本発明は、TCRα鎖タンパク質とTCRβ鎖タンパク質とからなるTCR複合体を提供する。本発明のTCR複合体の典型的な特徴は、HLA-A*24:02に拘束された野生型WT1ペプチドおよびHLA-A*24:02に拘束された変異型WT1ペプチドに対する認識である。
本発明は、また、上記TCRα鎖タンパク質をコードするDNA、および上記TCRβ鎖タンパク質をコードするDNAを提供する。これらDNAを発現ベクターに組み込んで細胞に導入することにより、上記TCRα鎖タンパク質、上記TCRβ鎖タンパク質、またはこれらタンパク質からなるTCR複合体を、細胞において発現させることができる。これらタンパク質を細胞に発現させるためのベクターとしては、本実施例に示したベクターを用いることができるが、それに制限されない。例えば、2種類の遺伝子をIRESを介して単一mRNAから翻訳可能なベクターpIRES(TaKaRa Bio社)やMammalian PowerExpress System(東洋紡)のベクターなどを利用することもできる。ベクターは、上記TCRα鎖タンパク質をコードするDNAと上記TCRβ鎖タンパク質をコードするDNAのいずれかを発現可能に含有するベクターであってもよく、両者を発現可能に含有するベクターであってもよい。上記ベクターを導入する細胞としては特に制限はなく、目的に応じて種々の細胞を用いることができる。細胞への遺伝子導入は、エレクトロポレーション法など当業者に公知の方法で行うことができる。
また、本発明は、上記TCRα鎖タンパク質に特異的に結合する抗体、上記TCRβ鎖タンパク質に特異的に結合する抗体、およびこれらタンパク質からなるTCR複合体に特異的に結合する抗体を提供する。
本発明は、また、上記TCR複合体を結合して多量体化した分子を提供する。当該分子の調製は、例えば、次の通り、行うことができる。TCRの細胞外領域をコードするDNAを発現ベクターに組み込み、人工的にTCRα鎖およびβ鎖の組み換えタンパク質を作製し、TCRα鎖あるいはβ鎖のC末端を酵素反応を利用してビオチン化する。ビオチン化されたTCR複合体をカラムクロマトグラフィー法で精製し、これをアビジンと反応させることで多量体化した分子を調製できる。アビジンを予めFITC(fluorescein isothiocyanate)、PE(Phycoerythrin)あるいはAPC(allophycocyanin)などの蛍光物質で標識することでフローサイトメーターや蛍光顕微鏡を用いて、特異的なMHC/ペプチド複合体を発現する細胞の検出が可能である。scTCRは、TCRα鎖およびβ鎖の細胞外領域を短鎖ペプチドリンカーを利用して連結させ一本のタンパク質として発現させることで調製できる。TCRテトラマーと同様にC末端側をビオチン化して多量体化することも可能であり、また、IgGの可変領域に組み込んで多量体化することも可能である。
本発明は、また、HLA-A*24:02に拘束された野生型WT1ペプチドまたはHLA-A*24:02に拘束された変異型WT1ペプチドを検出するためのキットであって、以下の(a)から(h)の少なくとも1つの構成要素を含むキットを提供する。

ここで「HLA-A*24:02に拘束された野生型WT1ペプチド」および「HLA-A*24:02に拘束された変異型WT1ペプチド」とは、上記の通り、これらペプチドが細胞表面上に存在する場合、および単離もしくは精製された分子(例えば、多量体化されたTCR複合体にこれらペプチドが拘束された分子)として存在する場合の双方を含む意である。本発明のキットにおいては、さらに、当該キットの使用説明書が含まれていてもよい。
本発明は、また、HLA-A*24:02に拘束された野生型WT1ペプチドを結合して多量体化した分子またはHLA-A*24:02に拘束された変異型WT1ペプチドを結合して多量体化した分子の品質管理方法であって、当該分子と上記本発明の形質転換細胞との反応性を確認する工程を含む方法を提供する。本発明の品質管理方法における反応性の確認は、例えば、本実施例8や9に記載の方法で実施することができる。その結果、反応性が維持されている場合には、当該分子の品質が維持されていると評価することができ、一方、反応性が低下した場合には、当該分子の品質が劣化したと評価することができる。なお、反応性の維持は、例えば、陽性率の維持および/またはMFI(平均蛍光強度、mean fluorescence intensity)の維持を指標に判定することができる。また、いずれかの指標の低下で、その品質の欠陥点を推認し、改善策を立てることができる。
