Abstract

본 발명은 오미자 잎 추출물을 유효성분으로 포함하는 아토피 피부염의 예방, 개선 또는 치료용 조성물에 관한 것이다.
아토피는 최근 들어, 유병률이 급격히 증가함에 따라 그 위험성이 크게 부각되고 있는 과민증(hypersensitivity)의 일종으로, 면역학적 관점에서는 면역글로불린E(immunoglobulin E)와 알레르겐(allergen)의 면역반응에 의한 주요증상을 동반하는 유전경향이 강한 알레르기성 질환군으로 정의된다. 특히, 아토피의 대표적 증상인 아토피 피부염(atopic dermatitis)은 전 인구의 0.5~1%, 어린이의 5~10%가 심각한 증상을 호소하고 있지만, 유전적인 요인과 면역계 결핍에 관련 있는 것으로 추정될 뿐 아직 정확한 원인은 밝혀진 바 없고, 환경 및 식생활 개선을 통해 다소 완화되는 것으로 기대될 뿐 근본적인 치료방법이 전무한 실정이다.
그러나, 아토피 피부염은 개인차를 보이기는 하지만, 대부분 극심한 가려움증을 비롯한 피부건조증, 발진, 진물, 부스럼딱지, 비늘 같은 껍질이 있는 피부(인비늘) 등을 수반하며, 이로 인한 정서적 불안, 스트레스, 긴장, 좌절, 분노의 감정과 함께 기타 알레르기 질환인 두드러기, 금속 알레르기, 천식이나 알레르기성 비염 등을 동반하는 경우가 빈번한 바, 오늘날 국민건강을 위협하는 심각한 사회문제로 떠오르고 있다. 한편, 현재까지 아토피 피부염에 효과가 있는 것으로 알려진 대표적 약물로는 스테로이드제(steroid), 항히스타민제(antihistamine), 항생제(antibiotic), 비스테로이드제를 비롯한 기타 진정제와 신경안정제 등을 들 수 있다.
한편, 오미자나무(Schizandra

)는 쌍떡잎 식물로 목련목 오미자과의 낙엽 덩굴식물이다. 주로 산골짜기에서 자라며, 줄기는 갈색이고 나무를 기어 오르는 성질이 있다. 잎은 어긋나고 넓은 타원형, 긴 타원형 또는 달걀 모양이며 뒷면 잎맥 위에는 털이 있고 가장자리에 치아 모양의 톱니가 있다. 꽃은 6∼7월에 피고 단성화이며 약간 붉은빛이 도는 황백색이다. 꽃이 핀 다음 암꽃의 꽃턱은 길이 3∼5cm로 자라서 열매가 수상으로 달린다. 열매는 장과(漿果)로 거의 둥글고 이삭 모양으로 여러 개가 달린다. 8∼9월에 홍색으로 익으며 1∼2개의 홍갈색 종자가 들어 있다. 어린 순은 나물로 먹는다. 열매에 신맛, 단맛, 쓴맛, 짠맛, 매운맛의 다섯 가지 맛이 섞여 있어 오미자라 하며 약용한다. 한방에서 자양(滋養), 강장, 진해(鎭咳), 거담(祛痰), 지한(止汗) 등의 효력이 있어 해수·유정(遺精), 구갈, 도한(盜汗), 급성간염 등에 처방한다. 민간에서는 오미자차를 만들어 마시며 술도 담근다.
오미자 관련 선행기술로는 한국공개특허 제2015-0027676호에 오미자 추출물을 포함하는 대사증후군 관련 질환의 예방 또는 치료용 조성물이 공개되어 있고, 한국공개특허 제2017-0017388호에 오미자 줄기 추출물을 함유하는 신경계 질환의 예방 또는 치료용 조성물이 개시되어 있으나, 본 발명의 오미자 잎 추출물을 유효성분으로 포함하는 아토피 피부염의 예방, 개선 또는 치료용 조성물은 개시된 바 없다.
