Abstract
本發明之課題係提供在含有S-氟白普洛芬(S-flurbiprofen)及作為經皮吸收促進劑之薄荷油之外用劑組成物中,抑制S-氟白普洛芬含量之經時性減少,S-氟白普洛芬的保存安定性優異的外用劑組成物。
本發明之解決手段為在含有S-氟白普洛芬及作為經皮吸收促進劑之薄荷油之外用劑組成物中,摻合選自二個中鏈脂肪酸鍵結至丙二醇之酯及三個中鏈脂肪酸鍵結至甘油之酯所組成群組之1種以上。
Description
本發明係關於含有S-氟白普洛芬及薄荷油之外用劑組成物,其可應用於醫藥品之領域。
一般而言,氟白普洛芬係具有消炎鎮痛作用,可使用為外用劑組成物之有效成分。
氟白普洛芬係消旋體2-(2-氟-4-聯苯基)丙酸,但僅將活性本體之S體進行光學分割而得之S-氟白普洛芬,相較於消旋體而言,消炎鎮痛作用係格外地優異(專利文獻1)。
作為含有S-氟白普洛芬之外用劑組成物,在專利文獻1中,已提案含有作為經皮吸收促進劑之薄荷油及賽弗索(Sefsol)之外用劑組成物。然而,此外用劑組成物係由於S-氟白普洛芬之化學性反應,在保存時可認出有效成分之經時性減少,而有進一步改善之餘地。
此外,已知一種牙膏組成物,其係填充於在最內層具有合成樹脂層之容器中之牙膏組成物,且含有包含氟白普洛芬在內之芳基丙酸系非類固醇抗炎症劑(藥效
劑)(專利文獻2)。此處,該藥效劑對合成樹脂係親和性極高,為了防止在保管中吸附或穿透至容器內面而使該藥效劑濃度經時性減少,係含有一定量的非離子界面活性劑及一定量的丙二醇脂肪酸酯等。專利文獻2係關於牙膏組成物,其並未揭示或暗示含有作為經皮吸收促進劑之薄荷油之外用劑組成物。

本發明之目的係提供在含有S-氟白普洛芬、及作為經皮吸收促進劑之薄荷油之外用劑組成物中,抑制S-氟白普洛芬含量之經時性減少,S-氟白普洛芬的保存安定性優異的外用劑組成物。本發明之其他目的係在含有S-氟白普洛芬及作為經皮吸收促進劑之薄荷油之非水系外用劑組成物中,抑制經時性S-氟白普洛芬含量之減少。
本發明者等人進行深入研究之結果,發現在含有S-氟白普洛芬及作為經皮吸收促進劑之薄荷油之外用劑組成物中,僅限於摻合某種特定的多元醇之中鏈脂肪酸
酯,即,選自二個中鏈脂肪酸鍵結至丙二醇之酯及三個中鏈脂肪酸鍵結至甘油之酯所組成群組之1種以上之情況,會抑制S-氟白普洛芬含量之經時性減少,S-氟白普洛芬即便在長期保存中仍安定,遂完成本發明。
即,本發明係提供:(1)一種外用劑組成物,其係包含S-氟白普洛芬或其藥學上可容許之鹽、薄荷油、以及選自二個中鏈脂肪酸鍵結至丙二醇之酯及三個中鏈脂肪酸鍵結至甘油之酯所組成群組之1種以上;(2)如(1)所記載之外用劑組成物,其中,二個中鏈脂肪酸鍵結至丙二醇之酯為二辛酸丙二醇酯;(3)如(1)所記載之外用劑組成物,其中,三個中鏈脂肪酸鍵結至甘油之酯為三癸酸甘油酯;(4)如(1)~(3)中任一項所記載之外用劑組成物,其中,選自二個中鏈脂肪酸鍵結至丙二醇之酯及三個中鏈脂肪酸鍵結至甘油之酯所組成群組之1種以上的摻合量係相對於S-氟白普洛芬或其藥學上可容許之鹽的摻合量而言以重量計為0.1倍量~10倍量;(5)如(1)~(3)中任一項所記載之外用劑組成物,其中,選自二個中鏈脂肪酸鍵結至丙二醇之酯及三個中鏈脂肪酸鍵結至甘油之酯所組成群組之1種以上的摻合量係相對於S-氟白普洛芬或其藥學上可容許之鹽的摻合量而言以重量計為0.45倍量~10倍量;(6)如(1)~(5)中任一項所記載之外用劑組成
