Description
- Die vorliegende Erfindung betrifft neue Morphin-Verbindungen, Verfahren zu ihrer Herstellung und ihre Verwendung in der Medizin.
- Bis jetzt wurde die Existenz von mindestens drei Populationen von Opioid-Rezeptoren (mu, delta und kappa) nachgewiesen und beschrieben und alle drei scheinen im zentralen und peripheren Nervensystem von vielen Arten, einschließlich des Menschen vorhanden zu sein (Lord J. A. H. et al., Nature, 1997, 267, 495).
- Eine Aktivierung von allen drei Opioid-Rezeptor-Subtypen kann in Tiermodellen zu Antinozizeption führen. Im Besonderen haben Studien mit peptidischen delta-Agonsiten gezeigt, dass eine Aktivierung des delta-Rezeptors in Nagetieren und in Primaten Antinozizeption hervorrufen kann, und dass beim Menschen klinische Analgesie ausgelöst werden kann (D. E. Moulin et al., Pain, 1985, 23, 213). Es gibt Anzeichen dafür, dass delta-Agonisten eine niedrigere Neigung aufweisen, die normalerweise bei mu- und kappa-Aktivierung auftretenden Nebenwirkungen zu zeigen (Galligan et al., J. Pharm. Exp. Ther., 1984, 229, 641).
- P. S. Portoghese et al., J. Med. Chem., 1988, 31, 836-841 offenbart Binalthorphimin-verwandte zweiwertige Liganden und ihre Aktivität als kappa-Opioid-Rezeptor-Antagonisten.
- P. S. Portoghese et al., J. Med. Chem., 1991, 34, 1715-1720 offenbart peptidomimetsische delta-Opioid-Rezeptor-Antagonisten, die mit Naltrindol verwandt sind.
- US 5,223,507 und US 5,225,417 (G. D. Searle & Co.) offenbaren bicyclisch kondensierte Morphin-Verbindungen, von denen berichtet wird, dass sie delta-Opioid-Agonisten sind, die als Analgetika therapeutischen Nutzen haben.
- WO 94/07896 (Toray Ind. Inc.) offenbart Indol-kondensierte Morphin-Verbindungen, die als Immunsuppressiva, antiallergische und antiinflammatorische Mittel hilfreich sind.
- Wir haben nun eine neue Klasse von substituierten mono-heterocyclisch kondensierten Morphin- Derivaten entdeckt, die wirkungsvolle und selektive delta-Opioid-Agonisten und -Antagonisten sind und welche deshalb von potentiellem therapeutischem Nutzen als Analgetika, Immunsuppressiva, die die Abstoßung eines Organtransplantats und transplantierten Hautgewebes verhindern, antiallergische und antiinflammatorische Mittel, Neuronprotektiva, Mittel zur Behandlung von Drogen- und Alkoholmissbrauch, Gastritis, Diarrhöe, kardiovaskulären und respiratorischen Erkrankungen, Husten, Geisteskrankheit, Epilepsie und, im Allgemeinen, zur Behandlung von jenen pathologischen Zuständen, die üblicherweise mit Agonisten und Antagonisten des delta-Opioid-Rezeptors behandelt werden können, sind.
- Die vorliegende Erfindung stellt eine Verbindung, oder ein Solvat oder Salz davon, der Formel (I) bereit:

- Beispiele für R&sub4; und R&sub5; sind ein Wasserstoffatom, eine Hydroxygruppe, eine Acetyloxygruppe, eine Methoxygruppe, eine O-Phenylgruppe oder R&sub4; und R&sub5; bilden zusammen eine Sauerstoffgruppe, oder R&sub4; bildet zusammen mit R&sub3; eine Methylendioxygruppe.
- Ein Beispiel für R&sub8; ist ein Wasserstoffatom.
- Besonders bevorzugte Verbindungen nach Formel (I) sind jene, bei denen R&sub6; CONEt&sub2;, CON(i-Pr)&sub2; oder COO-i-Bu, R&sub7; eine Methylgruppe und R&sub9; ein Wasserstoffatom, eine Methylgruppe oder CH&sub2;COOH ist.
- Die Verbindungen nach Formel (I) oder ihre Salze oder Solvate liegen bevorzugt in pharmazeutisch verträglicher oder im Wesentlichen reiner Form vor. Pharmazeutisch verträgliche Form bedeutet, unter anderem, einen pharmazeutisch verträglichen Reinheitsgrad, bei dem normale pharmazeutische Zusatzstoffe, wie zum Beispiel Verdünnungsmittel und Träger unberücksichtigt bleiben und die kein Material einschließt, das bei normalem Dosierungsgrad als giftig angesehen wird.
- Eine im Wesentlichen reine Form schließt im Allgemeinen mindestens 50% (ausschließlich die normalen pharmazeutischen Zusatzstoffe), bevorzugt 75%, stärker bevorzugt 90% und noch stärker bevorzugt 95% der Verbindung der Formel (I) oder deren Salz oder Solvat ein.
- Eine bevorzugte pharmazeutisch verträgliche Form ist die kristalline Form, einschließlich einer solchen Form in einem Arzneimittel. Im Falle von Salzen und Solvaten müssen auch die zusätzlichen Ionen- und Lösungsmitteleinheiten ungiftig sein.
- Beispiele von pharmazeutisch verträglichen Salzen einer Verbindung der Formel (I) schließen Säureadditionssalze mit herkömmlichen pharmazeutischen Säuren, wie zum Beispiel Maleinsäure, Chlorwasserstoffsäure, Bromwasserstoffsäure, Phosphorsäure, Essigsäure, Fumarsäure, Salicylsäure, Zitronensäure, Milchsäure, Mandelsäure, Weinsäure, Bernsteinsäure, Benzoesäure, Ascorbinsäure und Methansulfonsäure ein.
- Die Verbindungen der Formel (I) können in mehr als einer stereoisomeren Form vorkommen, und die Erfindung erstreckt sich auf alle diese Formen und auch auf Gemische davon, einschließlich Racemate.
