Description
Det er kjent forskjellige purinderivater, innbefattet guaninderivater, som har virkning som hemmere av purin-nukleosidfosforylase (PNP-4), ifølge US-patentsøknad 767.202 av 22/8-1985, nå patent 4.772.606 av 20/9-1988.
Utvalgte tidligere kjente guaninderivater er også beskrevet i nevnte patent.
Mer spesifikt er pyrrolo[3,2-d]pyrimidin-4-oner med følgende formel kjent:
Forbindelsen med formel 1 er beskrevet i J. Org. Chem., 1979, 44:3826 og forbindelsene med formel 2, 3 og 4 er beskrevet av J.D. Stoeckler et al., Cancer Res., 1986, 46:1774). Ifølge sistnevnte har forbindelser med formel 2, 3 og 4 PNP-virkning. Disse forbindelsene skiller seg fra de foreliggende ved en aryl-eller heteroarylsubstituent i stedet for metylgruppen i forbindelsen med formel 1 og sukkerdelen i forbindelser med formel 2, 3 og 4. Forbindelsene fremstillet i henhold til foreliggende oppfinnelse er således ikke forbindelser som er selvsagte varianter av disse.
Foreliggende oppfinnelse angår fremstillingen av en forbindelse med formel (I)
hvor Ar er (i) fenyl som er usubstituert eller substituert med alkoksy med 1-4 karbonatomer, (ii) 2- eller 3-tienyl, (iii) 2-eller 3-furanyl; eller et farmasøytisk akseptabelt syre- eller baseaddisjonssalt derav.
Forbindelser fremstillet i henhold til oppfinnelsen, kan benyttes for behandling av autoimmunsykdommer; så som artritt, systemisk lupus erythematosus, inflammatoriske tarmsykdommer, juvenil diabetes, myastenia gravis, multippel sklerose, gikt og giktisk artritt, samt av psoriasis, virusinfeksjoner og cancer eller avstøtning av transplantater, som omfatter en immunomodulator- eller antiavstøtnings-effektiv mengde; så som en T-celle cytotoksisk mengde, av en forbindelse med formel I med et farmasøytisk akseptabelt bæremiddel.
(A) I henhold til foreliggende oppfinnelse fremstilles nye forbindelser med formel (I) ved at en forbindelse med formel (II)
hvor Ar er som ovenfor definert; behandles med Na2S204 og i basisk oppløsning, så som IN NaOH, under oppvarming, fortrinnsvis til 90 °C, for å oppnå forbindelser med formel I. Tilslutt kan forbindelsen med formel I benyttes for å fremstille et farmasøytisk akseptabelt syreaddisjons- eller basesalt derav.
(B) En forbindelse med formel II med den ovenfor angitte definisjon, kan fremstilles ved behandling av en forbindelse
med formel III
med (1) en base så som IN NaOH-oppløsning ved romtemperatur, og deretter (2) med en syre, så som saltsyre.
(C) En forbindelse med formel III, med den ovenfor angitte definisjon, kan fremstilles ved å omsette en forbindelse med
formel IV
med en forbindelse med formel (V) hvor Ar er som ovenfor definert, i nærvær av natriumhydrid i dimetylformamid, eller av kalium-t-butoksyd i DMSO, eller under lignende betingelser. Det kan noen ganger være nødvendig å beskytte N- eller 0-gruppene i mellomproduktene med passende kjente beskyttelsesgrupper. Innføring og fjerning av slike oksygen- og nitrogen-beskyttelsesgrupper er velkjent innen den organiske kjemi; se for eksempel (1) "Protective Groups in Organic Chemistry," J.F.W. McOmie, red. Eksempler på hensiktsmessige oksygen-beskyttelsesgrupper er benzyl, t-butyldimetylsilyl, metyl, isopropyl, etyl, tert-butyl, etoksyetyl og lignende. Beskyttelse av en N-H-holdig del er nødvendig i noen av de beskrevne fremgangsmåter. Passende nitrogen-beskyttelsesgrupper er benzyl, trifenylmetyl, trialkyl-silyl, trikloretylkarbamat, trikloretoksykarbonyl, vinyloksy-karbamat og lignende.
