Abstract

Pronalazak obuhvata postupak prečišćavanja kamptotecina ili 11-alkoksikamptotecina, u kome se smeša jednog od ovih prirodnog proizvoda koja i nečistoće dovodi u kontakt sa rastvaračem koji je u stanju da iz nje odvoji drugačije alkaloide, kao i nečistoće koje ometaju hidrogenaciju ovih jedinjenja u odgovarajuće tetrahidro analoge. Kamptotecin ili 11-alkoksikamptotecin se zatim odvaja iz smeše. Takodje je obuhvaćen postupak za hidrogenaciju ovako prečišćenog kamptotecina ili 11-alkoksikamptotecina u odgovarajuće tetrahidro analoge, u kome se polazni materijal dovodi u kontakt sa vodonikom i katalizatorom za hidrogenaciju, u prisustvu moderatora. 11-alkoksi-1,2,6,7-tetrahidro-kamptotecini dobijeni ovim postupkom su nova jedinjenja. Pronalazak dalje obuhvata postupak za oksidaciju tetrahidro analoga, koji obuhvata dovodjenje tetrahidrokamptotecina ili 11-alkoksitetrahidrokamptotecina u kontakt sa oksidacionim sredstvom u tečnom reakcionom medijumu. 10-Hidroksi-11-alkoksikamptotecini dobijeni ovim postupkom su nova jedinjenja. Konačno, pronalazak obuhvata postupak za dobijanje u vodi rastvornih derivata kamptotecina, supstituisanih u 9-položaju grupom formule -CH2R1, gde R1 predstavlja -OR2, -SR3 ili -NR4R5. 10-Hidroksi analog prvo reaguje sa jedinjenjem formule CH2(NR4R5)2, a zatim se aminska grupa, po potrebi, prevodi u grupu -OR2 ili -SR3. U vodi rastvorni analozi kamptotecina predstavljaju nova jedinjenja i korisni su za inhibiranje rasta ćelija tumora koje su osetljive na takve analoge.
