh)을 통과하는 관 크로마토그래피를 이용하여 정제하여 녹색 고체 화합물을 획득하였다. 정제 시에는 전개액으로 5 % MeOH/CH2Cl2를 사용하였다. 그리고 녹색 고체 화합물은 다시 HPLC(high performance liquid chromatography)로 정제하여 최종적으로 화합물 20(5 mg, 12 %)을 획득하였다. 제조된 화합물 20을 NMR로 관찰한 결과 “1H NMR (CD3OD, 500 MHz, 298 K): d 8.32 (d, J = 14.5 Hz, 2H), 7.30 (d, J = 15 Hz, 2H), 7.09 (s, 2H), 5.18 (s, 2H), 3.62-3.55 (m, 8H), 3.24 (s, 3H), 2.98-2.94 (m, 4H), 2.77-2.66 (m, 2H), 2.63-2.53 (m, 2H), 2.09-1.93 (br, m, 8H), 0.67 (d, J = 14.0 Hz, 6H).”로 확인되었다.
실시예 20: 아미노-실라-파이로닌 화합물의 광물리적 특성 확인
상기 실시예 3 내지 19와 동일한 방법으로 제조된 화합물 4 내지 20, 즉, 아미노-실라-파이로닌 화합물의 흡수 특성 및 형광 방출 특성을 확인하기 위하여, 5 μM 농도의 화합물 4 내지 화합물 12 중 어느 하나를 포함하는 에탄올, 디옥산, 디클로로메탄, 아세토니트릴, 및 1 % DMSO를 포함하는 인산염완충용액을 통과 길이 1 cm의 석영 셀에 채워 HP8453 자외선/가시광선 분광광도계를 이용하여 자외선/가시광선(UV/Vis) 흡수 스펙트럼을 측정하고, 최대 흡수파장 값을 도 1a에 나타내었다. 또한, 10 μM 농도의 화합물 13 내지 화합물 21 중 어느 하나를 포함하는 에탄올, 아세토니트릴, 및 1 % DMSO를 포함하는 인산염완충용액을 1 cm의 석영 셀에 채워 동일한 방법으로 자외선/가시광선(UV/Vis) 흡수 스펙트럼을 측정하고, 최대 흡수파장 값을 도 1b에 나타내었다.
실시예
21: 세포를 이용한 현미경 영상화 확인
상기 실시예 3 내지 11과 동일한 방법으로 제조된 화합물 4 내지 12를 세포의 영상화에 사용가능한지 확인하기 위하여, 화합물들을 HeLa 세포에 처리한 후에 일광자 또는 이광자 현미경을 이용하여 영상을 획득하였다. 보다 자세하게는, HeLa 세포를 10 % 우태아 혈청(fetal bovine serum, FBS) 및 1 %의 항생제가 첨가된 Dulbecco's Modified Eagles Medium(DMEM) 배지에 접종하고, 5 % CO2-95 % air 및 37 ℃의 조건으로 60 mm 배양 접시에 2 × 106 세포의 밀도(density)로 배양하였다. 그리고 배양된 세포에 10 μM의 화합물 4 내지 화합물 12를 각각 처리하고 30분간 반응시킨 후에 배양액을 제거하고, 인산염완충용액(phosphate buffered saline, PBS)을 이용하여 3회 세척하여 배양액을 깨끗이 제거하였다. 그리고 4 % 포름알데히드(formaldehyde) 용액을 넣어주어 세포를 고정시키고 일광자 현미경 또는 이광자 현미경(TCS SP5 II, Leica, Germany)을 이용하여 관찰하였다. 그 결과는 도 2a에 나타내었다.
도 2a에 나타난 바와 같이, 화합물 4 내지 화합물 12 중 어느 하나의 화합물을 처리한 세포를 일광자 형광 현미경으로 관찰한 결과 488 nm 여기 파장으로 498-800 nm의 채널에서 형광이 관찰되었으며, 이광자 형광 현미경으로 관찰한 결과 900 nm의 여기 파장으로 청색 채널(430-480 nm), 녹색 채널(500-550 nm), 황색 채널(565-605 nm), 및 적색 채널(625-675 nm)에서 형광이 관찰되는 것을 확인하였다.