Iblant er det også nødvendig å beskytte to forskjellige oksygenatomer med ulike beskyttelsesgrupper slik at én av gruppene selektivt kan fjernes mens den andre forblir på plass. Benzyl- og t-butyldimetylsilylgrupper benyttes på denne måte, idet den ene kan fjernes i nærvær av den andre. Således fjernes benzyl ved katalytisk hydrogenolyse og t-butyldimetylsilyl ved omsetning med f.eks. tetra-n-butylammoniumfluorid.
De beskrevne reaksjonsproduktene isoleres ved konvensjonelle teknikker som ekstraksjon, destillasjon, kromatografi og lignende.
De ovenfor beskrevne salter av forbindelser I, fremstilles ved å omsette den passende base med en støkiometrisk mengde av de sure forbindelser med formel I for å oppnå farmakologisk akseptable salter derav.
Forbindelsene med formel I kan også fremstilles i hydrati-serte eller solvatiserte former.
Fremgangsmåtene, startende med (C) via (B) og (A), kan utføres i ett trinn. Det er imidlertid å foretrekke at produktet III og produktet II fraskilles før det neste prosesstrinn.
Forbindelsene med formel I og mellomproduktene II og III forekommer i tautomere former som puriner eller guaniner slik som vist nedenfor. Begge former omfattes av de her angitte formler, og det skjelnes ikke mellom dem i denne beskrivelse.

Ved en foretrukket utførelsesform av f remgSå^ ioåtén i henhold til oppfinnelsen, fremstilles 2 ,6-diamino1-^ *5-*-dihydro-7-(fenylmetyl) -4H-pyrrolo[3 ,2-d]pyrimidin-4-on, 2 ,6\-d'ismino-3,5-dihydro-7-(2-tienylmetyl)-4H-pyrrolo[3,2-d]-pyrimYdin-4-on eller 2,6-diamino-3,5-dihydro-7(3-tienylmetyl)-4H-pyrrV>lo[3,2-d]pyrimidin-4-on. Sistnevnte av disse tre er særlig for vetrukket.
Forbindelsen med formel I fremstilles generelt etter fremgangsmåten vist i det følgende skjema.
Utgangsmaterialene med formel IV og V er enten kommersielt tilgjengelige eller syntetiserbare etter kjente metoder. Forbindelsene med formel IV kan for. eksempel fremstilles etter en fremgangsmåte beskrevet i "Synthetic Procedures in Nucleic Acid Chemistry", Vol. 1 (1973) s. 94, og forbindelsene med formel V kan fremstilles analogt med fremgangsmåter beskrevet i US-patent 4.279.903 og referanser i dette.
Betingelsene for omsetning av forbindelsene V og IV i prosess (C), omfatter dråpevis tilsetning av natriumhydrid-oljesuspensjon til en oppløsning av forbindelse V i et opp-løsningsmiddel, så som dimetylformamid (DMF), etterfulgt av tilsetning av en oppløsning av forbindelse IV, også i et oppløsningsmiddel som DMF, under tørr nitrogenatmosfære. Den resulterende reaksjonsblanding oppvarmes til 60°-90°C i 0,5-2 timer og surgjøres deretter til ca. pH 3-4 med 1-6N HC1. Kondensasjonen i prosess (C) kan også foretas i et oppløsnings-middel, så som DMSO, og i nærvær av t-butoksyd.
Betingelsene for prosess (B) for behandling av produktet III fra (C), består i først å behandle en oppløsning av forbindelsen med, for eksempel, IN NaOH ved romtemperatur og deretter gjøre blandingen sur med 4N HC1 for å utfelle produktet

I prosess (A) behandles en oppløsning av forbindelsen II i IN NaOH med natriumditionitt ved 70-95°C, fortrinnsvis ved
90°C, i 10-60 minutter, fortrinnsvis 25-40 minutter, hvorpå den surgjøres igjen til pH 3 eller 4 for å gi krystaller av produktet I.