物,其係非水系組成物;(7)如(6)所記載之外用劑組成物,其係貼布劑。
藉由本發明,能夠提供在含有S-氟白普洛芬及作為經皮吸收促進劑之薄荷油之外用劑組成物中,抑制經時性S-氟白普洛芬含量之減少,S-氟白普洛芬即便在長期保存中仍安定性優異的外用劑組成物。特定而言,能夠在含有S-氟白普洛芬及作為經皮吸收促進劑之薄荷油之非水系外用劑組成物中,抑制經時性S-氟白普洛芬含量之減少。
圖1係表示將實施例1~2及比較例1~6之試料於50℃之條件下保存時之S-氟白普洛芬殘存率的推移之圖。
圖2係表示將實施例1~2及比較例1~6之試料於65℃之條件下保存時之S-氟白普洛芬殘存率的推移之圖。
圖3係表示將實施例1、3~6之試料於50℃之條件下保存時之S-氟白普洛芬殘存率的推移之圖。
圖4係表示將實施例1、3~6之試料於65℃之條件下保存時之S-氟白普洛芬殘存率的推移之圖。
以下,針對本發明之態樣進行詳述。
本發明係提供一種外用劑組成物,其係包含S-氟白普洛芬或其藥學上可容許之鹽、薄荷油、以及選自二個中鏈脂肪酸鍵結至丙二醇之酯及三個中鏈脂肪酸鍵結至甘油之酯所組成群組之1種以上。此處所謂的中鏈脂肪酸係指碳數8~12之飽和脂肪酸,較佳為辛酸、癸酸等。
作為本發明中所使用之二個中鏈脂肪酸鍵結至丙二醇之酯,可列舉二辛酸丙二醇酯、二(癸醯基/癸酸)丙二醇酯、二癸酸丙二醇酯、二月桂酸丙二醇酯等。該等之中,較適宜為二辛酸丙二醇酯。
作為本發明中所使用之三個中鏈脂肪酸鍵結至甘油之酯,可列舉三辛酸甘油酯、三異辛酸甘油酯、三(癸醯基/癸酸)甘油酯、三癸酸甘油酯、三月桂酸甘油酯等。該等之中,較適宜為三癸酸甘油酯。
作為本發明中所使用之二個中鏈脂肪酸鍵結至丙二醇之酯及三個中鏈脂肪酸鍵結至甘油之酯的摻合量,係相對於S-氟白普洛芬或其藥學上可容許之鹽的摻合量而言以重量計,較佳為0.1倍量~10倍量,更佳為0.45倍量~10倍量,再佳為0.45倍量~3.5倍量。在未滿0.1倍量時,會損及S-氟白普洛芬之經時性減少抑制效果,若超過10倍量,則會損及作為製品之使用性。
本發明中所使用之「S-氟白普洛芬或其藥學上可容許之鹽」可藉由日本專利特公昭62-6536號公報所記載之方法取得。
「S-氟白普洛芬或其藥學上可容許之鹽」較佳係使用
S-氟白普洛芬。
作為「藥學上可容許之鹽」,較佳係可列舉鈉鹽、鉀鹽。
S-氟白普洛芬或其藥學上可容許之鹽的含量係相對於外用劑組成物整體而言通常為0.1~10重量%,較佳為0.5~5重量%。
本發明中所使用之薄荷油係指將薄荷Mentha arvensis Linne var.piperascens Milinvaud(Labiatae)之地上部進行水蒸氣蒸餾並將所獲得之油冷卻,且去除固形份而得之精油,就薄荷醇(C10H20O)而言係包含30.0%以上。主成分為1-薄荷醇、薄荷酮、蒎烯、莰烯、薄荷烯酮、檸檬烯、倍半萜烯等。
薄荷油的含量係相對於外用劑組成物整體而言通常為0.1~10重量%,較佳為0.5~5重量%。
本發明中所使用之「非水系」係指未加水之系統,但可容許隨附進入之水分。
本發明中所使用之「保存安定性」係指使外用劑組成物歷經長期保存時S-氟白普洛芬呈化學上安定之性質。
本發明之外用劑組成物能夠以貼布劑(亦稱為硬膏劑)、糊劑、軟膏劑、乳霜劑、洗劑等各種外用製劑之形式提供。較佳的劑型為非水系外用製劑,例如貼布劑、軟膏劑,特佳為貼布劑。此等製劑能夠適宜使用適於各劑型之添加劑或基劑,並依照日本藥局方等所記載之通常的方法予以製造。
