Description
Foreliggende oppfinnelse angår dimere forbindelser, slike forbindelser som farmasøytiske midler, farmasøytiske preparater inneholdende slike, anvendelse av slike for fremstilling av medikamenter for behandling av sykdom samt en in vitro metode for påvisning av influensavirus.
BAKGRUNN FOR OPPFINNELSEN
Enzymer med evnen til å spalte N-acetyl-neuraminsyre (NANA), også kjent som sialinsyre, fra andre karbohydrater er til stede i mange mikroorganismer. Disse omfatter bakterier så som Vibrio cholerae, Clostridium perfringens, Streptococcus pneumoniae og Arthrobacter sialophilus og virus så som influensavirus, parainfluensavirus, kusmavirus, Newcastle sykdomsvirus og Sendai-virus. Mesteparten av disse virusene er av ortomyksovirus- eller paramyksovirus-grupper og bærer neuraminidase-aktivitet på overflaten av viruspartikkelen. Mange av disse neuraminidase-inneholdende organismer er viktige patogener for mennesker og/eller dyr og noen, så som influensavirus og Newcastle sykdomsvirus, forårsaker sykdommer av enorm betydning.
Det har lenge vært antatt at inhibitorer av neuraminidase kunne forhindre infeksjon av neuraminidase-bærende virus. Mesteparten av de kjente neuraminidase-inhibitorer er analoger av neuraminsyre, så som 2-deoksy-2,3-dehydro-N-acetylneuraminsyre (DANA) og noen derivater derav (Meindl et al, Virology, 1974 58 457). Vår internasjonale patentpublikasjon nr. WO 91/16320 beskriver flere analoger av DANA som er aktive mot viral neuraminidase og det har vært vist spesielt at 4-guanidino-2-deoksy-2,3-dehydro-N-acetylneuraminsyre (Forbindelse (A), kodenummer GG167) er anvendelig ved behandling av influensa A og B (N. Engl. J. Med., 1997 337 874-880). Andre patentsøknader beskriver forskjellige nærbeslektede sialinsyrederivater (f.eks. PCT-publikasjoner nr. WO 95/18800,
nr. WO 95/20583 og nr. WO 98/06712) og anti-virale makromolekylære konjugater av GG167 er også beskrevet (internasjonal patentsøknad nr. PCT/AU97/00771).

I tillegg til de sialinsyrebaserte inhibitorer nevnt ovenfor, er andre typer av meget aktive inhibitorer av influensavirus neuraminidase også beskrevet, spesielt de basert på 5- og 6-leddede karbocykliske ringsystemer (f.eks. internasjonale patentpublikasjoner nr. WO 96/26933 og nr. 97/47194).
Nylig har internasjonal patentpublikasjon nr. WO 97/06157, nr. WO
98/06712 og europeisk patentsøknad nr. 0823428 beskrevet visse derivater av forbindelse (A) hvor den normale sialinsyre 7-hydroksygruppe blir erstattet med forskjellige andre funksjonaliteter, som hemmer multiplisering av influensavirus.
Vi har nå overraskende funnet at når to neuraminidase-bindende forbindelser er hensiktsmessig bundet sammen gjennom en region av molekylet som ikke er involvert i binding til det aktive sete, viser de resulterende dimerer fremragende anti-viral aktivitet. Spesielt har vi funnet at, selv om en ekstra substituent bundet til forbindelse (A) i 7-stilling generelt forårsaker en liten reduksjon i anti-influensaaktivitet, når to slike 7-substituerte molekyler av forbindelse (A) begge er bundet til en egnet spacer-gruppe, kan anti-influensaaktiviteten være sterkt forbedret. Selv om vi ikke ønsker å være bundet eller begrenset av noen foreslått mekanisme for den observerte effekt, antas det at de dimere forbindelser ifølge foreliggende oppfinnelse har forbedret anti-influensaaktivitet fordi de kan binde til to separate neuraminidase-molekyler og derved forårsake aggregering av neuraminidase-tetramerer og/eller influensavirionene.
