derselben und Verwendung derselben
Die Erfindung betrifft neue Platin(IV)-diaminkomplexe, ein
Verfahren zu ihrer Herstellung, ein Verfahren für die Herstellung von Heilmitteln, die solche Komplexe enthalten und
für die Behandlung von Krebs,wie bösartige Geschwulste oder
Tumoren, geeignet sind und die mittels des genannten Verfahrens erhaltenen Heilmittel.
Aus der Literatur ist es bekannt, dass Platin-diaminkomplexe sowohl auf Basis von zweiwertigem Platin wie auf Basis von
vierwertigem Platin für die Behandlung von Krebs geeignet sind. Siehe beispielsweise den Artikel von B. Rosenberg
und L. Van Camp, Cancer Research 30, 1970, S. 1799 bis 1802.
Eine einschlägige Veröffentlichung betreffend die Verwendung
von Platin-diaminkomplexen mit zweiwertigem Platin, beispielsweise
das Cis-Platindiamin-dichlorid, für die Verwendung von
Krebs ist beispielsweise der Artikel von A.P. Zipp und S.G. Zipp in J.Chem.Ed., 54 (12) (1977), Seite 739. Dieser
Artikel beschreibt die Verwendung von Cis-Platin-diamindichlorid
für die Behandlung von Krebs. Im Artikel wird festgestellt, dass die Platinverbindungen ein breites Spektrum
als Antitumorwirkstoffe zeigen, dass sie aber auch wichtige Nachteile aufweisen, speziell hinsichtlich ihrer Nierentoxizität.
Um dieser Nierentoxizität zu begegnen, wird eine Kombination des Cis-Platin-diamindichlorides mit einer anderen
Substanz, die Verwendung einer grossen Flüssigkeitsmenge oder andere Vorkehrungen, um die Nieren zu spülen, vorgeschlagen.

Im Journal of Clinical Hematology and Oncology, 7(1) (1977), Seiten 114-134 werden eine grosse Zahl von Platin-diaminkomplexen
aufgeführt. Unter diesen Verbindungen wird auch das Cis-Platin-dichlordiamin für die Behandlung von Krebs
genannt. Auch dieser Artikel gibt jedoch als wichtigsten Nachteil der Verbindungen ihre Nierentoxizität an.
Auch in Chem. and Eng. News, Juni 6, 1977, Seiten 29-30,
wird das Cis-Platin-diamindichlorid und seine Verwendung als
Krebsbehandlungsmittel beschrieben. Auch gemäss diesem Artikel stellt die Nierentoxizität der wichtigste Nachteil
der Verbindung dar.
Aus dem Artikel in Cancer Chemotherapy Reports, Teil 1, Vol.59, Nr. 3, Mai/Juni 1975, Seiten 629-641, folgt ebenfalls
die Nierentoxizität von Cis-Platin-diamindichlorid (II). Wegen dieser Nierentoxizität und wegen des darauf beruhenden
niederen therapeutischen Indexes wurden andere Platinkomplexe im Hinblick auf ihre Wirksamkeit bei der Krebsbehandlung
untersucht.
In den nicht vorveröffentlichten NL-Patentanmeldungen
78.07334 und 79.04740 werden neue Platin (II)-diaminkomplexe beschrieben. Diese sind ebenfalls geeignet für die Behandlung
von Krebs, zeigen aber niedere bis gar keine Nierentoxizitäten.
Die genannten Patentanmeldungen betreffen Komplexe mit zweiwertigem Platin, die Liganden aufweisen, welche zwei Koordinationsstellen

Der zwei Koordinationsstellen absättigende Ligand ist ein eventuell substituiertes Propandiamin. Die genannten Verbindungen
zeigen, wegen der Natur der Substituenten R., R_ ,
R-. und R. eine tiefe oder gar keine Nierentoxizität.

Hinsichtlich der Verwendung von Platin-diajninkomplexen mit
vierwertigem Platin für die Behandlung von Krebs ist der einschlägigen Literatur das Folgende zu entnehmen:
Der oben genannten Artikel von Rosenberg und Van Camp beschreibt zum ersten Mal die Antitumoraktivität eines Platin(IV)
komplexes, des Cis-Platin(IV)-diamin-tetrachlorides gemäss
Formel (2).

Diese Verbindung wird, zusammen mit einer grossen Zahl anderer Platin(IV)-komplexe, auch durch M.L.Tobe und
A.R. Khokhar im Journal Clinical Hematol.Oncol. , 7 (1M1977),
Seiten 114-134, beschrieben. Als eine Koordinationsstelle absättigende Liganden dienen zwei primäre Amine. Solche Verbindungen
entsprechen der Formel (3).