본 발명은 상기와 같은 요구에 의해 도출된 것으로서, 본 발명은 오미자 잎 추출물을 유효성분으로 포함하는 아토피 피부염의 예방, 개선 또는 치료용 조성물을 제공하고, 본 발명의 오미자 잎 추출물이 세포독성이 거의 없으며, 항히스타민 효과와 NO 소거활성이 우수하고, 비만세포에서 사이토카인(TNF-α, IL-6)의 분비량을 감소시키며, 인간각질세포(HaCaT cell)에서의 케모카인(TARC, RANTES)의 분비량을 현저하게 감소시킨다는 것을 확인함으로써, 본 발명을 완성하였다.

본 발명은 오미자 잎 추출물을 유효성분으로 포함하는 아토피 피부염의 예방, 개선 또는 치료용 조성물에 관한 것으로, 본 발명의 유효성분인 오미자 추출물은 대식 세포와 비만 세포에서의 항히스타민 효과 및 NO 생성억제 효과가 있는 것이다. 특히, 오미자 잎 추출물은 오미자 열매 또는 줄기 추출물에 비해 항히스타민 활성이 우수하며, 비만세포에서 사이토카인(TNF-α, IL-6)의 분비량을 감소시키고, 인간각질세포(HaCaT cell)에서의 케모카인(TARC, RANTES)의 분비량을 현저하게 감소시키므로, 본 발명의 조성물을 아토피 피부염의 예방, 개선 또는 치료용 조성물로 유용하게 활용할 수 있다.
도 1은 본 발명의 오미자 잎 추출물과, 비교예의 오미자 열매 추출물 및 오미자 줄기 추출물을 처리한 MC9 마우스의 비만세포의 세포생존율(%)을 분석한 결과이다.
도 2는 본 발명의 오미자 잎 추출물과, 비교예의 오미자 열매 추출물 및 오미자 줄기 추출물을 처리한 비만세포에서의 항히스타민 효과를 확인한 결과이다. ***, ****은 오미자 잎 추출물이 아무것도 처리하지 않은 음성대조군(Con)에 비해 현저하게 히스타민 함량이 감소하였다는 것으로, ***은 p<0.001이고, ****은 p<0.0001임을 의미한다.
도 3는 본 발명의 오미자 잎 추출물과, 비교예의 오미자 열매 추출물을 처리한 비만세포에서의 NO(%) 소거 활성을 확인한 결과이다. NMMA는 양성대조군으로, 50μM의 N-모노메틸 아르기닌(N-monomethyl arginine)을 처리한 것이다.

본 발명은 오미자 잎 추출물을 유효성분으로 포함하는 아토피 피부염의 예방 또는 치료용 약학 조성물에 관한 것이다.
상기 오미자 잎 추출물은 하기의 단계를 포함하는 방법에 의해 제조되는 것일 수 있으나, 이에 한정하지 않는다:
1) 오미자 잎 건조 분말에 추출용매를 가하여 추출하는 단계;
2) 단계 1)의 추출물을 여과하는 단계; 및
3) 단계 2)의 여과한 추출물을 감압 농축하고 건조하여 추출물을 제조하는 단계.
상기 단계 1)에서 추출용매는 물, C1~C4의 저급 알코올 또는 이들의 혼합물인 것이 바람직하며, 더 바람직하게는 에탄올 추출물이지만 이에 한정하지 않는다.
상기 제조방법에 있어서, 오미자 잎의 추출은 여과법, 열수 추출, 침지 추출, 환류 냉각 추출 및 초음파 추출 등의 당 업계에 공지된 모든 통상적인 방법을 이용할 수 있다. 상기 단계 3)의 감압농축은 진공 감압 농축기 또는 진공회전증발기를 이용하는 것이 바람직하나 이에 한정하지 않는다. 또한, 건조는 감압건조, 진공건조, 비등건조, 분무 건조 또는 동결 건조하는 것이 바람직하나 이에 한정하지 않는다.
