5 La actividad protectora antiviral de N-(4-{[4-amino-2-butil-1H-imidazo[4,5-c]quinolin-1-il]oxi}butil)octadecanamida (Comp. del Ejemplo 1) se evaluó en ratones macho Balb/c (Charles River, Wilmington, MA) infectados intranasalmente con H1N1 A/Puerto Rico/8/34 adaptado a ratones (obtenido de la American Type Culture Collection, Manassas, VA). Cuatro semanas antes de la infección, grupos de 10 ratones cada uno se inmunizaron con 1. PBS;

Los antígenos se pueden cosuministrar con un compuesto de Fórmula I, por ejemplo en mezcla en una composición farmacéutica según la presente invención. Tales composiciones farmacéuticas pueden incluir el compuesto de Fórmula I en liposomas. Esto puede permitir que el compuesto de Fórmula I alcance, por ejemplo, a las células presentadoras de antígeno al mismo tiempo o aproximadamente al mismo tiempo que el antígeno. En algunas realizaciones, una composición farmacéutica según la presente invención puede ser una formulación de dosificación tópica convencional (por ejemplo, una crema, un ungüento, una formulación de aerosol, una pulverización no de aerosol, un gel, o una loción). Se ha mostrado que el compuesto de Fórmula I induce la producción de TNF- como se describe anteriormente. La capacidad para inducir la producción de TNF indica que el compuesto de Fórmula I es útil como un modificador de la respuesta inmunitaria que puede modular la respuesta inmunitaria de muchas maneras diferentes, haciéndolo útil en el tratamiento de una variedad de trastornos. Otras citocinas cuya producción se puede inducir mediante la administración del compuesto de Fórmula I incluyen generalmente interferones de tipo I (por ejemplo, INF-), IL-6, IL-8, IL-10, IL-12, MIP-1, MCP-1, y una variedad de otras citocinas. La actividad de IRM del compuesto de Fórmula I también puede incluir la inducción de la producción de citocinas mediante células T, activando células T específicas de un antígeno, y/o activando células dendríticas. Además, la actividad de IRM del compuesto de Fórmula I puede incluir proliferación y diferenciación de linfocitos B. El mecanismo para la actividad antivírica y antitumoral del compuesto de Fórmula I puede ser debido en parte sustancial a la potenciación del sistema inmunitario mediante inducción de diversas citocinas importantes (por ejemplo, al menos una de factor de necrosis tumoral, interferones, o interleucinas). Se ha mostrado que tales compuestos estimulan una liberación rápida de ciertas citocinas derivadas de monocitos/macrófagos, y también son capaces de estimular células B para que segreguen anticuerpos que desempeñan un papel importante en estas actividades antivíricas y antitumorales de los compuestos de IRM.
Se entenderá que en el tratamiento de las enfermedades mencionadas anteriormente, por ejemplo, el compuesto de Fórmula I se puede usar en combinación con otras terapias, tales como los agentes activos mencionados anteriormente, y otros procedimientos (por ejemplo, quimioablación, ablación por láser, crioterapia, y extirpación quirúrgica).

Una cantidad eficaz para tratar o inhibir una infección vírica es una cantidad que provocará una reducción en una o más de las manifestaciones de la infección vírica, tales como lesiones víricas, carga viral, velocidad de producción del virus, y mortalidad, en comparación con los animales del control no tratados. La cantidad precisa que es eficaz para tal tratamiento variará según factores conocidos en la técnica, pero se espera que sea una dosis de alrededor de 100 ng/kg a alrededor de 50 mg/kg, en algunas realizaciones alrededor de 10 g/kg a alrededor de 5 mg/kg. En algunas de estas realizaciones, el método incluye administrar suficiente compuesto para proporcionar una dosis de alrededor de 10 g/kg a alrededor de 5 mg/kg al sujeto, por ejemplo una dosis de alrededor de 100 g/kg a alrededor de 1 mg/kg. En algunas realizaciones de los métodos descritos aquí, el compuesto de Fórmula I se puede administrar, por ejemplo, desde una única dosis a múltiples dosis por semana, aunque en algunas realizaciones los métodos de la presente invención se pueden llevar a cabo administrando el compuesto de Fórmula I a una frecuencia fuera de este intervalo. En algunas realizaciones, el compuesto de Fórmula I se puede administrar desde alrededor de una vez por mes a alrededor de cinco veces por semana. Puesto que el compuesto de Fórmula I se puede formular para proporcionar niveles sistémicos reducidos del compuesto a la vez que induce niveles elevados de citocinas, se cree que es muy útil para proporcionar una respuesta inmunitaria local potenciada, a la vez que minimiza los efectos secundarios sistémicos indeseables.
