以下に本願発明にかかる化合物の一般製造法を示すが、これらに限定されるものではない。また、本発明化合物の一般製造法における各原料化合物・各種試薬は、塩あるいは溶媒和物(特に水和物)を形成していてもよい。

X2はヨウ素原子、臭素原子、塩素原子、メタンスルホニルオキシ基、p-トルエンスルホニルオキシ基等の脱離基を意味する。

本工程は、溶媒中、化合物(1A-1)と化合物(1A-2)とを、塩基、およびパラジウム触媒存在下、反応させることにより、化合物(1B-1)を製造する工程である。
本工程は、国際公開第04/037822号パンフレット(製造方法5中の工程5-E、実施例47)、後述の実施例2中の(2f)等に記載された反応条件を参考にして行うことができる。
一般式(1A-1)で表される化合物は実施例2中の(2e)等により製造することができる。
一般式(1A-2)で表される化合物としては、国際公開第04/037822号パンフレット(製造例29、33)や後述する製造例1C(3)、2C(6)、実施例16(b)、17(d)等に記載された反応条件により製造することができる化合物を用いることができる。また、一般式(1A-2)で表される化合物として、購入可能な化合物、または購入可能な化合物から当業者が通常行う方法により容易に製造することができる化合物を用いることもできる。
本反応は、窒素、アルゴン等の不活性気体の気流下または雰囲気下でも行うことができる。
本反応に用いる溶媒としては、出発原料をある程度溶解するものであり、かつ、反応を阻害しないものであれば、特に制限はないが、例えば、メタノール、エタノール、n-ブタノール等のアルコール系溶媒、テトラヒドロフラン、1,2-ジメトキシエタン、メチル-tert-ブチルエーテル、シクロペンチルメチルエーテル、ジエチルエーテル、ジイソプロピルエーテル、ジブチルエーテル、ジシクロペンチルエーテル、1,2-ジメトキシエタン、1,4-ジオキサン等のエーテル系溶媒、ベンゼン、トルエン、キシレン、メシチレン等の芳香族炭化水素系溶媒、N,N-ジメチルホルムアミド等のアミド系溶媒、ヘプタン、ヘキサン等の脂肪炭化水素系溶媒、1-メチル-2-ピロリジノン、水またはこれらの混合溶媒等を用いることができ、好適には、水、アルコール系溶媒、芳香族炭化水素系溶媒、エーテル系溶媒またはこれらの混合溶媒であり、より好適にはエタノールとトルエンあるいは1,2-ジメトキシメタンと水の混合溶媒である。
本工程における上記塩基としては、出発原料、使用する溶媒等により異なり、特に限定されないが、例えば水酸化リチウム、水酸化ナトリウム、水酸化カリウム、水酸化バリウム、炭酸リチウム、炭酸ナトリウム、炭酸カリウム、炭酸水素ナトリウム、炭酸水素カリウム、炭酸セシウム、フッ化セシウム、フッ化カリウム等の無機塩基、イミダゾール、ピリジン等の環状系塩基またはトリエチルアミン、N,N-ジイソプロピルエチルアミン等の有機アミン等が挙げられるが、好適には炭酸ナトリウム、炭酸カリウム等である。
上記パラジウム触媒としては、出発原料、使用する溶媒等により異なり、また反応を阻害しない限りにおいて特に限定されないが、好適にはテトラキス(トリフェニルホスフィン)パラジウム(0)、パラジウム(II)アセテート/トリフェニルホスフィン、パラジウム(II)アセテート/2-ジシクロヘキシルホスフィノ-2’,4’,6’-トリイソプロピルビフェニル、塩化パラジウム(II)、トリス(ジベンジリデンアセトン)ジパラジウム(0)/トリ-tert-ブチルホスフィン、ジクロロ[1,1’-ビス(ジフェニルホスフィン)-フェロセン]パラジウム(0)等が挙げられる。
反応温度は、通常、出発原料、溶媒、その他反応に用いる試薬によって異なり、好適には、0℃~溶媒の還流温度(反応容器中の内温)であり、より好適には、60℃~100℃である。
反応時間は、通常、出発原料、溶媒、その他反応に用いる試薬、反応温度によって異なり、好適には、試薬を加えた後、上記温度にて1~48時間であり、より好適には、1~6時間である。
化合物(1A-2)は、化合物(1A-1)に対して、1~5倍モル当量用いることができるが、好適には、1~3倍モル当量である。
上記塩基は、化合物(1A-1)に対して、1~10倍モル当量用いることができるが、好適には2~5倍モル当量である。
上記パラジウム触媒は、化合物(1A-1)に対して、0.01~1倍モル当量用いることができるが、好適には0.05~0.1倍モル当量である。
本工程は、溶媒中、化合物(1B-1)と化合物(1B-2)とを塩基存在下、反応させることにより化合物(1C-1)を製造する工程である。
本工程は、国際公開第04/037822号パンフレット(製造方法5中の工程5-A、製造例24)、後述の実施例2中の(2g)等に記載された反応条件を参考にして行うことができる。
本反応は、窒素、アルゴン等の不活性気体の気流下または雰囲気下でも行うことができる。
化合物(1B-2)としては、後述する製造例1Bに記載された反応条件等により製造することができる化合物を用いることができる。化合物(1B-2)として、購入可能な化合物、または購入可能な化合物から当業者が通常行う方法により容易に製造することができる化合物を用いることもできる。
本反応に用いる溶媒としては、出発原料をある程度溶解するものであり、かつ、反応を阻害しないものであれば、特に制限はないが、例えば、アセトニトリル等のニトリル系溶媒、テトラヒドロフラン、1,2-ジメトキシエタン、メチル-tert-ブチルエーテル、シクロペンチルメチルエーテル、ジエチルエーテル、ジイソプロピルエーテル、ジブチルエーテル、ジシクロペンチルエーテル等のエーテル系溶媒、ベンゼン、トルエン等の芳香族炭化水素系溶媒、N,N-ジメチルホルムアミド等のアミド系溶媒、ヘプタン、ヘキサン等の脂肪炭化水素系溶媒、ジメチルスルホキシドまたはこれらの混合溶媒等を用いることができ、好適には、N,N-ジメチルホルムアミドあるいはジメチルスルホキシドである。
本工程における塩基としては、出発原料、使用する溶媒等により異なり、特に限定されないが、例えば水酸化リチウム、水酸化ナトリウム、水酸化カリウム、炭酸リチウム、炭酸ナトリウム、炭酸カリウム、炭酸水素ナトリウム、炭酸水素カリウム、炭酸セシウム等の無機塩基、カリウム-tert-ブトキシド、ブチルリチウム、メチルリチウム、リチウムビストリメチルシリルアミド、ナトリウムビストリメチルシリルアミド、カリウムビストリメチルシリルアミド等の有機金属系塩基、水素化リチウム、水素化ナトリウム、水素化カリウム等のハイドライド系塩基、イミダゾール、ピリジン、4-ジメチルアミノピリジン等の環状系塩基またはトリエチルアミン、N,N-ジイソプロピルエチルアミン等の有機アミン等が挙げられるが、好適には水素化ナトリウムあるいは水酸化ナトリウム等である。
反応温度は、通常、出発原料、溶媒、その他反応に用いる試薬によって異なり、好適には、0℃~80℃(反応容器中の内温)である。
反応時間は、通常、出発原料、溶媒、その他反応に用いる試薬、反応温度によって異なり、試薬を加えた後、上記反応温度にて1~8時間撹拌するのが好適である。
化合物(1B-2)は、化合物(1B-1)に対して1~5倍モルの量を用いることができる。
塩基は、化合物(1B-1)に対して1~5倍モル当量用いることができ、好適には、1.5倍モル当量である。
本工程は、化合物(1C-1)を酸処理することにより化合物(1D-1)を製造する工程である。