Forbindelsene I, fremstillet i henhold til foreliggende oppfinnelse, har vist seg å ha signifikant enzym-hemmende og cytotoksisk aktivitet. I purin-nukleosidfosforylase (PNP-4) enzymforsøk, oppnås en ICsg-verdi ved en dosering på ca. 0,9-3,18 pmol for enkelte av forbindelsene. PNP-4 aktiviteten av forbindelsene I bestemmes radiokjemisk ved å måle dannelsen av [<14>C]-hypoksantin fra [<14>C]inosin [Biomedicine, 33, 39 (1980)] ved å benytte humane erytrocytter som enzymkilde. En in vivo hemming av purin-nukleosidfosforylase-bestemmelse (HPLC-1) benyttes i det vesentlige som beskrevet i "Annals of New York Academy of Sciences", Vol. 451, s. 313 (1985) for ytterligere å illustrere aktiviteten av forbindelsene I. Ved en standardtest (HTBA-1) [Science, 214. 1137, (1981)] ble de samme forbindelsene funnet å være selektivt cytotoksiske for T-celler i nærvær av 2'-deoksyguanosin i et tilsvarende konsentrasjonsområde og ugiftige overfor B-celler i nærvær av samme mengde 2'-deoksyguanosin, hvilket viser at forbindelsene I kan benyttes slik som omtalt her. Siden fjerning eller modulering av T-celler er gunstig ved behandling av autoimmune lidelser, vil disse selektivt T-celle cytotoksiske forbindelser være nyttige ved behandlingen. 8-aminoguanosin, en kjent PNP-hemmer, har vist seg å være effektiv som hemmer av hudavstøtning ved trans-plantasjon hos hunder [J.B. Benear et al., Transplantation, 1986, 41:274]. Klinisk har det vist seg at modulering og/eller fjerning av T-celler av "throacic duct drainage", lymphapheresis eller total lymfebestråling, har gitt delvis eller fullstendig lindring av reumatoid artritt hos pasienter som har vært helt uhelbredelige ved andre terapiformer (A. Tanay et al., Arthritis and Rheumatism, Vol. 30, No. 1. p. 1 (1987). S. Strober et al., Annual of Internal Medicine, V-102, No. 4, 441-449 (1985); H.G. Nusslein et al., Arthritis and Rheumatism, V-28, No. 11, 1205-1210 (1985); E. Brahn et al., ibid, V-27, No. 5, 481-487 (1984) og J. Karsh et al., ibid, V-24, No. 7, 867-873 (1981)). Cyklosporin A, en T-celle modulator, har hatt gunstige effekter ved behandling av juvenil diabetes (A. Assan et al., The Lancet, January 12, p. 67, (1985). For tiden er dessuten cyklosporin A det foretrukne medikament for å hindre trans-plantasjonsavstøtninger (R.M. Merion et al., New Eng. J. Med.,
(1984), 148). I det siste har cyklosporin A vist seg egnet for behandling av psoriasis. Det synes dessuten som cyklosporin-terapi klart nedsetter aktiverte T-celler ved psoriaslesjoner. Det er derfor rimelig å anta at vellykket behandling av psoriasis er basert på modulering av T-celleaktivitet (se C.N. Ellis et al., JAMA. V-256, No. 22, Dec. 12, 1986, pp. 3110-3116). Endelig, har cyklosporin A vist seg å ha effekt ved reumatoid artritt (M.E. Weinblatt et al., Arthritis and Rheumatism, V-30, No. 1, pp. 11-17 (January, 1987); 0. Forre et al., Arthritis and Rheumatism, V-30, No. 1, pp. 88-92 (January, 10
1987); M. Dougados et al., Arthritis and Rheumatism, Vol. 30. No. 1, pp. 83-87 (January, 1987).
Representative eksempler med forbindelser fremstillet i henhold til oppfinnelsen, er vist i den etterfølgende tabell som viser de ovenfor diskuterte virkninger.