Aehnliche Verbindungen werden auch in der NL-Patentanmeldung
78.10431 beschrieben. Auch hier liegen immer eine Koordinationsstelle
absättigende Liganden vor. Auch diese Verbindungen entsprechen der Formel (3), wobei R die allgemeine

Platin(IV)-komplexe mit zwei Koordinationsstellen absättigende
Aminliganden, bei denen die Amingruppen durch zwei C-Atome (Aethylengruppe) voneinander getrennt sind, sind im Journal
Clinical Hematol.Oncol., 7(1) (1977), Seiten 231-241 und
in der NL-Patentanmeldung 79.03048 beschrieben. Die Verbindungen entsprechen der Formel (4)

Platin(IV)-komplexe mit zwei Koordinationsstellen absättigenden
Aminosäureliganden, in denen zudem das Platin teilweise mit Stickstoff und teilweise mit Sauerstoffgruppen komplexiert
ist, sind in der NL-Patentanmeldung 79.03050 beschrieben.
Die vorliegende Erfindung betrifft Platin(IV)-diaminkomplexe
der Formel (5)

Die genannten Komplexe sind im Patentanspruch 1 charakterisiert.
Entsprechend ihrer hohen Antitumoraktivität und ihrer
niederen Nierentoxizität sind Verbindungen gemäss der Formel (6), speziell solche, in denen mindestens eine der Gruppen
R1 oder R mehr als ein C-Atom aufweisen, bevorzugt.
Speziell bevorzugte Verbindungen sind diejenigen der Formel (7 bis 13) und ausgezeichnet speziell bevorzugt sind die
beiden Verbindungen Cis-Dichlor-trans-dihydroxy-1,1-bis-(aminomethyl/cyclohexanplatin(IV)
der Formel (7) und cistetrachlor-1,1-bis(aminomethyl)-cyclohexanplatin
(IV) der Formel (8).
In den Formeln (5) und (6) sind die anionischen Gruppen X bevorzugterweise Chlor, Brom, Iod oder das Sulphatradikal.
Die gleiche anionische Gruppe kann aber auch das substituierte oder nicht substituierte Carboxylradikal wie Azetat oder
substituiertes Azetat, Oxalat, Malonad oder substituiertes Malonat oder das 4-carboxyphthalat sein. Die anionische
Gruppe Y ist (unabhängig von X) bevorzugterweise Chlor, Brom oder Iod, die Hydroxylgruppe oder die Nitratgruppe.
Die Erfindung erstreckt sich auch auf die Herstellung mittels an sich bekannter Verfahren der genannten Verbindung, auf
ein Verfahren für die Herstellung der diese Verbindungen enthaltenden Heilmittel sowie auf die derart erhaltenen Heilmittel.

Ausgedehnte Untersuchungen haben gezeigt, dass die erfindungsgemässen
Verbindungen eine hohe therapeutische Aktivität gegen Krebs zeigen. Im Gegensatz zu denjenigen
Platinkoraplexverbindungen, die bis anhin bekannt worden sind und praktisch ausschliesslich für die Krebsbehandlung
eingesetzt worden sind, beispielsweise im Gegensatz zum Cis-Platin-diaminchlorid (PDD), hat es sich gezeigt, dass die
erfindungsgemässen Verbindungen eine niedere oder gar keine Nierentoxizität aufweisen.
Die Erfindung wird nun anhand der folgenden Beispiele erläutert.
Die Komplexe werden allgemein so hergestellt, dass zuerst das Platin(II)-produkt erhalten wird, welches dann mittels
eines Oxydierungsmittels zur entsprechenden Platin(IV)-Verbindung umgesetzt wird.
Das Platin (II)-produkt der allgemeinen Formel Cis-LPtCl_,
in der L für das Diamin als Zweikoordinationsstellen absättigender
Ligand im Komplex, wird gemäss der Methode von G.L. Johnson, Inorg.Synth. VIII, Seiten 242-244 hergestellt.
Vom gesuchten Diamin wird zuerst das di-HCl-Salz hergestellt.
Dieses wird dann in Wasser gelöst und mit einer aequimolaren Menge K3PtCl4 versetzt. Die Mischung wird dann auf 950C
erwärmt und mit einer aequimolaren Menge NaOH im Wasser so schnell versetzt, dass der PH bei ungefähr 6 verbleibt.
Der ausgefallene, hellgelbe Feststoff wird abfiltriert, mit Wasser gewaschen und dann getrocknet. Das Produkt kann durch
Rekristallisation aus DMF gereinigt werden.

Das CiS-LPtCl2(II) wird mit gasförmigem Chlor zu CiS-LPtCl4
-(IV) oder mit 30
Cl2(IV) oxydiert.

