Abstract

Description
技术领域
本发明涉及促进角膜内皮细胞粘附的药剂(agent),它用于角膜内皮细胞的粘附、维护或保护。
背景技术
当从角膜(在眼球的最前部的透明组织)进入的光线到达视网膜激发视网膜神经细胞时,认识到视觉信息,并且所产生的电信号经由视神经被传输到脑视觉皮层。为了具有良好的视力,该角膜需要是透明的。当利用角膜内皮细胞的泵吸功能和阻隔功能将水含量保持在恒定水平时,维持角膜的透明度。
人的角膜内皮细胞的密度在出生时是约3000个细胞/1mm2。一旦损伤,该细胞丧失再生的能力。在由于各种原因引起的角膜内皮的功能性障碍所造成的内皮角膜营养不良和大疱角膜病中,角膜会发生水肿和变得不透明,并且该视力明显下降。目前,大疱角膜病是通过其中角膜的上皮、主质和内皮的整个三层结构被移植的穿透性角膜成形术来治疗的。然而,角膜的捐献在日本是不够的,并且虽然有大约5500名的角膜移植术等待患者,但是每年在日本进行约2700例角膜移植。
近年来,仅仅移植损伤组织的“部分移植”的想法已经引起人们的注意以减少排异反应和手术后并发症的风险,因此获得更好的视觉功能。在角膜移植中,已经进行了仅仅移植角膜上皮的上皮移植,移植口腔粘膜而不是移植角膜上皮的已培养的口腔粘膜上皮移植,移植主质组织的深层式板层角膜移植术等等。也考虑了仅仅移植角膜内皮的方法。对于角膜内皮的移植,由在胶原层培养的角膜内皮层组成的角膜内皮状片是已知的(参见专利文件1和2)。然而,至于角膜内皮细胞,特别地从人衍生的那些,因为角膜捐献者的数量是有限的并且离体培养是困难的,因此以移植所需的数量生产培养细胞需要花费时间和成本。
人胚胎茎(ES)细胞显示出高自复制性和多潜能性,并且从药物应用方面引起人们的注意。然而,它具有急剧减少的细胞数的实际问题,因为在培养步骤中分散细胞的手术容易引起细胞死亡。最近已经发现在人ES细胞的培养过程中发生的细胞死亡是由Rho激酶(ROCK)的活化所引起,并且ROCK的抑制显著地抑制细胞死亡,和报道说,通过使用ROCK抑制剂如Y-27632等,人ES细胞的大量培养和脑细胞的产生能够可行(非专利文件1)。
Rho激酶(ROCK)抑制剂已知具有各种作用。例如,专利文件3公开了Rho激酶抑制剂促进角膜轴索形成并且Rho激酶抑制剂用作恢复角膜感知的药剂。另外,专利文件4公开了Rho激酶抑制剂对于视网膜的神经节细胞具有轴突长出促进作用,并且用于视觉功能紊乱的医治。

本发明的效果
促进本发明的角膜内皮细胞的粘附的药剂含有Rho激酶抑制剂作为活性成分。促进本发明的角膜内皮细胞的粘附的药剂促进角膜内皮细胞的粘附和使得形成具有良好细胞形态和高细胞密度的角膜内皮细胞层。因此,它可用作伴有角膜内皮功能紊乱的疾病如大疱角膜病和角膜内皮炎的治疗剂或预防剂。本发明的促进角膜内皮细胞的粘附的药剂可用作在伴有角膜内皮功能紊乱的疾病的治疗或预防中保护角膜内皮的药剂。此外,本发明的促进角膜内皮细胞的粘附的药剂可用作在与眼内手术如白内障手术、玻璃体手术等等相关的角膜内皮功能紊乱,由增大的眼内压引起的角膜内皮功能紊乱(特别地青光眼侵袭),或由接触透镜引起的不充足的氧气所引起的角膜内皮功能紊乱的治疗或预防中保护角膜内皮的药剂。因为本发明的培养介质或角膜保存溶液含有Rho激酶抑制剂,所以角膜内皮细胞能够很好地培养、维持或保存,并确保了角膜内皮制剂的稳定的供应、维持或保存。

实施本发明的最佳模式
本发明的促进角膜内皮细胞的粘附的药剂(以下有时简写为“本发明的粘附促进剂”)含有Rho激酶抑制剂作为活性成分。
在本发明中,Rho激酶指与Rho的活化一起活化的丝氨酸/苏氨酸激酶。例子包括ROKα(ROCKII:Leung,T.et al.,J.Biol.Chem.,270,29051-29054,1995),p160ROCK(ROKβ,ROCK-I:Ishizaki,T.et al.,TheEMBO J.,15(8),1885-1893,1996)和具有丝氨酸/苏氨酸激酶活性的其它蛋白质。
在本发明中,一种类型的Rho激酶抑制剂可以单独被含有,或几种类型可以根据需要而相结合被含有。

