Description
本発明は、難溶性薬物の易溶性固形製剤およびその製造方法に関する。さらに詳しくは、2-(3-{4-アミノ-6-[(1-メチル-1H-ピラゾール-4-イル)アミノ]-1,3,5-トリアジン-2-イル}-2-(ヒドロキシメチル)フェニル)-6-シクロプロピル-8-フルオロイソキノリン-1(2H)-オン(化合物A)の固体分散体を含有することを特徴とする易溶性固形製剤およびその製造方法に関する。
分子標的治療薬として種々のプロティンキナーゼに対する阻害剤が癌および免疫炎症性疾患などに広く用いられており、例えば、ブルトンチロシンキナーゼ(Bruton’s tyrosine kinase; BTK)に対して阻害作用を有する薬剤(BTK阻害剤)のイムブルビカ(商標、一般名、イブルチニブ)は、再発又は難治性の慢性リンパ性白血病治療薬として使用されている。
化合物Aは、本願発明者らよって新たに創製されたBTK阻害剤であり、BTKを介した異常な細胞応答に関連していることが知られている疾患、例えば、自己免疫疾患、炎症性疾患、骨疾患、リンパ腫のような癌等に対する薬剤として有用な化合物である(後記特許文献1)。
化合物Aは、本願発明者らよって新たに創製されたBTK阻害剤であり、BTKを介した異常な細胞応答に関連していることが知られている疾患、例えば、自己免疫疾患、炎症性疾患、骨疾患、リンパ腫のような癌等に対する薬剤として有用な化合物である(後記特許文献1)。
しかしながら化合物Aは、水に対する溶解度が極めて低いため、例えば、化合物Aを充填したカプセル剤あるいは通常の錠剤などの固形製剤として経口投与した場合、溶出性が悪く、そのため血中に十分に吸収されず、バイオアベイラビリティー(生物学的利用能)が低いという欠点を有しており、化合物AをBTK阻害剤として経口投与製剤として用いるためには、経口吸収性に優れた易溶性固形製剤が求められている。
難溶性薬物の易溶性固形製剤の製造方法としては種々の方法が知られており、例えば、難溶性薬物を不活性担体(以下、単に担体という)中に非晶質状態で分散させて、いわゆる固体分散体として易溶性固形製剤を製造する方法などが知られている。
固体分散体の製造方法としては、薬物と担体とを溶媒に溶解した後、噴霧乾燥し、固体分散体とする方法(スプレードライ法)、薬物と担体とをエクストルーダーに投入し、難溶性薬物の融点以上に加熱しながら剪断力を加えて共溶融させた後にダイから押し出し、冷却、固化する方法(ホットメルトエクストルージョン法)または薬物と担体とを溶媒で溶解後、貧溶媒中に添加して析出させ、乾燥して固体分散体とする方法(共沈法)など種々の製造方法が知られているが、化合物Aを有効成分として含有する易溶性固形製剤およびその製造方法に固体分散体の製造技術を用いた製剤は報告されていない。
難溶性薬物の易溶性固形製剤の製造方法としては種々の方法が知られており、例えば、難溶性薬物を不活性担体(以下、単に担体という)中に非晶質状態で分散させて、いわゆる固体分散体として易溶性固形製剤を製造する方法などが知られている。
固体分散体の製造方法としては、薬物と担体とを溶媒に溶解した後、噴霧乾燥し、固体分散体とする方法(スプレードライ法)、薬物と担体とをエクストルーダーに投入し、難溶性薬物の融点以上に加熱しながら剪断力を加えて共溶融させた後にダイから押し出し、冷却、固化する方法(ホットメルトエクストルージョン法)または薬物と担体とを溶媒で溶解後、貧溶媒中に添加して析出させ、乾燥して固体分散体とする方法(共沈法)など種々の製造方法が知られているが、化合物Aを有効成分として含有する易溶性固形製剤およびその製造方法に固体分散体の製造技術を用いた製剤は報告されていない。
本発明は、化合物Aの固体分散体を含有することを特徴とする易溶性固形製剤およびその製造方法を提供することを課題とする。
本発明者らは、化合物Aの易溶性固形製剤およびその製造方法について種々検討を重ねた結果、化合物Aを特定の担体中にスプレードライ法を用いて非晶質化させることにより得られる化合物Aの固体分散体が化合物Aの溶出性を顕著に改善させることができ、当該固体分散体を含有する易溶性固形製剤が本発明の課題を解決できることを見出して本発明を完成させた。
本発明の化合物Aを有効成分として含有する易溶性固形製剤およびその製造方法によれば、化合物Aを担体中に非晶質化させ固体分散体とすることにより溶出性を顕著に改善させることによって、経口吸収性やバイオアベイラビリティーを向上させ得ることが、本発明の易溶性固形製剤を用いた溶出試験および動物(イヌ)を用いて行った経口投与試験によって確かめられた。このことから、本発明の化合物Aを有効成分として含有する易溶性固形製剤は、BTK阻害剤として優れた効果を奏し得る。
本発明は、以下のようにして達成される。
本発明は、本発明者らによって見出された製造方法によって製造される化合物A(特許文献1)と、通常、固体分散体の担体として使用可能な種々の高分子ポリマー(担体)とを有機溶媒に溶解し、次いで、スプレードライ法により化合物Aを担体中に非晶質化させ、得られた担体中において化合物Aが非晶質状態であることを確認後、得られた固体分散体を通常の製剤化工程に付すことによって目的とする化合物Aを含有する易溶性固形製剤を製造することにより達成される。
すなわち、本発明に用いられる化合物Aの担体としては、薬学的に許容される医薬品添加物として使用することができる種々の水溶性ポリマー(例えば、ヒプロメロース、ヒドロキシプロピルセルロース、メチルセルロース、ポリビニルピロリドン、コポビドンなど)および腸溶性ポリマー(例えば、ヒプロメロースフタル酸エステル、ヒプロメロース酢酸エステルコハク酸エステル、乾燥メタクリル酸コポリマーLDなど)を用いることができ、何れの場合にも、化合物Aは固体分散体のなかで非晶質状態となり、化合物A(原薬)に比較して溶出性が顕著に改善され、バイオアベイラビリティーが低いという欠点が改善される。
本発明は、本発明者らによって見出された製造方法によって製造される化合物A(特許文献1)と、通常、固体分散体の担体として使用可能な種々の高分子ポリマー(担体)とを有機溶媒に溶解し、次いで、スプレードライ法により化合物Aを担体中に非晶質化させ、得られた担体中において化合物Aが非晶質状態であることを確認後、得られた固体分散体を通常の製剤化工程に付すことによって目的とする化合物Aを含有する易溶性固形製剤を製造することにより達成される。
すなわち、本発明に用いられる化合物Aの担体としては、薬学的に許容される医薬品添加物として使用することができる種々の水溶性ポリマー(例えば、ヒプロメロース、ヒドロキシプロピルセルロース、メチルセルロース、ポリビニルピロリドン、コポビドンなど)および腸溶性ポリマー(例えば、ヒプロメロースフタル酸エステル、ヒプロメロース酢酸エステルコハク酸エステル、乾燥メタクリル酸コポリマーLDなど)を用いることができ、何れの場合にも、化合物Aは固体分散体のなかで非晶質状態となり、化合物A(原薬)に比較して溶出性が顕著に改善され、バイオアベイラビリティーが低いという欠点が改善される。
なかでも化合物Aとヒプロメロースフタル酸エステル(HPMCP(商標))との固体分散体の場合に他の固体分散体に比較して溶出性が特に改善され、当該固形組成物(固体分散体)をカプセルに充填して経口投与した場合、バイオアベイラビリティーが低いという欠点が改善され、また、固形製剤中の非晶質の化合物Aが保存中も結晶形への転移を起こしにくく、保存安定性にも優れている。
本発明に好適に用いられるヒプロメロースフタル酸エステル(HPMCP)は、ヒプロメロース(ヒドロキシプロピルメチルセルロース;HPMC)に疎水性のカルボキシベンゾイル基を導入した構造を有しており、そのためHPMCPは水に溶解せず、有機溶媒に溶解する。また、カルボキシベンゾイル基のカルボキシル基によって胃内では溶解せず、通常、薬物の最大吸収部位である小腸で溶解させることができることから腸溶性コーティング剤としても用いられている。
本発明に好適に用いられるヒプロメロースフタル酸エステル(HPMCP)は、ヒプロメロース(ヒドロキシプロピルメチルセルロース;HPMC)に疎水性のカルボキシベンゾイル基を導入した構造を有しており、そのためHPMCPは水に溶解せず、有機溶媒に溶解する。また、カルボキシベンゾイル基のカルボキシル基によって胃内では溶解せず、通常、薬物の最大吸収部位である小腸で溶解させることができることから腸溶性コーティング剤としても用いられている。
また、小腸は、十二指腸、空腸および回腸からなる人体の臓器のなかで最長の臓器で、薬物に応じてそれぞれ吸収部位が異なることから、小腸内での溶出をコントロールするためにHPMCPのカルボキシベンゾイル基の含有量をコントロールしたpH5.0前後で溶解するタイプ(HP-50)とpH5.5前後で溶解するタイプ(HP-55)のHPMCPが市販されているが、化合物Aの担体としては、pH5.5前後で溶解するタイプ(HP-55)のHPMCPが好ましい。
本発明において化合物Aが担体中に非晶質状態で存在していることは、粉末X線回析測定および温度変調DSC分析などにより確認することができる。
本発明の易溶性固形製剤は、化合物Aと薬学的に許容される医薬品添加物として使用することができる種々の水溶性および腸溶性ポリマーを担体として用いることができるが、HPMCPが好ましく、特にHPMCP(HP-55)が好ましい。

担体は、化合物A(1重量部)に対して、0.5から5重量部、好ましくは0.8から3重量部、特に好ましくは1重量部の割合で、有機溶媒に溶解後、スプレードライ法により化合物Aを非晶質化させ、得られた固形組成物(固体分散体)中において化合物Aが非晶質状態であることを確認後、通常の製剤化方法によって化合物Aを含有する易溶性固形製剤を製造することができる。

さらに、本発明の易溶性固形製剤は、必要に応じて、カプセル剤、錠剤、顆粒剤、細粒剤などの剤形にすることができる。また、各種製剤化のために、種々の薬学的に許容される賦形剤、崩壊剤、滑沢剤、着色剤、矯味矯臭剤を必要に応じて適宜配合することができる。
例えばこれら配合剤としては、ラクトース、コーンスターチなどのデンプン、微結晶性セルロースなどの結晶セルロース、ヒドロキシプロピルセルロース、カルシウムカルボキシメチルセルロース、タルク、ステアリン酸マグネシウムなどがあげられる。

本発明に用いられる有機溶媒としては、特に限定されないが、通常、化合物Aと担体とを溶解する、例えばジクロロメタンとメタノールとの混合溶媒を用いることができる。混合比は体積比で、ジクロロメタン1に対してメタノール0.5~2、好ましくは0.8~1.2である。
本発明製剤の用量は、疾患の種類、重症度、患者の年齢、性別、および体重などに従って適宜増減させることができるが、通常は成人において1日あたり10mg~2400gの範囲であり、それを1日、1回、または2回もしくは3回に分割して経口投与することができる。
特に、本発明の一実施態様であるカプセル剤は、疾患の種類、症状や年齢に合わせた服用量の調節が容易であるため、本発明の製剤の剤形として好適であるが、剤形は特に限定されない。
本発明製剤の用量は、疾患の種類、重症度、患者の年齢、性別、および体重などに従って適宜増減させることができるが、通常は成人において1日あたり10mg~2400gの範囲であり、それを1日、1回、または2回もしくは3回に分割して経口投与することができる。
特に、本発明の一実施態様であるカプセル剤は、疾患の種類、症状や年齢に合わせた服用量の調節が容易であるため、本発明の製剤の剤形として好適であるが、剤形は特に限定されない。
以下に実施例および試験例を挙げて本発明をさらに具体的に説明する。
なお、本発明の化合物Aの易溶性固形製剤およびその製造方法は、各実施例の記載に限定されるものではない。

試験例1(溶出試験)
本発明製剤の効果を化合物Aの種々の固体分散体を用いて評価した。
実施例1から実施例8の固体分散体(表1参照)を溶出試験に供した。なお、対照として、化合物A(原薬)についても同様に溶出試験を行った。
溶出試験は、日本薬局方溶出試験第2液(pH6.8)を溶出試験液とし、上記固体分散体各40mgに当該溶出試験液(30ml、37℃)を加え、経時的にサンプリングを行い(5,15,30,60および120分)、化合物Aの溶出量をUV測定によって求めることにより行った。測定結果を図1に示す。
本発明製剤の効果を化合物Aの種々の固体分散体を用いて評価した。
実施例1から実施例8の固体分散体(表1参照)を溶出試験に供した。なお、対照として、化合物A(原薬)についても同様に溶出試験を行った。

したがって、特に化合物AとHPMCP(HP-55)との固体分散体からなる本発明の易溶性固形製剤の場合には、薬物が吸収されやすい小腸において、化合物Aの過飽和状態が長時間維持され、バイオアベイラビリティーが低いという欠点が改善される。
試験例2(溶出試験)
実施例9の固体分散体の溶出試験を試験例1と同様にして行った。
その結果、実施例9の固体分散体は、担体の量を実施例1の3分の1に減量したにも拘わらず実施例1の固体分散体とほぼ同様の溶出性の改善がみられた。

化合物A(1g)をジクロロメタンとメタノール各250mlの混合溶媒に溶解させた後、HPMCP(HP-55)3gを加えて溶解し、フィード液を調製した。ついで、フィード液を入熱温度75℃、排熱温度50℃、噴霧窒素流量350(L/h)、フィード流量5(mL/min)でミニスプレードライヤーを用いて噴霧乾燥を行い、得られた粉末を室温で20時間真空乾燥工程に付すことにより収率約82%で化合物Aの固体分散体が得られた。
得られた固体分散体は、粉末X線回折試験でハローパターンを示し結晶ピークは観察されず、温度変調DSC分析において融点ピークは観察されず、ガラス転移点(140.22℃)が観察され、HPMCP(HP-55)(担体)中で化合物Aは非晶質状態であることが確認された。

化合物A(4g)をジクロロメタンとメタノール各1000mlの混合溶媒に溶解させた後、HPMCP(HP-55)4gを加えて溶解し、フィード液を調製した。ついで、フィード液を、排熱温度が50℃となるように入熱温度をコントロールしながら噴霧窒素流量350(L/h)、フィード流量5(mL/min)でミニスプレードライヤーを用いて噴霧乾燥を行い、得られた粉末を室温で20時間真空乾燥工程に付すことにより収率約71%で化合物Aの固体分散体が得られた。
得られた固体分散体は、粉末X線回折試験でハローパターンを示し結晶ピークは観察されず、温度変調DSC分析において融点ピークは観察されず、ガラス転移点が観察され、HPMCP(HP-55)(担体)中で化合物Aは非晶質状態であることが確認された。

実施例9で得られた固体分散体30mgをカプセルに充填することにより、化合物A(15mg)を含有するカプセル剤を得た。

本発明の化合物Aを有効成分として含有する易溶性固形製剤およびその製造方法によれば、化合物Aを固体分散体とすることにより非晶質状態として溶出性を顕著に改善させることによって、経口吸収性やバイオアベイラビリティーを向上させ得ることが本発明の易溶性固形製剤を用いた動物(イヌ)による経口投与試験によって確かめられたことから、本発明の化合物Aを有効成分として、非晶質状態で含有する易溶性固形製剤は、BTK阻害剤として優れた効果を奏し得る。
Claims (5)
- 2-(3-{4-アミノ-6-[(1-メチル-1H-ピラゾール-4-イル)アミノ]-1,3,5-トリアジン-2-イル}-2-(ヒドロキシメチル)フェニル)-6-シクロプロピル-8-フルオロイソキノリン-1(2H)-オン(化合物A)が非晶質状態で存在する固体分散体を含有することを特徴とする、易溶性固形製剤。
- 該固体分散体が、化合物A(重量部1)に対し、ヒプロメロースフタル酸エステル(重量部0.8~3)を担体として含有する、請求項1の易溶性固形製剤。
- 該易溶性固形製剤がカプセル剤である、請求項1または2に記載の易溶性固形製剤。
- 該化合物Aおよびヒプロメロースフタル酸エステルを有機溶媒に溶解させた後、スプレードライ法により、ヒプロメロースフタル酸エステル中に非晶質化させた化合物を含有する固体分散体を製造することを特徴とする、請求項1~3のいずれかに記載の易溶性固形製剤の製法。
- スプレードライ法に用いる有機溶媒として、ジクロロメタンとメチルアルコールとの混合溶媒を用いる、請求項4に記載の製法。