-(IV) oder mit 30%-igem Wasserstoffperoxyd zu Cis-LPt-(OH)_-
Die Umsetzung zu CiS-LPtCl4(IV) wird durch G.B. Kaufman in
Inorg. Synth VII, Seiten 236-238 beschrieben.
Die Oxydation mittels Chlor geschieht so, dass das Cis-LPtClim
Wasser suspendiert wird. Durch die Suspension wird etwa 1 Stunde lang bei 70 bis 750C gasförmiges Chlor durchgeleitet,
wodurch der zweiwertige Platinkomplex zum vierwertigen oxydiert wird. Anschliessend wird Luft durch die
Suspension geleitet, um überschüssiges Chlor zu entfernen.
Dazu ist es nötig, die Luft etwa 5 Minuten lang durchzuleiten und eine Temperatur von etwa 700C beizubehalten. Die Mischung
wird abgekühlt, das Produkt abfiltriert, mit Wasser gewaschen und unter reduziertem Druck getrocknet.
Die Oxydierung zum Cis-LPt(OH)2Cl_(IV) wird dadurch erreicht,
dass eine Suspension von CiS-LPtCl2(II) etwa eine halbe Stunde
lang mit einem üeberschuss von 30%-igem Wasserstoffperoxyd
auf Siedetemperatur gehalten wird. Die Suspension wird dann abgekühlt, das Produkt abfiltriert, mit Wasser gewaschen und
unter reduziertem Druck getrocknet.
Die Umsetzung von CIs-LPt(OH)2Cl2(IV) zu CiS-LPt-Cl4(IV) kann
auch durch Erhitzen einer Suspension von CiS-LPt(OH)2Cl2(IV)
während fünf Minuten auf 1000C in konzentrierter Salzsäure geschehen.

Die letztgenannten zwei Reaktionen sind durch F. Basolo,
J.C. Bailar jr. und B. Rapp-Tarr im Journal of American Chemical Society, 72, 2433 (1950) beschrieben. Sie wurden
in der obigen Spezifikation leicht modifiziert (Siedetemperatur anstelle von 800C, Verwendung von 30%-igem H-O,, anstelle von
10%-igem H3O2 und 50 bis 70 Ueberschuss H2O3 anstelle von 10).
Cis-Dichlor-trans-dihydroxy-l,1-bis-(aminomethy1)cyclohexan
platinum(IV) gemäss Formel (7)
1,2 g Cis-Dichlor-1,1-bis(aminomethyl)-cyclohexan platin(II)
werden in 5 ml destilliertem Wasser duspendiert. Zur Suspension werden 25 ml 30%-iger Wasserstoffperoxyd gegeben.
Die Mischung wird eine halbe Stunde lang bei Raumtermperatur gerührt, anschliessend eine Stunde lang unter Rückflussbedingungen.
Die Suspension wird dann abgekühlt und der Feststoff abfiltriert. Dieser wird dann mit Wasser gewaschen und
unter reduziertem Druck getrocknet. Das Gewicht der leicht gelben, festen Substanz beträgt 0,45 g.

Cis-Tetrachlor-1,1-bis (aminoniethyl) -cyclohexan platin (IV)

1/2 g Cis-Dichlor-1,1-bis(aminomethyl)cyclohexan platin (II)
werden in 15 ml destilliertem Wasser suspendiert. Die Suspension wird auf 700C erwärmt, worauf, unter Rühren, eine
Stunde lang gasförmiges Chlor durch die Suspension geleitet wird. Das überschüssige Chlor wird anschliessend mittels
Durchleiten von Luft entfernt, wobei die Temperatur auf etwa 700C gehalten wird. Die Reaktionsmischung wird abgekühlt
und der ausgefallene Feststoff abfiltriert. Dieser wird dann mit Wasser gewaschen und unter reduziertem Druck
getrocknet.
Das Gewicht der erhaltenen, gelben festen Substanz betrug 0,9 g, was einer Ausbeute von 63% entspricht.

Das Cis-Dichlor-trans-dihydroxy-1,1-bis(aminomethyl) cyclohexan
platin (IV) wurde gemäss dem Verfahren von Beispiel 1 hergestellt. Anschliessend wurde Salzsäure zur Suspension
gegeben. Die Lösung wurde nun 5 Minuten lang auf 95-1000C
erwärmt und darauf abgekühlt. Das Produkt wurde abfiltriert und mit Wasser gewaschen.
Die Verbindung wurde sowohl mittels 1H-NMR- wie auch mittels
IR-Spektrum untersucht. Es zeigte sich, dass sie die gleichen Spektra aufwies wie die Verbindung aus dem Beispiel 2.

Diese Verbindung wurde mittels eines Verfahrens gewonnen, das analog demjenigen des Beispiels 2 war. Ausgegangen wurde

Die Auswage betrug 1,3 g, was einer Ausbeite von 69% entspricht.

- 6. Verwendung der Platin(IV)-diaminkomplexe der Formeln (5) und (6) gemäss Patentanspruch 1 oder 2 in Krebsheilmitteln.130047/0410