AKTIVITETSTABELL
In vivo undersøkelser etter de ovenfor beskrevne metoder, kan benyttes for å bestemme forbindelsenes virkning ved de respektive sykdomstilstander.
Siden T-celler spiller en sentral rolle ved immun-respons, vil de nye forbindelsene kunne benyttes ved immuno-reguleringen for å hindre transplantatavstøtning eller for å behandle psoriasis og autoimmune sykdommer, så som reumatoid artritt, systemisk lupus erythematose, inflammatorisk tarmsykdom, multippel sklerose, myastenia gravis, gikt eller giktisk artritt, juvenil diabetes, cancer og virussykdommer. Forbindelser med formel I vil derfor inngå i preparater som skal motvirke transplantasjonsavstøtning eller for behandling av lidelser som psoriasis hos mennesker, eller av autoimmunsykdommer karakterisert ved unormal immun-respons hos primater eller mennesker. De nye forbindelsenes egenskaper benyttes i så henseende ved å gi varmblodige dyr en virksom mengde av et farmasøytisk preparat som inneholder virkestoffet i minst 0,1 vektprosent.
Oppfinnelsen er i det følgende ytterligere illustrert gjennom de følgende eksempler.
Eksempel 1
A. 2- amino- ot- cyano- l. 6- dihydro- 5- nitro- Q!- ( fenylmetyl)- 6- okso- 4-pyrimidineddiksyre. etylester. En oppløsning av etyl 2-cyano-3-fenyl-propionat (JCS, 1944, 13) (6,0 g) i DMF (25 ml) tilsettes dråpevis til en suspensjon av natriumhydrid (1,2 g, 60% suspensjon i olje, vasket med heksan) i DMF (25 ml) under en atmosfære av tørr N2. Reaksjonsblandingen omrøres i 15 minutter ved romtemperatur, hvorpå en oppløsning av 2-amino-6-klor-5-nitro-4(3H)-pyrimidinon (Synthetic Procedures in Nucleic Acid, V-l, 1973, p. 94) (1,9 g, nylig krystallisert fra metanol) i DMF (50 ml) tilsettes til anionet. Reaksjonsblandingen oppvarmes til 65°C i 24 timer og surgjøres deretter til pH 3 med IN HC1.
Det meste DMF fjernes ved rotasjonsfordampning under høyvakuum. Den gjenværende olje fordeles mellom etylacetat (500 ml) og vann. Det organiske lag tørkes (Na2S04) og inndampes til et gummiaktig faststoff. Residuet utgnis med eter og frafiltreres. Det tørkes under vakuum for å gi 2-amino-a-cyano-1,6-dihydro-5-nitro-a-(fenylmetyl)-6-okso-4-pyrimidineddiksyre, etylester som et partielt hydrat (2,25 g, 62%).
B. 2- amino- l. 6- dihydro- 5- nitro- 6- okso- a-( fenylmetyl)- 4- pyrimidin-acetonitril. En opplesning av 2-amino-oc-cyano-l,6-dihydro-5-nitro-ot- (fenylmetyl) -6-okso-4 -pyrimidineddiksyre, etylester, fremstillet som angitt ovenfor (2,0 g) i IN NaOH (100 ml), ble omrørt ved romtemperatur i 1 time og surgjort med 4N HC1 (30 ml). Bunnfallet ble frafiltrert og tørket under vakuum for å gi 2-amino-l,6-dihydro-5-nitro-6-okso-a-(fenylmetyl)-4-pyrimidinacetonitril (600 mg, 36%). C. 2. 6- diamino- 3. 5- dihydro- 7- ffenylmetyl)- 4H- pyrrolo-T3. 