本工程は、国際公開第04/037822号パンフレット(製造方法5中の工程5-B、製造例24)、後述の実施例2中の(2g)等に記載された反応条件を参考にして行うことができる。
本工程は、溶媒不存在下、または溶媒存在下でも実施することができるが、溶媒を使用する場合、本反応に用いる溶媒としては、出発原料をある程度溶解するものであり、かつ、反応を阻害しないものであれば、特に制限はないが、例えば、テトラヒドロフラン、1,2-ジメトキシエタン、メチル-tert-ブチルエーテル、シクロペンチルメチルエーテル、ジエチルエーテル、ジイソプロピルエーテル、ジブチルエーテル、ジシクロペンチルエーテル、1,4-ジオキサン等のエーテル系溶媒、ベンゼン、トルエン等の芳香族炭化水素系溶媒、ヘプタン、ヘキサン等の脂肪炭化水素系溶媒、酢酸エチル、ジクロロメタン等のハロゲン系溶媒、アセトニトリルまたはこれらの混合溶媒等を用いることができ、好適には、ジクロロメタンである。
本工程における上記酸とは、塩酸、硫酸、トリフルオロ酢酸等を意味するが、好適には、トリフルオロ酢酸である。
反応温度は、通常、出発原料、溶媒、その他反応に用いる試薬によって異なり、好適には、0℃~40℃(反応容器中の内温)である。
反応時間は、通常、出発原料、溶媒、その他反応に用いる試薬、反応温度によって異なり、好適には、試薬を加えた後、上記反応温度にて1~8時間撹拌するのが好適である。
上記酸は、化合物(1C-1)に対して1モル以上の量を用いることができ、反応溶媒として、大過剰の酸を用いることもできる。
工程1Bおよび工程1Cは、下記記載の実施例2中の(2g)と同様に、各工程において特に精製することなく、反応液の簡単な処理、溶媒の留去の操作のみで次の工程の原料として使用することもできる。
本工程は、溶媒中、化合物(1D-1)と化合物(1D-2)とを、還元剤存在下、酸存在下または不存在下で反応させることにより化合物(1E-1)を製造する工程である。
本工程は、国際公開第04/037822号パンフレット(製造方法5中の工程5-C、製造例24)、実施例2中の(2g)等に記載された反応条件を参考にして行うことができる。
本反応は、窒素、アルゴン等の不活性気体の気流下または雰囲気下でも行うことができる。
化合物(1D-2)としては、上記に定義のように、アルデヒド化合物やケトン化合物が挙げられるが、後述する製造例1A等に記載された反応条件により製造することができる化合物、購入可能な化合物、または購入可能な化合物から当業者が通常行う方法により容易に製造することができる化合物を用いることができる。
本反応に用いる溶媒としては、出発原料をある程度溶解するものであり、かつ、反応を阻害しないものであれば、特に制限はないが、例えば、メタノール、エタノール等のアルコール系溶媒、テトラヒドロフラン、1,2-ジメトキシエタン、メチル-tert-ブチルエーテル、シクロペンチルメチルエーテル、ジエチルエーテル、ジイソプロピルエーテル、ジブチルエーテル、ジシクロペンチルエーテル等のエーテル系溶媒、酢酸等があげられ、これらは単独あるいは混合溶媒として用いることが可能である。
還元剤は、カルボニル化合物とアミン化合物との還元的アミノ化反応に通常用いられている還元剤を用いることができ、本反応は特に限定されないが、例えばαピコリンボラン、ナトリウム トリアセトキシボロヒドリド、水素化ホウ素ナトリウム等を挙げることができる。
本工程で用いる酸としては、トリフルオロ酢酸、酢酸などを挙げることができる。
反応温度は、通常、出発原料、溶媒、その他反応に用いる試薬によって異なり、好適には、0℃~溶媒の還流温度(反応容器中の内温)であり、より好適には、室温である。
反応時間は、通常、出発原料、溶媒、その他反応に用いる試薬、反応温度によって異なり、好適には、試薬を加えた後、上記温度にて1~48時間であり、より好適には、1~6時間である。
還元剤は、化合物(1D-1)に対し、0.5~3倍モル当量用いることができ、好適には1~2倍モル当量である
化合物(1D-2)は、化合物(1D-1)に対し、1~5倍モル当量用いることができ、好適には1~2倍モル当量である。
本工程は、化合物(1B-1)を酸処理することにより、化合物(2B-1)を製造する工程である。
本工程の反応条件としては、前記製造方法1、工程1Cに記載された反応条件や、後述の実施例12中の(12a)等に記載された反応条件を参考にして行うことができる。
本反応に用いる溶媒としては、出発原料をある程度溶解するものであり、かつ、反応を阻害しないものであれば、特に制限はないが、例えば、酢酸エチル、水またはこれらの混合溶媒等を用いることができる。
本工程における上記酸とは、塩酸、硫酸、トリフルオロ酢酸等を意味するが、好適には、塩酸の酢酸エチル溶液またはトリフルオロ酢酸である。
反応温度は、通常、出発原料、溶媒、その他反応に用いる試薬によって異なり、好適には、0℃~40℃である。
反応時間は、通常、出発原料、溶媒、その他反応に用いる試薬、反応温度によって異なり、好適には、試薬を加えた後、上記温度にて1~48時間であり、より好適には、1~6時間である。
上記酸は、化合物(1B-1)に対して、1モル以上の量を用いることができ、反応溶媒として、大過剰の酸を用いることもできる
本工程は、溶媒中、化合物(2B-1)と化合物(2B-2)とを、縮合剤存在下、反応させることにより、化合物(2C-1)を製造する工程である。
本工程は、後述の実施例12中の(12b)等に記載された反応条件を参考にして行うことができる。
一般式(2B-2)で表される化合物としては、購入可能な化合物、または購入可能な化合物から当業者が通常行う方法により容易に製造することができる化合物を用いることもできる。
本反応は、窒素、アルゴン等の不活性気体の気流下または雰囲気下でも行うことができる。
本反応に用いる溶媒としては、出発原料をある程度溶解するものであり、かつ、反応を阻害しないものであれば、特に制限はないが、例えば、テトラヒドロフラン、1,2-ジメトキシエタン、メチル-tert-ブチルエーテル、シクロペンチルメチルエーテル、ジエチルエーテル、ジイソプロピルエーテル、ジブチルエーテル、ジシクロペンチルエーテル、1,2-ジメトキシエタン、1,4-ジオキサン等のエーテル系溶媒、N,N-ジメチルホルムアミド等のアミド系溶媒、またはこれらの混合溶媒等を用いることができる。
本工程における上記縮合剤としては、出発原料、使用する溶媒等により異なり、特に限定されないが、例えば水溶性カルボジイミド(1-エチル-3-(3-ジメチルアミノプロピル)カルボジイミド 塩酸塩)、1-ヒドロキシベンゾトリアゾール、またはこれらの組み合わせ等である。
反応温度は、通常、出発原料、溶媒、その他反応に用いる試薬によって異なり、好適には、0℃~40℃である。
反応時間は、通常、出発原料、溶媒、その他反応に用いる試薬、反応温度によって異なり、好適には、試薬を加えた後、上記温度にて1~48時間である。
化合物(2B-2)は、化合物(2B-1)に対して、1~5倍モル当量用いることができるが、好適には、1~1.5倍モル当量である。
上記縮合剤は、化合物(2B-1)に対して、1~10倍モル当量用いることができる。
本工程は、溶媒中、化合物(2C-1)を、還元剤存在下、還元することにより、化合物(2D-1)を製造する工程である。
本工程は、後述の実施例12中の(12c)等に記載された反応条件を参考にして行うことができる。