貼布劑及糊劑等貼附劑可藉由調製含有S-氟白普洛芬、薄荷油、以及選自二個中鏈脂肪酸鍵結至丙二醇之酯及三個中鏈脂肪酸鍵結至甘油之酯所組成群組之1種以上之膏體,並依序疊合支持體層、膏體層(亦稱為黏著劑層)及剝離襯墊層而獲得。另外,貼布劑之膏體基劑係於下述詳述,但視需要亦可摻合黏著賦予劑、軟化劑、溶解劑等。此外,作為糊劑之膏體基劑,能夠使用聚丙烯酸鈉或羧甲基纖維素鈉(以下,簡稱為CMC-Na),視需要亦可摻合黏著賦予劑、增黏劑、交聯劑等。
作為貼布劑之膏體基劑,可列舉苯乙烯/異戊二烯/苯乙烯嵌段共聚物(SIS)、苯乙烯/丁二烯/苯乙烯嵌段共聚物(SBS)、苯乙烯/乙烯/丁烯/苯乙烯嵌段共聚物(SEBS)或苯乙烯/乙烯/丙烯/苯乙烯嵌段共聚物(SEPS)等,較佳為苯乙烯/異戊二烯/苯乙烯嵌段共聚物(SIS)。該SIS係能夠由市面上取得例如Clayton D1161JP(Clayton公司製)或Cariflex TR-1107、TR-1117(Shell化學製)等。膏體基劑於膏體層中之含量係較佳為1~50重量%,更佳為5~40重量%。
作為黏著賦予劑,可列舉松香酯樹脂、多萜烯樹脂、萜烯酚樹脂、石油樹脂等,但該等之中,較佳為松香酯樹脂,特佳為經去除低沸點餾份,並進一步進行氫化而得之松香酯樹脂(例如Ester Gum HG,荒川化學製)。另外,作為多萜烯樹脂,有安原油脂製之YS-Resin,作為萜烯酚樹脂,有安原樹脂製之YS Polystar,作為石油樹
脂,可使用日本Zeon製之Quintone、荒川化學製之Arkon、Exxon製之Escorez等。黏著賦予劑於膏體層中之含量係較佳為5~50重量%,特佳為5~30重量%。
作為軟化劑,可列舉流動石蠟等。
在膏體基劑中,可進一步任意摻合抗氧化劑、橡膠老化防止劑等。作為橡膠老化防止劑,可摻合例如p,p’-二胺基二苯基甲烷、醛醇-α-萘基胺、苯基-α-萘基胺、苯基-β-萘基胺、N,N’-二苯基-對伸苯基胺、N,N’-二-β-萘基-對伸苯基二胺、1,2-二氫-2,2,4-三甲基喹啉、2,6-二第三丁基-對甲酚、2,5-二第三丁基-氫醌、氫醌-單苄基醚、4,4’-二羥基-二苯基環己酮等1種以上。
另外,專利文獻1段落0011所記載之「公知的老化防止劑」亦相當於抗氧化劑。
貼附劑可藉由將前述膏體層塗展於柔軟的支持體層上而予以製造。此處,作為支持體,只要是膏體層不會浸透至裡面之柔軟的片狀物,則無論種類皆可使用。若具體例示能夠使用之片狀物,則可列舉織布、不織布等。支持體的厚度較適當係通常設為約1000μm以下,較佳為30~700μm之範圍。
前述剝離襯墊只要是能夠在直至貼附劑使用前被覆前述膏體層,並在使用時予以剝離而去除者即可,可列舉例如將聚對苯二甲酸乙二酯、聚萘二甲酸乙二酯等聚酯;聚乙烯、聚丙烯等聚烯烴;聚氯乙烯;聚偏氯乙烯:紙等纖維素或其衍生物;尼龍、鋁等作成薄膜狀者。此外,作為
此種剝離襯墊,從可自前述膏體層容易地進行剝離之觀點而言,較佳係使用藉由聚矽氧或氟樹脂(Teflon(註冊商標)等)等剝離劑施行表面塗覆(剝離處理)者。
軟膏劑可藉由使S-氟白普洛芬、薄荷油、以及選自二個中鏈脂肪酸鍵結至丙二醇之酯及三個中鏈脂肪酸鍵結至甘油之酯所組成群組之1種以上溶解或分散於白色凡士林、聚乙二醇等基劑,並緩冷至室溫而獲得。
