- BEREICH DER TECHNIK
- Die Erfindung betrifft ein neues Pyridin-1-oxid-3-carboxamidin-Derivat, welches als Zwischenprodukt bei der Herstellung des Wirkstoffes von pharmazeutischen Produkten zur Behandlung von diabetischen Komplikationen verwendet werden kann. Die Erfindung betrifft nämlich die Verbindung N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboxamidin und ihre optisch aktiven Enantiomere (R)-(–)-N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboxamidin und (S)-(+)-N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboxamidin. Darüber hinaus betrifft die Erfindung die Herstellung von N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboximidoylchlorid, welches als ein Wirkstoff von Medikamenten verwendet werden kann, und die Herstellung der optisch aktiven Enantiomere dieser Verbindung unter Verwendung der Verbindungen der Erfindung als Zwischenproduktsubstanzen.
- STAND DER TECHNIK
- N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-3-carboximidoylchlorid ist zusammen mit mehreren verwandten Verbindungen aus WO 97/16439 als ein wirksames Mittel, geeigneterweise zur Erhöhung der Chaperonexpression von Zellen, bekannt. In dieser Veröffentlichung wird die Verbindung als eine neue Verbindung definiert und die N-Oxid-Derivate der Verbindung werden auch beansprucht, aber das N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboximidoylchlorid wird nicht speziell erwähnt und sein Herstellungsverfahren wird auch nicht beschrieben.
- N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboximidoylchlorid wird in WO 00/50403 als eine neue Verbindung offenbart und beansprucht und auch sein Herstellungsverfahren wird dort beschrieben. Die Verbindung wird durch die Oxidation des N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-3-carboximidoylchlorids hergestellt. Im Laufe der Oxidation mit Persäuren werden das Bis-N-oxid-Derivat, welches an den N-Atomen von sowohl dem Piperidinring als auch dem Pyridinring oxidiert ist, oder das Piperidin-N-oxid- Derivat bevorzugt gebildet und deshalb muss die Oxidation mit Persäure in der Gegenwart einer starken Säure durchgeführt werden, um die Vorherrschaft der Bildung des Pyridin-N-oxid-Derivats im Laufe des Oxidationsverfahrens sicher zu stellen. Die Ausbeute dieses Verfahrens ist jedoch nicht zufriedenstellend. Die optisch aktiven Enantiomere des N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboximidoylchlorids werden auch in WO 00/50403 beschrieben. Sie werden über einen Weg, der ähnlich zu der Herstellung der Raceme-Verbindung ist, unter Verwendung der geeigneten optisch aktiven Ausgangssubstanzen hergestellt.

- Unter Berücksichtigung der Tatsache, dass das N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboximidoylchlorid und seine optisch aktiven Enantiomere nicht nur bei der Behandlung von diabetischen Komplikationen, vorwiegend von Retinopathie, Neuropathie und Nephropathie, nützlich sind, sondern gleichzeitig chronische Insulinresistenz verringern, sind diese Verbindung wertvolle Wirkstoffe in pharmazeutischen Produkten. Damit diese Verbindungen jedoch in der pharmazeutischen Industrie nützlich sind, ist ein einfacheres Verfahren für ihre Herstellung notwendig.
- OFFENBARUNG DER ERFINDUNG
- Wir haben gefunden, dass N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboxamidin der Formel (I), welches eine neue Verbindung ist, als Zwischenprodukt nützlich ist, wenn die einfache Herstellung von N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboximidoylchlorid in hoher Reinheit und in einer hohen Ausbeute möglich gemacht wird.
- Bezogen auf diese Beobachtung betrifft die vorliegende Erfindung das N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboxamidin und seine Säureadditionssalze. Die Erfindung betrifft auch die optisch aktiven Enantiomere der vorstehend erwähnten Verbindung, das (R)-(–)-N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboxamidin und das (S)-(+)-N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboxamidin, und die Säureadditionssalze dieser Verbindungen.