- Die Erfindung stellt auch ein Verfahren zur Herstellung einer Verbindung der Formel (I), oder eines Solvats oder Salzes davon bereit, umfassend Kondensierung einer Verbindung der Formel (a), wobei K ein Wasserstoffatom, ein Bromatom, COR&sub7;, =CHOH oder =NOH ist, mit einer Verbindung der Formel (b), wobei Q COR&sub7;, CHClR&sub7;, COR&sub7;, SH oder NH&sub2; ist und J =NNHPh, =O, =H&sub2; oder =CHR&sub7; ist, wobei R&sub7; und R&sub6; die vorstehende Bedeutung haben.
- und gegebenenfalls anschließend Umwandlung der Verbindung der Formel (I) in ein Solvat oder Salz davon.
- Bevorzugte Reaktionsbedingungen für K = H, Q = COR&sub7; und J = = NNHPh sind AcOH/Zn in Gegenwart von AcONa bei einer Temperatur in einem Bereich von 60-100ºC;
- Bevorzugte Reaktionsbedingungen für K = COR&sub7;, Q = SH und J = H&sub2; sind i) trockenes HCl in alkoholischem Medium bei RT, ii) starke Base, zum Beispiel MeONa in MeOH.
- Bevorzugte Reaktionsbedingungen für K = H, Q = CHClR&sub7; und J = O sind NaH in THF.
- Die Verbindungen nach Formel (I), oder Salze oder Solvate davon, können nach Verfahren, wie sie in den folgenden allgemeinen Reaktionsschemata aufgezeigt sind, oder durch Abänderungen davon, unter Verwendung von leicht verfügbaren Ausgangsmaterialien, Reagenzien und herkömmlichen synthetischen Verfahren, hergestellt werden. Wenn ein besonderes Enantiomer einer Verbindung der vorliegenden Erfindung gewünscht ist, kann es ausgehend vom gewünschten Enantiomer des Ausgangsmaterials und unter Verwendung von Reaktionen, die keine Racemisierungsprozesse zulassen, synthetisiert werden, oder es kann durch chirale Synthese oder durch Abtrennung mit einem chiralen Hilfsstoff, wobei das daraus resultierende diastereomere Gemisch aufgetrennt und der Hilfsstoff abgespalten wird, um die reinen, gewünschten Enantiomere zu erhalten, hergestellt werden. In einer anderen Ausführungsform, bei der das Molekül eine basische funktionelle Gruppe, wie zum Beispiel eine Aminogruppe oder eine saure funktionelle Gruppe, wie zum Beispiel Carboxygruppe, enthält, werden diastereomere Salze mit einer geeigneten optisch aktiven Säure oder Base gebildet, gefolgt von der Auftrennung der diastereomeren Salze durch fraktionierte Kristallisation und darauffolgender Freisetzung der reinen Enantiomere.

- Verbindungen der allgemeinen Formel (I), wobei R&sub6; = CH&sub2;WA ist, können aus Verbindungen der allgemeinen Formel (I) nach herkömmlichen chemischen Umsetzungen, die in der Literatur für R&sub6;-Reste wie Esteramide und Thioamide beschrieben sind, hergestellt werden.
- Die Verbindungen nach Formel (I) können in ihre pharmazeutisch verträglichen Salze, durch Umsetzung mit geeigneten organischen Säuren oder Mineralsäuren, umgewandelt werden. Solvate der Verbindungen der Formel (I) können durch Kristallisation oder Umkristallisation in einem geeigneten Lösungsmittel gebildet werden. Zum Beispiel können Hydrate durch Kristallisation oder Umkristallisation in wässrigen Lösungen oder in Lösungen in organischen Lösungsmitteln, die Wasser enthalten, gebildet werden.
- Auch können Salze oder Solvate von Verbindungen der Formel (I), die nicht pharmazeutisch verträglich sind, hilfreiche Intermediate bei der Herstellung von pharmazeutisch verträglichen Salzen oder Solvaten sein. Dementsprechend sind auch solche Salze oder Solvate Teil dieser Erfindung.
- Im Allgemeinen können Verbindungen der Formel (I), die als selektive delta-Rezeptor-Liganden wirken, hilfreich als Analgetika, Immunsuppressiva, die die Abstoßung eines Organtransplantats und transplantierten Hautgewebes verhindern, antiallergische und antiinflammatorische Mittel, Neuronprotektiva, zur Behandlung von Drogen- und Alkoholmissbrauch, zur Verringerung der Magensekretion, zur Behandlung von Diarrhöe, kardiovaskulären und respiratorischen Erkrankungen, Husten und Atemdepression, Geisteskrankheit, epileptischen Anfällen und anderen neurologischen Störungen (die im Folgenden als 'Zustände' bezeichnet werden) sein. Insbesondere zeigt die Aktivität der Verbindungen der Formel (I) als delta-Agonisten in Standard-Testverfahren, dass diese potentiellen therapeutischen Nutzen als Analgetika zur Milderung oder Unterdrückung von Schmerzen haben.
- Dementsprechend stellt die vorliegende Erfindung auch eine Verbindung der Formel (I), oder ein pharmazeutisch verträgliches Salz oder Solvat davon, zur Verwendung als therapeutisch wirksamen Stoff bereit.
- Die vorliegende Erfindung stellt weiterhin ein Arzneimittel bereit, umfassend eine Verbindung der Formel (I), oder ein pharmazeutisch verträgliches Salz oder Solvat davon, und einen pharmazeutisch verträglichen Träger.
- Die vorliegende Erfindung stellt auch die Verwendung einer Verbindung der Formel (I), oder ein pharmazeutisch verträgliches Salz oder Solvat davon, für die Herstellung eines Medikaments zur Behandlung der Zustände bereit.
- Ein Medikament und eine Zusammensetzung der vorliegenden Erfindung kann durch Vermischung einer Verbindung der Erfindung mit einem geeigneten Träger hergestellt werden. Es/Sie kann ein Verdünnungsmittel, ein Bindemittel, einen Füllstoff, ein Sprengmittel, einen Geschmackstoff, ein farbgebendes Mittel, ein Gleitmittel oder einen Konservierungsstoff in herkömmlicher Weise enthalten.
- Diese herkömmlichen Exzipienten können zum Beispiel bei der Herstellung von Zusammensetzungen von bekannten Mitteln zur Behandlung der Zustände verwendet werden.