본 발명의 조성물은 상기 유효성분 이외에 약학적으로 허용 가능한 담체, 부형제 또는 희석제를 더 포함할 수 있으며, 경구 또는 비경구의 여러 가지 제형일 수 있다. 제제화할 경우에는 보통 사용하는 충진제, 증량제, 결합제, 습윤제, 붕해제, 계면활성제 등의 희석제 또는 부형제를 사용하여 조제된다. 경구투여를 위한 고형 제제에는 캡슐제, 산제, 과립제, 정제, 환제 등이 포함되며, 이러한 고형 제제는 하나 이상의 화합물에 적어도 하나 이상의 부형제 예를 들면, 전분, 탄산칼슘, 수크로오스(sucrose) 또는 락토오스(lactose), 젤라틴 등을 섞어 조제된다. 또한, 단순한 부형제 이외에 스테아린산 마그네슘, 탈크 등과 같은 윤활제들도 사용된다. 경구 투여를 위한 액상 제제로는 현탁액, 에멀전, 시럽, 에어로졸 등이 해당되는데 흔히 사용되는 단순 희석제인 물, 리퀴드 파라핀 이외에 여러 가지 부형제, 예를 들면 습윤제, 감미제, 방향제, 보존제 등이 포함될 수 있다. 비경구 투여를 위한 제제에는 멸균된 수용액, 비수성 용제, 현탁제, 유제, 동결건조제제, 좌제가 포함된다. 비수성 용제 및 현탁 용제로는 프로필렌글리콜(propylene glycol), 폴리에틸렌 글리콜, 올리브 오일과 같은 식물성 기름, 에틸올레이트와 같은 주사 가능한 에스테르 등이 사용될 수 있다. 좌제의 기제로는 위텝솔(witepsol), 마크로골, 트윈(tween) 61, 카카오지, 라우린지, 글리세로 젤라틴 등이 사용될 수 있다. 비경구 투여 시 피부 외용 또는 복강 내, 직장, 정맥, 근육, 피하, 자궁 내 경막 또는 뇌혈관 내 주사 방식을 선택하는 것이 바람직하며, 가장 바람직하게는 피부 외용으로 사용한다.
본 발명에 따른 약학 조성물은 약제학적으로 유효한 양으로 투여한다. 본 발명에 있어서, "약제학적으로 유효한 양"은 의학적 치료에 적용 가능한 합리적인 수혜/위험 비율로 질환을 치료하기에 충분한 양을 의미하며, 유효량의 수준은 환자의 질환의 종류, 중증도, 약물의 활성, 약물에 대한 민감도, 투여 시간, 투여 경로 및 배출 비율, 치료기간, 동시 사용되는 약물을 포함한 요소 및 기타 의학 분야에 잘 알려진 요소에 따라 결정될 수 있다. 본 발명의 조성물은 개별 치료제로 투여하거나 다른 치료제와 병용하여 투여될 수 있고 종래의 치료제와는 순차적 또는 동시에 투여될 수 있으며, 단일 또는 다중 투여될 수 있다. 상기한 요소들을 모두 고려하여 부작용없이 최소한의 양으로 최대 효과를 얻을 수 있는 양을 투여하는 것이 중요하며, 이는 당업자에 의해 용이하게 결정될 수 있다.
본 발명의 조성물의 투여량은 환자의 체중, 연령, 성별, 건강상태, 식이, 투여시간, 투여방법, 배설률 및 질환의 중증도에 따라 그 범위가 다양하며, 일일 투여량은 오미자 잎 추출물의 양을 기준으로 0.01~2,000mg/kg이고, 바람직하게는 30~500mg/kg이고, 더욱 바람직하게는 50~300mg/kg이며, 하루 1~6회 투여될 수 있다. 본 발명의 조성물은 단독으로 또는 수술, 방사선 치료, 호르몬 치료, 화학 치료 및 생물학적 반응 조절제를 사용하는 방법들과 병용하여 사용할 수 있다.
