Description
Foreliggende oppfinnelse vedrører anvendelse av en ringsystemforbindelse for fremstilling av et medikament for å kontrollere østrogenproduksjon og/eller for å targetrette det østrogen-metabolske spor.
Steroid-forstadier eller pro-hormoner som har en sulfatgruppe
i 3-stillingen av steroidkjernen, heretter simpelthen omtalt som steroidsulfater, er kjent til å spille en viktig rolle som mellomprodukter i steroidmetabolisme i menneskekroppen, østronsulfat og dehydroepiandrosteron {DHA) sulfat er for eksempel kjent til å spille en viktig rolle som mellomprodukter i kroppens produksjon av østrogener som østron og østradiol. Østronsulfat er for eksempel særlig kjent til å representere en av de viktigste sirkulerende østrogen-forstadier særlig hos kvinner etter overgangsalder og østronsulfatase-aktivitet i brysttumorer er fra 100-1000 ganger større enn aktiviteten til andre enzymer involvert i østrogendannelse (James et al, Steroids, 50, 269-279 (1987)).
Østrogener som østron og østradiol og særlig overproduksjonen av disse er sterkt implisert i maligne tilstander som brystcancer, se Breast Cancer, Treatment and Prognosis: Ed. R.A. Stoll, s. 156-172, Blackwell Scientific Publications (1986), og kontroll av østrogenproduksjon er det spesifikke mål i en rekke anti-cancer-terapier, både kjemoterapi og operativ terapi, f.eks. ooforektomi og adrenalektomi. Når det gjelder endokrin terapi har forsøk til nå hatt en tilbøyelighet til å konsentrere seg om aromataseinhibitorer, dvs. forbindelser som inhiberer aromataseaktivitet, hvis aktivitet er involvert i omdannelsen av androgener som androstendion og testosteron til henholdsvis østron og østradiol som det vedlagte flytskjema for østrogenmetabolisme viser (fig. l).
I den nylig publiserte internasjonale patentsøknad WO91/13083 er det blitt foreslått å rette søkelyset mot et annet punkt i det østrogen-metabolske spor, eller heller to andre punkt, det vil si omdannelsen av DHA-sulfat og østronsulfat til henholdsvis DHA og østron ved steroid sulfatase-aktivitet og anvendelse av 3-monoalkyltiofosfonatsteroidestere som en steroid sulfatase-inhibitor, mer spesielt østron-3-monometyltiofos-f onat.
Foreliggende oppfinnelse vedrører anvendelse av en ringsystemforbindelse for fremstilling- av et medikament for å kontrollere østrogenproduksjon, hvor ringsystemforbindelsen omfatter en ring og en sulfamatgruppe med formelen' -
hvor hver av RL og R2 uavhengig er valgt fra H, alkyl, alkenyl, cykloaikyl og aryl, eller danner sammen alkylen som eventuelt inneholder ett eller flere heteroatomer eller grupper i alkylehkjeden; ■ hvor nevnte forbindelse er en inhibitor av et enzym med steroid sulfatase-aktivitet (EC 3.1.6.2.), og hvor, dersom sulfamatgruppen av nevnte forbindelse erstattes med en sulfatgruppe for å danne en sulfatforbindeIse og inkuberes med et steroid sulf atase-enzym (EC 3.1.6.2.) ved en pH 7,4 og 37°C, tilveiebringes en K,,, verdi på mindre enn 50 /iM. Oppfinnelsen vedrører også anvendelse av en ringsystemforbindelse for fremstilling av et medikament for å targetrette det østrogen-metabolske spor, hvor ringsystemf orbind-elsen omfatter en ring og en sulfamatgruppe med formelen
hvor hver av Rx og R2 uavhengig er valgt fra H, alkyl, alkenyl, cykloaikyl og aryl, eller danner sammen alkylen som eventuelt inneholder ett eller flere heteroatomer eller grupper i alkylenkjeden;
hvor nevnte forbindelse er en inhibitor av et enzym med steroid sulfatase-aktivitet (EC 3.1.6.2.), og hvor, dersom sulfamatgruppen av nevnte forbindelse erstattes med en
sulfatgruppe for å danne en sulfatforbindelse og inkuberes med et steroid sulfatase-enzym {EC 3.1.6.2.) ved en pH 7,4 og 3 7°C, tilveiebringes en K^, verdi på mindre enn 50 ( iK.