反応温度は、通常、出発原料、溶媒、その他反応に用いる試薬によって異なり、好適には、0℃~溶媒の還流温度(反応容器中の内温)である。
反応時間は、通常、出発原料、溶媒、その他反応に用いる試薬、反応温度によって異なり、好適には、試薬を加えた後、上記温度にて1~24時間である。
上記還元剤は、化合物(2C-1)に対して、1~10倍モル当量用いることができるが、好適には2~5倍モル当量である。
工程2Cは、下記記載の実施例12中の(12c)と同様に、各工程において特に精製することなく、反応液の簡単な処理、溶媒の留去の操作のみで次の工程の原料として使用することもできる。
本工程は、化合物(2D-1)と化合物(1D-2)とを反応させることにより、化合物(2E-1)を得る工程である。
本工程の反応条件としては、製造方法1、工程1Dと同様な条件、操作方法を適用できる。
上記各方法において、各工程の反応終了後、各工程の目的化合物は常法に従い、反応混合物から採取することができる。
例えば、反応混合物全体が液体の場合、反応混合物を所望により室温に戻すか、氷冷し、適宜、酸、アルカリ、酸化剤または還元剤を中和し、水と酢酸エチルのような混和せずかつ目的化合物と反応しない有機溶媒を加える。混合物を充分振とう後、静置し、分離する二層のうち目的化合物を含む層を分取する。次に、得られた層と混和せず目的化合物と反応しない溶媒を加え、目的化合物を含む層を洗浄し、当該層を分離する。当該層が有機層の場合、無水硫酸マグネシウムまたは無水硫酸ナトリウム等の乾燥剤を用いて乾燥し、溶媒を留去することにより、目的化合物を得ることができる。また、当該層が水層である場合、電気的に脱塩した後、凍結乾燥することにより、目的化合物を得ることができる。
上記方法で採取した目的化合物の純度を向上させるため、適宜、再結晶法、各種クロマトグラフィー法、蒸留法を実施することができる。
本発明の化合物を医薬として使用する場合、通常、本発明の化合物と適当な添加剤とを混和し、製剤化したものを使用する。ただし、前記は、本発明の化合物を原体のまま医薬として使用することを否定するものではない。
上記賦形剤としては、例えば乳糖、白糖、ブドウ糖、コーンスターチ、マンニトール、ソルビトール、デンプン、α化デンプン、デキストリン、結晶セルロース、軽質無水ケイ酸、ケイ酸アルミニウム、ケイ酸カルシウム、メタケイ酸アルミン酸マグネシウム、リン酸水素カルシウム等を挙げることができる。
上記結合剤としては、例えばポリビニルアルコール、メチルセルロース、エチルセルロース、アラビアゴム、トラガント、ゼラチン、シェラック、ヒドロキシプロピルメチルセルロース、ヒドロキシプロピルセルロース、カルボキシメチルセルロースナトリウム、ポリビニルピロリドン、マクロゴール等を挙げることができる。
上記滑沢剤としては、例えばステアリン酸マグネシウム、ステアリン酸カルシウム、フマル酸ステアリルナトリウム、タルク、ポリエチレングリコール、コロイドシリカ等を挙げることができる。
上記崩壊剤としては、例えば結晶セルロース、寒天、ゼラチン、炭酸カルシウム、炭酸水素ナトリウム、クエン酸カルシウム、デキストリン、ペクチン、低置換度ヒドロキシプロピルセルロース、カルボキシメチルセルロース、カルボキシメチルセルロースカルシウム、クロスカルメロースナトリウム、カルボキシメチルスターチ、カルボキシメチルスターチナトリウム等を挙げることができる。
上記着色剤としては、三二酸化鉄、黄色三二酸化鉄、カルミン、カラメル、β-カロチン、酸化チタン、タルク、リン酸リボフラビンナトリウム、黄色アルミニウムレーキ等、医薬品に添加することが許可されているものを挙げることができる。
上記矯味矯臭剤としては、ココア末、ハッカ脳、芳香散、ハッカ油、竜脳、桂皮末等を挙げることができる。
上記乳化剤または界面活性剤としては、ステアリルトリエタノールアミン、ラウリル硫酸ナトリウム、ラウリルアミノプロピオン酸、レシチン、モノステアリン酸グリセリン、ショ糖脂肪酸エステル、グリセリン脂肪酸エステル等を挙げることができる。
上記溶解補助剤としては、ポリエチレングリコール、プロピレングリコール、安息香酸ベンジル、エタノール、コレステロール、トリエタノールアミン、炭酸ナトリウム、クエン酸ナトリウム、ポリソルベート80、ニコチン酸アミド等を挙げることができる。
上記懸濁化剤としては、前記界面活性剤のほか、ポリビニルアルコール、ポリビニルピロリドン、メチルセルロース、ヒドロキシメチルセルロース、ヒドロキシエチルセルロース、ヒドロキシプロピルセルロース等の親水性高分子を挙げることができる。
上記等張化剤としては、ブドウ糖、塩化ナトリウム、マンニトール、ソルビトール等を挙げることができ、
上記緩衝剤としては、リン酸塩、酢酸塩、炭酸塩、クエン酸塩等の緩衝液を挙げることができる。
上記防腐剤としては、メチルパラベン、プロピルパラベン、クロロブタノール、ベンジルアルコール、フェネチルアルコール、デヒドロ酢酸、ソルビン酸等を挙げることができる。
上記抗酸化剤としては、亜硫酸塩、アスコルビン酸、α-トコフェロール等を挙げることができる。
上記安定化剤としては、一般に医薬に使用されるものを挙げることができる。

