II.用語

HMW−MAAに特異的に結合するヒトモノクローナル抗体およびその機能性フラグメントが、本明細書中に開示される。1つの例において、HMW−MAAは、以下に示されるアミノ酸配列を有する:
(1)抗体分子の一価の抗原結合フラグメントを含むフラグメントであるFabは、酵素パパインで抗体全体を消化することによって生成されることにより、インタクトな軽鎖および1つの重鎖の部分を生じ得る;
(2)抗体分子のフラグメントであるFab’は、抗体全体をペプシンで処理した後、還元することにより得られ、これによりインタクトな軽鎖および重鎖の一部がもたらされ得る;抗体1分子あたり2つのFab’フラグメントが得られる;

ヒトHMW−MAAに特異的に結合するヒトモノクローナル抗体またはその機能性フラグメントは、治療方法において使用され得る。いくつかの実施形態において、本明細書中に記載されるヒトモノクローナル抗体またはその機能性フラグメントは、治療用薬剤に結合体化され得る。免疫結合体としては、治療用薬剤が抗体に共有結合されている分子が挙げられるが、これに限定されない。治療用薬剤は、特定の標的分子または標的分子を有する細胞に対して向けられている特定の生物学的活性を有する薬剤である。当業者は、治療用薬剤が、ビンブラスチン、ダウノマイシンなどの様々な薬物、天然のまたは改変されたPseudomonas外毒素またはジフテリア毒素などの細胞毒、それ自体が薬理学的組成物を含む封入剤(例えば、リポソーム)、放射性物質(例えば、125I、32P、14C、3Hおよび35S)ならびに他の標識、標的化部分およびリガンドを含み得ることを認識する。

細胞株、細胞溶解産物および組織
ヒトメラノーマ細胞株Colo38、FO−1およびMelurならびにヒトBリンパ系細胞株LG2を、10%血清+栄養補助剤(BioWhittaker,Walkersville,MD)および2mM L−グルタミン(BioWhittaker)が補充されたRPMI1640培地(Tissue Culture Media Facility,Roswell Park Cancer Institute(RPCI),Buffalo,NY)中で維持した。ヒトメラノーマ細胞株M14およびSK−MEL−28、ヒト線維芽細胞FF2376、ヒト乳癌腫細胞株T47D、ヒト膀胱癌腫細胞株T24、ヒト前立腺癌腫細胞株PC3、ヒトBリンパ系細胞株JYおよびLKT13、ならびにラット神経細胞株B49を、10%ウシ胎児血清(FBS)(BioWhittaker)および2mM L−グルタミンが補充されたRPMI1640培地中で維持した。pcDNA3.1TM(+)のプラスミドDNA/完全長のHMW−MAA DNA構築物を用いてM14細胞をトランスフェクションした後にHMW−MAAを発現するM14/HMW−MAA細胞(Yangら、J Cell Biol 165:881−891,2004)を、10%FBS、2mM L−グルタミンおよび0.4mg/ml G418(Promega,Madison,WI)が補充されたRPMI1640培地中で生育した。37℃、5%CO2雰囲気中で細胞を培養した。報告されているように(Desaiら、Cancer Res 58:2417−2425,1998(本明細書中で参考として援用される))、細胞溶解産物を調製した。メラノサイト起源の病変は、Kumamoto University School of Medicine(Kumamoto,Japan)のDepartment of Dermatologyにおいて外科術を受けた患者から得た。メラノーマ病変の診断は、組織病理学的特徴に基づいた。以前に報告されているように(Desaiら、Cancer Res 58:2417−2425,1998(本明細書中で参考として援用される))、凍結組織切片およびホルマリン固定組織切片を調製した。
8週齢の雌BALB/cマウスをRPCIの動物コア施設から入手した。
