Foreliggende oppfinnelse er i feltet for medisinsk kjemi og vedrører pyrazolforbindelser som er proteinkinaseinhibitorer, spesielt inhibitorer av ERK, sammensetninger inneholdende slike forbindelser og deres anvendelse. Oppfinnelsens bakgrunn
Pattedyrs mitogen-aktiverte protein (MAP)1-kinaser er se-rin/treoninkinaser som medierer intracellulære signal transduksjonsveier (Cobb og Goldsmith, 1995, J Biol. Chem., 279, 14843; Davis, 1995, Mol. Reprod. Dev. 42, 459). Med-lemmer av MAP-kinasefamilien deler sekvensiell likhet og bevarte strukturelle domener, og inkluderer ERK (ekstracel-lulær signalregulert kinase), JNK (Jun N-terminal kinase)
og p38-kinasene. Aurora2 er en serin/treoninproteinkinase som har blitt implisert i humancancer, slik som kolon, bryst og andre faste tumorer. Denne kinase er antatt å være involvert i protein-fosforyleringstilfeller som regulerer cellecyklusen. Spesielt kan aurora2 spille en rolle i kontrollen av den presise segregasjon av kromosomer under mitose. Misregulering av
cellecyklusen kan føre til cellulær proliferasjon og andre

Glykogensyntasekinase-3 (GSK-3) er en serin/treoninprotein-kinase omfattende a- og p-isoformer som hver er kodet av distinkte gener [Coghlan et al., Chemistry & Biology, 7, 793-803 (2000); Kim og Kimmel, Curr. Opinion Genetics Dve., 10, 508-514 (2000)]. GSK-3 har blitt implisert i forskjellige sykdommer inklusive diabetes, Alzheimers sykdom, CNS-forstyrrelser slik som manisk depressive forstyrrelser og neurodegenerative sykdommer, og kardiomyocete hypertrofi [WO 99/65897; WO 00/38675; og Haq et al., J. (2000) 151, 117]. Disse sykdommer kan være forårsaket av, eller resulterer i, den abnormale virkning av visse celle-signal-veier hvor GSK-3 spiller en rolle.
KDR er en tyrosinkinasereseptor som også binder VEGF (vaskulær endotelial vekstfaktor) (Neufeld et al., 1999, FASEB J., 13, 9). Bindingen av VEGF til KDR-reseptoren fø-rer til angiogenese, som er fremveksten av kapillærer fra pre-eksisterende blodkar. Høye nivåer av VEGF finnes i forskjellige cancerer som forårsaker tumorangiogenese og til-later hurtig vekst av cancerceller. Derfor er suppresjon av VEGF-aktivitet en måte å hemme tumorvekst på, og det har blitt vist at dette kan oppnås ved å hemme KDR-reseptor-tyrosinkinase.
AKT, også kjent som proteinkinase B, er en serin/treonin-kinase som spiller en sentral rolle i fremming av overle-velsen av et bredt område av celletyper [Khwaja, A., Na-ture, s. 33-34 (1990)]. Det har blitt vist av Zang, et al, at humane ovarialcancerceller viser hevde nivåer av AKT-1 og AKT-2. Hemming av AKT induserer apoptose av disse humane ovarialcancerceller som demonstrerer at AKT kan være viktig mål for ovarialcancerbehandling [Zhang, Q. Y., et al, Oncogene, 19 (2000)] og andre proliferative forstyrrelser. AKT-veien har også blitt implisert i motoneuronal overlevelese og nerveregenerasjon [Kazuhiko, N., et al, The Journal of Neuroscience, 20 (2000)].

Beskrivelse av oppfinnelsen
Det har nå blitt funnet at forbindelser av denne oppfinnelse og sammensetninger derav er effektive som proteinkinaseinhibitorer, spesielt som ERK-inhibitorer. Disse forbindelser har den generelle formel I:

Som anvendt her, skal de følgende definisjoner gjelde med mindre annet er indikert. Likeledes er kombinasjoner av substituenter eller variable tillatt bare hvis slike kombinasjoner resulterer i stabile forbindelser.