乳霜劑係在溶解有油脂成分之油相中添加S-氟白普洛芬、薄荷油、選自二個中鏈脂肪酸鍵結至丙二醇之酯及三個中鏈脂肪酸鍵結至甘油之酯所組成群組之1種以上以及界面活性劑,並進行脫氣/加溫。對其添加經加溫之水相,進行高速攪拌(乳化)後,緩冷至室溫,藉此便可調製乳霜劑。
洗劑可藉由使S-氟白普洛芬、薄荷油、以及選自二個中鏈脂肪酸鍵結至丙二醇之酯及三個中鏈脂肪酸鍵結至甘油之酯所組成群組之1種以上溶解或分散於水、乙醇、多元醇或此等之混液中而獲得。
在此等外用劑中,除了前述成分以外,在不會損及本發明之效果之量的、質的範圍中,亦可摻合多元醇、油分、陰離子性界面活性劑、陽離子性界面活性劑、兩性界面活性劑、陽離子性高分子、陰離子性高分子、非離子性高分子、兩性高分子等高分子、多醣類、螯合劑、氧化防止劑、紫外線吸收劑、防腐劑、維生素等藥劑、香料等。
作為多元醇,可列舉丙二醇、二丙二醇、1,3-丁二醇、甘油、聚乙二醇類等。
作為非離子界面活性劑,可列舉山梨醇酐單油酸酯等山梨醇酐脂肪酸酯類、單硬脂酸甘油酯等甘油脂肪酸酯類、單辛酸丙二醇酯等丙二醇脂肪酸酯類、硬化蓖麻油衍生物、甘油烷基醚等親油性非離子界面活性劑、聚氧乙烯(以下,簡稱為POE)山梨糖醇單月桂酸酯等POE山梨糖醇脂肪酸酯類、POE甘油單硬脂酸酯等POE甘油脂肪酸酯類、POE單油酸酯等POE脂肪酸酯類、POE硬脂醯醚等POE烷基苯基醚類、普盧蘭尼克(Pluronic)等普盧蘭尼克型類、POE/POP鯨蠟醚等POE/POP烷基醚類、特洛尼克(Tetronic)等四POE/四POP乙烯二胺縮合物類、POE硬化蓖麻油、POE硬化蓖麻油單異硬脂酸酯等POE硬化蓖麻油衍生物、蔗糖脂肪酸酯等。
作為陰離子界面活性劑,可列舉皂用基質、月桂酸鈉等脂肪酸皂、月桂基硫酸鈉等高級烷基硫酸酯鹽、POE月桂基硫酸鈉等烷基醚硫酸酯鹽、POE油醚磷酸鈉等磷酸酯鹽、POE烷基烯丙醚羧酸鹽、高級脂肪酸酯磺酸鹽等。
作為兩性界面活性劑,可列舉2-椰油醯基-2-咪唑啉鎓氫氧化物-1-羧基乙氧基2鈉鹽等咪唑啉系兩性界面活性劑、磺基甜菜鹼等甜菜鹼系界面活性劑、N-月桂基β-丙胺酸等胺基酸鹽等。
作為陽離子性高分子,可列舉係聚(二甲基
二烯丙基銨鹵化物)型陽離子性聚合物等,且以稱為Merquat 100之商品名由美國Merck公司(Merck & Co.Inc)所販售者等。
作為陰離子性高分子,可列舉黃原膠、鹿角菜膠、藻酸鈉、***膠、果膠、羧基乙烯基聚合物等。
作為非離子性高分子,可列舉聚乙烯吡咯啶酮及乙烯吡咯啶酮與醋酸乙烯酯之共聚物、乙烯吡咯啶酮、醋酸乙烯酯、丙烯醯胺基丙烯酸酯之共聚物、羥乙基纖維素、羥丙基纖維素、羥丙基甲基纖維素、甲基纖維素、糊精、半乳聚糖、普魯蘭多醣(pullulan)等。
作為兩性高分子,可列舉係將丙烯酸二烷基胺乙酯、甲基丙烯酸二烷基胺基乙酯、二丙酮丙烯醯胺等與丙烯酸、丙烯酸烷基酯、甲基丙烯酸、甲基丙烯酸烷基酯等進行共聚,並經鹵化醋酸等進行兩性化而得者等且以稱為Yukaformer AM-75之商品名由三菱油化所販售者等。
作為多醣類,可列舉黏多醣,例如玻尿酸、硫酸軟骨素及該等之鹽等。
作為紫外線吸收劑,可列舉對胺基安息香酸等安息香酸系、甲氧基桂皮酸辛酯等桂皮酸系、2,4-二羥基二苯甲酮等二苯甲酮系等之紫外線吸收劑。
作為防腐劑,可列舉二丁基羥基甲苯、對羥基安息香酸烷基酯、安息香酸鈉、山梨酸鉀等泛用於醫藥品、醫藥部外品、化妝品之防腐劑。