실시예
22: 세포를 이용한 현미경 영상화 확인
상기 실시예 12 내지 18과 동일한 방법으로 제조된 화합물 13 내지 19를 세포의 영상화에 사용가능한지 확인하기 위하여, 화합물들을 A549 세포에 처리한 후에 일광자 또는 이광자 현미경을 이용하여 영상을 획득하였다. 보다 자세하게는, A549 세포를 10 % 우태아 혈청(fetal bovine serum, FBS) 및 1 %의 항생제가 첨가된 Dulbecco's Modified Eagles Medium(DMEM) 배지에 접종하고, 5 % CO2-95 % air 및 37 ℃의 조건으로 60 mm 배양 접시에 2 × 106 세포의 밀도(density)로 배양하였다. 그리고 배양된 세포에 1 μM의 화합물 13 내지 화합물 15, 또는 10 μM의 화합물 16 내지 19를 각각 처리하고 30 분간 반응시킨 후에 배양액을 제거하고, 인산염완충용액(phosphate buffered saline, PBS)을 이용하여 3회 세척하여 배양액을 깨끗이 제거하였다. 그리고 4 % 포름알데히드(formaldehyde) 용액을 넣어주어 세포를 고정시키고 일광자 현미경 또는 이광자 현미경(TCS SP5 II, Leica, Germany)을 이용하여 관찰하였다. 그 결과는 도 2b 및 도 2c에 나타내었다.
도 2b에 나타난 바와 같이, 화합물 13 내지 화합물 15 중 어느 하나의 화합물을 처리한 세포를 일광자 형광 현미경으로 관찰한 결과 488 nm 여기 파장으로 500-800 nm의 채널에서 형광이 관찰되었으며, 이광자 형광 현미경으로 관찰한 결과 900 nm의 여기 파장으로 500-800 nm 채널에서 형광이 관찰되는 것을 확인하였다.
또한, 도 2c에 나타난 바와 같이, 화합물 16 내지 화합물 19 중 어느 하나의 화합물을 처리한 세포를 일광자 형광 현미경으로 관찰한 결과 488 nm 및 633 nm의 여기 파장으로 각각 적색 채널(650-800 nm)과 녹색 채널(500-630 nm)에서 형광이 관찰되는 것을 확인하였고, 이광자 형광 현미경으로 관찰한 결과 900 nm의 여기 파장으로 황색 채널(565-605 nm) 및 적색 채널(625-675 nm)에서 형광이 관찰되는 것을 확인하였다.
상기 결과들을 통하여, 세포를 이용한 실험에서도 하기 화학식 1 또는 화학식 3을 기반으로 하는 다양한 화합물 4 내지 화합물 20, 모든 화합물이 세포 내에서 안정적으로 유사한 형광 방출 특성을 나타나는 것을 확인하였으며, 이를 통하여, 본 발명의 화합물 4 내지 화합물 20을 생체 영상화에 적용 가능하다는 것을 확인할 수 있었을 뿐만 아니라, 본 발명의 화합물들은 모두 일광자 및/또는 이광자 흡수 형광체로 사용 가능하다는 것을 확인할 수 있었다. 따라서, 본 발명은 하기 화학식 1로 표시되는 아미노-실라-파이로닌(Amino Si-pyronin) 화합물 또는 하기 화학식 3으로 표시되는 줄로리딘 기반 아미노-실라 파이로닌(julolidine-based amino-Si-pyronin) 화합물 구조를 기반으로 하여 R기를 변화시키는 것만으로도 다양한 형광 방출 화합물을 제조할 수 있다는 것을 확인하였으며, 이를 통하여 하기 화학식 1 또는 3의 구조를 기반으로 하여 제한 없이 다양한 형질을 가지는 형광 프로브를 제조할 수 있다는 것을 확인할 수 있었다.