Begrepet "alkyl" og "alkoksy" anvendt alene eller som del av en større enhet refererer til både rettede og forgrenede kjeder inneholdende ett til tolv karbonatomer. Begrepet "halogen" betyr F, Cl, Br eller I. Begrepet "heteroatom" betyr N, 0 eller S og skal inkludere enhver oksidert form av nitrogen og svovel, og den kvaterniserte form av ethvert basisk nitrogen.
Begrepet "aryl", som anvendt alene eller som del av en større enhet som i "aralkyl", refererer til aromatiske ringgrupper med seks til fjorten ledd, slik som fenyl, ben-zyl, 1-naftyl, 2-naftyl, 1-antracyl og 2-antracyl.
Det vil bli verdsatt av fagmannen at visse forbindelser av oppfinnelsen kan eksistere i tautomere former, alle slike tautomere former av forbindelsene er innenfor rammet av oppfinnelsen.
Med mindre annet er fastlagt, er strukturene beskrevet heri også ment å inkludere alle stereokjemiske former av struk-turen; dvs., R- og S-konfigurasjonene for hvert asymmetrisk senter. Derfor er enkle stereokjemiske isomerer samt enan-tiomere og diastereomere blandinger av de foreliggende forbindelser innenfor rammet av oppfinnelsen. Med mindre annet er fastlagt, er strukturer beskrevet heri også ment å inkludere forbindelser som bare er forskjellige i nærværet av ett eller flere isotopisk anrikede atomer. Når R<4> er R<5>, inkluderer eksempler på foretrukne R<5->grupper pyrrolidin-l-yl, morfolin-l-yl, piperidin-l-yl og pipera-zin-l-yl hvori hver gruppe eventuelt er substituert. Når R<4 >er -NHR<5> eller -N(R<5>)2, inkluderer foretrukne R<5->grupper ytterligere (CH2)yR<6>, (CH2)yR<7> og (CH2) yCH (R<6>) (R7) . Skjema I over viser en generell synteserute som ble anvendt for å fremstille forbindelsene av denne oppfinnelse når R<2 >er en eventuelt substituert fenylgruppe. I trinn (a), ble en eventuelt substituert benzoylklorid kombinert med forbindelse 1 i diklormetan og aluminiumtriklorid for å danne forbindelse 2. Dannelsen av amid A ble oppnådd ved å behandle forbindelse 2 med et amin 3^ i DMF. Når amin 3_ var et primært amin, fo-regikk reaksjonen ved omgivelsestemperatur. Når amin 3^ var et sekundært amin, ble reaksjonen varmet ved 50 °C for å oppnå fullstendig reaksjon og gi amid 4^
Dannelsen av enamin 5 i trinn (c) ble oppnådd ved å behandle amid _4 med (Me2N) 2-0t-Bu ved omgivelsestemperatur. Alternativt så ble også reaksjonen for å danne enamin 5_ i trinn (c) oppnådd ved å anvende dimetylformaid-dimetylace-tal (DMF-DMA). Reaksjonen som anvender DMF-DMA krever hevet temperatur for å gi enamin 5_ mens derimot reaksjonen ved å anvende (Me2N)2-OtBu har fordelen av å foregå ved omgivelsestemperatur for å gi enaminet 5_ i høyere renhet.