- Hier nachstehend betrifft „optisch aktives Enantiomer" eine Verbindung, deren optische Reinheit mindestens 80 %, bevorzugt mindestens 90 %, am stärksten bevorzugt mindestens 96 % beträgt, das heißt, die Verbindung enthält mindestens dieses Massenverhältnis des spezifizierten optisch aktiven Enantiomers. „Säureadditionssalze" betrifft Salze, welche aus den Verbindungen mit Mineral- und organischen Säuren durch das bekannte Verfahren hergestellt werden.
- Wie vorstehend erwähnt, können die Verbindungen der Erfindung als Zwischenprodukte bei der Herstellung von N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboximidoylchlorid verwendet werden, Deshalb betrifft die Erfindung die Verwendung von N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboxamidin und seine optisch aktiven Enantiomere bei der Herstellung von N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboximidoylchlorid und seinen optisch aktiven Enantiomeren.
- Die Verbindung N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboxamidin der Erfindung wird bevorzugt durch das folgende Verfahren hergestellt.
- 3-Cyanopyridin wird als eine Ausgangsverbindung verwendet und das 3-Cyanopyridin-1-oxid der Formel (II), welches aus der Literatur [J. Chem. Soc. 3680 (1959)] bekannt ist, kann durch Oxidation hergestellt werden. Eine Persäure wird als Oxidationsmittel, bevorzugt m-Chlorperbenzoesäure, verwendet. Das so erhaltene Produkt kann durch Kristallisation gereinigt werden, aber das Rohprodukt kann auch im nächsten Schritt der Synthese verwendet werden.
- Durch Umsetzen von 3-Cyanopyridin-1-oxid der Formel (II) mit Hydroxylamin wird 3-Pyridinamidoxim-1-oxid der Formel (III) hergestellt. Die Umsetzung wird in einer geeigneten wässrigen Lösung bei Raumtemperatur durch Umsetzen von 3-Cyanopyridin-1-oxid mit Hydroxylamin, welches im Überschuss zugegeben wird und in Wasser in situ von seinem Hydrochloridsalz mit Natriumbicarbonat freigesetzt wird, durchgeführt. Das Produkt fällt aus, deshalb kann es einfach isoliert und durch Kristallisation gereinigt werden.
- N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboxamidin der Formel (I) wird aus dem 3-Pyridinamidoxim-1-oxid der Formel (III) durch Umsetzen der Verbindung mit einem reaktiven 3-(1-Piperidino)-2-hydroxypropan-Derivat hergestellt. Als Reagenz kann ein 1-Halogen- oder 1,2-Epoxy-Derivat verwendet werden, aber bevorzugt sollte ein cyclisches Derivat, ein Halogenid von 2-Hydroxy-4-azoniaspiro[3,5]nonan als Reagenz verwendet werden. Das am stärksten bevorzugte Reagenz ist das 2-Hydroxy-4-azoniaspiro[3,5]nonanchlorid der Formel (IV). Die Umsetzung wird in einem alkalischen Medium unter Verwendung eines geeigneten Alkohols, bevorzugt eines Alkohols mit 1 bis 3 Kohlenstoffatomen, stärker bevorzugt Ethanol, als Lösungsmittel durchgeführt. Die Reagenzien können in jedweder Reihenfolge zugegeben werden. Bevorzugt wird das reaktive 3-(1-Piperidino)-2-hydroxypropan-Derivat in einem geringen Überschuss verwendet. Die Umsetzung wird bei einer erhöhten Temperatur, bevorzugt beim Siedepunkt des Lösungsmittels, durchgeführt.
- Die so erhaltene Verbindung der Formel (I) wird entweder als eine Base isoliert und als Zwischenprodukt bei der Herstellung des biologisch wirksamen N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboximidoylchlorids verwendet oder die Verbindung wird mit einer Mineral- oder organischen Säure in ein Säureadditionssalz umgewandelt. So können Mono- oder Dihydrochlorid, Maleat oder jedwedes andere Säureadditionssalz hergestellt werden, welche für die Verwendung der Verbindung als Zwischenprodukt bei der Herstellung des vorstehend erwähnten Endprodukts geeignet sind. Jedoch ist es nicht notwendig, das N-[2-Hydroxy-3-(1- piperidinyl)-propoxy]-pyridin-1-oxid-3-carboxamidin, welches als Zwischenprodukt verwendet wird, zu isolieren. Der nächste Schritt bei der Synthese von N-[2-Hydroxy-3-(1-piperidinyl)-propoxy)-pyridin-1-oxid-3-carboximidoylchlorid kann auch ohne die Isolierung der Verbindung der Formel (I) durchgeführt werden.
