Description
in der R ein Wasserstoffatom oder eine niedere Alkylgruppe, m eine ganze Zahl von 0 oder 1 und
Yi, Y2 und Yi jeweils ein Wasserstoffatom, eine gleiche
oder unterschiedliche gerade oder verzweigte niedere Alkylgruppe mit 1 bis 3 Kohlenstoffatomen, eine
niedere Alkoxygruppe, Carboxylgruppe, Carboalkoxygruppe, Hydroxylgruppe, Trifluormethylgruppe
oder ein Chloratom bedeuten.
Die erfindungsgemäßen Verbindungen sind neu und aus der Literatur bisher nicht bekannt. Sie haben eine
entzündungshemmende und eine den Serumcholesterinspiegel senkende Wirkung und dienen folglich
in entsprechendenden Zubereitungen als wirksame Antiphlogistika und als Verhütungs- oder Heilmittel
gegen Arteriosklerose.
Die erfindungsgemäßen Verbindungen können auf verschiedenartige Weise hergestellt werden. Ein typischer
Herstellungsweg besteht darin, ein reaktionsfähiges Zwischenprodukt der IH-Dihydropimarsäure
(111), beispielsweise ein Säurehalogenid oder Säureanhydrid,
vorzugsweise ein !"-Dihydropimaroylhalogenid,
in an sich bekannter Weise mit normalerweise der ein- bis dreifachen Molmenge eines Amins der
folgenden allgemeinen Formel
H2N(CHR)
umzusetzen, in der R, /n, Y1, Y2 und Y3 die vorstehend
angegebene Bedeutung haben. Bei dieser Umsetzung können gute Ergebnisse erzielt werden, wenn die 1- bis
l,5fache Molmenge einer anorganischen Base, wie Alkalicarbonat oder Alkalihydroxid, oder einer tertiären
organischen Base, wie N-Dialkylanilin oder
Triäthylamin, zur Dehydrohalogenierung eingesetzt werden.
Die Umsetzung erfolgt, ganz nach den jeweiligen Erfordernissen, unter Kühlung, unter Erhitzen ader
bei Raumtemperatur und ist gewöhnlich innerhalb von 1 bis 10 Stunden beendet. Die Umsetzung zwischen
dem Säurehalogenid und dem jeweils verwendeten Amin verläuft bei Raumtemperatur oder
unter Wasserkühlung rasch und ist üblicherweise innerhalb von drei Stunden beendet. Die Beendigung
der Reaktion kann leicht aus der Verminderung oder dem Verschwinden der Ausgangsstoffkomponenten
aus dem Silikagel-Dünnschichtchromatogramm abgelesen werden. Nach Beendigung der Reaktion
kann das gewünschte I8-Dihydropimarsäureamid-Derivat aus dem Reaktionsgemisch abgetrennt und
nach bekannten Methoden gereinigt werden. Falls ein mit Wasser mischbares Lösungsmittel bei der Re-
j5 aktion verwendet wird, wie Aceton, wird das entsprechende
Reaktionsprodukt durch Zusatz zu einer großen Menge Wasser (gewöhnlich der 10- bis
20fachen Menge) zur Gewinnung des gewünschten Rohproduktes abgetrennt.
Falls ein mit Wasser nicht mischbares Lösungsmittel verwendet wird, wie Benzol, Toluol, Xylol,
η-Hexan oder Chloroform, wird die Reaktionslösung zunächst mit verdünnter Mineralsäure und dann mit
Wasser gewaschen, wonach die organische Lösungsmittelschicht mit wasserfreiem Natriumsulfat oder
Magnesiumsulfat entwässert und filtriert wird. Schließlich wird das Filtrat eingeengt, wobei das Rohprodukt
der entsprechenden I8-Dihydropimarsäureamid-Derivate erhalten wird.
Das erhaltene Rohprodukt kann durch Umkristallisieren aus einem geeigneten Lösungsmittel, wie Methanol
oder Aceton, oder durch Passieren einer Silikagelsäule und Eluieren des Produktes mit Benzol
leicht gereinigt werden.