Dannelsen av pyrazolforbindelsen 6 i trinn (d) ble oppnådd ved behandlingen av enamin 5_ med hydrazinhydrat ved hevet temperatur. Forbindelsene med formel II syntetisert ved denne fremgangsmåten, som eksemplifisert i tabell 1, ble isolert ved preparativ HPLC (omvendt fase, 10—>90% MeCN i vann over 15 minutter). De foreliggende forbindelser er spesielt nyttige for å behandle en sykdomstilstand hvilken lindres ved anvendelsen av en inhibitor av ERK, JAK, JNK, Aurora, GSK, KDR, eller AKT. Som anvendt her, med mindre annet er indikert, refererer begrepene "ERK", "JAK", "JNK", "Aurora", "GSK", "KDR", og "AKT" til alle isoformer av de respektive enzymer inklusive, men ikke begrenset til ERK1, ERK2, ERK3, ERK4, ERK5, ERK6, ERK7, JAK1, JAK2, JAK3, JAK4, JNK1, JNK2, JNK2, Auro-ral, Aurora2, GSK3-alfa, GSK3-beta, KDR, AKT-1, AKT-2, og AKT-3.
Aktiviteten til forbindelsene som proteinkinaseinhibitorer, for eksempel som ERK-inhibitorer, kan undersøkes in vitro, in vivo eller i en cellelinje. Ved å anvende ERK som et eksempel, inkluderer in vitro-undersøkelser som bestemmer hemming av enten kinaseaktiviteten eller ATPase-aktiviteten til aktivert ERK. Alternative in vitro-undersøkelser kvan-tiviserer inhibitorens evne til å binde til ERK og kan må-les enten ved radiomerking av inhibitoren før binding, ved å isolere inhibitoren/ERK-komplekset og bestemme mengden radiomerke bundet, eller ved å kjøre et konkur-ranseseksperiment hvor nye inhibitorer inkuberes med ERK bundet til kjente radioligander. Forbindelsene av denne oppfinnelse er potente inhibitorer av ERK som bestemt ved enzymatisk undersøkelse. Disse forbindelser har også blitt vist å hemme ERK i en celleproliferasjonstest. Betingelsesdetaljene anvendt for både de enzymatiske og celleproliferative tester er angitt i eksemplene under.
Forbindelsene av denne oppfinnelse er også inhibitorer av JNK, Aurora, GSK, KDR, og AKT som bestemt ved enzymatisk undersøkelse. Proteinkinaseinhibitorene av denne oppfinnelse, eller far-masøytiske salter derav, kan formuleres i farmasøytiske sammensetningene for administrasjon til dyr eller mennes-ker. Forbindelser av den foreliggende oppfinnelse er også nyttige som inhibitorer av beslektede kinaser. Begrepet "beslektede kinaser" refererer til proteinkinaser med residuer hvilke er lignende de residuer som kler ERK-bindingstedet. Uten å ønske å være bundet av teori, spekulerer søkere på at denne hemmende aktivitet skyldes.den nære strukturelle likhet mellom det aktive sete i ERK og beslektede kinaser. Oppstilling av ERK-sekvensen med andre kinaser kan avledes fra kjente software-programmer slik som "beste tilpasning" program tilgjengelig fra Genetics Computer Group. Dette program anvender den lokale homologialgoritme beskrevet av Smith og Waterman i Advances in Applied Mathematics 2; 482

Beslektede kinaser hemmet av forbindelsene av denne oppfinnelse ville inneholde residuer, identifisert av standard proteinsekvensoppstillings software over, som tilsvarer ERK-residuene: 131, E33, G34, A35, Y36, G37, M38, V39, A52, K54, R67, T68, E71, L75, 184, 186, 1103, Q105, D106, L107, M108, E109, Dill, K114, D149, K151, S153, N154, L156, C166, og D167, med en likhets skåring på 80 % eller mer. Det har blitt funnet, fra interne røntgen krystall struk-tureksperimenter med ERK-bundede inhibitorer, at tre aminosyre-residuer i det ERK-aktive sete danner nøkkelhydrogen-bindingsinteraksjoner med disse inhibitortyper. Disse tre aminosyre-residuer er M108, D106 og Q105. Denne aminosyre-nummerering tilsvarer Swiss-Prot database nummeret for til-gang #P28482. Swiss-Prot databasen er en internasjonal pro-teinsekvens database distribuert av the European Bioinfor-matics Institute (EBI) i Geneva, Sveits. Ryggradatomene i M108 og D106, og de assosierte interaksjoner, er vanlige i alle kinaser. M108 danner både en hydrogenbindingsdonor og -akseptor og D106 danner en hydrogenbinding akseptor gjennom sin ryggrad CO. En inhibitor som kunne danne en hydrogenbinding til en eller flere av disse hydrogenbindingsgrupper innenfor det aktive sete ville forventes å binde til enzymet og derfor vise hemming.