OPPSUMMERING AV OPPFINNELSEN
I et første aspekt tilveiebringer oppfinnelsen en forbindelse med den generelle formel I
hvor
R er en azidogruppe, en hydroksygruppe, en usubstituert eller substituert guanidinogruppe eller en usubstituert eller substituert aminogruppe;
R<2> representerer COCH3l COCF3, S02CH3 eller S02CF3;
X representerer O, 0(C=0), NH, NHCO, 0(C=0)NH, 0(C=S)NH, NH(C=0)NH eller NH(C=S)NH;
og spacergruppen Y er en eventuelt substituert og/eller forgrenet gruppe opptil 100 atomer lang, med ryggradsatomer valgt fra gruppen bestående av karbon, nitrogen, oksygen og svovel;
eller et farmasøytisk akseptabelt derivat eller salt derav.
Fortrinnsvis er spacer-gruppen 8 til 100, mer foretrukket 10 til 50, enda mer foretrukket 12 til 30 atomer lang.
Mest foretrukket er at:
R er en usubstituert eller substituert guanidinogruppe, eller en usubstituert eller substituert aminogruppe;
R2 er acetyl eller trifluoracetyl;
X er 0(C=0)NH; og
spacergruppe Y er mellom 10 og 50 atomer lang.
Molekylære modellundersøkelser indikerer at spacer-gruppen kunne være så kort som 18,15 eller 10 atomer lang eller til og med være kortere enn dette. Fagfolk på området vil lett være i stand til å optimalisere spacer-lengden ved rutinemessige prøve og feile eksperimentering.
Generelt menes at når en variabel forekommer to ganger i formel (I), kan variabelen være lik eller forskjellig.
Når R er en amino- eller guanidinogruppe omfatter egnede substituenter alkyl, hydroksyalkyl, allyl, nitril, alkoksykarbonyl og acyl.
Egnede spacergrupper Y omfatter eventuelt substituerte lineære eller forgrenede hydrokarbonkjeder, peptider, oligosakkarider, polyaminosyrer, polyetylenglykolenheter, alkylamidoalkaner, alkylureidoalkaner, som alle kan anvendes alene, i multippel form eller i kombinasjon. Spacergruppen Y kan også eventuelt ha bundet til den en ekstra funksjonalitet for å forbedre de farmasøytiske eller farmakokinetiske egenskaper av forbindelsen. Slike funksjonaliteter omfatter lipofile hydrokarbongrupper, polyetylenglykol- (PEG) kjeder og peptider.
For formålene i denne beskrivelse skal betegnelsene "hydrokarbon", "alkan" eller "alkyl" omfatte mettede, umettede og cykliske hydrokarbongrupper, aromatiske ringer og kombinasjoner av slike grupper. Egnede substituenter på hydrokarbonkjeder omfatter Br, Cl, F, I, CF3, NH2, substituerte aminogrupper så som NH-acyl og alkoksygrupper så som metoksy og hydroksy og er fortrinnsvis F, Cl, hydroksy, alkoksy, amino, alkylamino eller karboksy.
Det vil forstås av fagfolk på området at forbindelsene med formel (I) kan modifiseres for å gi farmasøytisk akseptable derivater derav ved hvilken som helst av de funksjonelle gruppene i forbindelsene med formel (I). Av spesiell interesse som sådanne derivater er forbindelser modifisert i karboksylfunksjonen, hydroksylfunksjonene eller ved aminogrupper. Således omfatter forbindelser av interesse alkylestere, så som metyl-, etyl-, propyl- eller isopropylestere, arylestere, så som fenyl- og benzoyl-estere og acetylestere av forbindelsene med formel (I).
Det vil forstås av fagfolk på området at de farmasøytisk akseptable derivater av forbindelsene med formel (I) kan være derivatisert i mer enn én stilling.
Betegnelsen "farmasøytisk akseptabelt derivat" betyr hvilket som helst farmasøytisk akseptabelt salt, ester eller salt av slik ester av en forbindelse med formel (I) eller hvilken som helst annen forbindelse som, ved administrering til mottageren, kan gi en forbindelse med formel (I) eller en antiviralt aktiv metabolitt eller rest derav. Av spesiell interesse som derivater er forbindelser modifisert i sialinsyrens karboksy- eller glycerol-hydroksygrupper eller i amino- og guanidin-grupper.