HMW−MAA特異的マウスmAb149.53、225.28、763.74、TP61.5ならびにVF1−TP34およびVF1−TP41.2を、他で報告されているように(Wilsonら、Int J Cancer,28:293−300,1981;Giacominiら、Cancer Res 43:3586−3590,1983;Chenら、Cancer Res 51:4790−4797,1991;Temponiら、Cancer Res 52:2497−2503,1992)作製し、特徴づけた。この6種のmAbは、HMW−MAA+メラノーマ細胞への結合において互いを相互阻害(cross−inhibit)しないので、相互阻止(Cross−blocking)実験から、これらが別々かつ空間的に遠い抗原決定基を認識すると示された(Campoliら、Crit Rev Immunol 24:267−296,2004)。100KD特異的マウスmAb376.96(Imaiら、J Natl Cancer Inst 68:761−769,1982)、HLAクラスI抗原特異的マウスmAb TP25.99(Desaiら、J Immunol 165:3275−3283,2000)、c−myc腫瘍性タンパク質特異的マウスmAb 9E10(Evanら、Mol Cell Biol 5:3610−3616,1985)およびマウス抗id mAb MK2−23(Kusamaら、J Immunol 143:3844−3852,1989)が、以前に報告されている。HMW−MAA特異的ヒトscFv#28(Noronhaら、J.Immunol 161:2968−2976,1998)、#61(Desaiら、Cancer Res 58:2417−2425,1998)および#70(Noronhaら、J.Immunol 161:2968−2976,1998)ならびに抗−抗id scFv#119(Wangら、Idiotypes in Medicine:Autoimmunity,Infection and Cancer p.523,1997)を、それぞれメラノーマ細胞S5、精製HMW−MAA、メラノーマ細胞SK−MEL−28および抗id mAb MK2−23を用いたパニングによって、合成scFvライブラリー(#1)(Nissimら、Embo J 13:692−698,1994)から単離した。MAA特異的ヒトscFv F98およびW34は、それぞれメラノーマ細胞FO−1およびWM1158を用いたパニングによって半合成scFvライブラリー(de Kruifら、J Mol Biol 248:97−105,1995)から単離した。
意図されたCDR3を含む大きな半合成ファージディスプレイscFv抗体ライブラリーを、報告されているように(de Kruifら、J Mol Biol 248:97−105,1995)構築した。15アミノ酸のランダムな線状ペプチドをディスプレイするファージディスプレイペプチドライブラリーX15、および12アミノ酸のランダムなジスルフィド拘束ペプチドをディスプレイするLX−8(XCX8CX)を、報告されているように(Bonnycastleら、J Mol Biol 258:747−762,1996)構築した。
ペプチドは、RPCI Biopolymer Core Facility(Buffalo,NY)から購入した。システイン残基を含むペプチドP1C21を、6mMフェリシアン化カリウムを用いて環化し、逆相HPLCで精製した。その組成物およびジスルフィド結合形成を質量分析によって確認した。ペプチドの純度は、HPLCによって評価したとき、95%超だった。ペプチドを水で5mMの濃度に再構成し、分注し、−20℃で保存した。水に不溶性であるペプチドP1C21をジメチルスルホキシド(DMSO)で5mMの濃度に再構成した。ペプチドを、報告されているように(Grant,Current Protocols in Immunology p.9.2.8,2002)カップリング剤のマレイミドベンゾイル−N−ヒドロキシスクシンイミド(MBS)(Pierce)を用いてキーホールリンペットヘモシアニン(KLH)(Pierce)に結合体化した。
メラノーマ細胞に結合しているファージディスプレイscFv抗体を、報告されているように(Noronhaら、J Immunol 161:2968−2976,1998(本明細書中で参考として援用される))パニング法を利用してファージディスプレイscFv抗体ライブラリーから単離した。