2- d1pyrimidin- 4- on. Til en oppløsning av 2-amino-l,6-dihydro-5-nitro-6-okso-a-(fenylmetyl)-4-pyrimidin-acetonitril, fremstillet som ovenfor (10,0 g) i IN NaOH (600 ml), tilsettes natriumditionitt (35 g). Reaksjonsblandingen oppvarmes til 90°C i 35 minutter og surgjøres til pH 4 med 4N HC1 mens den enda er varm. Reaksjonsblandingen avkjøles i et isbad og bunnfallet frafiltreres. Det tørkes over P2O5 under vakuum (4,0 g). Det rå (undekarboksylerte) produkt oppløses i 300 ml konsentrert HC1 og filtreres raskt gjennom en glassfritte før utkrystallisering. Den resulterende suspensjon kokes i 5 minutter og avkjøles. Produktet 2,6-diamino-3,5-dihydro-7-(fenylmetyl)-4H-pyrrolo[3,2-d]pyrimidin-4-on frafiltreres (1,98 g, 23%), smp. >250°C (dekomp.). Eksempel 2 A. 2- amino- a- cyano- l. 6- dihvdro- 5- nitro- 6- okso- a-( 2- tienylmetvl)-4- pyrimidineddiksYre. metvlester. Natriumhydrid (4,5 g, 60% suspensjon i olje, vasket med heksan) suspenderes i tørr DMF (50 ml) under en atmosfære av tørr N2 og en oppløsning av metyl 2-cyano-3-(tienyl)propionat (US-patent 4.279.903) (22,0 g) i tørr DMF (50 ml) tilsettes dråpevis og resulterer i dannelse av en mørkeblå oppløsning. En oppløsning av nylig omkrystallisert 2-amino-6-klor-5-nitro-4(3H)-pyrimidinon (8,57 g) i DMF (75 ml) tilsettes i én porsjon. Reaksjonsblandingen oppvarmes til 60°C over natten og avkjøles, hvorpå den surgjøres til pH 5 med IN HC1. Den helles over i 1000 ml etylacetat og ekstraheres med vann (4 x 300 ml). Det organiske lag inndampes nesten til tørrhet, og residuet suspenderes i eter og frafiltreres. Råproduktet vaskes med heksan inntil grønnfarven fra vaskevæsken forsvinner. Faststoffet tørkes under vakuum for å gi 2-amino-cx-cyano-1,6-dihydro-5-nitro-6-okso-a-(2-tienylmetyl)-4-pyrimidin-eddiksyre, metylester.
B. 2- amino- l. 6- dihvdro- 5- nitro- 6- okso- a-( 2- tienvlmetyl)- 4-<py>rimidinacetonitril. En oppløsning av 2-amino-a-cyano-l,6-dihydro-5-nitro-6-okso-a-(2-tienylmetyl)-4-pyrimidineddiksyre, etylester, fremstillet som ovenfor, (5,0 g)'i IN NaOH (200 ml) omrøres ved romtemperatur i 90 minutter. Reaksjonsblandingen surgjøres til pH 1 med 4N HC1 og omrøres i 5 minutter. Reaksjonsblandingen nøytraliseres (pH 7) med IN NaOH og produktet 2-amino-1,6-dihydro-5-nitro-6-okso-cx-(2-tienylmetyl)-4-pyrimidin-acetonitril (4,1 g, 98%) frafiltreres.
C. 2 . 6- d. iamino- 3 . 5- dihydro- 7-( 2- tienylmetyl) - 4H- pyrrolo-T3. 2- dlpyrimidin- 4- on. Til en oppløsning av 2-amino-l,6-dihydro-5-nitro-6-okso-a-(2-tienylmetyl)-4-pyrimidinacetonitril fremstillet som ovenfor (4,0 g) i IN NaOH (250 ml), tilsettes natriumditionitt (17 g), og reaksjonsblandingen oppvarmes til 90°C i 20 minutter. I fremdeles varm tilstand surgjøres reaksjonsblandingen til pH 2 med 4N HC1 og filtreres, hvorpå den avkjøles og nøytraliseres med IN NaOH. Det resulterende bunnfall frafiltreres og tørkes. Råproduktet tilsettes til en omrørt oppløsning av konsentrert HC1 (150 ml), og hydroklorid-saltet frafiltreres. Saltet oppløses i vandig NaOH og felles ut igjen med IN HC1. Omkrystallisasjon fra 2N HC1 gir det analytisk rene hydrokloridsalt monohydrat av 2,6-diamino-3,5-dihydro-7-(2-tienylmetyl)-4H-pyrrolo[3,2-d]pyrimidin-4-on (0,82 g, 19%), smp. 220-225°C (dekomp.).