- Wenn die Herstellung eines optisch aktiven Produkts gewünscht ist, kann die Verbindung der Formel (I) vor der Umwandlung durch Umsetzen davon mit einer optisch aktiven Säure, welche zur Bildung eines Diastereomerensalzpaares geeignet ist, durch die bekannten Aufspaltungsverfahren aufgespalten werden. Wenn das Salz mit der erforderlichen optischen Reinheit erhalten wurde, kann die optisch aktive Base davon freigesetzt werden. Danach kann, falls notwendig, ein Säureadditionssalz aus der Base mit einer Mineral- oder organischen Säure hergestellt werden. Entweder die Base oder das Salz können dann für den nächsten Schritt des Verfahrens der Erfindung verwendet werden.
- Gemäß der Erfindung wird N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboxamidin der Formel (I) durch Diazotierung in der Gegenwart von Chlorwasserstoffsäure zu N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboximidoylchlorid umgewandelt. Die Diazotierung wird durch das normale Verfahren bei einer Temperatur von zwischen –5°C und 0°C mit der langsamen Zugabe eines Alkalinitrits, bevorzugt von Natriumnitrit, durchgeführt. In der Gegenwart von Chlorwasserstoffsäure zersetzt sich das so erhaltene Diazoniumsalz bei der Temperatur der Diazotierung zu N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboximidoylchlorid der Formel (V). Dann wird das Reaktionsgemisch während Kühlen alkalisch gemacht und das Produkt wird in der Form einer Base durch das normale Verfahren isoliert. Die erhaltene Base kann ferner gereinigt werden, falls notwendig, oder mit einer Mineral- oder organischen Säure in ein Säureadditionssalz umgewandelt werden. Bevorzugt werden Maleat oder Citrat aus der Verbindung der Formel (V) gebildet, aber sie kann auch in das Hydrochlorid, Dihydrochlorid oder jedwedes pharmazeutisch verträgliche Säureadditionssalz umgewandelt werden.
- Die erforderlichen optisch aktiven Enantiomere des N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboximidoylchlorids der Formel (V), welches durch Diazotierung hergestellt wurde, werden durch Aufspaltung aus der Raceme-Verbindung hergestellt. Die Aufspaltung wird wieder durch die Bildung eines Diastereornerensalzpaares, bevorzugt mit Dibenzoylweinsäure unter Verwendung von ihrer geeigneten optisch aktiven Form für die Salzbildung, durchgeführt.
- Wenn die vorstehend beschriebene Diazotierung von N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboxamidin der Formel (I) an einem optisch aktiven Enantiomer durchgeführt wird, wird das Enantiomer des N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboximidoylchlorids der Formel (V) mit gegenteiliger Drehung und zufriedenstellender optischer Reinheit gebildet. Deshalb wird gemäß einer anderen Version des Verfahrens der Erfindung das optisch aktive N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboximidoylchlorid der Formel (V) durch Durchführen der vorstehend beschriebenen Diazotierung an dem geeigneten optisch aktiven Enantiomer von N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboxamidin der Formel (I) hergestellt. Die so erhaltene Base kann ferner gereinigt werden, falls notwendig, oder ein Säureadditionssalz kann mit einer Mineral- oder organischen Säure gebildet werden.