可溶性scFv抗体は、報告されているように(Schierら、J Mol Biol 255:28−43,1996(本明細書中で参考として援用される))イソプロピル−β−D−チオガラクトピラノシド(IPTG)(Roche Applied Science,Indianapolis,IN)による誘導によって、個別のアンピシリン耐性大腸菌TG1感染コロニーから産生された。可溶性scFv抗体を、個別の細菌コロニーの培養物の培養上清(SNT)または細胞周辺腔(細胞周辺調製物,PP)から回収した。RPAS精製モジュール(Amersham)について報告された方法に従って、scFv抗体を、不溶化されたmAb 9E10におけるアフィニティークロマトグラフィによってSNTまたはPPから精製した。精製されたscFv抗体を、Centricon10(Millipore Corporation,Bedford,MA)を製造者の指示書に従って使用して、濃縮した。scFv抗体調製物の純度および活性を、それぞれphastSystemTM(Amersham)を利用するSDS−PAGE、および酵素結合免疫吸着測定法(ELISA)を利用する対応抗原を用いた試験によって評価した。
合成ペプチドおよびメラノーマ細胞に対する可溶性scFv抗体の反応性、ならびにメラノーマ細胞およびペプチドに対するマウス免疫血清の反応性を試験するELISAを、報告されているように(Desaiら、Cancer Res 58:2417−2425,1998;Desaiら、J Immunol 165:3275−3283,2000;Matsuiら、J Immunol 139:2088−2095,1987)行った。結果は、450nmにおける光学濃度(O.D.)の吸光度として表される。scFv C21によって認識される抗原決定基をマッピングする競合アッセイは、ビオチン化scFv C21(0.25μg/ウェル)をマウスmAbもしくはscFvPPの2倍希釈物と混合することによって、またはビオチン化mAb(450nmにおいて1.0と測定される吸光度をもたらす最適な量のもの)をscFvPPの2倍希釈物と混合することによって、行われた。その混合物(100μl/ウェル)を、96ウェル組織培養プレート(Falcon 3072,Becton Dickinson,Franklin Lakes,NJ)内においてHMW−MAA+細胞(2×105/50μlのRPMI1640培地)とともに4℃で1時間インキュベートした。標的細胞へのビオチン化scFv抗体およびビオチン化mAbの結合を、報告されているように(Noronhaら、J Immunol 161:2968−2976,1998)SA−HRPおよび基質との連続的なインキュベートによって測定した。結果は、HMW−MAA+細胞へのscFvまたはmAbの結合の、mAb、scFvまたはペプチド(競合物質またはインヒビター)によるパーセント阻害として表される。パーセント阻害は、式:%阻害=((インヒビターの非存在下におけるOD450−インヒビターの存在下におけるOD450)/インヒビターの非存在下におけるOD450)×100を用いて計算した。
ツニカマイシン(Sigma Chemical Co.)の存在下における125ヨウ素(Na125I;Amersham)または35Sメチオニン(トランス−35S−標識,ICN Biochemicals,Costa Mesa,CA)による細胞の標識を、報告されているように(Noronhaら、J Immunol 161:2968−2976,1998;Desaiら、J Immunol 165:3275−3283,2000)行った。標識された細胞の可溶化、免疫沈降、SDS−PAGE、オートラジオグラフィおよびフルオログラフィを、ウサギ抗マウスIgG抗体でコーティングされたプロテインAの代わりにGammon Bind+セファロース(Amersham)を使用したことを除いて、報告されているように(Desaiら、Cancer Res 58:2417−2425,1998)行った。
ビオチン化scFv C21を用いたpVIIIライブラリーX15およびLX−8のマイクロパニングを、本質的には報告されているように(Desaiら、J Immunol 165:3275−3283,2000(本明細書中で参考として援用される))96ウェルマイクロタイタープレート(Falcon 3076,Becton Dickinson)において行った。パニングの1回目は、TBS50中の1×1012ファージ粒子および1μg/ウェルの濃度のビオチン化scFv C21を用いて行った。その後の3回のパニングは、1×1010ファージ粒子のファージ投入および0.1μg/ウェルのビオチン化scFv C21を利用して行った。