Eksempel 3
A. 2- amino- a- cyanc— 1. 6- dihvdro- 5- nitro- 6- okso- a-( 3- tienvlmetvl)-4- pvrimidineddiksyre. et<y>lester. Natriumhydrid (5,7 g; 60% suspensjon i olje, vasket med heksan) suspenderes i tørr DMF
(50 ml) under en atmosfære av tørr N2 og tilsettes dråpevis en oppløsning av metyl 2-cyano-3-(3-tienyl)propionat (US-patent 4.279.903) (28,0 g) i DMF (50 ml). 2-amino-6-klor-5-nitro-4(3H)-pyrimidinon (9,1 g, nylig omkrystallisert) i DMF (50 ml) tilsettes. Reaksjonsblandingen oppvarmes til 70°C over natten, avkjøles og surgjøres deretter til pH 4 med IN HC1 under avkjøling i isbad. Reaksjonsblandingen fortynnes til 1000 ml med kaldt vann og det resulterende bunnfall frafiltreres. Det renses med heksan/etylacetat og tørkes for å gi 2-amino-a-cyano-1,6-dihydro-5-nitro-6-okso-cx-(3-tienylmetyl)-4-pyrimidin-eddiksyre, etylester (Ar = 3-Th) (8,8 g, 52%).
B. 2- amino- l. 6- dihydro- 5- nitro- 6- okso- a-( 3- tienylmetyl)-4-pyrimidinacetonitril. En oppløsning av 2-amino-a-cyano-l,6-dihydro-5-nitro-6-okso-a-(3-tienylmetyl)-4-pyrimidin-eddiksyre, etylester i IN NaOH (200 ml) omrøres i 2 timer ved romtemperatur og surgjøres deretter til pH 1 ved dråpevis tilsetning av konsentrert HCl. Den resulterende suspensjon oppvarmes (45°C) i 2 minutter og avkjøles. Deretter justeres pH til 3 med NH4OH. Faststoffet frafiltreres, vaskes med vann og tørkes under vakuum for å gi 2-amino-l,6-dihydro-5-nitro-6-okso-cx-(3-tienylmetyl) -4-pyrimidinacetonitril (3,03 g).
C. 2. 6- diamino- 3. 5- dihydro- 7-( 3- tienylmetyl)- 4H- pyrrolor3. 2-dlpvrimidin- 4- on. Til en oppløsning av 2-amino-l,6-dihydro-5-nitro-6-okso-a-(3-tienylmetyl)-4-pyrimidinacetonitril fremstillet som ovenfor (5,0 g) i IN NaOH (300 ml) tilsettes natriumditionitt (20 g). Reaksjonblandingen oppvarmes til 90°C i 30 minutter og surgjøres deretter (pH=l) i fremdeles varm tilstand med konsentrert HCl. Reaksjonsblandingen avkjøles og nøytraliseres med ammoniumhydroksyd. Det resulterende bunnfall frafiltreres, vaskes med kaldt vann og tørkes under vakuum. Råproduktet tilsettes i små porsjoner under omrøring til 100 ml konsentrert HCl, hvoretter hydrokloridet frafiltreres. Produktet omkrystal-liseres fra IN HCl for å gi 2,6-diamino-3,5-dihydro-7-(3-tienylmetyl)-4H-pyrrolo[3,2-d]pyrimidin-4-on som monohydro-kloridsaltet (2,35 g), smp. >185°C (dekomp.).
