Description
- Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung α,β-ungesättigter Ketolactone, die als synthetische Zwischenprodukte für Prostaglandine nützlich sind.
- Während der Untersuchung verschiedener Arten von Prostaglandindenvaten wurde gefunden, daß viele Prostaglandine existieren, die das Prostansäureskelett aufweisen, worin mindestens ein Halogenatom, insbesondere ein Fluoratom, in der 16- oder 17-Stellung vorhanden ist [16(17)-Mono- oder 16,16(17,17)-Dihalogenprostaglandine], die durch die folgende Formel (α-Kette) (ω-Kette)
- dargestellt werden, die charakteristische pharmakologische Eigenschaften besitzen.
- Das Corey-Verfahren ist seit langer Zeit bekannt, und es ist noch ein typisches Syntheseverfahren für Prostaglandine.
- Das Corey-Verfahren umfaßt die Stufen der Herstellung eines α,β-ungesättigten Ketolactons (III) aus dem Corey-Lacton über den Corey-Aldehyd (II): 1,2-Dimethoxyethan (DME)
- [worin Ar eine aromatische Gruppe bedeutet).
- Das Corey-Lacton (I) wird zu dem Corey-Aldehyd (II) unter Verwendung eines Komplexes aus Pyridin und Chromtrioxid oxidiert (sogenannte Collins- Oxidation), gefolgt von einer Umsetzung zwischen dem Aldehyd und einem Anion, das durch Umsetzung von Dimethyl-(2-oxoalkyl)phosphonat mit Natriumhydrid erzeugt wurde, wobei das α,β-ungesättigte Ketolacton (III) gebildet wird.
- Es wird angenommen, daß diese Reaktionen einfach für die Synthese von 16(17)-Mono- oder 16,16(17,17)-Dihalogenprostaglandinen verwendet werden können. Wenn jedoch Dimethyl-(2-oxoalkyl)phosphonate mit ein oder zwei Habgenatomen an der 3-Stellung als Ersatz für das Phosphonat verwendet werden, nimmt die Ausbeute an dem gewünschten α,β-ungesättigten Ketolacton auf einen Wert unter 10% ab.
- Es wurden viele Versuche unternommen, um Reaktionen in Anwesenheit von Natriumhydrid und einer Kupferverbindung oder einer Thalliumverbindung durchzuführen, um die Ausbeute zu verbessern. Die Verwendung der Kupferverbindung ergibt jedoch nicht die gewünschte Verbesserung in der Ausbeute, wohingegen bei der Thalliumverbindung Verbesserungen der Ausbeute in gewissem Ausmaß erhalten werden, diese ist jedoch toxisch und sehr teuer. Es besteht daher ein Bedarf für eine weitere Verbesserung bei dieser Reaktionsstufe.
- Der vorliegenden Erfindung liegt die Aufgabe zugrunde, ein Verfahren zur Herstellung eines α,β-ungesättigten Ketolactons (III), das mit einem oder zwei Halogenatomen in der Stellung substituiert ist, welche der 16- oder 17- Stellung des Prostaglandinskeletts entspricht, wenn sich dieses von α,β-ungesättigten Ketolactonen ableitet, zur Verfügung zu stellen.
- Wie zuvor erwähnt, ist die Ausbeute an α,β-ungesättigtem Ketolacton niedrig, wenn es durch Umsetzung eines Corey-Aldehyds mit Dimethyl-(2-oxoalkyl)phosphonat, substituiert mit ein oder zwei Halogenatomen in der 3-Stellung, unter Verwendung von Natriumhydrid und einer Kupferverbindung hergestellt wird.
- Gemäß der vorliegenden Erfindung wird die Ausbeute an α,β-ungesättigtem Ketolacton verbessert, wenn anstelle einer Kupferverbindung eine Zinkverbindung verwendet wird.
- Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von α,β-ungesättigten Ketolactonen, welches die Umsetzung eines Bicyclolactonaldehyds mit Dimethyl-(2-oxoalkyl)phosphonat, welches mit mindestens einem Halogenatom in der 3-Stellung der Alkylgruppe substituiert ist, in Anwesenheit eines Alkalimetallhydrids und einer Zinkverbindung umfaßt.