Farmasøytisk akseptable salter av forbindelsene med formel (I) omfatter de avledet fra farmasøytisk akseptable uorganiske og organiske syrer og baser. Eksempler på egnede syrer omfatter saltsyre, bromhydrogensyre, svovelsyre, salpetersyre, perklorsyre, fumarsyre, maleinsyre, fosforsyre, glykolsyre, melkesyre, salicylsyre, ravsyre, toluen-p-sulfonsyre, vinsyre, eddiksyre, sitronsyre, metansulfonsyre, maursyre, benzosyre, malonsyre, naftalen-2-sulfonsyre og benzensulfonsyrer. Andre syrer så som oksalsyre, som ikke selv er farmasøytisk akseptable, kan være anvendelige ved fremstilling av salter anvendelige som mellomprodukter for å oppnå forbindelser ifølge foreliggende oppfinnelse og deres farmasøytisk akseptable syreaddisjonssalter.
Salter avledet fra passende baser omfatter alkalimetall- (f.eks. natrium), jordalkalimetall- (f.eks. magnesium), ammonium- og NR4<+-> (hvor R er Ci-4alkyl) salter.
Forbindelsene med formel (I) har antiviral aktivitet. Spesielt er disse forbindelser inhibitorer av viral neuraminidase i ortomyksovirus og paramyksovirus, for eksempel den virale neuraminidase i influensa A og B, parainfluensa, kusma og Newcastle sykdom.
I et andre aspekt tilveiebringer oppfinnelsen således en forbindelse ifølge foreliggende oppfinnelse, fortrinnsvis en forbindelse med formel (I) eller et farmasøytisk akseptabelt derivat derav, for anvendelse som et aktivt terapeutisk middel ved behandling av ortomyksovirus- og paramyksovirus-infeksjoner.
I et tredje aspekt tilveiebringer oppfinnelsen anvendelse av en forbindelse med formel (I) eller et farmasøytisk akseptabelt salt eller derivat derav, for fremstilling av medikament for behandling av en viral infeksjon, for eksempel ortomyksovirus- og paramyksovirus-infeksjoner hos et pattedyr.
I en foretrukket utførelsesform av dette aspekt ved foreliggende oppfinnelse tilveiebringes en anvendelse for behandling av influensa A eller B hos et pattedyr.
Fortrinnsvis er pattedyret et menneske.
Det vil forstås av fagfolk på området at referanse her til behandling omfatter forebygging mot infeksjon så vel som behandling av etablerte infeksjoner eller symptomer.
Forbindelsene ifølge foreliggende oppfinnelse kan også anvendes ved diagnostiske metoder in vitro, spesielt metoder for deteksjon av influensavirus. For anvendelse ved slike metoder kan det være fordelaktig å binde en forbindelse ifølge foreliggende oppfinnelse til et merke.
Det vil videre forstås at mengden av en forbindelse ifølge foreliggende oppfinnelse som er nødvendig for anvendelse ved behandling vil variere ikke bare med den spesielle forbindelse som velges, men også med administreringsveien, typen av lidelsen som behandles og alderen og tilstanden til pasienten og vil i siste instans bestemmes av behandlende lege eller veterinær. Generelt vil imidlertid en egnet dose være i området fra ca. 0,01 til 100 mg/kg kroppsvekt pr. dag, fortrinnsvis i området 0,05 til 10 mg/kg/dag, mest foretrukket i området 0,1 til 1 mg/kg/dag.
Behandling blir fortrinnsvis startet før eller ved begynnelsen av infeksjon og fortsatt inntil virus ikke lenger er til stede i luftveiene. Imidlertid er forbindelsene også effektive når de blir gitt etter infeksjon, for eksempel etter fremkomst av etablerte symptomer.
Hensiktsmessig blir behandling gitt 1-4 ganger daglig og fortsatt i 3-7 dager etter infeksjon, f.eks. 5 dager, avhengig av den spesielle forbindelsen som anvendes.
Den ønskede dose kan presenteres i en enkel dose eller som oppdelte doser administrert med passende mellomrom, for eksempel som to, tre, fire eller flere underdoser pr. dag.
Forbindelsen blir hensiktsmessig administrert i enhetsdoseform, for eksempel inneholdende 1 til 1000 mg, hensiktsmessig 2 til 200 mg, mest hensiktsmessig 50 til 100 mg aktiv bestanddel pr. enhetsdoseform.
Mens det er mulig at, for anvendelse i terapi, en forbindelse ifølge foreliggende oppfinnelse kan administreres som det rå kjemikalium, er det foretrukket å presentere den aktive bestanddel som et farmasøytisk preparat.