上記経口剤は、上記添加剤を適宜組み合わせて製剤化する。なお、必要に応じてこれらの表面をコーティングしてもよい。
上記外用剤は、上記添加剤のうち、特に賦形剤、結合剤、矯味矯臭剤、乳化剤、界面活性剤、溶解補助剤、懸濁化剤、等張化剤、防腐剤、抗酸化剤、安定化剤または吸収促進剤を適宜組み合わせて製剤化する。

混合物を充分振とう後、有機層を分取し、有機層を飽和食塩水にて洗浄し、無水硫酸マグネシウムで乾燥した。混合物をろ過し、次いでろ液中の溶媒を減圧下で留去した。残渣をシリカゲルカラムクロマトグラフィー(n-ヘプタンと酢酸エチルの混合溶媒:n-ヘプタン/酢酸エチル=3/1→2/1)にて精製した。
得られた残渣と水-エタノール(2:1,275mL)の混合物に、4℃でポタシウム ヒドラジンカルボジチオエート(Heterocycles,vol.23,No.12,1985,3099-3106)を加え、20時間撹拌した。混合物を、グラスフィルターにて吸引ろ過し、残渣を水で洗浄し、減圧下で乾燥した。得られた残渣とジクロロメタン(400mL)の混合物に、0℃でエチルマロニルクロライド(8.86mL,66.6mmol)を滴下し、室温で30分撹拌した。混合物中の溶媒を減圧下で留去した。残渣をシリカゲルカラムクロマトグラフィー(n-ヘプタンと酢酸エチルの混合溶媒:n-ヘプタン/酢酸エチル=1/1→1/3)で精製し、標記化合物(11.7g,27.6mmol)を得た。

残渣に、ジクロロメタン(5mL)、トリフルオロ酢酸(1mL)を順次加えて、室温で1時間撹拌した。混合物中の溶媒を、減圧下で留去した。室温で、残渣に、メタノール(6mL)、テトラヒドロ-4H-ピラン-4-オン(0.03mL,0.312mmol)、酢酸(1mL)を順次加え、次いで、0℃でα-ピコリンボラン(33.4mg,0.312mmol)を加え、1時間撹拌した。混合物に氷冷下で、混合物の液性がほぼ中性となるように5N水酸化ナトリウム水溶液を加えて、次いで酢酸エチルを加えた。
混合物を充分振とう後、有機層を分取し、無水硫酸マグネシウムで乾燥した。混合物をろ過し、次いでろ液中の溶媒を減圧下で留去した。残渣をシリカゲルカラムクロマトグラフィー(n-ヘプタンと酢酸エチルの混合溶媒:n-ヘプタン/酢酸エチル=2/3)にて精製し、標記化合物(61mg,0.125mmol)を得た。

混合物を充分振とう後、有機層を分取し、有機層を飽和食塩水にて洗浄し、無水硫酸マグネシウムで乾燥した。混合物をろ過し、次いでろ液中の溶媒を減圧下で留去した。
得られた残渣とエタノール(900mL)の混合物に、水浴下、攪拌しながらヒドラジン水和物(60.7mL,1.25mol)を加え、室温で13時間攪拌した。混合物をろ過し、ろ液中の溶媒を減圧下で留去した。
得られた残渣に1,4-ジオキサン(1L)、5N塩酸(200mL)を順次加え、60℃で4時間攪拌した。混合物を室温に戻し、混合物中の溶媒を減圧下で留去した。得られた残渣に水を加え、この混合物をろ過し、ろ取された残渣とろ液を得た。残渣は、更に水で洗浄後、減圧下で乾燥させ、標記化合物(42.5g,200mmol)を得た。
得られた上記ろ液に酢酸エチルを加えた。混合物を充分振とう後、有機層を分取し、有機層を飽和食塩水にて洗浄し、無水硫酸マグネシウムで乾燥した。混合物をろ過し、次いでろ液中の溶媒を減圧下で留去した。得られた残渣にジエチルエーテルを加え、析出した固体をろ取し、減圧下で乾燥させることにより、さらに、標記化合物(2.6g,12.3mmol)を得た。