- Der Bicyclolactonaldehyd (1), der ein Ausgangsmaterial für die vorliegende Erfindung ist, wird durch die folgende Formel (1)
- dargestellt [worin A eine Hydroxylschutzgruppe für Hydroxylgruppen bedeutet, beispielsweise
- (worin X&sub3; ein Wasserstoffatom oder ein Halogenatom bedeutet, R&sub4;, R&sub5; und R&sub6; eine Alkylgruppe mit C&sub1;-C&sub4;-Kohlenstoffatomen oder eine Phenylgruppe bedeuten, R&sub7; eine Alkylgruppe mit C&sub1;-C&sub4;-Kohlenstoffatomen bedeutet, die eine Alkoxyoder Silylgruppe enthalten kann, und R&sub1;&sub0; eine Alkylgruppe mit C&sub1;-C&sub4;-Kohlenstoffatomen bedeutet, und worin B eine Einfachbindung oder eine Methylengruppe -CH&sub2;- bedeutet]. Dieser Bicyclolactonaldehyd (1) kann aus dem Corey- Lacton (I) durch Oxidationsreaktion unter Verwendung von Dimethylsulfoxid (im folgenden einfach als DMSO-Oxidation bezeichnet) erhalten werden. Beispiele dieser DMSO-Oxidation sind die sogenannte Pfilzner-Moffatt-Oxidation, bei der DMSO, Dicyclohexylcarbodiimid, Trifluoressigsäure und Pyridin verwendet werden, die sogenannte Swern-Oxidation, bei der DMSO, Oxalylchlorid und Triethylamin verwendet werden, und die sogenannte Parikh-Doering-Oxidation, bei der DMSO, ein Komplex aus Schwefeltrioxid und Triethylamin verwendet werden.
- Die DMSO-Oxidation erlaubt, daß die Reaktion leicht kontrolliert werden kann und die Alkohole zu den entsprechenden Aldehyden in hohen Ausbeuten oxidiert werden können, da die gebildeten Aldehyde vor der weiteren Oxidation zu der entsprechenden Carbonsäure geschützt sind.
- Dieser Bicyclolactonaldehyd (1) kann ebenfalls durch Oxidation, einer ähnlichen Reaktion wie oben beschrieben, von Bicyclolacton, welches eine Alkylseitenkette, verlängert um eins in der Zahl der Kohlenstoffatome, hergestellt werden, indem das Corey-Lacton (I) den folgenden gezeigten Reaktionen unterworfen wird.
- Für die Kohlenstoffzahl-Erhöhungsreaktion wird zuerst eine Austrittsgruppe (beispielsweise eine Tosylgruppe) in das Corey-Lacton (1) mit einer geeigneten Schutzgruppe (beispielsweise einer 4-Phenylbenzoylgruppe) für die 7-Hydroxylgruppe eingeführt, und anschließend wird mit einer Cyanid erzeugenden Verbindung umgesetzt, wobei eine Nitrilverbindung erhalten wird. Die Hydroxylschutzgruppe wird aus der Nitrilverbindung eliminiert, und die Cyanogruppe wird dann zu der Carboxylgruppe hydrolysiert. Dann wird nach Einführung einer weiteren Schutzgruppe (beispielsweise der Acetylgruppe) für die 7-Hydroxylgruppe die Carboxylgruppe reduziert, wobei ein Bicyclolacton erhalten wird, das eine Alkylseitenkette aufweist, die in der Zahl der Kohlenstoffatome um eines verlängert wurde.
- Ein Merkmal der vorliegenden Erfindung besteht darin, daß die Kondensationsreaktion zwischen Bicyclolactonaldehyd (1) und Dimethyl-(2-oxoalkyl)phosphonat unter Bildung des α,β-ungesättigten Ketolactons (3) in Anwesenheit eines Alkalimetallhydrids und einer Zinkverbindung durchgeführt wird.
- Als Alkalimetallhydride können die Hydride der Alkalimetalle oder Erdalkalimetalle verwendet werden, und Beispiele davon umfassen Natriumhydrid, Kaliumhydrid oder Calciumhydrid, wobei Natriumhydrid besonders bevorzugt ist.
- Zinkhalogenide, wie Zinkchlorid, Zinkbromid, oder Zinksalze organischer Säuren, wie Zinkacetat, können als Zinkverbindungen verwendet werden, und die Halogenide sind besonders bevorzugt.
- Die Menge an Alkalimetallhydrid, die verwendet wird, beträgt bevorzugt etwa 1 Moläquivalent, bezogen auf die Menge an Dimethyl-(2-oxoalkyl)phosphonat (2), und die Menge an Zinkverbindung beträgt bevorzugt 0,5 bis 1 Moläquivalent, bezogen auf die Menge an Dimethyl-(2-oxoalkyl)phosphonat (2).
- Die Reaktionslösungsmittel sind nicht besonders spezifiziert, aber Lösungsmittel wie Tetrahydrofuran, Dimethoxyethan, Dichlormethan, Ethylether, 1,4-Dioxan, Benzol und Toluol sind bevorzugt.
- Die Menge an Reaktionslösungsmittel liegt bevorzugt im Bereich von 5 bis 30 ml, bezogen auf 1 g Corey-Aldehyd (1), und der Bereich von 10 bis 20 ml ist besonders bevorzugt.