- Bevorzugt liegt ein Arzneimittel der Erfindung in der Form von Einzeldosierungen vor und es ist zur Verwendung im medizinischen oder tiermedizinischen Bereich aufbereitet. Zum Beispiel können solche Formulierungen, in abgepackter Form zusammen mit einem geschriebenen oder gedruckten Verwendungshinweis, als ein Mittel zur Behandlung der Zustände vorliegen.
- Der geeignete Dosierungsbereich der Verbindungen der Erfindung hängt von der Verbindung, die verwendet wird, und dem Zustand des Patienten ab. Er wird unter anderem auch von der Beziehung der Wirksamkeit der Verbindung zur Absorptionsfähigkeit der Verbindung und der Häufigkeit und dem Weg der Verabreichung abhängen.
- Die Verbindung oder Zusammensetzung der Erfindung kann für jede Art der Verabreichung formuliert werden, und ist bevorzugt in der Form von Einzeldosierungen oder in einer Form, in der sich der menschliche Patient die Einzeldosierungen selber verabreichen kann. Vorteilhafterweise ist die Zusammensetzung zur oralen, rektalen, topischen, parenteralen, intravenösen oder intramuskulären Verabreichung geeignet. Die Formulierungen können so aufgebaut sein, dass sie eine verlangsamte Freisetzung des aktiven Bestandteils bewirken.
- Zusammensetzungen können zum Beispiel in der Form von Tabletten, Kapseln, Beuteln, Phiolen, Pulvern, Granula, Pastillen, rekonstituierbaren Pulvern oder flüssigen Formulierungen, wie zum Beispiel Lösungen oder Suspensionen, oder Zäpfchen sein.
- Die Zusammensetzungen, zum Beispiel jene, die zur oralen Verabreichung geeignet sind, können herkömmliche Exzipienten, wie Bindemittel, zum Beispiel Sirup, Gummi arabicum, Gelatine, Sorbitol, Tragant oder Polyvinylpyrrolidon; Füllstoffe, wie zum Beispiel Laktose, Zucker, Maisstärke, Calciumphosphat, Sorbitol oder Glycin; Tablettierungsgleitmittel, wie zum Beispiel Magnesiumstearat; Sprengmittel, wie zum Beispiel Stärke, Polyvinylpyrrolidon, Natriumstärkeglykolat oder mikrokristalline Cellulose; oder pharmazeutisch verträgliche Härtemittel, wie Natriumlaurylsulfat beinhalten.
- Feste Zusammensetzungen können über herkömmliche Verfahren von Mischen, Abfüllen, Tablettieren oder Ähnlichen erhalten werden. Wenn große Mengen von Füllstoffen verwendet werden, kann das aktive Mittel in diesen Zusammensetzungen durch sich wiederholende Mischungsvorgänge verteilt werden. Wenn die Zusammensetzung in der Form einer Tablette, eines Pulvers oder einer Pastille ist, kann jeder Träger, der zur Formulierung von festen Arzneimitteln geeignet ist, verwendet werden, wie zum Beispiel Magnesiumstearat, Stärke, Glukose, Laktose, Saccharose, Reismehl und Kreide. Tabletten können nach Verfahren, die aus dem Stand der Technik bekannt sind, beschichtet sein, insbesondere können sie einen Magensaft-resistenten Überzug aufweisen. Die Zusammensetzung kann auch in der Form einer ingestiven Kapsel, zum Beispiel aus Gelatine, welches die Verbindung enthält, sein und, wenn gewünscht, zusätzlich einen Träger oder andere Exzipienten umfassen.
- Zusammensetzungen zur oralen Verabreichung als Flüssigkeiten können in der Form von zum Beispiel Emulsionen, Sirupen oder Elexieren sein oder können als trockenes Produkt zur Verfügung gestellt werden, das mit Wasser oder anderen geeigneten Vehikeln vor der Verwendung zubereitet wird. Solche flüssigen Zusammensetzungen können herkömmliche Zusatzstoffe, wie zum Beispiel suspendierende Mittel, wie zum Beispiel Sorbitol, Sirup, Methylcellulose, Gelatine, Hydroxyethylcellulose, Carboxymethylcellulose, Aluminiumstearatgel, hydrierte Speisefette; Emulsionsmittel, wie zum Beispiel Lecithin, Sorbitanmonooleat oder Gummi arabicum; wässrige oder nicht-wässrige Vehikel, die Speiseöle, wie zum Beispiel Mandelöl, fraktioniertes Kokusnussöl, ölige Ester, wie zum Beispiel Ester von Glycerin oder Propylenglykol oder Ethylalkohol, Glycerin, Wasser oder normale Salzlösung einschließen; Konservierungsstoffe, wie zum Beispiel Methyl- oder Propyl-p-hydroxybenzoat oder Sorbinsäure; und, wenn gewünscht, herkömmliche Geschmacks- und farbgebende Mittel umfassen.
- Die Verbindungen der vorliegenden Erfindung können auch über nicht-orale Wege verabreicht werden. Im Einklang mit allgemein gebräuchlichen pharmazeutischen Verfahren können die Zusammensetzungen zum Beispiel zur rektalen Verabreichung als ein Zäpfchen formuliert sein.
- Sie können auch in einer injizierbaren Form in einer wässrigen oder nicht-wässrigen Lösung, Suspension oder Emulsion, in einer pharmazeutisch verträglichen Flüssigkeit, zum Beispiel in sterilem pyrogenfreiem Wasser oder einem parenteral verträglichem Öl oder einem Gemisch von Flüssigkeiten, formuliert sein. Die Flüssigkeit kann bakteriostatische Mittel, Antioxidantien oder andere Konservierungsstoffe, Puffer oder gelöste Stoffe, die die Lösung isotonisch mit Blut machen, Verdickungsmittel, suspendierende Mittel oder andere pharmazeutisch verträgliche Zusatzstoffe enthalten. Solche Formulierungen werden in der Form von Einzeldosierungen, wie zum Beispiel in Ampullen oder Injektionsvorrichtungen für den einmaligen Gebrauch, oder in der Form von Mehrfachdosierungen, wie zum Beispiel in einer Flasche, aus der die angemessene Dosis entnommen werden kann oder in einer festen Form oder einem Konzentrat, die/das zur Herstellung einer injizierbaren Formulierung verwendet werden kann, bereitgestellt.