这些当中,(+)-反式-4-(1-氨基乙基)-1-(4-吡啶基氨基甲酰基)环己烷,1-(5-异喹啉磺酰基)高哌嗪和其可药用盐是优选使用的,因为它们在角膜内皮细胞的粘附的促进上是特别优异的。作为化合物的盐,药物学上可接受的酸加合盐是优选的。该酸的例子包括无机酸,如盐酸,氢溴酸,硫酸等等,有机酸,如甲磺酸,富马酸,马来酸,苦杏仁酸,柠檬酸,酒石酸,水杨酸等等。更优选的是(+)-反式-4-(1-氨基乙基)-1-(4-吡啶基氨基甲酰基)环己烷2盐酸盐和1-(5-异喹啉磺酰基)高哌嗪盐酸盐。
在本发明中,“角膜内皮细胞的粘附的促进”的例子包括在角膜内皮细胞之间的粘附的促进和角膜内皮细胞对培养底物的粘附的促进两者。
本发明的粘附促进剂提供对于从哺乳动物(例如,人,小鼠,大鼠,仓鼠,兔子,猫,狗,牛,绵羊,猴子等等)的角膜组织中所分离的角膜内皮细胞或对于从其分离的并且传代(passaged)的角膜内皮细胞的粘附促进作用。因为本发明的粘附促进剂在来源于人的角膜内皮细胞的粘附促进作用上是优异的,来源于人的角膜内皮细胞被认为特别难以培养和传代(passage),人来源的角膜内皮细胞是优选的靶。
角膜内皮细胞在维持角膜的透明度上起着作用。当内皮细胞的密度在某个范围下降时,该角膜产生溶胀并且变得无法维持透明度,据此产生了角膜内皮功能紊乱。本发明的粘附促进剂促进角膜内皮细胞的粘附,使得具有良好细胞形态和高细胞密度的角膜内皮细胞层能够形成,并且进一步显示出对角膜内皮细胞的凋亡抑制作用。因此,它可用作伴有角膜内皮功能紊乱的疾病如大疱角膜病和角膜内皮炎的治疗剂或预防剂。另外,本发明的粘附促进剂可用作与眼内手术如白内障手术、玻璃体手术等等相关的角膜内皮功能紊乱,由增大的眼内压引起的角膜内皮功能紊乱(特别地青光眼侵袭),或由因为接触透镜导致的不充足的氧气所引起的角膜内皮功能紊乱的治疗剂或预防剂。
尽管本发明的粘附促进剂没有特别限制,只要它具有适合于局部给药于眼睛的剂型就行,但是它优选以前房内注射或眼内灌注流体形式配制。它们能够通过在本领域中广泛使用的普通方法来制备。当它以前房内注射或眼内灌注流体形式被局部施用于眼睛时,Rho激酶抑制剂和角膜内皮细胞在身体内接触,并且促进角膜内皮细胞的粘附。
例如,当本发明的粘附促进剂用作前房内注射或眼内灌注流体时,稳定剂(例如,亚硫酸氢钠,硫代硫酸钠,依地酸钠,柠檬酸钠,抗坏血酸,二丁基羟基苯等等),增溶剂(例如,甘油,丙二醇,聚乙二醇,聚氧化乙烯氢化蓖麻油等等),悬浮剂(例如,聚乙烯基吡咯烷酮,羟丙基甲基纤维素,羟甲基纤维素,羧甲基纤维素钠等等),乳化剂(例如,聚乙烯吡咯烷酮,大豆磷脂,蛋黄卵磷脂,聚氧化乙烯氢化蓖麻油,聚山梨酸酯80等等),缓冲剂(例如,磷酸盐缓冲剂,乙酸盐缓冲剂,硼酸盐缓冲剂,碳酸盐缓冲剂,柠檬酸盐缓冲剂,三羟甲基氨基甲烷缓冲液,谷氨酸,ε-氨基己酸等等),增稠剂(例如,水溶性纤维素衍生物比如甲基纤维素,羟乙基纤维素,羟丙基甲基纤维素,羧甲基纤维素等等,硫酸软骨素钠,透明质酸钠,聚羧乙烯,聚乙烯醇,聚乙烯吡咯烷酮,聚乙二醇等等),防腐剂(例如,苯扎氯铵,氯化苄乙氧铵,双氯苯双胍己烷葡糖酸盐,氯丁醇,苄醇,脱氢醋酸钠,对-羟基苯甲酸酯,依地酸钠,硼酸等等),等渗剂(例如,氯化钠,氯化钾,甘油,甘露糖醇,山梨糖醇,硼酸,葡萄糖,丙二醇等等),pH调节剂(例如,盐酸,氢氧化钠,磷酸,乙酸等等),清凉剂(例如,L-薄荷醇,d-莰酮,d-2-莰醇,薄荷油等等)等等能够作为添加剂添加进去。这些添加剂的添加量取决于添加剂的种类和用途以及其它因素来改变,并且以能够实现添加剂的目的的浓度添加。
活性成分在本发明的粘附促进剂中的量通常是0.00001-1w/v%,优选0.00001-0.1w/v%,更优选0.0001-0.01w/v%。尽管剂量和给药频率将取决于症状,年龄,体重和给药形式而变化,但是,当它用作成年人的前房内注射剂时,例如,含有0.0001-0.1w/v%、优选0.0001-0.01w/v%的活性成分的制剂通常能够通过0.01-0.1mL/每次给药的方式每天给药几次,优选1-2次,更优选1次。