또한, 본 발명은 오미자 잎 추출물을 유효성분으로 포함하는 아토피 피부염의 예방 또는 개선용 건강기능식품 조성물에 관한 것이다.
상기 건강기능식품 조성물은 분말, 과립, 환, 정제, 캡슐, 캔디, 시럽 및 음료 중에서 선택된 어느 하나의 제형으로 제조되는 것이 바람직하지만 이에 한정하는 것은 아니다. 본 발명의 건강기능식품 조성물을 식품첨가물로 사용하는 경우, 상기 오미자 잎 추출물을 그대로 첨가하거나 다른 식품 또는 식품 성분과 함께 사용될 수 있고, 통상적인 방법에 따라 적절하게 사용될 수 있다. 유효 성분의 양은 그의 사용 목적(예방, 건강 또는 치료적 처치)에 따라 적합하게 결정될 수 있다. 일반적으로, 식품 또는 음료의 제조시에 본 발명의 조성물은 원료에 대하여 15 중량부 이하, 바람직하게는 10 중량부 이하의 양으로 첨가된다. 그러나 건강 및 위생을 목적으로 하거나 또는 건강 조절을 목적으로 하는 장기간의 섭취의 경우에는 상기 양은 상기 범위 이하일 수 있으며, 안전성 면에서 아무런 문제가 없기 때문에 유효성분은 상기 범위 이상의 양으로도 사용될 수 있다. 상기 식품의 종류에는 특별한 제한은 없다. 상기 추출물을 첨가할 수 있는 식품의 예로는 육류, 소시지, 빵, 초콜릿, 캔디류, 스낵류, 과자류, 피자, 라면, 기타 면류, 껌류, 아이스크림류를 포함한 낙농제품, 각종 수프, 음료수, 차, 드링크제, 알코올 음료 및 비타민 복합체 등이 있으며, 통상적인 의미에서의 건강기능식품을 모두 포함한다.
본 발명의 조성물을 건강 음료로 사용할 경우, 통상의 음료와 같이 여러 가지 향미제 또는 천연 탄수화물 등을 추가 성분으로서 함유할 수 있다. 상술한 천연 탄수화물은 포도당, 과당과 같은 모노사카라이드, 말토스, 슈크로스와 같은 디사카라이드, 텍스트린, 사이클로텐스트린과 같은 폴리사카라이드, 자일리톨, 소르비톨, 에리트리톨 등의 당알콜이다. 감미제로서는 타우마틴, 스테비아 추출물과 같은 천연 감미제나, 사카린, 아스파르탐과 같은 합성 감미제 등을 사용할 수 있다. 상기 천연 탄수화물의 비율은 본 발명의 조성물 100g당 일반적으로 약 0.01~0.04g, 바람직하게는 약 0.02~0.03g이다. 상기 외에 본 발명의 조성물은 여러 가지 영양제, 비타민, 전해질, 풍미제, 착색제, 펙트산 및 그의 염, 알긴산 및 그의 염, 유기산, 보호성 콜로이드 중점제, pH 조절제, 안정화제, 방부제, 글리세린, 알코올, 탄산음료에 사용되는 탄산화제 등을 함유할 수 있다. 그 밖에 본 발명의 조성물은 천연 과일쥬스, 과일쥬스 음료 및 야채 음료의 제조를 위한 과육을 함유할 수 있다. 이러한 성분은 독립적으로 또는 혼합하여 사용할 수 있다. 이러한 첨가제의 비율은 크게 중요하진 않지만 본 발명의 조성물은 100 중량부 당 0.01~0.1 중량부의 범위에서 선택되는 것이 일반적이다.
또한, 본 발명은 오미자 잎 추출물을 유효성분으로 포함하는 아토피 피부염 개선용 화장료 조성물에 관한 것이다.
