Claims (1)
- 【特許請求の範囲】 1.植物細胞におけるウィルス性単一サブユニット RNAポリメラーゼの発現の ための核発現構造体であって、 転写の5′から3′方向に次の操作可能的に連結された成分: 植物細胞核において機能的なプロモーター、前記ウィルス性 RNAポリメラーゼ を植物プラスチドオルガネラ中に運ぶことができるトランジットペプチドのため のコード配列、前記ウィルス性 RNAポリメラーゼのためのコード配列及び植物細 胞核において機能的な転写終結領域を含んで成り、ここで前記トランジットペプ チド及び RNAポリメラーゼコード配列が同じ翻訳読み取り枠に存在することを特 徴とする構造体。 2.前記ポリメラーゼが、コリファージT7 RNAポリメラーゼ、コリファージT3 RNAポリメラーゼ、サルモネラファージ SP6 RNAポリメラーゼ、プソイドモナス ファージgh-1 RNAポリメラーゼ、クレブシエラファージ K11 RNAポリメラーゼ、 シトロバクターファージViIII RNAポリメラーゼ、及びセラチアファージIV RNA ポリメラーゼから成る群から選択される請求の範囲第1項記載の構造体。 3.前記プロモーターが構成的な発現を提供する請求の範囲第1項記載の構造 体。 4.前記プロモーターが組織選択性又は発生的に調節された発現を提供する請 求の範囲第1項記載の構造体。 5.前記プロモーターが植物種子組織において選択的に発現される遺伝子から である請求の範囲第4項記載の構造体。 6.前記プロモーターが植物スターチ貯蔵器官において選択的に発現される遺 伝子からである請求の範囲第4項記載の構造体。 7.前記プロモーターが植物花組織において選択的に発現される 遺伝子からである請求の範囲第4項記載の構造体。 8.前記プロモーターが植物果実組織において選択的に発現される遺伝子から である請求の範囲第4項記載の構造体。 9.植物プラスチドオルガネラの形質転換及び形質転換されたプラスミドを含 んで成る植物細胞の選択のためのポリヌクレオチドであって、前記プラスチドの ゲノムに対して相同性の DNA領域、及びマーカー遺伝子を発現するプラスチドオ ルガネラを含んで成る植物細胞の選択のための前記マーカー遺伝子の発現のため の構造体の他に、 転写の5′から3′の配向に次の操作可能的に連結された成分:ウィルス性サ ブユニット RNAポリメラーゼ特異的プロモーター、対象の DNA配列及び前記プロ モーターからの転写を終結できる転写終結領域を含んで成るポリヌクレオチド。 10.前記プロモーター及び前記転写終結領域が同じウィルスゲノムからである 請求の範囲第9項記載のポリヌクレオチド。 11.前記プロモーターが、コリファージT7 RNAポリメラーゼ、コリファージT3 RNAポリメラーゼ、サルモネラファージSP6 RNAポリメラーゼ、プソイドモナス ファージgh-1 RNAポリメラーゼ、クレブシエラファージK11 RNAポリメラーゼ、 シトロバクターファージViIII RNAポリメラーゼ、及びセラチアファージIV RNA ポリメラーゼから成る群から選択されたウィルス性単一サブユニットポリメラー ゼに対して特異的である請求の範囲第9項記載のポリヌクレオチド。 12.前記対象の DNA配列が前記プラスチドオルガネラに生来のmRNAに対して相 補的な配列の転写のために向けられている請求の範囲第9項記載のポリヌクレオ チド。 13.前記対象の DNA配列がセンス配向でタンパク質コード配列の発現を提供す る請求の範囲第9項記載のポリヌクレオチド。 14.前記対象の DNA配列が2又はそれ以上の構造遺伝子の発現のためのオペロ ンを含んで成る請求の範囲第9項記載のポリヌクレオチド。 15.請求の範囲第1〜8のいづれか1項記載の核発現構造体を含んで成る植物 又は植物細胞。 16.請求の範囲第9〜14のいづれか1項記載のポリヌクレオチドを含んで成る 植物又は植物細胞。 17.請求の範囲第1〜8のいづれか1項記載の核発現構造体及び請求の範囲第 9〜14のいづれか1項記載のポリヌクレオチドを含んで成る植物細胞であって、 前記核発現構造体によりコードされる前記ウィルス性単一サブユニット RNAポリ メラーゼが前記ポリヌクレオチドの前記ウィルス性単一サブユニット RNAポリメ ラーゼ特異的プロモーターに対して特異的であることを特徴とする植物細胞。 18.植物プラスチドオルガネラに対象の DNA配列の転写を提供するための方法 であって、 次の細胞を含んで成る植物を成長せしめることを含んで成り、ここで前記植物 細胞中の核が、転写の5′から3′の方向に次の操作可能的に連結された成分: 前記核において機能的なプロモーター、プラスチドトランジットペプチドのた めのコード配列、ウィルス性単一サブユニット RNAポリメラーゼのためのコード 配列、及び前記核において機能的な転写終結領域を含んで成る DNA構造体を含ん で成り、ここで前記トランジットペプチドコード配列及び前記 RNAポリメラーゼ コード配列が同じ翻訳読み取り枠において存在し、そして 前記プラスチドオルガネラのゲノムが、転写の5′から3′の配向に次の操作 可能的に連結された成分:前記ウィルス性単一サブユニット RNAポリメラーゼに 対して特異的なプロモーター、前記対象 の DNA配列、及び転写終結領域を含んで成る DNA構造体を含んで成ることを特徴 とする方法。 19.前記ウィルス性単一サブユニット RNAポリメラーゼが、コリファージT7 R NAポリメラーゼ、コリファージT3 RNAポリメラーゼ、サルモネラファージSP6 RN Aポリメラーゼ、プソイドモナスファージgh-1 RNAポリメラーゼ、クレブシエラ ファージ K11 RNAポリメラーゼ、シトロバクターファージViIII RNAポリメラー ゼ、及びセラチアファージIV RNAポリメラーゼから成る群から選択される請求の 範囲第18項記載の方法。 20.前記核において機能的なプロモーターが、組織選択性又は発生的に調節さ れた発現を提供する請求の範囲第18項記載の方法。 21.前記核において機能的なプロモーターが、構成的な発現を提供する請求の 範囲第18項記載の方法。