- Die Verbindungen der vorliegenden Erfindung können auch über Inhalation, über den nasalen oder oralen Weg, verabreicht werden. Solch eine Verabreichung kann über eine Formulierung als Spray, umfassend eine Verbindung der Erfindung und einen geeigneten Träger, gegebenenfalls suspendiert in zum Beispiel einem Kohlenwasserstoff-Treibmittel, durchgeführt werden.
- Bevorzugte Spray-Formulierungen umfassen feinstzerkleinerte Verbindungspartikel zusammen mit einem oberflächenaktiven Mittel, einem Lösungsmittel oder einem dispergierendem Mittel, um die Sedimentation der suspendierten Partikel zu verhindern. Bevorzugterweise beträgt die Partikelgröße der Verbindung etwa 2 bis 10 Mikrometer.
- Eine weitere Art der Verabreichung der Verbindungen der Erfindung umfasst transdermale Abgabe, unter Verwendung einer Formulierung in der Form eines Hautpflasters. Eine bevorzugte Formulierung umfasst eine Verbindung der Erfindung, dispergiert in einem Haftklebstoff, der an der Haut klebt, wobei die Verbindung durch Diffusion aus dem Klebstoff durch die Haut an den Patienten abgegeben wird. Für eine konstante perkutane Absorptionsgeschwindigkeit können Haftklebstoffe, wie sie nach dem Stand der Technik bekannt sind, verwendet werden, wie zum Beispiel natürlicher Kautschuk oder Silikon.
- Wie oben erwähnt, hängt die wirkungsvolle Dosis der Verbindung von der speziellen Verbindung, die verabreicht wird, dem Zustand des Patienten und der Häufigkeit und dem Weg der Verabreichung ab. Normalerweise wird eine Einzeldosis 20 bis 1000 mg und bevorzugt 30 bis 500 mg, insbesondere 50, 100, 150, 200, 250, 300, 350, 400, 450 oder 500 mg umfassen. Die Zusammensetzung kann einmal oder mehrmals am Tag, zum Beispiel zweimal, dreimal oder viermal täglich verabreicht werden und die tägliche Gesamtdosis für einen 70 kg schweren Erwachsenen wird normalerweise im Bereich von 100 bis 3000 mg liegen. In einer anderen Ausführungsform wird die Einzeldosis 2 bis 20 mg aktiven Bestandteil beinhalten und mehrfach verabreicht werden, um, wenn gewünscht, die vorherbestimmte tägliche Dosis zu erreichen.
- Von den Verbindungen der Erfindung werden keine toxikologischen Effekte erwartet, wenn sie gemäß der Erfindung verabreicht werden.
- Die vorliegende Erfindung stellt auch ein Verfahren zur Behandlung und/oder Vorbeugung der Zustände bei Säugern, insbesondere beim Menschen, bereit, das die Verabreichung einer wirksamen Menge einer Verbindung der Formel (I), oder eines pharmazeutisch verträglichen Salzes oder Solvates davon, an einen Säuger, der einer solchen Behandlung oder Vorbeugung bedarf, umfasst.
- Die Aktivität der Verbindungen der vorliegenden Erfindung als selektive delta-Liganden wurde über Radioligand-Bindungstests, wie im Folgenden beschrieben, bestimmt.
- Die Gehirnmembrane von Mäusen wurden, wie von Kosterlitz (Br. J. Pharmacol., 1981, 73, 939) beschrieben, aufbereitet. Die Bindung des bevorzugten delta-Liganden [³H]-[D-Ala²,D-Leu&sup5;]- enkephalin (DADLE) wurde bei seiner KD-Konzentration (1,3 nM) in Gegenwart von 40 nM des unmarkierten mu-Liganden [D-Ala²,MePhe&sup4;,Gly-ol&sup5;]-enkephalin (DAMGO) untersucht. Die Bindung des mu-Liganden [³H]-DAMGO (Eur. J. Pharmacol., 1989, 166, 213) und des kappa- Liganden [³H]-U69593 (Excerpta Medica, 1990, 211) wurde bei 0,5 nM durchgeführt. Die nicht spezifische Bindung wurde für alle drei Tritium-markierte Liganden in Gegenwart von Naloxon (10 uM) bestimmt. Die Bindungsdaten wurden als Prozent der Hemmung angegeben und über folgende Gleichung gefittet: f(x) = 100·X/(IC&sub5;&sub0; + X), wobei X die Konzentrationswerte des 'kalten' Arzneimittels sind. Die erhaltenen IC&sub5;&sub0;-Werte wurden zur Berechnung der Inhibitorkonstanten (Ki), gemäß der Beziehung nach Cheng und Prusoff (Biochem. Pharmacol., 1973, 22, 3099) verwendet.
- Die delta-Agonist/Antagonist-Aktivität der Verbindungen der vorliegenden Erfindung wurde in einem cAMP-Biotest an der NG108-15-Zelllinie, wie unten beschrieben, bestimmt.