- Der Vorteil der Erfindung ist, dass die Verwendung der Verbindung der Formel (I) der Erfindung als Zwischenprodukt die Herstellung von hoch reinem N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboximidoylchlorid, welches wertvolle biologische Wirkungen aufweist, möglich macht. Im Gegensatz zu dem Verfahren, welches im in der Einleitung erwähnten WO 00/50403 beschrieben wird, wobei diese Verbindung durch die Oxidation von N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-3-carboximidoylchlorid hergestellt wird, muss bei diesem vorliegenden Verfahren das Auftreten der Produkte von kompetitiven Umsetzungen nicht in Betracht gezogen werden. Durch das Verfahren der Erfindung wird die Basenform von N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboximidoylchlorid in hoher Reinheit erhalten. Bis zu diesem Zeitpunkt war dies nur durch eine lange Reinigung oder durch Freisetzung der Basenform aus dem Maleatsalz möglich.
- Ein weiterer Vorteil der Erfindung ist, dass das N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboxamidin durch das Zusammenlegen der vorstehend beschriebenen und in den folgenden Beispielen detaillierter veranschaulichten Schritte ohne die Isolierung und/oder Reinigung von jedem Zwischenprodukt hergestellt werden kann, wobei noch eine ausreichende Reinheit erhalten wird. Auf diese Weise wird die Herstellung des Zwischenprodukts N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboxamidin unter den Umständen einer pharmazeutischen Herstellung möglich, was die industrielle Herstellung des biologisch wirksamen N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboximidoylchlorids möglich macht.
- BESTE WEISE ZUR DURCHFÜHRUNG DER ERFINDUNG
- Die Erfindung wird durch die folgenden Beispiele veranschaulicht.
- Beispiel 1.
- Die Herstellung von 3-Cyanopyridin-1-oxid (Verbindung der Formel II)
- 86 g (0,378 Mol) 76 %ige m-Chlorperbenzoesäure werden in 730 ml Dichlormethan bei 20 bis 25°C gelöst und 220 ml der Lösung von 38,3 g (0,378 Mol) 3-Cyanopyridin in Dichlormethan werden bei 20 bis 28°C zugegeben. Das Reaktionsgemisch wird bei 20 bis 24°C für 24 Stunden gerührt. Am Ende der Umsetzung wird das Lösungsmittel abgedampft. Der Abdampfrückstand wird in 430 ml Methyl-tert-butylether digeriert, der Niederschlag wird abfiltriert, gewaschen und getrocknet. 59 g Rohprodukt werden erhalten.
- Durch zweifache Umkristallisation des Rohprodukts in heißem Ethanol werden 36,6 g (80 %) reines 3-Cyanopyridin-1-oxid erhalten, welches bei 174 bis 176,5°C schmilzt (Literatur: 174 bis 175°C; J. Chem. Soc. 3680 (1959)).
- Beispiel 2.
- Die Herstellung von 3-Pyridinamidoxim-1-oxid (Verbindung der Formel III).
- 25,41 g (0,366 Mol) Hydroxylamin-Hydrochlorid und 36,6 g (0,305 Mol) 3-Cyanopyridin-1-oxid werden in 540 ml Wasser gelöst, dann werden 30,72 g (0,366 Mol) Natriumhydrogencarbonat in kleinen Portionen zugegeben. Das Reaktionsgemisch wird für 2 Stunden bei 20 bis 25°C gerührt. Die Suspension wird filtriert, der Niederschlag wird mit Wasser gewaschen, getrocknet und in einem Gemisch von 9:1 von Methanol und Wasser umkristallisiert. Der Niederschlag, der sich durch Kühlen abscheidet, wird abfiltriert, gewaschen und getrocknet. 37,3 g (80 %) der Titelverbindung, welche bei 212 bis 215°C während Zersetzung schmilzt, werden erhalten.

