Abstract
Description
本発明は、Wilms腫瘍の癌抑制遺伝子WT1の産物に基づく癌抗原に関する。この癌抗原は、白血病、骨髄異形成症候群、多発性骨髄腫、悪性リンパ腫などの血液の癌、又は固形癌、例えば胃癌、大腸癌、肺癌、乳癌、胚細胞癌、肝癌、皮膚癌、膀胱癌、前立腺癌、子宮癌、子宮頸癌、卵巣癌等、並びにさらにはWT1を発現するすべての癌に対する抗癌ワクチンとして有用である。
背景技術
異物を排除するための免疫機構には、一般に、抗原を認識して抗原提示細胞として機能するマクロファージ、該マクロファージの抗原提示を認識して種々のリンホカインを産生して他のT−細胞等を活性化するヘルパーT−細胞、該リンホカインの作用により抗体産生細胞に分化するB−リンパ球等が関与する液性免疫と、抗原の提示を受けて分化したキラーT−細胞(細胞傷害性T細胞(CTL)とも称する)が標的細胞を攻撃し破壊する細胞性免疫とがある。
現在のところ、癌の免疫は主として、キラーT−細胞が関与する細胞性免疫によるものと考えられている。キラーT−細胞による癌免疫においては、主要組織適合抗原(Major Histocompatibility Complex;MHC)クラスI(MHCクラスI抗原、ヒトの場合はHLA抗原と呼ばれる)と癌抗原との複合体の形で提示された癌抗原を認識した前駆体T−細胞が分化増殖して生成したキラーT−細胞が癌細胞を攻撃し、破壊する。この際、癌細胞はMHCクラスI抗原と癌抗原との複合体をその細胞表面に提示しており、これがキラーT−細胞の標的とされる(Cur.Opin,Immunol.,5,709,1993;Cur.Opin,Immunol.,5,719,1993;Cell,82,13,1995;Immunol.Rev,146,167,1995)。
標的細胞である癌細胞上にMHCクラスI抗原により提示される前記の癌抗原は、癌細胞内で合成された抗原蛋白質が細胞内プロテアーゼによりプロセシングされて生成した約8〜12個のアミノ酸から成るペプチドであると考えられている(Cur.Opin,Immunol.,5,709,1993;Cur.Opin.Immunol.,5,719,1993;Cell,82,13,1995;Immunol.Rev.,146,167,1995)。
現在、種々の癌について抗原蛋白質の検索が行われているが、癌特異抗原として証明されているものは少ない。
Wilms腫瘍の癌抑制遺伝子WT1(WT1遺伝子)は、Wilms腫瘍、無紅彩、泌尿生殖器異常、精神発達遅滞などを合併するWAGR症候群の解析からWilms腫瘍の原因遺伝子の1つとして染色体11p13から単離された(Gessler,M.ら、Nature,Vol.343,p.774−778(1990))ものであり、ゲノムDNAは約50kbで10のエキソンから成り、そのcDNAは約3kbである。cDNAから推定されるアミノ酸配列は、配列番号:1に示す通りである(Mol.Cell.Biol.,11,1707,1991)。
WT1遺伝子はヒト白血病で高発現しており、白血病細胞をWT1アンチセンスオリゴマーで処理するとその細胞増殖が抑制される(特開平9−104627号公報)ことなどから、WT1遺伝子は白血病細胞の増殖に促進的に働いていることが示唆されている。さらに、WT1遺伝子は、胃癌、大腸癌、肺癌、乳癌、胚細胞癌、肝癌、皮膚癌、膀胱癌、前立腺癌、子宮癌、子宮頸癌、卵巣癌等の固形癌においても高発現しており(特願平9−191635)、WT1遺伝子は白血病及び固形癌における新しい腫瘍マーカーであることが判明した。
WO 00/06602には、WT1遺伝子発現生成物の部分から成る幾つかの癌特異抗原ペプチドが記載されており、その内の有望なものとしてDbと称し、次のアミノ酸配列:Cys Met Thr Trp Asn Gln Met Asn Leu(配列番号:2)(本発明において「WT1ワイルドペプチド」と称する)が記載されている。
発明の開示
従って、本発明は、すでに知られている癌特異抗原ペプチドに比べてより活性の高く、癌ワクチンとして有望なペプチドを提供しようとするものである。