상기 조성물은 용액, 현탁액, 유탁액, 페이스트, 겔, 크림, 로션, 파우더, 비누, 계면활성제-함유 클렌징, 오일, 분말 파운데이션, 유탁액, 파운데이션, 왁스 파운데이션 및 스프레이 중에서 선택된 어느 하나의 제형으로 제조되는 것이 바람직하지만 이에 한정하는 것은 아니다.
본 발명의 화장료 조성물의 제형이 용액 또는 유탁액의 경우에는 담체 성분으로서 용매, 용매화제 또는 유탁화제가 이용되고, 예컨대 물, 에탄올, 이소프로판올, 에틸 카보네이트, 에틸 아세테이트, 벤질 알코올, 벤질 벤조에이트, 프로필렌글리콜, 1,3-부틸글리콜 오일, 글리세롤 지방족 에스테르, 폴리에틸렌 글리콜 또는 소르비탄의 지방산 에스테르가 있다.
본 발명의 화장료 조성물의 제형이 현탁액인 경우에는 담체 성분으로서 물, 에탄올 또는 프로필렌 글리콜과 같은 액상 희석제, 에톡실화 이소스테아릴 알코올, 폴리옥시에틸렌 소르비톨 에스테르 및 폴리옥시에틸렌 소르비탄 에스테르와 같은 현탁제, 미소결정성 셀룰로오스, 알루미늄 메타히드록시드, 벤토나이트, 아가 또는 트라칸트 등이 이용될 수 있다.
본 발명의 화장료 조성물의 제형이 페이스트, 크림 또는 겔인 경우에는 담체 성분으로서 동물섬유, 식물섬유, 왁스, 파라핀, 전분, 트라가칸트, 셀룰로오스 유도체, 폴리에틸렌 글리콜, 실리콘, 벤토나이트, 실리카, 탈크 또는 산화아연 등이 이용될 수 있다.
본 발명의 화장료 조성물의 제형이 파우더 또는 스프레이인 경우에는 담체 성분으로서 락토스, 탈크, 실리카, 알루미늄 히드록시드, 칼슘 실리케이트 또는 폴리아미드 파우더가 이용될 수 있고, 특히 스프레이인 경우에는 추가적으로 클로로플루오로히드로카본, 프로판-부탄 또는 디메틸 에테르와 같은 추진체를 포함할 수 있다.
본 발명의 화장료 조성물의 제형이 계면-활성제 함유 클렌징인 경우에는 담체 성분으로서 지방족 알코올 설페이트, 지방족 알코올 에테르설페이트, 설포숙신산 모노에스테르, 아세티오네이트, 이미다졸리늄 유도체, 메틸타우레이트, 사르코시네이트, 지방산 아미드 에테르 설페이트, 알킬아미도베타인, 지방족 알코올, 지방산 글리세리드, 지방산 디에탄올아미드, 식물성 유, 리놀린 유도체 또는 에톡실화 글리세롤 지방산 에스테르 등이 이용될 수 있다.
본 발명의 화장료 조성물은 유효성분 이외에 추가로 동일 또는 유사한 기능을 나타내는 아토피 피부염을 개선할 수 있는 성분을 1종 이상 함유할 수 있다.
본 발명의 화장료 조성물은 생분해성 마이크로 니들 또는 패치 등을 이용하여 피부에 직접 적용할 수 있다.
또한, 본 발명은 오미자 잎 추출물을 유효성분으로 포함하는 아토피 피부염 개선용 의약외품 조성물에 관한 것이다.