scFv C21を用いた4回目のパニングの後のX15およびLX−8ライブラリーからのランダムファージクローンを、コロニーを含むプレートからのニトロセルロースフィルターリフトをプローブするために10μg/mlのscFv抗体および5μg/mlのビオチン化mAb 9E10を使用することを除いて、報告されているように(Desaiら、J Immunol 165:3275−3283,2000(本明細書中で参考として援用される))免疫学的スクリーニングによって解析した。
ファージクローンのペプチドインサートの配列を、報告されているように(Desaiら、J Immunol 165:3275−3283,2000)ジデオキシヌクレオチドチェーンターミネーション法によって決定した。
0日目はプライミングために等体積のフロイント完全アジュバント、ならびに21、42、63、84および105日目はブーストのために等体積のフロイント不完全アジュバントと混合されたP1C21ペプチド−KLH結合体(50μg/注射)を用いて、BALB/cマウス(各群8匹)の皮下に(s.c.)免疫した。プロテアソームサブユニットMB1の配列に由来する関連性のないペプチドMB1194−208で免疫されたマウスをコントロールとして用いた。132日目に、マウスを照射(20K Rads)細胞(5×105細胞/マウス)のs.c.注射によってブーストした。最初の免疫化の1週間前および各免疫化の1週間後にマウスから採血した。
scFv抗体または免疫血清中の抗体で染色されたメラノーマ細胞のフローサイトメトリー解析を、報告されているように(Wangら、J Immunol Methods 294:23−35,2004(本明細書中で参考として援用される))行った。簡潔には、細胞(5×105)を、12.5μlのscFvPPまたは0.5μgのmAb 9E10(両方とも、総体積100μlの2%BSA−PBS中に希釈されたもの)または100μlの免疫マウス血清とともに4℃で1時間インキュベートした。次いで、細胞を0.5%BSA−PBSで2回洗浄し、最適量のヤギ抗マウスIg抗体のRPE標識F(ab’)2フラグメントとともに4℃において30分間インキュベートした。2回洗浄した後、細胞を2%ホルムアルデヒド中で固定し、FACScanTMフローサイトメーター(BD Biosciences,San Jose,CA)を用いて解析した。各サンプルについての凝集物および残骸を排除するために前方散乱ゲートおよび側方散乱ゲートを用いて合計10,000細胞を数えた。結果は、相対的蛍光強度として表される。
ペプチドP1C21またはコントロールペプチドで6回免疫されたマウスの右および左の後足蹠に、それぞれ、照射されたHMW−MAA+細胞Colo38(5×105細胞/注射/)およびHMW−MAA−細胞LG2(5×105細胞/注射/)を132日目にs.c.注射した。各足蹠の厚さを、以前に報告されているように(Luoら、J Immunol 174:7104−7110,2005(本明細書中で参考として援用される))記載の時間において測定し、計算した。
試験群において得られた結果間の差の統計的有意性を、スチューデントのt検定を用いて解析した。
scFv C21の分子モデルを、AbM(Accelrys,San Diego,CA)を用いて構築した。タンパク質データベース(Bernsteinら、J Mol Biol 112:535−542,1977)からのインフルエンザウイルスノイラミニダーゼ(1NMC)特異的scFv抗体(Tulipら、J Mol Biol 227:149−159,1992)およびリゾチーム特異的scFv抗体(Ayら、J Mol Biol 301:239−246,2000)の結晶構造を用いてVL−VH二量体を作製した。CDRを定義する分類およびナンバリングスキームは、Kabatら(Kabatら、In Sequences of Proteins of Immunological Interest 5:91,1991)に従った。CDRループのL1、H1、L2およびH2は、標準的な正規立体配座(canonical conformation)をとる。H3およびL3は、タンパク質データベース(INSIGHTII,Accelrys)とCONGEN(Bruccoleriら、Nature 335:564−568,1988)のループ検索の両方を用いて構築された。ペプチドP1C21およびP3C21の出発立体配座は、INSIGHTIIに構築されたループ検索方法を用いて決定された。ペプチドP1C21およびP3C21とscFv C21との推定上の結合相互作用は、ソフトウェアAUTODOCK(Morrisら、J Comput Aided Mol Des 10:293−304,1996)とINSIGHTIIとDOCK(DesJarlaisら、J Med Chem 29:2149−2153,1986)との組み合わせを用いて決定された。
