Abstract
1. Claims (29)
1. Способ применения активированной светом молекулы ионного канала, которая реагирует на световой стимул, где способ включает:
конструирование активированной светом молекулы ионного канала в клетке; и
активирование в ответ на световой стимул, который обеспечивается для молекулы ионного канала и который обладает свойствами, не активирующими ионный канал ChR2, молекулы ионного канала, что позволяет ионам проходить через активированную светом молекулу ионного канала.
2. Способ по п.1, в котором свойствами светового стимула являются длина волны и интенсивность.
3. Способ по п.1, в котором молекула ионного канала представляет единичный белковый компонент.
4. Способ по п.1, в котором клетка является стволовой клеткой, и стадию активирования проводят в целях регуляции судьбы клетки.
5. Способ по п.1, дополнительно включающий стадию оценки эффективности лечения, включающего стадию активирования, где оценку проводят как функцию мониторинга характеристик клетки.

9. Способ по п.1, дополнительно включающий стадию обеспечения одного из длительного потенцирования и длительного торможения двух популяций нейронов путем генерации спайков, по меньшей мере, в одной из двух популяций нейронов с использованием активированной светом молекулы ионного канала.
10. Способ по п.1, в котором световой стимул включает серию оптических импульсов, где продолжительность каждого импульса является достаточной для генерации отдельных потенциалов действия на каждый оптический импульс.
11. Способ по п.1, в котором ионы, проходящие через активированную светом молекулу ионного канала, включают ионы натрия.
12. Способ по п.1, в котором стадия конструирования включает, по меньшей мере, одно из трансдукции клетки с использованием вирусного вектора, содержащего активированную светом молекулу ионного канала, трансфекции клетки, микроинъецирования ДНК в клетку и генетического нацеливания активированной светом молекулы ионного канала на конкретные группы клеток.
13. Способ по п.1, дополнительно включающий стадию определения полученной концентрации ионов в клетке с использованием оптического сенсора.

25. Способ по п.24, в котором стимулирующее устройство представляет устройство для доставки света.
26. Способ по п.24, в котором режим стимуляции представляет примерно 90-130 Гц.
27. Способ по п.24, в котором режим стимуляции представляет примерно 20 Гц.
28. Способ по п.24, в котором локализованная деполяризация афферентных аксонов направлена на афферентные аксоны первичной двигательной коры.
29. Способ по п.23, дополнительно включающий стадию конструирования генетической последовательности, направленной на афферентные аксоны первичной двигательной коры, которая экспрессирует молекулу, содержащую ионный канал, который активируется в ответ на свет.