Ved et aspekt av oppfinnelsen muliggjøres behandling av østrogenavherigige tumorer i pattedyr, særlig mennesker,
samt behandling av brystcancer i pattedyr, særlig i kvinner.
Den foreliggende oppfinnelse vedrører således anvendelse av nye forbindelser som har forbedret steroid sulfataseinhiberende aktivitet både in vitro og in vivo og i enkelte tilfeller ekstremt høye aktivitetsnivåer., Disse forbindelser kan være sulfamsyreesterne av polycykliske alkoholer, som er polycykliske alkoholer hvor sulfatet derav er et substrat for enzymer med steroid sulfatase-aktivitet (EC 3.1.6.2), og N-alkyl-, N-cykloaikyl-, N-alkenyl- og N-aryl-derivatene av disse sulfamsyreesterne og. deres farmasøytisk tålbare salter.
Kort sagt kan forbindelsene anvendt i henhold til oppfinnelsen være dem med formel (I)
hvor R1 og R2 er som angitt i det foregående og gruppen -0-polysyklus er en rest av en polycyklisk alkohol hvor sulfatet av denne alkohol er et substrat for enzymer med steroid-sulfatase-aktivitet (EC 3.1.6.2.).
Som anvendt heri vil henvisningen til polycykliske alkoholer hvis sulfat er et substrat for enzymer med steroid sulfatase-aktivitet, referere til polycykliske alkoholer hvor sulfatet av disse, nemlig derivatene nied formelen:
ved inkubasjon med steroid sulfatase EC 3.1.6.2 ved pH 7,4 og 37°C, tilveiebringer en l^-verdi på mindre .-enn 50 fimol.
Kort beskrivelse av tegningene
Aktiviteten til. de foreliggende forbindelser som steroid sulfatase-inhibitorer er vist i de etterfølgende tegninger. Fig. l er et skjema som viser de metabolske spor, enzymer og steroid-mellomprodukter som er forbundet med dannelsen av østradiol in vivo. Fig. 2 er et histogram som viser den doseavhengige inhiberende virkning av østron-3-sulfamat på steroid sulfatase-aktivitet i humane MCF-7-celler in vitro. Fig. 3 er et histogram som viser den doseavhengige inhiberende virkning av østron-3-N,N-dimetylsulfamat på steroid sulfatase-aktivitet i humane MCF-7-celler in vitro. Fig. 4 er én kurve som sammenligner log dose-respons kurvene for østron-3-sulfamat og østron-3-N,N-dimetylsulfamat på steroid sulfatase-aktivitet i humane MCF-7-celler in vitro. Fig. 5 er en kurve som viser den doseavhengige inhiberende virkning av østron-3-sulfamat, sammen med dets IC50-verdi (konsentrasjon som kreves for å gi 50 % inhibering), på steroid sulfatase-aktivitet i humane placentamikrosomer in vitro.
Den ovennevnte polysyklus vil foretrukket inneholde, inklusive alle substituenter, maksimalt omtrent 4 0 karbonatomer, mer vanligvis høyst omtrent 30. Foretrukne polysykluser er dem som inneholder en steroidal ringstruktur, dvs. et cyklopentan-fenantrenskjelett. Sulfamylgruppene eller den substituerte sulfamylgruppe er foretrukket bundet til dette skjelett i 3-stilling, dvs. forbindelser med formel II:
hvor Rx og R2 er som angitt over og ringsystemet ABCD representerer en substituert eller usubstituert, mettet eller umettet steroidkjerne, foretrukket østron eller dehydroepiandrosteron.

Generelt kan steroidringsystemet ABCD inneholde en rekke ikke-interfererende substituenter. Spesielt kan ringsystemet ABCD inneholde en eller flere hydroksy, alkyl særlig lavere (Cj^-Cg) alkyl, f.eks. metyl, etyl, n-propyl, isopropyl, n-butyl, sec-butyl, tert-butyl, n-pentyl og andre pentylisomerer, og n-heksyl og andre heksylisomerer, alkoksy, særlig lavere (Cj^-Cg) alkoksy, f. eks. metoksy, etoksy, propoksy osv., alkinyl, f.eks. etinyl, eller halogen, f.eks. fluorsubsti-tuenter.