- Die NG108-15-Zellen lies man bei 37ºC in wasserdampfgesättigter Atmosphäre von 5% CO&sub2; und 95% Luft in DMEM (ohne Natriumpyruvat, unter Verwendung von 4500 mg/l Glukose), dem 10% fötales Kälberserum, das 2 mM Glutamin, 2% HATx50-Zusatz, 50 ug Streptomycin und 50 I.U. Penicillin pro ml enthielt, zugesetzt wurde, wachsen und die konfluenten Zellen wurden mit 1 M EDTA in Ca/Mg-freier, phosphatgepufferter Salzlösung unter mechanischem Rühren geerntet. Das Medium wurde jeden zweiten Tag ausgetauscht. Einen Tag vor dem Experiment wurden die Zellen in 17-mm-Kulturplatten (etwa 10 · 10&sup6; Zellen/Platte) ausgesät. Einen Tag danach wurde das Wachstumsmedium entfernt und die Zellen wurden zweimal mit einem modifiziertem Krebs-Ringer-Medium, das mit 200 mM hepes-NaOH, pH 7,4, das NaCl (125), KCl (5), KH&sub2;PO&sub4; (0,4), MgSO&sub4; und CaCl&sub2; (1,2), NaHCO&sub3; (25) und Glukose (12) enthielt (mmol/l), gepuffert wurde, gewaschen. Das Inkubationsmedium enthielt zusätzlich 1 mM 3-Isobutyl-1-methylxantin (IBMX). Die Versuche wurden bei Raumtemperatur durchgeführt. Nach einer Inkubation von 10 Minuten, bei denen IBMX einwirken konnte, wurden die NG108-15-Zellen für 10 Minuten 1 uM Forskolin und der zu testenden Verbindung ausgesetzt. Die Reaktion wurde durch Zusatz von kalter 0,4 N HClO&sub4; abgestoppt. Nach 15 Minuten wurde der kalte Überstand vorsichtig abgetrennt und unter Verwendung von 1 M K&sub2;CO&sub3; neutralisiert. Nach einer Inkubation bei 4ºC über Nacht wurden die Röhrchen bei 9000 Umdrehungen pro Minute für 5 Minuten zentrifugiert, und in einem 100 ul-Aliquot wurde der cAMP-Gehalt unter Verwendung des im Handel erhältlichen ¹²&sup5;I-cAMP-RIA-Kits (Amersham Inc.) bestimmt. Die Pellets der Originalplatten wurden in 0,5 N NaOH gelöst und der Proteingehalt wurde über das Verfahren nach Bradford (Anal. Biochem., 1976, 72, 248) bestimmt. Die Daten wurden auf den Proteingehalt bezogen.
- Die potentesten Verbindungen, die in der vorliegenden Erfindung beschrieben werden, zeigen Affinitäten für den delta-Rezeptor im Bereich von 0,5 bis 200 nM, mit delta-Selektivitäten in einem Bereich von 20- bis 1500-fach, bezogen auf die anderen Opioid-Rezeptor-Typen. Diese Verbindungen zeigten im cAMP-Inhibitionstest auch Eigenschaften als starke delta-Agonisten oder -Antagonisten. Selektive delta-Agonisten (antagonisiert mit dem selektiven delta- Antagonisten Naltrindol) zeigten IC&sub5;&sub0;-Werte im Bereich von 1 bis 500 nM. Zum Beispiel zeigt die Verbindung von Beispiel 10 folgende Werte: delta-Ki-Wert = 2,9 nM, mu-Ki-Wert/delta-Ki-Wert = 840 und kappa-Ki-Wert/delta-Ki-Wert = 600. Die Verbindung von Beispiel 1 zeigte eine Aktivität als Agonist bei der Inhibierung von Forskolin-stimuliertem cAMP in NG108-15-Zellen (IC&sub5;&sub0; = 15 nM), die vollständig vom selektiven delta-Antagonisten Naltrindol (100 nM) antgonisiert wurde.
- Die abdominale Konstriktion der Maus (MAC) (Proc. Soc. Exp. Biol. Med., 1957, 95, 729), das Schwanz-Zucken der Maus (MTF) (J. Pharm. Exp. Ther., 1941, 72, 74) und das Schwanz- Zucken der Maus in warmen Wasser (MTF-WW) (Life Sci., 1986, 39, 1795) wurden dazu verwendet die antinozizeptive Wirksamkeit der Verbindungen der vorliegenden Erfindung zu untersuchen.
- Die folgenden Ansätze 1 bis 7 verdeutlichen das synthetische Verfahren, mit dem neue Ketone der allgemeinen Formel (II) erhalten werden können, die als solche einen Teil der vorliegenden Erfindung darstellen. Insbesondere sind 4,5-Epoxy-17-methyl-3-vinylmorphinan-6-on, 4,5-Epoxy-3-(1-ethoxyvinyl)-17-methylmorphinan-6-on, 4,5-Epoxy-3-ethyl-17- methylmorphinan-6-on, 3-Brom-4,5-epoxy-14-hydroxy-17-methylmorphinan-6-on und 3-Brom- 4,5-epoxy-17-methylmorphinan-6-on neue Verbindungen und werden als Ausgangsmaterialien zur Herstellung der Verbindungen der Beispiele 28, 30, 33, 35, 41, 52, 61 und 64 verwendet. Andere Ketone, die als Ausgangsmaterialien verwendet werden, sind in der Literatur bekannt. Ansatz 8 verdeutlicht die Herstellung eines neuen Phosphonohydrazons der allgemeinen Formel (III), das als Ausgangsmaterial für die Herstellung der Verbindung von Beispiel 89 verwendet wurde. Beispiel 1 zeigt die Herstellung von Verbindungen der allgemeinen Formel (I) der vorliegenden Erfindung, ausgehend von den entsprechenden Ketonen der allgemeinen Formel (II) und den entsprechenden bekannten Hydrazonen der allgemeinen Formel (III). Die Beispiele 2, 49 und 52 zeigen die Herstellung von Verbindungen der allgemeinen Formel (I), die der Reihe nach durch chemische Umwandlung der entsprechenden Verbindungen der Formel (I) hergestellt wurden. Beispiel 105 beschreibt die Herstellung von Verbindungen der allgemeinen Formel (I), wobei n = 0, X = S und Y ein substituiertes Kohlenstoffatom ist. Die Beispiele, die hier beschrieben werden, wurden gemäß den gleichen Verfahren, wie sie für die Beispiele 1, 2, 49, 52 und 105 beschrieben werden, hergestellt. Die Verbindungen der Beispiele 1 bis 105 sind in der Chemischen Tabelle zusammengefasst.
- Allgemeines Verfahren zur Herstellung von Verbindungen der allgemeinen Formel (II), wobei R&sub3; CH=CH&sub2; und C(OEt)=CH&sub2; ist.
- 5,5 g (19,3 mmol) 4,5-Epoxy-3-hydroxy-17-methylmorphinan-6-on wurden unter Stickstoffatmosphäre in 20 ml Pyridin gelöst. Die Lösung wurde auf 0ºC gekühlt, und 3,56 ml (21,2 mmol) Trifluormethansulfonsäureanhydrid wurden hinzugetropft. Die Lösung wurde für 5 Minuten bei 0ºC gerührt und dann lies man die Temperatur über Nacht auf Raumtemperatur ansteigen. Das Reaktionsgemisch wurde auf Wasser gegossen, und die wässrige Phase wurde mit AcOEt extrahiert. Die organische Phase wurde über Na&sub2;SO&sub4; getrocknet und das Lösungsmittel unter Vakuum abgezogen. Das rohe Reaktionsgemisch wurde über Flash-Chromatographie, durch Elution mit einem Gemisch von CH&sub2;Cl&sub2;/MeOH/konz. NH&sub4;OH 90 : 7 : 0,7 gereinigt. Die Ausbeute der Titelverbindung lag bei 6,26 g.