본 발명에서, "의약외품 조성물"은 사람이나 동물의 질병을 치료, 경감, 처치 또는 예방할 목적으로 사용되는 섬유, 고무제품 또는 이와 유사한 것, 인체에 대한 작용이 약하거나 인체에 직접 작용하지 않으며, 기구 또는 기계가 아닌 것과 이와 유사한 것, 감염 예방을 위하여 살균, 살충 및 이와 유사한 용도로 사용되는 제제 중 하나에 해당하는 물품으로서, 사람이나 동물의 질병을 진단, 치료, 경감, 처치 또는 예방할 목적으로 사용하는 물품 중 기구, 기계 또는 장치가 아닌 것 및 사람이나 동물의 구조와 기능에 약리학적 영향을 줄 목적으로 사용하는 물품 중 기구, 기계 또는 장치가 아닌 것을 제외한 물품을 의미하며, 구체적으로 피부 외용제 또는 개인위생용품일 수 있으나, 이에 제한되지 않는다. 본 발명의 의약외품 조성물은 아토피 피부염의 개선 효과를 나타낼 수 있다면 다양한 중량%로 포함할 수 있으며, 바람직하게는 0.00001 내지 10 중량%로 포함할 수 있다.
이하, 실시예를 이용하여 본 발명을 더욱 상세하게 설명하고자 한다. 이들 실시예는 오로지 본 발명을 보다 구체적으로 설명하기 위한 것으로 본 발명의 범위가 이들에 의해 제한되지 않는다는 것은 당해 기술분야에서 통상의 지식을 가진 자에게 있어 자명한 것이다.
실시예
1. 오미자 잎 추출물의 제조
1kg의 오미자 잎 건조분말에 대하여, 15ℓ의 70%(v/v) 에탄올을 가하고, 85℃에서 3시간 동안 환류추출한 후, 여과한 액을 50℃에서 감압 농축, 건조하여 오미자 잎 추출물을 획득하였다.

생존률
분석
MC/9 세포에 24시간 동안 각각의 시료를 농도별로 희석하여 처리하고, MTT 어세이 기법으로 세포 생존율을 550nm에서 마이크로플레이트 리더(Molecular Devices, Sunnyvale, CA, USA)로 측정하였다.
그 결과, 도 1에 개시한 바와 같이 오미자 열매, 잎 및 줄기 추출물 모두 세포독성이 거의 나타나지 않았다.
실시예
3. 항히스타민 효과 분석
48웰 플레이트에 적정 농도로 분주한 MC/9 세포에 PMA(50nM)와 A23189(1μM)를 처리하고, 본 발명의 오미자 잎 추출물과 비교예로 오미자 열매 추출물 및 오미자 줄기 추출물을 농도별(25, 50, 100㎍/㎖)로 처리하여 24시간 동안 배양하였다. 배양 상층액을 취한 후 히스타민 ELISA 어세이 키트를 사용하여 제조사의 방법에 따라 히스타민(histamine) 양을 측정하였다.
그 결과, 도 2에 개시한 바와 같이 오미자 추출물 모두 농도 의존적으로 히스타민의 농도가 감소하는 효과가 나타났으며, 특히 오미자 잎 추출물의 경우, 오미자 열매 추출물 또는 오미자 줄기 추출물에 비해 현저하게 히스타민의 함량이 줄어든 것을 확인할 수 있었다.

4. NO 측정
48웰 플레이트에 적정 농도로 분주한 RAW 274.7 세포에 본 발명의 오미자 잎 추출물과 비교예로 오미자 열매 추출물을 농도별(25, 50, 100㎍/㎖)로 처리한 후 LPS(500ng/㎖)를 처리하고 24시간 동안 배양하였다. 배양 상층액을 취한 후 Griess 시약을 사용하여 제조사의 방법에 따라 NO 양을 측정하였다.
그 결과 도 3에 개시한 바와 같이, 오미자 잎 추출물은 LPS 처리에 의해 유도된 NO의 생성량을 현저하게 감소시켰다.

5. 사이토카인 분비 억제 효능 분석
(1) 세포 배양
마우스 비만세포(mast cell) MC/9 세포를 American Type Culture Collection(ATCC, Rockville, MD, USA)로부터 구입하여 10% heat-inactivated Fetal Bovine Serum(FBS), 100 units/ml 페니실린(penicillin), 100 μg/ml 스트렙토마이신(streptomycin)을 첨가한 DMEM 배지로 37℃, 5% CO2 환경에서 배양하였다.