Passende ikke-steroidale ringsystemer inkluderer: dietylstilboestrol, stilboestrol og andre ringsystemer som gir sulfater med l^-verdier på mindre enn 50 //mol med steroid sulfatase EC 3.1.6.2.
Når de er substituerte, kan de N-substituerte forbindelser anvendt i henhold til oppfinnelsen inneholde en eller to N-alkyl-, N-alkenyl-, N-cykloalkyl- eller N-arylsubstituenter, og inneholder foretrukket eller inneholder hver foretrukket maksimalt 10 karbonatomer. Når Rx og/eller R2 er alkyl, er de foretrukket dem hvor Rx og R2 hver uavhengig er valgt fra lavere alkylgrupper inneholdende fra 1 til 5 karbonatomer, dvs. metyl, etyl, propyl osv. Rx og R2 er foretrukket begge metyl. Når Rx og/eller R2 er aryl, er typisk fenyl og tolyl (-PhCH3; o-, m- eller p-) . Når R± og R2 representerer cykloaikyl, er de typisk cyklopropyl, cyklopentyl, cykloheksyl osv. Når R-l og R2 er bundet sammen, vil de typisk representere en alkylengruppe med en kjede med 4 til 6 karbonatomer, som eventuelt er avbrutt av et eller flere heteroatomer eller -grupper, f.eks. -0- eller -NH- til å gi en 5-, 6- eller 7-leddet heterosyklus, f.eks. morfolino, pyrrolidino eller piperidino.
Innenfor betegnelsen alkyl, cykloaikyl, alkenyl og aryl inkluderes substituerte grupper inneholdende, som substituenter deri, en eller flere grupper som ikke interfererer med den sulfataseinhiberende aktivitet av den aktuelle forbindelse. Eksempler på ikke-interfererende substituenter inkluderer hydroksy, amino, halo, alkoksy, alkyl og aryl.
De mest foretrukne forbindelser er dem med formel III og IV:
hvor Rx og R2 er H eller C-L-Cg alkyl, dvs. østron-3-sulfamat og dehydroepiandrosteron-3-sulfamat og deres N- (C^-Cj) -alkylderi-vater, særlig dimetylderivatene, Rx = R2 = CH3. Sulfamsyreesterne fremstilles ved å reagere den polycykliske alkohol, f.eks. østron eller dehydroepiandrosteron, med et sulfamoylklorid R1R2NS02C1, dvs. reaksjonsskjemaet I
Betingelser for å gjennomføre reaksjonen ifølge reaksjons-skjerna I er som følger: Natriumhydrid og et sulfamoylklorid tilsettes til en omrørt løsning av østron i vannfri dimetylformamid ved 0°C. Deretter ble reaksjonsblandingen oppvarmet til romtemperatur og omrør-ingen fortsettes i ytterligere 24 timer. Reaksjonsblandingen helles inn i en kald mettet løsning av natriumbikarbonat og den oppnådde vandige fase ekstraheres med diklormetan. De kombinerte organiske ekstrakter tørkes over vannfri MgS04. Filtrering etterfulgt av løsningsmiddelavdamping i vakuum og samtidig avdamping med toluen gir en uren rest som ytterligere renses ved flash-kromatografi.
Om nødvendig kan funksjonelle grupper i den polycykliske alkohol (sterol) beskyttes på kjent måte og beskyttelses-gruppen eller -gruppene kan fjernes etter endt reaksjon.
For farmasøytisk tilførsel kan steroid sulfatase-inhibitorene utformes på enhver passende måte ved anvendelse av konven-sjonelle farmasøytiske teknikker og farmasøytiske bærere, eksipienser, fortynningsmidler osv. og vanligvis for parenteral tilførsel. Omtrentlige effektive dosemengder er i området fra 100 til 800 mg/dag avhengig av de individuelle aktiviteter til de aktuelle forbindelser for en pasient med gjennomsnittlig kroppsvekt på 70 kg. Mer vanlige dosemengder for foretrukne og mer aktive forbindelser vil være i området fra 200 til 800 mg/dag, mer foretrukket fra 200 til 500 mg/dag og mest foretrukket fra 200 til 250 mg/dag. De kan gis i form av enkle doser, som oppdelte doser og/eller i multiple doser som varer over flere dager. For oral tilførsel kan de være i form av tabletter, kapsler, løsninger eller suspensjoner som inneholder fra 100 til 500 mg forbindelse pr. doseenhet. Alternativt og foretrukket vil forbindelsene være utformet for parenteral tilførsel i en passende bærer som kan tilføres parenteralt og med tilveiebringelse av en eneste daglig dosemengde i området fra 200 til 800 mg, foretrukket fra 200 til 500 og mer foretrukket fra 200 til 250 mg. Slik effektiv daglig dose vil imidlertid variere avhengig av den aktive bestanddels iboende aktivitet og av pasientens kroppsvekt, idet slike variasjoner er innenfor legens faglige bedømning.