混合物を充分振とう後、有機層を分取し、有機層を飽和食塩水にて洗浄し、無水硫酸マグネシウムで乾燥した。混合物をろ過し、次いでろ液中の溶媒を減圧下で留去した。残渣をシリカゲルカラムクロマトグラフィー(n-ヘプタンと酢酸エチルの混合溶媒:n-ヘプタン/酢酸エチル=9/1→1/2.3)にて精製し、標記化合物(30.7g,136mmol)を得た。

得られた残渣に1,4-ジオキサン(400mL)、濃塩酸(200mL)を順次加えて、60℃で1.5時間攪拌した。混合物中の1,4-ジオキサンを、減圧下で留去した。混合物の液性が弱酸性になるように、氷冷攪拌下で、水酸化ナトリウムを適宜加え、次いで酢酸エチルを加えた。
混合物を充分振とう後、有機層を分取し、有機層を飽和食塩水にて洗浄し、無水硫酸マグネシウムで乾燥した。混合物をろ過し、次いでろ液中の溶媒を減圧下で留去した。残渣をシリカゲルカラムクロマトグラフィー(n-ヘプタンと酢酸エチルの混合溶媒:n-ヘプタン/酢酸エチル=1/0→2.3/1)にて精製し、標記化合物(15.8g,103mmol)を得た。

混合物をろ過し、次いでろ液中の溶媒を減圧下で留去した。残渣をシリカゲルカラムクロマトグラフィー(n-ヘプタンと酢酸エチルの混合溶媒:n-ヘプタン/酢酸エチル=1/1)にて精製し、標記化合物(50.0mg,0.105mmol)を得た。

混合物をろ過し、次いでろ液中の溶媒を減圧下で留去した。残渣を(NH)シリカゲルカラムクロマトグラフィー(n-ヘプタンと酢酸エチルの混合溶媒:n-ヘプタン/酢酸エチル=4/1)にて精製した。目的物を含有するフラクションを集め、減圧下で溶媒を留去した後、得られた残渣にn-ヘプタンとジイソプロピルエーテルの混合溶媒を加えた。析出した固体をろ取し、少量のn-ヘプタンで洗浄した後、減圧下で乾燥させ、標記化合物(43.7mg,0.09mmol)を得た。

残渣にメタノール(6mL)を加え、次いでテトラヒドロフラン-3-カルボキサルデヒド(55.4μL,0.306mmol)、酢酸(1mL)、α-ピコリンボラン(32.7mg,0.306mmol)を順次加え、室温で1時間撹拌した。混合物に氷冷下、混合物の液性がほぼ中性になる程度に、5N水酸化ナトリウム水溶液を加え、次いで酢酸エチルを加えた。混合物を充分振とう後、有機層を分取し、有機層を飽和食塩水にて洗浄し、無水硫酸マグネシウムで乾燥した。混合物をろ過し、次いでろ液中の溶媒を減圧下で留去した。残渣をシリカゲルカラムクロマトグラフィー(n-ヘプタンと酢酸エチルの混合溶媒:n-ヘプタン/酢酸エチル=2/1→1/1)にて精製し、標記化合物(53.9mg,0.115mmol)を得た。

得られた残渣にジクロロメタン(0.6mL)を加え、次いでトリフルオロ酢酸(0.2mL)を加え、室温で2時間撹拌した。この混合物中の溶媒を窒素気流により留去した。得られた残渣にTHF(1mL)を加え、次いでテトラヒドロ-2H-ピラン-4-カルバアルデヒド(36mg,0.315mmol)、ナトリウム トリアセトキシボロヒドリド(66.8mg,0.315mmol)を順次加え、室温で2時間撹拌した。混合物に飽和炭酸水素ナトリウム水溶液を加え、次いで酢酸エチルを加えた。混合物を充分振とう後、有機層を分取し、溶媒を窒素気流により留去した。残渣をシリカゲルカラムクロマトグラフィー(n-ヘプタンと酢酸エチルの混合溶媒:n-ヘプタン:酢酸エチル=1/0→3/2)にて精製し、標記化合物(71.3mg,0.148mmol)を得た。

得られた残渣にジクロロメタン(2.5mL)を加え、次いでトリフルオロ酢酸(1mL)を加え、室温で1時間20分撹拌した。この混合物中の溶媒を減圧下で留去した。
得られた残渣にTHF(4mL)を加え、次いでテトラヒドロ-2H-ピラン-4-カルバルデヒド(29.4mg,0.257mmol)、ナトリウム トリアセトキシボロヒドリド(54.5mg,0.257mmol)を順次加え、室温で2時間撹拌した。混合物に氷冷下、飽和炭酸水素ナトリウムを加え、次いで酢酸エチルを加えた。混合物を充分振とう後、有機層を分取し、有機層を飽和食塩水にて洗浄し、無水硫酸マグネシウムで乾燥した。混合物をろ過し、次いでろ液中の溶媒を減圧下で留去した。残渣をシリカゲルカラムクロマトグラフィー(n-ヘプタンと酢酸エチルの混合溶媒:n-ヘプタン/酢酸エチル=9/1→3/2)にて精製し、標記化合物(75.0mg,0.16mmol)を得た。

得られた残渣にジクロロメタン(6mL)を加え、次いでトリフルオロ酢酸(1mL)を加え、室温で1時間撹拌した。この混合物中の溶媒を減圧下で留去した。
得られた残渣にTHF(10mL)を加え、次いでテトラヒドロ-4H-ピラン-4-オン(49μL,0.501mmol)、酢酸(1mL)、ナトリウム トリアセトキシボロヒドリド(106mg,0.501mmol)を順次加え、室温で14時間撹拌した。混合物に氷冷下、混合物の液性がほぼ中性になる程度に、5N水酸化ナトリウム水溶液を加え、次いで酢酸エチルを加えた。
混合物を充分振とう後、有機層を分取し、有機層を飽和食塩水にて洗浄し、無水硫酸マグネシウムで乾燥した。混合物をろ過し、次いでろ液中の溶媒を減圧下で留去した。残渣をシリカゲルカラムクロマトグラフィー(n-ヘプタンと酢酸エチルの混合溶媒:n-ヘプタン/酢酸エチル=1/1)にて精製し、標記化合物(98.0mg,0.19mmol)を得た。

