Description

Pharmaceutical composition and preparation method and application thereof
Technical Field
The invention relates to a pharmaceutical composition, a preparation method and application thereof.
Background
With the development of social economy and the conception of prepotency, children's health is the focus of attention. Respiratory and digestive diseases are common morbidity trends in children. In addition, for children with weak spleen and stomach, respiratory tract infection usually causes digestive system problems directly, such as gastrointestinal type common cold in children. The nose is the first line of the respiratory tract, 1 item of epidemiological investigation on western regions shows that the incidence rate of allergic rhinitis accounts for more than 30 percent, and then retrospective analysis of national allergic rhinitis within 10 years shows that the incidence rate is in a stable state. Allergic rhinitis is an allergic disease caused by a sensitization source and caused by inflammatory factors released by an immune system, is closely related to infantile asthma, and is discovered in early researches that the incidence rate of the infantile asthma suffering from the allergic rhinitis is higher than that of other infants suffering from the allergic rhinitis. Furthermore, with the opening of the two-fetus policy and the development of medical technology, the number of cesarean section is increasing year by year, and 1 in the recent epidemiological investigation of the jinmei on allergic rhinitis and asthma of the cesarean section and children patients, the cesarean section is related to allergic rhinitis and asthma of children, and the children patients with family allergy history are more obvious. Children with respiratory diseases are large in number and are easily accompanied by digestive diseases.
With the development of social economy, the acceleration of urbanization and environmental pollution, allergic diseases of children tend to increase year by year, allergic rhinitis is manifested by a large amount of watery nasal discharge, sneezing and the like, and diseases such as chronic rhinosinusitis, nasal polyp and the like can be caused by delayed period. Influences intelligence development and physical development, children suffer from rhinitis for a long time, and respiratory obstruction can lead the brain to lack of oxygen to cause dizziness, lethargy, concentration failure, learning achievement reduction and the like, and has obvious influence on the brain of the children, including cognitive ability, memory and psychological activities. In addition, children can breathe with the mouth due to unsmooth expiration, and pharyngitis is caused by direct stimulation to the pharyngeal cavity, so that the lips of the children are thickened, teeth grow outwards, and face deformity is caused. While western medicine mainly treats allergic rhinitis of children with symptomatic treatment such as antihistamine, hormone, decongestant and the like, the long-term repeated use of the western medicine can cause adverse reactions such as nasal concha atrophy, nasal cavity dryness and the like. The traditional Chinese medicine has good treatment effect on allergic rhinitis and chronic rhinosinusitis and has no other side effect.
The traditional Chinese medicine of the constitution of children considers that the children are easy to be deficient and easy to be excessive, and easy to be cold and easy to be hot, and the children cannot express the disease, so that the pediatrics is called as the dumb department, the excessive syndrome of the disease occurs rapidly, and the deficient disease can be diagnosed only by carefully discovering that a doctor looks through the body, but cannot be treated rapidly. Therefore, the prevention of the disease before onset is more important in pediatrics than the treatment of the disease immediately. And the guidelines of western medicine on respiratory diseases such as allergic rhinitis and asthma are turned over to eliminate allergens, and the western medicine is mainly used for symptomatic treatment. And allergic diseases occur repeatedly, and are mostly taken for a long time, and children are difficult to take medicine, and atomization and the like are often not matched, so that effective treatment is difficult.
In the domestic market, products aiming at the weakness of the spleen and the stomach of children such as 'fat baby powder' and 'spleen-strengthening paste' exist, but the products only aim at children with the weakness of the spleen and the stomach, but cannot be used for children with the symptoms of the weakness of the spleen and the stomach, which are affected by respiratory diseases and lung qi weakness, and at present, products for performing long-term intervention on allergic rhinitis and chronic rhinosinusitis of children are lacked.

1. General data
59 cases of the infants are all the pediatric outpatients and inpatients of subsidiary hospitals of Chengdu Chinese medicine university, and meet the diagnosis standard established in the national small respiratory tract conference in 1987.
Diagnostic criteria:
repeating upper respiratory tract infection every year for 7 times in 0-2 years old;
② repeatedly upper respiratory tract infection for 6 times per year in 3-5 years old;
③ repeated upper respiratory tract infection for 6-14 years old every 5 times a year.