I forbindelse med spesielle tilførsler mener man at steroid sulfatase-inhibitorene kan anvendes i kombinasjonsterapier, enten med andre sulfataseinhibitorer eller for eksempel i kombinasjon med en aromataseinhibitor som for eksempel 4-hydroksyandrostendion (4-OHA).
I det etterfølgende er det angitt eksempler og forsøksdata:
Eksempel 1
Fremstilling av østron-3-sulfamat
Natriumhydrid (60 % dispersjon, 2 ekv.) og sulfamoylklorid
(2 ekv.) ble tilsatt til en omrørt løsning av østron (l ekv.) i vannfri dimetylformamid ved 0°C. Deretter ble reaksjonsblandingen oppvarmet til romtemperatur og omrøringen ble fortsatt ytterligere 24 timer.
Reaksjonsblandingen ble helt inn i en kald mettet løsning av natriumbikarbonat og den oppnådde vannfase ble ekstrahert med diklormetan. De kombinerte organiske ekstrakter ble tørket over vannfri MgS04. Filtrering etter avdamping av løsnings-middel i vakuum og ko-avdamping med toluen ga en uren rest som renses ytterligere ved flash-kromatografi.

Prosedyren i eksempel 1 ble gjentatt med unntak av at sulfamoylklorid ble erstattet med den samme mengde N-metylsulfa-moylklorid.

Eksempel 4
Inhibering av steroid sulfatase- aktivitet i MCF- 7- celler med østron- 3 - sulfamat
Steroidsulfatase er definert som: Sterylsulfatase EC 3.1.6.2.
Steroid sulfatase-aktivitet ble målt in vitro ved anvendelse av intakte MCF-7 humane brystcancerceller. Denne hormon-avhengige cellelinje anvendes i stor grad for å studere kontrollen av human brystcancer-cellevekst. Den utviser signifikant steroid sulfatase-aktivitet (Maclndoe et al Endocrinology, 123, 1281-1287 (1988); Purohit & Reed, Int. J. Cancer, 50, 901-905 (1992)) og er tilgjengelig i U.S.A. fra the American Type Culture Collection (ATCC) og i U.K. (f.eks. fra The Imperial Cancer Research Fund). Celler ble holdt i Minimal Essential Medium (MEM) (Flow Laboratories, Irvine, Skottland) inneholdende 2 0 mM HEPES, 5 % føtalt bovint serum, 2 mM glutamin, ikke-essensiell aminosyrer og 0,075 * natriumbikarbonat. Opp til 30 25 cm<2> replikat-vevsdyrkingskolber ble inokulert med omtrent 1 x IO<5> celler/kolbe ved anvendelse av det ovennevnte medium. Cellene ble dyrket til 80 % konfluens og mediet ble skiftet hver tredje dag.