- a) Die Herstellung von N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboxamidin (Verbindung der Formel I)
- 13,5 g (0,34 Mol) Natriumhydroxid werden in 35 ml Wasser gelöst und die Lösung wird auf 10°C gekühlt. 60,75 g (0,34 Mol) 2-Hydroxy-4-azoniaspiro[3,5]nonanchlorid werden zugegeben und das Reaktionsgemisch wird für 40 Minuten bei 5 bis 10°C gerührt. 540 ml Ethanol und 40,5 g (0,26 Mol) 3-Pyridinamidoxim-1-oxid werden zugegeben. Das Reaktionsgemisch wird für 2 Stunden unter einer Rückflusskondensiervorrichtung erwärmt. Die Lösung wird gekühlt, das abgeschiedene Natriumchlorid wird abfiltriert, mit 100 ml Ethanol gewaschen und dann wird das Lösungsmittel eingedampft. Der Eindampfrückstand wird mit Diethylether digeriert, der Niederschlag, der sich im Kühleren abscheidet, wird abfiltriert, mit Ether gewaschen, getrocknet und in Isopropanol heiß kristallisiert. 47,4 g (62 %) der Titelverbindung, welche bei 130 bis 132,5°C schmilzt, werden erhalten.
IR: ν (KBr, cm–1): 3397, 3189, 2928, 1647, 1610, 1570, 1505, 1425, 1247, 1226, 1033, 942, 901, 782, 662, 531. - b) Die Herstellung von N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboxamidin-Monohydrochlorid
- Die erhaltene Base der Formel (I) wird in Ethanol gelöst und 1 Äquivalent ethanolische Chlorwasserstoffsäure-Lösung wird zugegeben. Die Lösung wird eingedampft und der Rückstand wird mit Isopropanol digeriert. Das abgeschiedene Salz wird abfiltriert, gewaschen und getrocknet. N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboxamidin-Monohydrochlorid wird erhalten. Schmelzpunkt: 142 bis 144,5°C.

Cl–- Gehalt gemäß Mohr (berechnet/gemessen): 10,7/10,45. - Beispiel 4.
- a) Die Herstellung von N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboximidoylchlorid (Verbindung der Formel V)
- Die Lösung von 6,1 g (0,088 Mol) Natriumnitrit in 40 ml Wasser wird tropfenweise bei –5 bis 0°C zu der Lösung von 20 g (0,068 Mol) N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboxamidin in 110 ml 1 M Chlorwasserstoffsäure, welche auf –5°C gekühlt ist, gegeben. Das Reaktionsgemisch wird für 1,5 Stunden bei zwischen –5°C und 0°C, dann bei intensiver Kühlung (t < 7°C) gerührt, das Gemisch wird mit einer 7 M NaOH-Lösung alkalisch gemacht, auf einen pH-Wert von 10,5 bis 11,5. Die Lösung wird dreimal mit 180 ml Dichlormethan extrahiert, die organischen Phasen werden vereinigt, über wasserfreiem MgSO4 getrocknet, filtriert, gewaschen und eingedampft. Das so erhaltene dicke Öl wird in 130 ml Methyl-tert-butylether digeriert. Die Suspension wird für 12 Stunden in einen Kühlschrank gestellt, am darauffolgenden Tag wird der Niederschlag abfiltriert, gewaschen und getrocknet. 18 g (85 %) der Titelverbindung, welche bei 90 bis 91,5°C schmilzt, werden erhalten.

- 50 g der wie vorstehend beschrieben hergestellten Base werden in 50 ml Aceton gelöst und eine Menge von einem Äquivalent (1,85 g) Maleinsäure wurde dazu gegeben. Der abgeschiedene Niederschlag wird abfiltriert, mit Aceton gewaschen und getrocknet. Das Produkt wird in Ethanol umkristallisiert. 6,0 g (87 %) N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboximidoylchlorid-Maleat (1:1) werden erhalten, welches bei 150,5 bis 154,5°C schmilzt.

- 0,64 g (2,0 mMol) (R,S)-N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboximidoylchlorid-Base werden in 20 ml Ethylacetat gelöst und 0,64 ml der etherischen Lösung von 3,2 M Chlorwasserstoffsäure werden während Rühren zugegeben. Ein klebriger Niederschlag wird gebildet. Nach dem Abdampfen des Lösungsmittels werden 15 ml Aceton zugegeben und es wird über Nacht in einen Kühlschrank gestellt. Der Niederschlag wird mit der Zugabe von einigen Tropfen Ethanol kristallisiert, dann wird das weiße hygroskopische Material schnell abfiltriert und in einem Exsikkator über Phosphorpentoxid getrocknet.