I et fjerde aspekt tilveiebringer oppfinnelsen således et farmasøytisk preparat omfattende en forbindelse med formel (I) eller et farmasøytisk akseptabelt salt eller derivat derav, sammen med én eller flere farmasøytisk akseptable bærere og eventuelt andre terapeutiske og/eller profylaktiske bestanddeler. Bæreren(e) må være "akseptable" i den forstand at de er kompatible med de andre bestanddelene i preparatet og ikke skadelig for mottageren derav.
Forbindelsene ifølge foreliggende oppfinnelse kan også anvendes i kombinasjon med andre terapeutiske midler, for eksempel andre anti-infeksiøse midler. Spesielt kan forbindelsene ifølge foreliggende oppfinnelse anvendes med andre antivirale midler. Oppfinnelsen tilveiebringer således i et femte aspekt en kombinasjon omfattende en forbindelse med formel (I) eller et farmasøytisk akseptabelt salt eller derivat derav sammen med et annet terapeutisk aktivt middel, spesielt et antiviralt middel.
Kombinasjonene referert til ovenfor kan hensiktsmessig presenteres for anvendelse i form av et farmasøytisk preparat og således utgjør slike preparater omfattende en kombinasjon som definert ovenfor sammen med en farmasøytisk akseptabel bærer derfor et ytterligere aspekt ved foreliggende oppfinnelse.
Egnede terapeutiske midler for anvendelse i slike kombinasjoner omfatter andre antiinfeksiøse midler, spesielt anti-bakterielle og antivirale midler så som de anvendt for å behandle respiratoriske infeksjoner. For eksempel kan andre forbindelser effektive mot influensavirus, så som sialinsyreanalogene referert til ovenfor, amantadin, rimantadin og ribavirin, inkluderes i slike kombinasjoner.
De individuelle komponenter i slike kombinasjoner kan administreres enten sekvensielt eller samtidig i separate eller kombinerte farmasøytiske preparater.
Når forbindelsene ifølge foreliggende oppfinnelse blir anvendt med et annet terapeutisk middel aktivt mot samme virus, kan dosen av hver forbindelse enten være den samme som eller forskjellig fra den anvendt når hver forbindelse blir anvendt alene. Passende doser vil lett forstås av fagfolk på området.
Farmasøytiske preparater omfatter de egnet for oral, rektal, nasal, topisk (omfattende buckal og sublingual), vaginal eller parenteral (omfattende intramuskulær, subkutan og intravenøs) administrering eller de i en form egnet for administrering til luftveiene (omfattende nasal-passasjene) for eksempel ved inhalering eller insufflasjon. Preparatene kan, når det passer, hensiktsmessig presenteres i adskilte doseenheter og kan fremstilles ved hvilken som helst av metodene velkjent på området farmasi. Disse metoder omfatter trinnet å bringe den aktive forbindelse i kontakt med flytende bærere eller findelte faste bærere eller begge deler og deretter, om nødvendig, forme produktet til det ønskede preparat.
Farmasøytiske preparater egnet for oral administrering kan hensiktsmessig presenteres som adskilte enheter så som kapsler, pulverkapsler eller tabletter hver inneholdende en forutbestemt mengde av den aktive bestanddel; som et pulver eller granuler; som en løsning, en suspensjon eller som en emulsjon. Den aktive bestanddel kan også presenteres som bolus, latverge eller pasta. Tabletter og kapsler for oral administrering kan inneholde konvensjonelle tilsetningsmidler så som bindemidler, fyllmidler, smøremidler, desintegreringsmidler eller fuktemidler. Tablettene kan belegges i henhold til metoder velkjent på området. Orale flytende preparater kan for eksempel være i form av vandige eller oljeaktige suspensjoner, løsninger, emulsjoner, siruper eller eliksirer eller kan presenteres som et tørt produkt for konstituering med vann eller annen egnet konstituent før anvendelse. Slike flytende preparater kan inneholde konvensjonelle additiver så som suspenderingsmidler, emulgeringsmidler, ikke-vandige konstituenter, som kan omfatte spiselige oljer eller konserveringsmidler.