Intakte monolag av MCF-7-celler i 25 cm<2> vevsdyrkingskolber in triplo ble vasket med Earle<1>s Balanced Salt Solution (EBSS fra ICN Flow, High Wycombe, U.K.) og inkubert i 3-4 timer ved 37°C med 5 pmol (7 x 10<5> dpm) [6,7-<3>H]østron-3-sulfat (spesifikk aktivitet 60 Ci/mmol fra New England Nuclear, Boston, Mass., U.S.A.) i serumfritt MEM (2,5 ml) sammen med østron-3-sulfamat (11 konsentrasjoner: 0; lfM; 0,01pM; 0,lpM; lpM; 0,01nM; 0,lnM; 1 nM; 0,01/xM; 0,1^M; ljiM) . Etter inkubasjon ble hver kolbe avkjølt og mediet (l ml) ble pipettert inn i separate rør inneholdende [14C] østron (7 x IO<3> dpm) (spesifikk aktivitet 97Ci/mmol fra Amersham International Radiochemical Centre, Amersham, U.K.). Blandingen ble rystet inngående i 30 sekunder med toluen (5 ml). Forsøk viste at >90 % [14C] østron og <0,1 % [<3>H]østron-3-sulfat var fjernet fra vannfasen ved denne behandling. En del (2 ml) av den organiske fase ble fjernet, avdampet og <3>H og <14>C innholdet i resten ble bestemt ved seintillasjonsspektrometri. Massen av hydrolysert østron-
3-sulfat ble beregnet fra de oppnådde <3>H-tellinger {korrigert for anvendte volumdeler medium og organisk fase og for utvinning av tilsatt [<14>C]østron) og den spesifikke aktivitet til substratet. Hver gruppe av forsøk omfattet inkubasjoner av mikrosomer fremstilt fra sulfatasepositiv human placenta (positiv kontroll) og kolber uten celler (for å bestemme tilsynelatende ikke-enzymatisk hydrolyse av substratet). Antallet cellekjerner pr. kolbe ble bestemt ved anvendelse av en Coulter Counter etter behandling av cellemonolagene med Zaponin. En kolbe i hver gruppe ble anvendt for å vurdere cellemembranstatus og levedyktighet ved anvendelse av Trypan Blue utelukkelsesmetoden (Phillips, H.J. {1973) In: Tissue culture and applications, [eds: Kruse, D.F. & Patterson, M.K.],- s. 406-408; Academic Press, New York) .
Data for østron-3-sulfamat er vist i tabell I og figurene 2 og 4. Resultater for steroid sulfatase-aktivitet er uttrykt som gjennomsnittet ± 1 standardavvik av det totale produkt (østron + østradiol) dannet under inkubasjonsperioden (20 timer) beregnet for 10<6> celler og, for verdier som viser statistisk signifikans, som en prosentvis reduksjon (inhibering) i forhold til inkubasjoner som ikke inneholdt østron-3-sulfamat. Uparet Student's t-test ble anvendt for å teste resultatenes statistiske signifikans.
Eksempel 5
Inhiberina av steroid sulfatase-aktivitet i MCF-7-cel ler ved østron- 3- N. N- dimetylsulfamat
En forsøksprosedyre som er identisk med den som er beskrevet i eksempel 4 ble anvendt for å oppnå resultater for østron-3-N,N-dimetylsulfamat med unntak av at inkubasjoner inneholdt østron-3-N,N-dimetylsulfamat (5 konsentrasjoner: 0; 0,001 /iM; 0,01 /iM; 0,1 jiVl; 1 ( M) i stedet for østron-3-sulfamat.
Resultatene for østron-3-N,N-dimetylsulfamat er vist i tabell II og figur 3 og er uttrykt på samme måte som henholdsvis tabell I og figur 2. I tillegg er log dose-respons kurven sammenlignet med østron-3-sulfamat i figur 4.
Eksempel 6
Inhiberinq av steroid sulfatase-aktivitet i MC F- 7- celler ved forbehandling med østron-3-sulfamat oa østron-3-M. N- dimetylsulf amat
En forsøksprosedyre som er lik den som er beskrevet i eksempel 4 ble anvendt for å bestemme virkningen av å forbehandle MCF-7-celler med henholdsvis østron-3-sulfamat og østron-3-N,N-dimetylsulfamat.
Intakte monolag ble først inkubert i 2 timer ved 37°C med
0,1 fiK østron-3-sulf amat, østron-3-N, N-dimetylsulf amat eller medium alene (kontroll). Mediet som badet cellene ble deretter fjernet ved avsugning og cellene ble vasket 3 ganger påfølgende med 5 ml medium hver gang. De oppnådde vaskede celler ble deretter resuspendert og inkubert i 3-4 timer ved 37°C i et medium som inneholdt 5 pmol (7 x 10<5> dpm) [6,7-<3>H]østron-3-sulfat. Alle andre trinn var identiske med dem som er beskrevet i eksemplene 3 og 4.
Resultater for østron-3-sulfamat og østron-3-N,N-dimetylsulfamat er vist i tabell III og er uttrykt på tilsvarende måte som tabell I.