本発明者らは、2003年に報告されたKaraniksらの論文(J.Immunol., 2003; 171: 4898-4904)を参考に、限界希釈条件下で効率よく癌抗原特異的CTLを増殖培養するMLPC法(mixed lymphocyte-peptide cultures)を様々な癌抗原由来ペプチドを用いて検証と改良を繰り返し、癌抗原特異的CTLの誘導法として利用してきた。本発明者らは、癌抗原特異的CTLの存在比率は、健常人由来の末梢血単核球(PBMC) 1×107個中に1個未満であることを経験的に見出している。例えば96-ウェルプレートの1個のウェルに5×105個のPBMCを添加してCTLを誘導した場合、96個のウェルの内、最大でも5個未満のウェルで特異的CTLの誘導が確認される。従って、この条件は限界希釈条件と言える。MLPC法はPBMCに抗原ペプチドを添加して培養するだけの方法であり、血液提供者の体内に存在するメモリーT細胞、あるいはメモリー/エフェクターT細胞を刺激して増殖させていると考えられる。従って試験管内で調製した抗原提示細胞を利用した場合に心配されるような、ナイーブT細胞の人為的なプライミングによる人為的なT細胞を刺激増殖してしまうリスクは低いと考えられる。本発明者らは、HLA-A*24:02 WT1ペプチド特異的CTL lineの誘導を、次の通り、本発明者らが経験的に見出している限界希釈条件下で行った。
37F8にはHLA-A*24:02 WT1テトラマー試薬と反応するTCRを有する細胞が97%の比率で存在することが明らかとなったが、HLA-A*24:02 WT1テトラマー試薬は人工的に合成されたHLA-A*24:02とβ2-ミクログロブリンとWT1ペプチドの3者複合体で構成されているため、実際の細胞表面上に発現している3者複合体をこのTCRが認識できるか確認する必要がある。さらにこのTCRを有するCTLが、TCRと3者複合体との結合により活性され、3者複合体を発現している細胞に対して細胞傷害性活性を発揮できるかどうかは、37F8由来のTCRのα鎖とβ鎖を人為的に導入発現させたリンパ球が生体内で機能するかを推測する上で重要である。そこで、HLA-A*24:02 WT1テトラマー試薬陽性細胞集団として検出可能なHLA-A*24:02拘束性WT1ペプチド特異的なCTL細胞集団(37F8)の細胞傷害性活性を測定した。
本発明者らは、遺伝子を取得する過程でPCRを繰り返し行うことによる予測不可能な変異の挿入を避けるために、遺伝子情報を抽出する前に取得可能な情報を得ておくべきと考えた。この目的のため、TCRβ鎖のVβ領域を特異的に認識することが可能な抗体を用いたフローサイトメトリーにより24種類のVβ領域をレパトア分析することが可能なキット(IOTest beta Mark TCR Vβ Repertoire Kit、Beckman Coulter社)を利用した。このキットに含まれているTCRVβ領域に特異的な抗体は、Vβ1、Vβ2、Vβ3、Vβ4、Vβ5.1、Vβ5.2、Vβ5.3、Vβ7.1、Vβ7.2、Vβ8、Vβ9、Vβ11、Vβ12、Vβ13.1、Vβ13.2、Vβ13.6、Vβ14、Vβ16、Vβ17、Vβ18、Vβ20、Vβ21.3、Vβ22、Vβ23の24種類である。このキットはTCRβ鎖のVβ領域の約70%を検出可能であり、TCRβ鎖のVβ領域のみの情報が判別できる。
−37F8からのHLA-A*24:02 WT1テトラマー試薬陽性細胞の単離−
