Abstract

Se provee un sistema mejorado para la produccion a gran escala de una proteina de fusion dimerica que consiste de la porcion de union a ligando extracelular del receptor del factor de necrosis de tumor (TNFR) humano (p75) unido a la porcion Fc de IgGI en el cultivo de celulas, en particular, en medios caracterizados por una o mas de: (i) una concentracion acumulada de aminoacidos mayor que aproximadamente 70 nM; (ii) una relacion molar de glutamina acumulada a asparagina acumulada inferior a aproximadamente 2; (iii) una relacion molar de glutamina acumulada a aminoacidos totales cumulados inferior a aproximadamente 0.2; (iv) una relacion molar de ion inorganico acumulado a aminoacidos totales acumulados de entre alrededor de 0.4 y 1; o (v) una concentracion combinada de glutamina acumulada y asparagina acumulada de entre aproximadamente 16 y 36 mM. El uso de dicho sistema permite altos niveles de produccion de proteina, y disminuye la acumulacion de ciertos factores indeseables, tales como amonio y/o lactato. Adicionalmente, se proveen metodos de cultivo que incluyen un cambio de temperatura, que habitualmente comprende una disminucion en la temperatura cuando el cultivo ha alcanzado alrededor de 20-80% de su densidad celular maxima. Alternativa o adicionalmente, la presente invencion provee metodos tales que, despues de alcanzar un pico, los niveles de lactato y/o amonio en el cultivo disminuyen con el tiempo.