Eksempel 7
Inhiberina av steroid sulfatase-aktivitet i placentamikrosomer ved østron- 3- sulfamat
Sulfatase-positiv human placenta fra fullbyrdede svangerskap (Obstetric Ward, St. Mary's Hospital, London) ble finskåret med sakser og vasket en gang med kald fosfatbuffer (pH 7,4, 50 mM) og deretter resuspendert i kald fosfatbuffer (5 ml/g vev). Homogenisering ble gjennomført med en Ultra-Turax homogenisator ved anvendelse av tre omganger av 10 sekunder separert med 2 minutters perioder for avkjøling på is. Kjerne- og cellebiter ble fjernet ved sentrifugering (4°C) ved 2000 g i 30 minutter og porsjoner (2 ml) av supernatanten ble lagret ved -20°C. Proteinkonsentrasjonen i supernatantene ble bestemt ved metoden etter Bradford (Anal. Biochem., 72, 248-254 (1976)).
Inkubasjoner (1 ml) ble gjennomført ved anvendelse av en proteinkonsentras jon på 100 //g/ml, substratkonsentrasjon på 20 ptM [6,7-3H] østron-3-sulfat (spesifikk aktivitet 60 Ci/mmol fra New England Nuclear, Boston, Mass., U.S.A.) og en inkuba-sjonstid på 2 0 minutter ved 3 7°C. Åtte konsentrasjoner av østron-3-sulfamat ble anvendt: 0 (dvs. kontroll); 0,05 fiVl; 0,1 HM; 0,2 jM; 0,4 /iM; 0,6 fiM; 0,8 jtM; 1,0 flVl. Etter inkubasjon ble hver prøve avkjølt og mediet (1 ml) ble pipettert inn i separate rør inneholdende [14C] østron (7 x IO<3> dpm)
{spesifikk aktivitet 97 Ci/mmol fra Amersham International-Radiochemical Centre, Amersham, U.K.). Blandingen ble rystet inngående i 3 0 sekunder med toluen {5 ml). Forsøk viste at
>90% [14C] østron og <0,1 % [3H] østron-3-sulf at var fjernet fra vannfasen ved denne behandling. En porsjon (2 ml) av den organiske fase ble fjernet, avdampet og <3>H- og <14>C- innholdet i resten ble bestemt ved seintillasjonsspektrometri. Massen av hydrolysert østron-3-sulfat ble beregnet fra oppnådde <3>H-tellinger (korrigert for volumdelene av anvendt medium og organisk fase og for gjenvinning av tilsatt [14C] østron) og den spesifikke aktivitet av substratet.
Resultatene for østron-3-sulfamat er vist i tabell IV og figur 5. Resultatene av steroid sulfatase-aktivitet er uttrykt i tabell IV som totalt produkt {østron + østradiol) dannet under inkubasjonsperioden (tid) og som en prosentvis reduksjon (inhibering) i forhold til inkubasjoner som ikke inneholdt østron-3-sulfamat og som virket som kontroll. Resultatene for steroid sulfatase-aktivitet er uttrykt i figur 4 som prosentvis reduksjon (inhibering) i forhold til kontroll mot konsentrasjonen av østron-3-sulfamat og inkluderer den beregnede IC50-verdi (dvs. konsentrasjonen av østron-3-sulfamat som gir 50 % inhibering i forhold til kontroll) på 0,07 /zM.
Eksempel 8
Inhibering av steroid sulfatase-aktivitet i levermikrosompreparater fra rotter behandlet med subku tan østron- 3- sulfamat Fire grupper av 3 hunn-Wistar-rotter (vekt fra 80-110 g) ble gitt subkutane injeksjoner på 100 fil (en gang daglig i 7 dager, baerermedium: propylenglykol) av enten:
Propylenglykol (bærerkontroll)
Østron-3-sulfamat (10 mg/kg/dag)
Østron-3-sulfat (10 mg/kg/dag) (substratkontroll) Østron-3-sulfat (10 mg/kg/dag) + østron-3-sulfamat
(10 mg/kg/dag)
På den åttende dag ble alle rottene avlivet og leverne ble fjernet ved dissekering. Levermikrosompreparater ble fremstilt ved en prosedyre som er analog med den som er beskrevet i eksempel 6 med unntak av at vevskilden var rottelever og at forsøk in duplo for å bestemme steroid sulfatase-aktivitet ble gjennomført ved anvendelse av [6,7-3H] østron-3-sulfat og [7-<3>H]dehydroepiandrosteron-3-sulfat som separate substrater. Resultater for steroid sulfatase-aktivitet er vist i tabell V og er uttrykt som totalt produkt dannet under inkubasjonsperioden i form av gjennomsnitt + l st.avvik. Resultater for inkubasjoner av vev oppnådd fra grupper av rotter behandlet med østron-3-sulfamat er også uttrykt som en prosentandel reduksjon (inhibering) i steroid sulfatase-aktivitet sammenlignet med deres respektive kontroller.