Forbindelsene ifølge oppfinnelsen kan også formuleres for parenteral administrering ved injeksjon, for eksempel bolus-injeksjon eller kontinuerlig infusjon og kan presenteres i enhetsdoseform i ampuller, forhåndsfylte sprøyter, små volum infusjons- eller multi-dose-beholdere med tilsatt konserveringsmiddel. Preparatene kan være i form av suspensjoner, løsninger eller emulsjoner i oljeaktige eller vandige konstituenter og kan inneholde formuleringsmidler så som suspenderings-, stabiliserings- og/eller dispergeringsmidler. Alternativt kan den aktive bestanddel være i pulverform, oppnådd ved aseptisk isolering av sterilt fast stoff eller ved lyofilisering fra løsning, for konstituering med en egnet konstituent, f.eks. sterilt, pyrogenfritt vann, før anvendelse.
For topisk administrering til epidermis kan forbindelsene ifølge oppfinnelsen formuleres som salver, kremer eller losjoner eller som et transdermalt plaster. Salver og kremer kan for eksempel formuleres med en vandig eller oljeaktig base med tilsetning av egnede fortyknings- og/eller gelleringsmidler. Losjoner kan formuleres med en vandig eller oljeaktig base og vil generelt også inneholde ett eller flere emulgeringsmidler, stabiliseringsmidler, dispergeringsmidler, suspenderingsmidler, fortykningsmidler eller fargemidler.
Preparater egnet for topisk administrering i munnen omfatter pastiller omfattende den aktive bestanddel i en smakstilsatt base, vanligvis sukrose og gummi akasie eller gummi tragant; pastiller omfattende den aktive bestanddel i en inert base så som gelatin eller sukrose og gummi akasie; og munnvann omfattende den aktive bestanddel i en egnet flytende bærer.
Farmasøytiske preparater egnet for rektal administrering hvor bæreren er et fast stoff er mest foretrukket presentert som enhetsdose-suppositorier. Egnede bærere omfatter kakaosmør og andre materialer vanlig anvendt på området og suppositoriene kan hensiktsmessig dannes ved blanding av den aktive forbindelse med myknet eller smeltet bærer(e) fulgt av avkjøling og forming.
Preparater egnet for vaginal administrering kan presenteres som pessarer, tamponger, kremer, geler, pastaer, skum eller spray-preparater inneholdende i tillegg til den aktive bestanddel slike bærere som er kjent på området som egnet.
For administrering til luftveiene, omfattende intranasal administrering, kan neuraminidase-inhibitorer administreres ved hvilken som helst av metodene og preparatene anvendt på området for administrering til luftveiene.
Således kan forbindelsene generelt administreres i form av en løsning eller en suspensjon eller som et tørt pulver.
Løsninger og suspensjoner vil generelt være vandige, for eksempel fremstilt fra vann alene (for eksempel sterilt eller pyrogenfritt vann) eller vann og et fysiologisk akseptabelt medløsningsmiddel (for eksempel etanol, propylenglykol eller polyetylenglykoler så som PEG 400).
Slike løsninger eller suspensjoner kan i tillegg inneholde andre tilsetningsmidler, for eksempel konserveringsmidler (så som benzalkoniumklorid), solubiliseringsmidler/overflateaktive midler så som polysorbater (f.eks. Tween 80, Span 80, benzalkoniumklorid), buffermidler, isotonisitetsregulerende midler (for eksempel natriumklorid), absorpsjonsforbedrere og viskositetsforbedrere. Suspensjoner kan i tillegg inneholde suspenderingsmidler (for eksempel mikrokrystallinsk cellulose, karboksymetylcellulose-natrium).
Løsninger eller suspensjoner blir påført direkte tii neselhulen ved konvensjonelle metoder, for eksempel med en dråpeteller, pipette eller spray. Preparatene kan gis i enkel eller multidoseform. I det sistnevnte tilfelle er midler for doseoppmåling fortrinnsvis inkludert. I tilfellet med en dråpeteller eller pipette kan dette oppnås ved at pasienten administrerer et passende, forutbestemt volum av løsningen eller suspensjonen. I tilfellet med en spray kan dette oppnås for eksempel ved hjelp av en forstøvnings-spraypumpe med oppmåling.
Administrering til luftveiene kan også oppnås ved hjelp av et aerosolpreparat hvor forbindelsen er gitt i en pakning under trykk med et egnet drivmiddel, så som klorfluorkarbon (CFC), for eksempel diklordifluormetan, triklorfluormetan eller diklortetrafluoretan, karbondioksyd eller annen egnet gass. Aerosolen kan hensiktsmessig også inneholde et overflateaktivt middel så som lecitin. Dosen av medikament kan kontrolleres ved anvendelse av en oppmålingsventil.