Ausbeute: 0,4 g (56,3 %)
Schmelzpunkt: 115 bis 122°C

- a) Die Herstellung von (R)-(+)-N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboximidoylchlorid durch die Aufspaltung der Raceme-Verbindung
- 54 g (0,172 Mol) Raceme-N-[2-hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboximidoylchlorid werden in 720 ml trockenem Ethanol gelöst. Nach dem vollständigen Lösen werden 64,7 g (0,172 Mol) (–)-Dibenzoyl-L-weinsäure-Monohydrat zugegeben. Nach dem Animpfen wird bei Raumtemperatur kristallisiert, dann wird der abgeschiedene Niederschlag abfiltriert, gewaschen und getrocknet. Der erhaltene Rückstand wird zweimal in trockenem Ethanol kristallisiert. 38 g (65 %) (R)-(+)-N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboximidoylchlorid-Dibenzoyl-L-weinsäure-Salz werden erhalten.
- Die so erhaltenen 38 g (0,056 Mol) Weinsäuresalz werden dann zu 230 ml 1 M K2CO3-Lösung gegeben und das Gemisch wird mit 3 × 225 ml Dichlormethan extrahiert. Die organischen Phasen werden vereinigt, über wasserfreiem MgSO4 getrocknet, mit Aktivkohle behandelt, abfiltriert und eingedampft. Die Verbindung wird in einer reinen Form aus der rohen Base durch Kristallisation mit Hexan erhalten. Ausbeute: 14,0 g (80 %). Schmelzpunkt: 91 bis 93 °C.

- 18,0 g rohe (Abdampfrückstand aus dem Verfahren a) oder isolierte Base werden in 62 ml Aceton gelöst und dann wird die Lösung von 6,6 g Maleinsäure in 46 ml Aceton zugegeben. Der Niederschlag, der sich durch Kühlen abscheidet, wird abfiltriert, mit 10 ml Aceton gewaschen und getrocknet. Der Niederschlag (etwa 23 g) wird in 120 ml heißem Ethanol kristallisiert. Der Niederschlag, der sich durch Kühlen abscheidet, wird abfiltriert, gewaschen und getrocknet.
Ausbeute: 20 g (82 %)
Schmelzpunkt: 132,0 bis 133,5°C
Enantiomerenverhältnis: 98/2 (HPLC-Messung an einer Chiral AGP 100 × 4mm-Säule)

- c) Die Herstellung von (R)-(+)-N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboximidoylchlorid-Citrat
- 2,43 g (7,75 mMol) (+)-(R)-N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboximidoylchlorid werden in 15 ml Aceton während sanftem Erwärmen gelöst, dann werden 5 ml der Lösung von 1,63 g (7,75 mMol) Zitronensäure in Aceton zugegeben. Die sich abscheidende weiße klebrige Substanz wird durch die Zugabe von 5 ml Methanol zu einem Pulver. Die Lösung wird bis zum folgenden Tag in einen Kühlschrank gestellt. Der Niederschlag wird abfiltriert, mit Aceton gewaschen und getrocknet. 3,99 g (98 %) Produkt werden erhalten. Schmelzpunkt: 163 bis 165°C.

- 0,88 g (2,8 mMol) (R)-(+)-N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboximidoylchlorid-Base (hergestellt aus ihrem Maleat unter Verwendung von Dichlormethan/10 %ige wässrige Natriumcarbonat-Lösung) werden in 25 ml Ethylacetat gelöst und 0,87 ml der etherischen Lösung von 3,2 M Chlorwasserstoffsäure werden während Rühren zugegeben.
- Nach dem Abdampfen des Lösungsmittels werden 15 ml Aceton zu dem restlichen klebrigen Öl gegeben und es wird mit der Zugabe von einigen Tropfen Ethanol kristallisiert. Die weiße Verbindung wird abfiltriert und mit Aceton und Diethylether gewaschen.
