45 La unión de oligonucleótidos antisentido o en sentido a las secuencias de ácido nucleico diana da como resultado la formación de dúplex que bloquean la transcripción o traducción de la secuencia diana por uno o varios medios, que incluyen el aumento de la degradación de los dúplex, terminación prematura de la transcripción o traducción, o por otros medios. Ejemplos específicos de compuestos antisentido preferidos útiles para inhibir la expresión de proteínas SMO mutantes incluyen los oligonucleótidos que contienen armazones modificados o enlaces internucleósido no naturales. mediada por CaPO4, electroporación, o utilizando vectores de transferencia génica tales como virus de Epsten-Barr. En un procedimiento preferido, se inserta un oligonucleótido antisentido o en sentido en un vector retrovírico adecuado M-MuLV, N2 (un retrovirus derivado de M-MuLV), o vectores de copia doble denominados DCT5A, DCT5B y DCT5C (véase el documento WO 90/13641).
5 Los oligonucleótidos antisentido o en sentido también se pueden introducir en una célula que contiene la secuencia de nucleótido diana por la formación de un conjugado con una molécula de unión al ligando, como se describe en el documento WO 91/04753. 15 secuencia de ácido nucleico diana por la formación de un complejo oligonucleótido-lípido, como se describe en el documento WO 90/10448. El complejo oligonucleótido en sentido o antisentido-lípido se disocia preferentemente en la célula por una lipasa endógena.

Las secuencias de nucleótido que codifican un SMO mutante también se pueden utilizar para construir sondas de hibridación para mapear el gen que codifica esa SMO y para el análisis genético de individuos con trastornos genéticos. Las secuencias de nucleótido que se proporcionan en el presente documento pueden mapearse en un cromosoma y en regiones específicas de un cromosoma utilizando técnicas conocidas, tales como hibridación in situ, análisis de unión con marcadores cromosómicos conocidos y cribado por hibridación con bibliotecas.
35 Un potencial antagonista del SMO mutante es una construcción de ARN o ADN antisentido que se prepara utilizando tecnología antisentido, en la que, por ejemplo, una molécula de ARN o ADN actúa bloqueando directamente la traducción de ARNm hibridándose con un ARNm dirigido y evitando la traducción de la proteína. Se puede utilizar una tecnología antisentido para controlar la expresión génica por medio de la formación de una hélice triple o un ADN o ARN antisentido, ambos métodos se basan en la unión de un polinucleótido al ADN o ARN. Por ejemplo, los ácidos nucleicos que codifican el SMO mutante en el presente documento, se utilizan para diseñar un oligonucleótido ARN de desde aproximadamente 10 a 40 pares de bases de longitud. Un oligonucleótido de ADN se diseña para que sea complementario a la región del gen implicado en la transcripción (hélice triple – véase Lee et al. (1979) Nucl. Acids Res. 6: 3073; Cooney et al. (1988) Science 241: 456; Dervan et al. (1991) Science 251: 1360),
45 evitando de esta manera la transcripción y la producción de SMO mutantes. El oligonucleótido ARN antisentido se hibrida al ARNm in vivo y bloquea la traducción de la molécula de ARNm en mutantes SMO (Okano (1991) Neurochem. 56: 560); OLIGODEOXYNUCLEOTIDES AS ANTISENSE INHIBITORS OF GENE EXPRESSION, CRC Press, Boca Raton, FL, 1988). Los oligonucleótidos descritos anteriormente también se pueden suministrar a las células de manera que el ARN o ADN antisentido se pueda expresar in vivo para inhibir la producción de SMO mutante. Cuando se utiliza el ADN antisentido, se prefieren los oligodesoxirribonucleótidos derivados del sitio de inicio de la traducción, por ejemplo entre -10 y +10 posiciones de la secuencia de nucleótido del gen diana.
Los potenciales antagonistas de SMO mutantes incluyen moléculas pequeñas que se unen al sitio ocupado por el GDC-0449 en el SMO de tipo silvestre, bloqueando de esta manera la actividad biológica del SMO mutante.
55 Ejemplos de moléculas pequeñas incluyen, pero no se limitan a, péptidos pequeños o moléculas tipo péptido, preferentemente péptidos solubles, y compuestos inorgánicos u orgánicos no peptídicos sintéticos.
Las ribozimas son moléculas enzimáticas de ARN capaces de catalizar la escisión específica de ARN. Las ribozimas actúan por hibridación específica de secuencia con el ARN diana complementario, seguido por escisión endonucleolítica. Los sitios de escisión específicos de ribozima con un potencial ARN diana se pueden identificar por técnicas conocidas. Para más detalles, véase, por ejemplo, Rossi (1994) Current Biology, 4: 469-471, y publicación PCT Nº WO 97/33551 (publicada el 18 de septiembre de 1997).

II. Proteínas
La invención proporciona proteínas SMO mutantes aisladas. La SMO humana de tipo silvestre se muestra en SEQ ID NO: 1. La SMO mutante humana se muestra en SEQ ID NO: 2 en la que el aminoácido 473 se muestra como “X”, 15 con respecto a la presente solicitud sirve para cualquier aminoácido distinto de ácido aspártico (D). En algunas realizaciones, la X es histidina (H), glicina (G), fenilalanina (F), tirosina (Y), leucina (L), isoleucina (I), prolina (P), serina (S), treonina (T), metionina (M), glutamina (Q) o asparagina (N). 20 expresión como se sabe bien en esa técnica y se transfectan en células huésped, que pueden ser células procariotas o eucariotas dependiendo del uso propuesto para la proteína. La mutante SMO de longitud completa o los fragmentos (en los cuales los fragmentos contienen al menos la parte del extremo carboxilo del dominio transmembrana 6 y el aminoácido 473 de SEQ ID NO: 2) se puede utilizar como inmunógenos para producir los anticuerpos de la invención.

30 En un aspecto, la invención proporciona anticuerpos que se unen al SMO, particularmente a SMO mutante. En una realización, un anticuerpo anti-SMO es un anticuerpo monoclonal. En una realización, un anticuerpo anti-SMO es un fragmento de anticuerpo, por ejemplo, un fragmento Fab, Fab’-SH, Fv, o F(ab’)2. En una realización, un anticuerpo anti-SMO mutante es un anticuerpo quimérico, humanizado o humano. En una realización, un anticuerpo anti-SMO está purificado. En ciertas realizaciones, una composición es una formulación farmacéutica para el tratamiento del

1. Fragmentos de anticuerpo
La presente invención engloba fragmentos de anticuerpo. Los fragmentos de anticuerpo se pueden generar por
40 medios tradicionales, tales como digestión enzimática, o por técnicas recombinantes. En ciertas circunstancias existen ventajas en la utilización de fragmentos de anticuerpo, más que anticuerpos completos. El tamaño menor de los fragmentos permite un aclaramiento rápido y puede dar lugar a un acceso mejor a tumores sólidos. Para revisar ciertos fragmentos de anticuerpo, véase Hudson et al. (2003) Nat. Med. 9: 129-134.
45 Se han desarrollado distintas técnicas para la producción de fragmentos de anticuerpo. Tradicionalmente estos fragmentos se derivaron por medio de digestión proteolítica de anticuerpos intactos (véase, por ejemplo, Morimoto et al., Journal of Biochemical y Biophysical Methods 24: 107-117 (1992); y Brennan et al., Science, 229: 81 (1985)). Sin embargo, estos fragmentos se pueden producir ahora directamente mediante células huésped recombinantes. Los fragmentos de anticuerpo Fab, Fv y scFv se pueden expresar y secretar a partir de E. coli, permitiendo por lo tanto la