Q105 glutaminresiduet er implisert i et delsett av kinaser som inkluderer ERK og JAK som bestemt ved undersøkelse av oppstillingsdataene oppnådd fra de ovennevnte software-programmer. Q105 sørger for en nøkkelhydrogenbindingsaksep-terende sidekjede CO. Moduleringsekspermenter viser at for både ERK og JAK er hydrogenbindingsdonoren i Ht-ringen innenfor hydrogenbindingsdistanse til Q105-residuer. På grunn av disse lignende aktive-sete interaksjoner, hemmer ERK-inhibitorene av den foreliggende oppfinnelse JAK også. Føl-gelig er disse forbindelser nyttige for behandling av JAK-medierte tilstander.

Begrepet "parenteral" som anvendt heri, inkluderer subkatane, intravenøse, intramuskulære, intra-artikulære, intra-synoviale, intrasternale, intraetkale, intrahepatiske, intralesjonale og intrakraniale injeksjons-eller infusjonsteknikker. Fortrinnsvis administreres sammensetningene oralt, intraperitonealt eller intravenøst. Sterilt injiserbare former av sammensetningene av denne oppfinnelse kan være en vandig eller en oljeaktig suspensjon. Disse suspensjoner kan formuleres i henhold til teknikker kjent i faget ved å anvende passende dispergerings-eller fuktemidler og suspensjonsmidler. Bærere for topisk administrasjon av forbindelsene av denne oppfinnelse inkluderer, men ikke begrenset til, mineralolje, parafinolje, vaselin, propylenglykol, polyok-syetylen, polyoksypropylenforbindelse, emulgerende voks og vann. For oftalmisk anvendelse kan de farmasøytiske sammensetningene formuleres som mikroniserte suspensjoner i isoton, pH-justert, steril saline, eller fortrinnsvis som løs-ninger i isoton, pH-justert, steril saline, enten med eller uten et konserveringsmiddel slik som benzylalkoniumklorid. For oftalmisk anvendelse kan de farmasøytiske sammensetninger alternativt formuleres i en salve slik som vaselin.
De farmasøytiske sammensetninger av denne oppfinnelse kan også administreres ved nasal aerosol eller inhalasjon. Slike sammensetninger fremstilles i henhold til velkjente teknikker i faget for farmasøytisk formulering og kan fremstilles som løsninger i saline, ved å anvende benzylalkohol eller andre passende konserveringsmidler, absorp-sjonsfremmere for å øke biotilgjengelighet, fluorkarboner og/eller andre konvensjonelle oppløsnings- eller dispergeringsmidler.

2,2,2-triklor-l-(4-fenylacetyl-lH-pyrrol-2-yl)-etanon (1): I en tørr kolbe ble fenylacetylklorid (1 ekvivalent) kombinert med 2-trikloracetylpyrrol (1 ekvivalent) i en mini-mumsmengde av diklormetan (DCM). 4 fenylacetyl-lH-pyrrol-2-karboksylsyre benzylamid ( 2): Til en løsning av forbindelse 1 (1 ekvivalent) i DMF, ved omgivelsestemperatur ble benzylamin (1,2 ekvivalenter) tilsatt. Eksempel 5
Vi har fremstilt andre forbindelser med formel II ved fremgangsmåter vesentlig like de beskrevet i eksemplene 1-4 over og de illustrert i Skjema I. De karakteristiske data for disse forbindelser er summert i tabell 3 under og inkluderer LC/MC, HPLC og <1>H-NMR-data.