또한, 화합물 16의 경우에는 적색 채널에서 형광 신호가 관찰되고, 화합물 13의 경우에는 황색 채널에서 형광 신호가 관찰되는 것을 확인하였다. 이를 통하여, 화합물 16은 화합물 13에서 파생된 화합물이기 때문에, 분자내 상호교환 반응으로 2차 아민 형태의 화합물 13의 형태가 전환된다면, 스토크스 이동(stokes shift)을 통하여 두 개의 형광이 동시에 방출될 수 있다는 것을 확인할 수 있었다. 따라서, 이를 이용하여 하나의 감지계가 기질을 검출할 때, 기질의 농도에 따라 그 비율이 점차 변해가는 비율 기준(ratiometric) 형광 프로브로 응용이 가능하며, 이를 통하여 본 발명의 화합물들을 비율 기반 생체 영상화에 사용 가능하다는 것을 확인할 수 있었다. 또한, 각각의 구조의 변화에 따라 서로 다른 형광이 관측되므로, 동시에 다양한 기질을 검출하는데 사용가능할 뿐만 아니라, 화학식 1 또는 3으로 표시되는 화합물의 일부 구조들을 변화시킴으로써 더욱 다양한 형질을 가지는 형광 프로브를 제조할 수 있다는 것을 확인할 수 있었다.
전술한 본 발명의 설명은 예시를 위한 것이며, 본 발명이 속하는 기술분야의 통상의 지식을 가진 자는 본 발명의 기술적 사상이나 필수적인 특징을 변경하지 않고서 다른 구체적인 형태로 쉽게 변형이 가능하다는 것을 이해할 수 있을 것이다. 그러므로 이상에서 기술한 실시예들은 모든 면에서 예시적인 것이며 한정적이 아닌 것으로 이해해야만 한다.
본 발명의 아미노-실라-파이로닌 화합물 기반 일광자 또는 이광자 흡수 형광체는 이를 기반으로 하여 다양한 특성을 가지는 형광 프로브의 개발이 가능하며, 단독으로 또는 특정 질환의 마커에 대한 표적을 결합시켜 다양한 질환을 영상화하여 진단하는데 사용 가능할 뿐만 아니라, 생체 내 특정 물질의 분석 및 영상화에 있어서 다양하게 적용가능하다.

- 제 4 항에 있어서,상기 화합물 또는 이들의 약학적으로 허용가능한 염은 일광자 흡수 형광체 또는 이광자 흡수 형광체인 것을 특징으로 하는, 화합물 또는 이들의 약학적으로 허용가능한 염.
- 제 4 항에 있어서,상기 화합물 또는 이들의 약학적으로 허용가능한 염은 625 nm 이상의 장파장 영역 또는 근적외선 영역에서 형광을 방출하는 형광체인 것을 특징으로 하는, 화합물 또는 이들의 약학적으로 허용가능한 염.
- 제 1 항 또는 제 4 항의 화합물 또는 이들의 약학적으로 허용가능한 염을 세포 또는 조직에 처리하고 관측하는 단계를 포함하는, 세포 또는 조직의 영상화(imaging) 방법.
- 제 7 항에 있어서,상기 관측하는 단계는 일광자 형광 현미경 또는 이광자 형광 현미경을 이용하는 것을 특징으로 하는, 방법.
- 제 1 항 또는 제 4 항의 화합물 또는 이들의 약학적으로 허용가능한 염을 유효성분으로 포함하는, 진단용 조성물.
- 제 1 항 또는 제 4 항의 화합물 또는 이들의 약학적으로 허용가능한 염을 체내 또는 조직에 투여하여, 진단용 형광 영상을 획득하는 방법.
- 제 1 항 또는 제 4 항의 화합물 또는 이들의 약학적으로 허용가능한 염의 진단 용도.
