Abstract
Se revelan péptidos de glucagon modificados, que tienen una mayor potencia en el receptor de glucagon respecto del glucagon nativo. Otra modificacion de los péptidos de glucagon formando puentes de lactama o la sustitucion del ácido carboxílicoterminal con un grupo amida produce péptidos que exhiben una actividad coagonista del receptor de glucagon/GLP-1. La solubilidad y la estabilidad de estos análogos de glucagon de gran potencia también se pueden mejorar por modificacion de lospolipéptidos por pegilacion, sustitucion de los aminoácidos carboxiterminales o la adicion de un péptido carboxiterminal seleccionado del grupo que consiste en SEQ ID N°: 26 (GPSSGAPPPS), SEQ ID N°:27 (KRNRNNIA) y SEQ ID N°:28 (KRNR). Reivindicacion 1: Un péptido de glucagon no nativo que comprende la secuencie, de SEQ ID N°:55 o un análogo de SEQ ID N°:55, en donde dicho análogo no es un péptido natural y dicho análogo difiere de SEQ ID N°:55 por 1 a 3 modificaciones deaminoácidos seleccionados de las posiciones 1, 2, 3, 5, 7, 10, 11, 13, 14, 17, 18, 19, 21, 24, 27, 28, y 29, en donde dicho péptido de glucagon exhibe al menos 20% de la actividad del GLP-1 nativo en el receptor GLP-1. Reivindicacion 56: Unacomposicion farmacéutica que comprende el agonista de glucagon de acuerdo con la reivindicacion 1, o una de sus sales farmacéuticamente aceptables, y un portador farmacéuticamente aceptable.