Alternativt kan forbindelsene gis i form av et tørt pulver, for eksempel en pulverblanding av forbindelsen i en egnet pulverbase så som laktose, stivelse, stivelsesderivater så som hydroksypropylmetylcellulose og po lyvi nyl py rro I id i n (PVP). Hensiktsmessig vil pulverbæreren danne en gel i nesehulen. Pulverpreparatet kan presenteres i enhetsdoseform, for eksempel i kapsler eller patroner av f.eks. gelatin eller "blister"-pakninger fra hvilke pulveret kan administreres ved hjelp av en inhalator.
I preparater ment for administrering til luftveiene, omfattende intranasale preparater, vil forbindelsen generelt ha en liten partikkelstørrelse, for eksempel i størrelsesorden 5 mikron eller mindre. En slik partikkelstørrelse kan oppnås ved metoder kjent på området, for eksempel ved mikronisering.
Når det er ønsket kan preparater tilpasset for å gi forsinket frigjøring av den aktive bestanddel, anvendes.
For denne beskrivelsen skal det klart forstås at ordet "omfattende" betyr "omfattende, men ikke begrenset til" og at ordet "omfatter" har en tilsvarende betydning.
DETALJERT BESKRIVELSE AV OPPFINNELSEN

Spesielle eksempler på forbindelser ifølge foreliggende oppfinnelse omfatter de med formel (la), hvor spacer-gruppen Y er som vist i Tabell 1 nedenfor.
Forbindelsene ifølge foreliggende oppfinnelse kan fremstilles ved metodene beskrevet nedenfor, hvor R og R<2> er som definert for formel (I).
Egnede monomere mellomproduktforbindelser med den generelle formel (II) kan fremstilles ved å følge metodene beskrevet i internasjonale patentpublikasjoner nr. WO 97/06157 og nr. WO 97/32214. Således, hvis gruppen i stilling 7 er et arylkarbonat (f.eks. Z = 4-nitrofenoksy), kan mellomproduktet anvendes for å fremstille 7-karbamat-derivater (Z = alkyl-NH) ved omsetning med forskjellige aminer (alkyl-NH2).

Til en løsning av 5-acetamido-7-(6'-aminoheksyl)-karbamoyloksy-4-guanidino-2,3,4,5-tetradeoksy-D-glycero-D-galakto-non-2-enopyranosonsyre (7) (50 mg, 0,1055 mmol) i en blanding av DMSO (1 ml) og pyridin (2,5 ml) ble satt 1,4-diisocyanatobutan (7,39 mg, 0,0527 mmol) og 4-dimetylaminopyridin (12,87 mg, 0,1055 mmol). Hele blandingen ble omrørt under argon ved 50°C i 7 dager. Blandingen ble filtrert og filtratet ble inndampet under høyvakuum til tørrhet. Residuet ble omrørt i aceton (2 x 20 ml) ved romtemperatur i 24 timer og filtrert. Filterkaken ble vasket med aceton (5 ml) og omkrystallisert fra en blanding av metanol og vann (7:3), hvilket ga tittelforbindelsen (2) som et hvitt, faststoff (18,6 mg, 32%).

Til en løsning av 5-acetamido-7-(6'-aminoheksyl)-karbamoyloksy-4-guanidino-2,3,4,5-tetradeoksy-D-glycero-D-galakto-non-2-enopyranosonsyre (7) (50 mg, 0,1055 mmol) i en blanding av DMSO (1 ml) og pyridin (2,5 ml) ble satt 4-dimetylaminopyridin (12,87 mg, 0,1055 mmol) og 1,12-diisocyanatododekan (13,31 mg, 0,0527 mmol). Hele blandingen ble omrørt under argon ved 50°C i 7 dager og deretter filtrert. Filtratet ble inndampet under høyvakuum til tørrhet. På lignende måte som den beskrevet i Eksempler 1 og 2, ble forbindelser (4), (5) og (6) hver fremstilt ved anvendelse av 6-aminoheksyl-7-karbamat-forbindelsen (7) eller beskyttede former av (7), som nøkkel-utgangsmaterialet. Hver forbindelse ble karakterisert ved dens massespektrum og Nmr-data.