Eksempel 4
A. 2- aroino- ot- cvano- p- f 2- furanvlmetvH - 1. 6- dihvdro- 5- nitro- 6- okso-4- pyrimidineddiksyre. et<y>lester. Natriumhydrid (8,4 g, 60% suspensjon i olje, vasket med heksan) suspenderes i tørr DMF (100 ml) under en atmosfære av tørr N2 og tilsettes dråpevis en oppløsning av metyl 2-cyano-3-(2-furanyl)propionat (US-patent 4.279.903) (37,6 g) i DMF (100 ml). Etter fullført tilsetning dannes en klar rød oppløsning. 2-amino-6-klor-5-nitro-4(3H)-pyrimidinon (omkrystallisert fra metanol) (13,34 g) i fast form tilsettes. Reaksjonsblandingen oppvarmes til 100°C i 1 time og til 65°C over natten. Reaksjonsblandingen avkjøles i et isbad, surgjøres med 10% vandig HCl og fortynnes med tilstrekkelig vann for utfelling av produktet 2-amino-a-cyano-a-(2-furanyl-metyl)-1,6-dihydro-5-nitro-6-okso-4-pyrimidineddiksyre, etylester. Produktet frafiltreres og renses med vann og eter (18,5 g).
B. 2- amino- a-( 2- furanylmetvl)- 1. 6- dihvdro- 5- nitro- 6- okso- 4-pyrimidinacetonitril. En oppløsning av 2-amino-a-cyano-a-(2-furanylmetyl)-1,6-dihydro-5-nitro-6-okso-4-pyrimidineddiksyre, etylester (5,0 g) i IN NaOH (200 ml) omrøres ved romtemperatur i 2 timer. Reaksjonsblandingen surgjøres (pH 1) ved dråpevis tilsetning av konsentrert HCl og omrøres ved romtemperatur i 2 minutter. Bunnfallet frafiltreres, vaskes med vann og tørkes for å gi 2-amino-a-(2-furanylmetyl)-1,6-dihydro-5-nitro-6-okso-4-pyrimidinacetonitril (3,02 g, 73%). C. 2. 6- diamino- 3. 5- dihydro- 7-( 2- furanylmetvll - 4H- pyrrolof3. 2-d1pyrimidin- 4- on. Til en oppløsning av 2-amino-a-(2-furanyl-metyl) -1,6-dihydro-5-nitro-6-okso-4-pyrimidinacetonitril (4,0 g) i IN NaOH (250 ml) tilsettes natriumditionitt (16 g), og reaksjonsblandingen oppvarmes til 90°C i 30 minutter. Reaksjonsblandingen avkjøles i isbad og nøytraliseres med 4N HCl. Det resulterende bunnfall frafiltreres og renses ved en serie gjentatte syre/base-utfeliinger. Produktet utfelles først på nytt fra IN NaOH ved surgjøring (pH 2) med en mettet oppløsning av oksalsyre. Denne NaOH/oksalsyre-utfelling gjentas. Den felles deretter ut igjen fra IN NaOH ved å justere pH til 11 med 4N HCl. Produktet, 2,6-diamino-3,5-dihydro-7-(2-furanylmetyl)-4H-pyrrolo[3,2-d]pyrimidin-4-on tørkes under vakuum.
Eksempel 5
Følgende forbindelse fremtilles etter fremgangsmåten i Eksempel 1C ved å benytte korresponderende utgangsmaterialer: 2,6-diamino-3,5-dihydro-7-(2'-metoksyfenylmetyl)-4H-pyrrolo[3,2-d]pyrimidin-4-on monohydroklorid med et tredje mol H20, smp. 215-240°C (dekomp.).
Claims (1)
1. Analogifremgangsmåte for fremstilling av terapeutisk aktive 4H-pyrrolo[3,2-d]-pyrimidin-4-oner med formel (I)
hvor Ar er (i) fenyl som er usubstituert eller substituert med alkoksy med 1-4 karbonatomer, (ii) 2- eller 3-tienyl, (iii) 2-eller 3-furanyl; eller et farmasøytisk akseptabelt syre- eller baseaddisjonssalt derav,
karakterisert ved behandling av en forbindelse med formel II
hvor Ar er som ovenfor angitt; med Na2S204 i en basisk oppløsning under oppvarming til fortrinnsvis 90°C, og eventuelt fremstilling av et farmasøytisk akseptabelt syreaddisjons- eller basesalt av forbindelsen med formel I.
