Claims (18)
1. Miš čiji genom obuhvaća:
(a) bar jedan nerearanžirani ljudski VH genski segment, bar jedan nerearanžirani ljudski DH genski segment, i bar jedan nerearanžirani ljudski JH genski segment koji je operativno povezan s genom koji kodira endogenu konstantnu regiju teškog lanca u lokusu endogenog teškog lanca;
(b) ne više od jedne, ili ne više od dvije, rearanžirane ljudske Vκ/Jκ sekvence koje su operativno povezane sa genom koji kodira konstantnu regiju endogenog κ lakog lanca u lokusu endogenog κ lakog lanca; i
(c) sekvenca ektopične nukleinske kiseline koja kodira funkcionalni mišji ADAM6a protein ili funkcionalni ortolog, funkcionalni homolog ili njegov funkcionalni fragment i sekvenca ektopične nukleinske kiseline koja kodira funkcionalni mišji ADAM6b protein ili funkcionalni ortolog, funkcionalni homolog ili njegov funkcionalni fragment, pri čemu su ADAM6 proteini, ortolozi, homolozi, ili fragmenti funkcionalni kod mužjaka miša, pri čemu je spomenuta funkcija povezana s fertilitetom mužjaka miša,
pri čemu mišjem genomu nedostaje:
funkcionalni endogeni gen za ADAM6 u endogenom lokusu ADAM6,
funkcionalni endogeni Vκ genski segment imunoglobulina, i
funkcionalni endogeni Jκ genski segment imunoglobulina.
2. Miš prema zahtjevu 1, pri čemu sekvence ektopične nukleinske kiseline su na poziciji koja nije u lokusu endogenog teškog lanca imunoglobulina.
3. Miš prema zahtjevu 1, pri čemu mišji genom obuhvaća ne više od dvije rearanžirane ljudske Vκ/Jκ sekvence.
4. Miš prema zahtjevu 3, pri čemu ne više od dvije rearanžirane ljudske Vκ/Jκ sekvence su izabrane iz grupe koja se sastoji iz rearanžirane ljudske Vκ1-39/Jκ sekvence i rearanžirane ljudske Vκ3-20/Jκ sekvence.
5. Miš prema zahtjevu 1, pri čemu mišji genom obuhvaća ne više od jedne rearanžirane ljudske Vκ/Jκ sekvence.
6. Miš prema zahtjevu 5, pri čemu ne više od jedne rearanžirane ljudske Vκ/Jκ sekvence je izabrana iz grupe koja se sastoji iz rearanžirane ljudske Vκ1-39/Jκ sekvence i rearanžirane ljudske Vκ3-20/Jκ sekvence.
7. Miš prema zahtjevu 6, pri čemu rearanžirana ljudska Vκ1-39/Jκ sekvenca je rearanžirana ljudska Vκ1-39/Jκ5 sekvenca.
8. Miš prema zahtjevu 6, pri čemu rearanžirana ljudska Vκ1-39/Jκ sekvenca je rearanžirana ljudska Vκ3-20/Jκ1 sekvenca.
9. Postupak za dobijanje antitijela koji obuhvaća:
(a) imuniziranje miša prema bilo kojem od zahtjeva 1 do 8;
(b) ekspresiju u stanici:
(i) nukleotidne sekvence ljudskog teškog lanca koja se sastoji iz sekvence varijabilnog regiona ljudskog teškog lanca izolirane iz spomenutog miša, koja je operativno povezana sa sekvencom gena konstantnog područja ljudskog teškog lanca;
(ii) nukleotidne sekvence ljudskog lakog lanca koja se sastoji iz sekvence varijabilnog regiona ljudskog κ lakog lanca koja je izolirana iz spomenutog miša, koja je operativno povezana sa sekvencom gena konstantnog područja ljudskog lakog lanca; i
(c) održavanje stanice pod uvjetima koji su dovoljni za ekspresiju potpuno ljudskog antitijela, i
(d) izoliranje antitijela.
10. Postupak za dobijanje antitijela prema zahtjevu 9, pri čemu nukleotidna sekvenca ljudskog teškog lanca 9(b)(i) je nukleotidna sekvenca prvog ljudskog teškog lanca, i postupak dalje obuhvaća:
(e) imunizaciju miša prema bilo kojem od zahtjeva 1 do 8;
(d) ekspresiju u stanici:
(i) nukleotidnu sekvencu drugog ljudskog teškog lanca koja se sastoji iz sekvence varijabilnog područja drugog ljudskog teškog lanca koja je izolirana iz spomenutog miša, operativno povezana sa sekvencom gena konstantnog područja ljudskog teškog lanca.
11. Mišja stanica čiji genom obuhvaća:
(a) bar jedan nerearanžirani ljudski VH genski segment, bar jedan nerearanžirani ljudski DH genski segment, i bar jedan nerearanžirani ljudski JH genski segment operativno povezan sa genom endogene konstantne područja teškog lanca u lokusu endogenog teškog lanca;
(b) ne više od jedne, ili ne više od dvije, rearanžirane ljudske Vκ/Jκ sekvence operativno povezane s endogenim genom konstantnog regija κ lakog lanca u endogenom lokusu κ lakog lanca; i
(c) sekvenca ektopične nukleinske kiseline koja kodira funkcionalni mišji ADAM6a protein ili funkcionalni ortolog, funkcionalni homolog ili njegov funkcionalni fragment i sekvenca ektopične nukleinske kiseline koja kodira funkcionalni mišji ADAM6b protein ili funkcionalni ortolog, funkcionalni homolog ili njegov funkcionalni fragment, pri čemu su ADAM6 proteini, ortolozi, homolozi, ili fragmenti funkcionalni kod mužjaka miša, pri čemu je spomenuta funkcija povezana s fertilitetom mužjaka miša,
pri čemu genomu mišje stanice nedostaje:
funkcionalni endogeni gen za ADAM6 u endogenom lokusu ADAM6,
funkcionalni endogeni Vκ genski segment imunoglobulina, i
funkcionalni endogeni Jκ genski segment imunoglobulina.
12. Mišja stanica prema zahtjevu 11, pri čemu je stanica ES stanica.
13. Mišja stanica prema zahtjevu 11, pri čemu ektopične sekvence nukleinskih kiselina su u poziciji koja nije u lokusu endogenog teškog lanca imunoglobulina.
14. Mišja stanica prema zahtjevu 11, pri čemu genom mišje stanice obuhvaća ne više od dvije rearanžirane ljudske Vκ/Jκ sekvence.
15. Mišja stanica prema zahtjevu 14, pri čemu ne više od dvije rearanžirane ljudske Vκ/Jκ sekvence su izabrane iz grupe koja se sastoji iz rearanžirane ljudske Vκ1-39/Jκ sekvence i rearanžirane ljudske Vκ3-20/Jκ sekvence.
16. Mišja stanica prema zahtjevu 11, pri čemu genom mišje stanice obuhvaća ne više od jedne rearanžirane ljudske Vκ/Jκ sekvence.
17. Mišja stanica prema zahtjevu 16, pri čemu ne više od jedne rearanžirane ljudske Vκ/Jκ sekvence je izabrana iz grupe koja se sastoji iz rearanžirane ljudske Vκ1-39/Jκ sekvence i rearanžirane ljudske Vκ3-20/Jκ sekvence.
18. Mišja stanica prema zahtjevu 17, pri čemu:
(a) rearanžirana ljudska Vκ1-39/Jκ sekvenca je rearanžirana ljudska Vκ1-39/Jκ5 sekvenca; ili