本発明者らは、上記の課題を解決すべく種々検討した結果、すでに知られている上記のアミノ酸配列(配列番号:2)の2番目のアミノ酸MetをTyrに変更したアミノ酸配列:Cys Tyr Thr Trp Asn Gln Met Asn Leu(配列番号:3)を有するペプチド(「WT1改変ペプチド」と称する)がより高い活性を有することを見出し、本発明を完成した。
従って本発明は、次のアミノ酸配列:Cys Tyr Thr Trp Asn Gln Met Asn Leu(配列番号:3)を含んで成り、9〜30個のアミノ酸から成るペプチド(WT1改変ペプチド)を提供する。配列番号:3に示すアミノ酸配列を含む、9〜12個のアミノ酸からなるポリペプチドが好ましく、配列番号:3に示すアミノ酸配列から成るペプチドがさらに好ましい。
本発明はさらに、上記のWT1改変ペプチドを有効成分とする癌ワクチンを提供する。
本発明はさらに、上記のペプチドをコードするDNAを有効成分とする、癌に対するDNAワクチンを提供する。
本発明はさらに、HLA抗原(MHCクラスI抗原)と上記ペプチドとの複合体の提示された抗原提示細胞を提供する。
本発明はさらに、HLA抗原と上記ペプチドとの複合体を認識する細胞傷害性T細胞を提供する。
発明の実施の形態
本発明のペプチドは、配列番号:3に示す9個のアミノ酸から成るアミノ酸配列を含み、9〜30個のアミノ酸から成るペプチドである。さらにHLA抗原に結合して提示されるという観点から、配列番号:3に示すアミノ酸配列を含み、アミノ酸9〜12個から成るペプチドが好ましい。その際、HLA抗原に結合して提示される抗原ペプチドの配列の規則性(モチーフ)を有するペプチドが、より好ましい(J.Immunol.,152,p3913,1994,Immunogenetics,41:p178,1995,J.Immunol.,155:p4307,1994,J.Immunol.,155:p4749,1995)。さらに、配列番号:3に示す9個のアミノ酸のアミノ酸配列から成るペプチドが最も好ましい。
なお、前記で「配列番号:3に示すアミノ酸配列を含むペプチド」とは、具体的には、例えば、配列番号:3に示すアミノ酸配列を含み、WT1(配列番号:1)上の該当位置(第235位〜第243位)、又はヒトWT1(NCBIデータベースAccession No.XP012009)上の対応位置よりN末端方向及び/又はC末端方向に伸長したペプチドであって、かつ癌抗原ペプチドとしての活性を有するものが挙げられる。
本発明の癌抗原ペプチドの活性測定法としては、例えばJ.Immunol.,154,p2257,1995に記載の方法が挙げられる。以下、本方法の概略につき、HLAの型がHLA−A24の場合を例にとり説明する。まず、HLA−A24抗原陽性のヒトから末梢血リンパ球を単離する。次に、この末梢血リンパ球に対してin vitroで本発明のペプチドを添加して刺激することにより、本発明のペプチドとHLA−A24との複合体の提示された抗原提示細胞を特異的に認識するCTL(細胞傷害性T細胞)を誘導する。
当該CTLの誘導は、例えば、抗原ペプチドとHLA−A24との複合体に反応してCTLが産生する種々のサイトカイン(例えばIFN−γ)の量を、例えばELISA法によって測定することにより、調べることができる。また、51CrやEuropiumで標識した抗原ペプチド提示細胞に対するCTLの傷害性を測定する方法(51Crリリースアッセイ、Int.J.Cancer.58,p317,1994、Europiumリリースアッセイ、J.Immunol.,154,p3991,1995)によっても調べることができる。さらに、後述の実施例を参考にして行うこともできる。
本発明はまた前記抗原を有効成分とする癌ワクチンに関する。このワクチンは、WT1遺伝子の発現レベルの上昇を伴う癌、例えば白血病、骨髄異形成症候群、多発性骨髄腫、悪性リンパ腫などの血液の癌、胃癌、大腸癌、肺癌、乳癌、胚細胞癌、肝癌、皮膚癌、膀胱癌、前立腺癌、子宮癌、子宮頸癌、卵巣癌等の固形癌の予防又は治療のために使用することができる。特にこのワクチンは、HLA−A24陽性の患者に適用し得るものである。このワクチンは、経口投与、又は非経口投与、例えば腹腔内投与、皮下投与、皮内投与、筋肉内投与、静脈内投与、鼻腔内投与等により投与することができる。