結合実験を、表面プラズモン共鳴ベースのバイオセンサー装置BIACORETM3000(Biacore AB,Uppsala,Sweden)を25℃において用いて行った。センサー表面におけるscFv C21の固定化を、製造者の指示書に従い、標準的なアミンカップリング手順に従って行った。簡潔には、0.2M N−エチル−N’−(ジメチルアミノプロピル)カルボジイミド(EDC)および0.05M N−ヒドロキシスクシンイミド(NHS)を含む35μlの溶液を、5μl/分の流速で注入することにより、センサーチップ表面上のカルボキシル基を活性化させた。scFv C21(40ng/mlの10mM NaOAc緩衝液,pH5.0)を、所望の結合タンパク質レベルに達するまで、20μl/分の流速でそのチップ表面の上に流した。未反応のscFv C21を洗い流し、35μlの1Mエタノールアミンを5μl/分で注入することによって、未反応の活性化された基をブロッキングした。scFv C21の最終的な固定化応答は、3,000RUだった。参照表面を、同じ条件下であるがscFv C21注入を行わずに同時に作製し、装置および緩衝液アーチファクトについて補正するためのブランクとして使用した。ペプチドを、20〜100μl/分の様々な流速において可変濃度で注射した。チップ上に固定化されたscFv C21へのペプチドの結合をリアルタイムでモニターした。SPRから得られる結合定数がアナライトの再結合または大量注入(mass transport)効果に影響されないことを排除するために、様々な流速(20〜100μl/分)で研究を反復した。両方の流速において得られた速度定数は、変化しないままだった。
HMW−MAA+細胞Colo38を用いた半合成ファージディスプレイscFv抗体ライブラリーのパニングによるscFv抗体の単離
Colo38細胞を用いたパニングによって半合成ファージディスプレイscFv抗体ライブラリーから単離された40種の可溶性scFv抗体の、HMW−MAA+細胞Colo38およびHMW−MAA−Bリンパ系細胞LG2を用いたELISAにおけるスクリーニングによって、Colo38細胞との選択的な反応性を有する23個のクローンが同定された。その23個のクローンを、Colo38、FF2376線維芽細胞およびT24膀胱癌腫細胞を用いて追加試験することにより、クローンC3、C21、C29およびC30が単離された(これらのすべてが、Colo38細胞との選択的な反応性を示した)。scFv C21が最大の反応性を示したので、さらなる解析のためにscFv C21を選択した。その他のクローンは、失ったので、特徴づけられなかった。
異なるHMW−MAA発現を有するヒト細胞株のパネルおよびHMW−MAAホモログを発現するラット神経細胞株B49を用いたELISAにおいて試験したとき、scFv C21は、メラノーマ細胞株Colo38、FO−1、MelurおよびSK−MEL−28だけと反応した。それらのすべてが、HMW−MAAを発現することから、scFv C21が、HMW−MAAに特異的であることが示唆される(図1)。この可能性は、3つの系統の証拠によって立証された。第1に、フローサイトメトリー解析は、scFv C21が、完全長HMW−MAAをコードするプラスミドDNAによる安定なトランスフェクションの後にHMW−MAAを発現するM14細胞を染色したが、HMW−MAAを発現しない親のM14細胞を染色しなかったことを示した(図2)。第2に、scFv C21は、125Iで標識されたColo38細胞のHMW−MAA構成要素の特徴的な電気泳動(elecrophoretic)プロファイルを有する2つの構成要素を免疫沈降させた。最後に、連続的な免疫沈降実験において、scFv C21は、HMW−MAA特異的マウスmAb763.74を用いて免疫除去されていたColo38細胞溶解産物からのいかなる構成要素も免疫沈降させなかった。scFv C21が、100KD MAA特異的mAb376.96で免疫除去されたColo38細胞溶解産物のHMW−MAA構成要素を免疫沈降させたので、この免疫除去は、特異的である(図3)。逆に、scFv C21によるColo38細胞溶解産物の免疫除去は、ほとんどを除去したが、すべての構成要素がmAb763.74によって免疫沈降されたわけではない。これらの結果は、mAb763.74によって認識されるほとんどであるがすべてではないHMW−MAA分子上のscFv C21によって認識される決定基の発現および/またはmAb763.74の会合定数よりも小さいscFv C21の会合定数を反映している。