For forbindelser hvor HPLC-metoden er angitt som "A" ble den følgende metode benyttet: en gradient av vann: MeCN, 0,1% TFA (95:5 -> 0:100) ble kjørt over 22 minutter med 1 ml/min og 214 nanometer. Eksempel 6
ERK- hemmingsundersøkelse
Forbindelser ble testet for hemmingen av ERK2 ved en spek-trofotometriskkoblet-enzymanalyse (Fox et al (1998) Protein Sei 7, 2249). I denne test ble en fast konsentrasjon aktivert ERK2 (10 nM) inkubert med forskjellige konsentrasjoner av forbindelsen i DMSO (2,5 %) i 10 minutter ved 30 °C i 0. 1 M HEPES-buffer, pH 7,5, inneholdende 10 mM MgCl2, 2,5 mM fosfoenolpyruvat, 200 uM NADH, 150 j^g/ml pyruvatkinase, 50 uM/ml laktatdehydrogenase og 200 jiM erktidpeptid. Forbindelser som har en aktivitet angitt som "A" ga en Ki-verdi under 1 mikromolar; forbindelser som har en aktivitet angitt som "B" ga en K—verdi mellom 1 og 5 mikromolar; og forbindelser som har en aktivitet angitt som "C" ga en Ki-verdi større enn 5 mikromolar.
ERK- hemming celleproliferasjonstest
Forbindelser ble testet for hemmingen av ERK2 ved en celle-prolif eras jonstest . I denne test ble et fullstendig medium fremstilt ved å tilsette 10 % føtalt bovinserum og pencil-lin/streptomycinløsning til RPMI 1640-medium (JRH Bioscien-ces). Koloncancerceller (HT-29-cellelinje) ble tilsatt til hver av 84 brønner i en 96-brønns plate med en såings-tetthet på 10.000 celler/brønn/150 \il. Cellene ble tillatt å feste seg til platen ved inkubering ved 37°C i 2 timer. En løsning av testforbindelse ble fremstilt i fullstendig medium ved seriefortynning for å oppnå de følgende konsentrasjoner: 20 uM, 6,7 uM, 2,2 ^iM, 0,74 ^M, 0,25 uM og 0,08 uM. Testforbindelsesløsningen (50 ^1) ble tilsatt til hver av 72 celleinneholdende brønner. Utvalgte forbindelser av denne oppfinnelse som hemmer ERK i koloncelle-proliferasjonstesten med en IC50 på mindre enn 10 uM inkluderer: 11-43, 11-48 og 11-45.
Eksempel 8
JAK- hemmingstest
Forbindelseshemming av JAK kan testes ved metoden beskrevet av G.R.Brown, et al, Bioorg. Med. Chem. Lett. 2000, vol. 10, s. 575-579 på den følgende måte. Til Maxisorb-plater, tidligere belagt ved 4°C med Poly(Glu, Ala, Tyr) 6:3:1, deretter vasket med fosfatbufret saline 0,05 % og Tween (PBST), tilsettes 2 uM ATP, 5 mM MgCl2 og en løsning av forbindelse i DMSO. Reaksjonen startes med JAK-enzym og platene inkuberes i 60 minutter ved 30°C. Platene vaskes deretter med PBST, 100 (il HRP-konjugert 4G10 antistoff tilsettes og platen inkuberes i 90 minutter ved 30°C. Platen vaskes igjen med PBST, 100 \ il TMB-løsning tilsettes, deretter inkuberes platene i nye 30 minutter ved 30°C. Svovel-syre (100 ul, IM) tilsettes for å stoppe reaksjonen og platen leses ved 450 nanometer for å oppnå optiske tettheter for analyse for å bestemme ICso-verdier.