N-ブチル-3-[2,6-ジメトキシ-4-(メトキシメチル)フェニル]-6-メトキシ-N-(テトラヒドロ-2H-ピラン-4-イル)ピラゾロ[5,1-b][1,3]チアゾール-7-アミン(6.0mg)とメタノール(1mL)の混合物に、臭化水素酸(8.84M、2.0μL)を加え、室温で一時間撹拌し、減圧下、溶媒を留去した。残渣にメタノール(1mL)を加え、減圧下、溶媒を留去した。得られた残渣に酢酸エチル(1mL)を加え、減圧下で溶媒を留去、乾燥し、標記化合物(6.0mg)を得た。
N-ブチル-3-[4-(エトキシメチル)-2,6-ジメトキシフェニル]-6-メトキシ-N-(テトラヒドロ-2H-ピラン-4-イル)ピラゾロ[5,1-b][1,3]チアゾール-7-アミン(1.02g)に、湯で加温したアセトン(2mL)を加え、次いで臭化水素酸(8.84M、240μL)を加え撹拌した。次いで混合物中の溶媒を窒素気流下にて一部留去し、さらに撹拌した。
この混合物を13時間減圧下室温で放置し、残渣中の溶媒を留去した。得られた残渣を細かく粉砕した後、アセトン(2mL)とヘプタン(2mL)を加え、次いで1分間超音波照射下(バス型ソニケーター)で振とうした。この混合物を遮光し、容器を密閉し1日撹拌した。
混合物中の析出物を、吸引ろ取し、次いで減圧下室温で放置し、残渣中の溶媒を留去した。この残渣にアセトン(20mL)を加え、次いで、ヘプタン(3mL)を加え数分間撹拌した。さらにヘプタン(5mL)を加え、容器を遮光し1日撹拌した後、混合物中の析出物を、吸引ろ取し、次いで減圧下室温で放置し、残渣中の溶媒を留去し、標記化合物(701.17mg)を得た。
N-ブチル-3-[4-(エトキシメチル)-2,6-ジメトキシフェニル]-6-メトキシ-N-(テトラヒドロ-2H-ピラン-4-イル)ピラゾロ[5,1-b][1,3]チアゾール-7-アミン(2.99g)に、湯で加熱したエタノール(7mL)とリン酸(14.6M、410μL)を加え、次いで、ヘプタンを1mLずつ合計30mL加えた。この混合物を遮光し1時間撹拌した。
混合物中の析出物を、吸引ろ取し、次いで減圧下室温で1週間放置し、残渣中の溶媒を留去し、さらに60℃で1.5時間、加熱乾燥し標記化合物(4.10g)を得た。
N-(2-シクロプロピルエチル)-3-[4-(エトキシメチル)-2,6-ジメトキシフェニル]-6-メトキシ-N-(テトラヒドロ-2H-ピラン-4-イル)ピラゾロ[5,1-b][1,3]チアゾール-7-アミン(757.0mg)とエタノール(10mL)と酢酸エチル(5mL)の混合物に、リン酸(14.6M、101μL)を加えた。この混合物を室温で5分間撹拌し、窒素気流下、溶媒を留去、乾燥し、標記化合物(856.0mg)を得た。
N-(2-シクロプロピルエチル)-3-[4-(エトキシメチル)-2,6-ジメトキシフェニル]-6-メトキシ-N-(テトラヒドロ-2H-ピラン-4-イル)ピラゾロ[5,1-b][1,3]チアゾール-7-アミン(16.2mg)と酢酸エチル(0.5mL)の混合物に、メタンスルホン酸(2.05μL)を加えた。この混合物を室温で一時間撹拌し、窒素気流下、溶媒を留去、乾燥し、標記化合物(19.2mg)を得た。
N-(2-シクロプロピルエチル)-3-[4-(エトキシメチル)-2,6-ジメトキシフェニル]-6-メトキシ-N-(テトラヒドロ-2H-ピラン-4-イル)ピラゾロ[5,1-b][1,3]チアゾール-7-アミン(59.6mg)とエタノール(2mL)と酢酸エチル(1mL)の混合物に、濃硫酸(6.18μL)を加えた。この混合物を室温で2時間撹拌し、窒素気流下、溶媒を留去、乾燥し、標記化合物(66.3mg)を得た。
N-(シクロブチルメチル)-3-[4-(エトキシメチル)-2,6-ジメトキシフェニル]-6-メトキシ-N-(テトラヒドロ-2H-ピラン-4-イル)ピラゾロ[5,1-b][1,3]チアゾール-7-アミン(13.4mg)と酢酸エチル(0.5mL)の混合物に、メタンスルホン酸(1.69μL)を加えた。この混合物を室温で一時間撹拌し、窒素気流下、溶媒を留去、乾燥し、標記化合物(15.9mg)を得た。
N-(シクロプロピルメチル)-3-[4-(エトキシメチル)-2,6-ジメトキシフェニル]-6-メトキシ-N-(テトラヒドロ-2H-ピラン-3-イル)ピラゾロ[5,1-b][1,3]チアゾール-7-アミン(42.0mg)とエタノール(2mL)と酢酸エチル(1mL)の混合物に、濃硫酸(4.46μL)を加えた。この混合物を室温で2時間撹拌し、窒素気流下、溶媒を留去、乾燥し、標記化合物(45.7mg)を得た。
3-[4-(エトキシメチル)-2,6-ジメトキシフェニル]-6-メトキシ-N-プロピル-N-(テトラヒドロ-2H-ピラン-3-イル)ピラゾロ[5,1-b][1,3]チアゾール-7-アミン(8.1mg)とエタノール(2mL)の混合物に、濃硫酸(0.88μL)を加えた。この混合物を室温で2時間撹拌し、窒素気流下、溶媒を留去、乾燥し、標記化合物(9.8mg)を得た。
N-(シクロブチルメチル)-3-[2,6-ジメトキシ-4-(メトキシメチル)フェニル]-6-メトキシ-N-(テトラヒドロ-2H-ピラン-4-イル)ピラゾロ[5,1-b][1,3]チアゾール-7-アミン(99.0mg)とエタノール(2mL)と酢酸エチル(1mL)の混合物に、濃硫酸(10.5μL)を加えた。この混合物を室温で2時間撹拌し、減圧下溶媒を留去、乾燥し、標記化合物(120.4mg)を得た。