N−グリコシル化インヒビターのツニカマイシンの存在下において、35S−メチオニンで固有に標識されたColo38細胞から免疫沈降されたHMW−MAA構成要素の強度が著しく低下するので、炭水化物が、scFv C21によって定義される決定基の発現において役割を果たすと見られる。N−グリコシル化の阻害は、mAb763.74と反応する220kD前駆体の蓄積によって示唆される(図4)。競合実験では、scFv C21によって定義される決定基と、マウスmAbのパネルによって定義される6つの決定基ならびにヒトscFv#28、#61および#70によって定義される決定基との空間的な関係を調べた。scFv C21およびmAb VF1−TP34は、メラノーマ細胞SK−MEL−28への結合において互いを部分的に阻害した。この阻害は、用量依存的である(図5)。対照的に、マウスmAb149.53、225.28、763.74、TP61.5およびVF1−TP41.2ならびにscFv#28、#61および#70は、SK−MEL−28細胞へのscFv C21の結合を阻害しなかった。これらの結果から、scFvC21によって定義される決定基は、mAb VF1−TP34によって定義される決定基と異なるものであって、それと空間的に近位であり、残りのマウスmAbならびにヒトscFv#28、#61および#70によって定義される決定基と異なるものであって、それらと空間的に遠位であることが示唆される。
scFvC21は、免疫ペルオキシダーゼ反応において、凍結されたメラノサイト病変を染色したが、ホルマリン固定されたパラフィン包埋メラノサイト病変を染色しなかった。この染色は、膜と細胞質の両方におけるものだった(図6)。scFvC21およびHMW−MAA特異的マウスmAb763.74による良性および悪性のメラノサイト病変の染色パターンの比較から、両方の抗体が、膜および細胞質のパターンで4つの色素性母斑を均一に染色することが示された。さらに、mAb763.74は、15個中14個の主要な病変を均一なパターンで染色し、scFvC21は、9個を均一なパターンで染色し、2個を不均一なパターンで染色した。この染色は、1つの病変において膜におけるものであり、4つでは膜および細胞質におけるものであり、6つでは細胞質におけるものだった。最後に、mAb763.74は、6つの転移性病変を均一なパターンで染色し、scFvC21は、3つを均一なパターンで、3つを不均一なパターンで染色した。この染色は、3つの病変において膜および細胞質におけるものであり、残りの3つでは細胞質におけるものだった。
scFvC21のヌクレオチド配列およびアミノ酸配列を標準的な方法に従って決定した。VH、VLおよびリンカーのヌクレオチド配列およびアミノ酸配列を以下に提供する。免疫グロブリン重鎖のヒンジ、CH2およびCH3ドメインのヌクレオチド配列も提供する。

実施例4:scFvペプチド模倣物の同定および特徴づけ
ファージディスプレイペプチドライブラリーLX−8およびX15から単離されたscFvC21に結合するペプチドの配列
免疫学的スクリーニングから、scFvC21が、ファージディスプレイペプチドX15ライブラリーからのパニングによって単離されたクローンの20%と強く反応するが、LX−8ライブラリーから単離されたクローンのいずれとも反応しないことが明らかになった。免疫スクリーニングから得られた陽性クローンのファージ上清は、ELISAにおいてもscFvC21と反応した。これらのクローンが、無関係なMAAを認識するscFv F98およびW34と反応しなかったので、この反応性は、特異的である。scFvC21と反応するクローンのうちの16のヌクレオチド配列解析から、解析された16クローンの88、6および6%においてそれぞれ配列SPSWYCPDCDKRPLV(P1C21)(配列番号7)、EARNWHDFPIHPRTL(P2C21)(配列番号8)およびRPYRYDPLGDLKSRH(P3C21)(配列番号10)が同定された。ペプチドP1C21およびP3C21の配列は、コンセンサス