Eksempel 9
JNK- hemmingstest
Forbindelser ble screenet på den følgende måte for sin evne til å hemme JNK ved å anvende en spektrofotometrisk koblet enzymtest. Til en prøvebruksbufferløsning inneholdende 0,1 M HEPES-buffer (pH 7,5), 20 mM MgCl2, 2,5 mM fosfoenolpyruvat, 200 uM NADH, 150 ug/ml pyruvatkinase, 50 (ig/ml laktatdehydrogenase og 200 uM EGF-reseptorpeptid med sekvens KRELVEPLTPSGEAPNQALLR), ble forskjellige konsentrasjoner av forbindelsen i DMSO og en fast konsentrasjon (10 nM) aktivert JNK tilsatt. Den resulterende blanding ble inkubert ved 30°C i 10 minutter, deretter ble reaksjonen initiert ved tilsettingen av 10 \ iM ATP. Absorbansminkningen ved 340 nM ved 30°C ble monitorert som en funksjon av tid og de resulterende data ble tilpasset til en konkurransehemmings-kinetisk modell for å bestemme Ki.

Eksempel 10.
Aurora- hemminqstest
Forbindelser ble screenet på den følgende måte for sin evne til å hemme Aurora ved å anvende en standard koblet enzymtest. Til en prøvebruksbufferløsning inneholdende 0,1 M HEPES 7,5, 10 mM MgCl2, 25 mM NaCl, 2,5 mM fosfoenolpyruvat, 300 uM NADH, 30 ul pyruvatkinase, 10 ug/ml laktatdehydrogenase, 40 uM ATP og 800 uM peptid (LRRASLG, American Peptide, Sunnyvale, CA) ble en 30 uM løsning av forbindelsen i DMSO tilsatt og den resulterende blanding inkubert ved 30°C i 10 minutter. Reaksjonen ble initiert ved tilsettingen av 10 ul av 70 nM Aurora og 1 mM DTT. Reaksjonshastighetene ble oppnådd ved å monitorere absorbans ved 340 nM over 5 minutter lesetid ved 37°C ved å anvende en BioRad Ultramark plateleser (Hercules, CA) . IC5o ble bestemt fra hastighetsdataene som en funksjon av inhibitorkonsentrasjon.
Tabell 6 viser resultatene for aktiviteten til utvalgte forbindelser av denne oppfinnelse i Aurora2-hemmingstesten. Forbindelsesnummerne tilsvarer forbindelsesnummerne i tabell 1. Forbindelser som har en aktivitet angitt som "A" ga en ICso-verdi under 5 mikromolar; forbindelser som har en aktivitet angitt som "B" ga en IC5o-verdi mellom 5 og 10 mikromolar; og forbindelser som har en aktivitet angitt som "C" ga en ICso-verdi større enn 10 mikromolar.
Eksempel 11
GSK- 3 hemmingstest
Forbindelser ble screenet på den følgende måte for sin evne til å hemme Glykogen synthase kinase 3 (GSK-3) ved å anvende en standard koblet enzymtest (Fox et al (1998) Protein Sei 7, 2249). Til en prøvebruksbufferløsning inneholdende 0,1 M HEPES 7,5, 10 mM MgCl2, 25 mM NaCl, 2,5 mM fosfoenolpyruvat, 300 uM NADH, 1 mM DTT, 30 jig/ml pyruvatkinase, 10 uxj/ml laktatdehydrogenase, 300 uM peptid (HSSPHQp-SEDEEE, American Peptide, Sunnyvale, CA) og 60 nM GSK-3 ble en 30 fiM løsning av forbindelsen i DMSO tilsatt og den resulterende blanding inkubert ved 30°C i 5 minutter. Reaksjonen ble initiert ved tilsettingen av 10 uM ATP. Reaksjonshastighetene ble oppnådd ved å monitorere absorbans ved 340 nM over 5 minutters lesetid ved 30°C ved å anvende en Molecular Devices-plateleser (Sunnyvale, CA). IC50 ble bestemt fra hastighetsdataene som en funksjon av inhibitorkonsentrasj onen.