作為維生素等藥劑,可列舉維生素C類、維生素E類等維生素類、胎盤素、各種胺基酸、生藥、消炎劑等泛用於醫藥品、醫藥部外品、化妝品之藥劑。
作為本發明之製劑例,係將貼布劑、糊劑、軟膏劑及洗劑之配方例示於表A。
貼布劑可藉由例如表B所示之原材料所構成。通常,可藉由將有效成分及添加劑視需要在加溫條件下進行練合,調製膏體,並將此膏體進行延展、裁斷,使其附著於支持體,以薄膜等襯墊材進行保護,並將此製劑填充於包裝袋中而予以製造。
以下,藉由實施例進一步詳細說明本發明。本發明並不受此實施例所限定。
依照下述表1之組成,將S-氟白普洛芬、薄荷油及多元醇中鏈脂肪酸酯量取至玻璃瓶中,將玻璃瓶使用加熱至65℃之水浴一面進行加熱一面進行溶解而調製各試料。
將前述試料分注於玻璃製的試驗管中,在設定成50℃或65℃作為嚴苛條件之恆溫機中進行保存。在保存開始前及保存開始後經過1、2、3個月時採取試料,測定試料中之S-氟白普洛芬殘存率(%)並評估安定性。
將各試料之摻合組成及將各試料於前述條件下進行保存後之S-氟白普洛芬殘存率(%)的推移示於表1。此處,對將各種多元醇中鏈脂肪酸酯進行一定量混合時之S-氟白普洛芬殘存率(%)的推移進行比較。此外,S-氟白普洛芬殘存率(%)的推移亦示於圖1及2。
實施例1及2之試料係含有二個中鏈脂肪酸鍵結至丙二醇之二辛酸丙二醇酯或三個中鏈脂肪酸鍵結至甘油之三癸酸甘油酯作為各多元醇中鏈脂肪酸酯。與含有僅一個中鏈脂肪酸鍵結至丙二醇或甘油之多元醇中鏈脂肪酸酯之比較例1~6之試料進行比較,實施例1及2之試料係即便在3個月保存後仍顯著地維持較高的S-氟白普洛芬殘存率,S-氟白普洛芬殘存率(%)之減少較緩慢。
針對在前述檢討中S-氟白普洛芬殘存率(%)之減少較緩慢之二辛酸丙二醇酯,針對使其摻合量變動成相對於S-氟白普洛芬而言以重量計為0.45~3.50倍量之情況,進一步進行檢討。將各試料之摻合組成及將各試料於前述條件下進行保存後之S-氟白普洛芬殘存率(%)的推移示於表2。此外,S-氟白普洛芬殘存率(%)的推移亦示於圖3及4。
在使二辛酸丙二醇酯相對於S-氟白普洛芬之摻合比例改變之實施例1及3~6之試料中,於所有摻合比例中即便在3個月保存後與比較例之試料進行比較,仍顯著地維持較高的S-氟白普洛芬殘存率。再者,上述摻合比例越大,則越抑制S-氟白普洛芬殘存率(%)之減少。
藉由本發明,能夠藉由在含有S-氟白普洛芬及作為經皮吸收促進劑之薄荷油之外用劑組成物中,摻合選自二個中鏈脂肪酸鍵結至丙二醇之酯及三個中鏈脂肪酸鍵結至甘油之酯所組成群組之1種以上,而提供抑制S-氟白普洛芬含量之經時性減少,S-氟白普洛芬的保存安定性優異的外用劑組成物。特定而言,能夠在含有S-氟白普洛芬及作為經皮吸收促進劑之薄荷油之非水系外用劑組成物中,抑制經時性S-氟白普洛芬含量之減少。

- 一種外用劑組成物,其係包含S-氟白普洛芬(S-flurbiprofen)或其藥學上可容許之鹽、薄荷油、以及選自二個中鏈脂肪酸鍵結至丙二醇之酯及三個中鏈脂肪酸鍵結至甘油之酯所組成群組之1種以上。
- 如請求項1之外用劑組成物,其中,二個中鏈脂肪酸鍵結至丙二醇之酯為二辛酸丙二醇酯。
- 如請求項1之外用劑組成物,其中,三個中鏈脂肪酸鍵結至甘油之酯為三癸酸甘油酯。
- 如請求項1至3中任一項之外用劑組成物,其中,選自二個中鏈脂肪酸鍵結至丙二醇之酯及三個中鏈脂肪酸鍵結至甘油之酯所組成群組之1種以上的摻合量係相對於S-氟白普洛芬或其藥學上可容許之鹽的摻合量而言以重量計為0.1倍量~10倍量。