さらに、本発明のワクチンの投与方法として、患者の末梢血から単核球を集め、その中から樹状細胞を取り出し、本発明のペプチドをパルスして患者に皮下投与などで患者に戻す方法も行われる。
本方法は、細胞療法、あるいはDC(樹状細胞)療法とも呼ばれるものであり、詳しくは後述の「抗原提示細胞」の項を参照されたい。
ワクチンは、前記有効成分としての投与ペプチドのほかに、医薬として許容されるキャリアー、例えば適当なアジュバンド(Clin−Micobiol.Rev.,7,277−289,1994)、例えば水酸化アルミニウムのごとき鉱物ゲル;リソレシチン、プルロニックポリオールのごとき界面活性剤;ポリアニオン;ペプチド;又は油乳濁液を含むことができる。あるいは、リポゾーム中へ混合し、又は多糖及び/又はワクチン中に配合される他の集合体を含むことができる。投与量は一般に、1日当り0.1μg〜1mg/kgである。
本発明ではまた、上記のポリペプチドワクチンをコードするDNAもワクチン(DNAワクチン)として使用することができる。すなわち、本発明のWT1改変ペプチドをコードする核酸を含有する核酸、好ましくはDNAを、適切なベクター、好ましくは発現ベクターに挿入した後、動物に投与することにより、癌免疫を生じさせることができる。このようなDNAワクチンの具体的手法については、WO00/06602やJ.Immunol.,160,P1717,1998などを参照されたい。
本発明はまた、HLA抗原と上記ペプチドとの複合体の提示された抗原提示細胞に関する。実施例において、本発明のペプチド刺激により強いcell Killing活性が認められているが、これは、末梢血単核球中に、本発明のペプチドとHLA抗原(HLA−A24抗原)との複合体の提示された抗原提示細胞が存在し、そして、この抗原提示細胞を特異的に認識するCTL(細胞傷害性T細胞)が誘導された結果に他ならない。このような、HLA抗原と本発明のペプチドとの複合体の提示された抗原提示細胞は、以下に述べるような細胞療法(DC療法)において有効に用いられる。
細胞療法において用いられる抗原提示細胞は、腫瘍患者から抗原提示能を有する細胞を単離し、この細胞に本発明のペプチドを体外でパルスして、HLA抗原と本発明のペプチドとの複合体を細胞表面に提示させることにより作製される。ここで「抗原提示能を有する細胞」とは、本発明のペプチドを提示することの可能なHLA抗原を細胞表面に発現している細胞であれば特に限定されないが、抗原提示能が高いとされている樹状細胞が好ましい。
また、前記抗原提示能を有する細胞にパルスされる本発明のペプチドは、ペプチドの形態のみならず、当該ペプチドをコードするDNAやRNAの形態であっても良い。
本発明の抗原提示細胞の具体的な調製法としては、例えばCancer Immunol Immunother.,46:82,1998、J.Immunol.,158,p1796,1997、Cancer Rcs.,59,p1184,1999などを参考にすることができる。樹状細胞を用いる場合は、例えば、腫瘍患者の末梢血からフィコール法によりリンパ球を分離し、その後非付着細胞を除き、付着細胞をGM−CSFおよびIL−4存在下で培養して樹状細胞を誘導し、当該樹状細胞を本発明のペプチドと共に培養してパルスすることなどにより、本発明の抗原提示細胞を調製することができる。
また、前記抗原提示能を有する細胞に、本発明のペプチドをコードするDNAやRNAを導入することにより本発明の抗原提示細胞を調製する場合は、例えばDNAの場合はCancer Res,.56:p5672,1996やJ.Immunol.,161:p5607,1998などを参考にして行うことができ、またRNAの場合は、J.Exp.Med.,184:p465,1996などを参考にして行うことができる。
前記抗原提示細胞は、腫瘍の治療剤の有効成分とすることができる。その際、抗原提示細胞を安定に維持するために、生理食塩水、リン酸緩衝生理食塩水(PBS)、培地等を含むことが好ましい。投与方法としては、静脈内投与、皮下投与、皮内投与が挙げられる。