Tabell 7 viser resultatene for aktiviteten til utvalgte forbindelser av denne oppfinnelse i GSK-3 hemmingstesten. Forbindelsesnummerne tilsvarer forbindelsesnummerne i tabell 1. Forbindelser som har en aktivitet angitt som "A" ga en ICso-verdi under 10 mikromolar; forbindelser som har en aktivitet angitt som "B" ga en IC5o-verdi mellom 10 og 20 mikromolar; og forbindelser som har en aktivitet angitt som "C" ga en ICso-verdi større enn 20 mikromolar.
Eksempel 12
KDR- hemmingstest
Forbindelser ble screenet for deres evne til å hemme KDR ved å anvende en standard koblet enzymtest (Fox et al.,Protein Sei., (1998) 7, 2249). Testene ble utført i en blanding av 200 mM HEPES 7,5, 20 mM MgCl2, 25 mM NaCl, 1 mM DTTT and 1,5 % DMSO. Sluttsubstratkonsentrasjoner i prøven var 300 uM ATP (Sigma Chemicals) og 10 uM poly E4Y (Sigma). Testene ble utført ved 37°C og 30 nM KDR. Sluttkonsentrasjoner av komponentene i det koblede enzymsystem var 2,5 mM fosfoenolpyruvat, 200 uM NADH, 30 uM/ml pyruvatkinase og 10 uM/ml laktatdehydrogenase.
En prøvebruksbufferoppløsning ble fremstilt inneholdende alle reagensene listet over, med unntak av ATP og testforbindelsen av interesse. 177 ul av bruksløsningen ble plas-sert i en 96 brønns plate etterfulgt av tilsetting av 3 ul med 2 mM DMSO standard inneholdende testforbindelsen (ende-lig forbindelseskonsentrasjon 30 uM). Platen ble preinkubert i omtrent 10 minutter ved 37°C og reaksjonen initiert ved tilsetning av 20 ul ATP (sluttkonsentrasjon 300 uM). Reaksjonshastigheter ble oppnådd ved å anvende en Molecular Devices plateleser (Sunnyvale, CA), over en 5 minutters lesetid ved 37 °C. Forbindelser som viser mer enn 50 % hemming versus standardbrønner inneholdende prøveblandingen og DMSO uten testforbindelse ble titrert for å bestemme IC50-verdier.
Utvalgte forbindelser av denne oppfinnelse som hemmer KDR ved 2 uM-konsentrasjon i testen over med en prosentvis hemming større enn 40% inkluderer: 11-43, 11-48, 11-304 og II-305.
Eksempel 13
AKT- hemmingstest
Forbindelse ble screenet for deres evne til å hemme AKT ved å anvende en standardkoblet-enzymtest (Fox et al., Protein Sei., (1998) 7, 2249). Tester ble utført i en blanding av 100 mM HEPES 7,5, 10 mM MgCl2, 25 mM NaCl, 1 mM DTT og 1,5 % DMSO. Endelige substratkonsentrasjoner i prøven var 170 uM ATP (Sigma Chemicals) og 200 uM peptid (RPRAATF, American Peptide, Sunnyvale, CA). Tester ble utført ved
30 °C og 45 nM AKT. Sluttkonsentrasjoner av komponentene i det koblede enzymsystem var 2,5 mM fosfoenolpyruvat, 300 uM NADH, 30 ug/ml pyruvatkinase og 10 ug/ml laktatdehydrogenase.