37F8は図1Aに示した通り、約97%の細胞集団がHLA-A*24:02 WT1テトラマー試薬に対して反応性を示す。図3に示したTCRVβ鎖のレパトア解析の結果、大多数の細胞はVβ5.1であるが、極少量のVβ5.3とVβ11を有する細胞が含まれている。Vβ5.3とVβ11を有する細胞はその存在比率から明らかに目的とするTCRを有する細胞集団ではなく、これらの細胞からの遺伝子情報が混入すれば、TCRのα鎖とβ鎖の正しい組合せを決定する上で大きな障害となる。特にPCR法では、プライマーの設計により、遺伝子増幅効率が大きく影響を受け、増幅効率の高いPCR産物が必ずしも鋳型中のcDNA量を反映しているとはいえない。特に多様性に富んだTCR遺伝子全長を取得するために設計するプライマーの設計自由度は、通常の定量的PCR用に供されるプライマーの設計自由度と比較して著しく低い。そこで、PCRを行う前に、37F8に含まれる約3%のHLA-A*24:02 WT1テトラマー試薬と反応しない細胞集団を排除することとした。
本発明者らは、IMGTに登録されているTCRの遺伝子情報を網羅的に分析し、全てのTCRのα鎖とβ鎖の全長をクローニングできるように多数のプライマーを設計した。この方法の優位点は1回のPCRで全長配列が取得できることである。この方法では、PCR産物を基に得られたDNA配列情報に従ってプライマーを設計する必要が無いため、変異が含まれる可能性は最小限にとどめられている。しかしながら、プライマーの組合せが膨大になり、少ないcDNAを用いて検証できるPCRの回数は限られている。そこで、PCR反応をお互いに阻害しないことを確認したプライマーを10〜11種類混合させたプライマーミックスを用いてPCRを実施し、遺伝子産物が得られたプライマーミックスの全ての組合せでPCRを実施して、全長配列が得られるように工夫した。プライマーミックスの組合せとPCRの反応温度条件の設定は、既にTCRのレパトアが報告されているJurkat細胞、未処理のヒトPBMC、および特定のVβ鎖を発現するT細胞の3種類の細胞群から調製したcDNAを用いて行った。特定のVβ鎖を発現するT細胞は、ヒトPBMCよりVβ鎖特異的抗体を用いて自動磁気分離装置で分離濃縮して調製した。
37F8由来の2種類のTCRα鎖(A12-3とA41)と1種類のβ鎖(B5-1)の正しい組合せを解明する為に、哺乳動物細胞用発現ベクターであるpcDNA3.1(Invitrogen社)とpEF6/Myc-His(Invitrogen社)にcDNAをサブクローニングした。コントロールとして、HLA-A*02:01 Mart-1特異的TCRのα鎖およびβ鎖のcDNAを人工合成し、同様に発現ベクターを構築した(J. Immunol., 2008; 181: 1063-1070)。
陽性像が得られた細胞集団WT1 TCR A12-3/B5-1に、各ベクターの耐性薬剤であるG418(Roche社)と、Blastisidin(Invitrogen社)を添加し、薬剤耐性のTCR遺伝子発現形質転換細胞株SK37を樹立した。SK37のMHCテトラマー試薬に対する反応性を検証した結果を図8に示した。X軸にFITC標識抗CD8抗体の染色強度(logスケール)を、Y軸にPE標識MHCテトラマー試薬の染色強度(logスケール)を示した。各ドットプロット上部に使用したMHCテトラマー試薬の種類を示した。各ドットプロットを四分割した右上に存在する抗CD8抗体陽性かつMHCテトラマー試薬陽性細胞の生細胞中での存在率(%)を示した。
SK37細胞を用いたHLA-A*24:02 WT1テトラマー試薬の評価の正確性を確認する目的で添加回収試験を行った。実験方法は、SK37とTCR遺伝子を導入していない親株細胞を混合し、HLA-A*24:02 WT1テトラマー試薬で染色後、その陽性率と混合率から期待される陽性率とを比較した。培養中のSK37と親細胞株から20μLの細胞浮遊液を分取し、20μLのTrypan Blue Stain 0.4%(Life Technologies社)を加え血球計算盤にて生細胞数をカウントした。SK37と親細胞株を混合し、SK37の存在比率が100%、50%、25%、15%、10%、5%、2.5%、1%、0%の細胞集団を調整した。各細胞集団から5×105個細胞をエッペンドルフチューブに分取し、400×gで5分間遠心処理後、上清を注意深く廃棄した。