Abstract

A találmány tárgyát Gram-negatív baktériumsejt képezi, amely egy - vadtípusú sejtben jelen lévő - kromoszomális gén vonatkozásában hiányos,mely gén az yfcK-gén natív szekvenciájával legalább 80%-ban azonosszekvenciát tartalmaz és aminopeptidázt kódol. A találmány tárgyátképezi továbbá egy Gram-negatív baktériumsejt, amely egy - vad típusúsejtben jelen lévő - kromoszomális gén vonatkozásában hiányos, melygén a b2324-aminopeptidáz natív szekvenciájával legalább 80%-banazonos szekvenciájú aminopeptidázt kódol. A találmány szerinti sejtek,ha heterológ polipeptidet kódoló nukleinsavat tartalmaznak,tenyésztést követően olyan polipeptidet termelnek, amely N-terminálisán nincs hasítva, és gyakorlatilag szennyezésként semtermelődik N-terminálisán hasított polipeptid. Szintén a találmánytárgyát képezi egy eljárás egy polipeptid N-terminális aminosavánaklehasítására, melynek során a polipeptidet olyan aminopeptidázzalérintkeztetik, amely a b2324-aminopeptidáz natív szekvenciájávallegalább 80%-ban azonos szekvenciát tartalmaz.