En prøvebruksbufferløsning ble fremstilt inneholdende alle reagensene listet over med unntak av AKT, DTT og testforbindelsen av interesse. 56 ul av bruksløsningen ble plas-sert i en 384 brønns plate etterfulgt av tilsetting av 1 ul 2 mM DMSO standard inneholdende testforbindelsen (endelige forbindelseskonsentrasjon 30 uM). Platen ble preinkubert i ca. 10 minutter ved 30°C og reaksjonen initiert ved tilsetting av 10 (il enzym (sluttkonsentrasjon 45 nM) og 1 mM DTT. Reaksjonshastigheter ble oppnådd ved å anvende en BioRad Ultramark-plateleser (Hercules, CA) over 5 minutters lesetid ved 30°C. Forbindelser som viser mer enn 50 % hemming versus standardbrønner inneholdende prøveblandingen og DMSO uten testforbindelse ble titrert for å bestemme IC50-ver-dier.
Utvalgte forbindelser av denne oppfinnelse som hemmer AKT inkluderer: 11-89, 11-94 og 11-305.
Selv om vi har beskrevet en rekke utførelser av denne oppfinnelse, er det åpenbart at våre basiseksempler kan endres for å gi andre utførelser som benytter forbindelsene og fremgangsmåtene av denne oppfinnelse. Derfor vil det bli verdsatt at rammen av denne oppfinnelse defineres av de vedlagte krav heller enn av de spesifikke utførelser som har blitt beskrevet eksempelvis.
Claims (14)
1. Forbindelse med formel I:

Anvendelse av en forbindelse ifølge krav 1-5 for fremstilling av en farmasøytisk sammensetning for behandling av en sykdomstilstand valgt fra cancer, slag, diabetes, hepatomegali, kardiovaskulær sykdom, Alzheimers sykdom, cystisk fibrose, viral sykdom, autoimmune sykdommer, aterosklerose, restenose, psoriasis, allergiske forstyrrelser, inflammasjon, nevrologiske forstyrrelser, en hormonrelatert sykdom, tilstander assosiert med organtransplantasjon, immunsvikts-forstyrrelser, destruktive benforstyrrelser, proliferative forstyrrelser, infeksiøse sykdommer, tilstander assosiert med celledød, trombinindusert plateaggregering, kronisk myelogen leukemi (CML), leversykdom, patologiske immuntilstander som involverer T-celleaktivering eller CNS-forstyrrelser.
9. Anvendelse av en forbindelse ifølge krav 8 hvori syk-domstilstanden er cancer.
10. Anvendelse ifølge krav 9, hvori canceren er valgt fra bryst-; ovarie-; cervix-; prostata-; testikkelcancer; cancer i genito-urinveissystem; esofagus-; larynkscancer, glioblastom; nevroblastom; mage-, hudcancer, keratoakantom; lungecancer, epidermoidkarsinom, storcelle-karsinom, små-celle-karsinom; lunge-adenokarsinom; ben-; coloncancer; adenom; cancer i pankreas; adenokarsinom; tyreoidacancer; follikulært karsinom; udifferensiert karsinom; papillært karsinom; seminom; melanom; sarkom; blærekarsinom; lever-karsinom og cancer i gallepassasjer; nyrekarsinom; myeloide forstyrrelser; lymfoide forstyrrelser; Hodgkins sykdom, "hårete" celler; bukkal hulroms- og farynks(oral)-, leppe-, tunge-, munn-, farynks-; tynntarms-; colon-rektums-, tykk-tarms-, rektumscancer; hjernecancer og cancer i sentralner-vesystemet eller levkemi.
11. Anvendelse av en forbindelse ifølge krav 8 hvori syk-domstilstanden er kardiovaskulær sykdom.
12. Anvendelse ifølge krav 11, hvori den kardiovaskulære sykdom er valgt fra restenose, kardiomegali, arterosklero-se, myokardinfarkt eller kongestiv hjertesvikt.
13. Sammensetning for belegning av en implanterbar anordning omfattende en forbindelse ifølge krav 1 og en bærer egnet for belegning av den implanterbare anordning.
14. En implanterbar anordning belagt med en sammensetning ifølge krav 13.