(2) 세포 독성
추출물의 세포 독성을 확인하고자 Cell Counting Kit-8(CCK-8) assay를 실시하였다. 96웰 플레이트(well plate)에서 MC/9 비만세포에 시료를 농도별(25. 50, 100, 200㎍/㎖)로 30분 동안 전처리한 다음, PMA(phorbol myristate acetate) 20nM과 칼슘 이오노포어(calcium ionophore)인 A23187을 0.5μM의 농도로 처리하고 배양하였다. 4시간 후에 배지에 CCK-8을 10% 농도로 첨가한 다음 2시간 동안 37℃에서 반응시킨 후, 마이크로플레이트 스펙트로포토미터(microplate spectrophotometer)(Biorad, Hercules, CA, USA)를 이용하여 450nm에서 흡광도를 측정하고, 대조군과의 비교를 통해 상대적인 세포 생존율(%)을 계산하였다.
(3) 사이토카인 분비 억제 효능 분석
배양된 MC/9 마우스 비만 세포를 96-웰 플레이트(well plate)에 접종하여 시료를 농도별(25. 50, 100, 200㎍/㎖)로 30분 동안 전 처리한 다음, PMA(phorbol myristate acetate) 20nM과 칼슘 이오노포어(calcium ionophore)인 A23187을 0.5μM의 농도로 처리하고 4시간 동안 배양하였다. 양성대조군으로 0.1 및 1μM의 덱사메타손(Dexamethasone)을 각각 처리하였다. 4시간 후에 배지에 분비된 사이토카인(TNF-α, IL-6)의 분비량을 ELISA kit (R&D systems Inc., Minneapolis, MN, USA)로 측정하였다. 100% 활성은 PMA와 A23187이 처리되지 않은 군에서 처리된 군 사이의 사이토카인 분비량의 차이로 계산하였다.
사이토카인 분비량(%)은 식 (1)에 적용하여 계산하였다.
식 (1): 사이토카인 분비량(%)= [(시료 처리군 사이토카인 분비량 - 무처리군 사이토카인 분비량) / (대조군 사이토카인 분비량 - 무처리군 사이토카인 분비량)] × 100.
모든 측정 결과는 평균(mean)과 표준편차(standard deviation; SD)로 나타내었으며, 실험군 간의 차이는 Student's t-test를 사용하여 통계학적 분석을 수행하였고, p < 0.05 값인 경우에 통계적으로 유의성이 있는 것으로 판정하였다.
그 결과, 도 4에 개시한 바와 같이 MC/9 마우스 비만세포에는 오미자 열매 추출물 및 오미자 잎 추출물이 세포독성이 없다는 것을 확인하였으며, 오미자 열매 추출물 및 오미자 잎 추출물 처리에 의해 사이토카인 분비량이 현저하게 감소하였고, 특히, 오미자 열매 추출물에 비해 오미자 잎 추출물에서의 사이토카인 분비량이 더 감소하였다(도 5).

(1) 세포 배양
인간 각질세포(human keratinocyte) HaCaT 세포를 10% heat-inactivated Fetal Bovine Serum(FBS), 100units/㎖ 페니실린(penicillin), 100㎍/㎖의 스트렙토마이신(streptomycin)을 첨가한 DMEM 배지로 37℃, 5% CO2 환경에서 배양하였다.
(2) 세포 독성 실험
추출물의 세포 독성을 확인하고자 Cell Counting Kit-8(CCK-8) assay를 실시하였다. 96웰 플레이트(well plate)에서 HaCaT 세포에 시료를 농도별 (25, 50, 100, 200㎍/㎖)로 전처리한 다음, TNF-α 10ng/㎖과 IFN-γ 10ng/㎖의 농도로 처리하고, 24시간 동안 배양하였다. 24시간 후 배지에 CCK-8을 10% 농도로 첨가한 다음 2시간 동안 37℃에서 반응시킨 후, 마이크로플레이트 스펙트로포토미터(microplate spectrophotometer)(Biorad, Hercules, CA, USA)를 이용하여 450nm에서 흡광도를 측정하고, 대조군과의 비교를 통해 상대적인 세포 생존율(%)을 계산하였다.