当角膜内皮细胞在试管中培养时,本发明的粘附促进剂也能够被添加到培养介质中。当通过向培养介质中添加Rho激酶抑制剂来继续培养时,Rho激酶抑制剂和角膜内皮细胞在活体外接触,并促进角膜内皮细胞的粘附。
本发明提供角膜内皮细胞的培养介质,该介质含有Rho激酶抑制剂。在本发明的培养介质中所含的Rho激酶抑制剂是如上所述。
本发明的培养介质能够含有一般用于内皮细胞(例如,DMEM(GIBCO BRL),血清(例如,胎牛血清(FBS)),生长因子(例如,b-FGF),抗生素(例如,青霉素,链霉素)等等的培养的介质。
在本发明的培养介质中Rho激酶抑制剂的浓度通常是1-100μM,优选5-20μM,更优选10μM。
本发明的培养介质通过增大角膜内皮细胞的粘附来防止细胞的脱落,并且使得形成具有良好细胞形态和高细胞密度的角膜内皮细胞层。因此,它能够优选用于本发明的角膜内皮制剂的下述生产方法中。另外,本发明的培养介质也用于维持角膜内皮细胞。
本发明提供含有Rho激酶抑制剂的角膜保存溶液。在本发明的角膜保存溶液中所含的Rho激酶抑制剂是如上所述。角膜保存溶液是用于保存从捐献者所分离的角膜移植物直至移植给接受者为止的液体。
本发明的角膜保存溶液的例子包括通常用于角膜移植术的保存溶液(角膜巩膜的外植体保存溶液(Optisol GS:注册商标),角膜移植术的眼球的保存溶液(EPII:注册商标)),盐水,磷酸盐缓冲盐水(PBS)等等,它们中的每一种含有Rho激酶抑制剂。
在本发明的角膜保存溶液中Rho激酶抑制剂的浓度通常是1-100μM,优选5-20μM,更优选10μM。
本发明的角膜保存溶液通过增大角膜内皮细胞的粘附来防止细胞的脱落,并且使得形成具有良好细胞形态和高细胞密度的角膜内皮细胞层。因此,它能够用作用于器官移植等等的角膜保存溶液。另外,本发明的保存溶液也用作角膜内皮细胞的低温保藏的保存溶液。为了低温保藏,甘油,二甲亚砜,丙二醇,乙酰胺等等可以进一步被添加加到本发明的保存溶液中。
本发明提供生产角膜内皮制剂的方法,它包括使用本发明的培养介质培养角膜内皮细胞的步骤。
该角膜内皮制剂特征性地含有底物和在底物上的角膜内皮细胞层,该层已经在试管中培养。
在本发明中,底物没有特别限制,只要它能够承载所培养的角膜内皮细胞层并且能够在移植之后在体内维持它的形状至少3天。另外,当在试管中培养角膜内皮细胞时底物可具有支架(scaffold)的作用,或在培养之后可仅仅起着承载角膜内皮细胞层的作用。优选地,底物用于培养角膜内皮细胞,并且作为支架,在培养完成之后直接用于移植。
上述底物的例子包括从天然物质如胶原、明胶、纤维素等等衍生的聚合物材料,合成聚合物材料如聚苯乙烯,聚酯,聚碳酸酯,聚(N-异丙基丙烯酰胺)等等,可生物降解的聚合物材料如聚乳酸,聚乙醇酸等等,氢氧磷灰石,羊膜等等。
尽管上述底物的形状没有特别限制,只要它支持角膜内皮细胞层并且适合于移植就行,但是它优选是片。当本发明的制剂是片时,它能够在移植过程中被切成适合于应用位点的尺寸之后使用。还有可能将小片卷起,然后将它从伤口的口缘***。优选的特定例子包括覆盖病变角膜内皮区域的约80%的圆形。也优选的是在上述圆的邻近部分中形成狭缝,这样它能够紧密地粘附于应用位点。
在优选的实施方案中,上述底物是胶原。作为胶原,可优选使用描述在JP-A-2004-24852中的胶原片。该胶原片能够根据描述在上述JP-A-2004-24852中的方法从例如羊膜制备。
上述角膜内皮细胞层优选显示出下列特性中的至少一种。更优选,它显示出下列特性中的两种或多种,更优选全部。
(1)该细胞层具有单个层状结构。这是在身体内角膜内皮细胞层的诸多特性中的一种。
(2)该细胞层具有约1,000-约4,000个细胞/mm2的细胞密度。特别地,当成年人是接受者(移植接受者)时,它优选是约2,000-约3,000个细胞/mm2。
(3)构成细胞层的细胞具有基本上六边形的平面图。这是构成在身体内的角膜内皮细胞层的细胞的诸多特性中的一种。本发明的制剂类似于在身体内的角膜内皮细胞层,显示出与先天的角膜内皮细胞层的功能类似的功能,并且也能够显示出活体内增殖能力。