Ausbeute: 0,8 g (81,6 %)
Schmelzpunkt: 127 bis 131 °C

- 0,65 g (2,0 mMol) (R)-(+)-N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboximidoylchlorid-Base (hergestellt aus ihrem Maleat unter Verwendung von Dichlormethan/10 %ige wässrige Natriumcarbonat-Lösung) werden in 25 ml Ethylacetat gelöst und 1,4 ml der etherischen Lösung von 3,2 M Chlorwasserstoffsäure werden während Rühren zugegeben.
- Nach dem Abdampfen des Lösungsmittels werden 15 ml Aceton zu dem restlichen klebrigen Öl gegeben und es wird mit der Zugabe von einigen Tropfen Ethanol kristallisiert. Die weiße Verbindung wird abfiltriert und mit kaltem Aceton und Diethylether gewaschen.
Ausbeute: 0,4 g (55,0 %)
Schmelzpunkt: 160 bis 164°C

- a) Die Herstellung von (S)-(+)-N-[2-hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboxamidin durch die Aufspaltung der Raceme-Verbindung
- 1,74 g (5,9 mMol) Raceme-N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboxamidin werden in 15 ml Ethanol gelöst und dann werden 2,1 g (5,9 mMol) (+)-Dibenzoyl-D-weinsäure zu der Lösung gegeben. Es wird bei Raumtemperatur kristallisiert, dann wird das abgeschiedene Salz abfiltriert, mit Ethanol gewaschen und getrocknet. Die 2,1 g Produkt werden dann zweimal in Ethanol kristallisiert, wobei 0,33 g (17 %) (S)-(+)-N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboxamidin-Dibenzoyl-D-weinsäure-Salz (Schmelzpunkt: 156 bis 158°C) erhalten wurden.
- Das (S)-(+)-N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboxamidin wird aus dem erhaltenen Salz durch Verteilung zwischen 2 M K2CO3-Lösung und Chloroform, Eindampfen der Chloroform-Lösung und Isolierung mit Ether freigesetzt.
Ausbeute: 0,11 g (13 %).
Schmelzpunkt: 123 bis 126°C.
IR: ν (KBr, cm–1): 3398, 3188, 2936, 2800, 1647, 1600, 1560, 1500, 1426, 1245, 1226, 1033, 942, 907, 800, 667.
Drehung: [α]D = + 4,5° (c = 1, Methanol). - Um die optische Reinheit des Produkts zu bestimmen, wird eine kleine Probe in die entsprechende Chlorverbindung, (S)-(–)-N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboximidoylchlorid, durch das in Beispiel 4/a beschriebene Diazotierungsverfahren umgewandelt und seine optische Reinheit wird durch HPLC an einer Chiral AGP-Säule bestimmt. Die durch dieses Verfahren bestimmte optische Reinheit des Produktes, welche durch das beschriebene Aufspaltungsverfahren erreicht wird, ist 96 %.
- b) Die Herstellung von (R)-(–)-N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboxamidin durch die Aufspaltung der Raceme-Verbindung
- Folgend dem in a) beschriebenen Verfahren, aber unter Verwendung von (–)-Dibenzoyl-L-weinsäure für die Aufspaltung in diesem Fall, beträgt die Ausbeute des erhaltenen Dibenzoyltartratsalzes 20 % und sein Schmelzpunkt ist 156,5 bis 158°C. Nach dem Freisetzen der Base daraus wird (R)-(–)-N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboxamidin mit einer optischen Reinheit von 96 % erhalten.
Drehung: [α]D = –4,5° (c = 1, Methanol). - Sein Schmelzpunkt und IR-Spektrum waren die gleichen wie jene des anderen Enantiomers.
- c) Die Herstellung von (S)-(+)-N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboxamidin-Monohydrochlorid
- Die (S)-(+)-N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboxamidin-Base, welche durch das Beispiel 6/a hergestellt wurde, wird in Isopropanol gelöst. Eine äquivalente Menge an isopropanolischer Chlorwasserstoffsäure-Lösung wird zugegeben. Das kristallisierte Monohydrochloridsalz wird abfiltriert und getrocknet.