1mLのFCMバッファーを添加し、再懸濁後、400×gで5分間遠心し、上清を注意深く廃棄した。20μLのFCMバッファーと10μLのClear Back Human FcR blocking reagentを加え良く攪拌後、室温にて5分間反応させた。10μLのHLA-A*24:02 WT1(野生型)テトラマー試薬あるいはHLA-A*24:02 WT1(変異型)テトラマー試薬を加え穏やかに攪拌後、冷蔵室で30分間反応させた。10μLのCD8(clone T8)-FITCを加え、冷蔵室で20分間反応させた。適量のFCMバッファーを加え400×gで5分間遠心した。上清を注意深く捨て、7-AADを1%添加したFCMバッファーを400μL加えて細胞を懸濁し、フローサイトメーターで細胞を取込み分析した。FCS/SSCドットプロット展開図中で選択した領域をR1とし、FCS/7-AADドットプロット展開図での生細胞領域(即ち、7-AAD陰性細胞集団)をR2として、「R1かつR2」の細胞集団でデータの解析を行った。
フローサイトメーターで使用するMHCテトラマー試薬の保存安定性を確認するためには、ポジティブコントロール細胞が必須である。ポジティブコントロール細胞は、常に試薬に対して同一の反応性を示すことが重要である。SK37のようにHLA-A*24:02 WT1テトラマー試薬が結合するTCR遺伝子が導入された安定的形質転換細胞は理想的なポジティブコントロール細胞といえる。本発明者らはSK37を利用して、MHC WT1テトラマー試薬の保存安定性試験の実施方法を検証した。MHCテトラマー試薬の保存安定性は、フローサイトメトリーで期待される陽性率が保たれる期間として評価できる。SK37の陽性率を5〜15%に調整し、定期的に試薬の希釈系列により反応性を確認し、HLA-A*24:02 WT1(野生型)テトラマー試薬陽性細胞とHLA-A*24:02 WT1(変異型)テトラマー試薬陽性細胞の陽性率とMFIの比を分析することで保存安定性の検証を行った。
HLA-A*24:02 WT1(野生型)テトラマー試薬は、使用しているWT1(野生型)ペプチドのHLAに対する結合性が低いため、製造後2日間以内に沈殿物が確認される。この不安定性は試薬をHPLCで分析した場合に、MHCテトラマー試薬の有効成分を示すピーク面積の減少から把握することも可能である。しかしながら、HPLCの分析データとフローサイトメーターを用いた染色性データとの関連性は、ポジティブコントロール細胞が存在しなかったため、長い間不明であった。そこで、HLA-A*24:02 WT1(野生型)テトラマー試薬を使用する直前に調整できる用時調製キットを開発し、用事調製後に2〜8℃に冷蔵保存した場合の経時的な反応性の変化を、SK37を用いて検証した。
本実施例では、実施例5で示したJurkat細胞亜株にHLA-A*24:02 WT1特異的なTCRを安定的に発現させた細胞株(以下、「JSK37」と称する)を利用し、該細胞におけるIL-2の産生を指標に、細胞表面のHLAに目的ペプチドが提示されていることを検査することが可能かを検証した。なお、Jurkat細胞は、これまでに、抗CD3抗体とPMAの刺激によりIL-2を産生することが報告されている(J. Immunology, 1984; 133: 1123-1128)。

- 配列番号:1に記載のアミノ酸配列からなるCDR1、配列番号:2に記載のアミノ酸配列からなるCDR2、および配列番号:3に記載のアミノ酸配列からなるCDR3を有するT細胞レセプターα鎖タンパク質と配列番号:5に記載のアミノ酸配列からなるCDR1、配列番号:6に記載のアミノ酸配列からなるCDR2、および配列番号:7に記載のアミノ酸配列からなるCDR3を有するT細胞レセプターβ鎖タンパク質とからなる、T細胞レセプター複合体。
- 配列番号:4に記載のアミノ酸配列を有するT細胞レセプターα鎖タンパク質と配列番号:8に記載のアミノ酸配列を有するT細胞レセプターβ鎖タンパク質とからなる、請求項1に記載のT細胞レセプター複合体。
- 配列番号:14に記載のアミノ酸配列を有するT細胞レセプターα鎖タンパク質と配列番号:15に記載のアミノ酸配列を有するT細胞レセプターβ鎖タンパク質とからなる、請求項1に記載のT細胞レセプター複合体。