The concept of repetitive upper respiratory tract infections: repeated upper respiratory tract infection is mostly caused by virus infection, is an inflammatory reaction of repeated attacks of the upper respiratory tract, and clinical symptoms mainly include repeated fever, rhinorrhea, jet printing, cough, red throat, tonsil swelling and the like.
29 control cases, 1 case of abscission. 13 men and 14 women; the age is 6 months to 10 years old, the average age is 4.8 years old, and the number of attacks per year is 5 to 13. The treatment group comprises 30 cases, 16 cases of men and 14 cases of women, the age is 7 months-10 years, the average age is 5.2 years, and the number of attacks per year is 6-13. The disease course of the two groups is 0.4-2 years. The difference between the two groups of children patients in terms of sex, age and disease course has no statistical significance (P is less than 0.05), and the children patients have comparability.
2. Method of treatment
Control group: the symptomatic treatment (such as defervescence treatment, cough relieving, phlegm reducing, fluid infusion and the like, and the antibiotic treatment can be given under the condition of bacterial infection.
The treatment group is administered 1 bag each time and 1 time each day with boiled water for 3 months continuously under conventional treatment.
3. Standard of therapeutic effect
The effect is shown: in the latter half year after the treatment course of 3 months, the attack times are reduced by more than 2/3 compared with before treatment, the symptoms are obviously relieved during attack, and the treatment course is obviously shortened.
The method has the following advantages: after the 3-month treatment course, the disease incidence frequency in the latter half year is reduced by more than 1/2, the symptoms are relieved during disease incidence, and the treatment course is shortened.
And (4) invalidation: the frequency of disease onset after treatment is reduced to 1/2, and the symptoms and the treatment course are not obviously improved.
The total effective rate is significant effect plus effective.
4. Results
TABLE 1 comparison of treatment results of the treated groups with the control group
Figure GDA0002817652870000051
Figure GDA0002817652870000061
As can be seen from Table 1, the total effective rate of the treatment group using the pharmaceutical composition of the invention to the children recurrent respiratory tract infection is as high as 86.7%, which is significantly better than 55.5% of the control group, and the pharmaceutical composition of the invention has excellent treatment effect on the children recurrent respiratory tract infection.
Test example 2 sensory evaluation of the pharmaceutical composition of the present invention
Firstly, the taste is compared by adding different sugar
The product is a children product, no additive is recommended, and the composition prepared in the embodiment 1 of the invention is powder, is not suitable for being flavored by maltose and honey, and is added and compared with different types of sugar. 2g of white sugar, 2g of brown sugar and 2g of rock candy are added into the prescription amount per day respectively for comparing the taste, and the results are shown in a table 2.
Table 2 mouthfeel of products with different sugar additions
Figure GDA0002817652870000062
Therefore, the taste of the composition is greatly influenced by adding different kinds of sugar, wherein when 2g of white sugar is used, the taste is burnt after being mixed with boiled water, the white sugar is dark brown paste, the taste is fine and smooth, the fragrance is natural, the sweetness is slightly bitter, the taste is similar to that of sesame paste, and the taste is good; when other saccharides are added, the taste is not good, for example, brown sugar is added, and the taste is bitter; when crystal sugar is added, the taste is slightly bitter and sticky.
Secondly, the mouthfeel of the white sugar added with different dosages is compared
The product is a children product, does not suggest to use additives, is added with white sugar for flavoring, and simultaneously optimizes the addition amount of the white sugar. The original bitterness and sweetness of the sugar were compared by adding 0.5g, 1g, 1.5g, 2g, 2.5g of white sugar to the recipe one day, respectively, and the results are shown in Table 3.
Table 3 mouthfeel of products with different amounts of white sugar added
Figure GDA0002817652870000063
Therefore, different amounts of the white sugar have influence on the mouthfeel of the composition, wherein when 2g of the white sugar is used, the bitter taste disappears, the sweetness is proper, and the mouthfeel is optimal.
Therefore, the composition has good mouthfeel and is suitable for children.
In conclusion, the composition has reasonable compatibility, can be used for relieving various symptoms caused by respiratory system diseases of children, has obvious curative effect on repeated respiratory tract infection of children and has high clinical cure rate; the composition has good safety, good taste and high pediatric acceptance, and provides a new medicine selection for clinic.