(4)细胞在细胞层中规则地排列。在身体内的角膜内皮细胞层中,构成该层的细胞规则排列,鉴于此原因,角膜内皮细胞被认为维持正常功能和高透明度并且该角膜被认为适当地显示出水控制功能。因此,具有该形态特征的本发明制剂预计显示出与在身体内角膜内皮细胞层的功能类似的功能。
本发明的生产方法包括使用本发明的培养介质培养角膜内皮细胞的步骤并且例如能够通过下列方法来进行。
<1>角膜内皮细胞的采集和在试管中的培养
角膜内皮细胞是通过普通方法从接受者他自己/她自己或合适捐献者的角膜中采集的。考虑到在本发明中的移植条件,可以制备外源的角膜内皮细胞。例如,Descemet膜和角膜组织的内皮细胞层是从角膜基质上剥离,放入到培养皿中,然后用中性蛋白酶(dispase)等等处理。通过这一方法,从Descemet膜上分出角膜内皮细胞。保留在Descemet膜上的角膜内皮细胞能够通过移液管移取和类似方式被取出。在Descemet膜的取出后,角膜内皮细胞在本发明的培养介质中培养。培养介质能够例如通过适当地添加FBS(胎牛血清),b-FGF(basic-fibloblast生长因子),和抗生素如青霉素、链霉素和类似物到商购DMEM(Dulbecco’s ModifiedEagle’s Medium)中,和进一步添加Y-27632或fasudil到其中而获得。优选使用在表面上具有I型胶原,IV型胶原,粘连蛋白,层粘连蛋白或牛角膜内皮细胞的细胞外基质等等的涂层的培养容器(培养皿)。或者,可以使用用商购涂层剂如FNC涂覆混合物(注册商标)等等处理的一般培养容器。通过该涂层和本发明的培养介质的结合使用,促进了角膜内皮细胞在培养容器的表面上的粘附,并且实现了良好生长。
尽管角膜内皮细胞的培养的温度条件没有特别限制,只要角膜内皮细胞生长就行,但是考虑到生长效率,例如,温度是约25-约45℃,优选约30-约40℃,和进一步优选是约37℃。该培养是在约5-10%CO2浓度的环境中在湿润条件下在普通细胞培养器中进行。
<2>传代培养
传代培养能够在经受培养的角膜内皮细胞的生长之后进行。当细胞达到亚融合或融合时优选进行传代培养。传代培养能够如下进行。该细胞能够通过用胰蛋白酶-EDTA等处理从培养容器的表面上分离,并且回收。本发明的培养介质被添加到回收的细胞中得到细胞悬液。在细胞的回收过程中或在回收之后优选进行离心处理。该离心处理获得具有高细胞密度的细胞悬液。优选的细胞密度是大约1-2×106个细胞/mL。作为这里的离心处理的条件,例如能够提及500rpm(30g)-1000rpm(70g),1-10分钟。
按照与在上述初始培养中相同的方式将细胞悬液接种在培养容器上,然后培养。尽管在传代过程中的稀释比率取决于细胞的条件,但是它是大约1∶2-1∶4,优选约1∶3。该传代培养能够在与上述初始培养的培养条件类似的培养条件下进行。尽管培养时间取决于所使用的细胞的条件来改变,但是它例如是7-30天。以上传代培养能够根据需要进行多次。通过使用本发明的培养介质,在培养的最初阶段中细胞粘附会增大,据此培养时间能够缩短。
<3>角膜内皮细胞层的制备
细胞悬液被接种在底物如胶原片等等上,然后培养。这里,接种细胞的数量被控制,使得最终生产的角膜内皮制剂具有具备所需细胞密度的细胞层。确切地说,接种细胞,使得形成具有约1,000-约4,000个细胞/mm2的细胞密度的细胞层。培养能够在与上述初始培养的那些条件类似的条件和其它类似条件下进行。尽管培养时间取决于所使用的细胞的条件来改变,但是它例如是3-30天。
通过按照上述方式进行培养,能够获得其中在底物上已形成了在试管中所培养的角膜内皮细胞层的角膜内皮制剂。
在本发明中,该角膜内皮制剂可以含有本发明的培养介质以维持角膜内皮细胞。另外,该角膜内皮制剂可以含有本发明的角膜保存溶液,直至它用于移植为止。本发明也提供本发明的角膜内皮制剂和培养介质或保存溶液的结合物。
由本发明的生产方法获得的角膜内皮制剂能够用作需要角膜内皮的移植的疾病的治疗用移植物,所述疾病例如是大疱角膜病,角膜水肿,角膜白斑,特别地,由于角膜营养不良、创伤或眼内手术导致的角膜内皮功能紊乱所引起的大疱角膜病。