本発明はさらに、HLA抗原と上記ペプチドとの複合体を認識する細胞傷害性T細胞(CTL)に関する。本発明のCTLは、以下の養子免疫療法において有効に用いられる。
すなわちメラノーマにおいては、患者本人の腫瘍内浸潤T細胞を体外で大量に培養して、これを患者に戻す養子免疫療法に治療効果が認められている(J.Natl.Cancer.Inst,.86:1159,1994)。またマウスのメラノーマにおいては、脾細胞をイン・ビトロで腫瘍抗原ペプチドTRP−2で刺激し、腫瘍抗原ペプチドに特異的なCTLを増殖させ、該CTLをメラノーマ移植マウスに投与することにより、転移抑制が認められている(J.Exp.Med.,185:453,1997)。これは、抗原提示細胞のHLA抗原と腫瘍抗原ペプチドとの複合体を特異的に認識するCTLをイン・ビトロで増殖させた結果に基づくものである。従って、本発明のペプチドを用いて、イン・ビトロで患者末梢血リンパ球を刺激して腫瘍特異的CTLを増やした後、このCTLを患者に戻す治療法は有用であると考えられる。
このように本発明のCTLは、腫瘍の治療剤の有効成分とすることができる。その際、CTLを安定に維持するために、生理食塩水、リン酸緩衝生理食塩水(PBS)、培地等を含むことが好ましい。投与方法としては、静脈内投与、皮下投与、皮内投与が挙げられる。
実施例
次に、実施例により、本発明のペプチドが癌抗原及び癌ワクチンとして有用なことを、明らかにする。
実施例1.
HLA−A*2402を有するヒトの末梢血単核球を分離し、これを24ウエルプレートに2×106細胞/ウエルの量で分配し、これにWT1ワイルドペプチド又はWT1改変ペプチドを20μMの濃度になるように添加し、1週間培養した。この際の培地として、45%RPMI、45%AIV、10%FCS、1×非必須アミノ酸、SM/PCGを用いた。上記の培養の後、細胞を2×106細胞/ウエルに調製し、レスポンダー(responder)細胞とした。
他方、上記の同じHLA−A*2402のヒトから、別途、末梢血単核球を分離し、前記それぞれ一方のペプチド20μMと共に4時間培養してペプチドパルスし、次に30Gyの放射線照射した後、細胞を4×106細胞/ウエルに調製し、スチムレーター(stimulator)細胞とした。
上記のようにして調製したレスポンダー細胞とスチムレーター細胞を混合し、さらにIL−2を50U/mlの濃度で加えて1週間培養した。この結果、得られた細胞の状態は次の通りであった。
次に、51Crリリース法に従ってKilling assayを行った(J.Immunol.164:1873,2000)。標的細胞として、C1R2402細胞、及び上記のペプチドでパルスしたC1R2402細胞を用い、これらのそれぞれの標的細胞(T)に、上記の通りにWT1ワイルドペプチド又はWT1改変ペプチドにより刺激した細胞(エフェクター細胞)(E)を、E:T比1、5又は20において作用させ、細胞溶解を測定した。結果を図1に示す。この図から明らかな通り、WT1ワイルドペプチドにより刺激した細胞に比べてWT1改変ペプチドで刺激した細胞の方が強いcell killing活性を示した。
実施例2.
内因性(endogeneous)にWT1抗原を発現する白血病細胞(標的細胞)に対する、WT1ワイルドペプチド又はWT1改変ペプチドにより刺激したエフェクター細胞のcell killing活性を51Crリリース法により試験した。標的細胞としてWT1+/A*2402+細胞(#1AML患者の白血病細胞)、WT1−/A*2402+細胞(#2AML患者の白血病細胞)、WT1+/A*2402−細胞(#3AML患者の白血病細胞)、及びWT1−/A*2402−細胞(#4AML患者の白血病細胞)を用いた。
実施例1で調製したエフェクター細胞(E)と上記の標的細胞(T)とを、E/T比20:1で混合し、4時間培養し、細胞溶解の程度を測定した。結果を図2に示す。
この図から明らかな通り、WT1ワイルドペプチド又はWT1改変ペプチドにより刺激された細胞のいずれもWT1+/A*2402細胞に対して細胞毒性活性を示したが、その活性はWT1改変ペプチドの方が高かった。
実施例3.