N-(シクロブチルメチル)-3-[2,6-ジメトキシ-4-(メトキシメチル)フェニル]-6-メトキシ-N-(テトラヒドロ-2H-ピラン-4-イル)ピラゾロ[5,1-b][1,3]チアゾール-7-アミン(26.2mg)と酢酸エチル(1.0mL)の混合物に、メタンスルホン酸(3.40μL)を加えた。この混合物を室温で一時間撹拌し、窒素気流下、溶媒を留去、乾燥し、標記化合物(31.2mg)を得た。
3-[2,6-ジメトキシ-4-(メトキシメチル)フェニル]-6-メトキシ-N-ペンチル-N-(テトラヒドロ-2H-ピラン-4-イル)ピラゾロ[5,1-b][1,3]チアゾール-7-アミン(12.2mg)と酢酸エチル(0.5mL)の混合物に、メタンスルホン酸(1.58μL)を加えた。この混合物を室温で一時間撹拌し、窒素気流下、溶媒を留去、乾燥し、標記化合物(14.5mg)を得た。
N-(シクロプロピルメチル)-3-[2,6-ジメトキシ-4-(メトキシメチル)フェニル]-6-メトキシ-N-(テトラヒドロ-2H-ピラン-4-イル)ピラゾロ[5,1-b][1,3]チアゾール-7-アミン(8.38mg)と酢酸エチル(0.5mL)の混合物に、塩酸/ジエチルエーテル溶液(1M、17.2μL)を加えた。この混合物を室温で1.5時間撹拌し、窒素気流下、溶媒を留去、乾燥し、標記化合物(9.22mg)を得た。
3-[2,6-ジメトキシ-4-(メトキシメチル)フェニル]-6-メトキシ-N-プロピル-N-(テトラヒドロ-2H-ピラン-4-イル)ピラゾロ[5,1-b][1,3]チアゾール-7-アミン(6.98mg)と酢酸エチル(0.5mL)の混合物に、塩酸/ジエチルエーテル溶液(1M、14.7μL)を加えた。この混合物を室温で1.5時間撹拌し、窒素気流下、溶媒を留去、乾燥し、標記化合物(7.84mg)を得た。
N-(シクロプロピルメチル)-3-[2,6-ジメトキシ-4-(メトキシメチル)フェニル]-6-メトキシ-N-(テトラヒドロ-2H-ピラン-3-イル)ピラゾロ[5,1-b][1,3]チアゾール-7-アミン(30.0mg)と酢酸エチル(0.6mL)の混合物に、リン酸(14.6M、4.00μL)を加えた。この混合物を、室温で5分間撹拌し、窒素気流下、溶媒を留去、乾燥し、標記化合物(35.8mg)を得た。
3-[2,6-ジメトキシ-4-(メトキシメチル)フェニル]-6-メトキシ-N-[(2-メチルシクロプロピル)メチル]-N-(テトラヒドロ-2H-ピラン-4-イル)ピラゾロ[5,1-b][1,3]チアゾール-7-アミン(14.6mg)と酢酸エチル(0.5mL)の混合物に、メタンスルホン酸(1.90μL)を加えた。この混合物を室温で一時間撹拌し、窒素気流下、溶媒を留去、乾燥し、標記化合物(17.4mg)を得た。
3-[2-クロロ-6-メトキシ-4-(メトキシメチル)フェニル]-6-メトキシ-N-プロピル-N-(テトラヒドロ-2H-ピラン-4-イル)ピラゾロ[5,1-b][1,3]チアゾール-7-アミン(10.7mg)と酢酸エチル(0.5mL)の混合物に、メタンスルホン酸(1.45μL)を加えた。この混合物を室温で一時間撹拌し、窒素気流下、溶媒を留去、乾燥し、標記化合物(12.8mg)を得た。
4-{7-[(シクロプロピルメチル)(テトラヒドロフラン-3-イルメチル)アミノ]-6-メトキシピラゾロ[5,1-b][1,3]チアゾール-3-イル}-3,5-ジメトキシベンゾニトリル(8.0mg)とメタノール(1mL)の混合物に、臭化水素酸(8.84M、2.4μL)を加え、室温で1時間撹拌し、減圧下、溶媒を留去した。この残渣にメタノール(1mL)を加え、減圧下、溶媒を留去し、得られた残渣に酢酸エチル(1mL)を加え、減圧下で溶媒を留去、乾燥し、標記化合物(8.0mg)を得た。
4-{7-[(シクロプロピルメチル)(テトラヒドロ-2H-ピラン-4-イルメチル)アミノ]-6-メトキシピラゾロ[5,1-b][1,3]チアゾール-3-イル}-3,5-ジメトキシベンゾニトリル(26.4mg)と酢酸エチル(0.5mL)の混合物に、塩酸/ジエチルエーテル溶液(1M、54.7μL)を加えた。この混合物を室温で1時間撹拌し、窒素気流下、溶媒を留去、乾燥し、標記化合物(28.4mg)を得た。
N-(シクロプロピルメチル)-6-メトキシ-N-(テトラヒドロ-2H-ピラン-4-イルメチル)-3-(2,4,6-トリメトキシフェニル)-ピラゾロ[5,1-b][1,3]チアゾール-7-アミン(6.51mg)と酢酸エチル(0.5mL)の混合物に、塩酸/ジエチルエーテル溶液(1M、13.4μL)を加えた。この混合物を、室温で1.5時間撹拌し、窒素気流下、溶媒を留去、乾燥し、標記化合物(7.41mg)を得た。
3-{4-[(シクロブチロキシ)メチル]-2,6-ジメトキシフェニル}-6-メトキシ-N-プロピル-N-(テトラヒドロ-2H-ピラン-4-イル)ピラゾロ[5,1-b][1,3]チアゾール-7-アミン(30.0mg)、酢酸エチル(0.6mL)の混合物に、リン酸(14.6M、4.00μL)を加えた。この混合物を室温で5分間撹拌し、窒素気流下、溶媒を留去、乾燥し、標記化合物(36.1mg)を得た。
本発明化合物について、CRF1受容体(CRFR1)への結合能を評価した。試験方法とその結果は以下の如くである。
<CRFR1結合実験>
(1)CRFR1発現細胞の作製:
CRFR1結合実験の実験材料にはヒトCRFR1を高発現した細胞の膜画分を用いた。CRFR1発現細胞は以下のように作製した。cDNAライブラリーとしてhuman brain(QuickCloneTM Clontech社)を用いてCRFR1の全長遺伝子をPCR法により得た。得られたDNA断片をクローニングベクターに挿入し、塩基配列を確かめた。正しい塩基配列をもつcDNAを発現ベクター(pcDNA3.1TM,Invitrogen社)につなぎ変えた。CRFR1発現ベクターをHEK293細胞に遺伝子導入し、G418(1mg/ml)を含んだ細胞培養液中で増殖した耐性細胞を限界希釈法によりクローン化した。クローン細胞から以下に示す結合実験により、単位蛋白量あたりの膜画分とsauvagineとの結合能が高いクローンを最終的に選択して実験に用いた。