- HLA-A*24:02に拘束された野生型WT1ペプチドおよびHLA-A*24:02に拘束された変異型WT1ペプチドを認識する、請求項1から3のいずれかに記載のT細胞レセプター複合体。
- 以下の(a)から(c)のいずれかに記載の組み合わせからなるDNA。
(a)配列番号:1に記載のアミノ酸配列からなるCDR1、配列番号:2に記載のアミノ酸配列からなるCDR2、および配列番号:3に記載のアミノ酸配列からなるCDR3を有するT細胞レセプターα鎖タンパク質をコードするDNAと配列番号:5に記載のアミノ酸配列からなるCDR1、配列番号:6に記載のアミノ酸配列からなるCDR2、および配列番号:7に記載のアミノ酸配列からなるCDR3を有するT細胞レセプターβ鎖タンパク質をコードするDNAとの組合せ
(b)配列番号:4に記載のアミノ酸配列を有するT細胞レセプターα鎖タンパク質をコードするDNAと配列番号:8に記載のアミノ酸配列を有するT細胞レセプターβ鎖タンパク質をコードするDNAとの組合せ

- 以下の(a)から(c)のいずれかに記載の組み合わせからなるベクター。
(a)配列番号:1に記載のアミノ酸配列からなるCDR1、配列番号:2に記載のアミノ酸配列からなるCDR2、および配列番号:3に記載のアミノ酸配列からなるCDR3を有するT細胞レセプターα鎖タンパク質をコードするDNAを発現可能に含有するベクターと配列番号:5に記載のアミノ酸配列からなるCDR1、配列番号:6に記載のアミノ酸配列からなるCDR2、および配列番号:7に記載のアミノ酸配列からなるCDR3を有するT細胞レセプターβ鎖タンパク質をコードするDNAを発現可能に含有するベクターとの組合せ
(b)配列番号:4に記載のアミノ酸配列を有するT細胞レセプターα鎖タンパク質をコードするDNAを発現可能に含有するベクターと配列番号:8に記載のアミノ酸配列を有するT細胞レセプターβ鎖タンパク質をコードするDNAを発現可能に含有するベクターとの組合せ
(c)配列番号:14に記載のアミノ酸配列を有するT細胞レセプターα鎖タンパク質をコードするDNAを発現可能に含有するベクターと配列番号:15に記載のアミノ酸配列を有するT細胞レセプターβ鎖タンパク質をコードするDNAを発現可能に含有するベクターとの組合せ - 請求項5に記載のDNAまたは請求項6もしくは7に記載のベクターが導入された形質転換細胞。
- 請求項5に記載のDNAまたは請求項6もしくは7に記載のベクターが導入された形質転換細胞であって、HLA-A*24:02に拘束された野生型WT1ペプチドを結合して多量体化した分子およびHLA-A*24:02に拘束された変異型WT1ペプチドを結合して多量体化した分子によって検出することができる形質転換細胞。
- リンパ球である、請求項8または9に記載の形質転換細胞。
- 請求項10に記載の形質転換細胞を有効成分とする、WT1陽性の癌を治療するための医薬組成物。
- 請求項1から4のいずれかに記載のT細胞レセプター複合体の細胞外領域を結合して多量体化した分子。
- HLA-A*24:02に拘束された野生型WT1ペプチドまたはHLA-A*24:02に拘束された変異型WT1ペプチドを検出または捕捉するための薬剤であって、請求項12に記載の分子を含む薬剤。
- HLA-A*24:02に拘束された野生型WT1ペプチドまたはHLA-A*24:02に拘束された変異型WT1ペプチドを検出するためのキットであって、以下の(a)から(e)の少なくとも1つの構成要素を含むキット。
(a)請求項1から4のいずれかに記載のT細胞レセプター複合体
(b)請求項5に記載のDNA
(c)請求項6または7に記載のベクター
(d)請求項8から10のいずれかに記載の形質転換細胞
(e)請求項12に記載の分子 - HLA-A*24:02に拘束された野生型WT1ペプチドを結合して多量体化した分子またはHLA-A*24:02に拘束された変異型WT1ペプチドを結合して多量体化した分子の品質管理方法であって、該分子と請求項8から10のいずれかに記載の形質転換細胞との反応性を確認する工程を含む方法。
