Opfindelsen angår en fremgangsmåde til udvinding af olieholdige kraftigt rensede phosphatidylcholiner med et højt indhold af essentielle fedtsyrer ved ekstraktion af råphosphatid med organiske opløsningsmidler, adsorption på aluminiumoxid eller en aluminiumoxidsøjle og opløsning af phosphatidylcholinet med alkohol.
Fra de tyske patentskrifter nr. 1.617.679 og nr. 1.617.680 kendes fremgangsmåder til udvinding af kraftigt renset phosphatidylcholin med et højt indhold af essentielle fedtsyrer af vegetabilske lecithi-ner ved absorption af phosphatiderne på aluminiumoxid og ekstraktion med alkohol, hvor de olieholdige råphosphatider opløses i eddikeester eller en dichloreret hydrocarbon med 1-2 carbonatomer eller blandinger af disse opløsningsmidler, hvorhos opløsningsmidlet kan indeholde indtil 6 volumen% alkohol, og opløsningen så under omrøringen behandles med mindst den femdobbelte mængde aluminiumoxid beregnet på råphosphatidindholdet, hvorpå det kraftigt rensende phosphatidylcholin ekstraheres med alkohol fra det adskilte aluminiumoxid.

Fra schweizisk patentskrift nr. 361.088 og amerikansk patentskrift nr. 2.945.869 kendes rensningsmetoder til udvinding af sojaphos-phatidfraktioner, der skal finde anvendelse som emulgatorer til fedtemulsioner til intravenøs anvendelse. Herved bliver alkoholiske opløsninger af forud afolierede råphosphatider behandlet med ΑΊ^Ο^,
MgO eller aktivt carbon for at gøre disse opløsninger fattige på kephalin og frem for alt for i vidt omfang at befri dem for de inosit-holdige phosphatider, som ved en intravenøs applikation til katte fremkalder en blodtrykssnkning.
Ved den kendte teknik bliver phosphatidet først afolieret ved anvendelse af sædvanlige .fedtopløsende opløsningsmidler, såsom chlorerede hydrocarboner, eddikesyreethylester eller lignende, hvorpå det af-fedtede materiale derpå bliver ekstraheret med alkohol for at fremstille den alkoholiske opløsning, som derpå bliver behandlet med et vilkårligt adsorberende stof. Herved opnås imidlertid en phospha-lidylcholin, som endnu indeholder kephalin som forurening.
Fra tysk patentskrift nr. 1.053.299 kendes desuden en fremgangsmåde til udvinding af colaminphosphorsyrediglyceridesterfrie naturlige cholinphosphorsyrediglyceridestere ved søjlechromatographi, hvorhos der som absorberende stof blandt andet også anvendes aluminiumoxid. Også ved denne fremgangsmåde anvendes en alkoholisk ekstrakt af forud afolierede råphosphatid. Denne forudgående afoliering -opnås ved ekstraktion med acetone gentagne gange.
I modsætning hertil fås ved fremgangsmåden ifølge opfindelsen phos= phatidyloholinerne med et højt indhold af essentielle fedtsyrer i molekylet sammen med den i udgangsphosphatidet tilstedeværende planteolie. Det har nemlig i praksis vist sig, at det meget ofte er vigtigt at fremstille olieagtige opløsninger af kraftigt rensede phos= phatidylcholiner. Dette gælder specielt til en anvendelse på det farmaceutiske område., når det drejer sig om at gøre kraftigt rensede phosphatidylcholiner tilgængelige for oral applikation f.eks i form af- gelatinekapsler. Nyere resorptionsstudier med radioaktivt mærket phosphatidylcholin har nemlig vist, at resorptionen ved oral applikationen af en olieholdig applikationsform af phosphatidylcholin med overvejende polyumættede fedtsyrer kan forøges flere gange i forhold 148111 3 til et vandholdigt præparat. Et fortrinligt oliemedium hertil viste sig at være den med phosphatiderne altid associerede naturlige planteolie såsom solsikke- eller sojaolie fortrinsvis i blanding med mono-og diglycerider,som blev fremstillet af de samme olier. På denne måde bliver det muligt umiddelbart at få flydende,d.v.s. hældbare phospha= tidylcholin-fraktioner,som også hyppigt er af stor betydning til andre anvendelser f.eks på kosmetik- og emæringslæreområdét.