本发明的粘附促进剂和角膜内皮制剂的给药患者包括,例如哺乳动物(例如,人,小鼠,大鼠,仓鼠,兔,猫,狗,牛,绵羊,猴子等等)。

实施例1
ROCK抑制剂对于兔子角膜内皮细胞的培养的影响的研究
从安乐死之后立即分离出的兔角膜组织上分离出粘附有角膜内皮细胞的Descemet膜。Descemet膜与中性蛋白酶II(1.2U/mi,RocheApplied Science)一起在37℃,5%CO2的条件下培养45分钟,然后通过移液管移取手术以机械方式分离出角膜内皮细胞。分离的角膜内皮细胞进行离心处理,ROCK抑制剂(+)组的细胞在添加有Y-27632(10μM)的用于角膜内皮的介质中进行搅拌以及ROCK抑制剂(-)组的细胞在没有添加Y-27632的用于角膜内皮的介质中进行搅拌,达到相同的浓度,并且这些细胞是以约2000个细胞/孔的密度被接种在96孔板上。作为用于角膜内皮的介质,使用添加有1%胎牛血清和2mg/ml bFGF(GibcoInvitrogen)的培养介质(DMEM,Gibco Invitrogen)。该板用FNC涂覆混合物(Athena ES)预处理10分钟。在从培养起始后的72小时,该培养介质被交换并且在72小时后,ROCK抑制剂(+)组和ROCK抑制剂(-)组都在不含Y-27632的用于角膜内皮的常规介质中培养,直到第5天为止。在培养起始后的24小时之后,兔子角膜内皮细胞的原始培养物的相差显微照片示于图1中。从培养起始之后的第1天到第3天,这些细胞是每24小时通过使用CellTiter96(注册商标)AQueous One Solution CellProliferation(Promega)由MTS([3-(4,5-二甲基噻唑-2-基)-5-(3-羧基甲氧基苯基)-2-(4-磺基苯基)-2H-四唑])分析法来计数(图2)。
在ROCK里抑制剂(+)组中(图1B),在细胞接种之后在24小时(1天)中的细胞粘附显著地增加到ROCK抑制剂(-)组(图1A)的约2.8倍(图2和3)。在第3天当细胞实现融合时,在两个组之间没有发现显著差异(图2)。从它可以清楚地看出,在兔子角膜内皮细胞的原始培养物中传代之后Y-27632具有在早期阶段增大细胞粘附的作用。另外,在使用兔子角膜内皮细胞的传代培养的研究中也获得类似的结果,因此,Y-27632被认为对于在原始培养物和传代培养中传代之后的早期细胞粘附起作用。
实施例2
Y-27632对于猴子角膜内皮细胞的培养的影响的研究
从为了其它目的被安乐死的猕猴(Macaca fascicularis)解离的角膜组织中分离出粘附有角膜内皮细胞的Descemet膜。对于ROCK抑制剂(+)组,Descemet膜放入到添加了Y-27632(10μM)的用于角膜内皮的介质中并在37℃,5%CO2的条件下培养10分钟。对于ROCK抑制剂(-)组,该膜被放入到没有添加Y-27632的用于角膜内皮的介质中并且在相同的条件下培养10分钟。作为用于角膜内皮的介质,使用与实施例1中相同的细胞培养介质。在培养之后获得的Descemet膜与中性蛋白酶(Dispase)II(1.2U/ml,Roche Applied Science)一起在37℃,5%CO2的条件下培养45分钟,然后通过移液管移取手术以机械方式分离出角膜内皮细胞。分离的角膜内皮细胞进行离心处理并在Y-27632(+)和Y-27632(-)达到相同浓度的用于角膜内皮的介质中搅拌,然后这些细胞以约20000个细胞/孔的密度被接种在12孔板上。该板用FNC涂覆混合物(Athena ES)预处理10分钟。在从培养起始后的72小时,该培养介质被交换并且在72小时后,ROCK抑制剂(+)组和ROCK抑制剂(-)组都在不含Y-27632的用于角膜内皮的常规介质中培养。
从图4,在从培养起始后的24小时之后在猕猴(Macaca fascicuIaris)角膜内皮细胞的原始培养物中,显然更多的细胞粘附于在ROCK抑制剂(+)中的板上(图4b),这是指与ROCK抑制剂(-)相比而言(图4A)。在达到融合之前的天数对于ROCK抑制剂(+)是3,而对于ROCK抑制剂(-)是7。从其中可以表明,在培养起始之后的早期阶段中Y-27632增加细胞粘附并且是有用的药物,该药物甚至在认为与兔子相比体外培养比较困难的猴子角膜内皮细胞中能够进行细胞培养。