- 2 g (4,8 mmol) von 4,5-Epoxy-17-methyl-3-trifluormethansulfonyloxymorphinan-6-on wurden unter Stickstoffatmosphäre in 25 ml Dimethylformamid gelöst und dann mit 1,46 ml (5 mmol) Vinyltributylzinn, 1,6 g (38,4 mmol) LiCl, 0,337 g (0,48 mmol) Bis(triphenylphosphin)palladium(II)chlorid und 0,5 g (1,9 mmol) Triphenylphosphin versetzt. Das Reaktionsgemisch wurde für 3 Stunden auf 100ºC erhitzt und wurde dann auf Wasser gegossen, und die wässrige Phase wurde mit AcOEt extrahiert. Die organische Phase wurde über Na&sub2;SO&sub4; getrocknet, und das Lösungsmittel wurde unter Vakuum abgezogen. Das rohe Reaktionsgemisch wurde über Flash-Chromatographie, durch Elution mit einem Gemisch von CH&sub2;Cl&sub2;/MeOH/konz. NH&sub4;OH 90 : 7 : 0,7 gereinigt. Die Ausbeule der Titelverbindung lag bei 1,1 g.

- 2,5 g (6,0 mmol) 4,5-Epoxy-17-methyl-3-trifluormethansulfonyloxymorphinan-6-on, 2,1 ml (6,2 mmol) (1-Ethoxyvinyl)tributylzinn, 2 g (48 mmol) LiCl, 0,42 g (0,6 mmol) Bis(triphenylphosphin)palladium(II)chlorid und 0,63 g (2,4 mmol) Triphenylphosphin in 25 ml Dimethylformamid wurden wie in Ansatz 2 beschrieben behandelt. Das rohe Reaktionsgemisch wurde über Flash-Chromatographie, durch Elution mit einem Gemisch von CH&sub2;Cl&sub2;/MeOH/konz. NH&sub4;OH 90 : 7 : 0,7 gereinigt. Die Ausbeute der Titelverbindung lag bei 1,95 g.

- 1,2 g (4,06 mmol) 4,5-Epoxy-17-methyl-3-vinylmorphinan-6-on wurden in 150 ml absolutem EtOH gelöst. 1 g 10% Pd auf Aktivkohle wurde hinzugefügt, und das Reaktionsgemisch wurde in einer Parr-Einrichtung bei 35 psi und Raumtemperatur für 8 Stunden hydriert. Der Katalysator wurde abfiltriert und das Lösungsmittel unter Vakuum abgezogen. Die Ausbeute der Titelverbindung lag bei 0,77 g.

- Allgemeines Verfahren zur Herstellung von Verbindungen der allgemeinen Formel (I), wobei R&sub3; = Br ist.
- 3,1 g (11,4 mmol) 4,5-Epoxy-17-methylmorphinan-6-ol wurden in 150 ml Eisessig gelöst und 11,4 ml einer 1 M Lösung von Br&sub2; in AcOH wurden zugetropft. Nach 1 Stunde wurde AcOH unter Vakuum abgezogen, der Rückstand in Wasser aufgenommen, die wässrige Lösung mit einer gesättigten NaHCO&sub3;-Lösung auf pH 7 eingestellt und mit AcOEt extrahiert. Die organische Phase wurde über Na&sub2;SO&sub4; getrocknet und das Lösungsmittel im Vakuum abgezogen. Die Ausbeute der Titelverbindung lag bei 3,6 g.

- 1,1 g (3,8 mmol) 4,5-Epoxy-14-hydroxy-17-methylmorphinan-6-on in 50 ml Eisessig wurden mit 3,8 ml einer 1 M Lösung von Br&sub2; in AcOH so wie in Ansatz 5 beschrieben behandelt. Die Ausbeute der Titelverbindung lag bei 1,1 g.
- I.R. (KBr): 3348, 2910, 1738 cm&supmin;¹.
- Eine Lösung von 1,6 ml DMSO in 4,8 ml CH&sub2;Cl&sub2; wurde unter Stickstoffatmosphäre bei -55ºC langsam zu einer Lösung von 0,9 ml Oxalsäuredichlorid in 21 ml CH&sub2;Cl&sub2; gegeben. Nach 2 Minuten wurden 3,6 g (10,3 mmol) 3-Brom-4,5-epoxy-17-methylmorphinan-6-ol in 20 ml CH&sub2;Cl&sub2; hinzugefügt, gefolgt, nach 15 Minuten, von 6,6 ml Et&sub3;N. Innerhalb 2 Stunden lies man das Reaktionsgemisch auf Raumtemperatur kommen, und dann wurde die Reaktion mit 50 ml H&sub2;O abgestoppt. Die Phasen wurden getrennt, und die organische Phase wurde über Na&sub2;SO&sub4; getrocknet und das Lösungsmittel im Vakuum abgezogen. Das rohe Reaktionsgemisch wurde über Flash-Chromatographie, durch Elution mit einem Gemisch von CH&sub2;Cl&sub2;/MeOH/konz. NH&sub4;OH 90 : 7 : 0,7 gereinigt. Die Ausbeute der Titelverbindung lag bei 2,87 g.
- I.R. (KBr): 2940, 1716, 1446 cm&supmin;¹.

- Bei 0-10ºC wurden zu einer Lösung von 5,0 g (0,0257 mol) Diethyl(2-oxopropyl)phosphonat in 25 ml Ethanol/Wasser (4 : 1) 7,1 g (0,0517 mol) K&sub2;CO&sub3; und 0,0257 mol Phenyldiazoniumchlorid hinzugefügt. Die erhaltene Suspension wurde so lange gerührt, bis sie Raumtemperatur erreicht hatte. Dann wurden 30 ml Wasser und 100 ml CH&sub2;Cl&sub2; hinzugefügt. Die organische Phase wurde über Na&sub2;SO&sub4; getrocknet und im Vakuum eingedampft. Die Ausbeute der Titelverbindung, ein rotes Öl, lag bei 7 g und wurde so in der darauf folgenden Stufe verwendet.