(1)で得られたクローン細胞を集め、氷冷したmembrane buffer(50mM Tris-HCl,5mM MgCl2,2mM EGTA,1mM DTT,protease inhibitor cocktail(COMPLETETM,Roche Diagnostics社),pH7.3)で懸濁し、氷冷下、Polytron(KINEMATICA)にて細胞を破砕(level 5,10秒間,2-5回,氷冷下)した後、遠心分離(2,000rpm、5分間、4℃)をし、上清を回収した。沈殿にmembrane bufferを加え、Polytron処理(同上述条件)し、遠心分離(同上述条件)にて得られた上清を先の上清と併せた。これを、遠心分離(13,000rpm(18,000xg),30分間,4℃)して細胞膜を得た。沈殿した細胞膜をmembrane bufferで懸濁し、Polytronにより細胞膜を破砕(level 5,10秒,3-5回、氷冷下)し、分散懸濁液とした。タンパク質定量を行った。
以下の(1)または(2)の方法により、細胞膜画分として使用した。
(1)上記分散懸濁液のタンパク質濃度が200μg/mlとなるように0.1%BSAを含むmembrane bufferにて希釈し細胞膜画分として使用した。
(2)上記分散懸濁液を凍結保存し、必要に応じて解凍後、再度分散し、希釈して細胞膜分とした。
CRFとの結合競合実験は96穴プレートを用いて、SPA(GE Healthcare社)法により行った。細胞膜画分蛋白5μg,SPA beads 1mgと100pMの125I-CRF(Perkin Elmer社)を被検化合物存在下で2時間以上、室温で放置し、遠心(1,200rpm(260×g),5分間,室温)後に各穴の放射活性をTopCount(登録商標;Perkin Elmer社)にて測定した。
4,000倍過剰量の非放射sauvagineを加えた場合の放射活性を非特異的な結合として各々の値から差し引き、被検化合物を加えてない放射活性を100%(control)とし、各値を%(% of control)で表した。被検化合物の濃度を横軸に、%(% of control)を縦軸にした結合阻害曲線よりIC50値を算出した。
下表に示すように本発明にかかる化合物は、CRFR1に対し優れた結合能を示した。
<マウス明暗箱試験による抗不安作用の評価>
(1)試験操作:
マウス明暗箱試験はBelzung C.,Misslin R.,and Vogel E.(参考文献;Behavioural effects of the benzodiazepine receptor partial agonist RO16-6028 in mice,Psychopharmacology,97,388-391,1989)らの方法を改変して行った。この試験に用いる試験装置は、フタ付き黒色のアクリル箱(暗箱;15x10x20cm)と上面開放の白色のアクリル箱(明箱;15×20×20cm)を黒色のアクリルのトンネル(10×7×4.5cm)で連結し、マウスが暗箱と明箱を自由に往来することが可能な明暗箱を用いた。但し、この試験装置において、行動観察のために明箱の前面(20×20cm)および背面(20×20cm)には透明なアクリルを用いた。明箱床面の照度が150Luxになるように照明を設定した後、5週齢の雄性Balb/cマウス(日本チャールスリバーより購入)を暗箱に導入して試験を開始した。この試験において、被検化合物は5%ジメチルスルホキシド、5% Cremopor EL、90%生理食塩水に懸濁し、試験開始の1時間前に試験動物へ経口投与した。
試験開始から5分間のマウスの行動を観察した。マウスの四肢が明箱の床上にある状態を明所滞在として明所滞在時間を測定し、抗不安作用の指標とした。明所滞在時間をvehicle投与群より有意に延長させた最小の投与量を求め、最小有効量(MED)とした。なお、vehicle投与群と被験化合物投与群間の統計学的有意性については、同一試験に複数の用量を設定した場合は一元配置分散分析後、Dunnett型多重比較によって解析し、1用量のみを設定した場合はMann-WhitneyのU検定によって解析した。
実施例化合物1、2、3、4、5、9、10、13および14は、マウス明暗箱試験による抗不安作用の評価に対し優れた効果を示し、30mg/kg(経口投与)で統計学的に有意な抗不安作用を示した。
Claims (4)
- R2が、テトラヒドロピラン-4-イル基、テトラヒドロピラン-3-イル基、(テトラヒドロピラン-4-イル)メチル基または(テトラヒドロフラン-3-イル)メチル基である請求項1記載の化合物もしくはその塩。
- R1がn-プロピル基、n-ブチル基、n-ペンチル基、シクロプロピルメチル基、シクロブチルメチル基、2-(シクロプロピル)エチル基または(2-メチルシクロプロピル)メチル基である請求項1または2記載の化合物もしくはその塩。
- 請求項1記載の化合物もしくはその塩を含有してなる、うつ病、抑うつ症状、不安症、過敏性腸症候群、睡眠障害、不眠症、アルコール依存症、アルコール禁断症状、薬物依存症、薬物禁断症状、ストレスに伴う胃腸機能異常、神経性食思不振症、摂食障害、術後イレウス、虚血性神経障害、脳卒中、興奮毒性神経障害、けいれん、てんかん、高血圧、統合失調症、双極性障害もしくは認知症の治療剤または予防剤。