实施例3
Y-27632对于人角膜内皮细胞的培养的影响的研究
在从US眼库获得的人角膜组织中,中心部分(直径7mm)已经用于角膜移植术和使用剩下的***角膜组织。分离已粘附有角膜内皮细胞的Descemet膜。对于ROCK抑制剂(+)组,Descemet膜被放入到添加了Y-27632(10μM)的用于角膜内皮的介质中并且在37℃、5%CO2的条件下培养10分钟。对于ROCK抑制剂(-)组,该膜被放入到没有添加Y-27632的用于角膜内皮的介质中并且在相同的条件下培养10分钟。作为用于角膜内皮的介质,使用与实施例1中相同的细胞培养介质。在培养之后的Descemet膜与中性蛋白酶(Dispase)II(1.2U/ml,Roche Applied Science)一起在37℃,5%CO2的条件下培养45分钟,然后通过移液管移取手术以机械方式分离出角膜内皮细胞。分离的角膜内皮细胞进行离心处理并在Y-27632(+)和Y-27632(-)达到相同浓度的用于角膜内皮的介质中搅拌,然后这些细胞以约10000个细胞/孔的密度被接种在已用FNC涂覆混合物(Athena ES)预处理的48孔板上。从***角膜组织获得的人角膜内皮细胞的数量是极其少的,因此,一只眼睛的捐献者角膜在一个孔中培养。用于该研究的捐献者角膜是:对于ROCK抑制剂(+)组,在捐献者(年龄69岁)的死亡之后的6天开始培养,和对于ROCK抑制剂(-)组,在捐献者(年龄51岁)死亡之后的7天开始培养。对于年龄和从捐献者的死亡到细胞培养所消逝的时间的影响被认为是几乎相同的。
在从培养起始后的72小时,该培养介质被交换,并且在72小时后,ROCK抑制剂(+)组和ROCK抑制剂(-)组都在不含Y-27632的用于角膜内皮的常规介质中培养。为了研究对于在培养起始后的早期阶段中的细胞粘附以及细胞形态的影响,用相差显微镜观察细胞并且在培养起始后用数字式照象机拍取照片(图5)。
从图5中,在培养起始后的第7天,由与正常角膜内皮细胞类似的细胞(均匀的多角形细胞)组成的融合和高度致密的单个细胞层是在ROCK抑制剂(+)组中形成的(图5B),而具有低密度的成纤维细胞状长条形内皮细胞仅仅在ROCK里抑制剂(-)组中以小岛形状存活(图5A)。从这些结果可以认为,在已知极其难以培养的人角膜内皮的原始培养中,具有在培养起始后的早期阶段中增大细胞粘附的作用的Y-27632在培养介质中的添加能够实现具有良好细胞形态和高细胞密度的角膜内皮细胞层的形成。

实施例4
Y-27632对于猴子角膜内皮细胞的原始培养的影响的研究

在培养起始后的第10天的猕猴(Macaca fascicularis)角膜内皮细胞的原始培养物中,通过促进在培养的早期阶段中的粘附,ROCK抑制剂(+)(图6B)显著地使培养的细胞的总面积增大到相当于ROCK抑制剂(-)(图6A)的约1.6倍(图6C)。从它可以表明Y-27632甚至在认为体外培养很困难的猴子角膜内皮细胞中是可用于原始培养的药物。
实施例5
对于猴子角膜内皮细胞而言的Y-27632的最佳浓度的研究
将按照与实施例2中相同的方式采集的猴子角膜内皮细胞(原始培养物)加入到各自以1、10、33和100μM添加Y-27632的用于角膜内皮的介质中,和加入到没有添加Y-27632的用于角膜内皮的介质中,并且搅拌到相同的浓度,然后这些细胞以约2000个细胞/孔的密度接种在96孔板上。在培养起始后的24小时,活细胞通过使用CellTiter-Glo(注册商标,Promega)(图7)来计数。
当在培养起始后的第24小时的角膜内皮细胞在含有10μMY-27632的介质中培养时,粘附于板上的细胞的数量是显著高的(图7),这是指与在没有添加Y-27632的用于角膜内皮的介质中以及含有不同浓度的Y-27632的用于角膜内皮的介质中的细胞培养相比而言。从其中表明,在10μM的浓度下Y-27632对于猴子角膜内皮细胞是最有效的。
实施例6
Y-27632对于猴子角膜内皮细胞的传代培养的影响的研究