Til en løsning av 5-acetamido-7-(6-aminoheksyl)-karbamoyloksy-4-guanidino-2,3,4,5-tetradeoksy-D-glycero-D-galakto-non-2-enopyranosonsyre (7) (76 mg, 0,16 mmol) i en blanding av DMF (7,5 ml) og pyridin (2,5 ml) ble satt etylendiamintetraeddiksyre-dianhydrid (20,5 mg, 0,08 mmol) og 4-(dimetylamino)pyridin (3,5 mg, 0,028 mmol). Hele blandingen ble omrørt ved 50°C i 18 timer, og deretter inndampet til tørrhet under høyvakuum. Residuet ble fordelt mellom diklormetan (20 ml) og vann (10 ml). Fremstilling av D- glutam- v- vl- a- vlaminacetvl. di- f7-( 6 - aminoheksvl)-karbamovloksv- 5- acetamido- 4- guanidino- 2. 3. 4. 5- tetradeoksv- D- glvcero- D-galakto- non- 2- enopyranosonsvrel som trifluoreddiksvresalt ( Forbindelse nummer ( 9))
N-Boc-D-glutam-a-ylamineddiksyre (25 mg, 0,082 mmol) ble oppløst i vann (0,25 ml) inneholdende trietylamin (16,6 mg, 0,164 mmol) og N-metylmorfolin (16,6 mg, 0,164 mmol). Den klare løsningen ble fortynnet med aceton (3 ml) og deretter avkjølt til -20°C i et tørris-aceton-bad. Til denne løsningen ble satt isobutylklorformiat (26,95 mg, 0,197 mmol). Reaksjonsblandingen ble omrørt ved -15° ± 2°C i 12 min. og deretter kombinert med en løsning av 5-acetamido-7-(6'-aminoheksyl)karbamoyloksy-4-guanidino- 2,3,4,5-tetradeoksy-D-glycero-D-galakto-non-2-enopyranosonsyre (7) (116,9 mg, 0,246 mmol) og trietylamin (24,9 mg, 0,246 mmol) i vann (2,5 ml). Den resulterende blandingen fikk omrøres ved romtemperatur i 3 timer, og ble deretter inndampet til tørrhet under redusert trykk. Residuet ble underkastet "flash" kolonne-kromatografi (silikagel, 2-propanol:eddiksyre:vann = 3:1:1 som elueringsmiddel), hvilket ga N-Boc-derivatet av tittelforbindelsen (9), som deretter ble behandlet med trifluoreddiksyre (2 ml) ved romtemperatur i 1 time og inndampet under vakuum til tørrhet. Residuet ble frysetørket, hvilket ga tittelforbindelsen (9) som trifluoreddiksyresalt (31 mg, 30,4 %).

Eksempel 6
Hemning av influensavirus- replikasion med forbindelser ifølge oppfinnelsen
Forbindelser ifølge foreliggende oppfinnelse ble testet for deres evne til å hemme replikasjonen av influensa A virus, ved å følge standardmetoden som er beskrevet i litteraturen (se for eksempel Watanabe et al, J. Virological Methods, 1994 48 257). Forsøket ble utført ved anvendelse av MDCK-celler og resultatene er vist i Tabell 2 nedenfor. Resultatene er vist som ID50, den minimum forbindelseskonsentrasjon som hemmer cytopatisk effekt med 50%
[(|ig/ml)], beregnet ved anvendelse av et regresjonsanalyseprogram for semi-log kurvetilpasning. Resultatene viser at dimere forbindelser (2), (3) og (4) alle er mer aktive mot influensa enn det monomere ligandmolekyl (7) og at forbindelse (2) ifølge foreliggende oppfinnelse er enda kraftigere enn den meget aktive forbindelse (A) [GG167]. Den terapeutiske indeksen for forbindelsene kan beregnes ved å dividere minimum cytotoksisk medikamentkonsentrasjon (MTC) med ID50.
Eksempel 7
Hemning av influensavirus- replikasion med forbindelser ifølge oppfinnelsen
Forbindelser ifølge oppfinnelsen ble testet for deres evne til å hemme replikasjonen av influensa A/Victoria/3/75 B010 i et standard CPE type forsøk lignende det beskrevet ovenfor i Eksempel 6. Resultatene for tre separate forsøk er vist i Tabell 3 nedenfor.
Det vil være klart for fagfolk på området at mens oppfinnelsen er