scFvC21に対するファージディスプレイペプチドの反応性を確証するために、ペプチドP1C21(環状)、P2C21(線状)およびP3C21(線状)を合成し、ELISAにおいてscFvC21と反応する能力および阻害アッセイにおいてHMW−MAA+メラノーマ細胞Colo38への結合を阻害する能力について試験した。0.25%グルタルアルデヒドPBSによってマイクロタイタープレート上に固定化された合成ペプチドP1C21およびP2C21は、ELISAにおいてscFvC21と特異的に反応したが、固定化された合成ペプチドP3C21は、反応しなかった(図7)。ペプチドが無関係なHMW−MAA特異的scFv#28と反応しなかったので、この結合は、特異的である。さらに、ペプチドP1C21およびP3C21は、用量依存的様式でHMW−MAA+細胞へのscFvC21の結合を阻害した(図8)。この両方のペプチドが、メラノーマ細胞Colo38へのscFv#28の結合を阻害しなかったので、この阻害は、特異的である(図8)。対照的に、P2C21は、メラノーマ細胞Colo38へのscFvC21の結合を阻害しなかった。可溶性ペプチドP3C21によってメラノーマ細胞へのscFvC21の結合が阻害されること、および固定化されたペプチドP3C21とscFvC21との反応性がないことは、このペプチドとscFvC21との結合が、立体構造的に感受性であることを示唆する。
ペプチドP1C21によるHMW−MAA模倣をさらに立証するために、BALB/cマウスにおいてHMW−MAA特異的な体液性免疫および細胞性免疫を誘発する能力を試験した。HMW−MAA+メラノーマ細胞Colo38と選択的に反応する抗体が、ペプチドP1C21による3回目の免疫の1週間後にBALB/cマウスから回収された血清中で検出された。これらの抗体の平均力価は、低かったが、無関係なMB1194−208ペプチドで免疫されたマウス由来の血清中のこれらの抗体の平均力価よりも有意に(p<0.05)高かった。さらに、メラノーマ細胞と反応する抗体の力価は、3回のさらなる追加免疫の後も変化しなかった。しかしながら、HMW−MAA+メラノーマ細胞と選択的に反応する抗体の力価は、ペプチドP1C21で免疫されたマウスを、HMW−MAA+メラノーマ細胞でブーストすることによって著しく増加した(図9)。
scFvC21へのペプチドP1C21およびP3C21の結合に対する構造的根拠を定義するために、scFvC21およびこれら2つのペプチドとの複合体の分子モデルを構築した。INSIGHTIIにおける限られた立体配座検索アルゴリズムを用いて、ペプチドP1C21およびP3C21の最初のモデルを構築した。scFvC21へのペプチド結合の最初の配向をDOCK解析によって決定した。最善の配向を、接触スコアならびに結合エネルギーに基づいて選択し、次いで最適な結合および相互作用を解析するためにINSIGHTIIを用いて最小化した(表1)。その最小化中、最初の2000サイクルについてはscFvC21を固定したままにした。続いて、CDRループおよびペプチドを動かした。
起きうる構造遷移および潜在的な接触残基と関連する影響をさらに特徴づけるために、分子モデリング研究に基づいてアミノ酸置換を線状ペプチドP3C21に導入した。ペプチドP3C21とscFvC21との反応性に対する選択された置換の影響を、Biacore(表2)および阻害解析(図8)によって実験的に試験した。
この実施例では、HMW−MAAの過剰発現を示す癌(本明細書中で「HMW−MAA陽性」癌とも呼ばれる)(メラノーマ、乳癌、前立腺癌および扁平上皮癌腫を含むがこれらに限定されない)を処置するためのHMW−MAA特異的ヒトモノクローナル抗体の使用が説明される。HMW−MAA陽性癌と診断された患者は、当該分野において標準的な手順に従って処置され得る。一般に、処置の選択肢としては、外科術、放射線治療、化学療法、免疫療法またはインターフェロン療法が挙げられる。