从安乐死的猕猴(Macaca fascicularis)解离的角膜组织中分离出粘附有角膜内皮细胞的Descemet膜。Descemet膜与中性蛋白酶II(1.2U/ml,Roche Applied Science)一起在37℃,5%CO2的条件下培养45分钟,然后通过移液管移取手术以机械方式分离出角膜内皮细胞。分离的角膜内皮细胞进行离心处理,然后接种在用于角膜内皮的介质上。实现融合的角膜内皮细胞在37℃,5%CO2的条件下用0.05%胰蛋白酶培养10分钟,然后传代培养。在4到6代后,该角膜内皮细胞被加入到96孔板上达到1/2,1/6或1/8的稀释比。对于ROCK抑制剂(+)组通过使用添加了Y-27632(10μM)的用于角膜内皮的介质和对于ROCK抑制剂(-)组通过使用没有添加Y-27632的用于角膜内皮的介质来继续进行传代培养。在传代后的24小时,按照与实施例5中同样的方式通过使用CellTiter-Glo(注册商标,Promega)(图8)来计数。
当在传代后的第24小时的已培养的猕猴(Macaca fascicularis)角膜内皮细胞在含有Y-27632的介质中传代培养时,粘附于板上的细胞的数量是显著高的(图8),这是指与在没有添加Y-27632的用于角膜内皮的介质相比而言。从其可表明,除了原始培养外,Y-27632对于猴子角膜内皮细胞的传代培养也是有效的。
实施例7
Y-27632对于在猴子角膜内皮细胞的传代培养中细胞形态的影响的研究
按照与实施例6中相同方式传代培养的猕猴(Macaca fascicularis)角膜内皮细胞在载片上次代培养至1/4的稀释比。在传代培养后的24小时,用相差显微镜观察细胞形态(图9A和9B)。此外,在载片上的角膜内皮细胞用4%仲甲醛在室温下固定10分钟,并且肌纤蛋白纤维由毒伞素荧光染色法(图10A和10B)来染色。
在传代后的第24小时的已培养的猕猴(Macaca fascicularis)角膜内皮细胞中,当在含有Y-27632的介质中进行传代培养时(图9B),细胞在载片上的粘附增加,促进了展平成角膜内皮细胞状细胞,并且与不含Y-27632的用于角膜内皮的介质相比也增加了细胞的凝聚(图9A)。另外,通过毒伞素荧光染色,当在含有Y-27632的介质中进行传代培养时(图10B),与不含Y-27632的用于角膜内皮的介质相比清楚地观察到肌纤蛋白应力纤维(图10A),并且已发现Y-27632促进细胞骨架的形成。

因为在细胞增殖周期G1中的细胞核中发现Ki67抗原,并且从S到M阶段,已表明Y-27632具有在猕猴(Macaca fascicularis)角膜内皮细胞的传代培养之后的早期阶段中加速细胞周期的作用并且是可用于有效细胞培养的药物。
实施例9
Y-27632对于猴子角膜内皮细胞的凋亡的影响的研究
按照与实施例6中相同方式传代培养的猕猴(Macaca fascicularis)角膜内皮细胞被次代培养至1/4的稀释比。添加了Y-27632(10μM)的用于角膜内皮的介质是用于ROCK抑制剂(+)组以及没有添加Y-27632的用于角膜内皮的介质是用于ROCK抑制剂(-)组。在传代培养的第1天,包括在介质中的那些细胞在内的全部细胞通过使用0.05%胰蛋白酶被采集并且通过使用Annexin V和PI(Propidium Iodide)进行流式细胞光度术,然后测定细调凋亡(图14和15)。
ROCK抑制剂(+)组显示了在传代培养的第1天凋亡细胞与全部细胞的比率的显著减少(图14),这是指与ROCK抑制剂(-)组相比。此外,每1×104个细胞中凋亡细胞的数量的比较揭示了在ROCK抑制剂(+)组中的显著减少,这是指与ROCK抑制剂(-)组相比(图15)。
从其中可以表明,Y-27632在猕猴(Macaca fascicularis)角膜内皮细胞的传代培养中具有抑制凋亡的作用。
实施例10
Y-27632对于猴子角膜内皮细胞的密度的影响的研究
