Abstract

La presente invencion se relaciona generalmente con plasmidos recombinantes y en particular plasmidos recombinantes geneticos que comprenden secuencias polinucleotidicas capaces de liberarse en forma circular, cerrada covalentemente a partir de una secuencia nucleotidica mas grande tal como, pero sin limitarse a un genoma de una celula eucariota. Preferiblemente, una vez liberada, la secuencia polinucleotidica se reconstituye en una forma que permite la expresion de la secuencia polinucleotidica. En una modalidad, la secuencia polinucleotidica reconstituida comprende una secuencia codificante con la totalidad o parte de un nucleotido extrano tal como, pero sin limitarse a una secuencia intronica u otra senal de maduracion insertada en la misma. La expresion y en particular la transcripcion de la secuencia codificante involucra la maduracion de la secuencia extrana. La liberacion y circularizacion generalmente es en respuesta a un estimulo tal como un estimulo mediado por una proteina. Mas particularmente, la proteina es una proteina viral o derivada de procariota o eucariota o bien una proteina regulada especifica por el desarrollo o por el tejido. El plasmido recombinante de la presente invencion es util particularmente para conferir resistencia genetica contra patogenos o inducir apoptosis u otros mecanismos de muerte celular utiles, por ejemplo, para tratar cancer o inducir esterilidad masculina o femenina en plantas. Los plasmidos recombinantes permiten, por lo tanto, expresion especifica de sitio o expresion regulada en el desarrollo de secuencias geneticas seleccionadas.