Ifølge den hidtil kendte teknik sker fremstillingen af en hældbar olieagtig opløsning af kraftigt renset phosphatidylcholin, som er egnet til påfyldning i gelatinekapsler^ved at man behandler en oliefri phosphatidylcholin,f.eks udvundet ved fremgangsmåden ifølge det tyske patentskrift nr. 1.617.679, igen med den ved rensningen opnåede naturlige planteolie eller en anden spiseolie eventuelt under tilsætning af monoglycerider eller frie fedtsyrer. Dette betyder i-midlertid,at man til fjernelse af kephalinets og inositphosphatidet først må fjerne den til phosphatidblandingerne endnu vedhæftende olie før eller vinder adsorptionsbehandlingen på A^O^.
De kendte fremgangsmåder, såvel fremgangsmåden ifølge DE-PS 1.053.299 som ifølge DE-PS 1.617.679 og DE-PS 1.617.680 er foruden at være forbundet med temmelig store udgifter også behæftet med yderligere ulemper. I første tilfælde sker der under afolieringen af de i handelen værende råphosphatider med acetone altid en dannelse af små mængder mesityloxid, som kemisk dannes ved addition af 2 molekyler acetone, og som på grund af sin toxicitet og sin karakteristiske egenlugt virker temmelig forstyrrende selv i ganske små mængder. I det andet tilfælde dannes der ved chromatographisk fraktionering af olie og phospholipid altid opløsningsmiddelblandinger, som før genanvendelsen skal underkastes en fraktioneret destillation.
Ved den praktiske gennemførelse af fremgangsmåden ifølge opfindelsen ekstraheres de i handelen værende endnu 20-40% olieholdige rå= lecithiner først ved temperaturer indtil 50°C med methanol, ethanol eller propanol,hvorhos de ved afkølingen af blandingen dannede to faser adskilles fra hinanden ved dekantering. Den alkoholrige overfase behandles derpå med aluminiumoxidet enten under omrøring eller ved at passere en med det adsorberende stof fyldt chromatographi= 4 148111 søjle. Ved eluering med alkoholen fås en olieholdig kephalin- og inositphosphatidfri phosphatidylcholin med et højt indhold af essen= tielle fedtsyrer.
Fremgangsmåden ifølge opfindelsen er ejendommelig ved, at det olieholdige råphosphatid ekstraheres med en lavere alkohol med 1-3 carbonato-mer, og den opnåede olieholdige phosphatidekstrat umiddelbart behandles med aluminiumoxidet.
Når henses til den kendte teknik kunne det ikke forudses, at en olieholdig alkoholisk phosphatidopløsning uden videre forbehandling er egnet til en chromatographisk separering på aluminiumoxid, hvorved opløsningens høje olieindhold på ingen måde påvirker adsorptionen af de sure phosphatider. Denne opdagelse er endnu mere overraskende, når man.tager olies ringe opløselighed i alkohol i betragtning, idet man måtte forvente, at der- under kontakten med aluminiumoxidet ville ske en udskilning af olie fra phosphatidfasen og dermed . opstå en blokering af det adsorberende stofs aktive centre. Man måtte også antage, at adsorptionen af phosphatidylcholin fra den alkoholiske opløsning ikke ville finde sted i tilstrækkelig omfang, da der ved de kendte fremgangsmåder blev elueret med dette opløsningsmiddel.
De ved gennemførelse af fremgangsmåden ifølge opfindelsen anvendte olieholdige alkoholiske ekstrakter kan fås ved én enkelt eller gentagende ekstraktionsbehandlinger af råphosphatideme med alkohol ved temperaturer indtil opløsningsmidlets kogepunkt hensigtsmæssigt ved 20-50°C. Separeringen af den derved dannede faseblanding kan ske ved dekantering eller centrifugering. Efter omrøring af den alkoholiske overfase med aluminiumoxid omrøres det fra opløsningen adskilte adsorberende stof . så med ethanol i nogle minutter og adskilles derpå i skilletragt. Udvaskningen med lidt ethanol kan gentages endnu engang, og aluminiumox= idet indeholdende uønskede følgelipider bliver enten kastet bort eller oparbejdet. I tilfældet med anvendelsen af en chromatographisøjle til adskillensen af de sure phospholipider fås de olieholdige phosphati= dylinkoncentrater. ved elueringen af søjlen med alkohol hensigtsmæs- sigt også med ethylalkohol. Derved kan man.om nødvendigt ved fraktionering af det fra søjlen opnåede eluat endnu i givet fald opnå en reduktion af olieindholdet i phosphatidylcholinfraktionen.