Pimarsäure (IV) ist in relativ hoher Konzentration in Kiefernharz enthalten. Die Säure wird, ohne aus
dem Harz isoliert zu werden, in ein anorganisches Alkalisalz, wie das Natriumsalz, umgewandelt. Dann
wird das Salz mit einem geeigneten Reduktions-
bo katalysator, wie Raney-Nickel, zu I814-Dihydropimarsäure
reduziert. Diese kann durch Umkristallisieren aus einem organischen Lösungsmittel, wie Dekahydronaphthalin,
leicht gereinigt werden. Dann wird zur Herstellung von !"-Dihydropimarsäure (111)
b5 mit Chlorwasserstoff die Doppelbindung umgelagert.
Nach dem Umkristallisieren kann das Produkt als Ausgangsstoff für die Verbindungen nach der Erfindung
eingesetzt werden. Es kann auch leicht da-
durch gereinigt werden, daß zunächst handelsübliches reduziertes Kiefernharz, wie Kolophonium, einer
Isomerisierung mit Chlorwasserstoff unterzogen und dann auf die beschriebene Weise weiterbehandelt wird.

einem Überschuß von Thionylchlorid und 1,0 g (3,3 mMol) ^-Dihydropimarsäure) in 5 ml Aceton
erhalten worden war. Nach dreistündigem Rühren wurde die Reaktionslösung in 30 ral 5%ige verdünnte
Salzsäure gegossen, und die ausgeschiedenen Kristalle wurden abfiltriert und mit Wasser gewaschen. Nach
dem Trocknen wurde das Produkt zweimal aus Äthylacetat umkristallisiert, filtriert und getrocknet. Es
wurden 1,27 g der Titelverbindung als farblose Blättchen erhalten. Ausbeute 87%, Fp. 139 bis 1410C.

Auf gleiche Weise wurden noch die folgenden Verbindungen hergestellt:
Der Ausgangsstoff ist somit sehr stabil und stellt eine billige und leicht erhältliche Verbindung dar.
Die überführung in wertvolle Arzneimittel bietet somit eine neue Verwendungsmöglichkeit dieser reichlich
vorhandenen und preiswerten Kiefernharzkoniponenten.
Die Herstellung des als Ausgangsmaterial verwendeten I8-Dihydropimarsäureanhydridsoder I8-Dihydropimaroylhalogenids
aus der Verbindung der Formel III kann in üblicher Weise !eicht erfolgen.
So kann das I8-Dihydropimarsäureanhydrid aus der
freien Säure mit einem Dehydratisierungsmittel, wie Essigsäureanhydrid oder Acetylchlorid, erhalten werden,
während die Bildung von I8-Dihydropimaroylhalogenid
leicht durch Umsetzen der freien Säure mit einem Halogenierungsmittel, wie PX5, PX3 oder
SOX2 (wobei X ein Halogenatom ist) vorgenommen werden kann.
Die Amine der allgemeinen Formel II, die der andere Ausgangsstoff für die Verbindungen nach der
Erfindungsind, sind Anilin, 2,6-Dimethylanilin, 2,4-Dimethylanilin,
2,4,6-TrimethylaniIin, 2-Methylanilin,
4 - Methylanilin, 2 - Äthylanilin, 2,6 - Diäthylanilin, 2,6 - Diisopropylanilin, m - Anisidin, ο - Anisidiri,
ο - Phenetidin, p-Phenetidin, 2,3-Dimethoxyanilin,
p-Aminosalicylsäure und ihre Ester,p-Aminobenzoesä.ure
und ihre Alkylester, ringsubstituierte Aniline (wie Trifiuormethylanilin, 3-Chlor-p-toluidin, 3,4-Dichloranilin
oder 2,6-Dichloranilin), ringsubstituierte Benzylamine (wie o-Methylbenzylamin, p-Methylbenzylamin,
m - Methylbenzylamin, ο - Methoxybenzylamin, o-Chlorbenzylamin, m-Chlorobenzylamin
oder p-Chlorbenzylamin) oder Λ-Niederalkylbenzylaminderivale(wieix-Äthylbenzylamin,
Y-Methylbenzylamin oder I-Phenylhexylamin).
Die Erfindung wird nachstehend anhand von Beispielen im einzelnen erläutert.
Herstellung von N-(2-Äthylphenyl)-l8-dihydropimarsäureamid
Ein Gemisch von 0,5 m!Triäthylamin,0,5 go-Äthylaniliin
(4,2 mMol) und 5 ml Aceton wurde unter Eiskühlung gerührt. Dann wurde dem Gemisch unter
Rühren eine Lösung zugesetzt, die durch Auflösen von rohem !"-Dihydropimaroylchlorid (erhalten aus

Die voi teilhaften Eigenschaften der beanspruchten Verbindungen gehen aus folgenden Vergleichsversuchen
hervor, bei denen die d>,n Blutcholesterinspiegel
senkende Wirksamkeit der erfindungsgemäßen χ Verbindungen und von bekannten Verbindungen
gleicher Wirkungsrichtung bei Ratten ermittelt wurde.
Männliche Wistar-Ratten mit einem Gewicht von 50 bis 60 g wurden in Gruppen von je sechs Tieren
eingeteilt. An eine Gruppe (Grundgruppe) wurde nur eine synthetische Normaldiät verabreicht, während
einer weiteren Gruppe (Vergleichsgruppe) eine 1% Cholesterin und 0,25% Natriumcholat enthaltende
synthetische Diät gegeben wurde. Den übrigen Gruppen (Testgruppen) wurde eine 1 % Cholesterin, 0,25%
Natriumcholat und eine vorgegebene Menge jeder zu untersuchenden Verbindung enthaltende synthetische
Diät gegeben.
Nachdem die Ratten in dieser Weise drei Tage lang gefüttert worden waren, ließ man sie über Nacht
fasten und entnahm dann Blut. Das Blutserum wurde abgetrennt, und der Cholesteringehalt des Serums
wurde mit einem automatischen Technicon-Analysegerät bestimmt.
Die den Blutcholesterinspiegel senkende Wirksamkeit
der Verbindungen wird nach der folgenden Formel errechnet:

In der vorstehenden Tabelle bedeutet *), daß der Serumcholesterinspiegel statistisch wesentlich niedriger
als der der Vergleichsgruppe bei der Risikowahrscheinlichkeit von 5% (p < 0,05) ist. Ebenso bedeutet
**), daß der Serumcholesterinwert wesentlich niedriger ist bei der Risikowahrscheinlichkeit von
1% {p < 0,01 ((Students t-Test).
Aus den Versuchsergebnissen wird folgendes deutlich: Bei der durch cholesterinhaltige Nahrung verursachten
Hypercholesterolämie zeigen die erfindungsgemäßen Verbindungen, wenn sie im Gemisch mit
der Nahrung verabreicht werden, eine stärkere Wirkung bei einer kleineren Dosis als die Linolsäureamide,
die als gleichartig wirkend angesehen werden, oder als /i-Sitosterin.
Claims (2)
- Patentansprüche:I. I8 - Dihydropimarsäureamid - Derivate der allgemeinen FormelY1in der R ein Wasserstoffatom oder eine niedere Alkylgruppe, m eine ganze Zahl von 0 oder 1, Yi, Y2 und Y3 jeweils ein WasserstofTatom oder eine gleiche oder unterschiedliche gerade oder verzweigte niedere Alkylgruppe mit 1 bis 3 Kohlenstoffatomen, eine niedere Alkoxygruppe, Carboxylgruppe, Carboalkoxygruppe, Hydroxylgruppe oder Trifluormethylgruppe oder ein Chloratom bedeuten.
- 2. Entzündungshemmendes und den Serumcholesterinspiegel senkendes Arzneimittel, enthaltend eine Verbindung gemäß Anspruch 1 neben den üblichen inerten Trägerstoffen und Verdünnungsmitteln.Die Erfindung betrifft I8-Dihydropimarsäureamid-Derivate der allgemeinen FormelY1