从安乐死的猕猴(Macaca fascicularis)解离的角膜组织中分离出粘附有角膜内皮细胞的Descemet膜。Descemet膜与中性蛋白酶II(1.2U/ml,Roche Applied Science)一起在37℃,5%CO2的条件下培养45分钟,然后通过移液管移取手术以机械方式分离出角膜内皮细胞。分离的角膜内皮细胞进行离心处理,然后接种在用于角膜内皮的介质上。实现融合的角膜内皮细胞在37℃,5%CO2的条件下用0.05%胰蛋白酶培养10分钟,然后传代培养。添加了Y-27632(10μM)的用于角膜内皮的介质是用于ROCK抑制剂(+)组以及没有添加Y-27632的用于角膜内皮的介质是用于ROCK抑制剂(-)组。在从培养起始后的72小时,该培养介质被交换,并且在72小时后,ROCK抑制剂(+)组和ROCK抑制剂(-)组都在不含Y-27632的用于角膜内皮的常规介质中培养。在每一次传代中,用相差显微镜拍取细胞的照片,测量细胞密度,以及研究Y-27632对于所培养的猴子角膜内皮细胞的密度的影响。
该传代重复7次,和该ROCK抑制剂(+)组在整个传代培养的期间显示出更高的细胞密度(图16),这是指与ROCK抑制剂(-)组相比。从这一点可以认为正是Y-27632的使用能够实现具有高密度的角膜内皮细胞的培养,其未来将可用于再生药物,如培养角膜内皮片的移植。
实施例11
fasudil对于猴子角膜内皮细胞的培养的影响的研究
从安乐死的猕猴(Macaca fascicularis)解离的角膜组织中分离出粘附有角膜内皮细胞的Descemet膜。Descemet膜与中性蛋白酶II(1.2U/ml,Roche Applied Science)一起在37℃,5%CO2的条件下培养45分钟,然后通过移液管移取手术以机械方式分离出角膜内皮细胞。分离的角膜内皮细胞进行离心处理,然后在用于角膜内皮的介质中传代培养。作为用于角膜内皮的介质,使用与实施例1中相同的细胞培养介质。在通过0.05%胰蛋白酶处理进行的传代培养过程中所收集的已培养的猴子角膜内皮细胞被分离和离心处理,并且在fasudil组中在添加了fasudil(10μM,SIGMA-ALDRICH)的用于角膜内皮的介质中和在对照组中在没有添加fasudil的用于角膜内皮的介质中搅拌至相同的浓度,然后这些细胞以约2000个细胞/孔的密度被接种到96孔板上。在培养起始后的24小时,粘附细胞通过使用CellTiter-Glo(注册商标)LumineScent CellViability Assay(Promega)(图1 7)来计数。通过使用fasudil(一种ROCK抑制剂)也发现了角膜内皮细胞的受促进的细胞粘附的效果。

角膜内皮片的制备用的含Rho激酶抑制剂的培养介质
根据常规方法制备下列培养介质。
fasudil 0.5mg
FBS 10mL
青霉素-链霉素溶液 1mL
FGF basic 200ng
DMEM 适量
总量 100mL
使用由Invitrogen制造的FBS,由Nacalai Tesque制造的青霉素-链霉素溶液(含有青霉素5000u/mL和链霉素5000μg/mL),由Invitrogen制造的FGF basic,和由Invitrogen制造的DMEM。
本申请是基于在日本提交的专利申请No2007-223141(申请日:2007年8月29日)和2008-016088(申请日:2008年1月28日),它们的内容全部引入这里供参考。
Claims (8)
1.Rho激酶抑制剂用于生产预防或治疗角膜内皮功能紊乱的制剂的用途。
2.根据权利要求1的用途,其中该Rho激酶抑制剂是选自(+)-反式-4-(1-氨基乙基)-1-(4-吡啶基氨基甲酰基)环己烷,1-(5-异喹啉磺酰基)高哌嗪和其可药用盐中的至少一种。
3.根据权利要求1的用途,其中该角膜内皮细胞源自于人。
4.根据权利要求1到3中任何一项的用途,其中所述制剂具有对于角膜内皮细胞的粘附促进作用、对于角膜内皮细胞的凋亡抑制作用和/或对于角膜的保护作用。
5.根据权利要求4的用途,其中所述制剂具有对于角膜内皮细胞的粘附促进作用。
6.根据权利要求4的用途,其中所述制剂具有对于角膜内皮细胞的凋亡抑制作用。
7.根据权利要求4的用途,其中所述制剂具有对于角膜的保护作用。
8.根据权利要求4的用途,其中所述制剂是前房内注射或眼内灌注流体的形式。