Schmelzpunkt: 164 bis 165,5°C
IR: ν (KBr, cm–1): 3400, 3317, 3191, 2948, 2862, 2710, 2690, 2655, 1651, 1562, 1429, 1407, 1308, 1232, 1112, 1052, 1010, 962, 944, 880, 797, 670. - d) Die Herstellung von (S)-(+)-N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboxamidin-Maleat
- Das Maleatsalz der (S)-(+)-N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboxamidin-Base, welche durch Beispiel 6/a hergestellt wurde, wird durch die Zugabe von äquivalenter Maleinsäure in einer isopropanolischen Lösung hergestellt.
Schmelzpunkt: 148 bis 150,5°C.

- Die Herstellung von (S)-(–)-N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboximidoylchlorid durch Diazotierung
- Durch Diazotierung gemäß dem in Beispiel 4 beschriebenen Verfahren, gefolgt von der Zersetzung des Diazoniumsalzes werden 5 g des (S)-(+)-N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboxamidins, welches unter Beispiel 6/a durch die Aufspaltung von N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboxamidin hergestellt wurde, zu (S)-(–)-N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboximidoylchlorid umgewandelt. 4,4 g (82 %) des Produkts werden erhalten. Die Charakteristika des erhaltenen Produkts sind die gleichen wie jene, welche für das andere Enantiomer in Beispiel 5 gegeben sind.
- Die Base wird mit Maleinsäure durch das in Beispiel 5 gegebene Verfahren umgesetzt und so wird das entsprechende Maleatsalz erhalten, dessen Charakteristika auch die gleichen sind, wie jene, welche in Beispiel 5 gegeben sind.
Claims (6)
- N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboxamidin und seine Säureadditionssalze.
- (R)-(–)-N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboxamidin und seine Säureadditionssalze.
- (S)-(+)-N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboxamidin und seine Säureadditionssalze.
- Verwendung von N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboxamidin und seinen optisch aktiven Enantiomeren zur Herstellung von N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboximidoylchlorid und seinen optisch aktiven Enantiomeren.
- Verfahren zur Herstellung von N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboximidoylchlorid und seinen Säureadditionssalzen, umfassend: a) die Diazotierung von N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboxamidin durch Umsetzen mit einem Alkalinitrit in der Gegenwart von Chlorwasserstoffsäure, oder b) die Diazotierung von N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboxamidin durch Umsetzen mit einem Alkalinitrit in der Gegenwart von Chlorwasserstoffsäure und die Umwandlung der erhaltenen Base in ein Säureadditionssalz.
- Verfahren zur Herstellung von (R)-(+)-N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboximidoylchlorid und (S)-(–)-N-[2-Hydroxy-3-(1- piperidinyl)-propoxy]-pyridin-1-oxid-3-carboximidoylchlorid und ihren Säureadditionssalzen, umfassend a) die Diazotierung von (R)-(–)-N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboxamidin oder (S)-(+)-N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboxamidin durch Umsetzen mit einem Alkalinitrit in der Gegenwart von Chlorwasserstoffsäure, oder b) die Aufspaltung von N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboxamidin und die Diazotierung des erhaltenen optisch aktiven (R)-(–)-Enantiomers oder (S)-(+)-Enantiomers in der Gegenwart von Chlorwasserstoffsäure durch Umsetzen mit einem Alkalinitrit, oder c) die Diazotierung von N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboxamidin in der Gegenwart von Chlorwasserstoffsäure durch Umsetzen mit einem Alkalinitrit und die Isolierung des erforderlichen (R)-(+)- oder (S)-(–)-Enantiomers durch Aufspalten des erhaltenen racemischen-N-[2-hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboximidoylchlorids, oder d) die Umwandlung von (R)-(+)-N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboximidoylchlorid oder (S)-(–)-N-[2-Hydroxy-3-(1-piperidinyl)-propoxy]-pyridin-1-oxid-3-carboximidoylchlorid, welche durch eine der Verfahrensvarianten unter a) bis c) hergestellt wurden, in ein Säureadditionssalz.