- Der Reaktionstemperaturbereich kann von 0 bis 50ºC betragen, und der Bereich von 10 bis 50ºC ist besonders bevorzugt.
- Die Reaktionszeit kann im Bereich von 4 bis 72 h liegen und liegt bevorzugt im Bereich von 24 bis 50 h.
- Die folgenden Beispiele erläutern die Erfindung, ohne sie zu beschränken.
- Oxalylchlorid (2 M in CH&sub2;Cl&sub2;, 176,7 ml) wurde zu Dichlormethan (530 ml) gegeben, und nach dem Abkühlen auf -78ºC wurde eine Lösung von Dimethylsulfoxid (50,7 ml; im folgenden als DMSO bezeichnet) in Dichlormethan (50,7 ml) tropfenweise im Verlauf von 20 min unter Rühren bei -78ºC zugegeben. In die oben erhaltene Lösung wurde tropfenweise eine Lösung des Corey-Lactons (A) (45,3 g)
- in Dichlormethan (200 ml) zugegeben, und nach dem Rühren bei -50ºC während 1 h wurde Triethylamin (101,3 ml) tropfenweise zugegeben, und anschließend wurde 1 h bei einer auf -20ºC erhöhten Temperatur gerührt. Die Lösung wurde in eine gesättigte wäßrige Lösung aus Ammoniumchlorid gegossen. Nach der üblichen Aufarbeitung wurde der Aldehyd (B)
- erhalten.
- Natriumhydrid (60%, 8,48 g) wurde in THF (300 ml) bei 0ºC suspendiert, dazu wurde eine Lösung aus Dimethyl-(3,3-difluor-2-oxoheptyl)phosphonat (54,8 g) in THF (100 ml) zugegeben, und das Gemisch wurde 20 min gerührt. Zu dem entstehenden Gemisch wurde Zinkchlorid (14,5 g) gegeben, und es wurde 2 h bei Raumtemperatur gerührt. Das Gemisch wurde auf 0&sup4;C abgekühlt, und anschließend wurde eine Lösung des oben beschriebenen Aldehyds in THF (200 ml) zugegeben, und dann wurde 18 h bei Raumtemperatur gerührt. Das Additionsanion, hergestellt aus Dimethyl-(3,3-difluor-2-oxoheptyl)phosphonat (22,8 g), Natriumhydrid (60%, 3,5 g) und Zinkchlorid (6,0 g), wurde zugegeben, und das Gemisch wurde 6 h bei Raumtemperatur gerührt, und anschließend wurde Essigsäure (12,1 ml) bei 0ºC zugegeben. Das nach der üblichen Aufarbeitung erhaltene Rohprodukt wurde auf einer Silicagelsäule gereinigt, und (1A,5R,6R,7R)- 6-[(E)-4,4-Difluor-3-oxo-1-octenyl]-7-tetrahydropyranyloxy-2-oxabicyclo[3.3.0]octan-3-on (im folgenden als α,β-ungesättigtes Ketolacton (C) bezeichnet) wurde erhalten.
- Ausbeute: 52,8 g (77%)
- Der Aldehyd (8) wurde erhalten, indem das Corey-Lacton (A) (2,37 g) der Swern-Oxidation unter Verwendung von Oxalylchlorid (1,8 ml), DM50 (2,9 ml) und Triethylamin (12,5 ml) in Dichlormethan unterworfen wurde.
- Dimethyl-(3,3-difluor-2-oxoheptyl)phosphonat wurde tropfenweise zu einer Lösung von Thalliumethoxid (0,06 ml) in Methylenchlorid (60 ml) gegeben, und die Lösung wurde 30 min gerührt. Die Reaktionstemperatur wurde auf 0ºC eingestellt, und eine Lösung des Aldehyds (8) in Dichlormethan (30 ml) wurde tropfenweise zugegeben, und anschließend wurde 5 h bei Raumtemperatur gerührt. Das nach der üblichen Aufarbeitung erhaltene Rohprodukt wurde an einer Säule aus Silicagel chromatographiert, wobei das α,β-ungesättigte Ketolacton (C) erhalten wurde.
- Ausbeute: 2,35 g (66%)
- Der Aldehyd (B) wurde erhalten, indem das Corey-Lacton (A) (1,25 g) der Swern-Oxidation unter Verwendung von Oxalylchlorid (0,88 ml), DMSO (1,42 ml) und Triethylamin (6,1 ml) in Dichlormethan unterworfen wurde.