148111 5 I begge tilfælde såvel efter omrøringen med aluminiumoxLd som efter søjlekromatografi befries de ved fremgangsmåden ifølge opfindelsen opnåede alkoholiske opløsninger eventuelt efter filtrering med en ringe mængde celite som filterhjælpemiddel i vakuum under en ind-aktiv beskyttelsesgas for opløsningsmidlet. Derved fås en meget ren olieholdig fraktion af naturlig phosphatidylcholin,som er kep= halinfri eller meget kephalinfattig med et indhold af essentielle fedtsyrer på 66 til over 70%.
Til extrationen af de olieholdige råphosphatider og til gennemførelse af den kromatografiske fremgangsmåde ifølge opfindelsen egner sig de lavere alkoholer med 1-3 carbonatomer fortrinsvis ethylalkohol også i blanding med små mængder vand ved et indhold af 85-96% alkohol, hvorved sidsnævnte koncentration som handelsprodukt er: særlig fordelagtig.
Som adsorberende stof kan anvendes basiske og neutrale aluminiumoxider fortrinsvis imidlertid basiske med aktivitetstrin 1-3 og ifølge Brock-mann standardiserede aluminiumoxider.
Fremgangsmåden ifølge opfindelsen gør det nu muligt at befri Olieholdige phosphatidfraktioner for kephalin og inositphosphatid ved en enkelt behandling med aluminiumoxid enten på en krcmatografisøjle eller ved omrøring under bevarelse af oliefasen og i kun ét fremstillings^· trin at fremstille olieholdige phosphatidylcholinfraktioner med et højt indhold af essentielle fedtsyrer.
De ved fremgangsmåden ifølge opfindelsen fremstillede produkter besidder på grund af deres høje phosphatidylcholinandel på mere end 90% af phosphatidfraktionen,ved et indhold på over 70% af essentielle fedtsyrer, inden for rammerne af den samlede fedtsyremængde,ved oral= applikation allerede i lav dosering af 2g pr. dag terapeutisk ved mange sygdomsbilleder såsom arteriesklerose, diabetes eller hyper= lipidæmien en værdifuld stofskiftevirkning, som f.eks. beror på sænkningen af sygdomsforhøjede blodfedtvsrdier.
De ved fremgangsmåden ifølge opfindelsen af sædvanlige råphosphatider udvundne kraftigt rensede olieholdige phosphatidylcholinfraktioner har typisk følgende analytiske sammensætning: - 6 148111

Fremgangsmåden ifølge opfindelsen belyses nærmere ved hjælp af de følgende eksempler.
Eksempel 1 a) Fremstilling af den olieholdige ethanoliske solsikkephosphatid= opløsning. · 1 kg solsikkephosphatid med et olieindhold på 35% omrøres med 5 liter ethanol i en time i en nitrogenatmosfære ved 40 C. Efter 1 times‘henstand ved stuetemperatur adskilles den specifik lette ethanolrige overfase i skilletragt fra den viskose underfase.
148111 7 b) Behandling med A120^ 1,5 1 af den ifølge a) udvundne ovérfase med et stofindhold på 75g og 30g solsikkeolie omrøres kraftigt med 300g Al20^ i 2,5 time ved stuetemperatur under N.2-beskyttelse. Efter bundfældning af det adsorberende stof dekanteres ethanolopløsningen fra, og A120^ eftervaskes to gange hver gang med 150 ml ethanol. De forenede ethanolopløsninger filtreres efter irøring af lg celite som filterhjælpestof, og opløsningsmidlet afdestilleres i vakuum under nitrogen ved en badtemperatur på 40°C.
Der fås 45g af en tyktflydende olieholdig phosphatidylcholinfraktion, som er praktisk taget kephalinfri og har følgende sammensætning:

Eksempel 2 1 liter af en som under eksempel la) beskrevet af sojabønneråphos= phatid ved ethanolekstraktion fremstillet overfase med et stofindhold på 53g, som indeholder 27$ sojaolie, omrøres kraftigt med 280g AlgO^ i fire timer i en inert gasatmosfære. Efter bundfældning af det adsor= berende stof fraskilles ethanolopløsningen, ΑΙ,,Ο^ eftervaskes to gange, hver gang med 150ml ethanol, og de forenede ethanoliske ekstrakter filtreres klare eventuelt tander tilsætning af celite. Efter afdampning af opløsningsmidlet i vakuum under en nitrogenbeskyttelsesgas fås 33g af en blød plastisk gulfarvet kephalinfri phosphitidylcholinfraktion, som ved tilsætning af olie eller partialglycerider med over 50$ umættede fedtsyrer kan overføres i en hældbar form og har følgende sammensætning:
