일반적으로 염증반응은 생체의 세포나 조직에 기질적 변화를 가져오는 침습으로 인한 손상을 수복 및 재생하기 위한 생체방어 반응과정이고, 이 반응과정에는 국소의 혈관, 체액의 각종 조직세포 및 면역세포 등이 작용한다. 정상적으로 외부 침입 균에 의하여 유도되는 염증반응은 생체를 보호하기 위한 방어 시스템인 반면, 비정상적으로 과도한 염증반응이 유도되면 다양한 질환들이 나타나게 되는데, 이러한 질환들을 염증질환이라 칭한다. 상기 염증질환은 외부자극에 의하여 활성화된 표적세포로부터 분비되는 다양한 염증 매개물질이 염증을 증폭 및 지속시켜 인체의 생명을 위협하는 질환이다. 이와 같은 염증에 의해 발생하는 염증성 질환에는 천식, 만성폐쇄성폐질환, 알레르기, 전신성 홍반성 루푸스, 경피증, 궤양성 대장염, 크론병, 아토피성 피부염, 건선, 아나필락시스, 피부염, 당뇨병성 망막증, 망막염, 황반 변성, 포도막염, 결막염, 관절염, 류마티스성 관절염, 강직성 척추염, 골관절염, 골다공증, 당뇨, 당뇨성 신장병, 신염, 신장염, 쇼그렌 증후군, 크론씨 병, 자가 면역 췌장염, 치주질환, 이식편대숙주질환, 만성 골반 염증 질환, 자궁내막염, 비염, 편도염, 중이염, 인후염, 방광염 및 만성 전립선염 등 다양한 질환이 포함된다.
한편, 나한과(Siraitia
grosvenori)는 중국 광동, 광서성의 고랭지에서 재배되는 박과의 다년생 초본으로 과실은 직경 4~5cm의 계란형 또는 구형이며 원래 중국 계림지방에서는 청량음료의 원료나 조미료로 사용돼 왔으며, 목이 아프거나 기침, 위나 장에 문제가 있을 경우 민간에서 사용되었다.
나한과 추출물에 관한 알려진 종래 기술의 일례로는 한국공개특허 제2015-0132654호에 나한과 추출물, 이의 분획물 또는 이로부터 분리된 화합물을 포함하는 염증성 질환의 예방 또는 치료용 약학적 조성물이 개시되어 있으나, 아직까지는 본 발명의 나한과 잔사 추출물을 유효성분으로 함유하는 염증의 예방, 개선 또는 치료용 조성물이 알려진바 없다.
본 발명은 상기와 같은 요구에 의해 도출된 것으로서, 본 발명은 나한과 잔사 추출물을 유효성분으로 함유하는 염증의 예방, 개선 또는 치료용 조성물을 제공하고, 본 발명에 따른 나한과 잔사 추출물이 세포 독성이 없으며, 염증에 의해 유도되는 NO의 생성, PGE2 및 IL-1β의 증가를 억제하는 효과가 있으며, 아토피 피부염이 유발된 세포에서 사이토카인 및 케모카인의 함량을 감소시킨다는 것을 확인함으로써, 본 발명을 완성하였다.
상기 목적을 달성하기 위하여, 본 발명은 나한과 잔사 추출물을 유효성분으로 함유하는 염증성 질환의 예방 또는 치료용 약학 조성물을 제공한다.
또한, 본 발명은 나한과 잔사 추출물을 유효성분으로 포함하는 염증의 예방 또는 개선용 건강기능식품 조성물을 제공한다.
또한, 본 발명은 나한과 잔사 추출물을 유효성분으로 포함하는 아토피 피부염 개선용 화장료 조성물을 제공한다.
본 발명은 나한과 잔사 추출물을 유효성분으로 함유하는 염증의 예방, 개선 또는 치료용 조성물에 관한 것으로, 본 발명의 유효성분인 나한과 잔사 추출물은 염증에 의해 유도되는 NO의 생성, PGE2 및 IL-1β의 증가를 억제하는 효과가 우수하고, 아토피 피부염 또는 피부염이 유발된 세포에서의 사이토카인 또는 케모카인의 함량을 유의미하게 감소시키므로, 염증의 예방, 개선 또는 치료용 의약품, 건강기능성식품 또는 화장료에 유용하게 활용될 수 있다.
도 1은 나한과 추출물과 본 발명의 나한과 잔사 추출물이 세포 생존율(%)에 미치는 영향을 확인한 결과이다.
도 2는 RAW 264.7 세포에서 LPS(lipopolysaccharide)의 처리에 의해 NO 생성량이 증가하며, 나한과 추출물과 본 발명의 나한과 잔사 추출물을 처리하여 LPS에 의해 증가한 NO의 생성량이 감소하는 것을 확인한 결과이다. ###는 LPS의 처리에 의해 NO 생성이 음성대조군(Control)에 비해 통계적으로 유의미하게 증가하였다는 것으로, p<0.001을 의미하고, ***는 LPS에 의해 증가한 NO 생성량이 본 발명의 나한과 잔사 추출물의 처리에 의해 통계적으로 유의미하게 감소하였다는 것으로, p<0.001이다.
도 3은 RAW 264.7 세포에서 LPS(lipopolysaccharide)에 의해 유도된 PGE2 생성량에 대하여, 나한과 추출물과 본 발명의 나한과 잔사 추출물을 처리하여 PGE2의 생성량을 감소시키는 것을 확인한 결과이다. **, ***는 LPS에 의해 증가한 PGE2 생성량이 본 발명의 나한과 잔사 추출물의 처리에 의해 통계적으로 유의미하게 감소하였다는 것으로, **는 p<0.01이고, ***는 p<0.001이다.
도 4는 RAW 264.7 세포에서 LPS에 의해 유도된 IL-1β 생성량에 대하여, 나한과 추출물과 본 발명의 나한과 잔사 추출물을 처리하여 IL-1β의 생성량을 감소시키는 것을 확인한 결과이다. ###는 LPS의 처리에 의해 IL-1β 생성이 음성 대조구(Control)에 비해 통계적으로 유의미하게 증가하였다는 것으로, p<0.001을 의미하고, ***는 LPS에 의해 증가한 IL-1β 생성량이 본 발명의 나한과 잔사 추출물의 처리에 의해 통계적으로 유의미하게 감소하였다는 것으로, p<0.001이다.
도 5는 나한과 잔사 추출물의 MC/9 비만 세포 독성을 확인한 결과이다.
도 6은 MC/9 비만 세포에서, 나한과 잔사 추출물의 처리에 따른 사이토카인의 분비량이 감소한 것을 확인한 결과이다. Control은 아무것도 처리하지 않은 정상군이고, PMACI는 PMA(phorbol myristate acetate)와 칼슘 이오노포어(calcium ionophore: CI)를 처리한 군이다. ###는 정상군 대비 PMACI 군의 TNF-α(A) 및 IL-6(B)의 함량이 통계적으로 유의미하게 증가한 것을 나타내며, p<0.001이고, ***는 PMACI군 대비 본 발명의 나한과 잔사 추출물 또는 양성대조군인 덱사메타손을 처리한 군의 TNF-α(A) 및 IL-6(B)의 함량이 통계적으로 유의미하게 감소하였다는 것으로, p<0.001임을 의미한다.
도 7은 나한과 잔사 추출물의 HaCaT 세포 독성을 확인한 결과이다.
도 8은 인간 각질세포인 HaCaT 세포에서, 나한과 잔사 추출물의 처리에 따른 케모카인의 분비량이 감소한 것을 확인한 결과이다. Control은 아무것도 처리하지 않은 정상군이고, TI는 피부염을 유발시키기 위한 TNF-α 및 IFN-γ를 처리한 군이다. ###는 정상군 대비 TI 군에서의 RANTES(A), TARC(B) 및 IL-8(C)의 함량이 통계적으로 유의미하게 증가한 것을 나타내며, p<0.001이고, *, **, ***는 TI군 대비 본 발명의 나한과 잔사 추출물 또는 양성대조군(데사메타손, 사이클로스포린 A 또는 실리마린)을 처리한 군의 RANTES(A), TARC(B) 및 IL-8(C)의 함량이 통계적으로 유의미하게 감소하였다는 것으로, *는 p<0.05이고, **는 p<0.01이며, ***는 p<0.001임을 의미한다.
본 발명은 나한과 잔사 추출물을 유효성분으로 함유하는 염증성 질환의 예방 또는 치료용 약학 조성물에 관한 것이다.
상기 나한과 잔사 추출물은
(1) 건조한 나한과에 추출용매를 가하여 추출 및 여과하여 상등액을 제거하는 단계; 및
(2) 상기 상등액을 제거하고 남은 나한과 잔사에 에탄올을 가하여 나한과 잔사 추출물을 획득하는 단계;를 포함하는 나한과 잔사 추출물의 제조방법으로 제조된 것이 바람직하지만, 이에 한정하지 않는다. 상기 추출용매는 물, C1~C4의 저급 알코올 또는 이들의 혼합물인 것이 바람직하며, 더 바람직한 추출용매는 물이지만 이에 한정하는 것은 아니다.
상기 염증성 질환은 천식, 만성폐쇄성폐질환, 알레르기, 전신성 홍반성 루푸스, 경피증, 궤양성 대장염, 크론병, 아토피성 피부염, 건선, 아나필락시스, 피부염, 당뇨병성 망막증, 망막염, 황반 변성, 포도막염, 결막염, 관절염, 류마티스성 관절염, 강직성 척추염, 골관절염, 골다공증, 당뇨, 당뇨성 신장병, 신염, 신장염, 쇼그렌 증후군, 크론씨 병, 자가 면역 췌장염, 치주질환, 이식편대숙주질환, 만성 골반 염증 질환, 자궁내막염, 비염, 편도염, 중이염, 인후염, 방광염 및 만성 전립선염 중에서 하나 이상의 염증성 질환인 것이 바람직하지만 이에 한정하지 않는다.
본 발명에 따른 나한과 잔사 추출물을 포함하는 약학조성물은 크림, 젤, 패취, 분무제, 연고제, 경고제, 로션제, 리니멘트제, 파스타제 또는 카타플라스마제의 제형인 것이 바람직하지만 이에 한정하지 않는다.
상기 나한과 잔사 추출물 이외에 약학적으로 허용 가능한 담체, 부형제 또는 희석제를 더 포함할 수 있으며, 나한과 잔사 추출물을 포함하는 약학 조성물에 포함될 수 있는 담체, 부형제 또는 희석제로는 락토즈, 덱스트로즈, 수크로스, 올리고당, 솔비톨, 만니톨, 자일리톨, 에리스리톨, 말티톨, 전분, 아카시아 고무, 알지네이트, 젤라틴, 칼슘 포스페이트, 칼슘 실리케이트, 셀룰로즈, 메틸 셀룰로즈, 미정질 셀룰로스, 폴리비닐피롤리돈, 물, 메틸히드록시벤조에이트, 프로필히드록시벤조에이트, 탈크, 마그네슘 스테아레이트 및 광물유를 들 수 있다. 제제화할 경우에는 보통 사용하는 충진제, 증량제, 결합제, 습윤제, 붕해제, 계면활성제 등의 희석제 또는 부형제를 사용하여 조제될 수 있다.
본 발명의 나한과 잔사 추출물의 바람직한 투여량은 환자의 상태 및 체중, 질병의 정도, 약물형태, 투여경로 및 기간에 따라 다르지만, 당업자에 의해 적절하게 선택될 수 있다. 그러나 바람직한 효과를 위해서, 본 발명의 나한과 잔사 추출물을 포함하는 약학조성물은 1일 0.0001 내지 100㎎/㎏으로, 바람직하게는 0.001 내지 10㎎/㎏으로 투여하는 것이 좋다. 투여는 하루에 한번 투여할 수도 있고, 수회 나누어 투여할 수도 있다. 상기 투여량은 어떠한 면으로든 본 발명의 범위를 한정하는 것은 아니다.
또한, 본 발명은 나한과 잔사 추출물을 유효성분으로 포함하는 염증의 예방 또는 개선용 건강기능식품 조성물에 관한 것이다.
본 발명의 건강기능식품 조성물은 분말, 과립, 환, 정제, 캡슐, 캔디, 시럽 및 음료 중에서 선택된 어느 하나의 제형으로 제조될 수 있으며, 이에 한정하지 않는다.
본 발명의 건강기능식품 조성물을 식품첨가물로 사용하는 경우, 상기 건강기능식품 조성물을 그대로 첨가하거나 다른 식품 또는 식품성분과 함께 사용될 수 있고, 통상적인 방법에 따라 적절하게 사용될 수 있다. 유효성분은 그의 사용 목적(예방 또는 개선)에 따라 적절하게 사용될 수 있다. 일반적으로, 식품 또는 음료의 제조시 본 발명의 건강기능식품 조성물은 원료에 대하여 15 중량부 이하, 바람직하게는 10 중량부 이하의 양으로 첨가된다. 그러나 건강을 목적으로 하는 장기간의 섭취의 경우에는 상기 양은 상기 범위 이하일 수 있으며, 안전성 면에서 아무런 문제가 없기 때문에 유효성분은 상기 범위 이상의 양으로 사용될 수 있다.
상기 건강기능식품의 종류에 특별한 제한은 없다. 상기 건강기능식품 조성물을 첨가할 수 있는 식품의 예로는 육류, 소시지, 빵, 초콜릿, 캔디류, 스낵류, 과자류, 피자, 라면, 기타 면류, 껌류, 아이스크림류를 포함한 낙농제품, 각종 스프, 음료수, 차 드링크제, 알콜 음료 및 비타민 복합제 등이 있으며, 통상적인 의미에서의 건강식품을 모두 포함한다.
또한, 본 발명의 건강기능식품 조성물은 식품, 특히 기능성 식품으로 제조될 수 있다. 본 발명의 기능성 식품은 통상적으로 첨가되는 성분을 포함할 수 있다. 예를 들어, 단백질, 탄수화물, 지방, 영양소 및 조미제를 포함한다. 예컨대, 드링크제로 제조되는 경우에는 유효성분 이외에 천연 탄수화물 또는 향미제를 추가 성분으로서 포함시킬 수 있다. 상기 천연 탄수화물은 모노사카라이드(예컨대, 글루코오스, 프럭토오스 등), 디사카라이드(예컨대, 말토스, 수크로오스 등), 올리고당, 폴리사카라이드(예컨대, 덱스트린, 시클로덱스트린 등) 또는 당알코올(예컨대, 자일리톨, 소르비톨, 에리쓰리톨 등)인 것이 바람직하다. 상기 향미제는 천연 향미제(예컨대, 타우마틴, 스테비아 추출물 등)와 합성 향미제(예컨대, 사카린, 아스파르탐 등)를 이용할 수 있다.
상기 건강기능식품 조성물 이외에 여러 가지 영양제, 비타민, 전해질, 풍미제, 착색제, 펙트산 및 그의 염, 알긴산 및 그의 염, 유기산, 보호성 콜로이드 증점제, pH 조절제, 안정화제, 방부제, 글리세린, 알콜, 탄산음료에 사용되는 탄산화제 등을 더 함유할 수 있다. 이러한 상기 첨가되는 성분의 비율은 크게 중요하진 않지만 본 발명의 건강기능식품 조성물 100 중량부에 대하여, 0.01 내지 0.1 중량부의 범위에서 선택되는 것이 일반적이다.
또한, 본 발명은 나한과 잔사 추출물을 유효성분으로 포함하는 아토피 피부염 개선용 화장료 조성물에 관한 것이다.
본 발명의 일 구현 예에 따른 화장료 조성물에 있어서, 상기 아토피 피부염의 개선용 화장료 조성물은 용액, 현탁액, 유탁액, 페이스트, 겔, 크림, 로션, 파우더, 비누, 계면활성제-함유 클렌징, 오일, 분말 파운데이션, 파운데이션, 왁스 파운데이션 및 스프레이 중에서 선택된 어느 하나의 제형으로 제조될 수 있으며, 피부외용 연고, 크림, 유연화장수, 영양화장수, 팩, 에센스, 헤어토닉, 샴푸, 린스, 헤어 컨디셔너, 헤어 트리트먼트, 젤, 스킨로션, 스킨소프너, 스킨토너, 아스트린젠트, 로션, 밀크로션, 모이스처 로션, 영양로션, 마사지 크림, 영양크림, 모이스처 크림, 핸드 크림, 파운데이션, 영양에센스, 선스크린, 비누, 클렌징폼, 클렌징로션, 클렌징크림, 바디 로션 및 바디 클렌저로 이루어지는 군으로부터 선택된 어느 하나의 제형을 가질 수 있으나, 이에 제한되지 않는다. 이들 각 제형으로 이루어진 화장료 조성물은 그 제형의 제제화에 필요하고 적절한 각종의 기제와 첨가물을 함유할 수 있으며, 이들 성분의 종류와 양은 당업자에 의해 용이하게 선정될 수 있다.
본 발명의 화장료 조성물의 제형이 페이스트, 크림 또는 겔인 경우에는 담체 성분으로서 동물섬유, 식물섬유, 왁스, 파라핀, 전분, 트라가칸트, 셀룰로오스 유도체, 폴리에틸렌 글리콜, 실리콘, 벤토나이트, 실리카, 탈크 또는 산화아연 등이 이용될 수 있다.
본 발명의 화장료 조성물의 제형이 파우더 또는 스프레이인 경우에는 담체 성분으로서 락토스, 탈크, 실리카, 알루미늄 히드록시드, 칼슘 실리케이트 또는 폴리아미드 파우더가 이용될 수 있고, 특히 스프레이인 경우에는 추가적으로 클로로플루오로히드로카본, 프로판-부탄 또는 디메틸 에테르와 같은 추진체를 포함할 수 있다.
본 발명의 화장료 조성물의 제형이 용액 또는 유탁액의 경우에는 담체 성분으로서 용매, 용매화제 또는 유탁화제가 이용되고, 예컨대 물, 에탄올, 이소프로판올, 에틸 카보네이트, 에틸 아세테이트, 벤질 알코올, 벤질 벤조에이트, 프로필렌글리콜, 1,3-부틸글리콜 오일, 글리세롤 지방족 에스테르, 폴리에틸렌 글리콜 또는 소르비탄의 지방산 에스테르가 있다.
본 발명의 화장료 조성물의 제형이 현탁액인 경우에는 담체 성분으로서 물, 에탄올 또는 프로필렌 글리콜과 같은 액상 희석제, 에톡실화 이소스테아릴 알코올, 폴리옥시에틸렌 소르비톨 에스테르 및 폴리옥시에틸렌 소르비탄 에스테르와 같은 현탁제, 미소결정성 셀룰로오스, 알루미늄 메타히드록시드, 벤토나이트, 아가 또는 트라칸트 등이 이용될 수 있다.
본 발명의 화장료 조성물의 제형이 계면-활성제 함유 클렌징인 경우에는 담체 성분으로서 지방족 알코올 설페이트, 지방족 알코올 에테르설페이트, 설포숙신산 모노에스테르, 아세티오네이트, 이미다졸리늄 유도체, 메틸타우레이트, 사르코시네이트, 지방산 아미드 에테르 설페이트, 알킬아미도베타인, 지방족 알코올, 지방산 글리세리드, 지방산 디에탄올아미드, 식물성 유, 리놀린 유도체 또는 에톡실화 글리세롤 지방산 에스테르 등이 이용될 수 있다.
이하, 실시예를 이용하여 본 발명을 더욱 상세하게 설명하고자 한다. 이들 실시예는 오로지 본 발명을 보다 구체적으로 설명하기 위한 것으로 본 발명의 범위가 이들에 의해 제한되지 않는다는 것은 당해 기술분야에서 통상의 지식을 가진 자에게 있어 자명한 것이다.
실시예
1. 나한과 추출물의 제조
건조 나한과는 200g을 분쇄한 후, 물 2L를 가하여 3시간 동안 환류 추출하였다. 추출액을 여과하고 남은 잔사에 다시 물 1L를 가하고 1시간 동안 환류 추출한 후 여과하였다. 1차 추출액과 2차 추출액을 합하여 농축, 건조하여 나한과 추출물을 제조하였다.

실시예 1에서 추출하고 남은 나한과 잔사를 건조하여 얻어진 건조 잔사 50g에 1L의 70%(v/v) 에탄올을 첨가한 후, 3시간 동안 환류 추출하였다. 추출액을 여과한 후, 농축, 건조하여 나한과 잔사 추출물을 제조하였다.

잔사
추출물의 항염증 효능 평가
1) MTT 어세이
생쥐 대식세포주인 RAW 264.7 세포를 96웰 플레이트에 적정 농도로 분주한 후, 상기 실시예 1 및 2에서 제조한 나한과 추출물과 나한과 잔사 추출물을 농도별(100, 200㎍/㎖)로 처리하여 24시간 동안 배양하였다. 배양이 끝난 세포에 MTT 용액을 첨가, 4시간 반응시킨 후 ELISA 리더를 이용하여 540nm의 흡광도를 측정하였고, 이후 대조군과의 비교를 통해 상대적인 세포생존율(% of control)을 계산하였다.
그 결과, 도 1에 개시한 바와 같이 나한과 추출물과 나한과 잔사 추출물은 세포독성이 없어 세포생존율에 영향을 미치지 않는다는 것을 확인하였다.
2) NO 측정
48웰 플레이트에 적정 농도로 분주한 RAW 274.7 세포에 LPS(400ng/㎖)를 처리하고, 상기 실시예 1 및 2에서 제조한 나한과 추출물과 나한과 잔사 추출물을 농도별(100, 200㎍/㎖)로 처리하여 24시간 동안 배양하였다. 배양 상층액을 취한 후 Griess 시약을 사용하여 제조사의 방법에 따라 NO 양을 측정하였다.
그 결과, 도 2에 개시한 바와 같이 본 발명의 나한과 잔사 추출물은 LPS에 의해 증가된 NO 생성량을 농도 의존적으로 감소시켰으나, 나한과 추출물은 LPS에 의해 증가된 NO 생성량의 감소에는 아무런 영향을 미치지 못하였다.
3) PGE2(Prostaglandin E2) 측정
48웰 플레이트에 적정 농도로 분주한 RAW 274.7 세포에 LPS(400ng/㎖)를 처리하고, 상기 실시예 1 및 2에서 제조한 나한과 추출물과 나한과 잔사 추출물을 농도별(100, 200㎍/㎖)로 처리하여 24시간 동안 배양하였다. 배양 상층액을 취한 후 PGE2 ELISA 키트를 사용하여 제조사의 방법에 따라 PGE2의 생성량을 측정하였다.
그 결과, 도 3에 개시한 바와 같이 본 발명의 나한과 잔사 추출물과 나한과 추출물 둘 다, LPS에 의해 증가된 PGE2 생성량을 감소시켰으며, 본 발명의 나한과 잔사 추출물이 나한과 추출물보다 효과적으로 PGE2 생성량을 감소시켰다.
5) IL-1β 측정
48웰 플레이트에 적정 농도로 분주한 RAW 274.7 세포에 LPS(400ng/㎖)를 처리하고, 상기 실시예 1 및 2에서 제조한 나한과 추출물과 나한과 잔사 추출물을 농도별(100, 200㎍/㎖)로 처리하여 24시간 동안 배양하였다. 배양 상층액을 취한 후 IL-1β ELISA 키트를 사용하여 제조사의 방법에 따라 IL-1β의 생성량을 측정하였다.
그 결과, 도 4에 개시한 바와 같이 본 발명의 나한과 잔사 추출물이 나한과 추출물에 비해 LPS에 의해 증가된 IL-1β 생성량을 현저하게 감소시키는 것을 확인하였다.

4. 마우스 비만세포인 MC/9 세포에서 나한과
잔사
추출물의 세포독성 및 사이토카인 분비 억제 효능 평가
(1) 세포 배양
마우스 비만세포(mast cell) MC/9 세포를 American Type Culture Collection(ATCC, Rockville, MD, USA)로부터 구입하여 10% heat-inactivated Fetal Bovine Serum(FBS), 100units/㎖의 페니실린(penicillin) 및 100㎍/㎖의 스트렙토마이신(streptomycin)을 첨가한 DMEM 배지로 37℃, 5% CO2 환경에서 배양하였다. 나한과 잔사 추출물의 세포 독성 및 사이토카인의 분비 억제 효능 평가에 사용하였다.
(2) 세포독성 분석
나한과 잔사 추출물의 MC/9 비만 세포 독성을 확인하기 위하여, CCK-8(Cell Counting Kit-8) 어세이를 실시하였다. 96웰 플레이트에서 MC/9 비만세포에 시료를 농도별(50, 100, 200㎍/㎖)로 처리하고 24시간 동안 배양하였다. CCK-8을 10% 농도로 첨가한 다음 2시간 동안 37℃에서 반응시킨 후, 마이크로플레이트 분광광도계(microplate spectrophotometer)(Biorad, Hercules, CA, USA)를 이용하여 450nm에서 흡광도를 측정하고, 대조군과의 비교를 통해 상대적인 세포 생존율(%)을 계산하였다.
그 결과, 도 5에 개시한 바와 같이, 나한과 잔사 추출물은 MC/9 비만 세포독성이 거의 없는 것으로 나타났다.
(3) 사이토카인 분비 억제 효능 분석
배양된 MC/9 마우스 비만 세포를 96웰 플레이트에 접종한 후, 아토피 유발 물질인 20nM의 PMA(phorbol myristate acetate)와 칼슘 이오노포어(calcium ionophore: CI)인 A23187을 0.5μM의 농도로 처리하고, 동시에 다양한 농도(50, 100, 200㎍/㎖)의 나한과 잔사 추출물을 처리하여 공동 배양하였다. 양성대조군으로 0.1μM 및 1μM의 덱사메타손(Dexamethasone)을 각각 처리하였다. 4시간 후에 배지에 분비된 사이토카인(TNF-α 및 IL-6)의 분비량을 ELISA kit(R&D systems Inc., Minneapolis, MN, USA)로 측정하였다. 100% 활성은 PMA와 A23187이 처리되지 않은 군에서 처리된 군 사이의 사이토카인 분비량의 차이로 계산하였다.
사이토카인의 분비량은 식 (1)에 적용하여 계산하였으며, 사이토카인의 억제율은 식 (2)에 적용하여 계산하였다.

모든 측정 결과는 평균(mean)과 표준편차(standard deviation; SD)로 나타내었으며, 실험군 간의 차이는 Student's t-test를 사용하여 통계학적 분석을 수행하였고, p < 0.05 값인 경우에 통계적으로 유의성이 있는 것으로 판정하였다.
그 결과, 도 6에 개시한 바와 같이, PMA(phorbol myristate acetate)와 칼슘 이오노포어(calcium ionophore: CI)를 처리한 군(PMACI)에서는 TNF-α 및 IL-6의 함량이 통계적으로 유의미하게 증가하였으며, 본 발명의 나한과 잔사 추출물 및 양성대조군인 덱사메타손을 처리한 군에서는 통계적으로 유의미하게 TNF-α 및 IL-6의 함량이 감소한 것을 확인하였다.

분비 억제 효능 평가
(1) 세포 배양
인간 각질세포(human keratinocyte) HaCaT 세포를 10% heat-inactivated Fetal Bovine Serum(FBS), 100 units/㎖의 페니실린(penicillin) 및 100㎍/㎖의 스트렙토마이신(streptomycin)을 첨가한 DMEM 배지로 37℃, 5% CO2 환경에서 배양하였다.
(2) 세포독성 분석
나한과 잔사 추출물의 인간 각질세포(human keratinocyte) HaCaT 세포독성을 확인하기 위하여, CCK-8(Cell Counting Kit-8) 어세이를 실시하였다. 96웰 플레이트에서 HaCaT 세포에 시료를 농도별(50, 100, 200㎍/㎖)로 처리하고 24시간 동안 배양하였다. CCK-8을 10% 농도로 첨가한 다음 2시간 동안 37℃에서 반응시킨 후, 마이크로플레이트 분광광도계(Biorad, Hercules, CA, USA)를 이용하여 450nm에서 흡광도를 측정하고, 대조군과의 비교를 통해 상대적인 세포 생존율(%)을 계산하였다.
그 결과, 도 7에 개시한 바와 같이, 나한과 잔사 추출물은 HaCaT 세포독성이 거의 없는 것으로 나타났다.
(3) 케모카인 분비 억제 효능 분석
배양된 인간 각질 세포를 6-웰 플레이트(well plate)에 접종한 후, 피부염증 반응을 유발시키기 위해 10ng/㎖의 TNF-α 및 10ng/㎖의 IFN-γ로 처리하고, 동시에 다양한 농도(50, 100 및 200㎍/㎖)의 나한과 잔사 추출물을 처리하여 공동 배양하였다. 양성대조군으로 덱사메타손(Dexamethasone; Dex), Cyclosporin A을 0.01, 0.1 및 1μM의 농도로 처리하고 6, 12 및 25μM의 실리마린(silymarin)을 각각 처리하였다. 24시간 후에 배지에 분비된 케모카인의 분비량을 ELISA kit (R&D systems Inc., Minneapolis, MN, USA)로 측정하였다. 100% 활성은 TNF-α 및 IFN-γ이 처리되지 않은 군에서 처리된 군 사이의 사이토카인 분비량의 차이로 계산하였다.
케모카인의 분비량은 식 (3)에 적용하여 계산하였으며, 케모카인의 억제율은 식 (4)에 적용하여 계산하였다.

Claims (9)
- 나한과 잔사 추출물을 유효성분으로 함유하는 염증성 질환의 예방 또는 치료용 약학 조성물.
- 제1항에 있어서, 상기 나한과 잔사 추출물은(1) 건조한 나한과에 추출용매를 가하여 추출 및 여과하여 상등액을 제거하는 단계; 및(2) 상기 상등액을 제거하고 남은 나한과 잔사에 에탄올을 가하여 나한과 잔사 추출물을 획득하는 단계;를 포함하는 방법으로 제조된 것을 특징으로 하는 염증성 질환의 예방 또는 치료용 약학 조성물.
- 제2항에 있어서, 상기 추출용매는 물, C1~C4의 저급 알코올 또는 이들의 혼합물인 것을 특징으로 하는 염증성 질환의 예방 또는 치료용 약학 조성물.
- 제1항에 있어서, 상기 염증성 질환은 천식, 만성폐쇄성폐질환, 알레르기, 전신성 홍반성 루푸스, 경피증, 궤양성 대장염, 크론병, 아토피성 피부염, 건선, 아나필락시스, 피부염, 당뇨병성 망막증, 망막염, 황반 변성, 포도막염, 결막염, 관절염, 류마티스성 관절염, 강직성 척추염, 골관절염, 골다공증, 당뇨, 당뇨성 신장병, 신염, 신장염, 쇼그렌 증후군, 크론씨 병, 자가 면역 췌장염, 치주질환, 이식편대숙주질환, 만성 골반 염증 질환, 자궁내막염, 비염, 편도염, 중이염, 인후염, 방광염 및 만성 전립선염 중에서 선택된 어느 하나 이상인 것을 특징으로 하는 염증성 질환의 예방 또는 치료용 약학 조성물.
- 제1항에 있어서, 상기 나한과 잔사 추출물 이외에 약학적으로 허용 가능한 담체, 부형제 또는 희석제를 더 포함하는 것을 특징으로 하는 염증성 질환의 예방 또는 치료용 약학 조성물.
- 나한과 잔사 추출물을 유효성분으로 포함하는 염증의 예방 또는 개선용 건강기능식품 조성물.
- 제6항에 있어서, 상기 조성물은 분말, 과립, 환, 정제, 캡슐, 캔디, 시럽 및 음료 중에서 선택된 어느 하나의 제형으로 제조되는 것을 특징으로 하는 염증의 예방 또는 개선용 건강기능식품 조성물.
- 나한과 잔사 추출물을 유효성분으로 포함하는 아토피 피부염 개선용 화장료 조성물.
- 제8항에 있어서, 상기 화장료 조성물은 용액, 현탁액, 유탁액, 페이스트, 겔, 크림, 로션, 파우더, 비누, 계면활성제-함유 클렌징, 오일, 분말 파운데이션, 파운데이션, 왁스 파운데이션 및 스프레이 중에서 선택된 어느 하나의 제형으로 제조되는 것을 특징으로 하는 아토피 피부염 개선용 화장료 조성물.
