Abstract
Se describe una celula bacterial gram-negativa que es deficiente en un gen cromosomal presente en el tipo silvestre de tal celula cuyo gen comparte al menos el 80% de identidad de secuencia con la secuencia nativa del gen yfcK y codifica para una aminopeptidasa. Alternativamente, una celula bacterial gram-negativa es deficiente en un gen cromosomal presente en el tipo silvestre de tal celula cuyo gen codifica una aminopeptidasa que comparte al menos 80% de identidad de secuencia con la secuencia nativa de la aminopeptidasa b2324. Cualquiera de estos tipos de celulas, cuando comprenden un acido nucleico que codifica para un polipeptido heterologo, produce un polipeptido no eliminado de N-terminal cuando se cultiva y el polipeptido recuperado, sin virtualmente el polipeptido eliminado de N-terminal producido como una impureza. Contrariamente, se proporciona un metodo para desdoblar un aminoacido N-terminal a partir de un polipeptido que comprende poner en contacto al polipeptido con una aminopeptidasa que comparte al menos 80% de identidad de secuencia con la secuencia nativa de la aminopeptidasa b2324.