- C&sub1;&sub3;H&sub1;&sub9;N&sub2;O&sub4;P
- I.R. (rein): 3498, 1716, 1666, 1268, 1026 cm&supmin;¹.
- N.M.R. 300 MHz (CDCl&sub3;): δ 12,9 (bs, 1H), 7,5-7,0 (m, 5H), 4,2 (m, 4H), 2,3 (s, 3H), 1,5 (m, 6H).
- 0,65 g (2,17 mmol) 7,8-Dihydrocodeinon Hydrochlorid und 2,2 g (6,51 mmol) N-Benzyl-N- isopropyl-2-phenylhydrazon-3-oxobutyramid wurden in einem Gemisch von 10 ml Eisessig und 0,54 g (6,51 mmol) CH&sub3;COONa gelöst. Das erhaltene Reaktionsgemisch wurde auf 60ºC erwärmt und dann wurde unter Stickstoffatmosphäre portionsweise 0,57 g (8,6 mmol) Zink- Staub zugegeben. Das Gemisch wurde für 2 Stunden am Rückfluß gekocht und dann auf Raumtemperatur abgekühlt. Die erhaltenen Salze wurden durch Dekantieren abgetrennt und mit Essigsäure gewaschen. Die sauren Lösungen wurden gesammelt und der pH-Wert auf 8 eingestellt und dann mehrmals mit AcOEt extrahiert. Die organische Phase wurde getrocknet und im Vakuum bis zur Trockene eingeengt. Der erhaltene Rückstand wurde durch Mittel- Druck-Chromatographie unter Verwendung von Kieselgel (15-25 u) und einem Gemisch von AcOEt/MeOH/NH&sub4;OH konz. 90 : 10 : 0,5 als Eluent gereinigt. Das Produkt wurde in einem Gemisch von Aceton/MeOH 1 : 1 aufgenommen, und die Lösung wurde mit HCl/Et&sub2;O angesäuert. Der erhaltene Niederschlag wurde abfiltriert, gewaschen und getrocknet. Die Ausbeute der Titelverbindung lag bei 0,5 g.
- M.P. = 304ºC Zersetzung.
- 1 g (2,3 mmol) [8R-(4bS*,8a,8aβ,12bβ)]-11-Diethylaminocarbonyl-7,10-dimethyl-1-methoxy- 5,6,7,8,12,12b-hexahydro-(9H)-4,8-methanobenzofuro[3,2-e]pyrrolo[2,3-g]isochinolin und 0,950 g (2,3 mmol) Lawesson-Reagenz wurden in 40 ml Toluol gelöst, und das Gemisch wurde für 4 Stunden am Rückfluss gekocht. Das Lösungsmittel wurde im Vakuum abgezogen, und der Rückstand wurde dann in CH&sub2;Cl&sub2; aufgenommen und mit gesättigter NaHCO&sub3;-Lösung gewaschen. Die organische Phase wurde über Na&sub2;SO&sub4; getrocknet. Nach dem Abdampfen des Lösungsmittels wurde der Rückstand durch Flash-Chromatographie an Kieselgel (CH&sub2;CL&sub2;/MeOH/NH&sub4;OH konz. 86 : 10 : 0,6) gereinigt, und das Produkt wurde dann in Aceton gelöst. Die erhaltene Lösung wurde mit HCl/Et&sub2;O angesäuert. Der Niederschlag wurde abfiltriert und in kochendem MeOH innerhalb 30 Minuten in Gegenwart von Aktivkohle gelöst. Die Aktivkohle wurde abfiltriert und die Lösung zur Trockene eingeengt. Der Rückstand wurde in kochendem Et&sub2;O zerkleinert, wodurch eine Ausbeute von 0,41 g der Titelverbindung erhalten wurde.
- M.P. > 250ºC.
- Die Verbindung von Beispiel 73 wurde nach dem gleichen Verfahren, wie oben beschrieben, hergestellt.
- 0,52 g (1,2 mmol) [8R-(4bS*,8α,8aβ,12bβ)]-11-Isopropylcarbonyl-7,10-dimethyl-1-methoxy- 5,6,7,8,12,12b-hexahydro-(9H)-4,8-methanobenzofuro[3,2-e]pyrrolo[2,3-g]isochinolin (Beispiel 34) wurden unter Stickstoffatmosphäre in 5 ml DMF gelöst. 0,051 g NaH (60%) wurden portionsweise hinzugefügt, wobei die Temperatur bei 0ºC gehalten wurde. Nach 30 Minuten wurde eine Lösung von 0,2 g MeI in 1 ml DMF dazugetropft. Die Reaktion wurde nach 1 Stunde durch Zugabe von zerstoßenem Eis abgestoppt. Die erhaltene Lösung wurde erschöpfend mit Et&sub2;O extrahiert. Die organischen Phasen wurden über Na&sub2;SO&sub4; getrocknet, und das Lösungsmittel wurde dann im Vakuum abgezogen. Der erhaltene Rückstand wurde in Aceton gelöst und die Lösung mit Et&sub2;O/HCl angesäuert. Das Lösungsmittel wurde wieder abgezogen und der erhaltene Rückstand in kochendem Et&sub2;O zerkleinert. Der Feststoff wurde abfiltriert, gewaschen und getrocknet. Die Ausbeute der Titelverbindung lag bei 0,25 g.
- M.P. 240-243ºC.
- Die Verbindungen der Beispiele 62 und 82 wurden nach dem gleichen Verfahren, wie oben beschrieben, hergestellt.
- Die Verbindungen der Beispiele 74, 79 und 80 wurden nach dem gleichen Verfahren, unter Verwendung von Ethylbromacetat als Alkylierungsmittel, hergestellt. Die erhaltenen Ethylester wurden unter sauren Bedingungen zu den entsprechenden Säurederivaten der obigen Beispiele hydrolysiert.