Claims (13)
1. Anvendelse av en ringssystearforbindelse for fremstilling av et medikament for å kontrollere østrogenproduksjon, hvor ringsystemforbindelsen omfatter en ring og en sulfamatgruppe med formelen
hvor hver av Rx og R2 uavhengig er valgt fra H, alkyl, alkenyl, cykloaikyl og aryl, eller danner sammen alkylen som eventuelt inneholder ett eller flere heteroatomer eller grupper i alkylenkjeden;
hvor nevnte forbindelse er en inhibitor av et enzym med steroid sulfatase-aktivitet (EC 3.1.6.2.), og hvor, dersom sulfamatgruppen av nevnte forbindelse erstattes med en sulfatgruppe for å danne en sulfatforbindelse og inkuberes med et steroid sulfatase-enzym (EC 3.1.6.2.) ved en pH 7,4 og 37°C, tilveiebringes en verdi på mindre enn 50 /iM.
2. Anvendelse av en ringsystemforbindelse for fremstilling av et medikament for å targetrette det østrogen-metabolske spor, hvor ringsystemforbindelsen omfatter en ring og en sulfamatgruppe med formelen
hvor hver av Rx og R2 uavhengig er valgt fra H, alkyl, alkenyl,, cykloaikyl og aryl, eller danner sammen alkylen som eventuelt inneholder ett eller flere heteroatomer eller grupper i alkylenkjeden;
hvor nevnte forbindelse er en inhibitor av et enzym med steroid sulfatase-aktivitet (EC 3.1.6.2.), og hvor, dersom sulfamatgruppen av nevnte forbindelse erstattes med en sulfatgruppe for å danne en sulfatforbindelse og inkuberes med et steroid sulfatase-enzym (EC' 3.1.6.2.) ved en pH 7,4 og 3 7° C, tilveiebringes en K,,, verdi på mindre enn 50 /xM.
3. Anvendelse som angitt i krav 1 eller 2, hvor forbindelsen er en forbindelse med formelen
hvor Rx og R2 er som definert i krav' l eller 2.
4 . Anvendelse som angitt i krav 3, hvor gruppen polysyklus er en rest av en polycyklisk alkohol.
5. Anvendelse som angitt i krav 3 eller 4, hvor gruppen polysyklus representerer resten av en sterol.
6. Anvendelse som angitt i krav 5, hvor sterolen er en 3-sterol.
7. Anvendelse som angitt i krav 5 eller 6, hvor sterolen er valgt fra gruppen bestående av østron, dehydroepiandrosteroner/ substituerte østroner og substituerte dehydroepiandrosteroner.
8. Anvendelse som angitt i ett eller flere av kravene 1 til 7, hvor Ri og R2 uavhengig er valgt fra H eller et C^C^ alkyl.
9. Anvendelse som angitt i ett eller flere av kravene 1 til 8, hvor Rx og R2 uavhengig er valgt fra H eller C-^Cg alkyl.
10. Anvendelse som angitt i ett eller flere av kravene 1 til 9, hvor R-l og R2 uavhengig er valgt fra H eller metyl.
11. Anvendelse som angitt i ett eller flere av kravene 1 til 10, hvor forbindelsen er hvilken som helst av østron-3-sulfamat, østron-3-N,N-dimetylsulfamat, østron-3-N-monometyl-sulfamat.
12. Anvendelse som angitt i ett eller flere av kravene 1 til 11, hvor minst en av R1 og R2 er H.
13. Anvendelse som angitt i krav 12, hvor Rx er H og R2 er H.