マウス:雌SCID/BALB/cマウス(C.B−Igh−1bIcrTac−Prkdcscid,6〜8週齢)は、NCIまたはTaconic Farms,Incから購入した。本研究のための実験は、Institutional Animal Care and Use Committeeによって承認されたものである。
フロー解析から、scFv−FcC21が、メラノーマ細胞株MDA−MB−435、MV3およびWM1158;神経膠腫細胞株LN444およびA1207ならびに頭頸部癌細胞株PCI30およびPCI13を含むヒト培養細胞株のパネルの細胞表面を特異的に染色したことが示されている。CSPG4−メラノーマ細胞株M14をネガティブコントロールとして用い、M14細胞株のCSPG4トランスフェクタントであるM14/CSPG4をポジティブコントロールとして用いた(図14〜15を参照のこと)。
scFv−Fc21は、腫瘍細胞の成長および遊走をインビトロにおいて阻害した(図22〜23)。この方法は、上記の実施例6に記載されている。
scFv−FcC21で処理されたMV3細胞は、低レベルのタンパク質キナーゼCアルファ(PKCα)およびFAKを有した。さらに、scFv−FcC21処置により、FAK、Erk1/2、PDK1(Aktの上流)およびAktのリン酸化/活性化が阻害された。Akt活性化の低下は、PI3K/Aktシグナル伝達経路の負の制御因子であるリン酸化/活性化PTENの増加を伴った(図24〜26を参照のこと)。
scFc−FcC21は、MV3細胞の実験的転移を有意に阻害した(図16)。代用マーカーのp−ヒストンH3を用いて腫瘍細胞の増殖速度についても転移性病変を評価した。scFc−FcC21の処置により、有糸***細胞の数が有意に減少した(図17)。
scFc−FcC21がSCIDマウスにおいてMV3腫瘍の腫瘍再発および自然転移を阻害する能力を、原発腫瘍の外科的除去後に試験した。scFc−FcC21で処置されたマウスは、コントロールscFv−Fcを投与されたマウスよりも有意に遅い速度の腫瘍再発および低いレベルの自然肺転移を有した(図18〜20)。さらに、scFc−FcC21で処置されたマウスから外科的に取り出された原発腫瘍を、メラノーマの成長および進行に関連する特異的シグナル伝達経路の活性化についても評価した。PKC−αおよびSrcシグナル伝達経路の活性化は、コントロールscFv−Fc119で処置されたマウスから取り出された原発腫瘍と比べて、scFv−FcC21で処置されたマウスから取り出された原発腫瘍において著しく低下した(図21)。

- 前記単離されたヒトモノクローナル抗体またはそのscFvが、直接標識されている、請求項15に記載の組成物。
- 前記単離されたヒトモノクローナル抗体またはそのscFvに特異的に結合する2次抗体が前記組成物と組み合わせて使用され、そして前記サンプルと接触され、そして該2次抗体の結合が検出され、ここで、コントロールサンプルへの該2次抗体の結合と比較した際の該サンプルへの該2次抗体の結合の増加が、該被験体における癌を検出するかまたは該被験体における癌の診断を確証する、請求項15または16に記載の組成物。
- 前記癌が、メラノーマ、乳癌、頭頸部扁平上皮癌腫、前立腺癌、卵巣癌、結腸癌、神経膠腫、胃癌または膵癌である、請求項15〜17のいずれかに記載の組成物。
- 前記コントロールサンプルが、癌を有しない被験体由来のサンプルである、請求項15〜18のいずれかに記載の組成物。
- 前記サンプルが、血液、尿、生検材料、血清、痰、血漿、または脳髄液サンプルである、請求項15〜19のいずれかに記載の組成物。
- 前記癌が、転移性である、請求項15〜20のいずれか1項に記載の組成物。
- 請求項1〜5のいずれかに記載の単離されたヒトモノクローナル抗体またはそのscFvをコードする単離された核酸分子。
- 前記ヒトモノクローナル抗体のVHドメインが、配列番号1のヌクレオチド配列を含む、請求項22に記載の単離された核酸分子。
- 前記ヒトモノクローナル抗体のVLドメインが、配列番号3のヌクレオチド配列を含む、請求項22または請求項23に記載の単離された核酸分子。
- プロモーターに作動可能に連結された、請求項22〜24のいずれか1項に記載の単離された核酸分子。
- 請求項22〜25のいずれか1項に記載の単離された核酸分子を含む、発現ベクター。
- 請求項22〜25のいずれか1項に記載の単離された核酸分子または請求項26に記載の発現ベクターで形質転換された、単離された宿主細胞。