- Natriumhydrid (60%, 0,195 g) wurde in 30 ml THF suspendiert, und eine Lösung aus Dimethyl-(3,3-difluor-2-oxoheptyl)phosphonat (1,27 g) in TFH (5 ml) wurde tropfenweise zu der Suspension zugegeben, und anschließend wurde 1 h gerührt. Kupferiodid (0,929 g) wurde zu der Lösung zugegeben, und das Gemisch wurde 1 h bei Raumtemperatur gerührt. Nach dem Einstellen der Reaktionstemperatur auf 0ºC wurde eine Aldehydlösung (8) in THF (10 ml) tropfenweise zugegeben, und dann wurde über Nacht bei Raumtemperatur gerührt. Das nach der üblichen Aufarbeitung erhaltene Rohprodukt wurde an einer Säule aus Silicagel chromatographiert, wobei das α,β-ungesättigte Ketolacton (C) erhalten wurde.
- Ausbeute: 0,431 g (23%)
- Der Aldehyd (8) wurde erhalten, indem das Corey-Lacton (A) (0,523 g) der Swern-Oxidation gemäß dem gleichen Verfahren, wie in Vergleichsbeispiel 2 beschrieben, unterworfen wurde.
- Natriumhydrid (60%, 0,082 g) wurde in THF suspendiert, und eine THF- Lösung aus Dimethyl-(3,3-difluor-2-oxoheptyl)phosphonat (0,464 g) wurde zu der Suspension zugegeben, und das Gemisch wurde 30 min gerührt. Die Reaktionstemperatur wurde auf 0ºC eingestellt, und die THF-Lösung des Aldehyds (B), erhalten wie oben beschrieben, wurde zugegeben, und das Gemisch wurde über Nacht gerührt. Der nach der üblichen Aufarbeitung erhaltene Rückstand wurde an einer Säule aus Silicagel chromatographiert, aber kein α,β-ungesättigtes Ketolacton (C) wurde erhalten.
- Dimethyl-(3,3-difluor-2-oxononyl)phosphonat (3,918 g) und Natriumhydrid (60%, 0,547 g) wurden in THF (30 ml) während 15 min bei Raumtemperatur gerührt. Zinkchlorid (0,933 g) wurde zugegeben, und die Lösung wurde 2 h gerührt. Zu der Lösung wurde Corey-Aldehyd (1,69 g) in THF (20 ml) zugegeben, welche aus einem Corey-Lacton gemäß dem in Beispiel 1 beschriebenen Verfahren hergestellt wurde, und die Lösung wurde 50 h bei Raumtemperatur gerührt. Das bei der üblichen Aufarbeitung erhaltene Rohprodukt wurde chromatographiert, wobei (1S,5R,6R,7R)-6-[(E)-4,4-Difluor-3-oxo-1-decenyl]-7-tetrahydropyranyloxy-2-oxabicyclo[3.3.0]octan-3-on erhalten wurde.
- Ausbeute: 1,789 g (65%)
Claims (4)
1. Verfahren zur Herstellung α,β-ungesättigter Ketone
durch Umsetzung eines Bicyclolactonaldehyds mit einem
Dimethyl-(2-oxoalkyl)phosphonat, welches durch mindestens ein
Halogenatom in der 3-Stellung der Alkylgruppe substituiert
ist, in Anwesenheit eines Alkalimetallhydrids und einer
zinkverbindung.
2. Verfahren nach Anspruch 1, worin der
Bicyclolactonaldehyd durch die Formel
dargestellt wird [worin A eine Hydroxylschutzgruppe
bedeutet und B eine Einfachbindung oder -CH&sub2;- bedeutet] und das
Dimethyl-(2-oxoalkyl) phosphonat durch die Formel
dargestellt wird [worin X&sub1; und X&sub2; ein Wasserstoffatom oder
ein Halogenatom bedeuten, mit der Maßgabe, daß mindestens
eines von X&sub1; und X&sub2; ein Halogenatom bedeutet, R&sub1; eine
Alkylgruppe mit 1 bis 4 Kohlenstoffatomen bedeutet, R&sub2; eine
Alkylgruppe mit 1 bis 10 Kohlenstoffatomen bedeutet, die
eine Verzweigung, eine Doppelbindung, eine Alkoxygruppe,
eine Phenylgruppe oder eine Phenoxygruppe oder eine Gruppe
der Formel
(worin Y eine Einfachbindung oder ein Sauerstoffatom
darstellt und R&sub3; ein Wasserstoffatom oder ein Halogenatom oder
eine Alkyl- oder eine halogenierte Alkylgruppe bedeutet)
enthalten kann].
3. Verfahren nach Anspruch 2, worin mindestens eines
von X&sub1; und X&sub2; ein Fluoratom bedeutet.
4. Verfahren nach Anspruch 1, worin die
α,β-ungesättigten Ketolactone durch die Formel
dargestellt werden [worin A, X&sub1;, X&sub2; und R&sub2; die oben
gegebenen Bedeutungen besitzen und Z =CH- oder -CH=CH- bedeutet].