実施例1と同様の実験を別のHLA−A*2402陽性の健常人の末梢血単核球から調製したエフェクター細胞を用いて試験した。結果を図3に示す。
この図から明らか通り、実施例1と同様にWT1ワイルドペプチドにより刺激した細胞に比べてWT1改変ペプチドにより刺激された細胞の方が強い細胞傷害活性を示した。
実施例4.
WT1ワイルドペプチド又はWT1改変ペプチドで刺激したエフェクター細胞の内因性(endogeneous)にWT1抗原を発現する肺癌由来の癌細胞株(標的細胞)に対する細胞傷害活性を51Crリリース法に従って試験した。標的細胞としてRERF−LCAI(WT1+/A*2402+)、LC1sq(WT1+/A*2402+)、11−18(WT1−/A*2402+)、LK87(WT1+/A*2402−)を用いた。
実施例1と同様な方法により調製したエフェクター細胞(E)と51Crで標識した上記の標的細胞(T)とを、実施例2と同様にE/T比20:1で混合して4時間培養し、細胞溶解の程度を測定した。結果を図4に示す。
この図から明らかな通り、WT1ワイルドペプチド又はWT1改変ペプチドにより刺激された細胞のいずれもWT1+/A*2402+の標的細胞に対してのみ細胞傷害活性を示したが、その活性はWT1改変ペプチドの方が高かった。
実施例5.
WT1ワイルドペプチド又はWT1改変ペプチドで刺激したエフェクター細胞がHLAクラスIに拘束性のCD8陽性キラー細胞であることを抗体を用いたブロッキングアッセイにより確かめた。抗体としては、抗HLAクラスI抗体、抗HLAクラスII抗体、抗CD8抗体を用いた。実施例1と同様な方法により調製したエフェクター細胞(E)、51Crで標識したC1R2402又はWT1ワイルドペプチドをパルスしたC1R2402細胞を標的細胞(T)とし、E/T比20:1で抗体とともに混合して4時間培養し、51Crリリース法に従って細胞溶解の程度を測定した。結果を図5に示す。
この図から明らかな通り、WT1ワイルドペプチド又はWT1改変ペプチドにより刺激された細胞のいずれも抗HLAクラスI抗体及び抗CD8抗体により細胞傷害活性が阻害されており、細胞傷害活性を示す細胞は、HLAクラスIに拘束性のCD8陽性キラー細胞であることが示された。
実施例6.
WT1改変ペプチドとWT1ワイルドペプチドのHLA−A*2402への結合親和性を調べた。C1RA2402細胞を酸緩衝液(131mM クエン酸、66mMリン酸ナトリウム、290m osmol、pH3.3)で1分間処理した後、0.5%のウシ血清アルブミンを含むDMEM培養液を加えて中和した。細胞は、培養液で洗浄した後、200nMのβ2−マイクログロブリン(シグマ社)と0.5%のウシ血清アルブミンを含むDMEM培養液で2×106細胞/mlの濃度に懸濁した。15μlの細胞懸濁液を各種濃度のWT1ペプチドを含む50μlの培養液と混合し、室温で4時間インキュベートした。細胞は洗浄後、FITCで標識したHLA−A24に対するモノクローナル抗体(クローン名7A12)で染色し、フローサイトメーターFACSでHLA−A24発現量を解析した。同様の操作をHLA−A*2402に結合することが報告されているメラノーマ抗原のpmel 15の抗原ペプチド(Ala Tyr Gly Leu Asp Phe Tyr Ile Leu)(配列番号:4)(J.Immunol.,154:5994,1995)についても行い、これをスタンダードとして、文献(Immunogenetics,51:816,2000)に記載の方法によりWT1ペプチドの解離定数(Kd)を 算出した。結果を表2に示す。
この表から明らかな通り、WT1改変ペプチドはWT1ワイルドペプチドよりもHLA−A*2402への結合親和性が強かった。
産業上の利用可能性
上記の結果から、本発明のペプチドは確かに癌抗原として機能し、癌細胞に対するキラーT−細胞(癌細胞傷害性T細胞)を誘導増殖させたことが立証された。従って、本発明の癌抗原ペプチドは、WT1遺伝子の発現の上昇を伴う白血病及び固形癌に対する癌ワクチンとして有用である。

- 配列番号:3に示すアミノ酸配列を含む9〜12個のアミノ酸から成る、請求項1に記載の癌抗原ペプチド。