- 0,3 g (0,58 mmol) [8R-(4bS*,8α,8aβ,12bβ)]-1-Brom-11-diisopropylaminocarbonyl-7,10- dimethyl-5,6,7,8,12,12b-hexahydro-(9H)-4,8-methanobenzofuro[3,2-e]pyrrolo[2,3-g]isochinolin (Beispiel 30) wurden unter Stickstoffatmosphäre in 10 ml Eisessig gelöst, und es wurden 0,15 g (1,8 mmol) AcONa zugegeben. Das Reaktionsgemisch wurde auf 80ºC erwärmt und 0,23 g (3,5 mmol) Zink-Staub wurden portionsweise zugegeben. Das Reaktionsgemisch wurde unter Rückfluss 4 Stunden lang erwärmt, dann wurde auf Eis gegossen, der pH-Wert mit konz. NH&sub4;OH auf 9 eingestellt und mit CH&sub2;Cl&sub2; extrahiert. Die organische Phase wurde über Na&sub2;SO&sub4; getrocknet und das Lösungsmittel im Vakuum abgezogen. Das rohe Reaktionsgemisch wurde durch Flash-Chromatographie, durch Elution mit einem Gemisch von CH&sub2;Cl&sub2;/MeOH/konz. NH&sub4;OH 90 : 7 : 0,7 gereinigt. Der erhaltene Feststoff wurde in Et&sub2;O zerkleinert. Das Titelprodukt wurde in einer Ausbeute von 0,15 g erhalten.
- Eine Lösung von 7-Acetyl-4,5-epoxy-1-methoxy-17-methylmorphinan-6-on (1 g, 2,9 mmol) (J. Med. Chem., 1982, 25, 983) in MeOH (40 mL) wurde auf -10ºC gekühlt, und in dieses System wurde ein Strom von trockenem HCl eingeleitet, bis Sättigung eintrat (etwa 1 Stunde). Dann wurde Thioglycolsäure (0,4 mL, 5,8 mmol) zugegeben, und das Einleiten von HCl bei 10ºC wurde für weitere 4 Stunden fortgesetzt. Das Gemisch wurde für 6 Tage bei Raumtemperatur stehen gelassen. Dann wurde das Lösungsmittel im Vakuum abgezogen, der Rückstand mit konz. NH&sub4;OH behandelt und mit AcOEt extrahiert. Die organische Phase wurde abgetrennt, mit Wasser gewaschen, getrocknet (Na&sub2;SO&sub4;) und im Vakuum eingeengt. Das Rohprodukt wurde so direkt in der nächsten Stufe verwendet.
- Eine 2 N Lösung von MeONa (10 ml) würde zu einer Lösung des obigen Produkts in 12 ml MeOH gegeben und unter Stickstoff für 24 Stunden stehen gelassen. Das Lösungsmittel wurde abgezogen und der Rückstand mit eiskaltem Wasser behandelt. Das Gemisch wurde mit 6 N HCl angesäuert (pH = 1). Nach dem Waschen mit AcOEt wurde der pH-Wert der wässrigen Phase mit NaOH auf 9 eingestellt, und es wurde mit AcOEt extrahiert. Das Lösungsmittel wurde getrocknet, abgedampft, und das Rohprodukt wurde durch Kieselgel-Chromatographie (CH&sub2;Cl&sub2;/MeOH/konz. NH&sub4;OH 95 : 5 : 0,5) gereinigt. Das erhaltene Produkt wurde mit HCl/Et&sub2;O behandelt und es wurden 55 mg Ausbeute der Titelverbindung erhalten. CHEMISCHE TABELLE

[8R-(4bS*,8α,8aβ,12bβ)]-11-Methoxycarbonyl-7,10-dimethyl-1-methoxy-
5,6,7,8,9,12b-hexahydro-4,8-methanobenzofuro-[3,2-e]thieno[2,3-g]isochinolin
Hydrochlorid.
4. Arzneimittel umfassend eine Verbindung gemäß einem der Ansprüche 1 bis 3 und einen
pharmazeutisch verträglichen Träger.
5. Verbindung gemäß einem der Ansprüche 1 bis 3 zur Verwendung als therapeutisch
wirksamer Stoff.
6. Verbindung gemäß einem der Ansprüche 1 bis 3, zur Verwendung als Analgetikum,
Immunsupressivum, um die Abstoßung eines Organstransplantats und transplantierten
Hautgewebes zu verhindern, antiallergisches und antiinflammatorisches Mittel,
Neuronprotektivum, zur Behandlung von Drogen- und Alkoholmissbrauch, zur
Verringerung der Magensekretion, zur Behandlung von Diarrhöe, kardiovaskulären und
respiratorischen Erkrankungen, Husten und Atemdepression, Geisteskrankheit,
epileptischen Anfällen und anderen neurologischen Störungen.
7. Verwendung einer Verbindung gemäß einem der Ansprüche 1 bis 3 zur Herstellung eines
Medikaments zur Verwendung als Analgetikum, Immunsupressivum, um die Abstoßung
eines Organtransplantats und transplantierten Hautgewebes zu verhindern,
antiallergisches und antiinflammatorisches Mittel, Neuronprotektivum, zur Behandlung
von Drogen- und Alkoholmissbrauch, zur Verringerung der Magensekretion, zur
Behandlung von Diarrhöe, kardiovaskulären und respiratorischen Erkrankungen, Husten
und Atemdepression, Geisteskrankheit, epileptischen Anfällen und anderen
neurologischen Störungen.
8. Verfahren zur Herstellung einer Verbindung nach Formel (I) wie in Anspruch 1 definiert
oder ein Solvat oder Salz davon, umfassend Kondensierung einer Verbindung der Formel
(a) , wobei K ein Wasserstoffatom, ein Bromatom, COR&sub7;, =CHOH oder =NOH ist, mit
einer Verbindung der Formel (b), wobei Q COR&sub7;, CHClR&sub7;, COR&sub7;, SH oder NH&sub2; ist und J
=NNHPh, =O, =H&sub2; oder =CHR&sub7; ist, wobei R&sub7; und R&sub6; wie in Anspruch 1 definiert sind
und gegebenenfalls anschließend Umwandlung der Verbindung der Formel (I) in ein
Solvat oder Salz davon.