배양된 keratinocyte 인간 각질 세포를 6웰 플레이트(well plate)에 접종하여 시료를 농도별 (25, 50, 100, 200㎍/㎖)로 전처리한 다음, TNF-α 10ng/㎖과 IFN-γ 10 ng/㎖의 농도로 처리하고, 24시간 동안 배양하였다. 양성대조군으로 0.1 및 1μM의 덱사메타손(Dexamethasone)과 12 및 25㎍/㎖의 실리마린(silymarin)을 각각 처리하였다. 24시간 후에 배지에 분비된 케모카인(TARC, RANTES)의 분비량을 ELISA 키트(R&D systems Inc., Minneapolis, MN, USA)로 측정하였다. 100% 활성은 TNF-α와 IFN-γ(TI)이 처리되지 않은 군에서 처리된 군 사이의 케모카인 분비량의 차이로 계산하였다.
케모카인 분비량(%)은 식 (2)에 적용하여 계산하였다.
식 (2): 케모카인 분비량(%)=[(시료 처리군 케모카인 분비량 - 무처리군 케모카인 분비량) / (대조군 케모카인 분비량 - 무처리군 케모카인 분비량)] × 100.
모든 측정 결과는 평균(mean)과 표준편차(standard deviation; SD)로 나타내었으며, 실험군 간의 차이는 Student's t-test를 사용하여 통계학적 분석을 수행하였고, p < 0.05 값인 경우에 통계적으로 유의성이 있는 것으로 판정하였다.
그 결과, 도 6에 개시한 바와 같이 인간 각질세포에는 오미자 열매 추출물 및 오미자 잎 추출물이 세포독성이 없다는 것을 확인하였으며, 오미자 열매 추출물 및 오미자 잎 추출물 처리에 의해 케모카인 분비량이 감소하였으며, 특히, 오미자 열매 추출물에 비해 오미자 잎 추출물에서의 케모카인 분비량이 통계적으로 유의하게 더 감소하였다(도 7).
Claims (8)
- 오미자 잎 추출물을 유효성분으로 포함하는 아토피 피부염의 예방 또는 치료용 약학 조성물.
- 제1항에 있어서, 상기 오미자 잎 추출물의 용매는 물, C1~C4의 저급 알코올 또는 이들의 혼합물인 것을 특징으로 하는 아토피 피부염의 예방 또는 치료용 약학 조성물.
- 제1항에 있어서, 상기 오미자 잎 추출물 이외에 추가로 담체, 부형제 또는 희석제를 더 포함하는 것을 특징으로 하는 아토피 피부염의 예방 또는 치료용 약학 조성물.
- 오미자 잎 추출물을 유효성분으로 포함하는 아토피 피부염의 예방 또는 개선용 건강기능식품 조성물.
- 제4항에 있어서, 상기 조성물은 분말, 과립, 환, 정제, 캡슐, 캔디, 시럽 및 음료 중에서 선택된 어느 하나의 제형으로 제조되는 것을 특징으로 하는 아토피 피부염의 예방 또는 개선용 건강기능식품 조성물.
- 오미자 잎 추출물을 유효성분으로 포함하는 아토피 피부염 개선용 화장료 조성물.
- 제6항에 있어서, 상기 조성물은 용액, 현탁액, 유탁액, 페이스트, 겔, 크림, 로션, 파우더, 비누, 계면활성제-함유 클렌징, 오일, 분말 파운데이션, 유탁액, 파운데이션, 왁스 파운데이션 및 스프레이 중에서 선택된 어느 하나의 제형으로 제조되는 것을 특징으로 하는 아토피 피부염 개선용 화장료 조성물.
- 오미자 잎 추출물을 유효성분으로 포함하는 아토피 피부염 개선용 의약외품 조성물.
